4. Efficacy and Safety of Radiotherapy Plus Relugolix in Men With Localized or Advanced Prostate Cancer

Abstract

IMPORTANCE: Combination androgen deprivation therapy (ADT) with radiotherapy is commonly used for patients with localized and advanced prostate cancer. OBJECTIVE: To assess the efficacy and safety of the oral gonadotropin-releasing hormone antagonist relugolix with radiotherapy for treating prostate cancer. DESIGN, SETTING, AND PARTICIPANTS: This multicenter post hoc analysis of patients with localized and advanced prostate cancer receiving radiotherapy in 2 randomized clinical trials (a phase 2 trial of relugolix vs degarelix, and a subset of the phase 3 HERO trial of relugolix vs leuprolide acetate) included men who were receiving radiotherapy and short-term (24 weeks) ADT (n = 103) from 2014 to 2015 and men receiving radiotherapy and longer-term (48 weeks) ADT (n = 157) from 2017 to 2019. The data were analyzed in November 2022. INTERVENTIONS: Patients receiving short-term ADT received relugolix, 120 mg, orally once daily (320-mg loading dose) or degarelix, 80 mg, 4-week depot (240-mg loading dose) for 24 weeks with 12 weeks of follow-up. Patients receiving longer-term ADT received relugolix, 120 mg, orally once daily (360-mg loading dose) or leuprolide acetate injections every 12 weeks for 48 weeks, with up to 90 days of follow-up. MAIN OUTCOMES AND MEASURES: Castration rate (testosterone level <50 ng/dL [to convert to nmol/L, multiply by 0.0347) at all scheduled visits between weeks 5 and 25 for patients receiving short-term ADT and weeks 5 and 49 for patients receiving longer-term ADT. RESULTS: Of 260 patients (38 Asian [14.6%], 23 Black or African American [8.8%], 21 Hispanic [8.1%], and 188 White [72.3%] individuals), 164 (63.1%) received relugolix. Relugolix achieved castration rates of 95% (95% CI, 87.1%-99.0%) and 97% (95% CI, 90.6%-99.0%) among patients receiving short-term and longer-term ADT, respectively. Twelve weeks post–short-term relugolix, 34 (52%) achieved testosterone levels to baseline or more than 280 ng/dL. Ninety days post longer-term ADT, mean (SD) testosterone levels were 310.5 (122.4) (106.7) ng/dL (relugolix; n = 15) vs 53.0 ng/dL (leuprolide acetate; n = 8) among the subset assessed for testosterone recovery. Castration resistance-free survival was not statistically different between the relugolix and leuprolide acetate cohorts (hazard ratio, 0.97; 95% CI, 0.35-2.72; P = .62). Adverse events grade 3 or greater for short-term or longer-term relugolix (headache, hypertension, and atrial fibrillation) were uncommon (less than 5%). CONCLUSIONS AND RELEVANCE: The results of these 2 randomized clinical trials suggest that relugolix rapidly achieves sustained castration in patients with localized and advanced prostate cancer receiving radiotherapy. No new safety concerns were identified when relugolix was used with radiotherapy.

ABSTRACTIntroductionThe introduction of targeted therapy and immunotherapy has tremendously changed the clinical outcomes and prognosis of cancer patients. Despite innovative pharmacological therapies and improved radiotherapy (RT) techniques, patients continue to suffer from side effects, of which oral mucositis (OM) is still the most impactful, especially for quality of life.Areas CoveredWe provide an overview of current advances in cancer pharmacotherapy and RT, in relation to their potential to cause OM, and of the less explored and more recent literature reports related to the best management of OM. We have analyzed natural/antioxidant agents, probiotics, mucosal protectants and healing coadjuvants, pharmacotherapies, immunomodulatory and anticancer agents, photobiomodulation and the impact of technology.Expert OpinionThe discovery of more precise pathophysiologic mechanisms of CT and RT-induced OM has outlined that OM has a multifactorial origin, including direct effects, oxidative damage, upregulation of immunologic factors, and effects on oral flora. A persistent upregulated immune response, associated with factors related to patients’ characteristics, may contribute to more severe and long-lasting OM. The goal is strategies to conjugate individual patient, disease, and therapy-related factors to guide OM prevention or treatment. Despite further high-quality research is warranted, the issue of prevention is paramount in future strategies.

12. Economic Risk Assessment of Future Debris Flows by Machine Learning Method

Abstract

A reliable economic risk map is critical for effective debris-flow mitigation. However, the uncertainties surrounding future scenarios in debris-flow frequency and magnitude restrict its application. To estimate the economic risks caused by future debris flows, a machine learning-based method was proposed to generate an economic risk map by multiplying a debris-flow hazard map and an economic vulnerability map. We selected the Gyirong Zangbo Basin as the study area because frequent severe debris flows impact the area every year. The debris-flow hazard map was developed through the multiplication of the annual probability of spatial impact, temporal probability, and annual susceptibility. We employed a hybrid machine learning model—certainty factor-genetic algorithm-support vector classification—to calculate susceptibilities. Simultaneously, a Poisson model was applied for temporal probabilities, while the determination of annual probability of spatial impact relied on statistical results. Additionally, four major elements at risk were selected for the generation of an economic loss map: roads, vegetation-covered land, residential buildings, and farmland. The economic loss of elements at risk was calculated based on physical vulnerabilities and their economic values. Therefore, we proposed a physical vulnerability matrix for residential buildings, factoring in impact pressure on buildings and their horizontal distance and vertical distance to debris-flow channels. In this context, an ensemble model (XGBoost) was used to predict debris-flow volumes to calculate impact pressures on buildings. The results show that residential buildings occupy 76.7% of the total economic risk, while road-covered areas contribute approximately 6.85%. Vegetation-covered land and farmland collectively represent 16.45% of the entire risk. These findings can provide a scientific support for the effective mitigation of future debris flows.

13. Denosumab improves trabecular bone score in relationship with decrease in fracture risk of women exposed to aromatase inhibitors

Abstract

Purpose: Trabecular bone score (TBS) is a gray-level textural metric that has shown to correlate with risk of fractures in several forms of osteoporosis. The value of TBS in predicting fractures and the effects of bone-active drugs on TBS in aromatase inhibitors (AIs)-induced osteoporosis are still largely unknown. The primary objective of this retrospective study was to assess the effects of denosumab and bisphosphonates (BPs) on TBS and vertebral fractures (VFs) in women exposed to AIs. Methods: 241 consecutive women (median age 58 years) with early breast cancer undergoing treatment with AIs were evaluated for TBS, bone mineral density (BMD) and morphometric VFs at baseline and after 18–24 months of follow-up. During the study period, 139 women (57.7%) received denosumab 60 mg every 6 months, 53 (22.0%) BPs, whereas 49 women (20.3%) were not treated with bone-active drugs. Results: Denosumab significantly increased TBS values (from 1.270 to 1.323; P< 0.001) accompanied by a significant decrease in risk of VFs (odds ratio 0.282; P= 0.021). During treatment with BPs, TBS did not significantly change (P= 0.849) and incidence of VFs was not significantly different from women untreated with bone-active drugs (P= 0.427). In the whole population, women with incident VFs showed higher decrease in TBS vs. non-fractured women (P= 0.003), without significant differences in changes of BMD at any skeletal site. Conclusions: TBS variation predicts fracture risk in AIs treated women. Denosumab is effective to induce early increase of TBS and reduction in risk of VFs.

Abstract

Approximately 450 000 children are born with familial hypercholesterolaemia worldwide every year, yet only 2·1% of adults with familial hypercholesterolaemia were diagnosed before age 18 years via current diagnostic approaches, which are derived from observations in adults. We aimed to characterise children and adolescents with heterozygous familial hypercholesterolaemia (HeFH) and understand current approaches to the identification and management of familial hypercholesterolaemia to inform future public health strategies.

15. Genomic insights into a Proteus mirabilisstrain inducing avian cellulitis

Abstract

Proteus mirabilis, a microorganism distributed in soil, water, and animals, is clinically known for causing urinary tract infections in humans. However, recent studies have linked it to skin infections in broiler chickens, termed avian cellulitis, which poses a threat to animal welfare. While Avian Pathogenic Escherichia coli(APEC) is the primary cause of avian cellulitis, few cases of P. mirabilisinvolvement are reported, raising questions about the factors facilitating such occurrences. This study employed a pan-genomic approach to investigate whether unique genes exist in P. mirabilisstrains causing avian cellulitis. The genome of LBUEL-A33, a P. mirabilisstrain known to cause this infection, was assembled, and compared with other P. mirabilisstrains isolated from poultry and other sources. Additionally, in silicoserogroup analysis was conducted. Results revealed numerous genes unique to the LBUEL-A33 strain. No function in cellulitis was identified for these genes, and in silicoinvestigation of the virulence potential of LBUEL-A33’s exclusive proteins proved inconclusive. These findings support that multiple factors are necessary for P. mirabilisto cause avian cellulitis. Furthermore, this species likely employs its own unique arsenal of virulence factors, as many identified mechanisms are analogous to those of E. coli. While antigenic gene clusters responsible for serogroups were identified, no clear trend was observed, and the gene cluster of LBUEL-A33 did not show homology with any sequenced Proteusserogroups. These results reinforce the understanding that this disease is multifactorial, necessitating further research to unravel the mechanisms and underpin the development of control and prevention strategies.

16. Clonal relationship, virulence genes, and antimicrobial resistance of Morganella morganiiisolated from community-acquired infections and hospitalized patients: a neglected opportunistic pathogen

Abstract

Morganella morganiiis a bacterium belonging to the normal intestinal microbiota and the environment; however, in immunocompromised individuals, this bacterium can become an opportunistic pathogen, causing a series of diseases, both in hospitals and in the community, being urinary tract infections more prevalent. Therefore, the objective of this study was to evaluate the prevalence, virulence profile, and resistance to antimicrobials and the clonal relationship of isolates of urinary tract infections (UTI) caused by M. morganii, both in the hospital environment and in the community of the municipality of Londrina-PR, in southern Brazil, in order to better understand the mechanisms for the establishment of the disease caused by this bacterium. Our study showed that M. morganiipresents a variety of virulence factors in the studied isolates. Hospital strains showed a higher prevalence for the virulence genes zapA, iutA, and fimH, while community strains showed a higher prevalence for the ireAand iutAgenes. Hospital isolates showed greater resistance compared to community isolates, as well as a higher prevalence of multidrug-resistant (MDR) and extended-spectrum beta lactamase (ESBL)-producing isolates. Several M. morganiiisolates from both sources showed high genetic similarity. The most prevalent plasmid incompatibility groups detected were FIB and I1, regardless of the isolation source. Thus, M. morganiiisolates can accumulate virulence factors and antimicrobial resistance, making them a neglected opportunistic pathogen.

19. Diagnostic accuracy and safety of EUS-guided end-cutting fine-needle biopsy needles for tissue sampling of abdominal and mediastinal lymphadenopathies: a prospective multicenter series.

Abstract

The role of the newer EUS fine-needle biopsy needles in lymphadenopathies (LAs) is still under evaluation. We aimed to evaluate the diagnostic accuracy and adverse event rate of EUS-guided fine-needle biopsy sampling (EUS-FNB) in diagnosing LAs. From June 2015 to June 2022, all patients referred to 4 institutions for EUS-FNB of mediastinal and abdominal LAs were enrolled. Twenty-two–gauge Franseen tip or 25-gauge fork-tip needles were used. The criterion standard for positive results was surgery or imaging and clinical evolution over a follow-up of at least 1 year. One hundred consecutive patients were enrolled, consisting of those with a new diagnosis of LA (40%), presence of LA with a previous history of neoplasia (51%), or suspected lymphoproliferative disease (9%). EUS-FNB was technically feasible in all LA patients with 2 to 3 passes (mean, 2.62 ±.93). The overall sensitivity, positive predictive value, specificity, negative predictive value, and accuracy for EUS-FNB were 96.20%, 100%, 100%, 87.50%, and 97.00%, respectively. Histologic analysis was feasible in 89% of cases. Cytologic evaluation was performed in 67% of specimens. A statistical difference between the accuracy of the 22-gauge or 25-gauge needle (P =.63) was not found. A subanalysis on lymphoproliferative disease revealed a sensitivity and accuracy of 89.29% and 90.0%, respectively. No adverse events were recorded. EUS-FNB with new end-cutting needles is a valuable and safe method to diagnose LAs. The high quality of histologic cores and the good amount of tissue allowed a complete immunohistochemical analysis of metastatic LAs and precise subtyping of the lymphomas. Abstract

Drug-coated balloons (DCB) represent 1 of the most promising innovations in interventional cardiology and may represent a valid alternative to drug-eluting stents. Currently, some sirolimus-coated balloons (SCB) are being investigated for several coronary artery disease applications. This study sought to understand the role of a novel SCB for the treatment of coronary artery disease. EASTBOURNE (All-Comers Sirolimus-Coated Balloon European Registry) is a prospective, multicenter, investigator-driven clinical study that enrolled real-world patients treated with SCB. Primary endpoint was target lesion revascularization (TLR) at 12 months. Secondary endpoints were procedural success, myocardial infarction (MI), all-cause death, and major adverse clinical events (a composite of death, MI, and TLR). All adverse events were censored and adjudicated by an independent clinical events committee. A total population of 2,123 patients (2,440 lesions) was enrolled at 38 study centers in Europe and Asia. The average age was 66.6 ± 11.3 years, and diabetic patients were 41.5%. De novo lesions (small vessels) were 56%, in-stent restenosis (ISR) 44%, and bailout stenting occurred in 7.7% of the patients. After 12 months, TLR occurred in 5.9% of the lesions, major adverse clinical events in 9.9%, and spontaneous MI in 2.4% of the patients. The rates of cardiac/all-cause death were 1.5% and 2.5%, respectively. The primary outcome occurred more frequently in the ISR cohort (10.5% vs 2.0%; risk ratio: 1.90; 95% CI: 1.13-3.19). After multivariate Cox regression model, the main determinant for occurrence of the primary endpoint was ISR (OR: 5.5; 95% CI: 3.382-8.881). EASTBOURNE, the largest DCB study in the coronary field, shows the safety and efficacy of a novel SCB in a broad population of coronary artery disease including small vessels and ISR patients at mid-term follow-up. 21. The effect of two sessions of combined jump and sprint training per week on fitness parameters in soccer players. A randomized controlled trial.

Abstract

This study aimed to investigate the effect of a combined jump and sprint training program, two sessions a week for 6 weeks, on sprinting, change of directions (COD) and jumping performance in semiprofessional soccer players. Twenty soccer players were enrolled in this randomized controlled trial (age 20 ± 2 years, body mass 74.3 ± 5.9 kg). Players were randomized into two groups such as training group (TG, n = 10 players) or control group (CG, n = 10 players). Physical tests were performed before and after 6 weeks of training such as sprint 10 m, sprint 30 m, 505-COD test and standing long jump (LJ). The two groups performed the same training except for the combined jump and sprint training performed twice a week by TG. After 6 weeks of training, between-group analysis reported statistical difference in favor of the TG in sprint 10 m (p = 0.015, η² = 0.295, large), sprint 30 m (p < 0.001, η² = 0.599, large), in 505-COD (p = 0.026, η² = 0.154, large), and LJ (p = 0.025, η² = 0.027, small). These data indicate that combined sprint and jump training, when performed twice a week, for the duration of 6 weeks, in addition to the regular team training, can improve specific physical performance in male soccer players. This study has shown that a volume increment of 10% after 3 weeks of training can be a suitable training dose progression and that a combination of 64-70 jumps and 675-738 m of sprinting training per session can yield benefits in sprint, COD and jump performance. Abstract

Here we describe highly compact, click compatible, and photoactivatable dyes for super-resolution fluorescence microscopy (nanoscopy). By combining the photoactivatable xanthone (PaX) core with a tetrazine group, we achieve minimally sized and highly sensitive molecular dyads for the selective labeling of unnatural amino acids introduced by genetic code expansion. We exploit the excited state quenching properties of the tetrazine group to attenuate the photoactivation rates of the PaX, and further reduce the overall fluorescence emission of the photogenerated fluorophore, providing two mechanisms of selectivity to reduce the off-target signal. Coupled with MINFLUX nanoscopy, we employ our dyads in the minimal-linkage-error imaging of vimentin filaments, demonstrating molecular-scale precision in fluorophore positioning.

25. Assessment of exposure to perfluorinated industrial substances and risk of immune suppression in Greenland and its global context: a mixed-methods study

Perfluoroalkyl substances (PFASs) are ubiquitous global contaminants that do not readily biodegrade and are therefore routinely found worldwide in wildlife, humans, and the environment. There is a paucity of global assessments to understand regional and continental differences in exposure to PFASs and the associated health risks, including those for Indigenous Arctic communities who consume high trophic marine diets. We aimed to estimate the long-term exposure of dietary PFASs from consumption of polar bear and ringed seal meat and establish its association with blood serum concentrations of PFASs in Inuit in Ittoqoortoormiit (Scoresby Sound), East Greenland. We also aimed to assess the risk of immune suppression on the basis of European Food Safety Authority (EFSA) thresholds for weekly intake and blood serum concentrations of PFASs. Last, we conducted a worldwide risk assessment based on blood concentrations of PFASs emphasising Arctic exposure in a global context.

26. An Interpretation of Articles 18-23 of Directive (EU) 2019/790: Open Issues in Contractual Copyright and Related Rights Law

Abstract

Although creators have always been placed at the centre of the copyright system, they generally face difficulties in being fairly remunerated for their work. Articles 18-23 Directive (EU) 2019/790 require Member States to establish a series of protective measures in favour of authors and performers who license or transfer the exclusive economic rights to their works or performances to third parties for the purpose of exploitation. These measures constitute an absolute novelty at the European Union level and at the level of most Member States, and present a potential which must not be wasted. However, these provisions are in many ways complex and ambiguous, so that their effectiveness could be undermined in various ways. This article focuses on some still open issues concerning Arts. 18-23 Directive (EU) 2019/790 and attempts to provide the most appropriate interpretation in order to achieve the intended goals. In doing so, particular attention is paid to previous fragmented national and European experiences in the field of contractual copyright and related rights law.

29. Serum testosterone mirrors inflammation parameters in females hospitalized with COVID-19

Abstract

Background: While low testosterone (T) was described as a predictor of unfavorable coronavirus-disease 19 (COVID-19) outcome in men, data concerning the role of T in women with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection are scant and limited to small cohorts. This study investigated the relationship between serum T values and outcomes of COVID-19 in a large female hospitalized cohort. Methods: One-hundred-sixty-eight adult women (median age 77, range 18–100 years; 154 in post-menopause) hospitalized for COVID-19 were assessed for PaO2/Fio2 ratio, serum T and inflammatory parameters. Results: Median duration for hospital stay was 14.2 days (range 1–115) with overall mortality of 26% (n= 44). Subjects who died were significantly older (p&lt; 0.001), had significantly more comorbidities (p= 0.015) and higher serum T (p= 0.040), white blood cells (p= 0.007), c-reactive protein (CRP; p&lt; 0.001), interleukin-6 (IL-6; p&lt; 0.001), procalcitonin (PCT; p&lt; 0.001), lactate dehydrogenase (LDH; p= 0.001), D-dimer (p= 0.035), fibrinogen (p= 0.038) and lower serum free-triiodothyronine (FT3; p&lt; 0.001) and luteinizing hormone (LH; p= 0.024) values. In post-menopausal women, significant associations were observed between T levels and serum CRP (rho: 0.23; p= 0.002), IL-6 (rho: 0.41; p&lt; 0.001), LDH (rho: 0.34; p&lt; 0.001), D-Dimer (rho: 0.21; p= 0.008), PCT (rho: 0.26; p= 0.001) and HDL cholesterol (rho:  – 0,22, p= 0.008). In multivariate regression analyses, serum T maintained the significant association with mortality after correction for age, coexistent comorbidities and serum LH and FT3, whereas it was lost after correction for inflammatory parameters. Conclusion: In females, high serum T levels might be a mirror of inflammatory phenotype and worse COVID-19 course.

Abstract

Vacuum extraction is the most common choice to assist vaginal delivery, but there are still concerns regarding the neonatal injuries it may cause. This study aimed to evaluate the rate of intracranial injuries assessed by cranial ultrasound (cUS) among infants born by vacuum extraction, and the relationship with maternal and perinatal factors. This was a single-center retrospective study carried out in a level-3 neonatal unit. A total of 593 term and late preterm infants born by vacuum-assisted delivery were examined with a cUS scan within 3 days after birth. Major head injuries were clinically silent and occurred in 2% of the infants, with a rate of intracranial haemorrhage of 1.7%. Regardless of obstetric factors, the risk of cranial injury was increased in infants requiring resuscitation at birth (p = 0.04, OR 4.1), admitted to NICU (p = 0.01, OR 5.5) or with perinatal asphyxia (p < 0.01, OR 21.3). Maternal age ≥40 years correlated both with adverse perinatal outcomes (p < 0.05) and the occurrence of major injury (p = 0.02, OR 4.6). Overall, vacuum extraction is a safe procedure for neonates. Head injuries are usually mild and asymptomatic, and with spontaneous recovery. However, the rate of major cranial injuries in our cohort warrants further investigation to support a cUS screening, particularly for infants requiring respiratory support at birth. 32. Intrapartum use of zidovudine in a large cohort of pregnant women living with HIV in Italy.

Abstract

Background: Intravenous administration of zidovudine (ZDV) during labour is a key step for vertical HIV transmission (VT) prevention, but there is no evidence of benefit when maternal HIV-RNA at delivery is <50 copies/mL. The aim of this study is evaluating the appropriateness of intrapartum ZDV use in Italy.Methods: Observational study including mother-infant pairs with perinatal HIV exposure during 2002-2019, enrolled in the Italian Register for HIV Infection in Children. Univariable and multivariable logistic regression were used to evaluate factors associated with VT.Results: A total of 3,861 infants, born from 3,791 pregnancies were included. The frequency of ZDV use was 79.9%, 92.1%, 93.7% and 92.8% when HIV-RNA was not available, ≥400 copies, between 50 and 399 copies, and <50 copies/mL. Thirty-three out of 3861 (0.85%) infants were subsequently diagnosed with HIV, 25/3861 (0.6%) of them born to mothers receiving intrapartum ZDV, and 31 (93.9%) to mothers with HIV-RNA ≥50 copies/mL or not available. In women with HIV-RNA < 50 copies/mL, ART discontinuation during pregnancy was the strongest risk factor for VT (odds ratio, OR, 23.1, 95%CI 2.4-219.3), while a higher gestational age (OR 0.6, 95%CI 0.4-0.8) and PEP administration to the newborn (aOR 0.004, 95%CI <0.0001-0.4) were protective factors. Intrapartum ZDV administration did not influence the final outcome in this group.Conclusions: In ART era, more transmission events may occur in utero, limiting value of intrapartum ZDV, particularly for women with suppressed HIV-RNA load. 34. COVID-19 pandemic impact on people with diabetes: results from a large representative sample of Italian older adults.

Abstract

Aims: Restrictions imposed to prevent SARS-CoV-2 transmission should be weighed against consequences on vulnerable groups' health. Lifestyles and disease management of older people with diabetes might have been differentially impacted compared to non-chronic individuals.Methods: A cross-sectional study (LOST in Lombardia) was conducted on a representative full sample of 4 400 older adults (17th-30th November 2020), collecting data on lifestyles, mental health and access to care before and during the pandemic.Results: We compared 947 (51.9%) people with diabetes and 879 (48.1%) healthy subjects reporting no chronic conditions. People with diabetes reported more frequently increased physical activity (odds ratio, OR 2.65, 95% confidence internals, CI 1.69-4.13), drinks/week reduction (OR 6.27, 95%CI 3.59-10.95), increased consumption of fruit (OR 2.06, 95%CI 1.62-2.63), vegetables (OR 1.41, 95%CI 1.10-1.82), fish (OR 2.51, 95%CI 1.74-3.64) and olive oil (OR 3.54, 95%CI 2.30-5.46). People with diabetes increased telephone contacts with general practitioners (OR 3.70, 95%CI 2.83-4.83), hospitalisations (OR 9.01, 95%CI 3.96-20.51), visits and surgeries cancellations (OR 3.37, 95%CI 2.58-4.42) and treatment interruptions (OR 1.95, 95%CI 1.33-2.86).Conclusions: Pandemic adverse effects occurred but are heterogenous in a population with chronic diseases, who seized the opportunity to improve health behaviours, despite health system difficulties guaranteeing routine care, within and beyond COVID-19. 36. Photoactivatable Large Stokes Shift Fluorophores for Multicolor Nanoscopy

Abstract

We designed caging-group-free photoactivatable live-cell permeant dyes with red fluorescence emission and ∼100 nm Stokes shifts based on a 1-vinyl-10-silaxanthone imine core structure. The proposed fluorophores undergo byproduct-free one- and two-photon activation, are suitable for multicolor fluorescence microscopy in fixed and living cells, and are compatible with super-resolution techniques such as STED (stimulated emission depletion) and PALM (photoactivated localization microscopy). Use of photoactivatable labels for strain-promoted tetrazine ligation and self-labeling protein tags (HaloTag, SNAP-tag), and duplexing of an imaging channel with another large Stokes shift dye have been demonstrated.

37. What Does It Take to Complete the Cape Epic?

J Strength Cond Res36(12): 3513–3520, 2022—This study aimed to describe the racing and training demands of the Cape Epic. Six male mountain bike riders (age: 39 ± 7 years, height: 181 ± 3 cm, and body mass: 78.7 ± 8.1 kg) trained for 4.5 months and took part in the Cape Epic. Training and racing data (prologue, stage 1, and 2) were analyzed, and riders were tested in the laboratory on 3 distinct occasions for maximal oxygen uptake (V̇O2max), maximal work rate (Ẇmax), and power output associated with the respiratory compensation point (RCPPO). Statistical significance was set at p≤ 0.05. With race durations of 1.5 ± 0.2, 6.5 ± 1.2, and 6.4 ± 1.4 hours for, respectively, prologue, stage 1, and 2, normalized power was higher in prologue (3.73 ± 0.72 W·kg−1) compared with stages 1 (3.06 ± 0.59 W·kg−1, p< 0.001) and 2 (2.94 ± 0.69 W·kg−1, p< 0.001). Riders spent more time in power zones 1 and 2 (as %RCPPO) and less time in zones 4 and 5, during stage 2 compared with prologue (all zones p≤ 0.028). Despite no changes in V̇O2max or Ẇmax, RCPPOincreased from midtraining (3.89 ± 0.61 W·kg−1) to prerace testing (4.08 ± 0.64 W·kg−1, p= 0.048). No differences were found between base and build training phases for time in power zones. In conclusion, the Cape Epic requires an ability to sustain high submaximal power outputs for several hours as well as an ability to repeat high-intensity efforts throughout the race. A well-balanced program, incorporating a pyramidal intensity distribution, may be used as a starting point for the design of optimal training approaches.

38. Recent Jet Measurements in ALICE

Abstract

The precise measurement of jets in pp collisions plays a critical role in the study of quantum chromodynamics (QCD). Measurements of jets in relativistic heavy-ion collisions are additionally useful for probing QCD matter in the regime of high densities and temperatures since the partons in the jet evolution interact strongly with the medium. This phenomenon is called jet quenching and is characterized by both jet energy loss and modification of the internal structure of the jet. The ALICE experiment is well suited for studies of jets and their corresponding internal structure due to the high precision tracking system. These proceedings report on an overview of recent ALICE jet results in both pp and Pb–Pb collisions. Results in pp collisions include measurements of generalized angularities of groomed and inclusive jets, a measurement of the Lund jet plane, and a measurement of jet axis differences. Results from Pb–Pb collisions include measurements of jet splittings, studies of subjet fragmentation, as well as a measurement of the jet nuclear modification factor made with machine learning techniques. These new results, when combined with the corresponding theoretical comparisons, provide information regarding the QCD processes of the parton shower and hadronization, as well as the underlying physics of jet quenching.

39. Suspect and non-target screening of chemicals of emerging Arctic concern in biota, air and human serum.

Abstract

Contaminants of emerging concern receive increasing attention in the Arctic environment. The aim of this study was to screen for chemicals of emerging Arctic concern (CEACs) in different types of Arctic samples including biota, air and human serum. We used a combination of gas chromatography (GC) and liquid chromatography (LC) with high resolution mass spectrometry (HRMS) for suspect and non-target screening (NTS). Suspect screening of 25 CEACs was based on published in-silico approaches for the identification of CEACs and revealed tetrabromophthalic anhydride (TBPA) in pilot whale and air, albeit with low detection frequencies (17 and 33%, respectively). An NTS workflow detected 49, 42, 31 and 30 compounds in pilot whale, ringed seal, air, and human serum, respectively, at confidence level 2 and 3. Although legacy POPs still dominated the samples, 64 CEACs were tentatively identified and further assessed for persistence (P), bioaccumulation (B), mobility (M), toxicity (T), and long-range transport potential (LRTP). While four PBT compounds were identified, 37 PMT substances dominated among these 64 compounds. Our study indicated that many chemicals of potential risk might be present in Arctic samples and would benefit from confirmation and further studies of their transport to and accumulation in the Arctic environment. 42. A general design of caging-group-free photoactivatable fluorophores for live-cell nanoscopy

Abstract

The controlled switching of fluorophores between non-fluorescent and fluorescent states is central to every super-resolution fluorescence microscopy (nanoscopy) technique, and the exploration of radically new switching mechanisms remains critical to boosting the performance of established, as well as emerging super-resolution methods. Photoactivatable dyes offer substantial improvements to many of these techniques, but often rely on photolabile protecting groups that limit their applications. Here we describe a general method to transform 3,6-diaminoxanthones into caging-group-free photoactivatable fluorophores. These photoactivatable xanthones (PaX) assemble rapidly and cleanly into highly fluorescent, photo- and chemically stable pyronine dyes upon irradiation with light. The strategy is extendable to carbon- and silicon-bridged xanthone analogues, yielding a family of photoactivatable labels spanning much of the visible spectrum. Our results demonstrate the versatility and utility of PaX dyes in fixed and live-cell labelling for conventional microscopy, as well as the coordinate-stochastic and deterministic nanoscopies STED, PALM and MINFLUX.
