ABSTRACTThere is growing interest in the use and utility of news media content analysis in tourism research. We report a content analysis of 153 news media articles on a topic of growing interest among tourism researchers globally: tourism and biosecurity. Our work is based on media reports from Aotearoa New Zealand news sources over the period 2009–2019, identifying five key themes. Together they illustrate how tourism, nature conservation, agriculture, horticulture and forestry are utterly and irretrievably interconnected in global flows of people and organisms. They also highlight the important role played by the many actors at policy and operational levels in tourism and border control that have a role to play in minimising potential impacts of biosecurity breaches. We conclude by discussing the value of media content analyses in tourism research stressing that the method, on its own, will reveal only part of the story, and should always be followed up with more intense and critical interrogation of the issues under study.

ABSTRACTThis article discusses the relationships among leadership, race and racism, and social constructionism within counseling leadership. Accordingly, we critically examine the dominant constructions of race and racism within leadership. Next, we propose seven social constructionist principles to inform leadership’s construction and praxis within counseling. Finally, we utilize social constructionism and leadership, further informed through race-centered and anti-racism literature, to further this profession’s constructions of race and racism within leadership and future race-centered and anti-racism leadership actions.

ABSTRACTLeadership as part of school counselor competence remains under-addressed in supervision. Grounded in prior research models, we introduce the Model for Supervision of School Counseling Leadership (MSSCL) which is a 10 x three x three framework. We provide examples for each of the 90 different leadership x Focus x Role combinations through an appendix that can be used in supervision, discuss the use of the MSSCL, and identify implications for practice, training, and future research.

Avian wing morphing allows dynamic, active control of complex flight manoeuvres. Previous linear time-invariant (LTI) models have quantified the effect of varying fixed wing configurations but the time-dependent effects of morphing between different configurations is not well understood. To fill this gap, I implemented a linear parameter-varying (LPV) model for morphing wing gull flight. This approach models the wing joint angles as scheduled parameters and accounts for nonlinear kinematic and gravitational effects while interpolating between LTI models at discrete trim points. With the resulting model, I investigated the longitudinal response associated with various joint extension trajectories. By optimizing the extension trajectory for four independent objectives (speed and pitch angle overshoot, speed rise time and pitch angle settling time), I found that the extension trajectory inherent to the gull wing does not guarantee an optimal response but may provide a sufficient response with a simpler mechanical implementation. Furthermore, the results indicated that gulls likely require extension speed feedback. This morphing LPV model provides insights into underlying control mechanisms, which may allow for avian-like flight in future highly manoeuvrable uncrewed aerial vehicles.

Birds morph their wing shape to accomplish extraordinary manoeuvres1–4, which are governed by avian-specific equations of motion. Solving these equations requires information about a bird’s aerodynamic and inertial characteristics5. Avian flight research to date has focused on resolving aerodynamic features, whereas inertial properties including centre of gravity and moment of inertia are seldom addressed. Here we use an analytical method to determine the inertial characteristics of 22 species across the full range of elbow and wrist flexion and extension. We find that wing morphing allows birds to substantially change their roll and yaw inertia but has a minimal effect on the position of the centre of gravity. With the addition of inertial characteristics, we derived a novel metric of pitch agility and estimated the static pitch stability, revealing that the agility and static margin ranges are reduced as body mass increases. These results provide quantitative evidence that evolution selects for both stable and unstable flight, in contrast to the prevailing narrative that birds are evolving away from stability6. This comprehensive analysis of avian inertial characteristics provides the key features required to establish a theoretical model of avian manoeuvrability.

Recent research reported changes in fetal growth due to increased methyl donor availability through periconceptual supplementation of the dam. We hypothesized that maternal supplementation of methionine (MET) or guanidinoacetic acid (GAA) during the periconceptual period will result in a methyl donor surplus or deficiency in maternal serum and ultimately influence fetal body and organ weight at d +63 gestation. The objectives of this study were to evaluate effects of methyl donor surplus or deficiency during the periconceptual period of gestation on metabolite concentrations in the heifer and the effects on early fetal development. Eighty MARC II heifers (n = 20 per treatment, initial BW = 346 ± 8 kg) were stratified by age and weight to one of four treatment groups to receive 100 g of supplement: ground corn carrier as control (CON), MET (10 g/d) in ground corn, GAA (40 g/d) in ground corn, and MET + GAA (10 g/d MET + 40 g/d GAA) in ground corn. Supplementation began 63 d before breeding (d –63) until d 63 of gestation (d +63), and serum samples were collected before feeding on d -63, at breeding, and d +63. Heifers were bred using male sexed semen from the same sire, and 35 heifers were confirmed pregnant (CON, n = 10; MET, n = 8; GAA, n = 7; MET + GAA, n =10) and harvested at d +63 of gestation to collect maternal and fetal samples. Data were analyzed as a 2 × 2 factorial design with 2 levels of MET and 2 levels of GAA. Methionine concentrations in maternal serum were greater (P= 0.05) in MET and MET + GAA supplemented heifers at d 0 and d +63 compared with CON and GAA at d -63, d 0, and d +63. There were no differences (P≥ 0.15) in the concentration of GAA or creatine due to supplementation; however, both were decreased at breeding (P <0.01) compared with d -63 and d +63. Fetuses from MET supplemented heifers had greater (P= 0.01) brain weight and Longissimus muscle than dams not receiving MET. We conclude that MET supplementation during the periconceptual period can alter methyl donor availability and increase fetal muscle development. USDA is an equal opportunity provider and employer.

Recent research has demonstrated that feeding rumen protected methionine (MET) products increases circulating methionine while feeding guanidinoacetic acid (GAA) may result in a methyl deficiency. Therefore, we hypothesized that maternal MET or GAA supplementation from 63 days before breeding until d 63 of gestation would increase fetal hepatic methyl donors or result in a fetal hepatic methyl donor deficiency, respectively. Eighty MARC II heifers (n = 20 per treatment; initial BW = 344 ± 8 kg; day -63) were weighed and assigned to one of four breeding groups and four treatments stratified by age and starting weight. Heifers were fed a total mixed ration to gain 0.68 kg/d and received 100 g of one of four treatments top dressed: control supplement of ground corn (CON), MET (10 g/d) in a ground corn carrier, GAA (40 g/d) in a ground corn carrier, or MET + GAA (10 g/d MET + 40 g/d GAA) in a ground corn carrier. At breeding (d 0), all heifers were bred to a single sire using male sexed semen. At d 63 of gestation, 35 heifers (CON n = 10, MET n = 8, GAA n = 7, MET + GAA, n = 10) confirmed pregnant with bull calves were humanely slaughtered and fetal liver tissues were collected and sent to Metabolon Inc. for global metabolomics analysis. Data were analyzed as a 2 x 2 factorial with 2 levels of GAA and 2 levels of MET with breeding group included in the model. For this abstract, only metabolites from the methionine cycle and adjacent pathways will be reported and only metabolites with significant P-values will be discussed (P≤ 0.05). There were no main effects or interactions for the abundance of methionine, GAA, creatine, SAM, SAH or homocysteine. Glycine was decreased and sarcosine increased in fetuses from MET supplemented dams. Serine was affected by a MET x GAA interaction being decreased in livers of CON, MET, and MET + GAA fetuses compared with GAA. Cystathione, S-methylcysteine, s-methylcysteine sulfoxide, and hypotaurine were greater in fetuses of MET supplemented heifers and pyridoxal phosphate was reduced in fetal liver of MET supplemented heifers. The abundance of urea was greater in GAA supplemented fetuses. These results indicate that fetuses from dams fed methionine may be able to maintain balance in methionine supply by shunting methionine through adjacent pathways including the transsulfuration pathway. We also interpret these data to imply that excess methionine is not utilized by the fetus and is metabolized to products that can be excreted. Lastly, GAA did not alter the fetal methyl supply. The objective of this study was to determine the dose-dependent response of one-carbon metabolite (OCM: methionine, choline, folate, and vitamin B12) supplementation on heifer dry matter intake on fixed gain, organ mass, hematology, cytokine concentration, pancreatic and jejunal enzyme activity, and muscle hydrogen peroxide production. Angus heifers (n= 30; body weight [BW] = 392.6 ± 12.6 kg) were individually fed and assigned to one of five treatments: 0XNEG: total mixed ration (TMR) and saline injections at days 0 and 7 of the estrous cycle, 0XPOS: TMR, rumen-protected methionine (MET) fed at 0.08% of the diet dry matter, rumen-protected choline (CHOL) fed at 60 g/d, and saline injections at days 0 and 7, 0.5X: TMR, MET, CHOL, 5-mg B12, and 80-mg folate injections at days 0 and 7, 1X: TMR, MET CHOL, 10-mg vitamin B12, and 160-mg folate at days 0 and 7, and 2X: TMR, MET, CHOL, 20-mg vitamin B12, and 320-mg folate at days 0 and 7. All heifers were estrus synchronized but not bred, and blood samples were collected on days 0, 7, and at slaughter (day 14) during which tissues were collected. By design, heifer ADG did not differ (P= 0.96). Spleen weight and uterine weight were affected cubically (P= 0.03) decreasing from 0XPOS to 0.5X. Ovarian weight decreased linearly (P< 0.01) with increasing folate and B12injection. Hemoglobin and hematocrit percentage were decreased (P< 0.01) in the 0.5X treatment compared with all other treatments. Plasma glucose, histotroph protein, and pancreatic α-amylase were decreased (P≤ 0.04) in the 0.5X treatment. Heifers on the 2X treatment had greater pancreatic α-amylase compared with 0XNEG and 0.5X treatment. Interleukin-6 in plasma tended (P= 0.08) to be greater in the 0XPOS heifers compared with all other treatments. Lastly, 0XPOS-treated heifers had reduced (P≤ 0.07) hydrogen peroxide production in muscle compared with 0XNEG heifers. These data imply that while certain doses of OCM do not improve whole animal physiology, OCM supplementation doses that disrupt one-carbon metabolism, such as that of the 0.5X treatment, can induce a negative systemic response that results in negative effects in both the dam and the conceptus during early gestation. Therefore, it is necessary to simultaneously establish an optimal OCM dose that increases circulating concentrations for use by the dam and the conceptus, while avoiding potential negative side effects of a disruptive OCM, to evaluate the long-term impacts of OCM supplementation of offspring programming.This study determined that disrupted one carbon metabolism negatively affects multiple physiological systems; however, supplementation strategies that do not negatively impact one-carbon metabolism will not negatively impact the heifer.The feeding of one-carbon metabolites (including methionine and B vitamins) has been shown to improve fetal growth and milk production in species such as mice, sheep, and dairy cattle. Extending this to beef cattle around the time of breeding is a growing area of research. Our group previously determined that one-carbon metabolite supplementation to beef heifers altered the abundance of circulating methionine-folate cycle intermediates in a dose-dependent manner. Therefore, we aimed to determine a whole-body response to one-carbon metabolite supplementation in heifers by measuring the effects on specific physiological systems as well as a total systemic response. We determined that treatments that negatively altered the methionine-folate cycle yielded a fundamental negative whole-body response to supplementation.

Fall-calving primiparous beef females [body weight (BW): 451 ± 28 (SD) kg; body condition score (BCS): 5.4 ± 0.7] were individually-fed 100% (control; CON; n= 13) or 70% (nutrient restricted; NR; n= 13) of estimated metabolizable energy and metabolizable protein requirements from day 160 of gestation to calving. Post-calving, all dams were individually-fed tall fescue hay supplemented to meet estimated nutrient requirements for maintenance, growth, and lactation in Calan gates until day 149 of lactation, which limited calves to milk only. From day 150 of lactation until weaning at day 243, dams and calves were group-fed in drylots. Dam BW and metabolic status were determined every 21 d, and BCS and backfat (BF) were determined every 42 d of lactation until weaning. Pre-weaning calf BW, size, and metabolic status were determined every 21 d. Data were analyzed with nutritional plane, calving date, and calf sex (when P < 0.25) as fixed effects. Circulating metabolites included day and nutritional plane × day as repeated measures. We previously reported that post-calving, NR dams were 64 kg and 2.0 BCS less than CON, but calf BW and size at birth were not affected. During the first 147 d of lactation, NR dams gained more (P < 0.01) BW than CON and increased (P < 0.01) BCS, while CON decreased (P ≤ 0.01) BCS and BF. Previously, NR dams had lower (P< 0.01) circulating triglycerides on day 1 of lactation, tended to have lower (P= 0.08) triglycerides on day 21, and had lower (P≤ 0.04) non-esterified fatty acids (NEFA) on days 21 and 243 than CON. Maternal glucose and urea N were not affected (P ≥ 0.73). At weaning, NR dams weighed 17 kg less (P = 0.15), were 0.67 BCS lower (P < 0.01), and tended to have less (P = 0.06) BF. Calves born to NR dams weighed less (P = 0.02) than CON by day 42 of age and were 13% smaller (P < 0.01) at weaning. Calf girth measures diverged (P≤ 0.05) by day 21 of age, and skeletal size measures were less (P≤ 0.08) for calves born to NR dams at most timepoints after day 63 of age. Calves born to NR dams tended to have lower (P = 0.09) circulating urea N pre-weaning than CON, but glucose, triglycerides, and NEFA were not affected (P ≥ 0.16). In summary, first-parity beef females that were nutrient restricted during late gestation experienced compensatory growth and gained body condition during lactation but were still thinner at weaning. Nutrient restriction reduced pre-weaning calf growth, likely due to decreased milk production.First-parity beef females that were nutrient restricted during late gestation and then fed to meet estimated nutrient requirements during lactation experienced compensatory growth but were still in poorer body condition at weaning than controls. Despite a similar birth weight, calves born to nutrient restricted dams diverged in body weight by day 42 of age and weighed less at weaning.Nutrient requirements increase substantially during late gestation in the beef female; however, poor forage nutrient availability can result in undernutrition. For heifers, the added nutrient requirements needed to continue growing during their first pregnancy and lactation pose an even greater challenge. It is plausible that lingering effects of late gestational nutrient restriction may exist for the dam and calf pre-weaning. We report that first-parity beef females that were nutrient restricted during late gestation and then fed to meet estimated nutrient requirements during lactation recovered quickly metabolically and experienced compensatory growth, but still had less body condition at weaning than controls. Late gestational nutrient restriction did not affect calf size at birth but resulted in calf body weight and size measures diverging early in life. Ultimately, nutrient restriction resulted in a 13% decrease in weaning weight, which was likely due to decreased milk production (in a companion paper). Despite this, metabolic status of calves born to nutrient restricted dams was not greatly altered. In summary, first-parity beef females that were nutrient restricted during late gestation prioritized partitioning nutrients to maternal growth and energy reserves over milk production during lactation, but dams were thinner at weaning, and pre-weaning calf growth was slowed.

Follicular progression during peripuberty is affected by diet. Vascular endothelial growth factor A (VEGFA) induces follicle progression in many species; however, there are limited studies to determine if diet may alter the effects of angiogenic VEGFA165-stimulated follicle progression or antiangiogenic VEGFA165b follicle arrest. We hypothesized that diet affects the magnitude of angiogenic and antiangiogenic VEGFA isoform actions on follicular development through diverse signal transduction pathways. To test this hypothesis, beef heifers in our first trial received Stair-Step (restricted and refeeding) or control diets from 8 to 13 months of age. Ovaries were collected to determine follicle stages, measure vascular gene expression and conduct ovarian cortical cultures. Ovarian cortical cultures were treated with phosphate-buffered saline (control), 50 ng/ml VEGFA165, VEGFA165b, or VEGFA165 + VEGFA165b. The Stair-Step heifers had more primordial follicles (P < 0.0001), greater messenger RNA abundance of vascular markers VE-cadherin (P < 0.0001) and NRP-1 (P < 0.0051) than controls at 13 months of age prior to culture. After culture, VEGFA isoforms had similar effects, independent of diet, where VEGFA165 stimulated and VEGFA165b inhibited VEGFA165-stimulated follicle progression from early primary to antral follicle stages. In vitro cultures were treated with VEGFA isoforms and signal transduction array plates were evaluated. VEGFA165 stimulated expression of genes related to cell cycle, cell proliferation, and growth while VEGFA165b inhibited expression of those genes. Thus, VEGFA isoforms can act independently of diet to alter follicle progression or arrest. Furthermore, follicle progression can be stimulated by VEGFA165 and inhibited by VEGFA165b through diverse signal transduction pathways.

The diagnosis of death using neurological criteria is an important legal method of establishing death in the UK. The safety of the diagnosis lies in the exclusion of conditions which may mask the diagnosis and the testing of the fundamental reflexes of the brainstem including the apnoea reflex. Extracorporeal membrane oxygenation for cardiac or respiratory support can impact upon these tests, both through drug sequestration in the circuit and also through the ability to undertake the apnoea test. Until recently, there has been no nationally accepted guidance regarding the conduct of the tests to undertake the diagnosis of death using neurological criteria for a patient on extracorporeal membrane oxygenation. This article considers both the background to and the process of guideline development.

Elevated arsenic in Bengal Basin aquifers threatens human health. Most deep (>150 m) groundwater in Pleistocene aquifers is low in arsenic; however higher concentrations have been reported in the southwest border region. Here, we establish that this extensive arsenic contamination at depth is not associated with well failure. A combination of geochemistry and flow modeling constrains the factors that contribute to arsenic contamination at depth in this region. Deep groundwater in the affected area is younger (2.0 ± 0.6 kyr) than deep, low‐arsenic groundwater elsewhere (12.0 ± 4.0 kyr) based on radiocarbon. Stratigraphic data indicate pre‐Holocene deposition of the contaminated aquifers, but few low‐permeability strata. Numerical modeling indicates that this stratigraphic anomaly permits a natural flow system that transports shallow groundwater to depth. Thus, in areas lacking low‐permeability layers, arsenic contamination can occur in pre‐Holocene aquifers and is probably not an early sign of future deep contamination in regions with interbedded low‐permeability strata. Exposure to arsenic in untreated groundwater pumped from millions of shallow wells across rural South Asia causes life‐threatening cardiovascular disease and cancers in adults and reduces intellectual function in children. Deep (>150 m) groundwater low in arsenic is currently the most effective mitigation option in Bangladesh. This study shows that high concentrations of arsenic observed in deep aquifers in the border area between Bangladesh and India are likely confined to that region due to the absence of clay layers vertically dividing the sandy aquifer. Flow modeling constrained by groundwater dating shows that this contamination is therefore of natural origin and unlikely to expand to wider areas in the near future. Feed costs comprise the majority of variable expenses in beef cattle systems making feed efficiency an important economic consideration within the beef industry. Due to the expense of recording individual feed-intake phenotypes, a genomic-enabled approach could be advantageous toward improving this economically relevant trait complex. A genome-wide association study (GWAS) was performed using 748 crossbred steers and heifers representing seven sire breeds with phenotypes for ADG and ADFI. Animals were genotyped with the BovineSNP50v2 BeadChip containing approximately 54,000 SNP. Both traits were analyzed using univariate SNP-based (BayesC) and haplotype-based (BayesIM) models and jointly using BayesIM to perform a bivariate GWAS. For BayesIM, a hidden Markov model (HMM) of haplotype segments of variable length was built where haplotypes were mapped to clusters based on local similarity. The estimated HMM was then used to assign haplotype cluster genotypes, instead of SNP genotypes, as latent covariates in a Bayesian mixture model. The number of haplotype clusters at each location was assumed to be either 8 (BayesIM8) or 16 (BayesIM16). A total of three univariate analyses for each trait and two bivariate analyses were performed. Posterior SD (PSD) for ADG were 0.28 (0.08), 0.37 (0.11), 0.37 (0.11), 0.35 (0.11), and 0.35 (0.12) for BayesC, BayesIM8, BayesIM16, BayesIM8 bivariate, and BayesIM16 bivariate, respectively. ADFI PSD were 0.30 (0.07), 0.44 (0.13), 0.42 (0.12), 0.38 (0.10), and 0.38 (0.10) for the same models. The top 1% of 1-Mb windows that explained the largest fraction of genetic variation in common between univariate SNP and haplotype models ranged from 24% to 40% and from 20% to 32% for ADG and ADFI, respectively. Spearmen rank correlations between molecular breeding values from SNP and haplotype-based models in the training data were similar for both traits (>0.96) suggesting that either model would lead to similar rankings of animals, although resolution of potential QTL appeared to be greater for BayesIM.

The objective of this study was to determine if feeding monensin would improve diet digestion, energy and nitrogen balance in bred heifers receiving a limit-fed corn stalk-based diet. Sixteen pregnant Meat Animal Research Center (MARC) III composite heifers were used in a 161-d completely randomized design. Heifers were randomly assigned to one of two treatments, no monensin (CON) or 150 mg/d monensin (MON), with eight heifers in each treatment group. Heifers were limit-fed a corn stalk-based diet at 100% of MEmrequirements. Effects of monensin on energy and nitrogen balance were determined via total fecal and urine collections and open-circuit respiration calorimetry. Total fecal and urine collection occurred on d 14, 42, and 161 of monensin feeding, and calorimetry measurements were made on d 0, 3, 14, 28, 42, and 161 of monensin feeding. DMI was not different (P= 0.94) for CON and MON heifers and, by design, increased (P< 0.01) from d 14 to d 161 of the trial to account for increasing fetal growth requirements. No differences (P= 0.91) in GE intake were observed between CON and MON heifers, and DE and ME intakes did not differ (P> 0.58) with monensin inclusion. DM, OM, NDF, and ADF digestion did not differ (P> 0.52) between treatments. Fecal, methane, urinary, and heat energy losses were not different (P> 0.16) for MON and CON heifers. Methane production was not different between treatments when expressed as daily liters of methane (P= 0.40); however, MON heifers produced 7% less (P= 0.03) methane per day than CON heifers when expressed as liters of methane produced on a metabolic body weight (MBW) basis. Furthermore, monensin had no effect (P= 0.36) on overall retained energy (RE). Nitrogen intake and excretion was not different (P> 0.13) between treatment groups. Results of this experiment indicate that adding monensin to limit-fed, corn stalk-based diets may not have a large effect on the energy and nitrogen balance of confined heifers.

Shortening the period of recording individual feed intake may improve selection response for feed efficiency by increasing the number of cattle that can be recorded given facilities of fixed capacity. Individual DMI and ADG records of 3,462 steers and 2,869 heifers over the entire intake recording period (range 62 to 154 d; mean 83 d; DMI83 and ADG83, respectively), DMI and ADG for the first 42 d of the recording period (DMI42 and ADG42, respectively), and postweaning ADG based on the difference between weaning and yearling weights (PADG) were analyzed. Genetic correlations among DMI42 and DMI83, ADG42 and ADG83, ADG42 and PADG, and ADG83 and PADG were 0.995, 0.962, 0.852, and 0.822, respectively. Four objective functions [feed:gain ratio in steers (FGS) and heifers (FGH); residual gain (RG); and residual feed intake (RFI)] based on DMI83 and ADG83 were considered. Indices using DMI42 and ADG42 (I42); DMI42 and PADG (IPW); and DMI42, ADG42, and PADG (IALL) were developed. Accuracy of the 5 EBV, 4 objectives, and 12 objective × index combinations were computed for all 12,033 animals in the pedigree. Accuracies of indices (IA) were summarized for animals with accuracies for objectives (OA) of 0.25, 0.5, 0.75, and 1. For the RG objective and animals with OA of 0.75, indices I42, IPW, and IALL had IA of 0.63, 0.55, and 0.67, respectively. Differences in IA increased with increased emphasis on ADG83 in the objective. Differences in IA between I42 and IPW usually increased with OA. Relative efficiency (RE) of selection on 42-d tests compared with 83 d was computed based on differences in IA and selection intensities of 5%, 25%, 50%, and 75% under the 83-d scenario, assuming 65% more animals could be tested for 42 d. For 25% selected for the RG objective, and animals with OA of 0.75, indices I42, IPW, and IALL had RE of 1.02, 0.90, and 1.10, respectively. As % selected, OA, and emphasis on DMI increased, RE increased. Relative efficiency varied considerably according to assumptions. One-half of the scenarios considered had RE > 1.15 with a maximum of 2.02 and 77% RE > 1.0. A shorter period of recording DMI can improve selection response for feed efficiency. Selection for the efficiency objectives would not affect PADG. It will be most effective if ADG over the period coinciding with intake recording and ADG over a much longer period of time are simultaneously included in a multiple-trait genetic evaluation with DMI and used in a selection index for efficiency.

The importance of the rumen microbiota on nutrient cycling to the animal is well recognized; however, our understanding of the influence of the rumen microbiome composition on feed efficiency is limited. The rumen microbiomes of two large animal cohorts (125 heifers and 122 steers) were characterized to identify specific bacterial members (operational taxonomic units [OTUs]) associated with feed efficiency traits (ADFI, ADG, and G:F) in beef cattle. The heifer and steer cohorts were fed a forage-based diet and a concentrate-based diet, respectively. A rumen sample was obtained from each animal via esophageal tubing and bacterial community composition was determined through 16S rRNA gene sequencing of the V4 region. Based on a regression approach that used individual performance measures, animals were classified into divergent feed efficiency groups. Within cohort, an extreme set of 16 animals from these divergent groups was selected as a discovery population to identify differentially abundant OTUs across the rumen bacterial communities. The remaining samples from each cohort were selected to perform forward stepwise regressions using the differentially abundant OTUs as explanatory variables to distinguish predictive OTUs for the feed efficiency traits and to quantify the OTUs collective impact on feed efficiency phenotypes. OTUs belonging to the families Prevotellaceae and Victivallaceae were present across models for heifers, whereas OTUs belonging to the families Prevotellaceae and Lachnospiraceae were present across models for steers. Within the heifer cohort, models explained 19.3%, 25.3%, and 19.8% of the variation for ADFI, ADG, and G:F, respectively. Within the steer cohort, models explained 27.7%, 32.5%, and 26.9% of the variation for ADFI, ADG, and G:F, respectively. Overall, this study suggests a substantial role of the rumen microbiome on feed efficiency responses.

Aims.There are growing calls to reduce, and where possible eliminate, the use of seclusion and restraint in mental health settings, but the attitudes and beliefs of consumers, carers and mental health professionals towards these practices are not well understood. The aim of this study was to compare the attitudes of mental health service consumers, carers and mental health professionals towards seclusion and restraint in mental health settings. In particular, it aimed to explore beliefs regarding whether elimination of seclusion and restraint was desirable and possible.Methods.In 2014, an online survey was developed and widely advertised in Australia via the National Mental Health Commission and through mental health networks. The survey adopted a mixed-methods design, including both quantitative and qualitative questions concerning participants’ demographic details, the use of seclusion and restraint in practice and their views on strategies for reducing and eliminating these practices.Results.In total 1150 survey responses were analysed. A large majority of participants believed that seclusion and restraint practices were likely to cause harm, breach human rights, compromise trust and potentially cause or trigger past trauma. Consumers were more likely than professionals to view these practices as harmful. The vast majority of participants believed that it was both desirable and feasible to eliminate mechanical restraint. Many participants, particularly professionals, believed that seclusion and some forms of restraint were likely to produce some benefits, including increasing consumer safety, increasing the safety of staff and others and setting behavioural boundaries.Conclusions.There was strong agreement across participant groups that the use of seclusion and restraint is harmful, breaches human rights and compromises the therapeutic relationship and trust between mental health service providers and those who experience these restrictive practices. However, some benefits were also identified, particularly by professionals. Participants had mixed views regarding the feasibility and desirability of eliminating these practices.

Aims.Clozapine is the most effective medication for treatment refractory schizophrenia. However, descriptions of the mental health and comorbidity profile and care experiences of people on clozapine in routine clinical settings are scarce. Using data from the 2010 Australian Survey of High Impact Psychosis, we aimed to examine the proportion of people using clozapine, and to compare clozapine users with other antipsychotic users on demographic, mental health, adverse drug reaction, polypharmacy and treatment satisfaction variables.Methods.Data describing 1049 people with a diagnosis of schizophrenia or schizoaffective disorder, who reported taking any antipsychotic medication in the previous 4 weeks, were drawn from a representative Australian survey of people with psychotic disorders in contact with mental health services in the previous 12 months. We compared participants taking clozapine (n= 257, 22.4%) with those taking other antipsychotic medications, on a range of demographic, clinical and treatment-related indicators.Results.One quarter of participants were on clozapine. Of participants with a chronic course of illness, only one third were on clozapine. After adjusting for diagnosis and illness chronicity, participants taking clozapine had significantly lower odds of current alcohol, cannabis and other drug use despite similar lifetime odds. Metabolic syndrome and diabetes were more common among people taking clozapine; chronic pain was less common. Psychotropic polypharmacy did not differ between groups.Conclusions.Consistent with international evidence of clozapine underutilisation, a large number of participants with chronic illness and high symptom burden were not taking clozapine. The lower probabilities of current substance use and chronic pain among clozapine users warrant further study.

Aims.Few studies have examined the experiences of carers of people with psychosis using a representative sample. Aiming to obtain generalisable results concerning carers in the context of increased emphasis on involving carers in Australian mental health service delivery and policy frameworks, this study recruited carers within the second Australian national survey of psychotic disorders (Survey of High Impact Psychosis, SHIP). Given that most SHIP participants had long-term illness and extended relationships with carers, the health and wellbeing of carers as a group were expected to be relatively stable. However, since it is unknown whether carers’ health and wellbeing would change, our main aim was to explore change and stability in carers’ health and wellbeing and the relationship between any changes experienced by individual carers and corresponding SHIP participants’ functioning over time.Methods.Ninety-eight caregivers of SHIP participants were recruited at baseline and completed validated instruments assessing their health and wellbeing. Seventy-eight carers were re-interviewed at 1-year follow-up. Clinical factors were extracted from the SHIP database. Wilcoxon matched-pairs signed-rank test and t-test were used to analyse changes in variables over time. Cross-lagged analyses were conducted to identify possible causative relationships in changes in SHIP participant and carer variables.Results.A substantial percentage of carers experienced social isolation (28.6%), psychological distress (37.7%) and poorer quality of life than population norms. There were no statistically significant changes between baseline and follow-up scores for almost all carers’ health and wellbeing variables, other than a poorer perception of their quality of life in relation to their physical health after 1 year. Cross-lagged analyses suggested that poorer functioning of people with psychosis influenced carers’ social isolation, grief and psychological distress.Conclusions.Findings show that carers’ perception of their health and wellbeing did not improve within current mental health service delivery frameworks over time. Carer's persistently poor health and wellbeing suggests a pressing need to enhance services that improve carers’ health and wellbeing especially their physical health and the functioning of people with psychosis whom they support.

Most of the research addressing feed efficiency and the microbiota has been conducted in cattle fed grain diets, although cattle evolved to consume forage diets. Our hypothesis was that the bacteria in the rumen and cecum differed in cattle that have a common feed intake but had different ^average daily body weight gains (ADG) on a forage diet. Heifers (n= 134) were 606 ± 1 d of age and weighed 476 ± 3 kg at the start of the 84-d feeding study. Heifers were offered ad libitum access to a totally mixed ration that consisted of 86% ground brome hay, 10% wet distillers grains with solubles, and 4% mineral supplement as dry matter. Feed intake and body weight gain were measured, and gain was calculated. Heifers with the least (n= 8) and greatest (n= 8) ADG within 0.32 SD of the mean daily dry matter intake were selected for sampling. Digesta samples from the rumen and cecum were collected, and subsequent 16S analysis was conducted to identify Amplicon Sequence Variants. There were no differences in Alpha and Beta diversity between ADG classification within sample sites (P> 0.05). Both sample sites contained calculated balances of sister clades using phylogenetic isometric log ratio transferred data that differed across ADG classification. These findings suggest that bacteria did not differ at the community level, but there was structural difference at the clade level.Bacteria of the rumen and cecum.Feed is one of the greatest costs associated with beef production. Modifying the efficiency that feed is used offers a potential mechanism to improve production efficiency. Bacteria in the gastrointestinal tract modify the nutrient content of the feed cattle eat before it is absorbed. The rumen and cecum are the two primary sites of fermentation in the digesta tract. Structure of the bacterial community at the clade level suggests that they differ with feed efficiency.

Fall-calving primiparous crossbred beef females [body weight (BW): 451 ± 28 (SD) kg; body condition score (BCS): 5.4 ± 0.7] were allocated by fetal sex and expected calving date to receive either 100% (control; CON; n= 13) or 70% (nutrient restricted; NR; n= 13) of metabolizable energy and metabolizable protein requirements for maintenance, pregnancy, and growth from day 160 of gestation to calving. Heifers were individually-fed chopped poor quality hay and supplemented to meet targeted nutritional planes based on estimated hay intakes. Dam BW, BCS, backfat, and metabolic status were determined pre-treatment, every 21 d (BW and metabolic status) or 42 d (BCS and backfat) during gestation, and post-calving. At birth, calf BW and size were measured, and total colostrum from the most full rear quarter was collected pre-suckling. Data were analyzed with nutritional plane, treatment initiation date, and calf sex (when P < 0.25) as fixed effects. Gestational metabolites included day and nutritional plane × day as repeated measures. During late gestation, CON dams gained (P < 0.01) maternal (non-gravid) BW and maintained (P ≥ 0.17) BCS and backfat, while NR dams lost (P < 0.01) maternal BW, BCS, and backfat. Circulating glucose, urea N, and triglycerides were less (P ≤ 0.05) in NR dams than CON at most late gestational timepoints after treatment initiation. Circulating non-esterified fatty acids were greater (P < 0.01) in NR dams than CON. Post-calving, NR dams weighed 63.6 kg less (P < 0.01) and were 2.0 BCS less (P < 0.01) than CON. At 1 h post-calving, NR dams had less (P = 0.01) plasma glucose and tended to have less (P = 0.08) plasma triglycerides than CON. Nutrient restriction did not affect (P ≥ 0.27) gestation length, calf birth weight, or calf size at birth. Colostrum yield was 40% less (P = 0.04) in NR dams than CON. Protein and immunoglobulin concentrations were greater (P ≤ 0.04), but free glucose and urea N concentrations were less (P ≤ 0.03), in colostrum of NR dams than CON. Colostrum total lactose, free glucose, and urea N were less (P ≤ 0.03) in NR dams than CON, but total protein, triglycerides, and immunoglobulins were not affected (P ≥ 0.55). In summary, beef heifers experiencing late gestational nutrient restriction prioritized partitioning nutrients to fetal growth and colostrum production over maternal growth. During undernutrition, fetal and colostral nutrient demands were largely compensated for by catabolism of maternal tissue stores.Beef heifers experiencing late gestational nutrient restriction prioritized partitioning nutrients to fetal growth and colostrum production over maternal body tissues. Nutrient restricted females lost body weight and body condition during late gestation, but calf size at birth and colostrum total nutrients and immunoglobulins, except lactose, did not differ from adequately-fed females.Nutrient requirements increase substantially during late gestation in the beef female. Even in well-managed herds, it is possible for females to be nutrient restricted during this time due to challenges of poor forage quality or availability and environmental stress. For heifers, the added nutrient requirements needed to continue growing pose an even greater challenge during their first pregnancy. However, little is known about how late gestational undernutrition impacts nutrient partitioning between maternal growth, the developing offspring, and colostrum production in beef heifers. Our data show that late gestational nutrient restriction in heifers slowed the expected maternal growth and instead maternal tissue stores were catabolized. Less nutrients were available in the maternal circulation, yet calf weight and size at birth were not affected. Late gestational nutrient restriction resulted in less colostrum produced by the dam and less lactose available to the offspring, but the total protein, fat, and immunoglobulins available in colostrum were not altered. In summary, beef heifers experiencing late gestational nutrient restriction prioritized partitioning nutrients to fetal growth and colostrum production over maternal growth and maintenance of body condition.

Most of the metabolizable energy that a cow uses during a production year is for maintenance; however, less is known about the heritability of maintenance compared to other traits that can be measured directly. Feed intake is a heritable trait in the mature cow and most of the feed consumed is used for maintenance. We hypothesized that maintenance energy was a heritable trait. Individual feed intake was measured for 84 or 85 d on 5 yr old pregnant cows (N= 887) from a pedigreed population of cattle that represent prominent breeds in the United States. Phenotypic mean (± SD) values were 654 ± 68 kg for cow body weight, 0.21 ± 0.24 kg/d for average daily gain, and 175 ± 17 d for midpoint fetal age. Dry matter intake averaged (± SD) 10.84 ± 1.41 kg/d. Metabolizable energy for maintenance was estimated by subtracting the metabolizable energy used for conceptus growth and tissue accretion from metabolizable energy intake. Metabolizable energy for maintenance averaged (± SD) 139 ± 18 ME kcal/d/BW kg0.75and had a heritability of 0.31 ± 0.11. Cows have a moderate heritability for maintenance suggesting an opportunity for selection.The metabolizable energy required for maintenance in cows is heritable suggesting an opportunity for genetic selection.Feed is one of the greatest costs of beef production. Most of the feed used annually by a cow is to maintain her body. A study was conducted measuring individual feed intake of mature pregnant cows. We have determined that the amount of energy that a cow uses to maintain her body is heritable suggesting that cows can be selected for differences in the energy required to maintain their bodies.

The objective of this study was to determine the dose of folate and vitamin B12in beef heifers fed rumen protected methionine and choline required to maintain increased B12levels and intermediates of the methionine-folate cycle in circulation. Angus heifers (n = 30; BW = 392.6 ± 12.6 kg) were individually fed and assigned to one of five treatments: 0XNEG: Total mixed ration (TMR) and saline injections at day 0 and 7 of the estrous cycle, 0XPOS: TMR, rumen protected methionine (MET) fed at 0.08% of the diet DM, rumen protected choline (CHOL) fed at 60 g/d, and saline injections at day 0 and 7, 0.5X: TMR, MET, CHOL, 5 mg B12, and 80 mg folate at day 0 and 7, 1X: TMR, MET CHOL, 10 mg vitamin B12, and 160 mg folate at day 0 and 7, and 2X: TMR, MET, CHOL, 20 mg B12, and 320 mg folate at day 0 and 7. All heifers were estrus synchronized but not bred, and blood was collected on day 0, 2, 5, 7, 9, 12, and 14 of a synchronized estrous cycle. Heifers were slaughtered on day 14 of the estrous cycle for liver collection. Serum B12concentrations were greater in the 0.5X, 1X, and 2X, compared with 0XNEG and 0XPOS on all days after treatment initiation (P< 0.0001). Serum folate concentrations were greater for the 2X treatment at day 5, 7, and 9 of the cycle compared with all other treatments (P≤ 0.05). There were no differences (P≥ 0.19) in hepatic methionine-cycle or choline analyte concentrations by treatment. Concentrations of hepatic folate cycle intermediates were always greater (P≤ 0.04) in the 2X treatment compared with the 0XNEG and 0XPOS heifers. Serum methionine was greater (P= 0.04) in the 0.5X and 2X heifers compared with 0XNEG, and S-adenosylhomocysteine (SAH) tended (P= 0.06) to be greater in the 0.5X heifers and the S-adenosylmethionine (SAM):SAH ratio was decreased (P= 0.05) in the 0.5X treatment compared with the 0XNEG, 0XPOS, and 2X heifers. The hepatic transcript abundance of MAT2Aand MAT2Bwere decreased (P≤ 0.02) in the 0.5X heifers compared with the 0XNEG, 0XPOS, and 2X heifers. These data support that beef heifers fed rumen protected methionine and choline require 20 mg B12and 320 mg folate once weekly to maintain increased concentrations of B12and folate in serum. Furthermore, these data demonstrate that not all supplementation levels are equal in providing positive responses, and that some levels, such as the 0.5X, may result in a stoichiometric imbalance in the one-carbon metabolism pathway that results in a decreased SAM:SAH ratio.This study determined the optimal doses for vitamin B12(20 mg injected weekly) and folate (320 mg injected weekly) injection to beef heifers during the periconceptual period when rumen protected methionine and choline were fed at 0.08% of dry matter and 60 g/d, respectively. These doses set the basis upon which future studies can be based when feeding and injecting one-carbon metabolites.The strategic inclusion of one-carbon metabolites, which include vitamins and minerals that are found in human prenatal vitamins, to beef cattle feeding and management protocols during the periconceptual period (the time around breeding) is a novel concept. Therefore, this study aimed to identify the feeding and injection doses of one-carbon metabolites in beef heifers to maintain increased circulating concentrations of one-carbon metabolites for use as a model from which other studies could base their treatments on. We determined that daily feeding of methionine and choline at 0.08% of dry matter and 60 g/d, respectively, and administration of vitamin B12and folate at 20 mg and 320 mg once per week, respectively resulted in sustained elevated concentrations of one-carbon metabolites.

Mitochondrial DNA copy number (mtDNA CN) is heritable and easily obtained from low-pass sequencing (LPS). This study investigated the genetic correlation of mtDNA CN with growth and carcass traits in a multi-breed and crossbred beef cattle population. Blood, leucocyte, and semen samples were obtained from 2,371 animals and subjected to LPS that resulted in nuclear DNA (nuDNA) and mtDNA sequence reads. Mitochondrial DNA CN was estimated as the ratio of mtDNA to nuDNA coverages. Variant calling was performed from mtDNA, and 11 single nucleotide polymorphisms (SNP) were identified in the population. Samples were classified in taurine haplogroups. Haplogroup and mtDNA type were further classified based on the 11 segregating SNP. Growth and carcass traits were available for between 7,249 and 60,989 individuals. Associations of mtDNA CN, mtDNA haplogroups, mtDNA types, and mtDNA SNP with growth and carcass traits were estimated with univariate animal models, and genetic correlations were estimated with a bivariate animal model based on pedigree. Mitochondrial DNA CN tended (P-value ≤0.08) to be associated with birth weight and weaning weight. There was no association (P-value >0.10) between mtDNA SNP, haplogroups, or types with growth and carcass traits. Genetic correlation estimates of mtDNA CN were −0.30 ± 0.16 with birth weight, −0.31 ± 0.16 with weaning weight, −0.15 ± 0.14 with post-weaning gain, −0.11 ± 0.19 with average daily dry-matter intake, −0.04 ± 0.22 with average daily gain, −0.29 ± 0.13 with mature cow weight, −0.11 ± 0.13 with slaughter weight, −0.14 ± 0.13 with carcass weight, −0.07 ± 0.14 with carcass backfat, 0.14 ± 0.14 with carcass marbling, and −0.06 ± 0.14 with ribeye area. In conclusion, mtDNA CN was negatively correlated with most traits investigated, and the genetic correlation was stronger with growth traits than with carcass traits.Mitochondrial DNA copy number is easily obtained as a by-product of low-pass sequencing, heritable, and could be a potential genetic indicator of growth in beef cattle.This study investigated mitochondrial DNA copy number (mtDNA CN) as a potential genetic indicator of growth and carcass traits in a composite beef cattle population. Mitochondrial DNA CN was estimated as the ratio of mtDNA to nuclear DNA coverages. Growth and carcass traits were available for between 7,249 and 60,989 individuals. Genetic parameters were estimated from an animal model based on pedigree. Genetic correlation estimates of mtDNA CN with growth and carcass traits were low to moderate and mostly negative. These indicate that selection for lower mtDNA would be associated with an increase in birth weight, weaning weight, post-weaning gain, average daily dry-matter intake, mature cow weight, slaughter weight, and carcass weight. Therefore, the by-product of whole-genome sequencing at a low depth could be used as an indicator trait for growth and carcass traits in genetic evaluations, but the genetic relationships are not likely strong enough to prioritize mtDNA ahead of routinely used indicator traits.

Negative attitudes toward disability cause difficulties in integrating persons with disabilities (PWDs) into society and limit their access to health care, education, employment, and leisure. Being aware of societal attitudes toward disability may help explain discrimination against PWDs and draw attention to the solutions needed to address these. Good measures of attitudes are vital for this purpose.
