Combining the killing ability of chemotherapy drugs on tumor cells with the inhibiting ability of genetic drugs on tumor cell growth, a dual drug delivery system loaded with therapy drugs and siRNA has gradually received more and more research and extensive attention. In this paper, we designed a DNA nano-assembly based on rolling circle amplification that can co-deliver doxorubicin (Dox) and siRNA simultaneously. In order to fully exploit the potential of the dual loading system in cancer treatment, we selected STAT3 gene as a target and used siRNA to target STAT3 of mRNA and reduce the STAT3 expression in mouse melanoma cell line (B16); meanwhile, Dox as a chemotherapy drug was combined with multivalent aptamers specifically targeting B16 to achieve efficient delivery of siRNA and Dox. The results showed that the synergistic delivery system could achieve high efficiency in targeting and inhibiting proliferation in mouse melanoma cells. In addition, the synergistic effect of the dual delivery system on apoptosis of cancer cells was significantly better than that of single drug delivery systems.

DNA nanotechnology is beginning to yield unique advantages in the area of drug delivery. For the dual-targeting and proliferation suppression of cancer cells, a "willow branch" DNA assembly based on rolling circle amplification (RCA) was built. Three single-stranded DNAs, including antibody modified cDNAs, aptamer cDNAs, and simple cDNAs, were employed in the DNA self-assembly, along with the RCA scaffolds (every 63 bases is a repeat unit). "Willow branch" DNA (WB DNA) assembly successfully linked multiple antibodies and aptamers together to achieve dual targeting of cancer cells. Binding of CD44 antibodies and S2.2 aptamers to receptors on the cell membrane inhibits both pathways, β-catenin signaling and nuclear factor-kappa B-specific transcription activity, through feedback regulation. Results demonstrated that WB DNA assembly could effectively exert multivalency clustering cell-surface receptors, modulating signal pathways and inhibiting proliferation. This study proposes a new approach for cancer dual-target and proliferation inhibition by clustering multivalent receptors.

14. Effect of Sealing Treatment on Corrosion Resistance of Arc-Sprayed Zn and Zn85-Al15 Coatings

This study investigated the corrosion morphology of arc-sprayed Zn and Zn85-Al15 coatings with and without sealing treatment under simulated defect conditions. The hole sealing treatment was carried out by Conventional Impregnation Sealing (CIS). The performance of two coatings was assessed by employing morphological analysis, chemical composition, and electrochemical studies. The results showed that the corrosion performance of two coatings with sealing treatments was better than that of the un-sealing coating. Through the double protection of coating and sealing treatment, the defect-free material has excellent anti-corrosion performance in the salt spray experiment. In the simulated defect environment, the Zn-Al coating has better corrosion resistance, and the corrosion products are denser and more stable near the defects, which reduces the overall corrosion rate of the coating. The electrochemical experiment results demonstrated that the Zn-Al coating exhibited a three times lower corrosion rate compared to the Zn coating in the immersion experiment, and the corrosion rate did not change with the thickness of the coating. The average adhesion values of the two different methods (chilled iron and alumina) were similar (~8 MPa), but after the salt spray test, the adhesion strength increased. The corrosion mechanisms of Zn coating and Zn-Al coating are analyzed and compared. In general, it was indicated that the Zn85/Al15 coating prepared by sealing treatment has better corrosion resistance than the matrix steel. This study can provide some reference for corrosion behavior in defect environments.

18. Risk factors for breakthrough urinary tract infection in children with vesicoureteral reflux receiving continuous antibiotic prophylaxis

BACKGROUND: To investigate the risk factors for breakthrough urinary tract infection (BT-UTI) in children with vesicoureteral reflux (VUR) receiving continuous antibiotic prophylaxis (CAP). METHODS: This was a single-centre cohort study (January 2016 to December 2019). The clinical data of 256 children with grade I–V VUR receiving CAP were analysed. In this study, exposure variables were sex, younger age at the initial diagnosis of UTI ≤12 months, high-grade VUR, bilateral VUR, aetiology, presence of renal scarring at the initial diagnosis, presence of renal function impairment at the initial diagnosis, ultrasound abnormalities, antibiotic used and bladder and bowel dysfunction (BBD). Outcome was BT-UTI. RESULTS: BT-UTI occurred in 81 out of 256 children with grade I–V VUR who received CAP, an incidence of 31.64%. Univariate analysis showed that younger age at the initial diagnosis of UTI (≤12 months), bilateral VUR, renal scarring on the dimercaptosuccinic acid (DMSA) scan at the initial diagnosis of UTI and BBD were correlated with the occurrence of BT-UTI. Multivariate analysis showed that younger age at the initial diagnosis of UTI (≤12 months) [hazard ratio (HR): 4.629; 95% confidence interval (CI): 1.302–16.462], bilateral VUR (HR: 2.078; 95% CI: 1.084–4.022) and BBD (HR: 3.194; 95% CI: 1.243–8.206) were independent risk factors for the occurrence of BT-UTI. CONCLUSIONS: For VUR children receiving CAP, younger age at the initial diagnosis of UTI (≤12 months), bilateral VUR, and BBD were independent risk factors for the occurrence of BT-UTI.

19. Study on Association Behavior and Solution Properties of Poly(Acrylic Acid-Alkyl Polyoxyethylene Acrylate) Amphiphilic Copolymers

- Abstract
The hydrophobically associating water-soluble polymers (HAWSPs), which possess both surfactant and polymer properties, have been the subject of considerable research. In this study, a novel HAWSP, poly(acrylic acid-alkyl polyoxyethylene acrylate) (P(AA-AAEOn)), was synthesized via aqueous polymerization of acrylic acid andalkyl polyoxyethylene acrylate (AAEOn) with different ethoxy groups. The structure of P(AA-AAEOn) was characterized by Fourier Transform Infrared spectroscopy (FTIR) and Nuclear Magnetic Resonance (NMR). Additionally, its association behavior in solution was evaluated by surface tension, dynamic light scattering, and rheology. The results indicated that P(AA-AAEOn) exhibited excellent surface activity, and its ability to reduce the water surface tension increased with the increase in the proportion of the hydrophobic group. When the concentration reached 0.01%, P(AA-AAEOn) in the solution changed from intramolecular association to intermolecular association. Furthermore, P(AA-AAEOn) also displayed remarkable shear resistance, shear recovery, and viscoelasticity, with a transition from energy elasticity to entropy elasticity in P(AA-AAEOn) molecules between 25℃ and 35℃.

BackgroundNowadays, the number of patients with non-valvular atrial fibrillation (NVAF) complicated by end-stage renal disease (ESKD) is increasing. There are significant challenges in anticoagulation with prescription drugs because of the high risk of bleeding and embolism among these patients. However, no randomized controlled trials (RCTs) of warfarin in combination with any non-vitamin K oral anticoagulant (NOACs) have been performed in patients with baseline creatinine clearance (CrCl) MethodsThe present systematic review and meta-analysis searched the databases of PubMed, Embase, the Cochrane Library, CNKI, CBM, and Google Scholar for relevant studies from inception to 1 June 2022, with the restriction of English and Chinese. Eligible cohort studies and RCTs that reported efficacy outcomes [composite of stroke and systemic embolism (SSE), ischemic stroke (ICS), and systemic embolization] or safety outcomes [major bleeding, intracranial hemorrhage (ICH), and gastrointestinal bleeding (GIB)] of rivaroxaban in NVAF patients with ESKD were enrolled. Two authors completed the data extraction and quality assessment work, respectively. The Cochrane Collaboration tool for assessing the risk of bias was used for RCTs, and the NEW-Castle Ottawa scale was used for study quality assessment for cohort studies. Dichotomous variables were calculated as risk factors with 95% confidence intervals (CIs), and meta-analysis was performed to probe the effect of research design, rivaroxaban dose, and controlled drug factors on outcomes.ResultsIn total, three studies were included for meta-analysis, involving 6,071 NVAF patients with ESKD, and two studies were included for qualitative analysis. All included studies were at low risk of bias. A meta-analysis demonstrated that mix-dose rivaroxaban caused no statistical discrepancy in the occurrence of thrombotic and bleeding events when compared to the control group (embolism, LogOR: −0.64, 95% CI: −1.05 to −0.23, P:0.25; bleeding, LogOR: −0.33, 95% CI: −0.63 to −0.03, P:0.15), and low-dose rivaroxaban produced similar results (embolism, LogOR: −1.04, 95% CI: −2.15 to 0.07, P:0.61; bleeding, LogOR: −0.81, 95% CI: −1.19 to −0.44, P:0.93).ConclusionIn this study, low-dose rivaroxaban (10 mg, once a day) may benefit more than warfarin in patients with NVAF and ESKD.Systematic review registrationhttps://www.crd.york.ac.uk/prospero/#recordDetails, identifier CRD42022330973.

Chayote is rich in non-starch polysaccharides. The physicochemical properties of non-starch polysaccharide from chayote were not study extensively. In the present research, non-starch polysaccharide of chayote (CP-CHE) extracted by conventional heating extraction method was obtained, and the preliminary physicochemical and functional properties of CP-CHE had been carried out. The results indicated that the CP-CHE was rich in galactan, glucoses and galacturonic acid. Also, CP-CHE had the negatively charged, a wide distribution of molecular weight and particle size. Additionally, CP-CHE solutions had shear thinned fluid property when the concentrations between 1% to 4%. The results of this study would be beneficial to the application of CP-CHE in food and improve the economic value of chayote.

25. Systematically uncovering the absorbed effective substances of Radix scutellaria-licorice drug pair in rat plasma against COVID-19 using a combined UHPLC-Q-TOF-MS analysis and target network pharmacology

- Abstract
Rationale: Radix scutellaria-Licorice drug pair (RSLDP), a frequently used herbal pair with the effect of clearing heat and detoxifying, is the commonly employed drug pair in TCM prescription for the treatment of COVID-19. Until now, the metabolism feature and anti-COVID-19 mechanism of RSLDP have not been fully elucidated. Methodology: In this study, a sensitive and rapid method was developed for the separation and identification of the absorbed constituents of RSLDP in the rat plasma by UHPLC-QTOF-MS. Besides, we optimized the conventional process ways of network pharmacology and proposed a new concept called target network pharmacology (T-NP). It used the absorbed constituents and the corresponding targets to generate compound-target network, and compare to the conventional network pharmacology, it could reduce the false positive results. Results: A total of 85 absorbed constituents were identified or tentatively characterized in dosed plasma,including 32 components in Radix Scutellaria,27 components in Licorice and 65 components in RSLDP. The results showed that the compatibility of Radix Scutellaria and Licorice increased the quantity of components in vivo. We found that 106 potential targets of 64 active compounds in RSLDP were related to COVID-19.And 12 targets (MAPK3,IL6,JUN,IL2,STAT3, AKT1,TNF,STAT1,EGFR,HSP9AA1,VEGFA,RELA) could be core targets for RSLDP in treating COVID-19. Conclusions: The metabolism feature of RSLDP in vivo was systematically uncovered. The combining use of the T-NP method could discover potential drug targets and disclose the biological processes of RSLDP, which will clarify the potential mechanisms of RSLDP in the treatment of COVID-19.

26. Effect of Electroacupuncture on Reproductive Disorders and Insulin Resistance in a Murine Polycystic Ovary Syndrome Model

Polycystic ovarian syndrome (PCOS) is a common, complex, and heterogeneous endocrine and metabolic disorder. There is no standardized treatment, and it therefore requires individualized therapies according to the symptoms and pathogenesis of each patient. The present study aimed to determine the effect of electroacupuncture at the acupoints Zusanli (ST36), Sanyinjiao (SP6), and Neiguan (PC6) on reproductive disorders and insulin resistance in a murine model of PCOS induced by dehydroepiandrosterone (DHEA). Vaginal smear analysis was used to determine mice estrous cycle; intraperitoneal glucose and insulin tolerance tests were adopted to analyze metabolic characteristics; enzyme-linked immunosorbent assay was used to measure hormone levels; gene expression was quantified with real-time PCR; hematoxylin and eosin staining was used to observe ovarian morphology. We observed disordered estrous cycle, polycystic ovarian morphology, and higher levels of homeostasis model assessment-insulin resistance (HOMA-IR) and testosterone (T), indicating successful modeling of PCOS. DHEA increased levels of estrogen (E2), progesterone (P), testosterone (T), luteinizing hormone (LH), and follicle-stimulating hormone (FSH), and EA treatment restored them to levels seen in the control group. EA reduced the days in estrus caused by DHEA, improved the abnormal sex hormone receptor genes, and attenuated the DHEA-induced histomorphological changes in mouse ovaries. The average expressions of the androgen receptor (AR), estrogen receptor (ER), luteinizing hormone receptor (LHR), and follicle-stimulating hormone receptor (FSHR) genes in the ovary greatly increased after DHEA treatment and significantly decreased in the DHEA + EA group. After EA treatment, the cystic follicle (CF) number was reduced and corpora lutea (CL) increased in the DHEA + EA group compared to the DHEA group. EA improved glucose intolerance and insulin intolerance. Statistical analysis of intraperitoneal glucose tolerance test-area under curve (IPGTT-AUC) glucose levels revealed a significant decrease in DHEA group mice compared to the control and DHEA + EA groups. EA was found to restore fasting blood glucose, fasting serum insulin, and HOMA-IR. In summary, our study suggests that EA has a remarkable effect in the DHEA-induced murine PCOS model. Management of EA could improve estrous cycle, hormonal disorders, abnormal sex hormone receptors in ovaries, ovary morphology, and insulin resistance in PCOS mice.

This study investigates the consensus problem of second-order nonlinear multi-agent systems (MASs) with actuator faults via a sliding mode control approach. The consensus error dynamic is given based on the relative states of the neighbors. Then, a sliding mode surface based on consensus errors is proposed, and the asymptotic stability of the sliding mode is proved using the Lyapunov theory. Furthermore, a sliding-mode fault-tolerant consensus protocol is proposed to compensate for actuator faults. According to the sliding mode control theory, the proposed sliding-mode fault-tolerant controller ensures that the consensus of the MASs can be reached in a finite time. This paper uses wavelet neurons instead of traditional neurons, and uses wavelet multi-resolution analysis to decompose the FPGA image scan of the ship engine. Because the neural network has the approximation ability of arbitrary functions, the wavelet transform is connected to the neural network to form a wavelet neural network. To test ship engines. The hardware design of GigE image acquisition and processing system based on FPGA was started. FPGA was used as the main control chip and Gigabit Ethernet was used as the transmission medium. The hardware circuit of the image data acquisition and image processing system was designed. It mainly includes the FPGA main control circuit and the FPGA Peripheral circuits. The high-speed image acquisition, transmission, storage, and display module circuit design is realized. Real-time monitoring and fault analysis of the engine's condition is performed by the FPGA image scanning method, and data of the engine's running state is pre-processed with the help of step tracking technology to make it a standard signal. The data is transmitted to the computer through NI's data acquisition card. Combining feature extraction such as information entropy, Fourier transform, EMD and wavelet neural network technology. The accuracy of the diagnosis results and the actual fault state is improved. It can enable the staff to monitor the running status of the engine in real time, improve the efficiency of engine fault diagnosis, reduce labour costs and maintenance costs, and thus realize intelligent, real-time and accurate status monitoring of the engine.

29. Second-order Stokes wave-induced dynamic response and instantaneous liquefaction in a transversely isotropic and multilayered poroelastic seabed

The ocean waves exhibit obvious non-linearity with asymmetric distribution of wave crests and troughs, which could induce significantly different effect on the seabed compared to the commonly used linear wave theory. In this paper, a semi-analytical solution for a transversely isotropic and multilayered poroelastic seabed under non-linear ocean wave is proposed by virtue of the dual variable and position (DVP) method. The ocean wave and seabed are, respectively, modelled using second-order Stokes theory and Biot’s complete poroelastodynamic theory. Then the established governing equations are decoupled and solved via the powerful scalar potential functions. Making use of the DVP scheme, the layered solutions are finally gained by combining the boundary conditions of the seabed. The developed solutions are verified by comparing with existing solutions. The selected numerical examples are presented to investigate the effect of main parameters on the dynamic response of the seabed and evaluate the corresponding liquefaction potential. The results show that the anisotropic stiffness and permeability, degree of saturation and stratification have remarkable influence on the dynamic response and liquefaction behavior of the seabed. The present solution is a useful tool to estimate the stability of transversely isotropic and layered seabed sediment in the range of non-linear ocean wave.

In the remote sensing field, synthetic aperture radar (SAR) is a type of active microwave imaging sensor working in all-weather and all-day conditions, providing high-resolution SAR images of objects such as marine ships. Detection and instance segmentation of marine ships in SAR images has become an important question in remote sensing, but current deep learning models cannot accurately quantify marine ships because of the multi-scale property of marine ships in SAR images. In this paper, we propose a multi-scale feature pyramid network (MS-FPN) to achieve the simultaneous detection and instance segmentation of marine ships in SAR images. The proposed MS-FPN model uses a pyramid structure, and it is mainly composed of two proposed modules, namely the atrous convolutional pyramid (ACP) module and the multi-scale attention mechanism (MSAM) module. The ACP module is designed to extract both the shallow and deep feature maps, and these multi-scale feature maps are crucial for the description of multi-scale marine ships, especially the small ones. The MSAM module is designed to adaptively learn and select important feature maps obtained from different scales, leading to improved detection and segmentation accuracy. Quantitative comparison of the proposed MS-FPN model with several classical and recently developed deep learning models, using the high-resolution SAR images dataset (HRSID) that contains multi-scale marine ship SAR images, demonstrated the superior performance of MS-FPN over other models.

The aim of this study was to establish a method for free vancomycin concentration determination in human plasma and apply it to clinical therapeutic drug monitoring (TDM). The unbound vancomycin in plasma was separated by the hollow fiber centrifugal ultrafiltration (HFCF-UF) technique and analyzed by HPLC. Chromatographic conditions were optimized, the specificity, linearity, precision, recovery and stability of the method were examined, and plasma samples of patients were measured. The standard curve for free vancomycin is y = 0.0277x - 0.0080 with good linearity within 0.25-50 μg·mL

33. Classifying directed and diffusive transport in short, noisy single-molecule trajectories with wMSD

- Abstract
Fluorescence imaging in combination with single-particle tracking analysis has emerged as a powerful tool to study and characterize the motion of proteins moving in biological media. One of the main challenges in this approach is to reliably distinguish between directed and diffusive transport, especially for short and often noisy trajectories showing distinct, time- and place-dependent modes of motility. In this contribution, we present a windowed Mean-Square Displacements classifier (wMSDc) that is able to reliably (i) identify periods of diffusive and directed transport, (ii) extract position-dependent diffusion coefficients and velocities, and (iii) identify the location of switches in direction or motility mode in short (< 50 time points) and noisy single-molecule trajectories. We compare the performance of this approach to a Hidden Markov Model (HMM) method and a Moment Scaling Spectrum based method (DC-MSS) previously published and show that, in most cases, its performance is superior. We present a wide range of applications: from the movement of whole organisms and cells to protein-DNA interactionsin vitroand motor-protein dynamicsin vivo.Statement of SignificanceExtracting quantitative parameters from single-particle trajectories is a challenging task, especially in biological samples, which often show a high degree of heterogeneity. Trajectories can reveal switches between different types of motion; directed, diffusive and sub-diffusive motion. Usually, the length of these trajectories and their localization precision are limited by the experimental conditions. Here, we present a novel approach to analyse single molecule trajectories, windowed Mean-Square Displacement classifier (wMSDc) to reliably distinguish between directed and diffusive transport in the short trajectories with a finite precision of localization and integration time typically obtained when imaging single fluorescent proteins in living cells or organisms. We show that, using simulated and a wide range of experimental trajectories, wMSDc is a reliable method to extract motility parameters such as diffusion coefficient and velocity.

35. Improving the tensile strength of non-keyhole friction stir lap welding joint of 2024-T4 Al alloy by radial basis function neural network and improved particle swarm optimization algorithm

The non-keyhole friction stir lap welding (N-KFSLW) technology assisted by the outer stationary shoulder and the inner upper half-thread rotating pin was proposed to obtain the welding joint without keyhole through one-time process. Choosing 2024 aluminum alloys as the research object, the formation, microhardness and tensile strength of N-KFSLW joint were investigated. The improved particle swarm optimization (IPSO) algorithm was newly developed and had the advantages of large convergence speed and strong search ability, by which the radial basis function (RBF) neural network was optimized to enhance its prediction accuracy. After that, the RBF and IPSO (IPSO-RBF) system was used to predict the joint strength and optimize the process parameters combination. The results showed that the lap joint had not only the SZ with the thickness almost equal to the thickness of upper sheet but also the cold lap with a very small height, thereby leading to the high tensile strength of joint. The optimized parameters of welding speed, rotating speed and pin type by the IPSO-RBF system were respectively 612 rpm, 80 mm/min, and upper half-thread pin, and the tensile strength of lap joint reached 11.88 kN/mm. The N-KFSLW technology assisted by upper half-thread pin provides an effective way to obtain the lap joint with high performance, and the IPSO-RBF system can be used to maximize the strength of welding joint.

Nanozymes have potential applications in many fields, and a novel copper-containing nanozyme with highly dispersity and uniformity was self-assembled for efficient degradation of various organic dyes in this work. In the nanozyme, histidine was used to coordinate with copper ions, and hydrogen peroxide was prone to Fenton-like reaction to generate hydroxylated copper oxide intermediates. The nanozyme showed good peroxidase-like activity, and also had the ability to catalyze the degradation of various organic dyes efficiently with good storage and recycling ability. Furthermore, the degradation kinetics and mechanism of nanozyme had been further studied, and found that hydroxyl radical and singlet oxygen play vital roles in the catalytic degradation process. Meanwhile, this nanozyme can efficiently degrade two organic compounds at the same time, and this system is capable of dealing with complex practical application scenarios where wastewater contains a variety of organic pollutants.

This study aimed to develop a novel pH-sensing biopolymer film based on red cabbage puree (RCP) incorporated with polyvinyl alcohol (PVA), which was utilized for monitoring fish freshness during storage at 25 °C. A homogenized RCP suspension with a mean particle size of 12.86 ± 0.03 μm and a total anthocyanin concentration of 292.17 ± 2.65 mg/L was directly used as a film-forming substance and anthocyanin source to blend with PVA, showing visual changes in color and ultraviolet-visible spectra within a pH of 2–12. Rheological and microstructural studies certified the strong interactions and good compatibility between the RCP and PVA, resulting in better mechanical properties and water resistance of the composite film than those of a pure RCP film, but without affecting its pH sensitivity. When used for fish freshness monitoring at 25 °C, the developed RCP/PVA film presented visible color differences from purple to yellow, which corresponded to the spoilage threshold of the total volatile basic nitrogen and the total viable count in fish samples. The study highlights that anthocyanin-rich purees of fruits and vegetables, in this case red cabbage puree, can be fully utilized to develop eco-friendly pH-sensing indicator films for intelligent food packaging.

BackgroundThe duration of initial corticosteroid therapy in newly diagnosed Idiopathic nephrotic syndrome (INS) is about 3 months. Our study was designed to test the feasibility of a shorter duration of corticosteroid therapy in newly diagnosed INS who show a quicker response.MethodsPatients who responded within 10 days (Group A) received 8 weeks of corticosteroid therapy as compared to 12–14 weeks of standard therapy in those who responded between >10 days to 28 days (Group B), and follow up for 52 weeks. The primary endpoint is time to first relapse after treatment completion. (NCT03878914, March 18, 2019)ResultsA total of 33 children with INS were enrolled and the follow-up data were analyzed. The clinical and laboratory characteristics of patients in both groups were similar. No significant difference was found in time to first relapse [65(14.5, 159) days for Group A vs. 28(17, 61.5) days for Group B, P = 0.371], the incidence of frequently relapsing nephrotic syndrome [6/18 (33.3%) vs. 5/10(50%), P = 0.644] or requirement for alternative immunosuppressant [4/18 (22.2%) vs. 1/10 (10%), P = 0.769]. Group A received similar corticosteroid dose compare with Group B (3511 ± 2421 mg/m2 vs. 4117 ± 2556 mg/m2, P = 0.524). Frequency and severity of corticosteroid-related complications was similar in both groups.ConclusionsThe time to first relapse and the number of relapses per patient were comparable between the two groups. However, more patients in Group A relapsed and the mean total dose of prednisolone for the study period was very similar between the two groups.

Our previous study extracted and identified an antibacterial peptide that was named NP-6. Herein, we investigated the physicochemical properties of NP-6, and elucidated the mechanisms underlying its antimicrobial activity against Staphylococcus aureus. The results showed that the hemolysis activity of NP-6 was 2.39 ± 0.13%, lower than Nisin A (3.91 ± 0.43%) at the same concentration (512 µg/mL). Negligible cytotoxicity towards RAW264.7 cells was found when the concentration of NP-6 was lower than 512 µg/mL. In addition, it could keep most of its activity in fetal bovine serum. Moreover, transmission electron microscopy, confocal laser scanning microscopy, and flow cytometry results showed that NP-6 can destroy the integrity of the bacterial cell membrane and increase the membrane permeability. Meanwhile, NP-6 had binding activity with bacterial DNA and RNA in vitro and strongly inhibited the intracellular β-galactosidase activity of S. aureus. Our findings suggest that NP-6 could be a promising candidate against S. aureus.

To evaluate the effect of imrecoxib on CYP2C11 enzyme activity, mRNA, and protein expression, a UPLC method was established. Tolbutamide was selected as the CYP2C11 enzyme-specific probe drug and incubated with imrecoxib in rat liver microsomes. The yield of 4-hydroxytolbutamide was measured using UPLC to investigate the effect of imrecoxib on CYP2C11 enzyme activity. Imrecoxib (10 mg/kg) was administered intragastrically twice daily. After 1, 7, and 14 days of administration, the liver tissues were analyzed. The expression of CYP2C11 enzyme mRNA was determined using reverse transcription-polymerase chain reaction, and its protein expression was determined using Western blot analysis. The compression creep behavior of 8030 aluminum alloys at intermediate temperature was studied under the deformation temperature of 200∼250 °C and compression stress ranging from 20 to 40 MPa. The apparent stress exponent (na), the true stress exponent (nt) and the activation energy Qc for creep were na = 3.6, nt = 3 and Qc = 145.5 kJ/mol, respectively, indicating that the dislocation viscous glide controlled by lattice self-diffusion was the dominant creep mechanism of the samples at 200 °C-20 MPa. A creep constitutive equation was introduced with the threshold stress σ0 = 5.1 MPa. The microstructure characterization of selected samples was investigated by transmission electron microscopy (TEM), indicating that the sub-grain contours became ambiguous and the dislocations with long curved morphologies were homogeneously distributed within grain inners after creep for 100h.
