This study aimed to investigate the dynamic changes in SARS-CoV-2-specific CD4+ T-cell subsets and neutralizing antibody after three consecutive doses of inactivated COVID-19 vaccines (BBIBP-CorV) among PLWH.Methods: The blood samples were collected from 165 PLWH, including 66 PLWH in the 3-month interval between the second and third dose (cohort 1) and 99 PLWH in the 5-month interval (cohort 2). Blood collection for immunogenicity analysis was performed at 1-month post-2nd vaccination, pre-3rd vaccination, and within 2-month post-3rd vaccination. Wilcoxon matched-pairs signed-rank test was applied to compare the SARS-CoV-2-specific CD4+ T cell subsets and neutralizing antibody level at different time points. The relationship among CD4+ T-cells, Tregs subpopulations and SARS-CoV-2-specific neutralizing antibody level were evaluated with Spearman non-parametric correlation test.Results: No serious adverse reactions were found among PLWH. After two-dose or three-dose inactivated COVID-19 vaccination, the absolute counts of CD4+ T-cells and Tregs subpopulations (CD4+CD25HighCD127Low Tregs, CD45RA+ rTregs and CD45RO+ eTregs) increased in two cohorts. Satisfactory SARS-CoV-2-specific neutralizing antibody responses to the third-dose vaccination were found in two cohorts, including significantly enhanced neutralizing antibody level and high neutralizing antibody seroconversion rate. In addition, SARS-CoV-2-specific neutralizing antibody level were positively associated with the absolute counts of CD4+ T-cells and Tregs subpopulations as well as the frequency of CD45RO+ eTregs in PLWH after three doses of vaccinations.Conclusion: The three doses of inactivated COVID-19 vaccination were both safe and effective to increase SARS-CoV-2-specific CD4+ T-cells and neutralizing antibody in two PLWH cohorts with different inoculation intervals.Keywords: CD4+ T-cells, neutralizing antibody, inactivated COVID-19 vaccines, PLWH

Silicosis is an occupational lung disease caused by inhaling silica dust. The disease is characterized by early lung inflammation and late irreversible pulmonary fibrosis. Here we report the effect of Baicalin, a main flavonoid compound from the roots of Chinese herbal medicine Huang Qin on silicosis in a rat model. Results showed Baicalin (50 or 100 mg/kg/day) can mitigate the silica-induced lung inflammation and reduce the harm of alveolar structure and the blue region of collagen fibers in rat lung at 28 days after administration. At the same time, Baicalin also diminished the level of interleukin-1β (IL-1β), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) and transforming growth factor-β1 (TGF-β1) in lung tissues. The protein expression of collagen I (Col-1), α-smooth muscle actin (α-SMA) and vimentin were down-regulated while E-cadherin (E-cad) was increased in Baicalin-treated rats. In addition, the Toll Like Receptor 4 (TLR4)/ nuclear factor kappaB (NF-κB) pathway was enabled at 28 days after silica infusion, and the treatment of Baicalin diminished the expression of TLR4 and NF-κB in the lungs of rat with silicosis. These results suggested that Baicalin inhibited the pulmonary inflammatory and fibrosis in a rat model of silicosis, which could be attributed to inhibition of the TLR4/NF-κB pathway.

The protein-protein interaction between neuronal nitric oxide syntheses (nNOS) and the carboxy-terminal PDZ ligand of nNOS (CAPON) is a potential target for the treatment of ischemic stroke. Our previous study had identified ZLc-002 as a promising lead compound for inhibiting nNOS-CAPON coupling. To find better neuroprotective agents disrupting the ischemia-induced nNOS-CAPON interaction, a series of N-cyclohexylethyl-[A/G]-[D/E]-X-V peptides based on the carboxy-terminal tetrapeptide of CAPON was designed, synthesized, and evaluated in this study. Herein, we reported an affinity-based fluorescence polarization (FP) method using 5-carboxyfluorescein (5-FAM) labeled CAPON (496-506) peptide as the probe for high-throughput screening of the small-molecule inhibitors of the PDZ domain of nNOS. N-Cyclohexylethyl-ADAV displayed the most potent affinity for the nNOS PDZ domain in the FP and isothermal titration calorimetry (ITC) (ΔH = -1670 ± 151.0 cal/mol) assays. To improve bioavailability, lipophilicity, and membrane permeability, the Asp methylation was employed to get N-cyclohexylethyl-AD(OMe)AV, which possesses good blood-brain barrier (BBB) permeability in vitro parallel artificial membrane permeability assay (PAMPA)-BBB (Pe = 6.07 cm/s) and in vivo assays. In addition, N-cyclohexylethyl-AD(OMe)AV (10 mg/kg body weight, i.v., immediately after reperfusion) substantially reduced infarct size in rats, which was measured 24 h after reperfusion and subjected to 120 min of middle cerebral artery occlusion (MCAO).

To improve the decreased recognition performance for truck license plate in complex mine environment, which is caused by such factors as polluted, damaged, a robust license plate recognition method is proposed. Firstly, several candidate locations of license plate in the truck image are obtained by utilizing features of both edge and color. Furthermore, the parameter of license plate area intersection ratio is defined to find the precise localization among those above candidates. Then, a character segmentation scheme based on the gray projection method and morphological operator is used to remove those interference factors, such as frame, rivet and stain and uneven illumination. Finally, to increase the robustness of characters recognition by CNN, a self-built license plate character dataset is constructed, which contains various polluted samples obtained by manual simulation. As shown by the experimental results, the proposed method has higher accuracy in license plate location, and obtains an improvement on character recognition accuracy by 4% compared to other existed methods.

Tumor cells alter their protein expression profile to survive from the tumor metastasis. Fibronectin 1 (FN1 gene coding protein) is a member of the glycoprotein family that has been shown to play an important role in cancer metastasis. However, its effects on melanoma metastasis are still unclear.Methods:We detected the FN1 expression between metastatic cells and primary cells by using Western blot and RT-qPCR assays. And, we analyzed the expressed feature of FN1 in different tissues and examined the clinical relevance of upregulated FN1 in melanoma progression by bioinformatic analysis. Furthermore, we downregulated the expression of FN1 by small interfering RNA technique to reveal the effect of FN1 on melanoma phenotype and expression of related genes. Finally, we used bioinformatics to reveal the possible mechanism of FN1 regulating melanoma progression.Results:We reported that the expression of FN1 was changed during melanoma metastasis. In this study, we established two metastatic cell lines of melanoma through mouse model, and found that metastatic cells exhibited stronger mesenchyme phenotype and possessed higher FN1 expression level compared to primary cells. Besides, we examined the clinical relevance of upregulated FN1 in tumor progression. Small interfering RNA (siRNA)-mediated downregulation of FN1 suppressed the migration, invasion, adhesion, proliferation capabilities and induced apoptosis of melanoma cells. We detected a diminished EMT-related gene signature including increased expression of E-cadherin and decreased expression of N-cadherin and Vimentin. Downregulation of FN1 also increased Bax/Bcl-2 ratio which might result in apoptosis of melanoma cells. Bioinformatics analysis revealed that FN1 most likely involved in focal adhesion and PI3K-Akt signaling pathway to regulate EMT process and apoptosis.Conclusions: Taken together, these findings demonstrated a role of FN1 in promoting melanoma metastasis by inhibiting apoptosis and regulating EMT.Keywords: cancer metastasis, epithelial-mesenchymal transition, survival protein, migration, invasion

This paper aims at the fuel cell and battery hybrid automobile, based on one bus parameters, considers their own characteristics of fuel cell and battery and power demand when automobiles start, accelerate, climb, brake and other different working conditions, calculate the hybrid bus system parameters that match the fuel cell/battery., and ADVISOR is used is to verify simulation. The results show that the parameters of power drive system of this electric automobile are reasonable, and can meet the requirements of dynamic design indexes.

The overall performance of 5G mobile communication network has made great progress compared with 4G network. In order to ensure the expected utility of 5G communication system in practical application, this paper will focus on the future development trend of this communication system and its research on key technologies involved in the application process to provide theoretical support to relevant practitioners.

This paper briefly introduces some common short-region wireless communication technologies, comprehensively compares the application characteristics of each technology, and summarizes the application prospect of these technologies in equipment support.

For the design of air core reactors, the inductance calculation is an important technical parameter. Because air core reactors usually work in the three-phase condition, in this paper, the calculation method of the inductance of three-phase air reactors is obtained by using the method of equal division. Firstly, the magnetic flux, the induced potential and the self inductance and mutual inductance of the single-phase air reactor can be calculated accurately by using the method of equal division, Then, the division of different methods was taken according to the structure characteristics of three-phase reactors, and the calculation method of the mutual inductance between coils can be worked out for the three-phase air reactor in the different arrangements. This method is high accurate, simple and practical. The calculation example was given.

Screw stepping motors are widely used in many high-precision fields including manufacturing, precision calibration, and precision positional movement. Hence, research on the precise control of the screw stepping motor has important value in engineering application. In this paper, an ultra-low-power 16-bit MSP430F437 single chip microcomputer is used as the processor, combined with the subdivision drive system technology; the host computer is programmed with C#, and the communication between the host computer and the single chip microcomputer is established through Bluetooth, and a human-computer interaction interface is well established; the host computer sends a command to the single chip microcomputer to drive the screw stepping motor so as to control the speed and displacement of the load motion driven by the screw stepping motor.

This paper reported the visible light decoloration rate of the solution, mixed methylene blue with the lixivium obtained in removing iron from quartz sand by ultrasonic treatment. The effect of the concentration of ethanedionic acid, time of treatment by ultrasound, the initial concentration of methylene blue and the volume of lixivium were investigated in details. The primary result showed that it was apparent improved in decoloration efficiency after mixed. And the decoloration rate improved more highly after visible light irradiation. The decoloration rate of mixed solution containing 20 ppm methylene blue got to 99.5% under the condition of c(H2C2O4) 5g/L, treating by ultrasonic for 2.5h, using 30mL lixivium. It also had certain affection for the CODCr of lixivium. This process realized the multipurpose utilization of liquid waste. Ontology heterogeneity is the primary obstacle for interoperation of ontologies. Ontology mapping is the best way to solve this problem. The key of ontology mapping is the similarity computation. At present, the method of similarity computation is imperfect. And the computation quantity is high. To solve these problems, an ontology-mapping framework with a kind of hybrid architecture is put forward, with an improvement in the method of similarity computation. Different areas have different local ontologies. Two ontologies are taken as examples, to explain the specific mapping framework and improved method of similarity computation. These two ontologies are about classes and teachers in a university. The experimental results show that using this framework and improved method can increase the accuracy of computation to a certain extent. Otherwise, the quantity of computation can be decreased.

The aim of the present study was to investigate the effect of Chinese soft-shelled turtle whole egg powder (TE) on cholesterol metabolism in Sprague–Dawley rats to determine whether it has a cholesterol-lowering effect. Forty male Sprague–Dawley rats were fed a high-fat diet supplemented with TE (0, 0·75, 1·50 or 3·00 g/kg body weight) administrated by gavage for 24 weeks. Serum total cholesterol (TC), HDL-cholesterol (HDL-C), LDL-cholesterol (LDL-C) and faecal total bile acids levels were determined by enzymatic methods. Faecal steroid concentrations were measured by GC. Means and standard deviations were calculated where appropriate for values, and the data were tested by one-way ANOVA. After 24 weeks of feeding a high-fat diet with TE supplementation, serum TC and LDL-C levels, liver cholesterol and liver lipid levels were reduced in rats. TE supplementation did not affect the faecal output, but significantly increased steroid concentrations in faeces, indicating increased steroids excretion. The faecal bile acid excretion was also increased as evidence by elevated mRNA level of liver cytochrome P450, family 7, subfamily A, polypeptide 1. Our results demonstrated that the TE does have a cholesterol-lowering effect by increasing the excretion of total bile acids and neutral steroids.

Based on the Monte Carlo simulation conjoined with the embedded atom method (EAM) potentials, a double species model is employed to study the Ni75-3x/4Al25-x/4Crx (x < 3) structure, i.e. impure Ni3Al together with a grain boundary structure at the equilibrium. At the grain boundary, the model shows that Ll(2)-Ni3Al transforms into fcc-Ni when x increases from 0 to 0.40; then fcc-Ni transforms into Ll(2)-Ni3Cr when x increases from 0.40 to 2.40.

The analysis of the colored pottery figurines from Yangling Tombs of the Han Dynasty was realized by X-ray diffraction, emission spectroscopy, infrared spectroscopy, X-ray fluorescence and fiber optics reflectance spectroscopy (FORS). Consequent results showed that the colored components were respectively mercuric(II) sulphide, ferric (III) oxide, and carbon, among which mercuric(II) sulphide was an artificial pigment.

Monte Carlo simulation is used to study the low energy He ion challenging in a (17,0) single-wall nanotube and its rope. The simulation shows that the channelling critical angle Psi(C) = 48E(-1/2) (E is incident energy) for a 1.31-nm-diameter initial beam into the nanotube, while Psi(C) = 45E(-1/2) for the 1.31-nm-diameter initial beam into its rope.

Under 2 keV Xe ion bombardment of a 63Cu69.165Cu30.9 isotope mixture, isotope fractionation difference between small and large polar emission angles, δ(15°)-δ(75°), was calculated by use of a dynamic Monte Carlo program. Calculation results show that δ(15°) - δ(75°) is large at both the zero fluence and the high fluence, then it minimizes at a low fluence of 1.6×1015 ions/cm2 between two fluence limits. An important cause of this minimum is that the surface composition gradient of 63Cu maximizes at the fluence of 1.6×1015 ions/cm2.

Cannon barrel life is an important tactical characteristic of the cannon to maintain combat capability, and the inside diameter wear of the cannon barrel is one of the most important appraising standards of cannon barrel life. In this paper, a cannon barrel inner diameter gauge is designed based on the CCD imaging and computer image processing technique. The main idea of design and the working procedure and the design principle of the main components of the inner diameter gauge are introduced, in the end the various measurement errors are analyzed.

Aurora protocol of Xilinx Company is one that can be upgraded, small-scale chain layer protocol, can be used for the high speed serial data transmission of point to point. The paper mainly introduces relevant knowledge of chain layer of Aurora, and explains how to make use of RocketIO in Virtex-II Pro to realize Aurora protocol.

Four groups of mice were dissected at various time intervals from 0.5 to 144 hours after their intravenous injection of uranium. The uranium distribution and deposition in the tissues of kidney, lung, marrow, liver, heart, muscle and blood have been studied using the fission track technique (FTT). Our experimental results show that trace uranium is more likely to be deposited in kidneys and lungs than in marrow and liver. The presence of uranium in the blood is so transient that blood-uranium level cannot be used to estimate the degree of the storage of uranium in the body. However, the mice injected with high dose uranium die soon because the uranium poisons the heart severely.

The modified dynamic TCIS (Transport and atomic Collision cascade of energetic Ions in Solids) Monte Carlo program including the radiation-enhanced Gibbsian segregation (REGS) is employed to estimate the compositional distributions at surface of the Au0.56Cu0.44 alloy under 2ke V Ar+ ion bombardment at high dose. The calculation results exhibit that the Au composition enriches in the first layer (74 at.%), depletes in the second layer (37 at.%) and gradually rises with depth to the bulk composition. The effect of the relation between sputter depth and segregation layer on the surface concentration of Au0.56Cu0.44 is also discussed.

The recognition system of license-plate can improve the traffic situation. Sometimes the image of license-plate isn't clear, and some characters are difficult to identify. By image fusion, the clarity of license-plate can be improved. Because the ratio of standard character is unchanging, it is used to identify characters. First obtaining character from image of license-plate with segmentation technique, then match it with standard character. At last, characters can be identified. According to these ideas, a complete recognition system can be built

Channeling phenomena of He, Ne, Ar and Kr ions at energy (200–5000 eV) in single-wall carbon nanotubes (SWCNTs) are investigated by molecular dynamics simulation with analytical potentials. The critical angles for the particles to be channeled in an SWCNT are analyzed. In the incident energy range of 200–5000 eV, it is found that the ion energy dependence of the critical angle obeys an improved Lindhard equation which is closely related to the ratio of nuclear charge number to atomic mass Z/M. The critical angle for different types of ions channeling in SWCNTs is determined by both the atomic nuclear charge and mass.

The low energy sputtering angular distributions are simulated in inert gas ion-metal target combinations. It is found that the angular distribution of PKAs trends to an under-cosine shape, while the angular distribution of SKAs shows either a cosine one or an over-cosine one; the formation of the under-cosine sputtering angular distribution seems to come from the smaller mass ratio and the lower incident energy.

A series of novel danshensu amide derivatives were synthesized, and the protective effects of all the compounds on rat myocardial cell lines H9C2 by hypoxia were investigated. The results showed that all the seven compounds could significantly increased cell viability compared with hypoxia group. Among these compounds, 3-(3,4-dimethoxyphenyl)-2-hydroxy-N-propylpropanamide (6) exhibited good activities, with cell viability reached 94.2 % compared to the normal. A Monte Carlo simulation is used to study the angular distribution of sputtered atoms from Fe and Lu targets bombarded by Ne, Ar and Kr ions at energies of 0.2 keV and 0.5 keV. The sputtered atoms are distinguished as primary knock-on atoms (PKA) and secondary knock-on atoms (SKA) in the simulation. It is found that the angular distribution of the PKAs tends to an under-cosine shape, while the angular distribution of the SKAs shows either a cosine shape or an over-cosine shape. The calculations also give the contributions of the PKAs and the SKAs to sputtering yields. Based on the computations, the mechanism of the under-cosine angular ejection distribution at low energy sputtering is discussed.
