Key message Geography and tree diameter were the major factors in determining oleoresin yield in Sindora glabra. The tangential diameter and area of secretory canals are reliable indicators of the oleoresin amount. Abstract Sindora glabra is a native tropical plant of China that exudes copious amounts of oleoresin from wounds on the stem. Elucidation of the mechanism affecting the oleoresin yield is of particular importance for development and selection of superior germplasm. In this study, the variation in oleoresin yield and anatomical traits of the stem of S. glabra were investigated in four main natural populations in Hainan. The results showed that annual oleoresin yield varied considerably among populations. The oleoresin yield exhibited an increasing trend with the increment of tree stem diameter until the diameter was over 80 cm. Moreover, oleoresin production in the wet season was higher than in dry season. The analysis of oleoresin chemical composition indicated that the four populations belong to the same chemotype despite the amount of four types of sesquiterpenes varying greatly among areas. Anatomical analysis revealed that quantitative traits of secretory canals including tangential diameter, area, density and frequency were significantly different among populations and tree diameters. Furthermore, the tangential diameter and area of secretory canals were positively correlated with oleoresin yield. These results suggested that the tangential diameter and area of secretory canals are reliable indicators of the oleoresin amount and can be used for selection purposes in S. glabra.

Objective: To explore the effect of CDKN2B antisense RNA 1 (CDKN2B-AS1) on the proliferation, clone formation, and invasion of nasopharyngeal carcinoma (NPC) cells by regulating miR-98-5p/E2F transcription factor 2 (E2F2) axis. Methods: The expressions of CDKN2B-AS1, miR-98-5p, and E2F2 in NPC tissues and cell lines (SUNE-1, 5-8F, 6-10B, and HK-1) as well as in peritumoral normal tissues and cell line NP69 were determined by qRT-PCR. Subcellular localization of CDKN2B-AS1 was detected using the fluorescence in situ hybridization assay. The targeting relationships between CDKN2B-AS1 and miR-98-5p as well as between miR-98-5p and E2F2 were analyzed by the dual-luciferase reporter assay and RNA binding protein immunoprecipitation assay. The proliferation, clone formation and invasion of 5-8F cells were measured using the CCK-8 assay, Clone formation assay, and transwell assay, respectively. Background SPL (SQUAMOSA-promoter binding protein-like) proteins form a large family of plant-specific transcription factors that play essential roles in various aspects of plant growth and development. They are potentially important candidates for genetic improvement of agronomic traits. However, there were limited information about the SPL genes in Jatropha curcas, an important biofuel plant. Results In Jatropha, 15 JcSPL genes were identified. Phylogenetic analysis revealed that most of the JcSPLs were closely related to SPLs from woody plant rather than herbaceous plant and distantly related to monocotyledon SPLs. Gene structure, conserved motif and repetitive sequence analysis indicated diverse and specific functions of some JcSPL genes. By combination of target prediction and degradome sequencing analysis, 10 of the 15 JcSPLs were shown to be targets of JcmiR156. Quantitative PCR analysis showed diversified spatial-temporal expression patterns of JcSPLs. It is interesting that the expression levels of JcSPL3 were the highest in all tissues examined in 7- or 10-year-old plants and exhibited increasing trend with plant age, suggesting its important role in the regulation of age development in Jatropha. Overexpression of JcSPL3 in Arabidopsis resulted in earlier flowering time, shorter silique length and reduced biomass of roots. Conclusions Through comprehensive and systematic analysis of phylogenetic relationships, conserved motifs, gene structures, chromosomal locations, repetitive sequence and expression patterns, 15 JcSPL genes were identified in Jatropha and characterized in great detail. These results provide deep insight into the evolutionary origin and biological significance of plant SPLs and lay the foundation for further functional characterization of JcSPLs with the purpose of genetic improvement in Jatropha.

Mytilaria laosensis is a fast-growing tropical broadleaf tree that is extensively used for wood production and has significant ecological benefits. To investigate the genetic diversity and population structure of M. laosensis, eight major natural popu­lations in China were analyzed by using simple sequence repeat (SSR) markers. A total of 88 microsatellite-containing fragments were obtained by the method of magnetic bead enrichment, among which 26 pairs of SSR primers were scree­ned out and used to generate a total of 190 alleles among 152 individuals. The average of observed number of alleles, Shannon’s information index and polymorphism information content per locus were 18.3, 1.1577 and 0.7759, respectively, implying a high level of genetic diversity in M. laosensis popu­lations. The variation within populations accounted for 81.74 % of total variation based on analysis of molecular variance. Clus­ter analysis divided the eight populations into four groups, among which five populations from the southern parts of Guangxi province were classified as one major group. Mantel test showed that there was highly significant correlation bet­ween Euclidean genetic distance and geographic distance, suggesting that geographic isolation contribute to the high genetic diversity of M. laosensis. Together, these could provide support for the feasibility of exploration and utilization of M. laosensis in subtropical areas of East Asia including Jiangxi, Hunan and Fujian province of China.

A novel esterase gene TLip was identified from the strain Thauera sp. and expressed at high levels in Escherichia coli. The TLip protein shared the highest identity (48%) to esterase TesA from Pseudomonas aeruginosa when compared to enzymes with reported properties. Phylogenetic analysis showed that TLip belongs to the GDSL family of bacterial lipolytic enzymes. TLip was an alkaline esterase with a broad optimal temperature range 37-50 °C and an optimal pH of 8.0. Substrate specificity assays showed that TLip preferred medium chain p-nitrophenyl esters (C6 -C12 ). Besides, the activity of TLip was strongly inhibited by Cu2+ but greatly enhanced by Triton X-100 and Tween 80. Thermostability assay revealed that TLip was stable without loss of activity at 37 °C and still retained 69% activity at 50 °C after 2 H of incubation. Together, these provided a good candidate for further exploration of TLip as a promising biocatalyst in industry.

Sindora glabra, one of the second-class national protective plants in China, has important ecological and commercial values. To understand the genetic diversity and structure of S. glabra, eleven natural populations from four areas of Hainan Island in China were analyzed by inter-simple sequence repeat (ISSR) markers. Thirteen primers were screened out and used to amplify 157 DNA samples. A total of 122 bands were obtained, among which 114 (93.4%) bands were polymorphic. Genetic parameters including average number of effective alleles (1.547), Nei's gene diversity (0.321), Shannon's information index (0.482), and gene differentiation coefficient (0.1944) revealed a high level of genetic diversity maintained in S. glabra populations. The variation within populations accounted for 85.6% of total variation based on analysis of molecular variance. Genetic distance analysis showed that 11 populations could be divided into three groups and populations from the same areas were classified as one group, suggesting that high genetic diversity of S. glabra was attributable to geographic isolation. Together, introduction of germplasm from distant natural distribution areas would be a sound strategy for germplasm resource conservation of S. glabra and selection of elite individuals from populations of far relationship for hybridization is of great importance for breeding improvement programs in future.

A male-specific SCAR DNA marker was developed using a RAPD DNA marker specific for male plants of Salacca zalacca var. zalacca (salak palm). The marker is 1579 bp long and has a GC content of 38.5 %. Its sequence contains 1 or 2 open reading frames, indicating the marker is probably a coding region. No highly similar sequences were found in a search of the GenBank database. Sexes were identified using the SCAR DNA marker for three kinds of seedlings grouped by the number of seeds per fruit (1, 2 or 3). The sex ratio of female to male did not differ significantly from 1:1 for the three kinds of seedlings, implying that the number of seeds per fruit is not a reliable index to identify the sex of a seedling.

Terpenes serve important physiological and ecological functions in plants. Sindora glabra trees accumulate copious amounts of sesquiterpene-rich oleoresin in the stem. A transcriptome approach was used to determine the unique terpene biosynthesis pathway and to explore the different regulatory mechanisms responsible for the variation of terpene content among individuals. Analysis of de novo-assembled contigs revealed a complete set of genes for terpene biosynthesis. A total of 23,261 differentially expressed unigenes (DEGs) were discovered between high and low oil-yielding plants. DEG enrichment analysis suggested that the terpene biosynthesis process and the plant hormone signal transduction pathway may exert a major role in determining terpene variation in S. glabra. The expression patterns of candidate genes were further verified by quantitative RT-PCR experiments. Key genes involved in the terpene biosynthesis pathway were predominantly expressed in phloem and root tissues. Phylogenetic analysis and subcellular localization implied that S. glabra terpene synthases may evolve from a common ancestor. Furthermore, two sesquiterpene synthase genes, SgSTPS1 and SgSTPS2, were functionally characterized. SgSTPS1 mainly generated β-caryophyllene from farnesyl pyrophosphate. SgSTPS2 is a versatile enzyme that catalyzes the formation of 12 sequiterpenes from farnesyl pyrophosphate and synthesis of three monoterpenes using geranyl pyrophosphate. Together, these results provide large reservoir for elucidating the molecular mechanism of terpene biosynthesis and for exploring the ecological function of sesquiterpenes in S. glabra.

Graphene is a material that has superior mechanical, electrical, and thermal properties. It has drawn the attention of many the scientific researchers for this purpose. In this project, three different types of fillers, graphene nanoplatelets (GNPs)-, multi-walled carbon nanotubes (MWCNTs)-, and expanded graphite (EG)-reinforced epoxy nanocomposites were prepared by shear mixing. Different shear mixing speeds and shear mixing times were used during the preparation of the nanocomposites with 0.1 wt% loading of the fillers. The effects of various types of fillers and different shear mixing speeds and durations on mechanical and electrical properties of the final composites were examined. The GNPs-reinforced epoxy nanocomposite was the only one that showed a 13% improvement in elastic modulus as compared to pure epoxy when the shear mixing conditions were 3000 rpm for 2 hours.The project also studied the effects of different loadings of GNPs and the addition of acetone as a solvent on the final mechanical, electrical and thermal properties of the composites (with the fixed shear mixing speed and time). The tensile strength of the composites reduced drastically when the loading of GNPs increased while the elastic modulus show some increase with the growth in GNP loading. The GNPs-reinforced composites did not show the percolation threshold even with 5 wt% (with the ratio to the weight of epoxy) loading of the GNPs. The GNPs-reinforced epoxy composites showed an 116% improvement in the thermal conductivity as compared to the pure epoxy samples when the GNPs loading was 5 wt%. The results also showed that the samples prepared with the addition of acetone had higher thermal diffusivity than the samples prepared without acetone.

Many stems are subject to shoot drying in the plantations of Calamus simplicifolius Wei at the Nanmeiling Base of the Forestry Bureau of Baisha County, Hainan Province. As a palm, stems of C. simplicifolius stop growing and later die if the shoots are dry. Generally, shoot drying means reduced cane production and reduced profit for a rattan plantation. We aimed to quantify the incidence of shoot drying and determine its reason. We sampled eight 20 m × 20 m plots in the C. simplicifolius plantations. In each plot, we counted stems and categorized them as drying shoot or normal shoot, and mother stem or sucker stem. We measured stem length and categorized them as drying shoot and normal shoot for mother stems. We cut open each shoot-drying stem to determine the cause of drying from drying part to fresh part. The frequency of mother stems (259 stem·ha−1), was significantly lower than sucker stems (588 stem·ha−1). The percentage of shoot drying was 33.8% for mother stems, not significantly higher than that for sucker stems (18.9%). The mother shoot stems were significantly shorter if they were drying than if normal. 80% of stems with a drying shoot were hollow within the leaf sheath, whereas 10% hosted lively mature larvae (one larvae per shoot) and 10% hosted several lively young larvae. Mature larvae were identified as the Red Palm Weevil, Rhynchophorus ferrugineus Oliver. To confirm the present of the weevil, three pheromone traps for the weevil were set up in the plantations and adult weevils were trapped on the next day and identified as the Red Palm Weevil. We conclude that shoot drying of C. simplicifolius is probably caused by the Red Palm Weevil.

Hainan Island is the biggest supplier of domestic rattan canes in China and is considered the best area to develop rattan plantation in large-scale. This paper discusses the rattan production in Hainan Island on the basis of comprehensively collected information on rattan through interviewing with farmers, middle man, manufacture and official, on-site observation and literatures. The stock of available rattan is more than 25×106kg, but the real production annually is 4×105 kg. The area of rattan plantation established during 2000–2005 was about 10 000 hm2, however, they don not produce yet. Four indigenous rattan species (C. Simplicifolius, C. Faberii, C. Tetradactylus, D. margaritae) and three kinds of exotic canes are mainly utilized in Hainan island. Large quality of canes is treated as timber regulated by government, tax should be paid and a license should be applied from the government if canes are transport out of Hainan Island. The production-to-consumption flow of rattan in Hainan Island was drawn out based on the investigation. Constraints and advantages of the rattan sector in Hainan are identified and recommendations are proposed finally.

A greenhouse pot experiment was conducted to study the effects of nitrogen fertilization on Calophy- llum inophyllum seedlings grown with 0, 50, 100, 150, 200, 300, 400 and 600 mg N per seedling according to exponential functions. Seedling height, root collar diameter, leaf area and total biomass increased with increasing fertilization from 0 to 200 mg N per seedling and decreased with further increase in fertilization from 300 to 600 mg N per seedling. The net photosynthetic rate, stomatal conductance, intercellular CO2 concentration and transpiration rate of C. inophyllum seedlings showed a unimodal parabolic trend, with peak values of 7.29 mmol·m−2·s−1, 0.071 mol·m−2·s−1, 220 mmol·mol−1 and 1.34 mmol·m−2·s−1, respectively, when the rate of fertilization was 200 mg N per seedling. Photosynthetic gas exchange parameters were significantly different among nitrogen treatments. Based on the critical values of leaf N and P concentration and N/P ratio, the optimum amount of nitrogen of C. inophyllum seedlings was 200–400 mg per seedling for leaf N and P concentration, and 100–400 mg per seedling for N/P ratio. It was concluded that 200–400 mg N per seedling was the most suitable nitrogen range for C. inophyllum seedlings.

The aim of this study was to investigate the utility of datasets composed of texture measures and other features for the classification of vegetation cover, specifically wetlands. QUEST decision tree classifier was applied to a SPOT-5 image sub-scene covering the typical wetlands area in Three River Sources region in Qinghai province, China. The dataset used for the classification comprised of: (1) spectral data and the components of principal component analysis; (2) texture measures derived from pixel basis; (3) DEM and other ancillary data covering the research area. Image textures is an important characteristic of remote sensing images; it can represent spatial variations with spectral brightness in digital numbers. When the spectral information is not enough to separate the different land covers, the texture information can be used to increase the classification accuracy. The texture measures used in this study were calculated from GLCM (Gray level Co-occurrence Matrix); eight frequently used measures were chosen to conduct the classification procedure. The results showed that variance, mean and entropy calculated by GLCM with a 9*9 size window were effective in distinguishing different vegetation types in wetlands zone. The overall accuracy of this method was 84.19% and the Kappa coefficient was 0.8261. The result indicated that the introduction of texture measures can improve the overall accuracy by 12.05% and the overall kappa coefficient by 0.1407 compared with the result using spectral and ancillary data.
