Tensile tests were made on commercially pure Al (A1050), Al-4.4 wt.% Mg (A5182) and Al-4 wt.% Cu (A2017) alloys by varying the test temperature and the strain rate. Textures developed at various stages of the tensile deformation were investigated with the orientation distribution function analysis. During the tensile test of pure Al with a strain rate of 3 × 10–4s –1at 20 °C, the tensile axis readily rotated toward the <111> stable end orientation. However, such a rotation occurred only at the later stage of the tensile deformation near the tensile stress, where the stress– strain curve became flat, and where work hardening was almost saturated. It is suggested that the size and the morphology of the dislocation cell, which had developed up to this stage, played a significant role in the subsequent texture development. In fact, the addition of Mg and Cu, which inhibited the development of the well-defined cell structure due to the Portevin –Le Chatelier effect, strongly suppressed the rotation of the tensile axis toward <111>. In all the materials investigated, textures were not affected by the strain rate and also by the temperature of the tensile test.

Recent longitudinal studies have shown the influence of multiple tooth loss on cognitive impairment, and earlier studies suggested that periodontal disease was related to cognitive decline. Tooth loss is associated with reduced masticatory function, which may affect stimulation of the central nervous system and dietary intake. Although some studies have reported a relationship between chewing ability and cognitive function, no studies have examined this area in terms of objective oral function. The aim of this study was to examine the association of occlusal force with cognitive decline in the preclinical stage among older people with higher-level functional capacity. This cross-sectional study for community-dwelling older people living in urban and rural areas in Japan examined 994 persons in the 70-y group (age range, 69–71 y) and 968 persons in the 80-y group (age range, 79–81 y). Retention of higher-level competence was defined according to the Tokyo Metropolitan Institute of Gerontology Index of Competence. Cognitive function was measured with the Japanese version of the Montreal Cognitive Assessment (MoCA-J). Oral status and function were assessed by the number of remaining teeth, periodontal pocket depth, and maximal occlusal force. Associations between the MoCA-J score and occlusal force were examined by bivariate and multivariate analysis. Approximately one-half of the participants retained higher-level functional capacity and were included in the analysis. Multiple regression analysis showed that occlusal force was significantly related to cognitive function after controlling for possible predictors including age, sex, socioeconomic status, medical condition, and handgrip strength. The number of remaining teeth and periodontal pocket depth were not significantly associated with cognitive function. Among community-dwelling older people with retained competence, maximal occlusal force was positively associated with their cognitive function. These results suggest that oral function might be a predictor for preclinical cognitive decline.Knowledge Transfer Statement: Multiple regression analysis showed that occlusal force was significantly related to cognition after controlling for possible predictors including handgrip strength as an indicator of general muscle strength, suggesting the independence of oral function. The number of remaining teeth did not have this association. The majority of older people have lost teeth and have received prosthodontic treatment, and their occlusal force is determined not only by the number of remaining teeth but also by prosthetic rehabilitation. These results can be used by clinicians focusing on prevention of tooth loss among the entire population, as well as to encourage partially edentulous and fully edentulous patients to restore their oral function with prostheses in order to eliminate a possible risk factor for cognitive impairment.

Ethyl tertiary-butyl ether (ETBE) is a motor fuel oxygenate used in reformulated gasoline. The current use of ETBE in gasoline or petrol is modest but increasing. To investigate the effects of ETBE on splenocytes, mice were exposed to 0 (control), 500 ppm, 1750 ppm, or 5000 ppm of ETBE by inhalation for 6 h/day for 5 days/wk over a 6- or 13-week period. Splenocytes were harvested from the control and exposed mice, and the following cell phenotypes were quantified by flow cytometry: (1) B cells (PerCP-Cy5.5-CD45R/B220), (2) T cells (PerCP-Cy5-CD3e), (3) T cell subsets (FITC-CD4 and PE-CD8a), (4) natural killer (NK) cells (PE-NK1.1), and (5) macrophages (FITC-CD11b). Body weight and the weight of the spleen were also examined. ETBE-exposure did not affect the weight of the spleen or body weight, while it transiently increased the number of RBC and the Hb concentration. The numbers of splenic CD3+, CD4+, and CD8+ T cells, the percentage of CD4+ T cells and the CD4+/CD8+ T cell ratio in the ETBE-exposed groups were significantly decreased in a dose-dependent manner. However, ETBE exposure did not affect the numbers of splenic NK cells, B cells, or macrophages or the total number of splenocytes. The above findings indicate that ETBE selectively affects the number of splenic T cells in mice.

We previously reported that the forest environment enhanced human natural killer (NK) cell activity, the number of NK cells, and intracellular anti-cancer proteins in lymphocytes, and that the increased NK activity lasted for more than 7 days after trips to forests both in male and female subjects. To explore the factors in the forest environment that activated human NK cells, in the present study we investigate the effect of essential oils from trees on human immune function in twelve healthy male subjects, age 37–60 years, who stayed at an urban hotel for 3 nights from 7.00p.m. to 8.00a.m. Aromatic volatile substances (phytoncides) were produced by vaporizing Chamaecyparis obtusa(hinoki cypress) stem oil with a humidifier in the hotel room during the night stay. Blood samples were taken on the last day and urine samples were analysed every day during the stay. NK activity, the percentages of NK and T cells, and granulysin, perforin, granzyme A/B-expressing lymphocytes in blood, and the concentrations of adrenaline and noradrenaline in urine were measured. Similar control measurements were made before the stay on a normal working day. The concentrations of phytoncides in the hotel room air were measured. Phytoncide exposure significantly increased NK activity and the percentages of NK, perforin, granulysin, and granzyme A/B-expressing cells, and significantly decreased the percentage of T cells, and the concentrations of adrenaline and noradrenaline in urine. Phytoncides, such as α-pinene and β-pinene, were detected in the hotel room air. These findings indicate that phytoncide exposure and decreased stress hormone levels may partially contribute to increased NK activity.

We previously reported that a forest bathing trip enhanced human NK activity, number of NK cells, and intracellular anti-cancer proteins in lymphocytes. In the present study, we investigated how long the increased NK activity lasts and compared the effect of a forest bathing trip on NK activity with a trip to places in a city without forests. Twelve healthy male subjects, age 35–56 years, were selected with informed consent. The subjects experienced a three-day/two-night trip to forest fields and to a city, in which activity levels during both trips were matched. On day 1, subjects walked for two hours in the afternoon in a forest field; and on day 2, they walked for two hours in the morning and afternoon, respectively, in two different forest fields; and on day 3, the subjects finished the trip and returned to Tokyo after drawing blood samples and completing the questionnaire. Blood and urine were sampled on the second and third days during the trips, and on days 7 and 30 after the trip, and NK activity, numbers of NK and T cells, and granulysin, perforin, and granzymes A/B-expressing lymphocytes in the blood samples, and the concentration of adrenaline in urine were measured. Similar measurements were made before the trips on a normal working day as the control. Phytoncide concentrations in forest and city air were measured. The forest bathing trip significantly increased NK activity and the numbers of NK, perforin, granulysin, and granzyme A/B-expressing cells and significantly decreased the concentration of adrenaline in urine. The increased NK activity lasted for more than 7 days after the trip. In contrast, a city tourist visit did not increase NK activity, numbers of NK cells, nor the expression of selected intracellular anti-cancer proteins, and did not decrease the concentration of adrenaline in urine. Phytoncides, such as alpha-pinene and beta-pinene were detected in forest air, but almost not in city air. These findings indicate that a forest bathing trip increased NK activity, number of NK cells, and levels of intracellular anti-cancer proteins, and that this effect lasted at least 7 days after the trip. Phytoncides released from trees and decreased stress hormone may partially contribute to the increased NK activity.

In order to explore the effect of forest bathing on human immune function, we investigated natural killer (NK) activity; the number of NK cells, and perforin, granzymes and granulysin-expression in peripheral blood lymphocytes (PBL) during a visit to forest fields. Twelve healthy male subjects, age 37–55 years, were selected with informed consent from three large companies in Tokyo, Japan. The subjects experienced a three-day/two-night trip in three different forest fields. On the first day, subjects walked for two hours in the afternoon in a forest field; and on the second day, they walked for two hours in the morning and afternoon, respectively, in two different forest fields. Blood was sampled on the second and third days, and NK activity; proportions of NK, T cells, granulysin, perforin, and granzymes A/B-expressing cells in PBL were measured. Similar measurements were made before the trip on a normal working day as the control. Almost all of the subjects (11/12) showed higher NK activity after the trip (about 50% increased) compared with before. There are significant differences both before and after the trip and between days 1 and 2 in NK activity. The forest bathing trip also significantly increased the numbers of NK, perforin, granulysin, and granzymes A/B-expressing cells. Taken together, these findings indicate that a forest bathing trip can increase NK activity, and that this effect at least partially mediated by increasing the number of NK cells and by the induction of intracellular anti-cancer proteins.

AbstractElectron heat transport in the low-collisonality electron cyclotron heating plasma is investigated to clarify the effect of neoclassical transport optimization on the thermal plasma transport in the Large Helical Device (LHD). Five configurations are realized by shifting the magnetic axis position in major radius: 3.45, 3.53, 3.6, 3.75, and 3.9 m. A clear effective helical ripple (which is a quantitative measure of the neoclassical transport optimization) dependency on the enhancement factor of the global energy confinement relative to ISS95 is observed. Local heat transport analyses show a higher electron temperature and a lower heat transport in the neoclassical transport optimized configuration at half the minor radius. The comparisons of the experimental total heat fluxes with that of the neoclassical transport by DCOM/NNW suggest that the neoclassical transport plays a significant role in the heat transport and that the neoclassical transport optimization is effective in improving the plasma confinement in the low-collisionality LHD plasma.

A new machining method is proposed to obtain sculptured mirror surfaces by applying elliptical vibration cutting. The tool is vibrated elliptically unlike rotated end mills and fed along the sculptured surface in the proposed method. A 3 DOF ultrasonic vibration tool is developed, which can generate an arbitrary ultrasonic elliptical vibration in the 3D space so that it is suitable to machine the 3D sculptured surfaces. A precision machine tool is also developed, and the proposed method is successfully applied to mirror surface machining of hardened die steel.

AbstractBackground: Synovial sarcoma is a soft tissue sarcoma rarely seen in the head and neck. Due to its rarity and morphologic variations, diagnosis is difficult in most cases. Method: A case of synovial sarcoma arising in the upper neck is presented. The detection of the specific chromosomal translocation t(X;18)(p11.2;q11.2) is also described. Result: A specific chimeric gene, the SSX-SYT fusion gene, was identified in the formalin-fixed paraffin-embedded surgical specimen using the reverse-transcription polymerase chain reaction (RT-PCR) technique. Conclusion: Synovial sarcoma contains a characteristic chromosomal translocation, which serves as a useful diagnostic tool. RT-PCR technique has enabled to detect this specific translocation not only in fresh tissues but also in archival paraffin-embedded specimens.Copyright © 2004 S. Karger AG, Basel

A series of novel 5-amino-6-fluoro-1-[(1R,2S)-2-fluorocyclopropan-1-yl]-8-methylquinolones bearing fluorinated (3R)-3-(1-aminocyclopropan-1-yl)pyrrolidin-1-yl substituents at the C-7 position (2−4) was synthesized to obtain potent drugs for infections caused by Gram-positive pathogens, which include resistant strains such as methicillin-resistant Staphylococcus aureus (MRSA), penicillin-resistant Streptococcus pneumoniae (PRSP), and vancomycin-resistant enterococci (VRE). These fluorinated compounds 2−4 exhibited potent antibacterial activity comparable with that of a compound bearing a non-fluorinated (3R)-3-(1-aminocyclopropan-1-yl)pyrrolidine moiety at the C-7 position (1) and had at least 4 times more potent activity against representative Gram-positive bacteria than ciprofloxacin (CPFX), gatifloxacin (GFLX), or moxifloxacin (MFLX). Among them, the 7-[(3S,4R)-4-(1-aminocyclopropan-1-yl)-3-fluoropyrrolidin-1-yl] derivative 3 (=DQ-113), which showed favorable profiles in preliminary toxicological and nonclinical pharmcokinetic studies, exhibited potent antibacterial activity against clinically isolated resistant Gram-positive pathogens.

The reaction of [Cp*(CO)2Fe{P(NHPh)(OMe)2}]PF6 with NaNH2 yielded the P-iron-bonded iminophosphorane Cp*(CO)2Fe{P(NPh)(OMe)2}, with deprotonation from the NHPh group on the phosphorus. The ruthenium analogue, Cp*(CO)2Ru{P(NPh)(OMe)2}, was also prepared in a similar manner. The spectroscopic properties of the metallaiminophosphoranes are quite similar to those of their P&dbd;O analogue, i.e., phosphonate complexes. The X-ray structure analyses revealed that the P−N bond in Cp*(CO)2Fe{P(NPh)(OMe)2} is comparable in length to that of the ruthenium analogue, Cp*(CO)2Ru{P(NPh)(OMe)2}, and both are considerably shorter than a P−N single bond. These observations strongly suggest that the P−N bonds in the P-metalated iminophosphoranes have substantial double-bond character. The structural features of the metallaiminophosphorane of iron resemble those of the starting cation complex, indicating that their electronic structures are basically similar. The iron-iminophosphorane is thermally stable compared with its Cp analogue, which is not stable enough to isolate, suggesting that both steric and electronic factors are responsible for the stability of the Cp* complexes.

Novel peptidomimetics of the antifungal cyclic peptide Rhodopeptin were synthesized. As with the cyclic peptides, the presence of all three Rhodopeptin side chains was found to be indispensable for peptidomimetic activity. We discovered new compounds exhibiting greater antifungal activity and improved physiochemical properties in comparison to the parent compounds.

Novel nonpeptide peptidomimetics of the antifungal cyclic peptide Rhodopeptin were designed utilizing hydantoin, benzimidazole, d-glucosamine, quinolone, and benzodiazepine units as scaffolds. The scaffolds were chosen on the basis of their potential to improve the physiochemical properties of the peptidomimetics as well as their ability to bear the requisite Rhodopeptin side-chain moieties with the proper three-dimensional orientation.

In Japan, haemodialysis (HD) is usually performed with patients in the supine position. However, the effects of changing posture on major blood components have not been investigated in HD patients. It is possible that several fluid components change rapidly when patients change from the upright to the supine position. We therefore investigated the effects of posture on blood component analysis.

Two types of hydration of bovine serum albumin, BSA, were found by microwave impedance dispersion according to Suzuki et al. (J. Phys Chem. 1996, 100, 7279). At 25.0 °C the number of the strongly bound water molecules, Ns, was 606 and that of the weakly bound water molecules, Nw, was 393 per one BSA molecule. Elevation of the temperature decreased Ns continuously, whereas it increased Nw discontinuously at 30 °C. Addition of an anesthetic halothane increased Nw at the expense of Ns. At halothane 2.5 mM, Ns decreased by 300, and Nw increased by about 350. The electrical conductance of BSA solution showed a minimum at about 1.0 mM halothane. The halothane concentration that showed the minimum electrical conductance coincided with the maximum in Nw. Because the volume fraction of BSA is less than 2.5%, the conductance change is attributable to the change of bulk ion concentrations. The initial decrease of conductance represents the suppression of ionization by halothane. The increase of conductance above 1.0 mM halothane indicates unfolding of the protein, exposing polar moieties to water, thus increasing the ion concentration.

Abstract.: This paper presents an approach, called the ``topology-oriented approach,'' to numerically robust geometric algorithms. In this approach, the basic part of the algorithm is described in terms of combinatorial and topological computation primarily; this description guarantees robustness of the algorithm because combinatorial and topological computation is never contaminated with numerical errors. However, this part of the algorithm is usually nondeterministic, the flow of processing containing many alternative branches. Hence, numerical computation is used in order to choose the branch that seems the most promising to lead to the correct answer. The algorithm designed in this way is robust and simple. The basic idea of this approach as well as the basic properties of the resulting algorithms is shown with examples.

Structure−activity relationships of the west amino acid modified analogues of rhodopeptins, novel antifungal tetrapeptide isolated from Rhodococcus species Mer-N1033, have been investigated. Among the analogues synthesized, 2,2-difluoro and 2-hydroxy derivatives retained the antifungal activity with better physical properties, i.e., solubility or acute toxicity.

Structure−activity relationships of the east and south amino acid modified analogues of rhodopeptins, novel antifungal cyclic tetrapeptides isolated from Rhodococcus species Mer-N1033, have been investigated. It was observed that a basic amino acid moiety (lysine or ornithine) as the east amino acid and a hydrophobic and bulky neutral amino acid (i.e., γ-methylleucine) as the south amino acid were indispensable structure motifs for antifungal activity of rhodopeptin analogues.

Drosophila DLG (Discs Large Tumor Suppresser Protein) is a component of septate junctions, and disruption of its gene leads to over growth of imaginal discs. Homologs of Drosophila DLG recently isolated from mammalian tissue have been classified as members of the MAGUK (Membrane Associated GUanylate Kinase) superfamily of proteins. Using a modified RT-PCR method applied to rat tissues, we have isolated cDNA clones encoding a novel MAGUK family member that we have named rDLG6. Immunoblot and immunohistochemical analyses revealed that rDLG6 protein is predominantly expressed in brain. GST pull-down assays showed that the PDZ domain of rDLG6 protein binds to the C-terminus of the AMPA (α-Amino-3-hydroxy-5-Methyl-isoxazole-4-Propionic Acid) receptor GluR2 subunit.

Regulation of viral RNA levels in infected cells is considered important in the investigation of viral transcription and replication. Amounts of Borna disease virus (BDV) RNAs were increased in confluent persistently BDV-infected MDCK cells (MDCK/BDV) cells, while maintained at low levels in growing cells. The amount of 1.9-kb RNA without cap formation and polyadenylation at the 5′ and 3′ ends respectively were remarkably increased (200% per day) in confluent MDCK/BDV cells. Both the full-length genomic and anti-genomic RNAs were increased accompained by 1.9-kb RNA, suggesting the transcription of the 1.9-kb RNA was important for replication of BDV. Ribavirin has an inhibitory effect on replication and transcription of BDV at concentrations from 1 to 10 μg/ml [Mizutani T et al., Arch Virol (1998)143: 2 039–2 044]. BDV transcripts were decreased with ribavirin treatment and increased after its removal which indicated that ribavirin has a reversible inhibitory effect on BDV transcription. Furthermore, BDV transcription was also decreased by two agents, RMNPA and EICAR, which selectively inhibit enzyme activity related to cap formation at the 5′ end of mRNA. On the contrary, when the growing MDCK/BDV cells were treated with actinomycin D, transcripts of BDV RNA were increased for 24 h. These agents and culture conditions in this study were found to be useful tools for up-and down-regulation of BDV transcription in persistently BDV-infected cells.

On the basis of a simple and unified phenomenological method for treating the excess volume of mixing in multicomponent systems, formulas were developed for the partial molar volume of a copolymer (or a mixture of two polymers) in a mixture of two solvents and for the volume contraction factor in the bulk-copolymerization of two monomers. The possible dependence of the “net” volume of a copolymer on its primary structure was taken into account. The proposed formulas were partly checked against some experimental data, and found satisfactory. The previously proposed formulas for the copolymerization contraction factor seem to be less generally applicable.

A method is proposed to determine the rigid structure as well as the three dimensional motion of an object from a sequence of orthographically projected images. It is assumed that the velocities as well as the positions of the points attached to the object are observable in the images. The “instantaneous rigidity condition” wich states that the relative position vector between any two points is orthogonal to the corresponding relative velocity vector is derived from the condition of rigidity. Assuming further that the rotational velocity component is constant throughout the period of observation, each of the projected relative velocity vectors is shown to move along one of the elliptical trajectories, all of which are similar to one another. The orientations of the longer axes are common to every trajectory. Then one can determine the equations of the ellipses by observing only two points in three views or three points in two views. The solution is obtained from a set of linear equations. The length of the longer axis enables one to determine the relative velocity. The instantaneous rigidity condition is then used to obtain the relative position. The special cases where the rotational axis is either perpendicular to or parallel with the image plane are discussed. The above results are discussed in relation to the relevant psychophysical observations as well as theoretical studies.

The host defense of germfree (GF) mice against primary infection with Listeria monocytogenes was compared with that of specific-pathogen-free (SPF) mice. In SPF mice, the numbers of bacteria in the peritoneal cavity, liver, and spleen decreased gradually to undetectable levels by day 8 after intraperitoneal infection with a sublethal dose (2 X 10(3) CFU) of L. monocytogenes. On the other hand, the elimination of bacteria in these organs of GF mice was significantly impaired at this stage after inoculation. We have reported previously that T cells coexpressing L-selectin and CD44 play an important role in protection against L. monocytogenes through trafficking to sites of inflammation. Consistent with our previous findings, the number of unique L-selectin+ CD44+ T cells in the peritoneal cavity was remarkably increased on day 8 after infection in SPF mice, whereas such an increase was not evident in GF mice at this stage. Listeria-specific T-cell proliferation was normally detected in the lymph node cells of GF mice inoculated with L. monocytogenes, whereas the T-cell-proliferative response of the peritoneal exudate cells of GF mice was significantly impaired compared with that of SPF mice. These results suggest that the priming of T cells against listerial antigens normally occurs in the peripheral lymphoid organs of GF mice but the trafficking of the activated T cells to the inflamed sites may be severely impaired in GF mice, resulting in increased susceptibility to infection with L. monocytogenes.

The expression of proliferative cell nuclear antigen (PCNA) in follicular tumours of the thyroid was examined by immunohistochemistry. Both usual nonoxyphilic cell follicular tumours (non-OCT) and oxyphilic cell tumours (OCT) were subdivided into benign, indeterminate, encapsulated carcinoma, and widely invasive carcinoma types. Among non-OCT the percentages of PCNA-positive cells in benign tumours, encapsulated carcinomas, and widely invasive carcinomas was 2.5%–8.6%, 11.8%–39.1%, and 18.6%–20.0%, respectively. There was a statistically significant difference between benign tumours and encapsulated or widely invasive carcinomas, as in previous studies. A value of 10% was appropriate to distinguish benign from malignant lesions. PCNA-positive cells in indeterminate-type non-OCT were not significantly different from those in benign tumours, ranging from 4.3%–19.6%, and occurring at more than 10% in three of six tuours. Among OCT the positivity was less than 10% in benign tumours (4.5%–7.8%) and more than 10% in malignant tumours (14.1%–35.9%) and all the eight indeterminate tumours (12.5%–27.3%), with a statistically significant differences between the benign tumour and each of the latter types. These results indicate that the examination of PCNA is valuable in diagnosis of thyroid follicular tumours and that the use of similar diagnostic criteria may be warranted in both non-OCT and OCT.

The concentration and the localization of transforming growth factor (TGF)-α and epidermal growth factor (EGF) in the submandibular glands (SMGs) of male Wister rats of different ages (postnatal 0 to 10 weeks of age) were examined. Highest levels of TGF-α were seen early, at postnatal day 0; the levels dropped thereafter in an age-dependent manner, while EGF was not detectable before the third postnatal week. Immunoreactive localization of EGF was restricted to the granules of the granular convoluted tubule (GCT) cells in the mature SMGs, whereas TGF-α was observed throughout postnatal development over the entire duct system. TGF-α was demonstrated in the cytoplasm at early stages when the GCT granules were not observed and was also located on the granules at the late stage, as was the case for EGF, indicating that TGF-α is colocated with EGF in the mature SMG. These results demonstrate the differences between the expression of TGF-α and that of EGF in the developing rat SMG.

Decay processes of H2^- anions in γ-ray-irradiated solid parahydrogen were studied by using an ESR spectrometer. The following interesting results were obtained. First, the initial yields of ortho-H2^- anions in the γ-irradiated solid parahydrogen were 3 times as large as those of para-H2^- anions. Second, the yields of para-H2^- anions decrease faster than those of ortho-H2^- anions upon storage of the irradiated samples at 4.2 K. Third, the decay rate of H2^- anions is accelerated by the addition of HD and D2 molecules. Fourth, H2^- anions at 2.2 K decay faster than at 4.2 K. According to the parity conservation rule in a homonuclear diatomic molecule, the energy of ortho-H2^- anions at the ground state is lower than that of para-H2^- anions, whereas that of ortho-H2 molecules is higher than that of para-H2 molecules at low temperatures. The first and second results are ascribed to para → ortho conversion of H2^- anions. The third result indicates that nearby HD and D2 accelerate the decay of H2^- anion or broaden the spectra by superhyperfine interactions. The fourth result suggests a coherent tunneling mechanism of decay of H2^- anions in the solid parahydrogen.

It is well known that cytoskeleton and karyoskeleton proteins are associated with changes in cell shape and with the rearrangement of the dynamic structures involved in cell division and motility. In higher vertebrates, there are three major skeletal protein groups: microfilaments, microtubules and intermediate filaments, each representing a multigene family. Some of these skeletal proteins are expressed in a temporally- and spatially-specific fashion, and they establish cell-specific cytoplasmic and nucleoplasmic organization during development. Here we report the cDNA cloning of a novel 60 kDa skeletal protein from mouse spermatocytes, termed MNS 1 (meiosis-specific nuclear structural protein), whose computer-predicted protein configuration indicates long a-helical coiled-coil domains flanked by non-helical terminal domains. Functional characterization of MNS1 by ectopic expression in culture cells indicated that it is a detergent-and high salt-resistant skeletal protein which is involved in organization of the nuclear or perinuclear architecture. The MNS1 protein is specifically expressed at the pachytene stage during spermatogenesis, so that its function may involve the determination and maintenance of the appropriate nuclear morphology during meiotic prophase.

The renin-angiotensin system is important in cardiovascular remodeling. Although a variant of the angiotensinogen gene is associated with an increased generation of angiotensinogen, it is unclear how this genetic variant might influence the activity of the renin-angiotensin system and thereby contribute to the predisposition to coronary artery disease (CAD). The relation between genetic polymorphisms in the gene-encoding angiotensinogen and the risk of CAD in middle-aged Japanese men was investigated. Two polymorphisms in exon 2 of the angiotensinogen gene, M235T and T174M, were analyzed in 327 patients with CAD and 352 matched control subjects. The genotype distribution of both polymorphisms did not differ between patients with CAD and control subjects. No combination of genotypes of the two polymorphisms was associated with CAD. Results indicate that the M235T and T174M variants of the angiotensinogen gene are not associated with CAD in Japanese men. (Am Heart J 1997;134:260-5.)

Conformational properties of isolated diblock copolymer molecules comprising an incompatible polymer pair, poly-A and poly-B, were examined by Monte Carlo calculations, light scattering, and intrinsic viscosity analysis. It was established that the mean-square radius of gyration K of the K block (K = A or B) is, in all cases, almost the same as the radius H-K of the homopolymer identical to the K block. On this basis, light scattering and viscosity data for polystyrene-poly(methyl methacrylate diblock copolymers which had been strictly characterized with respect to the molecular weight and compositional heterogeneity were analyzed with atiention to the heterogeneity. It was found that the parameter σ, which is a function of the meansquare distance between the centers of mass of the two blocks, 2σ= /(H-A+ H-B) is very close to the average of the σ values for the homopolymers in all types of solvents examined.
