This paper proposes a strategy for the facile preparation of NiS nanowires via ion exchange reaction. This special synthesis process enables NiS nanowires to form a tightly connected network of transported electrons and a hierarchical porous structure with a large specific surface area.

Cilia are ubiquitous eukaryotic organelles responsible for cellular motility and sensory functions. The ciliary axoneme is a microtubule-based cytoskeleton consisting of two central singlets and nine outer doublet microtubules. Cryo-electron microscopy-based studies have revealed a complex network inside the lumen of both tubules composed of microtubule-inner proteins (MIPs). However, the functions of most MIPs remain unknown. Here, we present single-particle cryo-EM-based analyses of the Tetrahymena thermophila native doublet microtubule and identify 42 MIPs. These data shed light on the evolutionarily conserved and diversified roles of MIPs. In addition, we identified MIPs potentially responsible for the assembly and stability of the doublet outer junction. Knockout of the evolutionarily conserved outer junction component CFAP77 moderately diminishes Tetrahymena swimming speed and beat frequency, indicating the important role of CFAP77 and outer junction stability in cilia beating generation and/or regulation.

C/EBPα is an essential transcription factor involved in regulating the expression or function of certain cell-cycle regulators, including in breast cancer cells. Although protein arginine methyltransferases have been shown to play oncogenic roles in a variety of cancers, little is known about the role of arginine methylation in regulating the antiproliferation activity of C/EBPα. Here, we report that the protein arginine methyltransferase 1 (PRMT1) is overexpressed in human breast cancer and that elevated PRMT1 correlates with cancer malignancy. RNA-sequencing analysis revealed that knockdown of PRMT1 in breast cancer cells is accompanied by a decrease in the expression of pro-proliferative genes, including cyclin D1. Furthermore, tandem affinity purification followed by mass spectrometry identified PRMT1 as a component of the C/EBPα complex. C/EBPα associated with and was methylated by PRMT1 at three arginine residues (R35, R156, and R165). PRMT1-dependent methylation of C/EBPα promoted the expression of cyclin D1 by blocking the interaction between C/EBPα and its corepressor HDAC3, which resulted in rapid growth of tumor cells during the pathogenesis of breast cancer. Inhibition of PRMT1 significantly impeded the growth of cancer cells from patients with triple-negative breast cancer. This evidence that PRMT1 mediates C/EBPα methylation sheds light on a novel pathway and potential therapeutic target in breast cancer.Significance:This study provides novel mechanistic insight of the role of the arginine methyltransferase PRMT1 in breast cancer pathogenesis.

Redox electrolytes for supercapacitors (SCs) have recently sparked widespread interest. Due to the redox reactions within electrolytes, they can achieve high capacitance and long cycle stability. However, the energy density of SCs with redox electrolytes is limited by the narrow applied electrochemical window due to the irreversible side reaction of redox mediators at high potential. To overcome this issue, a redox mediator with a high redox potential, tetrachloridehydroquinone (TCHQ), is added to organic electrolytes to obtain a broad electrochemical window. TCHQ is designed to undergo a dehydrogenation reaction catalyzed by N-doped activated carbon to provide capacitance. The pyrrole N atoms have the highest electrocatalytic activity based on the theoretical calculation of reaction overpotential with predicted reaction pathways due to their Lewis basicity. Benefitting from that, TCHQ shows promising reversibility with a larger electrochemical window (up to 2.7 V). As a result, a higher energy density is obtained when compared to commercial SCs. This study proposes a strategy for designing redox mediators and interfaces of SCs with high energy density and a calculation method of dehydrogenation reaction electrocatalysis.

Lithium-sulfur battery (LSB) has high energy density but is limited by the polysulfides shuttle and dendrite growth during cycling. Herein, a free-standing cellulose nanofiber (CNF) separator is designed and fabricated in isopropanol/water suspension through vacuum filtration progress. CNFs with abundant polar oxygen-containing functional groups can chemically immobilize the polysulfides, and suppress the formation of the dendrites by controlling the surface morphology of the SEI on lithium metal in LSB. The isopropanol content in a suspension can fine-tune the pore structure of the membrane to achieve optimal electrochemical performance. The prepared separator displays integrated advantages of an ultrathin thickness (19 μm), lightweight (0.87 mg cm–2), extremely high porosity (98.05%), and decent electrolyte affinity. As a result, the discharge capacity of the LSB with CNF separator at the first and 100th cycle is 1.4 and 1.3 times that of PP separator, respectively. Our research provides an environmental-friendly and facile strategy for the preparation of multifunctional separators for LSBs.

Compounds 5 and 7 exhibited nitric oxide release inhibitory activities with IC50 values of 7.5 and 10.6 μM, respectively, and compounds 9 and 14 at the sub-MIC level significantly decreased the MIC of oxacillin against MRSA from 32.0 to 1.0 and 0.5 μg mL−1, respectively.

Background Bacterial viruses, phages, play a key role in nutrient turnover and lysis of bacteria in terrestrial ecosystems. While phages are abundant in soils, their effects on plant pathogens and rhizosphere bacterial communities are poorly understood. Here, we used metagenomics and direct experiments to causally test if differences in rhizosphere phage communities could explain variation in soil suppressiveness and bacterial wilt plant disease outcomes by plant-pathogenic Ralstonia solanacearum bacterium. Specifically, we tested two hypotheses: (1) that healthy plants are associated with stronger top-down pathogen control by R. solanacearum-specific phages (i.e. ‘primary phages’) and (2) that ‘secondary phages’ that target pathogen-inhibiting bacteria play a stronger role in diseased plant rhizosphere microbiomes by indirectly ‘helping’ the pathogen. Results Using a repeated sampling of tomato rhizosphere soil in the field, we show that healthy plants are associated with distinct phage communities that contain relatively higher abundances of R. solanacearum-specific phages that exert strong top-down pathogen density control. Moreover, ‘secondary phages’ that targeted pathogen-inhibiting bacteria were more abundant in the diseased plant microbiomes. The roles of R. solanacearum-specific and ‘secondary phages’ were directly validated in separate greenhouse experiments where we causally show that phages can reduce soil suppressiveness, both directly and indirectly, via top-down control of pathogen densities and by alleviating interference competition between pathogen-inhibiting bacteria and the pathogen. Conclusions Together, our findings demonstrate that soil suppressiveness, which is most often attributed to bacteria, could be driven by rhizosphere phage communities that regulate R. solanacearum densities and strength of interference competition with pathogen-suppressing bacteria. Rhizosphere phage communities are hence likely to be important in determining bacterial wilt disease outcomes and soil suppressiveness in agricultural fields.

Carbon materials are considered as the ideal electrode materials for supercapacitors due to their diverse structure and nature. However, their poor frequency response is an obstacle to their application in high-frequency supercapacitors. Herein, an ultra-high temperature graphitization process at 2800 °C is proposed to fabricate carbon nanotubes/graphene hybrid films that are successfully employed as the electrode materials of high-frequency supercapacitors. By rational hybridization, the carbon nanotubes/graphene interlinked networks offer fast ion transport paths. Importantly, via a graphitization process at 2800 °C, the as-obtained hybrid films exhibit an ultrahigh in-plane conductivity of 491.81 S cm−1 and favorable out-plane conductivity of 27.98 mS cm−1. Such design brings the as-constructed high-frequency supercapacitors an unprecedented phase angle (up to −56.23°) and area capacitance (up to 230.56 μF cm−2) at 120 Hz, and their cut-off frequency can be nearly 30 times higher than that of films carbonization at 1600 °C. Such increases, further supported by density functional theory (DFT) calculations, are partly attributable to enhancements of ion response arising from the repair of edge defects of graphene. These findings will provide a new method in designing the structure of carbon electrodes for enhancing SCs frequency response performances.

Five new polychlorinated bibenzyls (1–5) along with 3 known compounds (6–8) were isolated from the stems and leaves of Rhododendronminutiflorum. The chemical structures of all the isolates were determined by spectroscopic methods, and compounds 1 and 2 were further verified by single-crystal X-ray diffraction analyses. Compounds 1–5 were halogenated compounds which bear three to five chlorine atoms in their chemical structures. Biologically, compounds 2, 5 and 6 showed varying degrees of toxicity toward the Asian citrus psyllid (Diaphorinacitri) with LD50 values 27.15, 17.02 and 16.20 mg/L, respectively. These values were comparable to the positive control matrine (LD50 = 11.86 mg/L), which were calculated using observations on day 6. Meanwhile, compound 4 had α-glucosidase inhibitory activity with IC50 value of 17.87 ± 0.74 μM. Graphical Abstract

The incorporation of boron into carbon material can significantly enhance its capacity performances. However, the origin of the promotion effect of boron doping on electrochemical performances is still unclear, in part due to the inadequate exposure of boron configurations resulting from the complexity of traditional carbon materials. To overcome this issue, herein, a series of boron-doped graphene with highly-exposed boron configurations are prepared by tuning annealing temperature. Then the correlation between boron configurations and the electrochemical performances is investigated. The combination of density-functional theory (DFT) computation and NH3-TPD/Py-FTIR indicates that the BCO2 configuration formed on the surface of graphene is easier to accept lone-pair electrons than BC2O and BC3 configurations due to the stronger Lewis acidity. Such an electronic structure can effectively reduce the number of unstable electron donors and stabilize the electrochemical interface, which is proved by NMR, and critical for improving the electrochemical performances. Further experiments confirm that the optimized BG800 with the largest amount of BCO2 configuration presents ultralow leak current, improved cyclic stability, and better rate performance in SBPBF4/PC. This work would provide an insight into the design of high-performance boron-doped carbon materials towards energy storage.

Intestinal ischemia-reperfusion (II/R) injury is a common type of tissue and organ injury, secondary to intestinal and mesenteric vascular diseases. II/R is characterized by a high incidence rate and mortality. In the II/R process, intestinal barrier function is impaired and bacterial translocation leads to excessive reactive oxygen species, inflammatory cytokine release, and even apoptosis. A large number of inflammatory mediators and oxidative factors are released into the circulation, leading to severe systemic inflammation and multiple organ failure of the lung, liver, and kidney. Acute lung injury (ALI) is the most common complication, which gradually develops into acute respiratory distress syndrome and is the main cause of its high mortality. This review summarizes the signal transduction pathways and key molecules in the pathophysiological process of ALI induced by II/R injury and provides a new therapeutic basis for further exploration of the molecular mechanisms of ALI induced by II/R injury. In particular, this article will focus on the biomarkers involved in II/R-induced ALI.

Designing molecules with specific scaffolds can facilitate the discovery and optimization of lead compounds. Some scaffold-based molecular generation models have been developed using deep-learning methods based on specific scaffolds, although incorporating scaffold generalization is expected to achieve scaffold hopping. Moreover, most of the existing models focus on the 2D shape of the scaffold and overlook the stereochemical properties of the compound, especially for natural products. In this study, we optimized the scaffold-based molecular generation model designed by Lim et al. (Chemical Science 2020, 11, 1153-1164). Real-time ultrafast shape recognition with pharmacophore constraints (USRCAT) was introduced into the model to search for molecules similar to the 3D conformation and pharmacophore of the input scaffold sourced from the training set; the searched molecules were then used as new scaffolds to execute scaffold hopping. The optimized model could generate new molecules with the same chirality as the input scaffold. Furthermore, the probability distribution of the molecular structure and various physicochemical properties were analyzed to evaluate the model's generation capability. We thus believe that the optimized model can provide a basis for medicinal chemists to explore a wider chemical space toward optimization of the lead compounds and to screen the virtual compound library.

Colorectal cancer (CRC) is the third most common malignant tumor in the world. CRC has high morbidity and mortality rates and it is a serious threat to human health. Ferroptosis is a unique form of iron-dependent oxidative cell death that is usually accompanied by iron accumulation and lipid peroxidation. Ferroptosis has attracted worldwide attention since it was first proposed. It plays an important role in the development of a variety of diseases, such as tumors, ischemia/reperfusion injury, nervous system diseases, and kidney damage, and it may serve as a new therapeutic target. This article reviews the mechanism of ferroptosis and the possibility to target ferroptosis pathways in CRC, providing new ideas for the diagnosis and treatment of CRC.

Morphine, the most widely used analgesic, relieves severe pain by activating the μ-opioid receptor (MOR), whereas naloxone, with only slight structural changes compared to morphine, exhibits inhibitory effect, and is used to treat opioid abuse. The mechanism by which the MOR distinguishes between the two is unclear. Molecular dynamics (MD) simulations on a 1-μs time scale and metadynamics-enhanced conformational sampling are used here to determine the different interactions of these two ligands with MOR: morphine adjusted its pose by continuously flipping deeper into the pocket, whereas naloxone failed to penetrate deeper because its allyl group conflicts with several residues of MOR. The endogenous peptide ligand endomorphin-1 (EM-1) underwent almost no significant conformational changes during the MD simulations. To validate these processes, we employed GIRK4S143T, a MOR-activated Gβγ-protein effector, in combination with mutagenesis and electrophysiological recordings. We verified the role of some key residues in the dynamic recognition of naloxone and morphine and identified the key residue I322, which leads to differential recognition of morphine and naloxone while assisting EM-1 in activating MOR. Reducing the side chain size of I322 (MORI322A) transformed naloxone from an inhibitor directly into an agonist of MOR, and I322A also significantly attenuated the potency of MOR on EM-1, confirming that binding deep in the pocket is critical for the agonistic effect of MOR. This finding reveals a dynamic mechanism for the response of MOR to different ligands and provides a basis for the discovery of new ligands for MOR at the atomic level.

Humans are simultaneously exposed to numerous of environmental brominated flame retardants (BFRs). We aim to explore the overall associations of BFRs mixture on metabolic syndrome (MetS) and its components and further identify significant chemicals.This study included 4641 adults from the National Health and Nutrition Examination Survey (NHANES) in 2007-2016. The weighted logistic regression was conducted to estimate the association of a single BFR exposure with MetS and its components. Meanwhile, the weighted quantile sum (WQS) regression and Bayesian kernel machine regression (BKMR) were adopted to evaluate the overall associations of BFRs mixture on MetS and its components, and to identify significant chemicals. We also evaluated potential associations modified by sex.In the weighted logistic regression model, PBB153 were positively associated with MetS in a dose-dependent manner (PThe present study suggested exposure to BFRs mixture was positively associated with MetS and its components in adults, and PBB153, PBDE28 and PBDE209 were the significant chemicals. However, prospective cohort studies are still needed to confirm the causal effect between BFRs mixture and MetS.

Hashimoto's thyroiditis (HT) is the most common autoimmune disease involving the thyroid gland. HT often clinically manifest as hypothyroidism due to the destruction of thyroid cells mediated by humoral and cellular immunity. The pathogenesis of HT is a complex process in which environmental factors, hereditary inclination, trace elements immune factors, cytokines, and DNA and miRNA all play an important role. Herein, we summarize the precision factors involved in the pathogenesis of HT and offer an update over the past 5 years to provide a theoretical basis for further investigation of the relevant targets for HT treatment.

Combination therapy such as photodynamic therapy (PDT)-enhanced chemotherapy is regarded as a promising strategy for cancer treatment. Boron-dipyrromethene (BODIPY), as close relatives of porphyrins, was widely used in PDT. However, poor water solubility, rapid metabolism by the body and lack of targeting limits its clinical application. Lenvatinib, as the first-line drug for molecular-targeted therapy of liver cancer, restricted its clinical application for its side effects. Herein, to achieve the synergy between PDT and chemotherapy, we synthesized two halogenated BODIPY, BDPBr2 and BDPCl2, which were prepared into self-assembly nanoparticles with lenvatinib, and were encapsulated with Pluronic F127 through the nanoprecipitation method, namely, LBPNPs (LBBr2 NPs and LBCl2 NPs). The fluorescence quantum yields of LBPNPs were 0.73 and 0.71, respectively. The calculated loading rates of lenvatinib for LBBr2 NPs and LBCl2 NPs were 11.8 and 10.2%, respectively. LBPNPs can be hydrolyzed under weakly acidic conditions (pH 5.0) to generate reactive oxygen species (ROS), and the release rate of lenvatinib reached 88.5 and 82.4%. Additionally, LBPNPs can be effectively taken up by Hep3B and Huh7 liver cancer cells, releasing halogenated BODIPY and lenvatinib in the acidic environment of tumor cells to enhance the targeting performance of chemotherapeutics. Compared with free lenvatinib and separate halogenated BODIPY, LBPNPs can inhibit tumor growth more effectively through pH-responsive chemo/photodynamic synergistic therapy and significantly promote the cascade of caspase apoptotic protease. This study shows that LBPNPs can be a promising nanotheranostic agent for synergetic chemo/photodynamic liver cancer therapy.

A series of unsymmetrical 2-(2,4-bis(bis(4-methoxyphenyl)methyl)-6-MeC6H2N)-6-(1-(arylimino)ethyl)pyridine-iron halides has been synthesized and characterized. The molecular structure of two representative species was determined by the single-crystal X-ray diffraction. Activated with either MAO or MMAO, the precatalysts displayed high activity, reaching 2.19×107 g PE (mol Fe)−1 h−1 at 60 °C in ethylene polymerization. The microstructural analysis of the polymers obtained indicates highly linear polyethylene containing a vinyl chain end.

The SiC whiskers/hollow carbon microspheres (SiCw/HCMS) were successfully synthesized by a combination of the spray drying technology and the carbothermal reduction method. The anchoring of SiCw on HCMS shells improves the thermostability of composites and induces the formation of new heterointerfaces between SiC whiskers and HCMS. Experimental characterizations together with DFT calculations show that compared to intrinsic defects and functional groups in SiCw/HCMS, the formed heterointerfaces trigger the separation of positive and negative charges and induce large dipole moments, resulting in the intensive dielectric polarization loss. Consequently, SiCw/HCMS samples achieve an excellent electromagnetic wave absorption, where the reflection loss value of SiCw/HCMS-1450 reaches to −48.60 dB at 8.0 GHz with a thickness of 2.6 mm and the maximum effective absorbing bandwidth is 4.34 GHz. This work investigates a deep insight into the relationship between interfacial polarization loss and the electromagnetic wave attenuation and designs a novel absorbent that exhibits great potential for electromagnetic wave absorption at high-temperature environments.

High-entropy alloys (HEAs) are proposed as potential structural materials for advanced nuclear systems, but little is known about the response of matrix chemistry in HEAs upon irradiation. Here, we reveal a substantial change of matrix chemical concentration as a function of irradiation damage (depth) in equiatomic NiCoFeCr HEA irradiated by 3 MeV Ni ions. After ion irradiation, the matrix contains more Fe/Cr in depth shallower than ~900–1000 nm but more Ni/Co from ~900–1000 nm to the end of the ion-damaged region due to the preferential diffusion of vacancies through Fe/Cr. Preferential diffusion also facilitates migration of vacancies from high radiation damage region to low radiation damage region, leading to no void formation below ~900–1000 nm and void formation around the end of the ion-damaged region at a fluence of 5 × 1016 cm−2 (~123 dpa, displacements per atom, peak dose under full cascade mode). As voids grow significantly at an increased fluence (8 × 1016 cm−2, 196 dpa), the matrix concentration does not change dramatically due to new voids formed below ~900–1000 nm.

Objective To investigate the relationship between preoperative proteinuria and postoperative acute kidney injury (AKI). Methods We performed a search on databases included PubMed, Embase, the Cochrane Library, and Web of Science, from December 2009 to September 2020. Data extracted from eligible studies were synthesized to calculate the odds ratio (OR) and 95% confidence interval (CI). A fixed or random effects model was applied to calculate the pooled OR based on heterogeneity through the included studies. Results This meta-analysis of 11 observational studies included 203,987 participants, of whom 21,621 patients suffered from postoperative AKI and 182,366 patients did not suffer from postoperative AKI. Primary hepatocellular carcinoma (HCC) is one of the most frequently occurring pernicious tumors in the world. It is typically very insidious in the early stages with no obvious symptoms. Its development and metastasis are very rapid. Upon diagnosis, most patients have already reached a local advanced stage or have established distant metastases. The treatment of HCC is limited, with poor prognosis and short natural survival time. In order to improve the efficiency of early diagnosis, it is particularly significant to choose economic and effective diagnosis methods. Ultrasound, magnetic resonance imaging, and computed tomography are usually used in the clinic, but these methods are extremely limited in the diagnosis of HCC. Tumor markers have become the main effective early clinical diagnosis method. Potential serum tumor markers include alpha fetoprotein heterogeneity, Golgi protein 73, phosphatidylinositol proteoglycan (GPC-3), osteopontin, abnormal prothrombin, and heat shock protein. These tumor markers provide new ideas and methods for the diagnosis of HCC. A combination of multiple markers can make up for the deficiency of single marker detection and provide a new strategy for the prognosis and auxiliary diagnosis of HCC. This review introduces protein tumor markers utilized over the past five years.
