Controlled charge flows are fundamental to many areas of science and technology, serving as carriers of energy and information, as probes of material properties and dynamics, and as a means of revealing or even inducing broken symmetries. Emerging methods for light-based current control offer promising routes beyond the speed and adaptability limitations of conventional voltage-driven systems. However, optical manipulation of currents at nanometer spatial scales remains a basic challenge and a key step toward scalable optoelectronic systems and local probes. Here, we introduce vectorial optoelectronic metasurfaces as a new class of metamaterial in which ultrafast charge flows are driven by light pulses, with actively-tunable directionality and arbitrary patterning down to sub-diffractive nanometer scales. In the prototypical metasurfaces studied herein, asymmetric plasmonic nanoantennas locally induce directional, linear current responses within underlying graphene. Nanoscale unit cell symmetries are read out via polarization- and wavelength-sensitive currents and emitted terahertz (THz) radiation. Global vectorial current distributions are revealed by spatial mapping of the THz field polarization, also demonstrating the direct generation of elusive broadband THz vector beams. We show that a detailed interplay between electrodynamic, thermodynamic, and hydrodynamic degrees of freedom gives rise to these currents through rapidly-evolving nanoscale forces and charge flows under extreme spatial and temporal localization. These results set the stage for versatile patterning and optical control over nanoscale currents in materials diagnostics, nano-magnetism, microelectronics, and ultrafast information science.

In the field of aviation, the contrate gear is an important component, the material of the contrate gear is the basis for ensuring the reliability and high performance of the aircraft engine, the commonly used material for contrate gear is 20Cr2Ni4A. In order to fully understand the electrolytic machining of the toothed groove of the contrate gear, a multiphysics coupling model of the electric field, the gas-liquid two-phase flow field and the temperature field was established to study the distribution of bubble rate, temperature, conductivity and current density in the processing gap. Simulation results show that the temperature and bubble rate gradually increase along the process direction in the processing gap, the conductivity and current density of the electrolyte gradually decrease.

Hypertension is a common cardiovascular disease. Zhengan Xifeng Decoction (ZGXFD), a classic prescription for adjuvant treatment of hypertension, but its clinical application characteristics and biological information have not been comprehensively analysed. This study is based on real-world data from 7571 electronic medical records of hypertension patients treated by ZGXFD. The Apriori algorithm was used to obtain the coupled herbs of ZGXFD. A convolutional neural network was designed to measure the dose characteristic information of herbs. The topological features of the protein‒protein interaction data were used to analyse the biological information of herbs. The K-nearest neighbour model integrates the above characteristics of herbs into the same framework to observe the composition adjustment laws and mechanism of ZGXFD from multiple dimensions. Eighty-seven coupled herbs with dose characteristics were obtained. The results revealed that ZXGFD regulates cytokines and reduces the inflammatory response and metabolic disorder to achieve the purpose of adjuvant therapy. Moreover, machine learning model is used to analyze real-world data that include clinical and molecular biological data with hierarchical characteristics, which provides a micro-biological explanation for the clinical application of herbs.

There is currently no way for subway air-conditioning systems to adjust to changes in passenger capacity in terms of air-supply volume and air-supply temperature. In this paper, a study of the setting of the air-supply volume and controlling the temperature in subway trains was conducted. Based on the fact that the air-supply volume in a subway train needs to meet the requirements of cooling load and humidity load, as well as fresh-air volume, the minimum critical air-supply volume curve was determined on the basis of theoretical calculations under different passenger capacities. Under different passenger capacity and air-supply-volume conditions, the PMV (predicted mean vote) distribution of the subway train was simulated and calculated, and the results indicated that increasing the air-supply volume would not be sufficient to achieve PMV = 0. Therefore, in accordance with the theoretical formulation of the PMV, multiple linear regression was used to fit the temperature-control curve for various passenger capacities. A simulation calculation of the subway train was used to assess the uniformity of the flow field and the temperature field within the subway train in accordance with EN14750. It is evident from the results that under the control curve of the air-supply volume and temperature under different passenger capacities, the uniformity of the flow field and the temperature field can meet the standard requirements. By using the air-supply volume and temperature-control curve proposed in this paper, it is possible to adjust the air-supply volume in real-time, control the temperature of the air-conditioning system of the subway train, improve the passenger comfort of the train, and reduce the energy consumption of the air-conditioning system.

An open resonator antenna for orbital angular momentum (OAM) generation and dual-mode application is proposed. The excitation of OAM mode is deduced by the open resonator theory and the circular array theory. In the cavity, eight aperture coupled patch antenna elements are used to excite two higher-order Laguerre–Gaussian HE21 modes with a 90° phase difference. Verifying by simulation and experiment, the proposed low-profile antenna can radiate left-hand or right-hand circularly polarized waves of OAM modes $l=\pm 1$ , which have a relatively low divergence angle of ±9° and high gain of around 13 dBi.

Introduction Hepatitis B virus (HBV) infection is associated with the onset of several major liver diseases. Inactive hepatitis B surface antigen (HBsAg) carriers (IHCs) may be successfully treated with PEGylated interferon-α2b (PEG-IFNα2b)-based antiviral therapy; however, studies on this treatment have been insufficient. In this study, we evaluated the efficacy and safety of PEG-IFNα2b treatment in IHCs. Methods Nineteen IHCs were treated with subcutaneous PEG-IFNα2b (180 μg/week) for 48 weeks (treatment group). Background: Although we previously revealed that DNA-dependent protein kinase catalytic subunit (DNA-PKcs) is overexpressed in pancreatic ductal adenocarcinoma (PDAC) and important for gemcitabine resistance, the role of DNA-PKcs in the progression and metastasis of PDAC remain unclear. To date, the upstream signaling events stimulating DNA-PKcs overexpression in PDAC are still not well characterized. Methods: Expression of DNA-PKcs was measured by western blot. The levels of miRNA-101 and lncRNA nuclear paraspeckle assembly transcript 1 (NEAT1) were detected by real-time PCR. Cell viability was determined by CCK-8. Cell migration and cell invasion were measured by transwell assay. The regulatory relationship between NEAT1 and miR-101 was determined by a luciferase assay. Results: DNA-PKcs expression was significantly elevated in human PDAC tissues and cells. DNA-PKcs overexpression was correlated with TNM stage and lymph node metastasis. Higher expression of DNA-PKcs was closely correlated with patients of worse overall survival (OS). DNA-PKcs knockdown suppresses malignant behaviors of PDAC cells. Further study showed that miRNA-101 level was decreased in PDAC tissues and cells, which could be responsible for DNA-PKcs overexpression and DNA-PKcs mediated oncogenic actions in PDAC cells. Moreover, NEAT1 functions as an oncogene influencing cell proliferation, migration and invasion in part by serving as a competing endogenous RNA (ceRNAs) modulating miR-101 expression, leading to up-regulation of DNA-PKcs. Conclusion: These findings suggest that NEAT1/miR-101-dependent up-regulation of DNA-PKcs promotes the malignant behaviors of PDAC cells. The NEAT1/miR-101/DNA-PKcs axis may serve as a viable prognostic marker and therapeutic target for PDAC.

In this study, through animal experiments, we investigated the effects of Qingli Gandan decoction on the IL-23/IL-17 signalling pathway and the expression of CD4+/IL-17A+ cells in EAU rats. We then predicted the potential mechanisms of the action. First, Lewis rats were divided using the random number table method into a (1) control group, (2) model group and (3) TCM group. The TCM group was given a Qingli Gandan prescription by gavage after an intervention model. Next, blood, aqueous humour and vitreous were collected from each group on days 4, 8, 12, 16 and 20 after immunisation. An enzyme linked immunosorbent assay (ELISA) was used to detect the protein concentrations of IL-17 and IL-23 in a serum, aqueous humour and vitreous of rats on different immunisation days, and the percentage of CD4+/IL-17A+ cells in rat spleen was detected by flow cytometry. The peak concentration of serum IL-17 protein in group 3 was statistically lower than that in group 2 (P < 0.05) on days 4, 8 and 12 of immunisation. However, the peak concentration of serum IL-23 protein in group 3 was statistically lower than in group 2 (P < 0.05) on days 8 and 12 of immunisation; in addition, on days 8 and 12, the protein concentrations of IL-23 and IL-17 in the aqueous humour group 3 were all lower than those in group 2. On days 4, 8 and 12, the protein concentrations of IL-23 and IL-17 in vitreous in group 3 were significantly lower than those in group 2. We found that the proportion of CD4+/IL-17A+ cells in the groups 2 and 3 increased on day 8 compared with group 1 (P < 0.05). The proportion of CD4+/IL-17A+ cells decreased in group 3 compared with group 2 (P < 0.05).At last, we think Qingli Gandan can reduce the onset of EAU in rats and treat it, presumably because of inhibiting the IL-23/IL-17 axis. It may inhibit the expression of CD4+/IL-17A+ cells and play a role in inhibiting the IL-23/IL-17 signalling pathway.

The Erd\H{o}s-Simonovits stability theorem is one of the most widely used theorems in extremal graph theory. We obtain an Erd\H{o}s-Simonovits type stability theorem in multi-partite graphs. Different from the Erd\H{o}s-Simonovits stability theorem, our stability theorem in multi-partite graphs says that if the number of edges of an $H$-free graph $G$ is close to the extremal graphs for $H$, then $G$ has a well-defined structure but may be far away to the extremal graphs for $H$. We show that short-range resonating-valence-bond correlations and long-range order can coexist in the ground state (GS) of a frustrated spin system. Our study comprises a comprehensive investigation of the quantum magnetism on the structurally disorder-free single crystal of Cu$_2$(OH)$_3$NO$_3$, which realizes the $s$ = 1/2 Heisenberg model on a spatially anisotropic triangular lattice. Competing exchange interactions determined by fitting the magnetization measured up to 55 T give rise to an exotic GS wavefunction with coexistence of the dominant short-range resonating-valence-bond correlations and weak long-range stripe order (ordered moment $M_0$ = $|\langle s_i^z\rangle|$ $\sim$ 0.02). At low temperatures, a first-order spin-flop transition is visible at $\sim$ 1-3 T. As the applied field further increases, another two magnetic-field-induced quantum phase transitions are observed at $\sim$ 14-19 and $\sim$ 46-52 T, respectively. Simulations of the Heisenberg exchange model show semi-quantitative agreement with the magnetic-field modulation of these unconventional phases, as well as the absence of visible magnetic reflections in neutron diffraction, thus supporting the GS of the spin system of Cu$_2$(OH)$_3$NO$_3$ may be approximate to a quantum spin liquid. MiniPDX® represents a novel in vivo drug sensitivity test with fast turnaround (7-10 days), using either fresh patient tumor samples or tissues from established PDX models. We systematically evaluated and compared the response rates of MiniPDX® assays and PDX assays pair-wise in 26 PDX models across 3 types of cancers to 12 clinically relevant regimens for chemical and targeting drugs. The results showed a high correlation between drug responses of the two assays, with sensitivity and specificity of 80% and 93%, respectively. More studies of different tumor types showed that MiniPDX drug sensitivity test results were consistent with clinical responses in most patients, which indicated that MiniPDX® models have great potential in guiding personalized cancer therapy. LIDE has completed over 3,000 MiniPDX® tests for clinical precision medicine, covering more than 50 indications. MiniPDX® was used to rank clinically approved drugs or drug regimens to guide the selection of the best individualized treatment strategy. At the same time, screening other compounds facilitates selection of the best drug candidates for further R&D. Importantly, performing MiniPDX® Mouse Trial using fresh tumor samples generated from clinic is beneficial for determination of potential clinical indications. Only thousands of fresh cells leftover from MiniPDX® preparation are sufficient to get genomic and transcription data by OncoVee™ K-cell technology. The combination of MiniPDX® assay and omics data would be very useful for determination of potential bio-markers in order to distinguish responders and non-responders in population with certain indication(s) and leveraged for patient stratification and selection criteria in clinical trial design. Currently, we are developing a new version of MiniPDX® assay for immunotherapy (IO-FIVE, Immuno-Oncology Fast In Vivo Efficacy test) by using fresh patient cancer cells with autologous immune cells to maximally mimic human tumor microenvironment before seeding in immune-deficient mice. Total cell viability and cell phenotyping are tracked before and after drug treatment to evaluate its in vivo efficacy within two weeks. More than 60 samples have been tested and the IO-FIVE functional assay can effectively distinguish the non-responder from responders for immunotherapeutic drugs. Objectives: The treatment of acute herpangina is inconsistent. We aim to evaluate the effectiveness and safety of interferon α-2b spray versus Ribavirin for this disease. Methods: A randomized, controlled trial was conducted in eight hospitals in China between 2016 and 2018. 668 patients (1–7 years old) were randomized into an experimental group (treated with Interferon α-2b spray) or control group (received Ribavirin Aerosol). Body temperature returning to normal within 72 h and remaining so for 24 h was the primary outcome; release of oral herpes and adverse events were the secondary outcomes. Results: (1) The average age of onset was 2.5 years old. (2) After 72 h treatment, body temperature of 98.5% patients in experimental group and 94.3% in control group returned to normal and remained so for 24 h (P = 0.004). The differences were greater at 48 h treatment (95.2% vs. 85.9%, P < 0.001) and at 24 h (77.5% vs. 66.5%, P = 0.001). (3) The rate of improved oral herpes in the experimental group was higher than that in control group (46.7% vs.37.1%, P = 0.011). No adverse reaction occurred. Conclusions: Local application of recombinant interferon α-2b spray showed better efficacy for acute herpangina in children. It was safe for use.

Background Surgical resection remains the best option for long-term survival in colorectal cancer (CRC); however, surgery can lead to tumor cell release into the circulation. Previous studies have also shown that surgery can affect cancer cell growth. The role of perioperative factors influencing long-term survival in patients presenting for CRC surgery remains to be investigated. Methods This retrospective single–center cohort study was conducted to collect the clinical data of patients who underwent elective laparoscopic resection for CRC from January 2014 to December 2015, namely clinical manifestations, pathological results, and perioperative characteristics. Survival was estimated using the Kaplan–Meier log-rank test. Univariable and multivariable Cox regression models were used to compare hazard ratios (HR) for death. Results A total of 234 patients were eligible for analysis. In the multivariable Cox model, tumor-node-metastasis (TNM) stage (stage IV: HR 30.63, 95% confidence interval (CI): 3.85–243.65; P = 0.001), lymphovascular invasion (yes: HR 2.07, 95% CI 1.09–3.92; P = 0.027), inhalational anesthesia with isoflurane (HR 1.96, 95% CI 1.19–3.21; P = 0.008), and Klintrup–Makinen (KM) inflammatory cell infiltration grade (low-grade inflammation: HR 2.03, 95% CI 1.20–3.43; P = 0.008) were independent risk factors affecting 5-year overall survival after laparoscopic resection for CRC. Conclusions TNM stage, lymphovascular invasion, isoflurane, and KM grade were independent risk factors affecting CRC prognosis. Sevoflurane and high-grade inflammation may be associated with improved survival in CRC patients undergoing resection.

The spin-1/2 kagom\'{e} Heisenberg antiferromagnet is generally accepted as one of the most promising two-dimensional models to realize a quantum spin liquid state. Previous experimental efforts were almost exclusively on only one archetypal material, the herbertsmithite ZnCu$_3$(OH)$_6$Cl$_2$, which unfortunately suffers from the notorious orphan spins problem caused by magnetic disorders. Here we turn to YCu$_3$(OH)$_{6.5}$Br$_{2.5}$, recently recognized as another host of a globally undistorted kagom\'{e} Cu$^{2+}$ lattice free from the orphan spins, thus a more feasible system for studying the intrinsic kagom\'{e} quantum spin liquid physics. Our high-resolution low-temperature thermal conductivity measurements yield a vanishing small residual linear term of $\kappa/T$ ($T\rightarrow 0$), and thus clearly rule out itinerant gapless fermionic excitations. Unusual scattering of phonons grows exponentially with temperature, suggesting thermally activated phonon-spin scattering and hence a gapped magnetic excitation, consistent with a $\mathbb{Z}_2$ quantum spin liquid ground state. Temporal focusing-based multiphoton excitation microscopy (TFMPEM) just provides the advantage of widefield optical sectioning ability with axial resolution of several micrometers. However, under the plane excitation, the photons emitted from the molecules in turbid tissues undergo scattering, resulting in complicated background noise and an impaired widefield image quality. Accordingly, this study constructs a general and comprehensive numerical model of TFMPEM utilizing Fourier optics and performs simulations to determine the superior spatial frequency and orientation of the structured pattern which maximize the axial excitation confinement. It is shown experimentally that the optimized pattern minimizes the intensity of the out-of-focus signal, and hence improves the quality of the image reconstructed using the Hilbert transform (HT). However, the square-like reflection components on digital micromirror device leads to pattern residuals in the demodulated image when applying high spatial frequency of structured pattern. Accordingly, the HT is replaced with Hilbert–Huang transform (HHT) in order to sift out the low-frequency background noise and pattern residuals in the demodulation process. The experimental results obtained using a kidney tissue sample show that the HHT yields a significant improvement in the TFMPEM image quality.

Chronic hepatitis B virus (HBV) infection can lead to fibrosis, liver cirrhosis, and primary hepatocellular carcinoma. Investigating host factors that regulate HBV replication helps to identify antiviral targets. In the current study, we identified Nicotinamide N-Methyltransferase gene (NNMT) as a novel factor that regulates HBV transcription. NNMT is up-regulated at both the mRNA and protein levels in HepG2.2.15 cells compared to HepG2 cells. Overexpression of NNMT reduces HBV replication in several cell models, while knockdown of NNMT enhances HBV DNA levels. Mechanistically, NNMT suppresses HBV DNA replication by inhibiting HBV RNA transcription. The region required for the inhibitory effect of NNMT was narrowed to nt 1672-1708 in enhancer II by luciferase assays. On the other hand, ChIP assays and EMSA results showed that NNMT does not bind to this region substantially, either directly or indirectly. Next, a collection of hepatic nuclear receptor transcription factors was screened to determine whether they were affected by NNMT overexpression. NR5A1, a positive regulator of HBV replication, decreased significantly after NNMT overexpression. Collectively, the findings of this study shed light on the regulation of HBV transcription.

One of the most desirable targets for HBV medications is the sodium taurocholate cotransporting polypeptide (NTCP), an entry receptor for the hepatitis B virus (HBV). N-myristoylated preS1 2-48 (Myrcludex B or Hepcludex), an NTCP-binding peptide from the large surface protein of HBV, has been developed as the first-in-class entry inhibitor. However, its relatively large molecular weight contributes to increased immunogenicity and antibody production. As a result, it is preferable to look for an NTCP-binding peptide with a smaller size. To do this, we developed a human cell surface display strategy and screened peptides based on preS1-21. PreS1-21 (genotype D) was extended by 7 random amino acids and fused with mCherry and FasL transmembrane domain. The pooled constructs were transfected into HEK293 cells by using the transposon/transposase system to create a library displaying various peptides on the cell surface with red fluorescence. On the other hand, we expressed NTCP protein fused with EGFP on HEK293 and used the membrane lysate containing NTCP-GFP as the bait protein to select peptides with increased NTCP affinity. After 7 cycles of selection, the deep sequencing results revealed that some polypeptides were more than 1,000 times enriched. Further screening of the mostly enriched 10 peptides yields the peptide preS1-21-pep3. Replacing the preS1-21 sequence of preS1-21-pep3 with those from different genotypes demonstrated that the consensus sequence of genotype A-F had the best performance. The peptide (Myr-preS1-21-pep3) was synthesized and tested on the HepG2-NTCP cell model. The results showed that Myr-preS1-21-pep3 is approximately 10 times more potent than the initial peptide Myr-preS1-21 in preventing HBV infection. In conclusion, we developed a new strategy for screening peptides binding to membrane proteins and identified a new NTCP-binding peptide with a much smaller size than Hepcludex.

Introduction Sepsis is a complication in acute-on-chronic liver failure (ACLF) patients associated with high rates of mortality and morbidity. Early diagnosis of sepsis in ACLF patients can improve prognosis. This study aimed to explore potential effective biomarkers for the early diagnosis of sepsis in ACLF patients. Methods Ninety-four ACLF patients with sepsis were enrolled from 10 hospitals across China from January 2015 to June 2016 as well as 49 ACLF patients without infection from Xiangya Hospital. Background Acute fatty liver of pregnancy (AFLP) is a potentially lethal condition of pregnant women with a high mortality rate. Potential predictors related to postpartum recovery time and prognostic factors of AFLP are still unclear. This study aimed to evaluate potential predictors for prognosis and postpartum recovery time of AFLP. Methods We retrospectively analyzed the clinical data of 76 AFLP patients in our hospital from 2002 to 2017 and investigated potential predictors using univariate analysis and multivariate logistic regression analysis. Results Hepatic encephalopathy (HE) was found to be associated with prognosis in AFLP patients (P = 0.005, OR = 26.844). The postpartum recovery time analysis showed that AFLP patients with a age P = 0.134, OR = 5.952). The postpartum recovery time of patients with liver failure (LF) was significantly prolonged compared to those without LF (P = 0.036, OR = 10.052). Cryoprecipitate, and plasma infusion showed no significant effect on prognosis or recovery time. Artificial liver support therapy (ALST) had no effect on prognosis, but it might affect postpartum recovery time with no statistical significance (P = 0.128, OR = 5.470). Conclusion HE is a potential predictor for prognosis of AFLP. LF is a potential predictor for postpartum recovery time.

Non-small-cell lung cancer (NSCLC) is known as one of the most lethal cancers, causing more than 1 million deaths annually worldwide. Therefore, the development of novel therapeutic drugs for NSCLC has become an urgent need. Herein, various mouse models provide great convenience not only for researchers but also for the development of antitumor drug. Meanwhile, TCM, as a valuable and largely untapped resource pool for modern medicine, provides research resources for the treatment of various diseases. Until now, cell-derived xenograft (CDX) model, patient-derived xenograft (PDX) model, syngeneic model, orthotopic model, humanized mouse model (HIS), and genetically engineered mouse models (GEMMs) have been reported in TCM evaluation. This review shows the role and current status of kinds of mouse models in antitumor research and summarizes the application progress of TCM including extracts, formulas, and isolated single molecules for NSCLC therapy in various mouse models; more importantly, it provides a theoretical exploration of what kind of mouse models is ideal for TCM efficacy evaluation in future. However, there are still huge challenges and limitations in the development of mouse models specifically for the TCM research, and none of the available models are perfectly matching the characteristics of TCM, which suppress the tumor growth through various mechanisms, especially by regulating immune function. Nevertheless, with fully functional immune system existing in syngeneic model and humanized mouse model (HIS), it is still suggested that these two models are more suitable for development of TCM especially for TCM extracts or formulas. Moreover, continued efforts are needed to generate more reliable mouse models to test TCM formulas in future research.

The aged seeds have a significant influence on seed vigor and corn growth. Therefore, it is vital for the planting industry to identify aged seeds. In this study, hyperspectral reflectance imaging (1,000–2,000 nm) was employed for identifying aged maize seeds using seeds harvested in different years. The average spectra of the embryo side, endosperm side, and both sides were extracted. The support vector machine (SVM) algorithm was used to develop classification models based on full spectra to evaluate the potential of hyperspectral imaging for maize seed detection and using the principal component analysis (PCA) and ANOVA to reduce data dimensionality and extract feature wavelengths. The classification models achieved perfect performance using full spectra with an accuracy of 100% for the prediction set. The performance of models established with the first three principal components was similar to full spectrum models, but that of PCA loading models was worse. Compared to other spectra, the two-band ratio (1,987 nm/1,079 nm) selected by ANOVA from embryo-side spectra achieved a better classification accuracy of 95% for the prediction set. The image texture features, including histogram statistics (HS) and gray-level co-occurrence matrix (GLCM), were extracted from the two-band ratio image to establish fusion models. The results demonstrated that the two-band ratio selected from embryo-side spectra combined with image texture features achieved the classification of maize seeds harvested in different years with an accuracy of 97.5% for the prediction set. The overall results indicated that combining the two wavelengths with image texture features could detect aged maize seeds effectively. The proposed method was conducive to the development of multi-spectral detection equipment.
