Background: In 2017, large-scale flowering, seeding, and dying events of dwarf bamboo (Sasa borealis) occurred in a wide range in central Japan for the first time in 120 years. This phenomenon of S. borealis, like the mast seeding of trees, could be expected to provide a large amount of food for seed-eating rodents and gradually affect their ecology and population dynamics. We captured rodents in survey plots in a secondary broad-leaved forest with the phenomenon from 2018 to 2019 to investigate the species, the number of individuals, growing stage, sex, and body mass. In addition, we also compared the capture data for 2 years (2018–2019) with that for the mast seeding year (2017) and 6 years before it (2011–2016). Results: The mast seeding of S. borealisgreatly increased the population size of rodents, especially Apodemus speciosusand A. argenteus. Conversely, Eothenomys smithiidid not show such an increase. Most of the captured rodent individuals were already adults at the time of new capture, and the proportion of male juveniles was extremely low. These results suggest that the mast seeding of S. borealiscreated unusually rich food availability for the population concentration of rodents due to their immigration. However, body mass was not significantly different before and after the mast seeding. In addition, the increased populations of the two Apodemusspecies did not decline 2 years after the S. borealismasting, contrary to the previously reported decrease of rodent populations after tree masting. Conclusions: Our results indicate that the mast seeding of S. borealisaffected the population dynamics of Apodemusspecies over a long time and their individual composition of different growth stages. However, it did not affect their body mass.

Pittosporum tobiraseedlings were inoculated with Fusarium solaniisolated from Scolytogenes birosimensis, a bark beetle that infects P. tobira.The fungus produced necrotic lesions in the phloem and a discolored zone in the sapwood. Seedlings inoculated with the fungus and grown under weak light conditions were killed within 12–30 days after inoculation. Isolates from both S. birosimensis, which has caused P. tobiradecline in Aichi and Fukuoka prefectures, and from the beetle under endemic conditions in Ibaraki Prefecture were both pathogenic to P. tobira.

Abstract: To clarify the effects of asynchronous seed production among tree species on the population of seed predators, we investigated the relationship between the annual variation in production of mature acorns and the insect damage in those acorns of two sympatric oak species, Quercus variabilis Blume and Quercus serrata Thunb. ex Murray, over 4 years at two study sites. The annual variation in acorn production was noticeable, with a coefficient of variation (CV) at the two sites of 1.05 and 0.80 for Q. variabilis and 0.87 and 0.73 for Q. serrata. Annual fluctuation in acorn production by Q. serrata was synchronized between the two sites. Since annual fluctuation in acorn production was not synchronized between the two species, the CVs for the total acorn production by both oak species (0.83 and 0.62 at the two sites) were lower than those for Q. variabilis and Q. serrata alone. The rate of predation by the specialist predators (Curculio weevils) on the acorns of both species was not related to the annual acorn crop size. Prolonged diapause of Curculio weevils might stabilize their populations. The rate of acorn predation by the generalist predators (tortricid moths) was also not related to the annual crop size. Asynchronous acorn production by the two oak species would help to stabilize the population.

The rates of hypocotyl and radicle survival and of germination success were investigated in mature acorns ofQuercus variabilisBlume in relation to endosperm loss due to seed insects. The acorns were damaged by curculio weevils (Coleoptera: Curculionidae) and moths, including tortricid moths (Lepidoptera: Tortricidae); the former were more abundant than the latter. Acorns damaged by curculio weevils showed a significantly lower germination rate when there was a large endosperm loss than the rate for sound acorns. The survival rate of the hypocotyl and radicle also decreased as endosperm loss increased. These results suggest that acorn germination is directly inhibited by damage to the hypocotyl and radicle, which depends on the amount of endosperm eaten by the weevils. However, the germination rate of acorns with hypocotyl and radicle was consistently high, irrespective of the degree of endosperm loss, which suggests that weevil-damaged acorns probably germinate, provided the hypocotyl and radicle survive until the cessation of damage.

Abstract: To confirm the pathogenicity of a blue stain fungus,Ceratocystis piceae (Mnch) Bakshi to the Japanese red pine (Pinus densiflora Sieb. et Zucc.), the responses of healthy young pine trees and stressed trees which were girdled by the half-circumferential girdling technique were investigated by the fungal inoculation test. Although neither of the pine trees inoculated withC. piceae in the non-girdled treatment nor the controls died, mortality of the trees girdled and inoculated withC. piceae was 28.6%. In the pine trees inoculated withC. piceae, the mean area of the necrotic lesion of the sapwood was significantly larger than that of the controls, and the mean of the water pressure potential of the xylem decreased, regardless of the girdling treatment. TheC. piceae was reisolated from the wood pieces near the inoculation points on the inoculated trees, but not from the controls. These results suggest that under strongly stressed conditions, the Japanese red pine trees might have been killed by heavy infestations ofC. piceae carried by bark beetles.

Abstract: The association between blue stain fungi andIps cembrae Heer (Coleoptera: Scolytidae) was investigated in the Japanese larch,Larix leptolepis Gordon, in the Nagoya University Forest, Aichi Prefecture, central Japan.I. cembrae had one or two generation(s) in a year in this study area. Two blue stain fungi,Ophiostoma piceae andLeptographium sp., were isolated from the body surface of both male adults in mating chambers and female adults in parent galleries, suggesting that this beetle species was a vector of these fungi. Although no blue stain fungi were isolated from non-stained wood, both fungi were isolated from the mating chambers, the center and the uppermost end of the galleries throughout the season. The fact thatO. piceae was consistently isolated with high frequency from adults and from their galleries strongly suggested that this species would be the principal blue stain fungus infecting the beetle-attacked larch trees.

The dynamics of the fungal symbionts in the gallery system and the mycangia of the ambrosia beetle, Xylosandrus mutilatus, were studied in relation to its life history using both isolation experiments and scanning electron microscopy (SEM). In the galleries, Ambrosiellasp. was predominant during the larval stages but its relative dominance gradually decreased during the development of the larvae. In contrast, yeasts (mainly Candidasp.) and Paecilomycessp. dominated continuously in the galleries after eclosion. Ambrosiellasp. was consistently stored in the mycangia in all adult stages, except in the teneral and overwintering adults when the other fungi were dominant. No fungal spores occurred in the mycangia of the adult beetles reared under aseptic conditions from the pupal stage, while only Ambrosiellasp. was stored in those reared from the teneral‐adult stage. These results suggest that: (i) Xmutilatusis associated with at least three fungal species, among which Ambrosiellasp. is the most essential food resource for development of the broods; (ii) immediately after eclosion, new female adults may take at least four associated fungal species, with no or incomplete selection, into their mycangia from the walls of the cradles; and (iii) conditions may well be produced in the mycangia of both matured and dispersing beetles whereby only the spores of Ambrosiellasp. can proliferate.

Exotic insects are constantly intercepted at U.S. ports-of-entry. Of these, wood-boring beetles, particularly xyleborine ambrosia beetles, are sometimes missed during port inspections and become established in the United States. Euwallacea validus (Eichhoff) and Euwallacea interjectus (Blandford) are morphologically similar Asian ambrosia beetle species that vary by their fungal associates and their potential to cause economic damage. Euwallacea validus and E. interjectus were first discovered in New York (1975) and Hawaii (1976), respectively. Euwallacea validus was collected multiple times from widely separated localities and is assumed to have spread throughout the eastern United States. The discovery of E. interjectus in Florida (2011) and Texas (2011) prompted our review of the E. validus specimens because of the potential misidentification of the species. In addition, using mitochondrial cytochrome oxidase I (COI) DNA data and phylogenetic analysis, we tested the hypothesis that multiple introductions account for the U.S. populations of E. interjectus and E. validus. Our review of 7,184 specimens revealed an earlier introduction to the mainland for E. interjectus, which was first collected from Louisiana in 1984. This species is distributed in the South while E. validus occurs in the North with a known area of syntopy in northeastern Georgia. The extent of the syntopy within the United States is unknown and further investigation is required. Phylogenetic analysis of 24 E. interjectus and 20 E. validus individuals resolved clades that associated with each species and gross geographic provenance. Four well-supported clades represented E. interjectus which included the following localities: 1) Hawaii and Thailand; 2) Vietnam, Taiwan, and Texas; 3) Okinawa (Japan); and 4) Japan and several southern U.S. states. One clade comprised all E. validus specimens from Japan and the mainland United States. Four and two haplotypes were found for the E. interjectus and E. validus specimens, respectively, in mainland United States. Except for the Texas specimen, the haplotypes differed by one nucleotide. The relationship of the haplotypes and their sequence similarity suggested that the provenance of E. validus and the majority of E. interjectus haplotypes was Japan while the Texas haplotype originated later and from a location near Taiwan. Given the high nucleotide sequence difference between the Hawaiian and Thai haplotypes, the exact origin of the Hawaiian E. interjectus is unknown but likely Southeast Asia. A broader investigation including more SE Asian individuals will help to further explain the introduction of E. interjectus into Hawaii and Texas.

The bark beetle Scolytogenes birosimensis Niijima is suspected to be involved in the decline of Pittosporum tobira (Thunb. ex Murray) Aiton in the coastal areas of Japan. We isolated fungi from adult S. birosimensis in nine different localities in Japan to assess their potential association and predict their contribution to the success of the beetle. Results from morphological identification of associated fungi showed that the beetle was associated with Fusarium solani and Candida spp. Furthermore, molecular analysis showed that F. solani was most closely related to the plant pathogenic fungus F. solani f. sp. mori. Fungal isolation from surface-sterilized, dissected beetles and scanning electron miscroscope (SEM) observation of the body surface suggested that the associated fungi were carried in the pits on the beetles' elytra. These findings contribute to the understanding of the relationships between S. birosimensis and its associated fungi.

We examined the genetic structure of populations of an ambrosia beetle, Xylosandrus brevis (Eichhoff) (Curculionidae: Scolytinae), to understand its colonization dynamics. We collected specimens from 20 sites in Japan and studied the genetic structure of these populations using portions of the mitochondrial cytochrome oxidase I gene (COI). A phylogenetic analysis showed three distinct lineages (clades A, B, and C) within X. brevis. Clade A had 65 haplotypes from all the populations, except for populations from Nagano (Chiisagata-gun) and the Ryukyu Islands (Ishigaki), whereas clade B had 14 haplotypes from nine populations (Yamagata, Saitama, Shimo-Minochi-gun, Chiisagata-gun, Toyota, Watarai-gun, Wakayama, Tottori, and Kochi), and clade C had 6 haplotypes from the Ryukyu Islands (Ishigaki) population (and nowhere else). Nested clade phylogeographic analysis showed 65 clade A haplotypes and 14 clade B haplotypes geographically structured in two clades, respectively. A contact zone was identified in the Chubu and Tokai areas. Our results suggest that gene flow and individual movement of X. brevis between the Ryukyu Islands and the other three main islands of Japan has been rare, and X. brevis may be divided into two populations, eastern-central and northern-western, with little gene flow between the four areas of western Japan (Kinki, Chugoku, Shikoku, and Kyushu).

Dissection of emerging adult xiphydriid woodwasps, Xiphydria ogasawarai Matsumura, occurring in eastern, central, and western Japan during a 2- to 4-yr period, revealed that adult females possessed mycangia filled with fungal spores, whereas the mycangia were absent in males. Two masses of the spore were located on the slit-like 1st and 2nd valvulae of the ovipositor sting directly below the subgenital plate, and were covered with the ventral membranes. This is a new type of mycangia in woodwasps. The spores were globular or ovoid in shape and ≈5-30 µm in diameter, and had no clamp connections. All isolates from the mycangia of the xiphydriid populations, originating from different sites and years, developed the same colonies on potato-dextrose agar. This suggests that female adults of X. ogasawarai carry a single fungal species in their mycangia. It was also found that X. ogasawarai produced a slime-like secretion from glandular organs in the body cavity (mucus reservoir), which is homologous to that present in the Siricinae.
