Abstract Young (3-month-old) and aged mice (18-month-old) were fed a diet containing Japanese herbal medicine (TJ-41 or TJ-48) for 5 months, and the effect of the herbal medicines were examined in terms of levels of monoamines and their metabolites in the brain of young and aged mice. In the aged mice, the levels of norepinepherine, serotonin and their metabolites in the brain were decreased in the cortex, hippocampus and hypothalamus. Feeding of diet containing TJ-48, but not TJ-41, enhanced the levels of some monoamines and their metabolites in the brains of aged mice, comparable to those of young mice. The results indicated that the improvement of levels of monoamines by Japanese herbal medicine was observed only in the aged mice, not in the young mice. The data have suggested the importance of the aged animals to see the effect of medicine on the functions of organs or systems.

Abstract:  To determine the efficacy of cyclosporin A (CysA) for the treatment of steroid-resistant interstitial pneumonitis (IP), we enrolled 25 patients with various rheumatic diseases and steroid-resistant IP in a pilot study [4 patients with rheumatoid arthritis (RA), 2 with systemic lupus erythematosus (SLE), 11 with polymyositis/dermatomyositis (PM/DM), 4 with systemic sclerosis (SSc), 1 with mixed connective tissue disease (MCTD), 3 with Sjögren syndrome (SS)]. Twelve patients (48%) showed a persistent response to CysA therapy, and 7 of them had PM/DM, including so-called amyopathic DM. Patients with a persistent response had moderately elevated lactate dehydroxygenase (LDH) levels, whereas patients who died had much higher LDH levels and hypoxia. Even patients with low blood levels of CysA achieved a persistent response. In responding patients, the symptoms, chest X-ray findings, arterial oxygen tension, and LDH level all improved after less than 4 weeks. In conclusion, CysA seem to be useful for treating patients with steroid-resistant IP, whose duration is short and severity is mild.

Abstract: We studied the characteristics of anti-Ki antibodies that react with a synthetic peptide (KILT) which has homology with SV40 large T antigen nuclear localization signal (SV40 T NLS). Immunoreactivity of antibodies to KILT was tested by enzyme-linked immunosorbent assay (ELISA), and this reactivity was compared with results obtained by immunoblotting using purified Ki and ELISA using purified or recombinant Ki antigen. The clinical significance of anti-KILT antibodies in lupus patients was also studied. Twenty percent of anti-Ki sera reacted with KILT, and all sera that reacted with KILT recognized both purified and recombinant Ki antigen in all our assay systems. A significant correlation was observed between reactivities in ELISA using KILT and with these using purified and recombinant Ki. Some sera with low titers by double immunodiffusion (DID) reacted with KILT, whereas high-titer anti-Ki sera showed a tendency to react with Ki antigen in different assays. The prevalence of discoid rash and sicca complex was higher in the anti-KILT-positive group. The amino acid sequence homologous to SV40 T NLS is an immunologically active epitope on the Ki antigen; reactivity to this epitope is associated with characteristic clinical features allied with anti-Ki antibodies in lupus patients.

AbstractRengeite, Sr4ZrTi4Si4O22, is a new member of the perrierite–chevkinite group found in the jades from the Itoigawa-Ohmi district, central Japan. It is monoclinic, P21/a, a= 13.97(1), b= 5.675(7), c= 11.98(1)Å, ß = 114.26(8)°, V= 866 Å3and Z= 2. The six strongest lines in the X-ray powder diffraction pattern are 3.12 (s) (40-3), 3.05 (vvs) (31-3), 2.99 (vs) (311), 2.84 (s) (020), 2.74 (s) (004), 2.20 (s) (31-5). Electron microprobe analysis gave SiO222.58, TiO229.88, ZrO29.49, Nb2O50.24, Ta2O50.07, Al2O30.20, FeO 0.10, CaO 0.43, SrO 34.32, BaO 0.13, La2O30.00, Ce2O30.38, Pr2O30.10, Nd2O30.29, Sm2O30.04, total 98.25 wt.%, corresponding to (Sr3.62Ca0.08Ce0.03Nd0.02Ba0.01Pr0.01)?3.76(Zr0.84Ti0.09Al0.04Fe0.02Nb0.02)?1.01Ti4.00Si4.11O22on the basis of O = 22. The unitcell parameters and chemical composition imply that rengeite is the Sr and Zr-analogue of perrierite or high-ß analogue of strontiochevkinite. It is transparent, dark brown with adamantine lustre. Its streak is pale greenish brown, and no cleavage was observed. The hardness is VHN100606–698 kg mm-2(Mohs 5–5.5). The calculated density is 4.12 g cm-3. It is strongly pleochroic from pale green to pale greenish brown where the REEcontents are <1 wt.% and pale violet to greenish brown where the REEcontents are between 3 and 10 wt.%. It occurs as anhedral grains in close association with titanite, zircon and tausonite in a pebble of blue titanian omphacite-jadeite rock from the seashore of Oyashirazu, Ohmi Town, in a boulder of lavender-coloured Ti-bearing jadeitite from the bed of the Kotaki-gawa river, Itoigawa City, and in a boulder of green jade from the bed of the Hime-kawa river, Itoigawa City, Niigata Prefecture, central Japan. Rengeite is considered to have crystallized by interaction between pre-existing minerals (rutile, anatase, titanite and zircon) and Sr-rich metamorphic fluid during later stage activity of high-P/Tmetamorphism. The name is for Mt. Renge near the locality and the Renge metamorphic belt where jadeitite deposits are found.

It is well known that IL-2 production of CD4(+) T cells from old mice (old T cells) is impaired. In this study, we have examined TCR complex zeta chain expression of old T cells and their TCR downstream signal transduction pathways stimulated with anti-CD3. Activation of protein tyrosine kinases, Fyn and ZAP-70, and turnover of inositol phosphates stimulated with anti-CD3 were severely impaired in old T cells, although levels of these proteins were comparable to those in young T cells. Increase in intracellular Ca2+ concentration in old T cells was also impaired. Old T cells starting the Ca(2+) oscillation by the anti-CD3 stimulation were severely decreased in number and the oscillation waves were broader in shape. T cells with zeta-FcvarepsilonRgamma heterodimer in the TCR-CD3 complex were increased in proportion in old T cells with a concomitant decrease in the T cells with zeta-zeta homodimer. The density of the TCR-CD3 complex on old T cells was confirmed to be comparable to that on young T cells. The impairment in TCR downstream signal transduction pathways and the increase in zeta-FcvarepsilonRgamma heterodimer in the TCR-CD3 complex were confirmed to be the situation in Th1 clones established from old mice. These results indicate that old T cells are impaired in response to TCR stimulation, because T cells with the TCR-CD3 complex containing the zeta-FcvarepsilonRgamma heterodimer are increased in proportion in old T cells.

Expression of mRNA protein tyrosine phosphatases (PTPs) was surveyed in an esophageal cancer cell line by RT-PCR using degenerate primers. The mRNAs for eight kinds of PTPs were expressed in the cell line. We examined mRNA expression of these PTPs in 12 cases of esophageal cancer by Northern analysis. Significant signals were obtained for three kinds of PTPs, PTP1B, PTPH1, and PTPD1. The magnitude of expression of each PTP was measured as the ratio of the signal intensity of each PTP to that of a control gene (NADPH), and the ratio was then compared to normal mucosa around the cancer lesion. Among the three kinds of PTPs, the expression of PTP1B mRNA was significantly depressed in cancer lesions compared with that in the surrounding normal mucosa. In contrast, the expression of PTPH1 mRNA was significantly increased in cancer lesions compared with that in normal mucosa. PTPD1 did not show any significant trend in comparisons of cancer and surrounding normal mucosa. The results suggest that PTP1B and PTPH1 are engaged in opposing signaling pathways, the tumor-suppressive and tumor-promoting pathways, respectively, in esophageal carcinogenesis.

CD24 antigen is a glycoprotein expressed on haematopoietic cells, including B cells, T cells and granulocytes and on non-haematopoietic cells, including neural cells, ganglion cells and the cells of the adrenal medulla. The antigen is also expressed on renal cell carcinoma, small cell lung carcinoma and neuroblastoma. We have cloned rat cDNA encoding core polypeptide of CD24 antigen from embryonic brain and shown that the core molecule is highly expressed in embryonic brain and non-neural tissues. Rat tissue and various human neoplastic cell lines were investigated for the gene expression of CD24 core polypeptide by in situ hybridization. The transcript was localized in gastrointestinal epithelia, ductal and acinar epithelia of the salivary gland, the bronchiolar epithelium, renal tubular epithelium, the epithelium of the oviduct, follicular cells of the thyroid, medullary cells of the adrenal gland, Auerbach's plexus, B blastoid cells in lymph nodes, hair follicles, and the sweat glands of the skin. Among the various human neoplastic cell lines investigated, the transcript was detected in squamous cell carcinoma of the lung, gastric carcinoma, colon carcinoma, choriocarcinoma and renal cell carcinoma. The result suggest that the core molecule of CD24 antigen may be expressed in a wider range of epithelial cells and carcinoma cell lines than has been reported. Furthermore, we show that gene expression of CD24 core polypeptide is confined to the proliferative zone of the gastrointestinal mucosa, suggesting that core molecule is transiently expressed on the surface of epithelial cells in the process of cellular maturation. We discuss a possible role for CD24 antigen in the maturation of epithelial cells in the gastrointestinal tract.

Fv-4 is a mouse gene that dominantly confers resistance to infection by ecotropic murine leukemia virus (MuLV). We previously demonstrated that mixed radiation bone marrow chimeras containing Fv-4r-bearing BALB/c-Fv- 4Wr (C4W) bone marrow and Fv-4r-bearing C3H/He (C3H) bone marrow grafted into C3H recipient mice (C4W+C3H-->C3H) were resistant to Friend leukemia virus (FLV)-induced leukemogenesis, even when they contained as high as 70% C3H-derived cells. This indicates that FLV- sensitive C3H-derived cells are rendered refractory to infection and/or transformation with FLV when they coexist in mice with Fv-4r-bearing cells. To investigate the mechanism of Fv-4 resistance to FLV-induced leukemogenesis, we first examined the expression of Fv-4r env antigen in the peripheral blood mononuclear cells (PBMC) of these chimeras. The Fv-4r env antigen was present not only on C4W-derived cells, but also on Fv-4r-bearing C3H-derived cells in C4W+C3H-->C3H mixed bone marrow chimeras. The Fv-4r env antigen that binds to the cells surface of C3H cells was found in sera from normal C4W mice, C4W-->C3H chimeras, and C4W+C3H-->C3H mixed chimeras. The serum Fv-4r env antigen binds to ecotropic MuLV receptors, shown by specific binding to transfectant mink cells expressing ecotropic MuLV receptor, but not to parental mink cells. To determine whether the binding of Fv-4r env antigen to the putative MuLV receptors would block FLV infection, C3H thymocytes or spleen cells that had been preincubated with C4W serum were mixed with FLV and the subsequent production of MuLV specific antigens was examined. C3H thymocytes or spleen cells treated with C4W serum became refractory to binding by FLV. These results provide evidence that the Fv-4r env antigen is released from C4W-derived cells in vivo and binds to cells expressing surface receptors for ecotropic MuLV, thereby protecting them from infection with FLV. The implication of these findings for gene therapy of retrovirus-induced disease such as acquired immune deficiency syndrome (AIDS) is discussed.

Immunohistochemical and clinicopathological studies were performed in 27 autopsy cases with indisputable DIC, which had been selected from 1,800 autopsy cases of elderly people based on the following two criteria; 1. presence of fibrin thrombi in glomeruli, and 2. presence of fresh patchy necrotic foci in myocardium and/or fibrin thrombi in splenic sinuses. A high incidence of liver lesions (22/27) was revealed in autopsy cases with indisputable DIC. The liver lesions could be classified into four groups. Group-I (Central degeneration) was characterized by massive precipitation of fibrin irregularly around the central vein, causing parenchymal damage. Group-II (Central necrosis), showed coagulation necrosis in the cental zone due to circulatory disturbance caused by either shock as a cause of DIC or abrupt cessation of blood flow into the lobules following fibrin thrombus formation in vessels of Glisson's sheath. Both group-I and -II showed a short clinical duration of DIC. Group-III (Sinusoidal thrombosis), showed the presence of fibrin thrombi in sinusoids with mild parenchymal damage and long clinical duration of DIC. Group-IV (No thrombosis), showed neither parenchymal damage nor fibrin thrombi in sinusoids, but a long clinical duration of DIC.

An immunohistological study of Alzheimer's brains was performed using antibodies to C-series gangliosides and microtubule-associated protein 5 (MAP5), and their staining patterns were compared with those of antibodies to tau and ß-amyloid precursor protein. Antibodies to C-series gangliosides and MAP5, both of which are known to preferentially expressed in the fetal brains, immunostained dystrophic neurites of senile plaques, neurofibrillary tangles and neuropil threads abundant in 3rd and 5th layers in the cerebral cortex, all of which are considered to be pathological hallmarks of Alzheimer's disease. The immunostaining patterns of these structures by antibodies to C-series gangliosides and MAP5 were similar to those by the antibody to tau. These three antibodies also immunostained some neurons in Alzheimer's brain, although their staining patterns were slightly different from one another; i.e., both diffuse and granular patterns were seen by the antibody to tau, but only granular pattern by the antibodies to C-series gangliosides and MAP5. These neurons immunostained by these three types of antibodies appeared to be the precursors of the classical neurofibrillary tangles, as positively stained neurons were not seen in the brains of non-demented cases. The presence of fetal antigens such as the C-series gangliosides and MAP5 in Alzheimer's brain may suggest that regeneration or sprouting of neurons is ongoing in association with the re-induction of gene expression characteristic for the brain in the early stage of development.

Effects of cholinergic agonists/antagonists on apoptosis and differentiation of murine thymus cells were investigated in two in vitro models. Treatment with 10−7M carbachol was found to counteract the effects of a cortical thymus epithelial cell line (TEC 1.4), on apoptosis and the ratio of CD4 CD8 DP/DN cells in cocultured fetal tjymus lobes. A medullary line (TEC 2.3) did not influence apoptosis in fetal thymus lobes. Both TEC lines had the same strong apoptotic effect on thymus cells in suspension, but only the effect of TEC 1.4 was counteracted by carbachol. This cholinergic influence on TEC 1.4 cells is mediated via nicotinic cholinergic receptors, since d-tubocurarine, but not atropine, effectively blocked the effect of carbachol. The results suggest that cholinergic signals to thymic epithelial cells may have regulatory influence on thymic differentiation and selection processes.

Previous reports demonstrated that the expression of thrombomodulin (TM) in endothelial cells was modulated by various agents. Although TM was down-regulated by endotoxin or cytokines, up-regulation of TM was accomplished when endothelial cells were stimulated with unphysiologically high concentrations of cyclic AMP derivatives or tumour-promoting phorbol esters. We investigated the expression of TM in human umbilical-vein endothelial cells (HUVECs) by physiological substances that can be released into the bloodstream. Histamine (0.1-10 microM, 1-48 h) increased TM activity, TM antigen in cell lysates and TM mRNA levels, but 5-hydroxytryptamine and bradykinin had no effect. Enhancement of TM activity by histamine was completely blocked by the H1-selective antagonist pyrilamine, whereas the H2-antagonist cimetidine had no effect, showing that histamine up-regulates TM activity via H1-receptors on HUVECs. Enhanced TM activity by histamine and the resultant increase in protein C activation might play a role in a feedback regulation for prevention of vascular thrombosis.

The immunoreactivity of anti-neuron-specific enolase (NSE) and anti-Leu-7 on formalin-fixed sections of human fetal salivary gland epithelium was determined by the avidin-biotin-peroxidase complex (ABC) method. In addition, expression of some neuropeptides such as vasoactive intestinal polypeptide (VIP), somatostatin (SRIF), and substance P in the human salivary gland epithelium during the gestational period was observed, whereas the other polypeptides examined, including glucagon, cholecystokinin (CCK), Leu-enkephalin, and calcitonin were absent. NSE and Leu-7 immunoreactivity in the fetal salivary gland epithelium was observed solitarily or in groups commonly restricted to the developing duct epithelium. Positive immunoreactivity was observed in 46 cases with NSE (73%) and 44 cases with Leu-7 (70%) in 63 fetal salivary glands examined. In contrast, the incidence of positive cases stained with neuropeptides was lower than those of NSE and Leu-7 immunoreactivity in the human fetal salivary gland epithelium. These findings indicate that certain neuropolypeptides, as well as VIP, SRIF, and substance P present in the human fetal salivary gland epithelium may play a significant role in the development of the gland.

Zusammenfassung: Zur direkten Spurenanalyse von Kupfer, Mangan, Silber und Blei in Stählen wird eine normale Graphit-Becherküvette verwendet, mit deren Hilfe kleine Proben direkt in fester Form eingebracht werden können. Die Absorptionsintensitäten und Verdampfungsgeschwindigkeiten hängen von der Temperatur und der Diffusionsgeschwindigkeit im Eisen ab. Die Oberfläche der Proben spielt eine wichtige Rolle bei der Verdampfung von Spurenelementen aus Festkörpern. Mehrfache Aufschmelzung für Kupfer und Mangan ist erforderlich bei Temperaturen von etwa 2080° C. Leicht atomisierbare Elemente (Ag, Pb) können mit einer einmaligen Heizstufe bestimmt werden. Die Verdampfung wird in diesem Fall auch von der Oberflächengröße der Probe beeinflußt. Die Nachweisgrenzen betragen 0,1 ng für Cu, 0,04 ng für Mn und 0,008 ng für Ag.

The full length of cDNA encoding murine ZAP-70 was obtained from a cDNA library of embryonic mouse thymus. Northern blot analysis indicated that the expression of ZAP-70 mRNA was most pronounced in the thymus and spleen and also slightly in the brain. Analysis by in situ hybridization revealed that the mRNA was expressed not only in T cells in the thymus and spleen, but also in nerve cells of the brain. In the thymus, the distribution of positive cells was different between newborn and young adult mice. In the newborn thymus, positive signal in thymocytes was localized in the subcapsular layer as well as in the medulla, while in the young adult thymus, it was localized exclusively in the medulla. Furthermore, the in situ hybridization revealed that positive signal was also seen in the liver, intestine, and lung of embryonic mice. These findings have suggested that ZAP-70 plays an important role in growth, differentiation, and function of not only T cells but also nerve cells and several embryonic tissues. The high-affinity receptor (R) for IL-5 consists of a unique alpha chain (IL-5R alpha) and a beta chain (beta c) that is shared with the receptors for IL-3 and granulocyte macrophage colony stimulating factor (GM-CSF). We defined two regions of IL-5R alpha for the IL-5-induced proliferative response, the expression of nuclear proto-oncogenes, and the tyrosine phosphorylation of cellular proteins including beta c, SH2/SH3-containing proteins and JAK2 kinase. In the studies described here, we demonstrate that IL-5, IL-3 or GM-CSF stimulation induces the tyrosine phosphorylation of JAK2, and to a lesser extent JAK1, and of STAT5. Mutational analysis revealed that one of the proline residues, particularly Pro352 and Pro355, in the membrane-proximal proline-rich sequence (Pro352-Pro353-X-Pro355) of the cytoplasmic domain of IL-5R alpha is required for cell proliferation, and for both JAK1 and JAK2 activation. In addition, transfectants expressing chimeric receptors which consist of the extracellular domain of IL-5R alpha and the cytoplasmic domain of beta c responded to IL-5 for proliferation and tyrosine phosphorylation of JAK1. Intriguingly, electrophoretic mobility shift assay analysis revealed that STAT5 was activated in cells showing either JAK1 or JAK2 tyrosine phosphorylation. These results indicate that activation of JAK1, JAK2 and STAT5 is critical to coupling IL-5-induced tyrosine phosphorylation and ultimately mitogenesis, and that Pro352 and Pro355 in the proline-rich sequence appear to play more essential roles in cell growth and in both JAK1/STAT5 and JAK2/STAT5 activation than Pro353 does.
