The applications of data on biological variation include assessment of the utility of population-based reference intervals, evaluation of the significance of change in serial results, and setting of analytical quality specifications. We investigated the biological variation of 19 biochemistry analytes and total T4, measured in serum from 7 clinically healthy domestic cats sampled once weekly for 5 weeks. Samples were frozen and analyzed in random order in the same analytical run. Results were analyzed for outliers, and the components of variance, subsequently generated by restricted maximum likelihood, were used to determine within-subject and between-subject variation (CVI and CVG, respectively), as well as analytical variation (CVA) for each analyte. Indices of individuality, reference change values, and analytical performance goals were calculated. The smallest CVI and CVG were found for calcium, chloride, and sodium, whereas the largest values were calculated for bile acids. Nine analytes (albumin, alkaline phosphatase, alanine aminotransferase, aspartate aminotransferase, cholesterol, creatinine, phosphate [phosphorus], total protein, total T4) demonstrated high individuality, indicating limited utility of population-based reference intervals. Individuality was low, and population-based reference intervals were thereby considered appropriate for 5 analytes (bile acids, calcium, fructosamine, glucose, potassium). The intermediate individuality observed for 4 analytes (creatine kinase, iron, magnesium, urea) indicated that population-based reference intervals should be used with caution. The efficiency of spermatogonial stem cell (SSC) isolation and culture from pubertal donors is currently poor primarily, because of contamination with other testicular cells. This study aimed to purify SSC-like cells using different extracellular matrixes and a discontinuous gradient density. In experiment 1, testes (n=6) were analyzed for histology and SSC-related protein expressions (laminin, SSEA-4, DDX-4 and GFRα-1). After enzymatic digestion, the cell suspension was plated onto either a laminin- or gelatin-coated dish. The number of SSC-like cells was determined at 15, 30 and 60 min of culture (experiment 2). Experiment 3 was performed to test whether or not the additional step of Percoll gradient density centrifugation could really improve purification of SSC-like cells. Testicular histology revealed complete spermatogenesis with laminin expression essentially at the basal lamina of the seminiferous tubules. SSEA-4 and GFRα-1 colocalized with DDX-4 in the spermatogonia. The relative percentage of SSC-like cells, as determined by cells expressing SSEA-4 (59.42 ± 2.18%) and GFRα-1 (42.70 ± 1.28%), revealed that the highest SSC-like cell purity was obtained with the 15-min laminin-coated dish compared with other incubation times and gelatin treatment (P<0.05). Percoll treatment prior to laminin selection (15 min) significantly improved SSC-like cell recovery (91.33 ± 0.14%, P<0.001) and purity (83.82 ± 2.05% for SSEA-4 and 64.39 ± 1.51% for GFRα-1, P<0.05). These attached cells demonstrated a typical SSC-like cell morphology and also expressed POU5F1, RET and ZBTB16 mRNA. Polyclonal immunoglobulin (Ig) G autoantibodies against insulin have been identified in sera of healthy cats. We purified and fractionated insulin-binding IgGs from cat sera by affinity chromatography and analyzed affinity of insulin-binding IgGs for insulin and their epitopes. Following the passing of fraction A, which did not bind to insulin, insulin-binding IgGs were eluted into two fractions, B and C, by affinity chromatography using a column fixed with bovine insulin. Dissociation constant (KD) values between insulin-binding IgGs and insulin, determined by surface plasmon resonance analysis (Biacore™system), were 1.64e-4 M for fraction B (low affinity IgGs) and 2e-5 M for fraction C (high affinity IgGs). Epitope analysis was conducted using 16 peptide fragments synthesized in concord with the amino acid sequence of feline insulin by an enzyme-linked immunosorbent assay. Fractions B and C showed higher absorbance (affinity) of the peptide fragment of 10 amino acid residues at the carboxyl-terminal of the B chain (peptide No. 19), followed by peptide fragments of 6 to 15 amino acid residues of the B chain (peptide No. 8). Fraction C showed a higher absorbance to 7 to 16 amino acid residues of the B chain (peptide No. 5) compared with the absorbance of fraction B. Polyclonal insulin-binding IgGs may form a macromolecule complex with insulin through the multiple affinity sites of IgG molecules. This study aimed to map the lymphatic drainage of the cranial thoracic mammary gland (T1) in clinically healthy domestic cats. The most important aspect was to identify the sentinel lymph node that drains the T1 mammary gland, leading to a new model of mammary tumor removal in the cat. The study was conducted on 10 female's domestic cats. The applied methods of investigation were: radiographic indirect lymphography with contrast agent (Optiray 350), injecting a coloring agent (Evans blue dye) in vivo followed by subject euthanasia and stratigraphic and regional dissection. For each cat, have been supervised behavior and physiological parameters (i.e. pulse, breathing, temperature). The cats were kept in individual cages, and all experiments were performed one day later after the accommodation of each animal. Drainage of the cranial thoracic mammary gland was found to be exclusively cranial in all subjects. The first lymph node station has been established that can be either the accessory axillary lymph node or the proper axillary lymph node when the former is missing. Taking into account internodal connections, the study has also underlined other lymph nodes involved in the drainage of the T1 mammary region. The stratigraphic dissection method complements radiographic indirect lymphography with contrast agent, by uncovering features that the latter is unable to underline. Stratigraphic dissection can only be used as an experimental tool, but the radiographic indirect lymphography is a very good method for clinical use. [ABSTRACT FROM AUTHOR]

The article discusses a study which evaluated the possible protection of resveratrol against lung injury induced by arsenic trioxide. The study included Chinese Dragon Li cats that were given resveratrol, and resveratrol plus arsenic trioxide. It found that resveratrol pretreatment can reverse the morphological and biochemical changes induced by arsenic trioxide in the lung.

With the use of perfusion tracers, in vivo examination of the regional cerebral blood flow in cats can be performed with single photon emission computed tomography (SPECT). Reliable perfusion data of normal, healthy cats are necessary for future clinical studies or other research use. Therefore, this dataset of the regional perfusion pattern of the normal feline brain was created. Twelve cats were used in this study. Technetium-99m-ethyl cysteinate dimer (99mTc-ECD) was injected intravenously and the acquisition, using a triple head gamma camera equipped with three multi-pinhole collimators (pinhole SPECT), was started 40 mins after tracer administration under general anaesthesia. Nineteen regions of interest were defined using 7T magnetic resonance images of the feline brain and a topographical atlas. Regional counts were normalised to the counts of two reference regions: the total brain and the cerebellum. The highest tracer uptake was noticed in the subcortical structures, and the lowest in the frontal cortex and the cerebellum. Also left–right asymmetry in the temporal cortex and a rostrocaudal gradient of 5% were observed. [ABSTRACT FROM AUTHOR]

An interplay between growth, glucose regulation and hypertrophic cardiomyopathy (HCM) may exist, but has not been studied in detail. The purpose of this study was to characterize morphometric features, insulin-like growth factor-1 (IGF-1) and glucose metabolism in Maine Coon cats with HCM. Body weight, body condition score (BCS), head length and width, and abdominal circumference were measured in Maine Coon cats >2 years of age. Echocardiography and thoracic radiography (for measurement of humerus length, and fourth and twelfth vertebrae length) were also performed. Blood was collected for biochemistry profile, DNA testing, insulin and IGF-1. Sixteen of 63 cats had HCM [myosin binding protein C (MYBPC)+, n = 3 and MYBPC−, n = 13] and 47/63 were echocardiographically normal (MYBPC+, n = 17 and MYBPC−, n = 30). There were no significant differences in any measured parameter between MYBPC+ and MYBPC− cats. Cats with HCM were significantly older (P <0.001), heavier (P = 0.006), more obese (P = 0.008), and had longer humeri (P = 0.02) compared with the HCM− group. Cats with HCM also had higher serum glucose (P = 0.01), homeostasis model assessment (HOMA) and IGF-1 (P = 0.01) concentrations, were from smaller litters (P = 0.04), and were larger at 6 months (P = 0.02) and at 1 year of age (P = 0.03). Multivariate analysis revealed that age (P <0.001), BCS (P = 0.03) and HOMA (P = 0.047) remained significantly associated with HCM. These results support the hypothesis that early growth and nutrition, larger body size and obesity may be environmental modifiers of genetic predisposition to HCM. Tracheal diameter can be assessed from a thoracic radiograph, with assessment of tracheal diameter in dogs based on ratios between tracheal diameter and a skeletal measurement. Reference intervals are not, however, available for the cat. Tracheal narrowing may cause significant clinical problems, although tracheal hypoplasia in dogs may be clinically silent, and is rarely reported in cats (both mesati- and brachycephalic). The tracheal diameter and trachea:thoracic inlet and trachea:rib ratios were calculated for populations of domestic shorthair (DSH) (n = 68) and Persian (n = 40) cats. This gave reference intervals for radiographic tracheal measurements in these breeds. It is proposed that the tracheal diameter in a normal DSH cat should be 18% of the diameter of the thoracic inlet, and compared to 20% in Persian cats. [ABSTRACT FROM AUTHOR]

Abstract: Objectives: To evaluate whether the addition of a protease inhibitor (PI) to feline plasma improves the temporal stability of N-terminal pro-B-type natriuretic peptide (NT-proBNP). Animals, materials and methods: Forty-four EDTA blood samples were collected from 42 cats with cardiac disease or hyperthyroidism. Samples were separated within 15 min of collection and the plasma stored at −80 °C. Samples were thawed and each separated into 2 aliquots, one of which was mixed with PI. Each was subdivided into 5 aliquots, which remained at room temperature (RT) for zero, 24, 48, 96 or 120 h before being returned to storage at −80 °C. NT-proBNP was measured using a commercially-available ELISA. Results: There was no difference in NT-proBNP measurements between plain and PI samples at time zero (P = 0.836) or 24 h (P = 0.293). At subsequent time-points NT-proBNP was higher in the PI than the plain samples (P < 0.05). An overall effect of time at RT was detected for plain (P < 0.001) and PI samples (P < 0.001). Conclusions: The addition of PI to feline plasma significantly reduces, but does not eliminate, degradation of NT-proBNP at RT. This degradation occurs over a time course comparable to postal transportation of samples for laboratory analysis and may alter the clinical interpretation of results. [Copyright &y& Elsevier]

The article provides information on a study which investigated the influences of storage time of the feline epididymis on epididymal semen qualities before and after cryopreservation to identify the optimal duration for low-temperature storage of the epididymis applicable to artificial insemination. The testes and epididymis of male mixed cats were placed in sterile physiological saline immediately after excision and stored at 4 degrees Celsius for various durations and then subjected to epididymal sperm collection. The results suggest that low-temperature storage of the epididymis until 24 hours may be useful for frozen sperm collected from the feline cauda epididymis.

This study investigated the recombinant human thyrotropin (rhTSH) stimulation test in healthy cats (group 1), cats with non-thyroidal illness (group 2) and cats with low serum total T4 (TT4) and azotaemia after 131I treatment (group 3). Serum TT4 responses and thyroidal pertechnetate uptake after administration of 25μg rhTSH IV were assessed. Baseline serum TT4 was significantly lower in group 3 compared with group 1, but not between other group pairs. Serum TT4 increased significantly in groups 1 and 2 but not in group 3 after rhTSH administration. Post-rhTSH serum TT4 concentrations differed significantly between groups 1 and 3 and groups 2 and 3, but not between groups 1 and 2. Thyroid/salivary gland uptake ratio (T/S uptake ratio) differed only significantly between groups 1 and 3. Stimulation with rhTSH is valuable to differentiate euthyroidism from iatrogenic hypothyroidism in cats. [Copyright &y& Elsevier]

The aims of this study were to evaluate whether a recently published study used to objectively monitor pleural fluid volumes in dogs could be successfully employed in cats and secondly to assess its accuracy. Eleven feline cadavers were selected. Using the trans-sternal view employed in dogs, linear measurements from the pleural surface of the midline of the sternebra at the centre of the heart to the furthest ventro-lateral point of both right and left lung edges were recorded. Isotonic saline was injected using ultrasound guidance into both right and left pleural spaces and the measurements were repeated using standard increments until 400ml total volume was reached. The mean measurement increased in a linear relationship with the cube root of fluid volume for all cats individually. An equation was produced to predict the volume of fluid from the mean linear measurement for all cats combined: but variability in the slope of the curve for individual cats limited the accuracy of the combined equation. Equations were derived to predict the constant and slope of the curve for individual cats using the thoracic measurements made, but the residual diagnostic graphs demonstrated considerable variability. As in dogs, good correlation was found between the ultrasonographic measurement and fluid volume within individual cats. An accurate equation to predict absolute pleural fluid volume was not identified. Further analysis with reference to thoracic measurements did not increase accuracy. In conclusion, this study does provide a method of estimating absolute pleural fluid volume in cats, which may be clinical useful for pleural fluid volume monitoring but this is yet to be validated in live cats. Practical relevance: Neurological diagnosis in veterinary practice can be very cAlthough gait disturbance is one of the most common neurological presentations in feline medicine, the clinical approach to the paralyzed cat can be challenging. After excluding orthopedic and cardiovascular diseases that may mimic a neurological condition, the clinician has to address a long list of different diseases that may affect the spinal cord and produce paresis. Clinical challenges: In many cases a definitive cause of spinal weakness in cats is difficult to prove. Even when treatable diseases are identified, the prognosis is very much dependent on the severity of the clinical signs and their chronicity. This review sets out to describe the specific approach, diagnosis and management of cats with spinal cord disease and to outline the most common diseases responsible. Patient group: Patients of either gender and all ages and breeds can be affected by spinal cord disease. Evidence base: Many diseases affecting the spinal cord of cats, which include fibrocartilaginous embolic myelopathy, intervertebral disc disease, exogenous spinal cord trauma, spinal cord lymphosarcoma and feline infectious peritonitis, are well described in the literature. Many of these descriptions, however, have been based on case reports or series. While there have been several retrospective studies that describe the characteristics and incidence of these diseases in cats, there are no long term treatment trials or outcome studies to assist with prognostic determinations. Practical relevance: A number of systemic diseases are associated with neurological deficits. Most systemic diseases that impact on the nervous system result in multifocal neurological signs; however, isolated deficits can also be observed. This article reviews the clinical signs, pathophysiology, diagnosis, treatment and prognosis of four important systemic diseases with neurological consequences: feline infectious peritonitis, toxoplasmosis, hypertension and hepatic encephalopathy. Clinical challenges: Early recognition of systemic signs of illness in conjunction with neurological deficits will allow for prompt diagnosis and treatment. While neurological examination of the feline patient can undoubtedly be challenging, hopefully the accompanying articles in this special issue will enable the clinician to approach these cases with more confidence. Evidence base: The veterinary literature contains numerous reports detailing the impact of systemic disease on the nervous system. Unfortunately, very few references provide detailed descriptions of large cohorts of affected cats. This review summarises the literature underpinning the four key diseases under discussion. Practical relevance: Although seizures occur less commonly in cats compared with dogs, they are one of the most common forms of neurological disease in the feline patient. Cats may experience both focal (partial) and generalized seizures and causes are divided into primary disorders, in which there is no underlying cause (ie, idiopathic epilepsy), and secondary disorders. Cats with secondary seizure disorders have either an underlying structural lesion or metabolic disease. Patient group: Seizures affect cats of all ages. Cats with idiopathic epilepsy tend to be younger (approximately 3.5 years) than cats with secondary seizure disorders (approximately 8 years). Audience: This review of feline seizures is directed at all veterinarians who treat cats, both in an emergency setting as well as in general practice. Clinical challenges: Refractory seizures are often a diagnostic and therapeutic challenge. A systematic approach to the seizuring cat is described, easing the task of diagnosing the cause of the seizures. In addition, novel antiepileptics are discussed, which can be used as add-on drugs in challenging feline seizure cases. Evidence base: Compared with the canine counterpart, the literature regarding treatment of feline seizures is less established. Recent clinical trials and studies are focusing on new treatment options for feline seizures. Specifically, these studies, some of which are ongoing, have led to the use of levetiracetam, zonisamide and pregabalin as add-on antiepileptics in cases that are refractory to phenobarbital. Practical relevance: Weakness is recognized somewhat infrequently in cats, but is an important manifestation of neurological disease. The clinician must perform a complete neurological examination to determine the neuroanatomic basis for the weakness. As for all species, the neuroanatomic diagnosis allows the clinician to generate an appropriate differential diagnosis, to design a diagnostic plan, to prognosticate, and ultimately to develop a treatment plan. Clinical challenges: The cause(s) of neurological weakness in the cat may be difficult to determine without access to advanced imaging modalities, cerebrospinal fluid analysis or electrodiagnostics. However, an accurate neuroanatomic diagnosis allows the clinician to pursue preliminary anomalous (vertebral anomalies), metabolic (eg, diabetes mellitus, electrolyte abnormalities) and neoplastic differentials via blood work, vertebral column and thoracic radiography, and abdominal ultrasound. Subsequently, referral to a specialty veterinary hospital may be warranted to pursue advanced neurodiagnostics. Audience: This review provides a framework for generating a neuroanatomic and differential diagnosis in the weak cat. It also discusses the pathogenesis and clinical signs associated with the most common neurological differentials for feline paresis. As such, it is aimed at both primary health care and specialty veterinarians. Patient group: The neurological conditions discussed in this review cause weakness in cats of all age groups. Practical relevance: Generalised ataxia is one of the most common neurological presentations identified in cats in practice. The causes can be subdivided into three forms on the basis of the neuroanatomical diagnosis: cerebellar, vestibular and sensory (spinal or general proprioceptive) ataxia. The type of ataxia will determine the diagnostic procedures and select the differential diagnoses, and an accurate neuroanatomical diagnosis is therefore essential. The differential diagnosis list can then be further tailored on the basis of patient signalment, clinical presentation and progression. Global importance: Irrespective of the world region, most of the causes of generalised ataxia in the cat are similar and many have been identified for a number of years (cerebellar hypoplasia has been recognised since the late 19th century). However, it is the advent of new technology, in particular veterinary access to magnetic resonance imaging, which has resulted in particularly rapid advances in our understanding, investigation and management of these different forms of ataxia. Audience: This article introduces the classification of patients with ataxia on the basis of their clinical presentation, discusses the common differential diagnoses associated with each form, and briefly reviews the more important diseases from a clinical perspective. Practical relevance: Neurological diagnosis in veterinary practice can be very challenging, especially as many animals with neurological signs present as emergencies. Nevertheless, even in the absence of specialist facilities for definitively diagnosing neurological disorders, a great deal of information can be gained with some basic knowledge and a logical stepwise approach. Clinical challenges: A lack of initial consideration as to where exactly the problem might be localised within the nervous system, and what kind of disease processes may be in operation there, is the most common cause of failure in the diagnosis of neurological conditions in cats. Too often, this presents a hurdle that pushes the clinician into neglecting the neurological evaluation in favour of making the best guess at which diagnostic tests may achieve a diagnosis. Audience: This article is aimed at all first opinion practitioners who see cats as, undoubtedly, whatever the presentation, the approach to a suspected neurological case can be daunting for even the calmest and most patient clinician. The enzyme rhodanese (EC 2.8.1.1) is an ubiquitous enzyme which is present in all living organisms, from bacteria to man. It is speculated that this enzyme plays a central role in cyanide detoxification. However, its wide tissue distribution suggests this enzyme might perform other functions beside cyanide detoxification. Although the distribution of rhodanese in different tissues of human and domestic animals has been studied, little is known about the pattern of distribution and physiological roles of this enzyme in the cat. The purpose of this investigation was to determine the enzyme levels and compare the distribution of this enzyme in different tissues of the cat. A selection of tissue samples was assayed for rhodanese activity. The protein content of tissue extracts and enzymatic activities were calculated as units per gram tissue and units per milligram protein of the tissue. Results showed that in terms of units per milligram protein of the tissue (specific activity of the enzyme), colon and rectum mucosal layers and testis were the richest sources of the enzyme followed by ovary, mucosal layer of jejunum and liver. With respect to units/gram tissue, liver followed by testis, colon and rectum mucosal layers, ovary and mucosa of jejunum exhibited highest activities. The results of this study will allow one to speculate on the involvement of rhodanese in several biochemical and physiological functions in different tissues and organs of this species. Optic flow analysis has been extensively investigated in several extrastriate visual areas of the primate, but it remains unclear whether and how the striate cortex is involved in the processing. In the present study 156 striate neurons in anaesthetized cats were tested with simulated optic flow patterns. More than 80% of the cells responded to the stimuli, about 30% significantly preferred translation to radiation and rotation, but few, if any, displayed clear preference for radial or circular motion. Most responsive neurons had fairly good direction sensitivity for their optimal stimuli. Generally, the differences between simple and complex cells were small and insignificant. Although translation obviously prevailed over radiation and rotation in driving more cells and eliciting stronger responses, the responsivity to complex flow fields was not predictable with the direction preference to planar motion. These results suggest that the striate contribution to optic flow analysis may go beyond the direction selectivity to local translation and there might be some kind of specific mechanism to complex motion features. The article discusses the results of a study that examined the effect of experimental hyperlipemia on insulin resistance in seven healthy cats. Topics discussed include higher serum triglyceride and free fatty acid concentrations when lipid-heparin was administered compared with saline, lower mean glucose infusion rate when lipid-heparin was administered and the conclusion that experimental hyperlipemia induced insulin resistance in cats.

The article presents the insights of veterinarian Elizabeth O'Brien regarding the care of cats in The Cat Clinic. O'Brien believes that deworming was required for caring cats without homes and it was her obligation to educate her clients regarding its importance to their pets. She adds that it will keep the animal free from internal parasites and benefits its owners' safety and one who had contacts with it.
