ABSTRACTLeukemia is a common blood cancer, whose treatment usually necessitates chemo/radiotherapy and bone marrow transplant. Hence, safer and more effective options are urgently needed. Mylabris, the dried body of blister beetles, has been used extensively in traditional Chinese medicine. This study applied bioinformatics and systematic pharmacology to investigate the mechanism of action of mylabris in the treatment of leukemia. Five effective components and 35 corresponding target proteins were identified by screening the TCMSP database; whereas 776 genes related to leukemia were selected using OMIM, GeneCards, and the Therapeutic Target Database. Eight genes common to mylabris and leukemia were identified. Protein-protein interaction network analysis and a component-target-pathway diagram identified TP53and PTENas key gene targets of mylabris in the treatment of leukemia. GO enrichment analysis pointed to DNA damage and cell cycle disorder caused by p53 signaling as the most significant processes; whereas KEGG enrichment pointed to the p53 signaling pathway. In summary, mylabris may exert a therapeutic effect on leukemia by triggering DNA damage, inducing apoptosis, as well as inhibiting the growth and proliferation of tumor cells through the regulation of TP53and PTEN. These findings provide a mechanistic rationale for the treatment of leukemia with traditional Chinese medicine.

Lightweight and high strength CFRP material has been applied to airspace industry to realize lightweight aircraft. Nowadays, composite T-joint is a common adhesive joint type used in aircraft CFRP load-bearing structure. This study employed T300/7901 prepreg and J-272 film as the adhesive in the deltoid of T-joints, and produce T-shaped specimens via co-curing. The start point of failure location is located at deltoid through pull-off test combined with digital image correlation system. In the pull-off test, the failure of the T-joint specimen, the deltoid of which was filled with T300/7901 prepreg and J-272 film, exhibited progressive failure mechanism from rib web to skin. Compared to adherence with T300/7901 prepreg, the initial failure load and toughness of composite T-joints adhered with J-272 film were increased 24.3% and 34.7%, in which J-272 film boosted the interfacial binding between the rib web and the skin.

•The adhesion between polymer and metal was enhanced by adding graphene flakes.•The bonding strength increased by an order of magnitude with only 0.5 wt% GFs.•The adhesion enhancement mechanism mainly relies on mechanical interlocking.

•The crystallinity of the composites is increased by about 10%.•Graphene flakes and polymer molecules exhibit a highly ordered state.•The fractional free volume is reduced from 6.52 vol% to 5.43 vol%.•The helium leak rate of composites is reduced by 98.6%.
