Industrial wood pellet demand and international trade have been growing rapidly, requiring producers to build new production facilities. The purpose of this paper is to illustrate the trade‐offs of different wood pellet production locations across the world within the next ten years and to improve the quality and viability of future wood pellet production location and supply chain decisions. To this end, a multi‐criteria decision analysis (MCDA) was performed. This approach enabled assimilation and synthesis of both qualitative and qualitative data of a comprehensive set of regions in which lies its originality.

The purpose of this study is to provisionally classify, based on the Biopharmaceutics Classification System (BCS), drugs in immediate-release dosage forms that appear on the World Health Organization (WHO) Essential Drug List. The classification in this report is based on the aqueous solubility of the drugs reported in commonly available reference literature and a correlation of human intestinal membrane permeability for a set of 29 reference drugs with their calculated partition coefficients. The WHO Essential Drug List consists of a total of 325 medicines and 260 drugs, of which 123 are oral drugs in immediate-release (IR) products. Drugs with dose numbers less than or equal to unity [Do = (maximum dose strength/250 mL)/solubility ≤ 1] are defined as high-solubility drugs. Drug solubility for the uncharged, lowest-solubility form reported in the Merck Index or USP was used. Of the 123 WHO oral drugs in immediate-release dosage forms, 67% (82) were determined to be high-solubility drugs. The classification of permeability is based on correlations of human intestinal permeability of 29 reference drugs with the estimated log P or CLogP lipophilicity values. Metoprolol was chosen as the reference compound for permeability and log P or CLogP. Log P and CLogP were linearly correlated (r ² = 0.78) for 104 drugs. A total of 53 (43.1%) and 62 (50.4%) drugs on the WHO list exhibited log P and CLogP estimates, respectively, that were greater than or equal to the corresponding metoprolol value and are classified as high-permeability drugs. The percentages of the drugs in immediate-release dosage forms that were classified as BCS Class 1, Class 2, Class 3, and Class 4 drugs using dose number and log P were as follows:  23.6% in Class 1, 17.1% in Class 2, 31.7% in Class 3, and 10.6% in Class 4. The remaining 17.1% of the drugs could not be classified because of the inability to calculate log P values because of missing fragments. The corresponding percentages in the various BCS classes with dose number and CLogP criteria were similar:  28.5% in Class 1, 19.5% in Class 2, 35.0% in Class 3, and 9.8% in Class 4. The remaining 7.3% of the drugs could not be classified since CLogP could not be calculated. These results suggest that a satisfactory bioequivalence (BE) test for more than 55% of the high-solubility Class 1 and Class 3 drug products on the WHO Essential Drug List may be based on an in vitro dissolution test. The use of more easily implemented, routinely monitored, and reliable in vitro dissolution tests can ensure the clinical performance of drug products that appear on the WHO Essential Medicines List. Although oral vaccination has numerous advantages over parenteral injection, degradation of the vaccine and low uptake by the gut associated lymphoid tissue (GALT) still complicate the development of efficient oral vaccines. However, previous studies in our laboratory demonstrated that chitosan microparticles can have suitable size, charge, loading and release characteristics for oral vaccination using ovalbumin as model vaccine.In this study, two different approaches were used to investigate the in vivouptake of chitosan microparticles by murine Peyer's patches. Firstly, a confocal laser scanning microscopy (CLSM) study was performed to visualize the uptake of fluorescent-labeled chitosan microparticles in the Peyer's patches after intragastrical feeding. Subsequently, the intestinal epithelial uptake of ovalbumin loaded chitosan microparticles was visualized using immunohistochemical staining of ovalbumin. Because the microparticles are biodegradable, this entrapped ovalbumin will be released after intracellular digestion in the Peyer's patches. CLSM visualization demonstrated that chitosan microparticles enhance the uptake of fluorescent-labeled ovalbumin by the epithelium of the Peyer's patches. No ovalbumin uptake by the intestinal epithelium was observed when the protein was administered without microparticles. Moreover, immunohistochemical visualization studies revealed that ovalbumin could only be transported into the Peyer's patches after association to chitosan microparticles. Since uptake by Peyer's patches is an essential step in oral vaccination, these in vivoexperiments demonstrate that chitosan microparticles are very promising vaccine delivery systems.

Octreotide acetate is a somatostatin analogue used for the control of endocrine tumors of the gastrointestinal (GI) tract and the treatment of acromegaly. The oral absorption of octreotide is limited because of the limited permeation across the intestinal epithelium. Both chitosan hydrochloride and N-trimethyl chitosan chloride (TMC), a quaternized chitosan derivative, are nonabsorbable and nontoxic polymers that have been proven to effectively increase the permeation of hydrophilic macromolecules across mucosal epithelia by opening the tight junctions. This study investigates the intestinal absorption of octreotide when it is coadministered with the polycationic absorption enhancer TMC. Caco-2 cell monolayers were used as an in vitro intestinal epithelium model, and male Wistar rats were used for in vivo studies. Octreotide with or without polymers (TMC; chitosan hydrochloride) was administered intrajejunally in rats, and serum peptide levels were measured by radioimmunoassay. All applications and administrations were performed at neutral pH values (i.e., pH = 7.4). In vitro transport studies with Caco-2 cells revealed an increased permeation of octreotide in the presence of TMC. Enhancement ratios ranged from 34 to 121 with increasing concentrations of the polymer (0.25–1.5%, w/v). In rats, 1.0% (w/v) TMC solution significantly increased the absorption of the peptide analogue, resulting in a 5-fold increase of octreotide bioavailability compared with the controls (octreotide alone). Coadministration of 1.0% (w/v) chitosan hydrochloride did not enhance octreotide bioavailability. These results in combination with the nontoxic character of TMC suggest that this polymer is a promising excipient in the development of solid dosage forms for the peroral delivery and intestinal absorption of octreotide. © 2000 Wiley-Liss, Inc. and the American Pharmaceutical Association J Pharm Sci 89:951–957, 2000

Purpose. To evaluate N-trimethyl chitosan chloride (TMC) of highdegrees of substitution as intestinal permeation enhancers for thepeptide drug buserelin in vitrousing Caco-2 cell monolayers, and toinvestigate TMCs as enhancers of the intestinal absorption of buserelinin vivo, in rats. Methods. TMCs were tested on Caco-2 cells for their efficiency toincrease the paracellular permeability of the peptide buserelin. For thein vivostudies male Wistar rats were used and buserelin wasadministered with or without the polymers intraduodenally. Both types ofexperiments were performed at pH 7.2. Results. Transport studies with Caco-2 cell monolayers confirmed thatthe increase in buserelin permeation is dependent on the degree oftrimethylation of TMC. In agreement with the in vitroresults, in vivodata revealed highly increased bioavailability of buserelin followingintraduodenal co-administration with 1.0% (w/v) TMCs.Intraduodenally applied buserelin resulted in 0.8% absolute bioavailability,whereas co-administrations with TMCs resulted in mean bioavailabilityvalues between 6 and 13 %. Chitosan HCl (1.0% pH = 7.2) did notsignificantly increase the intestinal absorption of buserelin. Conclusions. Both the in vitroand in vivoresults indicate that TMCsare potent mucosal permeation enhancers of the peptide drug buserelinat neutral pH values.

The thermal behaviour of human stratum corneum (SC) with various hydration levels was studied using differential thermal analysis DSC within the temperature range of −130 to 120°C. SC containing 20% water, resembling the intact condition, shows thermal transitions at around −20°C (representing water in skin), −10, 40, 70°C (representing skin lipids), 85°C (representing protein-associated lipids) and 100°C (representing skin protein). Dehydration of SC causes the transitions at −20 and 100°C to be invisible. Lipid extraction followed by dehydration eliminates all transitions. Further hydration produces a transition of water at around 0°C with a huge change in enthalpy. The perturbation effects of penetration enhancers fatty acids (FA) and propylene glycol (PG) were studied using DTA on SC after pretreatment with PG alone and FA/PG. The application of PG alone shifted the transitions at 70 and 85°C to lower temperatures. Additionally, the application to dehydrated stratum corneum removes the transitions at −10°C. Saturated fatty acids, e.g. nonanoic and decanoic acids, exert barely noticeable effects on the thermal behaviour of SC suggesting that they easily mix with the skin lipids. Thermal analysis also revealed that the cis-9- and 13-isomers of octadecenoic acid (monounsaturated fatty acids) form a separate domain containing mostly the pure fatty acids within the SC lipids and suppress the lipid transitions at 70/80°C. Polyunsaturated fatty acids linoleic and α-linolenic acids — form separate domains but do not completely suppress the SC lipid transitions at 70/80°C as monounsaturated acids do. This study suggests different ways of perturbation by various fatty acids.

Two different toxicity models were used to assess the relationship between the physicochemical properties of non-ionic surfactant vesicles (NSVs), and the safety of these vesicles for topical drug administration. The vesicles used in this study consisted of polyoxyethylene alkyl ethers (CnEOm) in which the number of C atoms (n) varied between 12 and 18 and the number of oxyethylene units (m) between 3 and 7. The physicochemical properties of the vesicles are described in terms of hydrophilic-lipophilic balance (HLB) values, and critical micelle concentrations (CMC), and the rigidity of the bilayers as determined by the gel-liquid transition temperatures and the cholesterol content of the bilayers. The first toxicity model, comprising the measurement of the ciliary beat frequency, is a tool to assess the safety of intranasally applied formulations. Studies using this ciliotoxicity model revealed that by increasing the length of the alkyl chain of the surfactant, a decrease in toxicity was observed. The opposite correlation was found if the length of the polyoxyethylene headgroup was increased. Furthermore, it was observed that gel-state vesicles produce less of an effect on the ciliary beat frequency than liquid state vesicles. The second toxicity model, comprising the determination of cell proliferation of human keratinocytes, is a method to assess skin irritancy. In contrast to the ciliotoxicity model the length of the polyoxyethylene headgroup and of the alkyl chains did not seem to have an effect on the safety of the vesicles. However, the bond by which the headgroup is linked to the alkyl chain, showed a very strong effect on the toxicity of the surfactant: oleyl-EO5ester vesicles were found to have an effect on the cell proliferation, which was one-sixteenth that of the oleyl-EO5ether vesicles. The cholesterol content did not appear to have an effect on the proliferation of the keratinocytes. Neither the HLB nor the CMC values appeared to have an effect on the safety of the NSV formulations as observed in both toxicity models.

The electrokinetic properties of the endotoxins of Escherichia coliand Serratia marcescenshave been examined. Both endotoxins are negatively charged, with the zetapotential being increased by the presence of cations whose relative influence resembles the Schulze-Hardy rule for colloid stability. Fe3+and Th4+ions are capable of reversing the negative charge of the endotoxin particles to positive. These cations were found to have a strong inhibitory effect on the activity of endotoxins in the limulus amoebocyte lysate test but the inhibitory effect did not parallel changes in the zetapotential because the effect occurred at concentrations too low to affect this parameter.

Hydrogel patches based on water swellable polyacrylates have been developed for long-term transdermal drug delivery. Two properties, relevant to the performance of hydrogel patches in-vivo have been investigated in humans over five days. These were: (i) the kinetics of water exchange between the skin and the patches; (ii) the skin compatibility of the patches. It was found that initially there was a gradually increasing uptake of water from the skin by the patches, but after about 20 h the water exchange followed a regular fluctuating pattern, peaking once a day and once a night. The skin compatibility of the patches was satisfactory, in that no redness or pustulation was noticed throughout the five days. This was most likely due to the capability of the patches to exchange water with the skin.

Endotoxins of Escherichia coli, Salmonella abortus equiand Serratia marcescenswere examined using freeze-etch electronmicroscopy and light scattering techniques, with emphasis on their aggregation state. These endotoxins show distinct features of particle size and mass and they may be affected by the presence or absence of Ca2+ions. No important differences were observed in the particle characteristics of the respective endotoxins in various infusion fluids.

The time dependent changes of the lamellar gel structure in a nonionic O/W cream were studied. It appeared that the changes were connected with alterations in the hydrophilic layers of this lamellar gel structure. The structure of the hydrocarbon layers did not change. The alterations were induced by an increasing hydration of the surfactant molecules on cooling from the preparation temperature to room temperature. Ageing of the cream involves a decrease of the thickness of the hydrophilic layers and a change of the distribution of the surfactant molecules, resulting in, among other things, a decrease of the release rate of a hydrophilic drug. Ageing of the cream can be prevented by using the appropriate amount of starting materials or by the use of polymerizable surfactants. In the former case a cream, from which a drug is slowly released, is obtained. On the other hand, creams containing polymerized surfactants can release drugs at a relatively high rate.

The structure of a nonionic O/W cream and its separate components were investigated by X-ray diffraction and microscopic methods. The results presented in this paper are combined with the results obtained with DSC in order to elucidate these structures. The very important role of water with respect to the formation of the colloidal structures is discussed. The influence of water on the polymorphic behaviour of cetylstearylalcohol is attributed to a good structural fit of itsa-modification and water. Hydration induces a tilt of the alkyl chains of poly(oxyethylene)20glycerylmonostearate (PGM20). For this surfactant an antiparallel arrangement of the molecules is proposed. The lamellar structures present in mixtures of cetylstearylalcohol, PGM20 and water are discussed extensively.

As part of a study polyoxyethylene alkyl ethers (CmEOn), water and decane, the phase diagram and the structures of the mesophases of pure C12EO6 and technical grade C12EO<7> were compared. The constructed phase diagrams of the two systems show a great resemblance except for one difference: the viscous isotropic phase is only present in the C12EO6 phase diagram.

The influence of the addition of liquid paraffin (LP) on the structure of the lamellar (La) and hexagonal (H1) mesophases formed in mixtures of water (W) and BRIJ 96 (B) was studied. Mesophases were identified using polarization microscopy and small angle x-ray diffraction (SAXD). Repeat spacings were also determined with SAXD. Depending on theW/B ratio, addtion ofLP toLa gives a large, almost linear one-dimensional swelling or an initial swelling followed by a gradual transition to H1.La with a highLP-content gives a diffraction pattern showing only the first order diffraction maximum, possibly a result of undulations of the layers. The lamellar structure, however, was confirmed using freeze fracture electron microscopy (FFEM). Addition ofLP to H1 gives an initial swelling followed by a transition to a transparent, highly viscous, isotropic phase, called the gel-phase (G). The diffraction pattern obtained fromG yields little information on its structure. A large swelling ofG withLP was observed. From the degree of swelling as a function of hydrocarbon content it was inferred that this phase consists of spherical aggregates forming a close-packed structure. Using FFEM, textures were visualized resembling those obtained from the isotropic mesophase (I1) in water-surfactant mixtures. Finally, geometrical factors are discussed that may play a role in the formation of the gel-phase.

The structure of a simple O/W cream and its separate components were investigated by differential scanning calorimetry. The results obtained for cetylstearylalcohol agree with those cited in literature. The thermal behaviour of the component poly(oxyethylene)20 glycerolmonostearate (= PGM20) was also investigated. It is pointed out that the thermal history of the PGM20 batch may play an important role in a study concerning the structure of this surfactant. To both components water was added to study the effects of hydration. Hydration affected the transition temperatures of either component. The melting enthalpy per unit mass of cetylstearylalcohol was not influenced, in contrast with the melting enthalpy of the hydrocarbon fraction in PGM20/water mixtures. It is supposed that hydration induces a tilt of the hydrocarbon chains with respect to the lattice plane in lamellar PGM20/water mixtures. Cooling experiments indicated that, in samples containing sufficient water, 2 water molecules were tightly bound to the polyoxyethylene chains of the surfactant. The properties of simple creams containing cetylstearylalcohol, PGM20 and water can be explained regarding their structure and considering the behaviour of the separate components.

As a part of a study of the colloidal structures present in nonionic ointments, the interaction between polyoxyethylene and water is investigated. A series of mixtures of polyoxyethylene 1550 and water is studied using differential scanning calorimetry. Heating as well as cooling experiments are performed. The effects of the heat of mixing, supercooling and melting point depression on the measured enthalpy changes are discussed. From the non-freezing water fraction it is concluded that 2 water molecules per oxyethylene unit are tightly bound to the polymer chain. The observed differences between the cooling and the heating curves lead to a possible explanation for the alterations in the samples occurring at low temperatures. A hydrate structure of polyoxyethylene at low temperatures is proposed. Finally, comment is made on the phase diagram of the system polyoxyethylene/water.

Calorimetric measurements have been carried out to determine to which extent water interacts with p-HEMA gels and with p-(HEMA+MAA) gels. It appeared, that 0.14 g water/g dry p-HEMA and 0.23 g water/g dry p-(HEMA+MAA) show strong negative enthalpy changes on sorption in the polymer, the enthalpy changes being -180 J/g and -360 J/g water respectively.

This study deals with effects of electrical (current density, frequency and duty cycle) and chemical (buffer pH and ionic strength) conditions on the flux of the octapeptide, 9-desglycinamide, 8-arginine-vasopressin (DGAVP), through dermatomed human skin. A pulsed constant current was applied during iontophoresis. The anode faced the anatomical surface of the skin samples inside the diffusion cells. The resistive and capacitative components of the equivalent electrical circuit of human skin could be calculated by fitting the voltage response to a bi-exponential equation. The skin resistance prior to iontophoresis varied between 20 and 60 k O.cm2. During iontophoresis a decrease of skin resistance and an increase of the series capacitances was observed, which were most pronounced during the first hour of iontophoresis; thereafter both quantities gradually levelled off to an apparent steady state value. The reduction of the resistance during iontophoresis increased non-linearly with increasing current density between 0.013–0.64 mA.cm-2. The steady state resistance and capacitances did not vary significantly with frequency and duty cycle of the current pulse. There was no pH dependence of skin resistance at steady state. Between pH 4 and 10, the steady state peptide flux had a bell-shaped pH-dependence with a maximum of 0.17 nmol.cm-2.h-1 at pH 7.4, which is close to the I.E.P. of the peptide. Lowering the ionic strength from 0.15 to 0.015 M NaCl increased the steady state flux at pH 5 and pH 8 by a factor 5 to 0.28 ± 0.21 and 0.48 ± 0.37 nmol.cm-2.h-1, respectively. Together these observations suggested that DGAVP is transported predominately by volume flow. At pH 6, at which 65% of the peptide carried a net single positive charge, the steady state flux increased with increasing current density (0.013–0.64 mA.cm-2) from 0.11 ± 0.03 to 0.19 ± 0.04 nmol.cm-2.h-1. Skin permeability during passive diffusion preceding iontophoresis at pH 6.0 was 2.9 ± 0.6 * 10-7 cm.h-7. In accordance with theoretical predictions based on the Nernst-Planck equation, to which a volume flow term was added, the flux was proportional to the mean voltage across the skin between 0.013 and 0.32 mA.cm-2.h-1. Variation of frequency or duty cycle did not result in significantly different peptide transport rates. From these studies it is concluded that DGAVP can be transported iontophoretically through human skin. The pH- and ionic strength-dependence of the iontophoretic peptide flux suggests that transport of DGAVP mainly occurs by volume flow. Furthermore, the flux of DGAVP appears to be controlled by the applied voltage rather than by the current density, as predicted by the Nernst-Planck equation.

The intercellular lipid bilayers of the stratum corneum provide the permeability barrier of the skin. To perform an electron microscopical examination of the ultrastructure of these bilayers, ruthenium tetroxide fixation is required. In this study an optimal fixation protocol was developed and selected upon comparing seven different fixation procedures, using glutaraldehyde (GA) and the postfixatives ruthenium red, osmium tetroxide (OsO4) and ruthenium tetroxide (RuO4) in combination with potassium ferrocyanide (K4Fe(CN)6) and potassium ferricyanide (K3Fe(CN)6). Instead of fixing skin with either OsO4or RuO4, these two fixatives were combined in one protocol. In addition, the use of RuRed was introduced and the influence of both K4Fe(CN)6and K3Fe(CN)6in combination with RuO4were examined. Furthermore, we compared two dehydration solvents, methanol and acetone.

Lamellar (La) and hexagonal (H1) mesophases, formed in mixtures of water and polyethylene oxide (PEO) oleylethers (average EO-numbers 5 (a), 8.5 (b) and 14 (c)) and pure hexaoxyethylene dodecylether (d), were investigated with small-angle X-ray diffraction. Analysis of repeat spacings shows a deviation from linear one- and two-dimensional swelling ofLa andH1, respectively, when a transition to a mesophase with a larger curvature of the interface is approached. With regard toLa, in this respect similar behaviour is observed for a pure surfactant (d) and a surfactant with an EO-length distribution (b). It is proposed that, through the formation of aggregates of limited extent, water filled pores in the mesophases arise. This is confirmed by analysis of the intensity ratio of diffraction maxima of first and second order inLa. Models for finite aggregates are introduced. These consist of (1) lamellae, infinite in one direction and with variable lengthL in a direction perpendicular to the first, with cylindrical edges and (2) cylinders with variable lengthL bounded by spherical caps. According to these models, under certain conditions, finite aggregates may be formed without an increase of the interfacial area per surfactant molecule. Applying the models to the regions where non-linear swelling is observed, reasonable values forL are found, which decrease progressively when the phase transition is approached.

Mixtures of polyoxyethyleneoleylether, with an average of 14 oxyethylene units per molecule, and water, were investigated at between 20‡ and 100‡C using polarization microscopy. A lamellar (La), hexagonal (H1), viscous isotropic (V1) and cubic (I1) mesophase were found and an outline of the pseudo binary phase diagram was constructed. The mesophases were investigated with freeze-fracture electron microscopy and micrographs of characteristic textures are presented.La andH1 displayed well known features, the repeat distances observed were in good agreement with findings from X-ray diffraction.H1 showed fracture planes with and without a ribbed pattern. Using specific decoration with ice crystals, fracture planes with the ribbed pattern were found to be more hydrophilic.V1 showed regular patterns, in some places forming a square lattice, in other places resembling a network of short rods with two-dimensional hexagonal symmetry. The repeat spacing observed in the textures was about 11 nm.I1 showed foremost fractured layers with a fine texture of spherical units in a two-dimensional hexagonal array. The centre-to-centre distance between these units was about 8–9 nm.

The structure of systems containing equal weights of cetylalcohol, stearylalcohol and the nonionic surfactant poly(oxyethylene)20-glycerolmonostearate is studied as a function of the water concentration. In this paper mainly results are presented obtained by small angle X-ray-diffraction. Up to ± 22% (w/w) water several coexisting phases were observed. From ± 22% to ± 55% water a lamellar structure is found. This structure consists of lipophilic bilayers formed by the hydrocarbon tails of cetylalcohol, stearylalcohol and the surfactant, alternated by hydrophilic layers formed by the polyoxyethylene glycerol chains, the hydroxyl groups of cetylalcohol and stearylalcohol and water. From the swelling behaviour the density of the gel structure (0.9 g/cm3) and the cross sectional area of the molecules in the bilayers (0.19 nm2) have been calculated. It is concluded that the hydrocarbon chains are in a crystalline, state and are placed perpendicular to the lattice plane. Upwards of ± 55% water the lamellar phase disperses in the excess of water.

Hydrogels are widely studied materials for the preparation of sustained release drug dosage forms. Their soft, tissue-like consistency and their high biocompatibility in a number of applications make them promising candidates for this purpose. The water and the polymer in the gel form intricate structures and much research has been devoted to the elucidation of these structures, and of the interactions involved in their formation. Simple, drug-loaded hydrogels normally give a matrix-type delivery profile, in which the release rate is proportional to the square root of time; a number of approaches has been used to change this profile to other types of delivery, for instance to zero-order release. A number ofin vivo tests using hydrogel delivery systems has given favourable results.

Within the Center for Bio-Pharmaceutical Sciences the release characteristics of drugs form the major research object of the Division of Pharmaceutical Technology. Transdermal systems are being developed that can supply a drug during several days. Irritation of the skin may be avoided by using hydrogels. To enable long-term transdermal application also the colloidal structure of creams and ointments is investigated. As most drugs are to be taken orally, however, a research project was also started to make cheap and easy to produce controlled release tablets. The results with a microporous polypropylene polymer are promising.

O/W creams with crystalline gel structures (Water Containing Hydrophilic Ointment DAB 8, Non-ionic Hydrophilic Ointment DAC and a stearate cream) were investigated by means of small angle X-ray diffraction, differential scanning calorimetry, thermogravimetry and freeze fracture technique in combination with Transmission Electron Microscopy (TEM).

The effects of non-ionic surfactant vesicles (NSVs) on human skin in vitro were studied in relation to the physico-chemical properties of the vesicles. The interactions between NSVs and skin were visualized using both freeze fracture electron microscopy and confocal laser scanning microscopy. The physico-chemical properties of the NSVs were varied in a systematic way, using a broad series of polyoxyethylene monoalkyl ether type surfactants (CnEOm). The number of oxyethylene units (m) was varied between 3, 7 and 10, and the number of carbon atoms (n) was either 12 or 18. Both the effects of liquid state vesicles composed of C12Eo3,7 and C9=9EO10 surfactants and gel state vesicles (C18EO37) were investigated. After the application of the NS V suspension on the stratum corneum surface two essentially different types of vesicle-skin interactions were visualized. Firstly, an interfacial interaction, involving the adsorption of vesicles, and the deposition of bilayer sheets on the outermost layers of the stratum corneum observed for all NSV formulations tested in this study. Secondly, effects on the ultrastructure of the stratum corneum were observed: the appearance of water pools observed for the liquid state vesicles only, and ultrastructural changes of the intercellular lipid domains are induced only observed after treatment with C12EO3 NSVs. Neither changes in the ultrastructure of the viable epidermis, nor changes in deeper skin layers were observed.
