The DNA helicase FANCJ is mutated in hereditary breast and ovarian cancer and Fanconi anemia (FA). Nevertheless, how loss of FANCJ translates to disease pathogenesis remains unclear. We addressed this question by analyzing proteins associated with replication forks in cells with or without FANCJ. We demonstrate that FANCJ-knockout (FANCJ-KO) cells have alterations in the replisome that are consistent with enhanced replication stress, including an aberrant accumulation of the fork remodeling factor helicase-like transcription factor (HLTF). Correspondingly, HLTF contributes to fork degradation in FANCJ-KO cells. Unexpectedly, the unrestrained DNA synthesis that characterizes HLTF-deficient cells is FANCJ dependent and correlates with S1 nuclease sensitivity and fork degradation. These results suggest that FANCJ and HLTF promote replication fork integrity, in part by counteracting each other to keep fork remodeling and elongation in check. Indicating one protein compensates for loss of the other, loss of both HLTF and FANCJ causes a more severe replication stress response.

Understanding the response of soil respiration to global warming in agro-ecosystem is crucial for simulating terrestrial carbon (C) cycle. We conducted an infrared warming experiment under conventional tillage (CT) and no-tillage (NT) farmland for winter wheat and summer maize rotation system in North China Plain (NCP). Treatments include CT with and without warming (CTW and CTN), NT with and without warming (NTW and NTN). The results indicated that warming had no significant effect on soil moisture in irrigated farmland of NCP (P>0.05). The elevated average soil temperature of 1.1–1.6°C in crop growing periods could increase annual soil CO2emission by 10.3% in CT filed (P>0.05), but significantly increase it by 12.7% in NT field (P<0.05), respectively. The disturbances such as plowing, irrigation and precipitation resulted in the obvious soil CO2emission peaks, which contributed 36.6–40.8% of annual soil cumulative CO2emission. Warming would enhance these soil CO2emission peaks; it might be associated with the warming-induced increase of autotrophic respiration and heterotrophic respiration. Compared with un-warming treatments, dissolved organic carbon (DOC) and soil microbial biomass carbon (MBC) in warming treatments were significantly increased by 11.6–23.4 and 12.9–23.6%, respectively, indicating that the positive responses of DOC and MBC to warming in both of two tillage systems. Our study highlights that climate warming may have positive effects on soil C release in NCP in association with response of labile C substrate to warming.

The hard silicon calcium stone slurry were synthesized by dynamic hydrothermal synthesis in this article, the influence of water solid ratio and carbon fiber on the performance of the calcium silicate thermal insulation material was studied. The results showed that when the water-solid ratio of 40, sodium dodecyl benzene sulfonate cited as a foaming agent, the sphericity of spherical particles was well, and xonotlite fiber developed well. The added carbon fiber could significantly increase the flexural and compressive strength and coefficient of thermal conductivity of the material. The microstructure of calcium silicate insulation material was observed and analyzed by scanning electron microscope.

In this article, the influence of the churning technology on the average diameter of xonotlite particle and the micro-structure of xonotlite particle during the hydrothermal synthesis progress of the heat insulating material of xonotlite were researched. The influence of fiber adding process on the strength and density of calcium silicate insulation material was studied. The complexity and the macro mechanical properties of the fiber dispersed in the matrix with different fiber adding way were discussed. The microstructure of xonotlite was observed and analyzed by scanning electron microscope and the related mechanisms were studied.
