Laser plasmas can be produced when high-power laser beams are focused in matter. A focused laser beam of TW(terawatt)-level high power has an extremely strong electric field, so neutral atoms are immediately ionized by the laser electric field, leading to a laser-produced plasma. The laser plasma can be produced by small table-top TW lasers based on the CPA (chirped-pulse amplification) technique, and now they are rather easily available even in university laboratories. In Korea, there are several CPA-based TW (or even petawatt) lasers in a few institutions, and they have been used for diverse laser plasma physics research and applications, including the laser acceleration for electrons and ions, high-power THz (tera-hertz) generation, advanced light sources, high-energy-density plasmas, plasma optics, etc. This paper reviews some of the laser plasma physics research and applications that have been performed in several universities and research institutes.

Injuries to the limb are the most frequent cause of permanent disability following combat wounds. We reviewed the medical records of 450 soldiers to determine the type of upper limb nerve injuries sustained, the rate of remaining motor and sensory deficits at final follow-up, and the type of Army disability ratings granted. Of 189 soldiers with an injury of the upper limb, 70 had nerve-related trauma. There were 62 men and eight women with a mean age of 25 years (18 to 49). Disabilities due to nerve injuries were associated with loss of function, neuropathic pain or both. The mean nerve-related disability was 26% (0% to 70%), accounting for over one-half of this cohort’s cumulative disability. Patients injured in an explosion had higher disability ratings than those injured by gunshot. The ulnar nerve was most commonly injured, but most disability was associated with radial nerve trauma. In terms of the final outcome, at military discharge 59 subjects (84%) experienced persistent weakness, 48 (69%) had a persistent sensory deficit and 17 (24%) experienced chronic pain from scar-related or neuropathic pain. Nerve injury was the cause of frequent and substantial disability in our cohort of wounded soldiers.Cite this article: Bone Joint J2014;96-B:254–8.

Natural polymers and their derivatives are attracting increasing interest as promising biodegradable materials that can meet the environmental and recycling demands from society. This study prepared biodegradable composites of cellulose diacetate and starch, and examined their physical and thermal properties. In addition, the morphology of the composites was examined by scanning electron microscopy. For melt processing, epoxidized soybean oil, as a lubricant, and triacetine, as a plasticizer, were added to the composites. The optimal conditions for the preparation of the biodegradable composites were determined. Increasing the amount of starch in the composites resulted in further enhancement of the processability of cellulose diacetate. The tensile strength and Young's modulus decreased, and the amount of elongation and Tgvalue increased with increasing amount of starch.

This paper reports the MOVPE growth of InN on a 3c-SiC/Si template formed by C+-ion implantation into Si(111). Properties of grown films on the template are compared with those grown on sapphire in the same run. By employing the nitridation process of the template at 900 °C for 30 min just before the GaN buffer growth, the buffer layer becomes to show uniform nucleation. This is due to the improved wettability of GaN layer on the template by the formation of Si–N bonds on the Si-polar SiC layer. The surface mophology of InN film grown on the nitrided template is similar to that for a film grown on a sapphire. The film on the nitrided template shows strong photoluminescence with a peak energy of 0.70 eV at room temperature. The carrier concentration and Hall mobility are 7.6 × 1018 cm–3 and 630 cm2/Vs, respectively. These data are comparable to those for InN grown on the sapphire (0.71 eV, 6.5 × 1018 cm–3 and 870 cm2/Vs). Polyaniline (PANI) and its various copolymers were synthesized using different monomers such as o‐methyl‐, o‐methoxy‐, o‐ethyl‐, o‐ethoxyaniline and sodium diphenylamine sulfonate by the chemical oxidation polymerization as air‐stable organic conducting polymers, and adopted as one of the most potential materials of electrorheological (ER) fluids, especially for the anhydrous system. A relatively low density, a controllable conductivity, and thermal stability are advantages of the PANI based ER system compared with other ER materials. An FT‐IR analysis was adopted to confirm their synthesis and a scanning electron microscopy (SEM) analysis indicated the shape of PANI derivatives was irregular. The effect of functional group of PANI on electric and electroresponsive properties of poly(aniline‐co‐ethoxyaniline) (COPA) particles were examined. A universal scaling equation of the yield stress was applied to these ER fluids and it was found that all data were collapsed successfully onto a single curve regardless of monomer type of the polyaniline synthesized. Polymer gel electrolytes were prepared by thermal crosslinking reaction of a series of acrylic end-capped poly(ethylene glycol) and poly(propylene glycol) [P(EG-co-PG)] having various geometries and molecular weights. Acrylic end-capped prepolymers were prepared by the esterification of low molecular weight (Mn: 1900–5000) P(EG-co-PG) with acrylic acid. The linear increase in the ionic conductivity of polymer gel electrolyte films was observed with increasing temperature. The increase in the conductivity was also monitored by increasing the molecular weight of precursor polymer. Nanocomposite electrolytes were prepared by the addition of 5 wt % of organically modified layered silicate (montmorillonite) into the gel polymer electrolytes. A composite material of a silica-based mesoporous molecular sieve, MCM-41, with conducting polyaniline (PANI) inside the uniformly aligned one-dimensional channels (PANI/MCM-41) was prepared and its nanocomposite formation was confirmed through an electrical conductivity measurement. This nanocomposite particle was adopted for a dispersed phase in electrorheological (ER) fluids, and the ER property was measured using a Couette-type rotational rheometer equipped with a high voltage generator. Suspension of PANI/MCM-41 showed ER properties more enhanced than those of MCM-41 or PANI alone as a result of the anisotropic polarization of the PANI/MCM-41 nanocomposite.

Copolymer particles consisting of acrylamide (AM) and acryloyloxyethyl dimethylbenzyl ammonium chloride (AODBAC) were prepared by dispersion polymerization in an aqueous solution of ammonium sulfate and in the presence of poly(acryloyloxyethyl trimethyl ammonium chloride) (PAOTAC) as the stabilizer. The average particle size ranged from 4 to 7 μm, and Mw and Mn were 2–6 ×10⁶ g/mol and 1–3 × 10⁶ g/mol, respectively. The effects of the AM/AODBAC ratio, monomer, initiator, salt, and stabilizer concentration on the particle size and molecular weight were studied. Increase of the AODBAC/AM ratio resulted in a decrease in the molecular weight and an increase in particle size. With the increase of the AODABA/AM ratio, the dispersion became less stable during long storage. Due to the presence of the salt, ionization of the quaternary ammonium groups in the dispersed particles is considered significantly suppressed. © 2002 Wiley Periodicals, Inc. Previously we reported the development of a novel expression system with Tat/TAR‐oriP vectors and HKB11 cell line, which supports high level protein expression (Cho et al. Cytotechnology2001, 37, 23–30). In the present study, we further demonstrated that HKB11 cells are suitable for high throughput expression (microgram scale) of genomic candidates in transient transfection system for in vitro evaluation of biological functions. HKB11 cells were also shown to support the production of milligram to gram quantities of protein drug candidates for in vivo evaluation of efficacy in various disease models. Stable HKB11 clones secreting high levels of a tissue factor (TF; 40–50 pg/c/d) and B‐domain deleted recombinant factor VIII (BDDrFVIII; 5–10 μU/c/d) were derived under serum‐free conditions. The specific productivity for these two proteins from the HKB11 cells was 10‐fold greater than those from CHO cells derived under the similar conditions. In conclusion, we have demonstrated that the HKB11 cell line is well‐suited for transient and long‐term production of recombinant proteins.

The spherical, monodisperse polymer microshperes (PAPMMAs) consisting of a poly(methyl methacrylate) (PMMA) core and a polyaniline (PANI) shell were prepared and adopted as dispersed materials for electrorheological (ER) fluids, in which electrical conductivity was originated primarily from the surface-coated conducting PANI layer. These surfaces were rough and irregular. Three different core sizes of PAPMMAs (2, 4.5, and 9 μm) with the same weight ratio of PANI, the larger core having the thicker PANI shell, were prepared. PAPMMAs that have similar shell thicknesses but different core sizes were also prepared in order to examine the particle size effect on ER performance. Performances of PAPMMA-based ER fluids were enhanced with increasing core particle size. The same result was obtained for the PAPMMA systems having the same shell thickness. Dielectric spectra of these ER fluids were found to provide additional information on both analyzing their electrical polarization properties and interpreting the flow behavior of the PAPMMA-based ER fluids.

Dispersion polymerization of acrylamide (PAM) has been successfully carried out in aqueous ammonium sulfate media by using poly(acryloyloxyethyl trimethylammonium chloride) (PAOTAC) as the polymeric stabilizer and 2,2'-azobis(2-methyl propionamidine) dihydrochloride (AIBA) as the initiator. The polymerization behaviors with varying concentrations of acrylamide, PAOTAC, AIBA, and ammonium sulfate were investigated. The reaction conditions for stable dispersion were concentrations of 5–10% for acrylamide, 0.6–1.8% for the stabilizer, 0.92–1.84 × 10⁻⁴ mol/L for the initiator, and 24–30% for the salt. The resulting conversion–time curves were S-shaped, as is typically observed in polymerization. Polydisperse spherical particles were formed in the system. An image analyzer photographed the size of the dispersed particles and their distribution was measured. To determine whether pathogenic Acanthamoeba culbertsoni trophozoites and lysate can induce cytopathic changes in primary-culture microglial cells, morphological changes were observed by transmission electron microscopy (TEM). In addition, the secretion of two kinds of cytokines, tumor necrosis factor alpha (TNF-alpha) and interleukin-1beta (IL-1beta), from microglial cells was observed. Trophozoites of pathogenic A. culbertsoni made contact with microglial cells and produced digipodia. TEM revealed that microglial cells cocultured with amoebic trophozoites underwent a necrotic process, accompanied by lysis of the cell membrane. TEM of microglial cells cocultured with amoebic lysate showed that the membranes of the small cytoplasmic vacuoles as well as the cell membrane were lysed. The amounts of TNF-alpha secreted from microglial cells cocultured with A. culbertsoni trophozoites or lysate increased at 6 h of incubation. The amounts of IL-1beta secreted from microglial cells cocultured with A. culbertsoni trophozoites at 6 h of incubation was similar to those secreted from the control group, but the amounts decreased during cultivation with A. culbertsoni lysate. These results suggest that pathogenic A. culbertsoni induces the cytopathic effects in primary-culture rat microglial cells, with the effects characterized by necrosis of microglial cells and changes in levels of secretion of TNF-alpha and IL-1beta from microglial cells.

To determine whether trophozoites and lysates of pathogenic Acanthamoeba spp. induce apoptosis in primary-culture microglial cells, transmission electron microscopic (TEM) examinations, assessment of DNA fragmentation by agarose gel electrophoresis, and the TdT-mediated dUTP nick-end labeling assay were performed. When a trophozoite of pathogenic Acanthamoeba culbertsoni came in contact with a microglial cell, the digipodium was observed by TEM. Nuclear chromatin condensation was observed in 10% of microglial cells, while it was not revealed when they were cocultured with weakly pathogenic Acanthamoeba royreba trophozoites. DNA fragmentation in microglial cells cocultured with the A. culbertsoni lysate was detected by electrophoresis, showing DNA ladder formation, whereas it was hardly observed in microglial cells cocultured with A. royreba. DNA fragmentation of microglial cells was also confirmed by flow cytometry analysis. The fluorescence of TdT-stained apoptotic bodies became intensely visible with microglial cells cocultured with the A. culbertsoni lysate. In contrast, with microglial cells cocultured with the A. royreba lysate, only a background level of fluorescence of TdT-stained apoptotic bodies was detected. These results suggest that some rat microglial cells cocultured with pathogenic A. culbertsoni undergo cytopathic changes which show the characteristics of the apoptotic process, such as nuclear condensation and DNA fragmentation.

Abstract: Electrorheological (ER) characteristics of ER fluids, composed of suspensions of semiconducting polyaniline and two different copolyaniline particles in silicone oil, were determined experimentally. Ionic sodium diphenylamine sulfonate and nonionic o-ethoxyaniline were used to synthesize copolyanilines, i.e. N-substituted copolyaniline and poly(aniline-co-o-ethoxyaniline), respectively. ER fluids composed of these three different polymers were compared with respect to their rheological properties under an applied electric field. It was found that ER fluids containing a copolymer with an ionic group exhibited the highest ER performance among polyaniline and its copolymer systems in a shear-rate region above 1 s−1.

Abstract: As one of the intrinsically polarizable materials used in electrorheological (ER) fluid, polyaniline was synthesized by the chemical oxidation of aniline with ammonium peroxysulfate. ER fluids were prepared by dispersing polyaniline particles in silicone oil, and their rheological properties were measured. The effect of the polymerization temperature of polyaniline on its ER characteristics was investigated. ER fluids with polyaniline particles synthesized at −10C (PA-10) showed the best ER performance (yield stress) compared with those synthesized at higher temperatures. A difference in the flow behavior of the ER fluids was also investigated through the dielectric spectra of ER fluids.

Ferritin-labeled antibodies and lectins have been used to visualize endocytic vesicles in erythrocytes. Distinguishing preformed endocytic vesicles from invaginated plasma membrane has proved difficult especially since antibodies and lectins by themselves induce these changes in membrane configuration. We have used a transmission electron microscope technique (TEM) utilizing positive colloidal iron staining, employing as our model erythrocytes treated with cationic amphipathic drugs. Our technique allows fixation of cells at any point during drug treatment before staining. The membrane lining of unsealed but not of sealed vesicles was coated with electron dense ferric oxide grains. The formation of sealed vesicles is evidence for the fluidity of membranes of the erythrocytes of adults. The data also suggest that the mechanism may not be identical with every cationic amphiphile.

A 94-kilodalton phosphoprotein known as IE94 is the only viral polypeptide synthesized in abundance under immediate-early conditions after infection by cytomegalovirus (CMV) strain Colburn in either permissive primate or nonpermissive rodent cells. The IE94 gene, which maps at coordinates 0.71 to 0.73 in the viral genome, contains a large intron in the 5' leader sequence, and its promoter regulatory region contains novel, multiple-palindromic, repeated elements. Two recombinant plasmids (pTJ148 and pTJ198) containing the 10.5-kilobase-pair HindIII-H DNA fragment from CMV (Colburn) were transfected into mouse Ltk- cells, by either linked or unlinked coselection in hypoxanthine-aminopterin-thymidine medium, together with herpes simplex virus thymidine kinase genes. With both procedures, constitutive synthesis of the IE94 immediate-early protein was detected in pools of Ltk+ cells by immunoprecipitation. Subsequently, we isolated a clonal Ltk+ cell line which expressed the [35S]methionine-labeled IE94 polypeptide in sufficient abundance to be visualized directly in autoradiographs after gel electrophoresis of total-cell-culture protein extracts. The IE94 polypeptide synthesized in the transfected cells was indistinguishable in size and overall net charge from that produced in virus-infected cells. In addition, the IE94 protein expressed in LH2p198-3 cells was phosphorylated (presumably by a cellular protein kinase) and generated similar phosphopeptide patterns after partial tryptic digestion to those obtained with the CMV IE94 protein from infected cells. The cell line contained two to four stably integrated copies of the IE94 gene and synthesized a single virus-specific mRNA of 2.5 kilobases detectable on Northern blots. A new antigen, detectable by indirect anticomplement immunofluorescence with monoclonal antibody against the human CMV IE68 protein, was present in the nuclei of more than 95% of the LH2p198-3 cells. This evidence suggests that (unlike most herpesvirus genes) the CMV IE94 gene, together with its complex promoter and spliced mRNA structure, may contain all of the regulatory elements necessary for strong constitutive expression in mammalian cells in the absence of other viral factors.

The carboxyl-terminal one-third of the Epstein-Barr virus nuclear antigen (EBNA-1) encoded by the BamHI restriction fragment K was synthesized in Escherichia coli by use of a high-expression plasmid. The resultant 28-kDa EBNA fusion polypeptide, comprising 5-10% of the total soluble bacterial protein, was purified to apparent homogeneity by phosphocellulose and hydroxylapatite column chromatography. Both rabbit monospecific antibodies and mouse monoclonal antibodies against 28-kDa EBNA gave nuclear immunofluorescence staining on Epstein-Barr virus (EBV)-infected lymphoblastoid cell lines and recognized the appropriate intact EBNA polypeptide bands on immunoblots. An ELISA with the purified 28-kDa EBNA as antigen was used to quantitate anti-EBNA antibody in human serum samples. The ELISA method was approximately 100-fold more sensitive than the classical anticomplement immunofluorescence assay. Anti-EBNA antibody was detected in sera from 100% of normal individuals who were seropositive for the viral capsid antigen, and low anti-EBNA titers were detected in serum from most patients with acute infectious mononucleosis. The assay gave the expected pattern of titers in sera from patients with rheumatoid arthritis, Burkitt lymphoma, or nasopharyngeal carcinoma, thus confirming the validity of this purified reagent for assessing EBNA antibody status. Approximately 10% of normal individuals and rheumatoid arthritis patients had anti-EBNA titers as high as those seen in nasopharyngeal carcinoma patients. In these high-titer individuals, greater than 1% of the total IgG are antibodies that recognize 28-kDa EBNA, which indicates that the carboxyl-terminal domain of EBNA is highly immunogenic.

Copolyaniline containing phenyl sulfonic acid sodium salt pendent, was chemically synthesized and used for its characterization as an electrorheological (ER) fluid. Its chemical structure and particle shape were analyzed using an FT-IR, 13C-NMR, and SEM. Copolyaniline particles have irregular and lamellar morphology. This ER fluid was prepared by dispersing polymer particles in silicone oil and was characterized by a rotational Haake rheometer with a high voltage generator. Typical ER behavior was found in the copolyaniline suspension.

Calcium-gated secretion of proteins involves the transfer of "reserve" granules, exocytotic vesicles that are cytoplasmic and, hence, plasma membrane-naive, from the cell interior to the surface membrane where they dock prior to fusion. Docking and subsequent priming steps are thought to require cytoplasmic factors. These steps are believed to induce fusion competence. We have tested this hypothesis by isolating reserve granules from sea urchin eggs and determining under which conditions these granules will fuse. We find that isolated reserve granules, lacking soluble cofactors, support calcium-dependent membrane fusion in vitro. Preincubation with adenosine 5'-3-O-(thio)triphosphate and guanosine 5'-3-O-(thio)triphosphate did not prevent fusion. Thus, isolated reserve granules have all the necessary components required for calcium-gated fusion prior to docking.

Epstein-Barr virus (EBV), isolated from P3HR-1 cells, induces early antigen and viral capsid antigen upon infection of human B-lymphoblasts. The strong early antigen- and viral capsid antigen-inducing activity is only observed in P3HR-1 virus preparations harboring particles with defective genomes, suggesting that this biological activity is directly associated with the defective DNA population. After infection of EBV genome-carrying Raji or EBV genome-negative BJAB cells, defective genomes of P3HR-1 EBV DNA are replicated in excess, depending on the multiplicity of infecting EBV particles. Hybridization of the DNA from such infected cells with 32P-labeled EBV DNA after HindIII cleavage reveals six hypermolar fragments. Mapping of these fragments shows that they form one defective genome unit containing four nonadjacent regions (alpha, beta, gamma, and delta) of the nondefective P3HR-1 EBV DNA. Two of the segments (alpha and beta) contain ca. 17 and 13 megadaltons, respectively, from the terminal regions of the P3HR-1 genome, whereas the two smaller segments (gamma and delta) contain ca. 3.7 and 3.0 megadaltons, respectively, originating from the central portion of the genome. In the defective molecule, the regions gamma and delta are present in the opposite orientation compared with nondefective P3HR-1 EBV DNA. Tandem concatemers are formed by fusion of the alpha and beta regions. Our model suggests that tandem concatemers of three defective genome units can be packaged into virions in P3HR-1 cells.

We have investigated the effects of membrane lipid composition on biological membrane fusion triggered by low pH and mediated by the baculovirus envelope glycoprotein gp64. Lysolipids, either added exogenously or produced in situ by phospholipase A2 treatment of cell membranes, reversibly inhibited syncytium formation. Lysolipids also decreased the baculovirus infection rate. In contrast, oleic and arachidonic acids and monoolein promoted cell-cell fusion. Membrane lipid composition affected pH-independent processes which followed the low-pH-induced change in fusion protein conformation. Inhibition and promotion of membrane fusion by a number of lipids could not be explained by mere binding or incorporation into membranes, but rather was correlated with the effective molecular shape of exogenous lipids. Our data are consistent with the hypothesis that membrane fusion proceeds through highly bent membrane intermediates (stalks) having a net negative curvature. Consequently, inverted cone-shaped lysolipids inhibit and cone-shaped cis-unsaturated fatty acids promote stalk formation and, ultimately, membrane fusion.

We used antiserum raised against the bacterially synthesized product of one of the open reading frames in Epstein-Barr virus (EBV) BamHI fragment M to demonstrate that this reading frame (BMRF1) codes for a nuclear protein of the diffuse early antigen (EA) class. In indirect immunofluorescence assays, the rabbit anti-BMRF1 antiserum gave nuclear staining in approximately 5% of Raji cells which had been treated with sodium butyrate, and positive fluorescence was observed in both acetone- and methanol-fixed cells. Uninduced Raji cultures contained less than 0.1% positive cells regardless of whether indirect immunofluorescence or anti-complement immunofluorescence was used. In immunoblot analyses, the rabbit serum identified a family of polypeptides of 46 to 55 kilodaltons (kDa) in total protein extracts from B95-8 cells or from butyrate-induced Raji cells. In both cell types, the dominant polypeptides were the 48- and 50-kDa species. This same family of polypeptides was identified when the immunoblots were reacted with the R3 monoclonal antibody, and we concluded that this antibody also recognized the product of the BMRF1 open reading frame. Fibroblast cell lines containing EBV BamHI fragment M were established by cotransfection of baby hamster kidney cells with BamHI-M and the gene for neomycin resistance. Aminoglycoside G418-resistant colonies which showed evidence for EBV antigen expression in immunofluorescence assays were selected, and clonal cell lines were established. After 3 to 4 months of passaging, constitutive synthesis of EA was no longer detectable in these cell lines either by immunofluorescence or by immunoblot analysis. However, in the one cell line examined, synthesis of the 48- to 50-kDa EA was induced by treatment of the culture with sodium butyrate. Thus, the regulation of expression of this EA in transfected fibroblasts is analogous to that seen in Raji lymphoblasts. We showed previously that BamHI fragment M also contains the coding sequences for a 60-kDa nuclear EA, and hence BamHI-M encodes two separate components of the diffuse EA complex.

Expression of a component of the Epstein-Barr virus early antigen (EA) complex has been studied in fibroblast cells transfected with both wild-type and P3HR-1 defective DNA fragments covering the BamHI-M-S region of the Epstein-Barr virus genome. Baby hamster kidney (BHK) cells transfected with the BglII-J fragment and stained with human serum that was positive for the diffuse component of EA [EA(D)] in an indirect immunofluorescence assay exhibited positive nuclear staining in 5% of the cell population. Cleavage of BglII-J before transfection with the restriction enzyme BglII, StuI, HindIII, or PvuII did not affect EA expression, whereas prior cleavage with BamHI or EcoRI reduced or eliminated synthesis of EA. These observations were confirmed by using individual cloned subfragments. A Bal 31 deletion clone (pTS1) in which the HindIII and StuI sites were eliminated retained activity, whereas a clone (pTS5) in which the deletion extended closer to the EcoRI site had greatly reduced activity. Transfection of the individual BamHI-M or BamHI-S fragments, which span BglII-J, also resulted in little or no EA expression. The 2.1-kilobase biologically active region defined by these experiments corresponds precisely to the BMLF1 open reading frame. Immunoblot analyses of BHK cells transfected with either P3HR-1 defective DNA clones or the BglII-J wild-type fragment identified the product of this EA(D) coding region as a family of polypeptides consisting of a major 60-kilodalton product and minor 45- and 50-kilodalton species. In latently Epstein-Barr virus-infected lymphocytes these early antigens are not expressed, but can be induced by treatment of the cultures with sodium butyrate or phorbol esters. Using the BglII-J and pTS6 clones that were positive in transient assays, we also established Neor coselected BHK and Vero cell lines which showed similar regulated expression of the 60-kilodalton EA(D) protein. In these cell lines constitutive expression of EA(D) was limited (0.1% positive by indirect immunofluorescence and undetectable by immunoblot analysis). However, expression of EA(D) could be induced by treatment with sodium butyrate. In the induced cultures, up to 30% of the cells were EA(D) positive by immunofluorescence, and there was a concomitant appearance of the 60-kilodalton EA(D) polypeptide.

Eight independent recombinant Epstein-Barr virus genomes, each of which was a transforming strain, were made by superinfecting cell lines containing Epstein-Barr virus DNA (Raji or B95-8 strain) with a nontransforming virus (P3HR1 strain). A knowledge of the constitution of each transforming recombinant allowed the localization of the defect in the genome of the nontransforming parent to a 12-megadalton sequence within the EcoRI A fragment. Within this region, the nontransforming virus has a deletion of the BamHI Y fragment and about half of the sequences in the adjacent BamHI H fragment. The present data suggest that this deletion is responsible for the nontransforming phenotype. Furthermore, mapping a deletion in one of the recombinant genomes allowed the conclusion that a sequence (comprising about 20% of the Epstein-Barr virus genome) from the center of BamHI-D to BamHI-I' is not necessary for the maintenance of transformation by Epstein-Barr virus.
