The aim of the present review is to summarize the methods used to estimate the turnover of various neurotransmitters of low molecular weight in the intact animal brain First, some introductory remarks and definitions are given. The following description of the terms used is derived mainly from (1969), (1957) and some references therein. The chemical compound whose quantitative behavior will be studied is called a substance and the behavior of the substance is studied in a system Thus, the turnover of the substance acetylcholine (ACh) may be studied in a system, such as the rat striatum or the whole mouse brain In vivo studies of the central nervous system are usually considered to be an open system, meaning that a precursor enters the system and metabolites leave the system. The behavior of a substance is not necessarily uniform in the system, and, therefore, a system may contain several pools or compartments of that substance. A compartment is defined as a quantity of a substance that has distinguishable and uniform kinetics of transformation The boundaries of a compartment may, but do not necessarily, conform to anatomical boundaries, and conversely, anatomical boundaries are not necessarily divisions of the compartments. Abstract: We assessed essential fatty acid (EFA) and B-vitamin status, together with their determinants, in 61 patients with schizophrenia and established whether those with poor status responded biochemically to the appropriate dietary supplements. As a group, the patients had high erythrocyte saturated fatty acids (FAs), monounsaturated FA and low polyunsaturated FA of the ω3 and ω6 series. Patients reporting not to take vitamin supplements had low vitamin B
12 and high homocysteine. Homocysteine variance proved best explained by folate in both the total group and male patients, and by vitamins B12 and B6 in females. Alcohol consumption and duration of illness are risk factors for low polyunsaturated FA status (P97.5 of reference range). Two patients exhibited biochemical EFA deficiency and seven showed biochemical signs of ω3/docosahexaenoic acid (DHA) marginality. Four patients exhibited moderate hyperhomocysteinemia with plasma values ranging from 57.5 to 74.8μmol/L. None of the five patients with either moderate hyperhomocysteinemia, biochemical EFA deficiency, or both, was predicted by their clinicians to have poor diets. That diet was nevertheless at the basis of these abnormalities became confirmed after supplementing 4 of them with B vitamins and with soybean and fish oils. We conclude that a subgroup of patients with schizophrenia has biochemical EFA deficiency, ω3/DHA marginality, moderate hyperhomocysteinemia, or combinations. Correction seems indicated in view of the possible relation of poor EFA and B-vitamin status with some of their psychiatric symptoms, but notably to reduce their high risk of cardiovascular disease. 4. Bergsonian Theses in a Contemporary (Neuro-) Biological Context

- Abstract
Henri Bergson's philosophy is based heavily on assumptions concerning biological, brain and mental functions. Most significant are his concepts of elan vital (vital impetus, driving force for life, ontology and evolution) and associated intuition and creativity, duration (duree) versus physical time, discreteness versus continuity of space and time, and his stance against association of ideas. This article describes some of Bergson's major assumptions and theses, together with comments thereon. Several general biological and neurobiological principles are examined with respect to their compatibility with Bergson's philosophy { and if so, how. I consider the following principles: random hazard functions, fast brain transitions, stochastic or random modeling, potential energy, entropy and isoenergicity. Whereas some of Bergson's concepts are well compatible with biological principles (including atemporal (intra)cerebral mechanisms of memory, consciousness, intuition and instinct), others (creative and vital mechanisms) might be viewed as being indirectly triggered by lifetimespanning processes, such as aging. Modern (neuro-) biology tends to incorporate discrete transitions into intuitively perceived continuity of life, as for instance those observed and modeled in stochastic mental and aging processes. Some concepts (e.g. timelessness of ideas, rejection of associationism in memory) are too strictly applied by Bergson, but are compatible with current neurobiology in a broader sense.

5. Nerve Growth Factor and Brain-Derived Neurotrophic Factor in Schizophrenia and Depression: Findings in Humans, and Animal Models

- Abstract
Depression and schizophrenia are major psychiatric disorders. Recently it has been documented that these diseases are characterized by deficits and / or loss of neurons in specific brain regions. Nerve growth factor and brainderived neurotrophic factor are endogenous biological mediators involved in neuronal survival and plasticity of dopaminergic, cholinergic, and serotonergic neurons in the central nervous system. Structural, biochemical, and molecular findings led to the hypothesis that these molecules play a role in the pathophysiology of psychiatric disorders and suggested that alterations in expression of neurotrophic factors could be responsible for neural maldevelopment and disturbed neural plasticity both in young, adult and aged subjects. Studies aimed at understanding the mechanisms regulating these events might be an important line of research for analyzing the etiopathogenesis of psychiatric disorders and eventually identifying new methods for diagnosis and new therapeutic strategies.

- Abstract
In the past few decades, our understanding of the central nervous system has evolved from one of an immune-privileged site, to one where inflammation is pathognomonic for some of the most prevalent and tragic neurodegenerative diseases. Current research indicates that diseases as diverse as multiple sclerosis, stroke and Alzheimer's disease exhibit inflammatory processes that contribute to cellular dysfunction or loss. Inflammation, whether in the brain or periphery, is almost always a secondary response to a primary pathogen. In head trauma, for example, the blow to the head is the primary event. What typically concerns the neurologist and neurosurgeon more, however, is the secondary inflammatory response that will ensue and likely cause more neuron loss than the initial injury. This paper reviews the basic neuroinflammatory mechanisms, the potential neurotoxic mediators during activation of microglia, the brain resident macrophages, and their role in neurodegeneration. Alzheimer's disease is taken as a prototype for exploring these mechanisms, as it expresses more than 40 inflammatory mediators, it is the most extensively studied disorder in terms of immune-related pathogenesis, and because of its importance as the most prevalent type of dementia. Tools for the visualization of these neuroinflammatory processes, both structural and mainly functional, are critically reviewed and discussed.

This paper summarizes the possible interrelation between peripheral and/or cerebral inflammation and depression. Often, depression is regarded as a consequence of life events, including disabling diseases. The question addressed here is whether activation of the inflammatory response system (IRS) can cause depression. Epidemiological studies suggest that depression can be precipitated by bacterial or viral infections. In depressed patients, peripheral markers of the IRS are often increased. There is some evidence that some forms of depression are caused by a viral infection of the limbic system. More consistent are the observations that depression in diseases with active cerebral inflammatory processes (e.g. multiple sclerosis, Alzheimer's disease) may concur. Direct evidence of a relation between depression and inflammation was found in post-mortem brain material of patients with a vascular depression. In both inflammatory brain diseases and in depression, a state-dependent increased hypothalamus–pituitary–adrenal axis activity is seen. Animals studies have shown that intact cerebral serotonin systems are required for the activation of the IRS following an endotoxin challenge and that long-term treatment with antidepressants may change such a response. Gender differences between the prevalence of depression and inflammatory diseases are similar, as more females are affected. We hypothesize that cerebral or peripheral activation of the IRS may contribute to the course of some antidepressant treatment-resistant depressions. Clinical trials combining antidepressants and drugs that reduce the activation of the IRS may provide evidence for such proposed depression subtypes.

- Abstract
A miniaturised flow-through biosensor with a cell volume of only a few nanoliters was developed in our laboratory. The biosensor can be directly coupled to a microdialysis or ultrafiltration probe. Sampling and continuous on-line monitoring can thus be carried out at submicroliter levels and as a consequence quantitative recoveries of the analyte of interest are achieved. Via this method excessive calibration procedures, as are necessary with conventional microdialysis, are avoided. Here, the construction and the performance of such a biosensor for the continuous on-line monitoring of glucose and lactate will be presented. The biosensor is based on the amperometric detection of hydrogen peroxide after conversion of the analyte of interest by an immobilised oxidoreductase enzyme. Immobilisation of the enzyme is performed through electropolymerisation of m-phenylenediamine. Strategies to improve the performance (e.g. linearity, selectivity and stability) of the miniaturised biosensor are discussed and ex vivo and in vivo experiments carried out thus far demonstrate the potential of this miniaturised flow-through biosensor.

- Abstract
Inflammatory mechanisms contribute to the pathophysiology of senile dementia of the Alzheimer type (sDAT). Previous studies have shown that 57Co single photon emission computed tomography (SPECT) is able to visualize inflammatory lesions, probably by means of the final common pathway of Ca2homeostasis disturbance in both neuronal degeneration and inflammation. The aims of this study were: (1) to detect 57Co SPECT changes in sDAT patients; (2) to correlate these findings with those of conventional neuroimaging techniques and neuropsychological testing (NPT); and (3) to compare 57Co SPECT findings in sDAT patients with those in other types of dementia. Six patients suffering from probable sDAT were included and compared with four patients suffering from other types of dementia. All patients had a magnetic resonance imaging (MRI) scan, NPT, 57Co and 99mTc-ethyl cysteinate dimer (ECD) SPECT scan. Perfusion SPECT images were semiquantitatively evaluated by comparison with an age-matched normal database, while 57Co SPECT scans were assessed qualitatively. MRI and 99mTc-ECD SPECT scans yielded conclusive results with regard to the exclusion of other pathologies and the confirmation of the diagnosis. Using visual analysis, 57Co SPECT scans were unable to show any regional raised uptake, irrespective of the disorder, depth or extent of the perfusion defects, presence of atrophy on MRI or the results of NPT.

SummaryMounting evidence suggests involvement of prenatal factors in the pathogenesis of schizophrenia. The objective of the present study was to provide evidence for the hypothesis on schizophrenia that abnormal fetal brain development leads to impaired sensorimotor gating of acoustic startle stimuli. To this purpose, pregnant rats were treated with methylazoxymethanol acetate on one of four subsequent days of gestation, from E9 to E12. These procedures inhibit early stages of cortical proliferation in the foetuses, resulting in adult brain abnormalities that were mainly found in the entorhinal region. At the adult age, habituation andprepulse inhibition (PPI) of startle responses to acoustic stimuli were investigated. Our results show impaired habituation and PPI in rats treated at embryonic days 10 and 11. It is concluded that sensorimotor gating deficits can result from a developmental abnormality involving the entorhinal region, supporting the hypothesis that similar deficits, often observed in schizophrenic subjects, may be the result of entorhinal cortex dysfunction.

21. The Extracellular Concentration of the Anti-epileptic Drug Valproate in the Rat Brain as Determined with Microdialysis and an Automated HPLC Procedure

The cellular and extracellular brain concentration of valproic acid in freely moving rats has been estimated after intravenous injection of sodium valproate. Some rats were provided with a stereotaxically implanted dialysis probe in the striatum and a cannula in the heart through which the drug was injected and which allowed regular removal of blood. In other rats tissue levels of valproic acid were determined 5 and 90 min after drug injection. Valproic acid was determined by an automated precolumn derivatization procedure followed by HPLC separation and fluorimetric detection. Extracellular concentration was proportional to the blood concentration at every time interval, indicating rapid exchange of the drug between the two compartments. About 50% of the striatal content of valproate was in the extracellular space. The experiments demonstrated the usefulness of microdialysis to estimate both the extracellular concentration and the average cellular drug levels, provided a sensitive analysis procedure is available.

26. Manganese-Induced Hydroxyl Radical Formation in Rat Striatum Is Not Attenuated by Dopamine Depletion or Iron Chelationin Vivo

The present studies were aimed at investigating the possible roles of dopamine (DA) and iron in production of hydroxyl radicals (·OH) in rat striatum after Mn2+intoxication. For this purpose, DA depletions were assessed concomitant within vivo2,3- and 2,5-dihydroxybenzoic acid (DHBA) formation from the reaction of salicylate with·OH, of which 2,3-DHBA is a nonenzymatic adduct. Following intrastriatal Mn2+injection, marked 2,3-DHBA increases were observed in a time- and dose-dependent fashion reaching maximum levels at 6–18 h and a plateau beyond 0.4 μmol (fourfold increase). The delayed increase of 2,3-DHBA levels suggests that Mn2+induces·OH formation in the living brain by an indirect process. The early DA depletion (2 h) and relatively late·OH formation (6 h) indicate independent processes by Mn2+. In addition, depletion of DA (about 90%) by reserpine pretreatment did not significantly alter Mn2+-induced 2,3-DHBA formation or the extent of DA depletion, suggesting that DA or DA autoxidation are not participating in Mn2+-induced·OH formationin vivo.Furthermore, Mn2+injection did not significantly alter the low molecular weight iron pool in striatum, and co-injections of the iron-chelator deferoxamine with Mn2+into striatum did not significantly attenuate Mn2+-induced 2,3-DHBA formation. These findings suggest no role of chelatable iron in generation of Mn2+-induced·OH, but do not exclude a role for mitochondrial heme–iron or peroxynitrite (Fe-independent) in Mn2+-induced·OH formation.

In PET studies we have shown the usefulness of cobalt radionuclides for the visualization and quantification of ischaemic damage in stroke. In the present study, we explored 57Co2+as a SPET tracer. Uptake of radioactivity was estimated by using a cobalt enhancement ratio defined as the ratio of cobalt uptake in the affected region versus a similar volume in the non-affected contralateral side. Clinical status was assessed with the Orgogozo stroke score at the time of scanning and at least 60 days after admission. Nineteen patients (11 men, 8 women) with a middle cerebral artery stroke were examined with 57Co2+SPET 0–30 days after stroke onset. Our investigations show enhanced cobalt uptake in the infarcted brain tissue in patients with a major stroke and little clinical improvement. There was a significant correlation between the cobalt enhancement ratio and the Orgogozo score at the time of scanning and discharge. Our results suggest that 57Co2+SPET is suitable for determining the extent of (possibly calcium-mediated) damage in stroke and in the assessment of potential therapeutic interventions.

30. The significance of the expression of Fos protein in the brain for the classification of antipsychotics

- Abstract
SummaryIn the past 6 years many reports have been published on the effects of antipsychotic drugs on regional c-fos gene expression in the brain. In animals this experimental approach has allowed the study of antipsychotic drug effects at the cellular level. The fos-gene, a member of the immediate early gene family, regulates the transcriptions for other, often unknown, genes. Some aspects of this mechanism have been previously described. In this report the significance of this technique for the classification of antipsychotic drugs is discussed. It appears that these drugs are effective both in the (meso)limbic brain areas (including nucleus accumbens, amygdala and septum) and also in the basal ganglia. The relative effects studied using c-fos expression of the atypical antipsychotics (eg. clozapine, risperidone) are higher in the limbic structures, as compared to results obtained with the typical antipsychotics (eg. haloperidol). The potential of the c-fos approach for further developments in this area are indicated.

31. Antidepressive interventions: on state and vulnerability of the brain

SummaryAn attempt is made to relate drug and non-drug antidepressive interventions to brain processes. In the present context two concepts are proposed: vulnerability towards depressogenic factors and depression as a state of the brain. Accordingly, it is assumed that the current antidepressants make the brain less vulnerable by inhibiting monoaminergic neurons and by reducing the number or efficacy of their postsynaptic receptors. In contrast, electroconvulsive therapie and sleep-deprivation affect primarily the state of the brain and can produce spectacular (but often short lasting) improvements. It is proposed that the combination of the two aspects may lead to fast and lasting antidepressive effects.

The probability that an individual will suffer from a major depressive episode is often considered to be influenced by risk factors such as gender, premature parental loss, exposure to pathogenic parental rearing, personality, a history of traumatic events, a previous history of major depression, low social support, recent stressful life events and difficulties and predisposing genetic influences. Although several studies suffer from methodological limitations, major depression (endogenous depression) is regarded as a multifactorial disorder and understanding its etiology requires the rigorous integration of several risk factors.

InleidingIn deze rubriek zal worden ingegaan op recente observaties betreffende de psychofarmakologie in de breedst mogelijke zin. Vaak zullen zaken uit de literatuur worden behandeld. De gekozen onderwerpen dienen altijd klinische relevantie te hebben. Ook zal deze rubriek gaan over gewoontes die in psychofarmakologische behandeling of in onderzoek zijn geslopen die volgens de auteur lang niet altijd logisch zijn. Soms zal deze rubriek een oproep bevatten om te reageren op een bijdrage gepubliceerd in dit of in een ander tijdschrift. Ook zal de lezer worden aangemoedigd hier zijn eigen ervaringen mee te delen. Het is niet de bedoeling in deze rubriek onderzoeken uitvoerig te rapporteren. Daarvoor staan andere pagina's van de Acta Neuropsychiatrica voor u open.

34. Evidence for a probenecid-sensitive transport system of acid monoamine metabolites from the spinal subarachnoid space

- Abstract
The location of probenecid-sensitive elimination mechanisms of monoamine metabolites from the cerebrospinal fluid of the cat was determined with ventriculo-cisternal or ventriculo-lumbar perfusion techniques. These techniques were described in detail. Levels of endogenous homovanillic acid and 5-hydroxyindoleacetic acid were assayed in the perfusate. Probenecid administration induced the most marked increase of the levels of the monoamine metabolites in the ventriculo-lumbar perfusates. It was concluded that probenecid blocked the transport of both metabolites from the spinal subarachnoid space.

- Abstract
Primary brain rumours are usually assessed by computed tomography (CT) and magnetic resonance imaging (MRI), sometimes in conjunction with positron emission tomography (PET). We used cobalt-55 (55Co) as a calcium (Ca) tracer to visualize decaying tumour tissue, based on the fact that Ca-influx is essential in both cell death and leukocyte activation. Net 55Co uptake may be the result of cell decay, leukocyte infiltration, (re)perfusion and the pharmacological profile of 55Co. Three patients with primary malignant brain tumours (first presentation) were studied with CT, MRI and Co-PET after the intravenous administration of 0.5 mCi 55Co. Histopathological diagnosis was obtained by biopsy or resection. Co-PET demonstrated each of the brain tumours and showed good topographical agreement with CT and MRI. Co-PET provided additional detail as to the site and size of the necrotic core and the peri-necrotic rim of decaying tumour. The 55Co uptake indices varied between 2.6 and 5.3. 55Co demonstrated uptake in decaying tissue, irrespective of the integrity of the blood-brain barrier. Neither necrotic nor viable tumour tissue showed affinity for 55Co. Since 55Co is readily applicable to both PET and single photon emission tomography (SPET), differences in the uptake mechanism and functional significance of the 55Co tracer are discussed in relation to 201T1 SPET. We present a (limited) pilot series of three patients to forward the claim of this new functional technique in nuclear neurology.

Summary A convenient method is described for light microscopic autoradiography at the cellular level for the visualization ofin vivo labeled dopamine receptors. (
3 H)N-n-propylnorapomorphine [(3 H)NPA] is administered to rats under conditions that are known to give specific and saturable accumulation in the striatum. Seventy minutes after intravenous administration, the brain is rapidly frozen and cut on a cryostate microtome. The sections are treated with formaldehyde vapor, defatted for 1 hour in xylene and dipped in liquid nuclear emulsion, exposed for 2 weeks and developed.- Published

Summary The enzyme monoamine oxidase was labelled in the rat striatum or substantia nigra with locally injected radioactive pargyline. The binding was prevented by a pretreatment with non-radioactive pargyline, or with a combination of clorgyline and deprenyl. Most of the MAO labelled with

Summary The origin of free 3-methoxy-4-hydroxyphenylethyleneglycol (MHPG, a major metabolite of norepinephrine) in rat cerebrospinal fluid (CSF) was investigated using brain and spinal cord perfusions with artificial CSF, electrical stimulation of the locus coeruleus (LC) and the technique of retrograde cell labelling with horseradish peroxidase (HRP). The rates of appearance of MHPG into ventricular-cisternal and lumbar-cisternal perfusates were about 12.4 and 19.2 ng/hour respectively. Probenecid (200 mg/kg i.p.) did not alter the MHPG output in either preparation. Intravenous administration of 3µg MHPG did not substantially enhance the outflow of the metabolite in the ventricular-cisternal perfusate, indicating that MHPG found in the perfusate is of central origin. Maximal activation of central norepinephrine (NE)-containing neurons of the LC by electrical stimulation induced a 50% increase of MHPG levels in the ventricular-cisternal perfusate. In addition HRP was found to be retrogradely transported from the lateral ventricle to these LC-neurons.

45. Protective effect of fasting upon cerebral hypoxic-ischemic injury

- Abstract
This study was designed to determine the effect of fasting upon cerebral hypoxic-ischemic injury. In the first part of the study the effect of fasting was determined for survival, brain tissue water and kation contents, and blood-brain barrier intregrity. In the second part of the study the administration of the substratesß-hydroxybutyrate (BHB) and glucose has been evaluated regarding their influence upon the effect of fasting. The study used the Levine-Klein model of unilateral carotid occlusion and hypoxia because it mimics clinical situations of ischemia with hypoxia. The data show that fasting did protect rats from developing brain infarction following hypoxia-ischemia. Hypoglycemia seems to be involved in the mitigation of ischemic blood-brain barrier disruption. The plasma glucose level seems to be not the only factor involved in the genesis of the tissue kation changes. Starvation-induced ketosis probably does not play a role in the protection mechanism.

- Abstract
The role of glucocorticoids in neuronal viability is controversial. Most studies which describe the effects of glucocorticoids on ischemic brain damage use surgical adrenalectomy to induce a reduction in plasma corticosterone levels. In the present study we used metyrapone, a corticosterone synthesis inhibitor, to examine the effects of acute manipulation of corticosterone levels on neuronal damage and seizures. Shortly before transient hypoxia/ischemia, animals were subcutaneously injected with sesame oil, metyrapone, corticosterone or corticosterone+metyrapone. Both the neuronal damage and the percentage animals with seizures were found to correlate well with plasma corticosterone levels. This relation between affected area, seizures and corticosterone levels was confirmed when the rats were re-arranged into animals with and without seizures. However, corticosterone administration in metyrapone treated rats did not result (compared to MET treatment) in an increased neuronal damage. This suggests that the beneficial effects of metyrapone may be regulated by a corticosterone independent mechanism. © 1997 John Wiley & Sons, Ltd.
