Little is known about the long-term effects of stillbirth on dairy cows. This study compared the behavior, serum and hair cortisol concentrations, production and quality of milk, and reproductive performance in primiparous dairy cows that delivered dead or live newborn calves. Forty-four Holstein cows housed in an earth-flooring corral (42.66 m2/cow) were studied for 90 days and classified into two groups: 29 cows with alive newborn calves (AC group) and 15 cows with dead newborn calves (DC group). On the first day of lactation and on the 30th, 60th, and 90th day, hair and serum cortisol concentrations were measured using radioimmunoassay as reliable stress indicators. Social, sexual, and maintenance behaviors were observed daily for 4 hours. Milk yield was measured three times a day in a milking parlor, and somatic cell counts were compared at 3 and 9 months of lactation. Milk quality and body condition were also recorded. Cows in the AC group spent more time lying, ruminating, and drinking than cows in the DC group, which received and exhibited significantly less aggression. No differences were observed between the groups in the proportion of time spent standing, walking, or eating. Serum and hair cortisol concentrations were significantly higher in DC cows than in AC cows on day 60 of lactation. No differences were observed in the body condition or reproductive performance. As expected, the milk quality was higher in AC cows than in DC cows. Our study shows that the delivery of dead newborn calves by primiparous cows has a clear negative effect on long-term behavior, hair and serum cortisol concentrations, and milk production. Introduction Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis (Mtb) that mainly affects the lungs. Along the course of pulmonary TB there are remarkable changes in the production of cytokines that cause endocrine changes. So far, it is not known the physiological and histological changes in the male reproductive system during pulmonary TB. Objective To investigate whether pulmonary TB produces histological alterations of the BALB/c mice reproductive organs, as well as abnormalities in spermatogenesis, serum testosterone concentrations and expression of testicular cytokines. Methods BALB/c mice were infected intratracheally with high dose Mtb strain H37Rv. Groups of six non infected and infected animals were euthanized on days 1, 3, 7, 14, 21, 28, 60, 90 and 120 post-infection. Bacillary loads were determined by counting colony forming units (CFUs) in lungs, testes, prostate and seminal vesicles. Histological sections were obtained from the same organs. Spermatozoids number and quality were assessed by spermatobioscopy. Serum testosterone concentrations were determined by radioimmunoanalysis (RIA) in control and infected mice in each time of sacrifice. Results Mtb only grew in lung tissue. Serum androgens showed a trend to decrease in the infected mice compared to the healthy animals, the difference turn into statistically significance at post infection day 120. The weight of the testis was not modified throughout the study, and no histopathological changes were found. However, we detected a significant decrease in the weight of the seminal vesicles and prostate starting at 28 days post-infection. Atrophy of the seminal vesicles and prostate epithelia were significant, beginning after 60 days of infection. Spermatobioscopy revealed hypospermia in the later stages of the disease. We have observed in the testes a local significant disbalance on the cytokine profile (increase of IL-6 and decrease of IL-10 and TGF-b levels) together with a very significant reduction of the body weight during late pulmonary TB. Conclusion Pulmonary TB affects the histophysiology of the male reproductive system due to hormonal changes, an imbalance of pro-inflammatory cytokine profile, and a wasting syndrome during late disease. Two jaguars were anesthetized with dexmedetomidine, ketamine, and isoflurane. Arterial blood samples analyzed shortly after darting revealed no abnormalities. Samples analyzed 2 and 1.5 hr after darting revealed moderate hyperkalemia in both animals (6.8 and 6.2 mEq/L, respectively). Shortly after hyperkalemia was recognized, one jaguar developed electrocardiographic abnormalities (sinoventricular rhythm and widened QRS complexes), and a few minutes later, suffered cardiopulmonary arrest. Resuscitation with chest compressions, intermittent positive-pressure ventilation, and epinephrine was successful, and autonomous breathing and circulation resumed within a few minutes. Anesthesia-related hyperkalemia has been reported in a variety of large felids but has not been reported previously in jaguars. In all reports, α-2 adrenergic agonists were used as part of the immobilization protocol. Due to the presumptively high incidence and mortality caused by this complication, frequent monitoring of electrolyte concentrations and prompt treatment of hyperkalemia is recommended when anesthetizing large felids, including jaguars. Tuberculosis (TB) caused by Mycobacterium tuberculosis mainly affects the lungs, but can spread to other organs. TB chronically activates the immune and endocrine systems producing remarkable functional changes.So far, it is unknown whether pulmonary non-disseminated TB cause changes in the female reproductive system and lung endocrinology. To investigate whether pulmonary TB produces immunoendocrine alterations of the female mice reproductive organs, and lung estradiol synthesis. BALB/c mice were infected intratracheally with Mycobacterium tuberculosis (Mtb) strain H37Rv. Groups of six non-infected and infected animals were euthanized on different days. Bacillary loads were determined in the lungs, ovaries and uterus. Immunohistochemistry and morphometry studies were performed in histological sections. Serum estradiol wasassayed, and supernatantfrom cultured lung cells was analyzed by Thin Layer Chromatography (TLC). Mtb only grew in lung tissue. Histopathology revealed abnormal folliculogenesis and decreased corpora lutea. Altered ovarian expression of IL-6, IL-1β was found. The infection increased serum estradiol. Estradiol synthesis by infected lung cells triplicate after 30 pi days.Aromatase immunostaining was found in the alveolar and bronchial epithelium, being stronger in the infected lungs, mainly in macrophages. Summary Background HIV-1 infection is associated with increased risk of tuberculosis and a safe and effective vaccine would assist control measures. We assessed the safety, immunogenicity, and efficacy of a candidate tuberculosis vaccine, modified vaccinia virus Ankara expressing antigen 85A (MVA85A), in adults infected with HIV-1. Methods We did a randomised, double-blind, placebo-controlled, phase 2 trial of MVA85A in adults infected with HIV-1, at two clinical sites, in Cape Town, South Africa and Dakar, Senegal. Eligible participants were aged 18–50 years, had no evidence of active tuberculosis, and had baseline CD4 counts greater than 350 cells per μL if they had never received antiretroviral therapy or greater than 300 cells per μL (and with undetectable viral load before randomisation) if they were receiving antiretroviral therapy; participants with latent tuberculosis infection were eligible if they had completed at least 5 months of isoniazid preventive therapy, unless they had completed treatment for tuberculosis disease within 3 years before randomisation. Participants were randomly assigned (1:1) in blocks of four by randomly generated sequence to receive two intradermal injections of either MVA85A or placebo. Randomisation was stratified by antiretroviral therapy status and study site. Participants, nurses, investigators, and laboratory staff were masked to group allocation. The second (booster) injection of MVA85A or placebo was given 6–12 months after the first vaccination. The primary study outcome was safety in all vaccinated participants (the safety analysis population). Safety was assessed throughout the trial as defined in the protocol. Secondary outcomes were immunogenicity and vaccine efficacy against Mycobacterium tuberculosis infection and disease, assessed in the per-protocol population. Immunogenicity was assessed in a subset of participants at day 7 and day 28 after the first and second vaccination, and M tuberculosis infection and disease were assessed at the end of the study. The trial is registered with ClinicalTrials.gov , number NCT01151189 . Findings Between Aug 4, 2011, and April 24, 2013, 650 participants were enrolled and randomly assigned; 649 were included in the safety analysis (324 in the MVA85A group and 325 in the placebo group) and 645 in the per-protocol analysis (320 and 325). 513 (71%) participants had CD4 counts greater than 300 cells per μL and were receiving antiretroviral therapy; 136 (21%) had CD4 counts above 350 cells per μL and had never received antiretroviral therapy. 277 (43%) had received isoniazid prophylaxis before enrolment. Solicited adverse events were more frequent in participants who received MVA85A (288 [89%]) than in those given placebo (235 [72%]). 34 serious adverse events were reported, 17 (5%) in each group. MVA85A induced a significant increase in antigen 85A-specific T-cell response, which peaked 7 days after both vaccinations and was primarily monofunctional. The number of participants with negative QuantiFERON-TB Gold In-Tube findings at baseline who converted to positive by the end of the study was 38 (20%) of 186 in the MVA85A group and 40 (23%) of 173 in the placebo group, for a vaccine efficacy of 11·7% (95% CI −41·3 to 44·9). In the per-protocol population, six (2%) cases of tuberculosis disease occurred in the MVA85A group and nine (3%) occurred in the placebo group, for a vaccine efficacy of 32·8% (95% CI −111·5 to 80·3). Interpretation MVA85A was well tolerated and immunogenic in adults infected with HIV-1. However, we detected no efficacy against M tuberculosis infection or disease, although the study was underpowered to detect an effect against disease. Potential reasons for the absence of detectable efficacy in this trial include insufficient induction of a vaccine-induced immune response or the wrong type of vaccine-induced immune response, or both. Abstract: Few studies have been carried out on the welfare of captive bottlenose dolphins (Tursiops truncatus). Of these, most include information on animals kept in closed facilities or pools. The aim of this study was to assess the welfare of captive bottlenose dolphins in open and closed facilities by measuring states of individual behavior and salivary cortisol concentrations. A total of 23 bottlenose dolphins were studied in 4 different dolphinaria. Dolphinaria A and B have closed facilities, whereas dolphinaria C and D have open facilities. A total of 152 hours of behavioral observations were analyzed using a combination of behavior and scan sampling to obtain information on individual time budgets. Salivary cortisol concentrations were measured using radioimmunoassay in 96 and 180 saliva samples of dolphins kept in closed and open facilities, respectively. In general, the results found that dolphins kept in open facilities spent less time floating (P < 0.05) and swimming in circular patterns than linear ones (P < 0.05) compared with dolphins in closed facilities. Dolphins kept in open facilities also had lower salivary cortisol concentrations than dolphins kept in closed facilities (P < 0.05). For this reason, we suggest that further research should include other welfare indicators such as reproductive function and health measurements to know more about the relationships between the design of pools and dolphin welfare. Summary: Objectives: A fundamental to global tuberculosis (TB) control is timely and accurate diagnosis of infectious cases of the disease. Among various methods, techniques based on nucleic acid amplification are the ones with promising prospects. The present study evaluates the diagnostic value of the recently developed IS6110-based loop-mediated isothermal amplification (LAMP) for detection of Mycobacterium tuberculosis complex (MTBC) in sputum specimens. Methods: In this cross-sectional study (2008–2009), IS6110-LAMP was evaluated on 101 sputum specimens from 93 highly suspected TB patients and compared to Amplicor MTB test and in-house IS6110-PCR and -nested PCR assays. Culture results or clinical recovery following anti-TB therapy was considered as a reference to prove the TB cases. Results: The overall sensitivity of IS6110-LAMP, Amplicor, nPCR, and PCR were respectively 89.6% (69/77 specimens; 95% confidence interval [CI], 80.5–95.4%), 76.6% (59/77 specimens; CI, 65.6–85.5%), 79.2% (61/77 specimens; CI, 68.5–87.6%) and 59.7% (46/77 specimens; CI, 47.9–70.8%). The specificity and positive predictive value (PPV) were 100% for all the tests, and the negative predictive value (NPV) of IS6110-LAMP, Amplicor, nPCR, and PCR were respectively 75%, 57.1%, 60%, and 43.6%. There was an excellent overall agreement between LAMP and nPCR (k 0.828), and between LAMP and Amplicor (k 0.746), in addition to a better tolerance of IS6110-LAMP to inhibitors present in clinical specimens. Conclusion: The better diagnostic performance of IS6110-LAMP compared to Amplicor (p = 0.009), nPCR (p = 0.013) and PCR (p < 0.0001) besides its rapidity, simplicity, and cost-effectiveness makes it a valuable method for the detection of MTBC in clinical samples, particularly in resource-limited settings. The article presents a study on the effects of adrenocotropic hormone (ACTH) challenge and age on hair cortisol concentrations in dairy cattle. The researchers compared cortisol concentrations in the hair of cows and calves to test whether cortisol accumulates in bovine hair after ACTH challenges and to determine any link between cortisol levels and hair color in Holstein cows. Assessing cortisol concentration in hair is said to be a useful non-invasive method for assessing stress in cattle.

Abstract: Urbanization generates human-made habitats with novel resources. Such change is not homegeneous, and includes different urban land uses characterized by specific habitat traits. In this study we explored the ecophysiologial response of an urban-exploiter species in three different urban land uses (urban, suburban, industrial). Our results show that corticosterone (obtained through fecal samples) and immunoglobulin (obtained through blood samples) concentrations did not differ significantly among the studied urban land uses. Corticosterone and immunoglobulin concentration showed both high and low values in urban and suburban areas, while industrial areas had low and high levels of corticosterone, and medium-to-low immunoglobulin concentration values. Also, we found a negative relationship between corticosterone and immunoglobulin concentrations in industrial areas. Based on corticosterone and immunoglobulin concentration thresholds established from previous studies, our results suggest that: (1) birds in the three studied urban conditions are both stressed and non-stressed; and (2) the immune system of industrial House Sparrows could not be responding successfully to pathogens. Not finding physiological relationships in urban and suburban areas underline that this species has a large capacity to respond to the different stress and immune challenges found in urban areas. However, our results suggest that the physiological condition of industrial House Sparrows could be compromised by differences in the frequency and intensity of the stressor agents faced by the birds in this land use category.
