The construction from scratch of synthetic cells by assembling molecular building blocks is unquestionably an ambitious goal from a scientific and technological point of view. To realize functional life-like systems, minimal enzymatic modules are required to sustain the processes underlying the out-of-equilibrium thermodynamic status hallmarking life, including the essential supply of energy in the form of electrons. The nicotinamide cofactors NAD(H) and NADP(H) are the main electron carriers fueling reductive redox reactions of the metabolic network of living cells. One way to ensure the continuous availability of reduced nicotinamide cofactors in a synthetic cell is to build a minimal enzymatic module that can oxidize an external electron donor and reduce NAD(P)+. In the diverse world of metabolism there is a plethora of potential electron donors and enzymes known from living organisms to provide reducing power to NAD(P)+ coenzymes. This perspective proposes guidelines to enable the reduction of nicotinamide cofactors enclosed in phospholipid vesicles, while avoiding high burdens of or cross-talk with other encapsulated metabolic modules. By determining key requirements, such as the feasibility of the reaction and transport of the electron donor into the cell-like compartment, we select a shortlist of potentially suitable electron donors. We review the most convenient proteins for the use of these reducing agents, highlighting their main biochemical and structural features. Noting that specificity toward either NAD(H) or NADP(H) imposes a limitation common to most of the analyzed enzymes, we discuss the need for specific enzymes─transhydrogenases─to overcome this potential bottleneck.

Biomolecules are frequently chiral compounds, existing in enantiomeric forms. Amino acids represent a meaningful example of chiral biological molecules. Both L- and D-amino acids play key roles in the biochemical structure and metabolic processes of living organisms, from bacteria to mammals. In this review, we explore the enantiospecific interaction between proteins and chiral amino acids, introducing theoretical models and describing the molecular basis of the ability of some of the most important enzymes involved in the metabolism of amino acids (i.e., amino acid oxidases, dehydrogenases, and aminotransferases) to discriminate the opposite enantiomers. Our analysis showcases the power of natural evolution in shaping biological processes. Accordingly, the importance of amino acids spurred nature to evolve strictly enantioselective enzymes both through divergent evolution, starting from a common ancestral protein, or through convergent evolution, starting from different scaffolds: intriguingly, the active sites of these enzymes are frequently related by a mirror symmetry.

Understanding the role of metal ions in biology can lead to the development of new catalysts for several industrially important transformations. Lanthanides are the most recent group of metal ions that have been shown to be important in biology, that is, in quinone‐dependent methanol dehydrogenases (MDH). Here we evaluate a literature‐known pyrroloquinoline quinone (PQQ) and 1‐aza‐15‐crown‐5 based ligand platform as scaffold for Ca2+, Ba2+, La3+ and Lu3+ biomimetics of MDH and we evaluate the importance of ligand design, charge, size, counterions and base for the alcohol oxidation reaction using NMR spectroscopy. In addition, we report a new straightforward synthetic route (3 steps instead of 11 and 33 % instead of 0.6 % yield) for biomimetic ligands based on PQQ. We show that when studying biomimetics for MDH, larger metal ions and those with lower charge in this case promote the dehydrogenation reaction more effectively and that this is likely an effect of the ligand design which must be considered when studying biomimetics. To gain more information on the structures and impact of counterions of the complexes, we performed collision induced dissociation (CID) experiments and observe that the nitrates are more tightly bound than the triflates. To resolve the structure of the complexes in the gas phase we combined DFT‐calculations and ion mobility measurements (IMS). Furthermore, we characterized the obtained complexes and reaction mixtures using Electron Paramagnetic Resonance (EPR) spectroscopy and show the presence of a small amount of quinone‐based radical., A pyrroloquinoline quinone (PQQ) based model system was used for the investigation of different factors that may have an impact on the oxidation reaction in the active sites of alcohol dehydrogenase enzymes. Using various metal salts, the influence of ion size and charge, as well as counterions was investigated. The study involves NMR and EPR analysis as well as advanced mass spectrometry techniques.

Reuse of leachates by recycling them for composting is part of the circular economy. However, directly returning compost leachates to the mixture might increase the concentration of contaminants in the stabilized mass. The application of the ultrasonic modification of leachate was aimed at increasing the activity of microorganisms and the availability of nutrients, and reducing the number of pathogenic species. The purpose of the work was to determine the impact of variable ultrasonic (time and amplitude) field parameters on the biochemical properties, and number of selected groups of microorganisms, of leachates from a composting process. The tests used short sonication times of 15, 30, 60, 90, and 120 s and vibration amplitudes of 15.25, 30.5, and 46 µm. The assessment was made on the basis of changes in numbers of microbial communities (mesophilic, thermophilic, Escherichia coli, Salmonella spp. and fungi) and enzymatic activity (dehydrogenases-DHA), as well as respiratory activity (AR). Based on the conducted research, it was found that the leachate sonication time of 60 s and amplitude 30.5 µm were the most effective. The above parameters were considered borderline, above which there were no significant differences in the values of the analyzed indicators.

Biocatalytic approaches have been investigated in order to isolate the enantiomers of Wieland-Miescher ketone (1) and of its alcoholic derivatives (cis-2 and trans-3). Specifically, two enzymes from our in-house metagenomic collection of oxidoreductases, IS2-SDR and Dm7?-HSDH, catalyzed the kinetic resolution of the starting racemic ketone 1 or its complete conversion into two diastereomeric products, respectively. Moreover, the kinetic resolution of the racemic cis-alcohol (2) was very efficiently obtained (E?2.000) by lipase PS catalyzed acetylation in dry acetone. All the products were isolated with ee>=95 %. Simple chemical elaborations of some of them allowed to isolate the missing enantiomers.

This study aimed to evaluate the fertilizing value of rye stillage used in the cultivation of winter triticale cv. ‘Grenado’. The research was performed in 2018 (autumn, before the application of the stillage), 2019, and 2020 (spring and autumn after stillage application) on Luvisoil at the depth levels of 0–20 and 20–40 cm. Each year, the basic soil parameters were analyzed, i.e., pH in 1M KCl, organic carbon (Corg), available phosphorus (P), potassium (K), magnesium (Mg), zinc (Zn), and copper (Cu). Enzymes were also accounted: alkaline phosphatase (AlP), acid phosphatase (AcP), and dehydrogenases (DEH). The use of stillage resulted in a significant increase in the content of P, K, and Mg and the activity of AlP, AcP, and DEH in the soil. It significantly increased the Corg content and did not have a significant effect on pH. The obtained results indicate that the response of the enzymatic activity to the distillery stillage depended on both the sampling season soil and the depth. However, it is necessary to systematically monitor the pH of the soil and at the same time to rationally apply mineral fertilization.

Oxygen diffusion into the soil ecosystem is imperative for the sustainability of life in the soil. This study evaluated selected microbial exudates as indices of soil respiration in a crude oil polluted soil ecosystem ex-situ using biochemical and physicochemical tools to determine the following parameters; total petroleum hydrocarbon (TPH), dehydrogenases, catalase, ATP, and pH. The result of the study at 1.5 – 3.5% contamination across days-zero to -28 following crude oil impact presented an augmented increase in TPH from 0.02 ± 0.00 to 0.10 ± 0.00 with an increase in the activity of soil dehydrogenases from 4.90 ± 0.01 to 8.80 ± 0.04 katal which was significant (p0.05). Conversely, the activity of soil catalase suffered inhibition from 0.20 ± 0.00 to 0.11 ± 0.00 katal. ATP synthesis decreased from control to 0.24 ± 0.00mg/100g on week one and increased to 0.60 ± 0.10mg/100mg significantly (p0.05) on week four with contamination as the pH was reduced from 5.8 ± 0.00 to 4.1 ± 0.00 significantly, thus creating hypoxia and a reducing environment, a demonstration of soil respiration.

The study objective was to determine the relationship of selected enzyme activities with carbon sequestration and N, P, K, Mg, Zn and Cu contents in Phaeozem soils. Soil samples were taken from a 10 ha area. A selection of their physical and chemical properties and the contents of the available forms of selected macro- and microelements were determined. The activities of dehydrogenases (DEH), catalase (CAT), peroxidases (PER), alkaline (AlP) and acid (AcP) phosphatase, β-glucosidase (BG) and proteases (PR) were also determined. The relationship between enzymatic soil fertility indices (AlP/AcP, BIF, GMea, TEI, BA12 and BA13) and selected soil parameters was also determined. The research used principal component analysis (PCA) to distinguish significantly correlated parameters of a Phaeozem used for agricultural purposes. The study area showed low TOC and K contents and average P and Mg contents. Significant positive correlations were found between the TOC content and activity of the tested enzymes, evidencing that soil enzymes are an important parameter in carbon sequestration and soil nutrient dynamics.

The aim of the study was to assess the quality of agricultural soils, which accumulate additional amounts of heavy metals from fertilization and modify their bioavailability, with the use of interdependencies between their biological and chemical properties conditioned by the distance from communication routes. Our results indicated that heavy metals had an impact on enzyme activity in soils and their accumulation was significantly related to the distance from the edge of the road, location of sampling sites, date of soil sampling, and years of research. It was found that the greatest amounts of zinc, cadmium, lead, and copper were accumulated at a distance of 5–20 m from the edge of the road. The highest enrichment factor and geoaccumulation index values were recorded for Pb, followed by Zn, Cu, and Cd. Principal component analysis and regression models showed that the activities of phosphatase and dehydrogenases seem to be the best bioindicators of contamination of roadside soils used for agricultural purposes. Since their activity is related to soil’s organic carbon content, inputs of organic fertilizers and crop residues should be ensured in the agroecosystems along roadsides.

The content of sulphur and copper (Cu), iron (Fe), manganese (Mn), zinc (Zn) and of assimilation pigments in the needles of Scots pine (Pinus sylvestris L.) in the forests of Bydgoszcz, Poland was determined. The content of those metals and the activity of dehydrogenases (DHA) in the rhizosphere of the trees was assayed. The average total sulphur (TS) content in 2-year-old pine needles was 832.4 mg kg−1 d.w. No significant correlation was found between TS and Cu, Fe, Mn and Zn in needles and the content of assimilation pigments indicating no phytotoxic effect of sulphur dioxide (SO2) and metals on Scots pine. The content of metals in the needles pointed to an inconsiderable degree of human impact. The soils in the surface layer were not contaminated with heavy metals. With the principal component analysis (PCA) two principal components were identified which accounted for 68% of the total change in variation. The variables that determined the principal components were the soil content of organic carbon (TOC), total nitrogen (TN), TS and sulphates (SO42−), the soil content of Mn, Zn, available forms to plants of Cu, Mn, and the content of Cu, Fe in needles.

This study demonstrates the indirect effects of non-thermal ambient air plasmas (NTP) on seed germination and plant growth. It investigates the effect of plasma-activated water (PAW) on 3-day-old seedlings of two important farm plants—barley and pea. Applying different types of PAW on pea seedlings exhibited stimulation of amylase activity and had no inhibition of seed germination, total protein concentration or protease activity. Moreover, PAW caused no or only moderate oxidative stress that was in most cases effectively alleviated by antioxidant enzymes and proved by in situ visualization of H2O2 and ˙O2−. In pea seedlings, we observed a faster turn-over from anaerobic to aerobic metabolism proved by inhibition of alcohol dehydrogenase (ADH) activity. Additionally, reactive oxygen/nitrogen species contained in PAW did not affect the DNA integrity. On the other hand, the high level of DNA damage in barley together with the reduced root and shoot length and amylase activity was attributed to the oxidative stress caused by PAW, which was exhibited by the enhanced activity of guaiacol peroxidase or ADH. Our results show the glow discharge PAW at 1 min activation time as the most promising for pea. However, determining the beneficial type of PAW for barley requires further investigation.

The aim of the study was to examine the features of phenotype and the levels of NAD(P)-dependent dehydrogenases activity of blood neutrophils in the prognosis of abdominal sepsis development in patients with widespread purulent peritonitis (WPP). 50 patients with WPP of community and hospital origin in the pre-operative period were examined. From the 5th to the 10th day of the postoperative period 35 patients (70%) had developed abdominal sepsis, 15 patients (30%) had absence of complications. As controls 67 respect healthy people were examined. The research blood neutrophils phenotype was performed by f low cytometry using a direct immunofluorescence whole peripheral blood. The levels of surface receptor expression was assessed by the mean fluorescence intensity. The NAD(P)-dependent dehydrogenases activity in the blood neutrophils studied using bioluminescence method. It was established that the inflammatory reaction in patients with WPP is characterized by neutrophilia and changes in the phenotype of blood neutrophils. The markers of the subsequent development of sepsis in WPP are less pronounced (in comparison with the indices of uncomplicated patients), an increase in the number of neutrophils, a decrease in the HLA-DR + -cell count against the background of a high level of neutrophils expressing a high affinity receptor for IgG. The patients without subsequent complications had the number of neutrophils with CD23 receptor expression is increased. Metabolism of neutrophils in patients with WPP is characterized by a decrease in the intensity of plastic processes due to low activity of glucose-6-phosphate dehydrogenase and an imbalance in the activity of the enzymes of the mitochondrial compartment. A feature of the neutrophil metabolism in patients with WPP without subsequent development of sepsis is a high activity of anaerobic lactate dehydrogenase reaction and a decrease in the activity of NADP-dependent decarboxylating malate dehydrogenase. The patients with WPP with subsequent development of sepsis had a high level of NAD-dependent out flow of substrates from the tricarboxylic acid cycle on the amino acid exchange reaction via glutamate dehydrogenase which can affect the activity of aerobic respiration of blood neutrophils. The established differences in the phenotype and activity of enzymes in the blood neutrophils in patients with WPP depending on the subsequent development of sepsis determine the possibility of creating a method for predicting the development of complications and developing immunoactive therapy in the postoperative period of WPP.

This work reports the study of ZnO-based anodes for the photoelectrochemical regeneration of the oxidized form of nicotinamide adenine dinucleotide (NAD+). The latter is the most important coenzyme for dehydrogenases. However, the high costs of NAD+ limit the use of such enzymes at the industrial level. The influence of the ZnO morphologies (flower-like, porous film, and nanowires), showing different surface area and crystallinity, was studied. The detection of diluted solutions (0.1 mM) of the reduced form of the coenzyme (NADH) was accomplished by the flower-like and the porous films, whereas concentrations greater than 20 mM were needed for the detection of NADH with nanowire-shaped ZnO-based electrodes. The photocatalytic activity of ZnO was reduced at increasing concentrations of NAD+ because part of the ultraviolet irradiation was absorbed by the coenzyme, reducing the photons available for the ZnO material. The higher electrochemical surface area of the flower-like film makes it suitable for the regeneration reaction. The illumination of the electrodes led to a significant increase on the NAD+ regeneration with respect to both the electrochemical oxidation in dark and the only photochemical reaction. The tests with formate dehydrogenase demonstrated that 94% of the regenerated NAD+ was enzymatically active.

In four study plots located in silver fir and Scots pine stands in Magura National Park (southeastern Poland), the relationships between the occurrence and biomass of epigeic carabids, the volume of deadwood and soil biochemical properties were investigated. Thirteen carabid beetle species from the genera Abax, Carabus, Molops and Pterostichus were captured. Rare epigeic carabid species in the fauna of Poland and Europe, such as Carabus glabratus (Paykull), Carabus sylvestris (Panzer) and Abax schueppeli (Germar), were recorded. The number of carabid individuals and species captured as well as the mean individual biomass index at different elevations and in forests of different tree compositions differed significantly. There were no correlations between deadwood volume, carabid abundance and the mean individual biomass of the carabid beetles. The mean individual Carabidae biomass increased with elevated pH, soil carbon content, soil dehydrogenase activity and the number of stumps.

The development of sustainable processes for the valorization of byproducts and other waste streams remains an ongoing challenge in the field of catalysis. Racemic borneol, isoborneol and camphor are currently produced from a‐pinene, a side product from the production of cellulose. The pure enantiomers of these monoterpenoids have numerous applications in cosmetics and act as reagents for asymmetric synthesis, making an enzymatic route for their separation into optically pure enantiomers a desirable goal. Known short‐chain borneol‐type dehydrogenases (BDHs) from plants and bacteria lack the required specificity, stability or activity for industrial utilization. Prompted by reports on the presence of pure (−)‐borneol and (−)‐camphor in essential oils from rosemary, we set out to investigate dehydrogenases from the genus Salvia and discovered a dehydrogenase with high specificity (E>120) and high specific activity (>0.02 U mg−1) for borneol and isoborneol. Compared to other specific dehydrogenases, the one reported here shows remarkably higher stability, which was exploited to obtain the first three‐dimensional structure of an enantiospecific borneol‐type short‐chain dehydrogenase. This, together with docking studies, led to the identification of a hydrophobic pocket in the enzyme that plays a crucial role in the stereo discrimination of bornane‐type monoterpenoids. The kinetic resolution of borneol and isoborneol can be easily integrated into the existing synthetic route from a‐pinene to camphor thereby allowing the facile synthesis of optically pure monoterpenols from an abundant renewable source. Highly Specific Borneol‐Type Dehydrogenase: A borneol‐ type dehydrogenase with high specificity for borneol and isoborneol was characterized from the genome of Salvia rosmarinus. The crystallographic structure was determined, showing an hydrophobic pocket that plays a crucial role in the stereo discrimination of bornane‐type monoterpenoids.

One of the challenges in biocatalysis is the development of stable and efficient bi-enzymatic cascades for bio-redox reactions coupled to the recycling of soluble cofactors. Aldo-keto reductase (LEK) and glucose dehydrogenase (GDH) can be utilized as the NADPH recycling system for economic and efficient biocatalysis of (R)-4-chloro-3-hydroxybutanoate ((R)-CHBE), an important chiral pharmaceutical intermediate. The LEK and GDH was efficiently co-immobilized in mesocellular siliceous foams (MCFs) under microwave irradiation (CoLG-MIA). while they were also co-immobilized by entrapment in calcium alginate without MIA as control (CoLG-CA). The relative activity of CoLG-MIA was increased to 140% compared with that of free LEK. The CoLG-MIA exhibited a wider range of pH and temperature stabilities compared with other preparations. The thermal, storage and batch operational stabilities of microwave-assisted immobilized LEK-GDH were also improved. The NADPH recycling system exhibited the potential as the stable and efficient catalyst for the industrial preparation of (R)-CHBE.

Gotovo svaki industrijski proces proizvodnje uključuje primjenu organskih otapala čija je uporaba odgovorna za onečišćenje okoliša te nepovoljna za zdravlje radnika i sigurnost procesa zbog svojstva toksičnosti, lake hlapljivosti, zapaljivosti i eksplozivnosti. Stoga, kako bi se ispunili zahtjevi zelene kemije za ekološkom prihvatljivošću i održivošću, traže se alternativna otapala. Eutektička otapala istaknula su se svojstvima poput niske hlapljivosti, netoksičnosti, nezapaljivosti, biorazgradivosti te jednostavnim metodama priprave. Nadalje, u polju biokatalize dehidrogenaze pokazuju veliki potencijal za dobivanje visoko vrijednih kiralnih spojeva koji se koriste u proizvodnji optički čistih lijekova. Cilj ovog rada bio je istražiti mogućnost primjene prirodnih eutektičkih otapala na bazi kolin-klorida kao medija za reakcije redukcije katalizirane dehidrogenazama. U pripravljenim otapalima određena je aktivnost alkohol dehidrogenaze te promjene u konformaciji iste. U istim otapalima također je ispitana stabilnost pripadajućih koenzima NADH/NAD+. Blago lužnata prirodna eutektička otapala koja su sadržavala etilen-glikol i ureu pokazala su stabilizirajući utjecaj na oba koenzima, no niti u jednom otapalu alkohol dehidrogenaza nije pokazala zadovoljavajuću aktivnost uz značajne promjene u konformaciji u odnosu na pufer kao referentno otapalo. Almost every industrial production process involves the use of organic solvents whose use is responsible for environmental pollution and unfavorable to workers' health and process safety, due to the properties of toxicity, volatility, flammability and explosiveness. Therefore, in order to meet the requirements of green chemistry for environmental safety and sustainability, alternative solvents are sought. Eutectic solvents stood out with properties such as low volatility, non-toxicity, non-flammability, biodegradability and simple preparation methods. Furthermore, in the field of biocatalysis, dehydrogenases show great potential for obtaining high-value chiral compounds used in the production of optically pure drugs. The aim of this study was to investigate the possibility of using natural eutectic solvents based on choline chloride as a medium for dehydrogenase-catalyzed reduction reactions. The activity of alcohol dehydrogenase and changes in its conformation was determined in the prepared solvents. The stability of the corresponding NADH/NAD+ coenzymes was also tested in the same solvents. Slightly alkaline natural eutectic solvents containing ethylene glycol and urea showed a stabilizing effect on both coenzymes, but in neither solvent did alcohol dehydrogenase show satisfactory activity with significant changes in conformation relative to buffer as reference solvent.

Zahvaljujući sposobnosti kataliziranja redukcija karbonilnih grupa u prokiralnim spojevima do kiralnih alkohola, koji su važni međuprodukti u proizvodnji lijekova i kemikalija, dehidrogenaze su enzimi od velike industrijske važnosti. U novije vrijeme cilj je industrijsku proizvodnju uskladiti s načelima zelene kemije, stoga se velike količine hlapljivih, zapaljivih i toksičnih organskih otapala nastoje zamijeniti sigurnijim otapalima za ljudsko zdravlje i okoliš. Nova generacija takvih otapala su eutektička otapala koje karakteriziraju niska hlapljivost, biorazgradivost i jednostavna sinteza. Cilj ovog rada bio je ispitati mogućnosti primjene eutektičkih otapala u reakcijama kataliziranim alkohol dehidrogenazom. Pripravljena su različita eutektička otapala, s kolin-kloridom u sastavu, u kojima je ispitana aktivnost te su praćene promjene u konformaciji enzima alkohol dehidrogenaze. U istim je otapalima također praćena stabilnost koenzima NADH i NAD+ koji su potrebni za aktivnost alkohol dehidrogenaze. Eutektička otapala na bazi kolin-klorida s ureom u sastavu najbolje su stabilizirala oba oblika nikotinamidnog koenzima, no niti jedno eutektičko otapalo nije se pokazalo kao povoljan medij za odvijanje enzimske reakcije alkohol dehidrogenaze. Due to their ability to catalyze the reduction of carbonyl groups in prochiral compounds to chiral alcohols, which are important intermediates in the production of pharmaceuticals and chemicals, dehydrogenases are enzymes of great industrial importance. Recently, a tendency is to coordinate industrial production with the principles of green chemistry, so large amounts of volatile, flammable and toxic organic solvents are being replaced by solvents which are safer for human health and the environment. A new generation of such solvents is deep eutectic solvents characterized by low volatility, biodegradability, and easy synthesis. The aim of this study was to investigate the possibilities of using deep eutectic solvents in alcohol dehydrogenase-catalyzed reactions. Various choline-chloride based deep eutectic solvents were prepared in which enzyme activity was measured and changes in the conformation of the enzyme alcohol dehydrogenase were monitored. In the same solvents, the stability of the coenzymes NADH and NAD+ required for alcohol dehydrogenase activity was also monitored. Choline-chloride based deep eutectic solvents containing urea best stabilized both forms of nicotinamide coenzyme, but neither deep eutectic solvent proved to be a suitable medium for the enzymatic reaction of alcohol dehydrogenase.

Cadmium (Cd) is a toxic metal occurring in the environment naturally. Almond mushroom (Agaricus brasiliensis) is a well-known cultivated edible and medicinal mushroom. In the past few decades, Cd accumulation in A.brasiliensis has received increasing attention. However, the molecular mechanisms of Cd-accumulation in A. brasiliensis are still unclear. In this paper, a comparative transcriptome of two A.brasiliensis strains with contrasting Cd accumulation and tolerance was performed to identify Cd-responsive genes possibly responsible for low Cd-accumulation and high Cd-tolerance. Using low Cd-accumulating and Cd-tolerant (J77) and high Cd-accumulating and Cd-sensitive (J1) A.brasiliensis strains, we investigated 0, 2 and 5 mg L-1 Cd-effects on mycelium growth, Cd-accumulation and transcriptome revealed by RNA-Seq. A total of 57,884 unigenes were obtained. Far less Cd-responsive genes were identified in J77 mycelia than those in J1 mycelia (e.g., ABC transporters, ZIP Zn transporter, Glutathione S-transferase and Cation efflux (CE) family). The higher Cd-accumulation in J1 mycelia might be due to Cd-induced upregulation of ZIP Zn transporter. Cd impaired cell wall, cell cycle, DNA replication and repair, thus decreasing J1 mycelium growth. Cd-stimulated production of sulfur-containing compounds, polysaccharides, organic acids, trehalose, ATP and NADPH, and sequestration of Cd might be adaptive responses of J1 mycelia to the increased Cd-accumulation. DNA replication and repair had better stability under 2 mg L-1 Cd, but greater positive modifications under 5 mg L-1 Cd. Better stability of DNA replication and repair, better cell wall and cell cycle stability might account for the higher Cd-tolerance of J77 mycelia. Our findings provide a comprehensive set of DEGs influenced by Cd stress; and shed light on molecular mechanism of A.brasiliensis Cd accumulation and Cd tolerance.

Pseudomonas aeruginosais a ubiquitous opportunistic pathogen which relies on a highly adaptable metabolism to achieve broad pathogenesis. In one example of this flexibility, to catalyze the NADH:quinone oxidoreductase step of the respiratory chain,P.aeruginosahas three different enzymes: NUO, NQR and NDH2, all of which carry out the same redox function but have different energy conservation and ion transport properties. In order to better understand the roles of these enzymes, we constructed two series of mutants: (i) three single deletion mutants, each of which lacks one NADH dehydrogenase and (ii) three double deletion mutants, each of which retains only one of the three enzymes. All of the mutants grew approximately as well as wild type, when tested in rich and minimal medium and in a range of pH and [Na+] conditions, except that the strain with only NUO (ΔnqrFΔndh) has an extended lag phase. During exponential phase, the NADH dehydrogenases contribute to total wild-type activity in the following order: NQR > NDH2 > NUO. Some mutants, including the strain without NQR (ΔnqrF) had increased biofilm formation, pyocyanin production, and killed more efficiently in both macrophage and mouse infection models. Consistent with this, ΔnqrFshowed increased transcription of genes involved in pyocyanin production.

Anxiety&ndash, depressive disorders (ADD) are a risk factor of cardiovascular mortality in patients with coronary artery disease (CAD). Acute coronary syndrome (ACS) is the main clinical manifestation of a progressing CAD. Metabolic processes disorder in platelets can be one of the causes of cardiovascular complications in patients with ACS and concomitant ADD. We studied platelets metabolism and prognostic informativity of NAD(P)-dependent dehydrogenases of platelets in ACS patients with ADD in terms of forecasting cardiovascular complications development over a year of observation. The levels of NAD- and NADP-dependent dehydrogenases of platelets were determined by means of a bioluminescent method during the first 24 h after admission to hospital and in dynamics in 10 days. Among 315 examined patients, ADD was found in 161 (51.1%). ACS patients with concomitant ADD had both cytoplasmic and mitochondrial processes impairment in platelets that consisted in a decrease of energy metabolism intensity, inhibition of anaerobic glycolysis reactions and lipid catabolism. After 12 months of follow-up, 41 (25.5%) cardiovascular complications were detected in the group of ACS patients with ADD and 20 (13.0%) in the group of ACS patients without ADD. According to the results of the analysis of the neural network based on NAD(P)-dependent dehydrogenases of platelets activity in ACS patients with ADD, indicators were obtained that are informative for predicting the development of recurrent cardiovascular complications.

The biological role of REEs has long been underestimated, and research has mainly focused on methanotrophic and methylotrophic bacteria. We have recently demonstrated that P. putida, a plant growth-promoting bacterium that thrives in the rhizosphere of various food crops, possesses a REE-dependent alcohol dehydrogenase (PedH), but knowledge about REE-specific effects on physiological traits in nonmethylotrophic bacteria is still scarce. This study demonstrates that the cellular response of P. putida to lanthanum (La3+) is mostly substrate specific and that La3+ availability highly affects the growth of cells on glycerol. Further, a novel route for glycerol metabolism is identified, which is initiated by PedE and/or PedH activity and provides a growth advantage to this biotechnologically relevant organism by allowing a faster onset of growth. Overall, these findings demonstrate that lanthanides can affect physiological traits in nonmethylotrophic bacteria and might influence their competitiveness in various environmental niches., Ever since the discovery of the first rare earth element (REE)-dependent enzyme, the physiological role of lanthanides has become an emerging field of research due to the environmental implications and biotechnological opportunities. In Pseudomonas putida KT2440, the two pyrroloquinoline quinone-dependent alcohol dehydrogenases (PQQ-ADHs) PedE and PedH are inversely regulated in response to REE availability. This transcriptional switch is orchestrated by a complex regulatory network that includes the PedR2/PedS2 two-component system and is important for efficient growth on several alcoholic volatiles. To study whether cellular responses beyond the REE switch exist, the differential proteomic responses that occur during growth on various model carbon sources were analyzed. Apart from the Ca2+-dependent enzyme PedE, the differential abundances of most identified proteins were conditional. During growth on glycerol—and concomitant with the proteomic changes—lanthanum (La3+) availability affected different growth parameters, including the onset of logarithmic growth and final optical densities. Studies with mutant strains revealed a novel metabolic route for glycerol utilization, initiated by PedE and/or PedH activity. Upon oxidation to glycerate via glyceraldehyde, phosphorylation by the glycerate kinase GarK most likely yields glycerate-2-phosphate, which is eventually channeled into the central metabolism of the cell. This new route functions in parallel with the main degradation pathway encoded by the glpFKRD operon and provides a growth advantage to the cells by allowing an earlier onset of growth with glycerol as the sole source of carbon and energy.

As a ubiquitous enzyme, succinic semialdehyde dehydrogenase contributes significantly in many pathways including the tricarboxylic acid cycle and other metabolic processes such as detoxifying the accumulated succinic semialdehyde and surviving in nutrient-limiting conditions. Here the cce4228 gene encoding succinic semialdehyde dehydrogenase from Cyanothece sp. ATCC51142 was cloned and the homogenous recombinant cce4228 protein was obtained by Ni-NTA affinity chromatography. Biochemical characterization revealed that cce4228 protein displayed optimal activity at presence of metal ions in basic condition. Although the binding affinity of cce4228 protein with NAD+ was about 50-fold lower than that of cce4228 with NADP+, the catalytic efficiency of cce4228 protein towards succinic semialdehyde with saturated concentration of NADP+ is same as that with saturated concentration of NAD+ as its cofactors. Meanwhile, the catalytic activity of cce4228 was competitively inhibited by succinic semialdehyde substrate. Kinetic and structural analysis demonstrated that the conserved Cys262 and Glu228 residues were crucial for the catalytic activity of cce4228 protein and the Ser157 and Lys154 residues were determinants of cofactor preference.

BACKGROUND: Approximately 20–40% of patients diagnosed with breast cancer will experience a recurrence up to 20 years after their original diagnosis. 17?-hydroxysteroid dehydrogenase 1 and 2 (HSD17B1 and HSD17B2, respectively) regulate intratumoral concentrations of oestradiol, which promotes growth and proliferation of hormone dependent tumours. Breast carcinomas with increased HSD17B1, without a corresponding increase in HSD17B2, expression may become resistant to tamoxifen therapy by producing locally higher concentrations of oestradiol, which compete with tamoxifen and its metabolites for binding to the oestrogen receptor (ER). MATERIALS AND METHODS: In this population-based case-control study, we included women diagnosed with stage I–III breast cancer between 1985 and 2001, aged 35–69 years, registered in the Danish Breast Cancer Group. We identified 541 cases of breast cancer recurrence among women with ER positive disease who were treated with tamoxifen for at least 1 year (ER+TAM+). We also enrolled 300 breast cancer recurrence cases among women with ER negative disease, not treated with tamoxifen, who survived at least 1 year (ER?/TAM?). Controls were recurrence-free breast cancer patients at the time of case diagnosis, matched to recurrence cases on ER/TAM status, date of surgery, menopausal status, stage, and county or residence. Expression of HSD17B1 and HSD17B2 were measured by immunohistochemistry on tissue microarrays. We fit logistic regression models to compute odds ratios (ORs) and 95% confidence intervals (CIs) associating the HSD17B1:HSD17B2 ratio (>1 vs. ?1)—and each enzyme’s independent expression—with recurrence. RESULTS: We found no association between the HSD17B1:HSD17B2 ratio and breast cancer recurrence in either ER/TAM stratum (ER+/TAM+: OR=1.03, 95% CI: 0.78, 1.40; ER?/TAM?: OR=1.02, 95% CI: 0.77, 1.19). Associations for expression of each individual enzyme were also near null. CONCLUSION: The ratio of HSD17B1 expression to HSD17B2 expression was not associated with breast cancer recurrence in this study.

Regarding to the increase in population and industry development, global energy demand tends to increase continuously. However, our main source of energy, the fossil fuels, like coal, oil and natural gas, are limited and start running out. Moreover, the combustion of such fossil-based energy carriers gives carbon dioxide (CO2) back to the atmosphere which is considered as the major cause of the rising of global temperature. Therefore, renewable, cleaner and more sustainable energy sources are urgently needed. Converting inexhaustible energy (such as solar and wind energy) into chemicals or artificial fuels offers transportable fuels which are independent from place and time. One promising approach is to convert exhausted CO2 into fuels, for example, formate, methane and methanol using renewable energies like solar and wind. Developed from such idea, this study is aimed to investigate electrochemical systems for the conversion of CO2 to value-added chemicals products by using bio-origin catalysts such as microorganisms and enzymes. Herein, microbial electrolysis cells containing Methylobacterium extorquens, acetogenic bacteria, were developed and their catalytic activities towards the conversion of CO2 to formate were monitored under applying of a constant potential. In this part, direct electron injection and mediator-assisted approaches were investigated. Neutral red, a biocompatible dye, was introduced into the systems as redox mediator in two ways: homogeneous (dissolved in the solution) and heterogeneous (electropolymerized onto the electrode). Electrochemical characterization of such systems was carried out by means of cyclic voltammetry and electrolysis showing electrocatalytic activities towards the CO2 reduction to formate. The performance was found to be enhanced with the aid of mediator in both systems. Further investigation was performed by combining two bioelectrodes, bioanode and biocathode, which were developed from sewage sludge containing mixed culture microorganism, together in bio-electrochemical systems. The systems were developed serving as a wastewater treatment system where organic substances were oxidized at bioanode and CO2 was reduced to methane (CH4) at biocathode. Such system was successfully built up and system performance was investigated by monitoring chemical oxygen demand values and CH4 amount. Additionally, electrode modifications for both electrodes using poly(neutral red) and chitosan approaches were carried out. The results revealed improved overall performance in both organics degradation and CH4 production. In addition, dehydrogenase enzymes were investigated as electrocatalysts in electrochemical catalytic system. The dehydrogenases are redox enzymes that catalyze redox reactions and require sacrificial electron donor such as nicotinamide adenine dinucleotide, reduced form (NADH). Avoiding using such costly cofactor, electrochemical injection of electrons from electrode directly into enzymes active sites is suggested. Efficient immobilization of enzymes onto the electrode was presented by chemical grafting of conjugated dehydrogenases directly onto graphene (as a conductive nanoplatform). Using an alginate hydrogel matrix, we subsequently coated this bio-nano-electrocatalyst onto the electrode. This immobilization technique was employed for single dehydrogenase for the reduction of acetaldehyde to ethanol and furthermore, of three dehydrogenases for the cascade reduction of CO2 all the way to methanol. The enzyme-graphene bio-nano-electrocatalysts show enhanced reductive current delivered into the system, leading to higher production rate. Fungal pathogens including Verticillium longisporum which causes verticillium wilt are among the serious diseases of crops that easily spread worldwide. One of the ways to prevent fungal infection is to use fungal biocontrol agents applied as a seed coating. This control agent, the non-pathogenic oomycete Pythium oligandrum, enters the soil together with the seed and acts symbiotically in the plant's root system. On the one hand, it stimulates the plant's defence mechanism by secreting elicitors, and on the other hand, by providing tryptamine, it stimulates growth and increases the plant's fitness with auxin. In this work we studied the effect of seed treatment of Solanum lycopersicum L. cv. Micro-Tom with three different isolates of the genus Pythium (including the commercially used isolate M1 and two yet unused isolates X42 and X48) on the activities of antioxidant and NADP(H)-dependent enzymes in the leaves of plants infected with fungal pathogen V. longisporum. Two weeks after pathogen inoculation, no significant difference was found in the studied enzymes except for increased glucose-6phosphate dehydrogenase and NADP-malate dehydrogenase (oxaloacetate decarboxylation) activity in plants treated with X42 isolate and increased shikimate dehydrogenase activity and antioxidant capacity in plants...

Species-specific sex pheromones play key roles in moth sexual communication. Although the general pathway of Type-I sex pheromone biosynthesis is well established, only a handful of genes encoding enzymes involved in this pathway have been characterized. Streltzoviella insularis is a destructive wood-boring pest of many street trees in China, and the female sex pheromone of this species comprises a blend of (Z)-3-tetradecenyl acetate, (E)-3-tetradecenyl acetate, and (Z)-5-dodecenyl acetate. This organism therefore provides an excellent model for research on the diversity of genes and molecular mechanisms involved in pheromone production. Herein, we assembled the pheromone gland transcriptome of S. insularis by next-generation sequencing and identified 74 genes encoding candidate key enzymes involved in the fatty acid biosynthesis, β-oxidation, and functional group modification. In addition, tissue expression patterns further showed that an acetyl-CoA carboxylase and two desaturases were highly expressed in the pheromone glands compared with the other tissues, indicating possible roles in S. insularis sex pheromone biosynthesis. Finally, we proposed putative S. insularis biosynthetic pathways for sex pheromone components and highlighted candidate genes. Our findings lay a solid foundation for understanding the molecular mechanisms underpinning S. insularis sex pheromone biosynthesis, and provide potential targets for disrupting chemical communication that could assist the development of novel pest control methods.

[cat] Aquesta tesi doctoral està enfocada a la investigació de nous agents amb potencial activitat antitumoral per inhibició enzimàtica. S’ha descrit la implicació de la G6PDH en el desenvolupament i progressió del càncer degut a la seva participació en la síntesi d’àcids nucleics. Es busca la preparació de compostos que actuïn inhibint de forma més selectiva les cèl·lules canceroses, actuant sobre els enzims implicats en el seu metabolisme, com és la G6PDH, per tal de millorar l’eficàcia i reduir els efectes adversos dels tractaments dels que disposem actualment. S’ha dut a terme la preparació sintètica de tres series de compostos diferents. La primera pertany a les arilmetilenindolinona i derivats, la segona consta de sistemes tiofen-condensats, dissenyats a partir de modelització i amb canvis estructurals a partir de farmacomodulacions modulatives i finalment la tercera sèrie, la corresponent a les urees. Els compostos s’han preparat mitjançant reaccions clàssiques de química orgànica i s’han purificat posteriorment mitjançant tècniques de separació (cromatografia). L’elucidació estructural corresponent s’ha dut a terme mitjançant tècniques de Ressonància Magnètica Nuclear (RMN), Espectroscòpia de masses (EM) i d’Infraroig (IR). Per tal d’estudiar l’activitat inhibitòria enzimàtica, s’han realitzat assajos d’inhibició directa de la G6PDH, mesurada a partir de la producció d’NADPH., [eng] This PhD has been focused on the discovery of new compounds with antitumoral potential activity for inhibition of enzymes, like G6PDH. The implications of G6PDH for the development and progression of cancer due to its participation in nucleic acid synthesis have been described. Our objective was to prepare compounds that can be able to inhibit enzymes such as G6PDH that involved in metabolism of cancer cells with a good level of selectivity which have a perfect efficacy without significant side effects. Synthetic preparation of three different compounds has been carried out. The first one has arilmetilenindolinone and its derivatives. The second one contains thiophen systems and designed by computational modulator and changed structure from pharmacomodulations and finally, the third series of compounds is based on the urea-derivatives. Compounds have been prepared using classical organic chemistry reactions and further purification with separation techniques (chromatography). Corresponding structural elucidation by NMR, Mass and IR. Finally, part of the biological evaluation of some of the compounds have been prepared and carried out using in vitro assays based on cellular viability on different prostate (PC-3) and leukemic (TPH-1, MOLT3, NB4, HL-60, K562, KU812 and OCIAML-3) cell lines using the MTT assay. The direct inhibition of G6PDH measured by the production quantity of NADPH was used to study enzymatic inhibition activity.

Enzymatic assays based on bacterial 3α-hydroxysteroid dehydrogenase are the method of choice for quantification of total bile acids (BAs) in serum. Although non-specific, it is generally considered precise and robust. The aim of this study was to investigate how changes in the BA spectrum might affect the reliability of the method. We measured standard solutions of twenty-three human and murine BAs using a commercial enzymatic assay and compared the measured vs. expected concentrations. Additionally, total BA concentrations in rat and human cholestatic samples with an abnormal BA spectrum were measured using an enzymatic assay, and a more specific LC-MS/MS method. We observed a great variability in the response of individual BAs in the enzymatic assay. Relative signal intensities ranged from 100% in glycocholic acid (reference) to only 20% in α-muricholic acid. The enzymatic assay markedly underestimated the BA concentrations in both human and rat cholestatic sera when compared to the LC-MS/MS assay. Our study indicated that the performance of an enzymatic assay largely depends on the BA spectrum, and the total concentration of BAs can be markedly underestimated. Samples with an atypical BA spectrum (viz. in rodents) should preferably be measured by other methods.

Background We identified the species of Leishmania isolated from traveling and migrant patients attended in a reference center from 2000 to 2015, we performed the georeferencing of these species in Rio de Janeiro (RJ) state and we had knowledge about the human flows between the likely location of infection (LLI) and place of residence (PR) in RJ state, Brazil. Methodology/Principal findings This is a retrospective cross-sectional study including 171 patients diagnosed with ATL. Google Maps, OpenStreetMap, and Bing Maps were tools used to georeference LLI and PR. For etiological identification, we used isoenzyme electrophoresis, polymerase chain reaction-restriction fragment length polymorphism (molecular target hsp70C with restriction enzymes HaeIII and BstUI), and sequencing of the internal transcribed spacer of ribosomal DNA. ARCGIS software was used to create maps of the geographic distribution of Leishmania species in the state and municipality of RJ, together with flows between the LLI and PR. Isolates from 104 patients were identified as: L. (Viannia) braziliensis (80.8%), L. (V.) naiffi (7.7%), L. (V.) guyanensis (6.7%), L. (Leishmania) amazonensis (1%), and genetic variants of L. (V.) braziliensis (3.8%). The flow maps showed that the LLI included 4 countries, 19 Brazilian states, and 18 municipalities of RJ state. The Brazilian states with the highest density of cases were Amazonas (n = 32), Bahia (n = 18), and Ceará (n = 15). Conclusions/Significance This work is the first contribution to the knowledge of the routes of Leishmania species introduced in RJ state by migrants and travelers patients. L. (V.) braziliensis, L. (V.) guyanensis, L. (V.) naiffi, L. (L.) amazonensis, and genetic variants of L. (V.) braziliensis were identified in RJ state. To determine whether the autochthonous transmission of these imported species is possible it is necessary the adaptation of these species to environmental conditions as well as the presence of reservoirs and phlebotomine vectors in this region., Author summary Leishmaniasis is one of the most important neglected parasitic diseases in humans worldwide. American Tegumentary Leishmaniasis (ATL) is a disease caused by protozoa of the genus Leishmania, which are injected into the skin by small insects known by sandflies. Many different Leishmania species may cause ATL, which can affect the skin and/or oral and nasal mucosa. In this study, we analyzed flows between the likely location of infection (LLI) and the place of residence (PR) in Rio de Janeiro (RJ) state and identified Leishmania species isolated from travelers and migrants attended between 2000 and 2015 in a reference hospital for the care of patients with ATL. This work is the first contribution to knowledge about the routes of Leishmania species introduced to RJ state by migrant and traveling patients. In addition to Leishmania (Viannia) braziliensis, which was the most common species, other species have also been introduced to RJ state, including L. (V.) guyanensis, L. (V.) naiffi, L. (Leishmania) amazonensis, and genetic variants of L. (V.) braziliensis. Knowledge of the migratory flow and geographic location of these imported Leishmania species may contribute to the surveillance and control of ATL in RJ state, Brazil.

This paper assesses the dissipation of napropamide and its impact on the activity of dehydrogenases, alkaline phosphatase, acid phosphatase, and urease in sandy clay loam. The experiment was carried out on soil samples with organic carbon content of 12.08 g·kg-1, total nitrogen content of 0.97 g·kg-1, and pH 5.24 with the following variable factors: (a) dose of Devrinol 450 SC formation (containing 450 g of napropamide in dm3): 0 (control), 0.5, 1, 2, 4, 8, and 16-fold hold of field dose; (b) day of experiment: 1, 7, 14, 28, 56, and 112. The half-life of napropamide ranged from 33.50 to 71.42 days. The use of napropamide at the dose recommended by the manufacturer and at the dose reduced by half appeared to exhibit low toxicity in relation to enzymes determined. In contrast, the application of elevated napropamide doses decreased the values of biochemical parameters of the soil in most cases. The Pearson correlation coefficients showed statistically significant negative correlation between the content of napropamide residues and the enzymatic activity of the soil.

The paper evaluates the contents of total forms of selected heavy metals (Zn, Cu, Ni and Pb) as well as the activity of catalase (CAT), dehydrogenases (DEH), alkaline phosphatase (AlP) and acid phosphatase (AcP) in mineral surface horizons of forest soils exposed to the effect of road traffic pollutions. The sampling locations (n = 24) were determined in the area covered by the Szubin Forest along the exit road from Bydgoszcz to Poznań (provincial road no. 223). Soil was sampled 25 m away from the traffic lane, from two depths, 5–20 cm (humus horizons) and 20–50 cm (eluvial horizons). The contents of the heavy metals analysed were in the order of Pb > Zn > Cu > Ni. Despite intensive road traffic, with the Integrated Pollution Index (IPI) calculated, there was found a low pollution with nickel, average with zinc and copper and high with lead only. However, under the Regulation of the Minister of Environment, heavy metal values recorded allow for classifying the soils analysed as soils unpolluted with those metals. In recent years the short-chain fatty acid, 4-phenylbutyrate (PB), has emerged as a promising drug for various clinical conditions. In fact, PB has been Food and Drug Administration-approved for urea cycle disorders since 1996. PB is more potent and less toxic than its metabolite, phenylacetate (PA), and is not just a pro-drug for PA, as was initially assumed. The metabolic pathway of PB, however, has remained unclear. Therefore, we set out to identify the enzymes involved in the β-oxidation of PB. We used cells deficient in specific steps of fatty acid β-oxidation and ultra-HPLC to measure which enzymes were able to convert PB or its downstream products. We show that the first step in PB oxidation is catalyzed solely by the enzyme, medium-chain acyl-CoA dehydrogenase. The second (hydration) step can be catalyzed by all three mitochondrial enoyl-CoA hydratase enzymes, i.e., short-chain enoyl-CoA hydratase, long-chain enoyl-CoA hydratase, and 3-methylglutaconyl-CoA hydratase. Enzymes involved in the third step include both short- and long-chain 3-hydroxyacyl-CoA dehydrogenase. The oxidation of PB is completed by only one enzyme, i.e., long-chain 3-ketoacyl-CoA thiolase. Taken together, the enzymatic characteristics of the PB degradative pathway may lead to better dose finding and limiting the toxicity of this drug.

In the recent decade, CO2 has increasingly been regarded not only as a greenhouse gas but even more as a chemical feedstock for carbon‐based materials. Different strategies have evolved to realize CO2 utilization and conversion into fuels and chemicals. In particular, biological approaches have drawn attention, as natural CO2 conversion serves as a model for many processes. Microorganisms and enzymes have been studied extensively for redox reactions involving CO2. In this review, we focus on monitoring nonliving biocatalyzed reactions for the reduction of CO2 by using enzymes. We depict the opportunities but also challenges associated with utilizing such biocatalysts. Besides the application of enzymes with co‐factors, resembling natural processes, and co‐factor recovery, we also discuss implementation into photochemical and electrochemical techniques.

Aim. To study the features of the state of hemostasis and platelet enzymes activity in acetylsalicylic acid (ASA) sensitive and resistant patients with acute coronary syndrome (ACS). Material and methods. The study included 53 patients (25 men and 28 women) with ACS during the first 24 hours and after 10 days. The control group included 50 healthy volunteers. Before treatment the patients were tested on the sensitivity and resistance to ASA. The indicators of vascularplatelet and plasma hemostasis were evaluated as well as the NAD(P)- dependent dehydrogenases activity in platelets was assessed by the bioluminescent method in the first day of ACS before antiplatelet therapy and on day 10. Results. Increase in spontaneous [1.72 U (1.28-2.72 U) and 1.60 U (1.49-2.78 U), respectively] and ADP-induced [24.4% (21.1-29.8%) and 19.2% (16.1-22.9%), respectively] platelet aggregation, von Willebrand factor activity [159.0% (108.0-190.0%) and 155.0% (149.0-185.1%), respectively] was found in ASA-resistant patients with ACS in 1 and 10 day. Besides the ASA-resistant patients with ACS had very low pentose phosphate cycle and lactate dehydrogenase aerobic activity. They also demonstrated, compared with ASA-sensitive patients, higher intensity of aerobic respiration and the level of NADP-dependent substrate exchange between the tricarboxylic acid cycle and reactions of amino acid metabolism. Conclusion. Despite the dual antiplatelet therapy with ASA and clopidogrel, risk of thrombotic events is saved in ASA resistant patients with ACS. The metabolic changes in platelet influence their aggregation activity and cause an inadequate response to the antiplatelet therapy in ACS patients.

The activity of key ferments in the Krebs cycle (SDH) and glycolysis (α-GPDH) in peripheral blood lymphocytes in animals and humans with cancer pathology is considered herein. Identified is a close relationship between the fermentative activity and non-specific integral reactions of an organism, the type of which can be regulated by factors of electromagnetic nature. Monitoring of signal energetic and adaptive reactions permits to adequately evaluate an efficacy of treatment and predict recovery.

Background (+)-Nootkatone is a highly valued sesquiterpenoid compound, exhibiting a typical grapefruit aroma and various desired biological activities for use as aromatics and pharmaceuticals. The high commercial demand of (+)-nootkatone is predominately met by chemical synthesis, which entails the use of environmentally harmful reagents. Efficient synthesis of (+)-nootkatone via biotechnological approaches is thus urgently needed to satisfy its industrial demand. However, there are only a limited number of studies that report the de novo synthesis of (+)-nootkatone from simple carbon sources in microbial cell factories, and with relatively low yield. Results As the direct precursor of (+)-nootkatone biosynthesis, (+)-valencene was first produced in large quantities in Saccharomyces cerevisiae by overexpressing (+)-valencene synthase CnVS of Callitropsis nootkatensis in combination with various mevalonate pathway (MVA) engineering strategies, including the expression of CnVS and farnesyl diphosphate synthase (ERG20) as a fused protein, overexpression of a truncated form of the rate-limiting enzyme 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase (tHMG1), and downregulating the squalene synthase enzyme (ERG9). These approaches altogether brought the production of (+)-valencene to 217.95 mg/L. Secondly, we addressed the (+)-valencene oxidation by overexpressing the Hyoscyamus muticus premnaspirodiene oxygenase (HPO) variant (V482I/A484I) and cytochrome P450 reductase (ATR1) from Arabidopsis thaliana. However, (+)-valencene was predominantly oxidized to β-nootkatol and only minor amounts of (+)-nootkatone (9.66 mg/L) were produced. We further tackled the oxidation of β-nootkatol to (+)-nootkatone by screening various dehydrogenases. Our results showed that the short-chain dehydrogenase/reductase (SDR) superfamily dehydrogenases ZSD1 of Zingiber zerumbet and ABA2 of Citrus sinensis were capable of effectively catalyzing β-nootkatol oxidation to (+)-nootkatone. The yield of (+)-nootkatone increased to 59.78 mg/L and 53.48 mg/L by additional overexpression of ZSD1 and ABA2, respectively. Conclusion We successfully constructed the (+)-nootaktone biosynthesis pathway in S. cerevisiae by overexpressing the (+)-valencene synthase CnVS, cytochrome P450 monooxygenase HPO, and SDR family dehydrogenases combined with the MVA pathway engineering, providing a solid basis for the whole-cell production of (+)-nootkatone. The two effective SDR family dehydrogenases tested in this study will serve as valuable enzymatic tools in further optimizing (+)-nootkatone production.

N-alkylated-α-amino acids are useful building blocks for the pharmaceutical and fine chemical industries. Enantioselective methods of N-alkylated-α-amino acid synthesis are therefore highly valuable and widely investigated. While there are a variety of chemical methods for their synthesis, they often employ stoichiometric quantities of hazardous reagents such as pyrophoric metal hydrides or genotoxic alkylating agents, whereas biocatalytic routes can provide a greener and cleaner alternative to existing methods. This review highlights the occurrence of the N-alkyl-α-amino acid motif and its role in nature, important applications towards human health and biocatalytic methods of preparation. Several enzyme classes that can be used to access chiral N-alkylated-α-amino acids and their substrate selectivities are detailed. Postprint

Non-methanotrophic bacteria such as methylotrophs often coexist with methane-oxidizing bacteria (methanotrophs) by cross-feeding on methane-derived carbon. Methanol has long been considered a major compound that mediates cross-feeding of methane-derived carbon. Despite the potential importance of cross-feeding in the global carbon cycle, only a few studies have actually explored metabolic responses of a bacteria when cross-feeding on a methanotroph. Recently, we isolated a novel facultative methylotroph, Methyloceanibacter caenitepidi Gela4, which grows syntrophically with the methanotroph, Methylocaldum marinum S8. To assess the potential metabolic pathways in M. caenitepidi Gela4 co-cultured with M. marinum S8, we conducted genomic analyses of the two strains, as well as RNA-Seq and chemical analyses of M. caenitepidi Gela4, both in pure culture with methanol and in co-culture with methanotrophs. Genes involved in the serine pathway were downregulated in M. caenitepidi Gela4 under co-culture conditions, and methanol was below the detection limit (< 310 nM) in both pure culture of M. marinum S8 and co-culture. In contrast, genes involved in the tricarboxylic acid cycle, as well as acetyl-CoA synthetase, were upregulated in M. caenitepidi Gela4 under co-culture conditions. Notably, a pure culture of M. marinum S8 produced acetate (< 16 μM) during growth. These results suggested that an organic compound other than methanol, possibly acetate, might be the major carbon source for M. caenitepidi Gela4 cross-fed by M. marinum S8. Co-culture of M. caenitepidi Gela4 and M. marinum S8 may represent a model system to further study methanol-independent cross-feeding from methanotrophs to non-methanotrophic bacteria.

Streptococcus pneumoniae displays increased resistance to antibiotic therapy following biofilm formation. A genome-wide search revealed that SP 0320 and SP 0675 (respectively annotated as 5-keto-D-gluconate-5-reductase and glucose dehydrogenase) contain the highest degree of homology to CsgA of Myxococcus xanthus, a signaling factor that promotes cell aggregation and biofilm formation. Single and double SP 0320 and SP 0675 knockout mutants were created in strain BS72; however, no differences were observed in the biofilm-forming phenotypes of mutants compared to the wild type strain. Using the chinchilla model of otitis media and invasive disease, all three mutants exhibited greatly increased virulence compared to the wild type strain (increased pus formation, tympanic membrane rupture, mortality rates). The SP 0320 gene is located in an operon with SP 0317, SP 0318 and SP 0319, which we bioinformatically annotated as being part of the Entner-Doudoroff pathway. Deletion of SP 0317 also resulted in increased mortality in chinchillas; however, mutations in SP 0318 and SP 0319 did not alter the virulence of bacteria compared to the wild type strain. Complementing the SP 0317, SP 0320 and SP 0675 mutant strains reversed the virulence phenotype. We prepared recombinant SP 0317, SP 0318, SP 0320 and SP 0675 proteins and confirmed their functions. These data reveal that disruption of genes involved in the degradation of ketogluconate, the Entner-Doudoroff pathway, and glucose dehydrogenase significantly increase the virulence of bacteria in vivo; two hypothetical models involving virulence triggered by reduced in carbon-flux through the glycolytic pathways are presented.

During the last decade the number of characterized F420-dependent enzymes has significantly increased. Many of these deazaflavoproteins share a TIM-barrel fold and are structurally related to FMN-dependent luciferases and monooxygenases. In this work, we traced the origin and evolutionary history of the F420-dependent enzymes within the luciferase-like superfamily. By a thorough phylogenetic analysis we inferred that the F420-dependent enzymes emerged from a FMN-dependent common ancestor. Furthermore, the data show that during evolution, the family of deazaflavoproteins split into two well-defined groups of enzymes: the F420-dependent dehydrogenases and the F420-dependent reductases. By such event, the dehydrogenases specialized in generating the reduced deazaflavin cofactor, while the reductases employ the reduced F420 for catalysis. Particularly, we focused on investigating the dehydrogenase subfamily and demonstrated that this group diversified into three types of dehydrogenases: the already known F420-dependent glucose-6-phosphate dehydrogenases, the F420-dependent alcohol dehydrogenases, and the sugar-6-phosphate dehydrogenases that were identified in this study. By reconstructing and experimentally characterizing ancestral and extant representatives of F420-dependent dehydrogenases, their biochemical properties were investigated and compared. We propose an evolutionary path for the emergence and diversification of the TIM-barrel fold F420-dependent dehydrogenases subfamily. Fil: Mascotti, María Laura. Background Glucose-6-Phosphate dehydrogenase (G6PD) deficiency is the most common enzymopathy worldwide, no reliable bedside diagnostic tests to quantify G6PD activity exist. This study evaluated two novel quantitative G6PD diagnostics. Methods Participants with known G6PD activity were enrolled in Bangladesh. G6PD activity was measured by spectrophotometry, Biosensor (BS; AccessBio/CareStart, USA) and STANDARD G6PD (SG; SDBiosensor, ROK). G6PD activity was measured repeatedly in a subset of samples stored at room temperature and 4˚C. Results 158 participants were enrolled, 152 samples tested by BS, 108 samples by SG and 102 samples were tested by all three methods. In comparison to spectrophotometry BS had sensitivity and specificity of 72% (95%CI: 53–86) and 100% (95%CI: 97–100) at 30% cut off respectively, while SG had a sensitivity of 100% (95%CI: 88–100) and specificity of 97% (95%CI: 91–99) at the same cut off. The sensitivity and specificity at 70% cut off activity were 71% (95%CI: 59–82) and 98% (95%CI, 92–100) respectively for BS and 89% (95%CI: 77–96) and 93% (95%CI: 83–98) respectively for SG. When an optimal cut-off was applied the sensitivity of the BS at 70 cut off rose to 91% [95%CI: 80–96] and specificity to 82% [95%CI: 83–89]; a diagnostic accuracy comparable to that of the SG (p = 0.879). G6PD activity dropped significantly (-0.31U/gHb, 95%CI: -0.61 to -0.01, p = 0.022) within 24 hours in samples stored at room temperature, but did not fall below 90% of baseline activity until day 13 (-0.87U/gHb, 95%CI: (-1.11 to -0.62), p Conclusion BS and SG are the first quantitative diagnostics to measure G6PD activity reliably at the bedside and represent suitable alternatives to spectrophotometry in resource poor settings. If samples are stored at 4˚C, G6PD activity can be measured reliably for at least 7 days after sample collection.

Mitochondria are organelles involved in the production of cellular energy, regulation of Ca2+ and redox signaling, and are critical for normal functioning of eukaryotic cells. The dysfunction of mitochondria has been implicated in a wide range of diseases, including metabolic and neurodegenerative disorders and different types of cancers. To better understand the role of mitochondria in healthy and disease states, the development of efficient and reliable tools for the assessment of mitochondrial function is particularly important. Janus green B (JG-B) is a supravital lipophilic cationic dye which, in its oxidized form, has a green-blue color. As JG-B is taken up and reduced by metabolically active mitochondria, the dye has been used for assessing the purity, integrity and metabolic activity of mitochondria with microscopy-based methods. Here we present a simple, time- and cost-efficient JG-B-based colorimetric assay for assessing mitochondrial function, activity and toxicity. The method is based upon reduction of JG-B by mitochondrial dehydrogenases to diethylsafranine, which is pink colored and has a maximum absorption at 550 nm. In this proof of principle study, using in vitro mitochondrial preparations isolated from rat brain, we provide evidence that monitoring JG-B conversion to diethylsafranine can be used as a reliable and robust indicator of mitochondrial activity and toxicity. Because of its simplicity and efficiency in terms of costs and time, this assay has a wide potential in analytical as well as therapeutic areas of biomedical research.

During germination at low temperatures, seeds rich in proteins may experience damage to their cytoplasmic membranes. The study aimed to investigate the influence of the germination temperature on growth, development, and yield of four cultivars of soybean, a typical thermophilic species. The seeds were germinated at 10, 15, and 25 °C in the dark. After 48 h, one part of the seeds was analyzed for their amylase and dehydrogenase activity, cell membrane permeability, and germination vigor. The other part was transferred into soil and cultivated up to yielding. Chlorophyll fluorescence, fresh (FW) and dry weight (DW) of shoots, pod and seed number, and seed DW were analyzed. The plants of cvs. ‘Abelina’, ‘Malaga’, and ‘Merlin’, germinating at low temperature, produced the highest number of seeds. Seed number negatively correlated with their DW and positively with the number of active reaction centers (RC/CSm) in all cultivars. In cvs. ‘Abelina’ and ‘Malaga’, the number of seeds also positively correlated with the index performance of photosystem II (PSII), which was the highest in all plants germinating at low temperature. We suggest cultivating cv. ‘Abelina’ in cooler regions, while cvs. ‘Petrina’ and ‘Malaga’ in warmer areas.

The present study aims to define the effects of Cold Atmospheric Pressure Plasma (CAPP) exposure on seed germination of an agriculturally important crop, soybean. Seed treatment with lower doses of CAPP generated in ambient air and oxygen significantly increased the activity of succinate dehydrogenase (Krebs cycle enzyme), proving the switching of the germinating seed metabolism from anoxygenic to oxygenic. In these treatments, a positive effect on seed germination was documented (the percentage of germination increased by almost 20% compared to the untreated control), while the seed and seedling vigour was also positively affected. On the other hand, higher exposure times of CAPP generated in a nitrogen atmosphere significantly inhibited succinate dehydrogenase activity, but stimulated lactate and alcohol dehydrogenase activities, suggesting anoxygenic metabolism. It was also found that plasma exposure caused a slight increment in the level of primary DNA damage in ambient air- and oxygen-CAPP treatments, and more significant DNA damage was found in nitrogen-CAPP treatments. Although a higher level of DNA damage was also detected in the negative control (untreated seeds), this might be associated with the age of seeds followed by their lower germination capacity (with the germination percentage reaching only about 60%).

Methanol is often considered as a non-competitive substrate for methanogenic archaea, but an increasing number of sulfate-reducing microorganisms (SRMs) have been reported to be capable of respiring with methanol as an electron donor. A better understanding of the fate of methanol in natural or artificial anaerobic systems thus requires knowledge of the methanol dissimilation by SRMs. In this study, we describe the growth kinetics and sulfur isotope effects of Desulfovibrio carbinolicus, a methanol-oxidizing sulfate-reducing deltaproteobacterium, together with its genome sequence and annotation. D. carbinolicus can grow with a series of alcohols from methanol to butanol. Compared to longer-chain alcohols, however, specific growth and respiration rates decrease by several fold with methanol as an electron donor. Larger sulfur isotope fractionation accompanies slowed growth kinetics, indicating low chemical potential at terminal reductive steps of respiration. In a medium containing both ethanol and methanol, D. carbinolicus does not consume methanol even after the cessation of growth on ethanol. Among the two known methanol dissimilatory systems, the genome of D. carbinolicus contains the genes coding for alcohol dehydrogenase but lacks enzymes analogous to methanol methyltransferase. We analyzed the genomes of 52 additional species of sulfate-reducing bacteria that have been tested for methanol oxidation. There is no apparent relationship between phylogeny and methanol metabolizing capacity, but most gram-negative methanol oxidizers grow poorly, and none carry homologs for methyltransferase (mtaB). Although the amount of available data is limited, it is notable that more than half of the known gram-positive methanol oxidizers have both enzymatic systems, showing enhanced growth relative to the SRMs containing only alcohol dehydrogenase genes. Thus, physiological, genomic, and sulfur isotopic results suggest that D. carbinolicus and close relatives have the ability to metabolize methanol but likely play a limited role in methanol degradation in most natural environments.
