Intracellular delivery of therapeutic proteins has increased advantages over current small-molecule drugs and gene therapies, especially in therapeutic efficacies for a broad spectrum of diseases. Hence, developing the protein therapeutics approach provides a needed alternative. Here, we designed a mesoporous silica nanoparticle (MSN)-mediated protein delivery approach and demonstrated effective intracellular delivery of the denatured superoxide dismutase (SOD) protein, overcoming the delivery challenges and achieving higher enzymatic activity than native SOD-conjugated MSNs. The denatured SOD-conjugated MSN delivery strategy provides benefits of reduced size and steric hindrance, increased protein flexibility without distorting its secondary structure, exposure of the cell-penetrating peptide transactivator of transcription for enhanced efficient delivery, and a change in the corona protein composition, enabling cytosolic delivery. After delivery, SOD displayed a specific activity around threefold higher than in our previous reports. Furthermore, the in vivo biosafety and therapeutic potential for neuron therapy were evaluated, demonstrating the biocompatibility and the effective antioxidant effect in Neuro-2a cells that protected neurite outgrowth from paraquat-induced reactive oxygen species attack. This study offers an opportunity to realize the druggable possibility of cytosolic proteins using MSNs.

Osteosarcoma (OS) is a type of tumor with high malignant behaviors. Increasing investigates have confirmed that long non-coding RNA HLA complex group 18 (lncRNA HCG18) acted as a tumor-promoting factor in multiple tumors. Nevertheless, the underlying mechanism of HCG18 on OS remains largely unclear. HCG18, miR-34a, and runt-related transcription factor 2 (RUNX2) expressions were detected by quantitative real-time PCR (RT-qPCR) or western blotting assays, respectively. The underlying tumorigenic phenotypes were detected by MTT, wound healing, transwell invasion, western blotting assays. Molecular interactions were verified by dual-luciferase report assay. HCG18 and RUNX2 were notably enhanced, whereas miR-34a was decreased in OS tumor tissues and cell lines. Functional experiments uncovered that HCG18 silencing significantly inhibited the capabilities of proliferation, migration, and invasion, while overexpression of HCG18 play the opposite roles. Furthermore, HCG18 directly bound to miR-34a, and miR-34a was confirm to be a negative regulator of RUNX2. Interestingly, the anti-tumor effects of HCG18 silencing were attenuated by miR-34a inhibitor and RUNX2 overexpression. Taken together, the present study suggested that HCG18 promoted the malignant biological behaviors of OS through regulating the miR-34a/RUNX2 pathway, implying HCG18 might serve as a new target for OS treatment.

ObjectiveThis study aimed to compare the effects of robot-assisted thoracic surgery (RATS), video-assisted thoracic surgery (VATS), and thoracotomy on the psychological status, medical coping mode, and quality of life of patients with lung cancer.MethodsA total of 158 patients with lung cancer were selected from the thoracic surgery center of a third-grade hospital in Hunan Province, China, from September to November 2020. The Self-Rating Anxiety Scale (SAS), Self-Rating Depression Scale (SDS), Medical Coping Modes Questionnaire (MCMQ), and Medical Outcomes Study (MOS) 36-item Short Form Health Survey (SF-36) were used to assess the effects of the surgical approaches on the study parameters before and 48–96 h after surgery. The t-test and analysis of variance were used to analyze the data.ResultsThe results revealed that the patients’ depression increased, their short-term quality of life decreased, and they tended to adopt a positive coping mode after surgery (p p p ConclusionSurgical approaches have little effect on the psychological status, medical coping modes, and quality of life of patients with lung cancer; however, their depression increased and quality of life decreased after the surgery.

Background Left-behind adolescents (LBAs) are adolescents aged 11–18 years who are separated from their parents and left behind in local cities by one or both parents for a period of more than 6 months. LBAs in rural areas are likely to engage in aggressive behavior, which can affect interpersonal relationships, reduce academic performance, and even lead to anxiety and depression. To our knowledge, no studies have examined the mediating effect of resilience and self-esteem on the relationship between negative life events and aggression among Chinese rural LBAs. Therefore, this study aimed to explore the relationship between negative life events and aggression among Chinese rural LBAs and how self-esteem and resilience mediate the association. Methods Using a stratified random sampling method, 1344 LBAs in Hunan Province of China were investigated. Information was collected by a self-designed sociodemographic questionnaire, Adolescent Self-Rating Life Events Checklist, Resilience Scale Chinese Adolescent, Rosenberg Self-Esteem Scale and Aggression Scales to assess the psychology of LBAs. Data analysis was conducted using descriptive statistics, Pearson correlation, and regression analysis to estimate direct and indirect effects using bootstrap analysis. Results Negative life events were significantly related to self-esteem (r = − 0.338), resilience (r = − 0.359), and aggression (r = 0.441). Aggression was directly affected by self-esteem (β = − 0.44) and resilience (β = − 0.34). Negative life events were not only directly related to aggression (β = 0.34, 95% CI: 0.275 ~ 0.398) but also showed an indirect effect on aggression through self-esteem and resilience. The direct effect, total effect and indirect effect of negative life events on aggression through self-esteem and resilience were 0.3364, 0.4344 and 0.0980, respectively. The mediating effect of self-esteem and resilience accounted for 22.56% of the relationship between negative life events and aggression. Conclusions We found that self-esteem and resilience mediated most negative life events on aggression. It is imperative for educators and families to improve LBAs’ self-esteem and resilience to reduce the occurrence of aggression. Future intervention studies should be designed to strengthen self-esteem and resilience.

The eco-environment of semi-arid regions is extremely sensitive and vulnerable to climate change and human activities. The realization of sustainability of eco-environments has long been a key issue. Consequently, we analysed the Chinese Loess Plateau (CLP) to identify solutions to the sustainable development of such regions. We first reviewed the eco-environmental changes in the CLP and discussed the impacts of human activities over the past 3000 years, and then summarized previous experiences on the protection and restoration of degraded eco-environments. Our study indicates that the increasing demand for cultivated land has led to the destruction of vegetation and subsequent worsening of soil erosion on the CLP, which in turn, has led to an increase in floods in the Yellow River basin. However, many positive measures, such as slope cropland abandonment and afforestation have been demonstrated to be effective in improving the eco-environment via reducing soil erosion, and decreasing sediment input and flood disasters in the Yellow River in recent decades. Nonetheless, emerging problems such as water shortage, food insecurity, rapid urban development, and over-exploitation of energy resources hinder the sustainable development of the eco-environment in the CLP. Given the historical and present eco-environmental situations of the CLP, we provide suggestions for the sustainable development of both the eco-environment and social economy, which could serve as guidelines for eco-environmental design and government policy-making in semi-arid regions globally.

Detection of neurotransmitters at the single-cell level is essential for understanding the related biological processes and neurodegenerative diseases. We report a dual-nanopore biosensor utilizing a DNA aptamer probe to specifically interact with dopamine, enabling detection of intracellular dopamine and dopamine efflux (extracellular dopamine) in a single pheochromocytoma (PC12) cell. We demonstrate the ability to form an intrapipette electric circuit with the dual-nanopore configuration, which is crucial to achieving both intracellular and extracellular dopamine detection. The sensor allowed rapid detection of dopamine in 10 min with a limit of detection of 0.4 nM. We show the dual-nanopore biosensor was able to monitor single-cell dopamine concentration change under different stimulations. The developed dual-nanopore biosensor represents a novel strategy for time-dependent monitoring of neuron behavior at the single-cell level and potentially can be extended to other platforms for single-cell analysis.

The unlimited possibilities of machine learning have been shown in several successful reports and applications. However, how to make sure that the searched results of a machine learning system are not tampered by anyone and how to prevent the other users in the same network environment from easily getting our private data are two critical research issues when we immerse into powerful machine learning-based systems or applications. This situation is just like other modern information systems that confront security and privacy issues. The development of blockchain provides us an alternative way to address these two issues. That is why some recent studies have attempted to develop machine learning systems with blockchain technologies or to apply machine learning methods to blockchain systems. To show what the combination of blockchain and machine learning is capable of doing, in this paper, we proposed a parallel framework to find out suitable hyperparameters of deep learning in a blockchain environment by using a metaheuristic algorithm. The proposed framework also takes into account the issue of communication cost, by limiting the number of information exchanges between miners and blockchain.

Background/aim Cetyltrimethylammonium bromide (CTAB), a quaternary ammonium surfactant, was shown to have antitumor effects in a cellular mode of head and neck squamous cell carcinoma (HNSCC), modulating apoptotic and cytotoxic processes. However, the mechanisms by which CTAB exerts its effects against the epithelial- mesenchymal transition in HNSCC remain poorly understood. In the present study, we investigated whether CTAB inhibits cellular mobility and invasiveness of hypopharyngeal squamous cell carcinoma (HPSCC) cells. Materials and methods WST-1, cell-cycle phase distribution, and wound healing, as well as transwell assays were conducted. Changes in protein expression patterns and related signaling pathways involved in effects of CTAB on HPSCC cell lines were evaluated by western blotting. Results Treatment of human HPSCC cell lines with CTAB significantly altered their morphology from spindle-like to cobblestone-like by diminishing mesenchymal-like phenotypic characteristics. CTAB also hindered cell functional properties, including migration and invasion, independently of cell viability. In addition, western blot results demonstrated that treatment with CTAB reduced expression of mesenchymal markers. Further investigation showed that CTAB treatment suppressed the phosphorylation of extracellular-regulated kinase 1/2, mechanistic target of rapamycin kinase and AKT serine/threonine kinase 1. CTAB also repressed the expression and phosphorylation levels of epidermal growth factor receptor (EGFR) and phosphatidylinositol-4,5-bisphosphate 3-kinase (PI3K), and the partial restoration of mesenchymal phenotype by EGF addition confirmed that CTAB inhibited migration and invasion in HPSCC cells by blocking the EGFR signaling pathway. Conclusion Our results suggest that CTAB is involved in the suppression of EGFR-mediated mesenchymal phenotype and the molecular mechanism by which CTAB obstructs HPSCC cell metastasis may represent a promising strategy for use in HPSCC treatment.

Microbiome plays an important role in evaluating soil quality for sustainable agriculture. However, the suitability of biological indicators in reclaimed farmland is less understood. Using high-throughput sequencing, we evaluated the soil microbial community of the newly created farmland (NF) after reclamation with two local high-yield farmlands (slope farmland (SF), check-dam farmland (CF)) on the Loess Plateau. Soil enzyme activities and the amount of culturable microorganism were also quantified to assess the soil quality. Results showed that the microbial diversity, cultural microorganism abundance, and soil enzyme activities indicated poor soil quality in NF. The dominant bacterial phyla were Proteobacteria, Bacteroidetes, Acidobacteria, and Cyanobacteria. The abundance of Acidobacteria was significantly lower in NF (13.31%) than in SF (27.25%) and CF (27.91%). Soil enzyme activities had a significant correlation with the abundance of culturable microorganism, Proteobacteria and Bacteroidetes, soil organic matter, total nitrogen, cation exchange capacity, and pH, suggesting that soil microbes have driven the formation of nutrition and further mediated crop growth. Therefore, the application of bacterial fertilizers could be a potential way to improve the soil quality of reclaimed farmland for crop growth.

The blood–brain barrier (BBB) is a highly selective cellular barrier that tightly controls the microenvironment of the central nervous system to restrict the passage of substances, which is a primary challenge in delivering therapeutic drugs to treat brain diseases. This study aimed to develop simple surface modifications of mesoporous silica nanoparticles (MSNs) without external stimuli or receptor protein conjugation, which exhibited a critical surface charge and size allowing them to cross the BBB. A series of MSNs with various charges and two different sizes of 50 and 200 nm were synthesized, which showed a uniform mesoporous structure with various surface zeta potentials ranging from +42.3 to −51.6 mV. Confocal microscopic results showed that 50 nm of strongly negatively charged N4-RMSN50@PEG/THPMP (∼−40 mV) could be significantly observed outside blood vessels of the brain in Tg(zfli1:EGFP) transgenic zebrafish embryos superior to the other negatively charged MSNs. However, very few positively charged MSNs were found in the brain, indicating that negatively charged MSNs could successfully penetrate the BBB. The data were confirmed by high-resolution images of 3D deconvoluted confocal microscopy and two-photon microscopy and zebrafish brain tissue sections. In addition, while increasing the size to 200 nm but maintaining the similar negative charge (∼40 mV), MSNs could not be detected in the brain of zebrafish, suggesting that transport across the BBB based on MSNs occurred in charge- and size-dependent manners. No obvious cytotoxicity was observed in the CTX-TNA2 astrocyte cell line and U87-MG glioma cell line treated with MSNs. After doxorubicin (Dox) loading, N4-RMSN50@PEG/THPMP/Dox enabled drug delivery and pH-responsive release. The toxicity assay showed that N4-RMSN50@PEG/THPMP could reduce Dox release, resulting in the increase of the survival rate in zebrafish. Flow cytometry demonstrated N4-RMSN50@PEG/THPMP had few cellular uptakes. Protein corona analysis revealed three transporter proteins, such as afamin, apolipoprotein E, and basigin, could contribute to BBB penetration, validating the possible mechanism of N4-RMSN50@PEG/THPMP crossing the BBB. With this simple approach, MSNs with critical negative charge and size could overcome the BBB-limiting characteristics of therapeutic drug molecules; furthermore, their use may also cause drug sustained-release in the brain, decreasing peripheral toxicity.

Reversing the immunosuppressive tumor microenvironment (TME) is a strategic initiative to sensitize cancer immunotherapy. Emerging evidence shows that cyclic diguanylate monophosphate (c-di-GMP or cdG) can induce the stimulator of interferon genes (STING) pathway activation of antigen-presenting cells (APCs) and upregulate expression of type I interferons (IFNs) to enhance tumor immunogenicity. In vitro anionic cdG revealed fast plasma clearance, poor membrane permeability, and inadequate cytosolic bioavailability. Therefore, we explored a comprehensive "in situ vaccination" strategy on the basis of nanomedicine to trigger robust antitumor immunity. Rhodamine B isothiocyanate (RITC) fluorescent mesoporous silica nanoparticles (MSN) synthesized and modified with poly(ethylene glycol) (PEG) and an ammonium-based cationic molecule (TA) were loaded with negatively charged cdG via electrostatic interactions to form cdG@RMSN-PEG-TA. Treatment of RAW 264.7 cells with cdG@RMSN-PEG-TA markedly stimulated the secretion of IL-6, IL-1β, and IFN-β along with phospho-STING (Ser365) protein expression. In vivo cdG@RMSN-PEG-TA enhanced infiltration of leukocytes, including CD11c+ dendritic cells, F4/80+ macrophages, CD4+ T cells, and CD8+ T cells within the tumor microenvironment (TME), resulting in dramatic tumor growth inhibition in 4T1 breast tumor-bearing Balb/c mice. Our findings suggest that a nanobased platform can overcome the obstacles bare cdG can face in the TME. Our approach of an in situ vaccination using a STING agonist provides an attractive immunotherapy-based strategy for treating breast cancer.

Muscles serve as a critical regulator of locomotion and damping, resulting in changes of soft tissue vibration. However, whether muscle fibre compositions of different individuals will cause different extents of soft tissue vibration during gait is unclear. Therefore, this study investigated the differences in lower extremity vibration frequencies among power-trained and non-power-trained athletes during walking and running. Twelve weightlifting athletes were assigned to the power-trained group and twelve recreational runners were assigned to the non-power-trained group. Accelerometers were used to detect soft tissue compartment vibration frequencies of the rectus femoris (RF) and gastrocnemius medialis (GMS) during walking and running. Results indicated that power-trained athletes, as compared to the non-power-trained, induced significantly (p 0.05) higher vibration frequencies in their soft tissue compartments during walking and running. This suggests that power-trained athletes, who have higher ratios of fatigable fast-twitch muscle fibres, may have induced higher soft tissue compartment vibration frequencies. As a result, there is a likelihood that power-trained athletes may recruit more fatigable fast-twitch muscle fibres during muscle tuning, causing dysfunctions during prolonged exercises.

The present study investigated the ecotoxicity of raw mining effluent from the largest molybdenum (Mo) open-pit mine in the Qinling mountains, China, and the treated effluent with neutralization and coagulation/adsorption processes, using zebrafish (Danio rerio). The results showed the following: (1) the mining effluent is acid mine drainage (AMD) and is highly toxic to zebrafish with a 96-h median lethal concentration (LC50) of 3.80% (volume percentage) of the raw effluent; (2) sublethal concentrations of the raw effluent (1/50, 1/10, and 1/2 96-h LC50) induced oxidative stress and osmoregulatory impairment, as reflected by the alterations in activities of superoxide dismutase and catalase and contents of malondialdehyde, and inhibition of Na+, K+-ATPase activity in gills and muscle after 28 days of sub-chronic exposure when compared with the unexposed group; and (3) the treatment of the raw effluent with neutralizer (NaOH) and adsorbent activated carbon reduced the acute lethal effect of raw effluent. The used endpoints including acute lethal and biochemical parameters related to oxidative stress and osmoregulatory impairment in zebrafish are cost-effective for toxicity assessment of AMD like the studied Mo mining effluent. Mining effluent management strategies extended by these results, i.e., the restriction of discharging raw and diluted effluent to adjacent waterways and the introduction of bio-monitoring system across all mining drainages in this area, were also proposed and discussed.

Background/aim Hepatocellular carcinoma (HCC) arises from hepatocytes, and is the most frequently occurring malignancy of primary liver cancer. In this study, we investigated the anti-metastatic effects of the quaternary ammonium compound, cetyltrimethylammonium bromide (CTAB), on HA22T/VGH HCC cells. Materials and methods According to our preliminary data, the effect of CTAB on cell cycle distribution, migration, invasion and the associated protein levels was examined using flow cytometry, wound-healing migration, Matrigel transwell invasion assay and western blotting under sub-lethal concentrations. Results CTAB treatment of HA22T/VGH cells casued dose-dependent mesenchymal-epithelial transition (MET)-like changes and impaired migration and invasion capabilities. In addition, CTAB reduced the levels of metastasis-related proteins including c-Met, phosphoinositide 3-kinase (PI3K), Akt, mammalian target of rapamycin (mTOR), ribosomal protein S6 kinase (p70S6K), Twist, N-cadherin, and Vimentin. Moreover, pretreatment with hepatocyte growth factor (HGF) rescued CTAB-mediated effects. Conclusion CTAB exhibited potent anti-EMT and anti-metastatic activities through the inhibition of migration and invasion of HA22T/VGH cells. CTAB interrupted the mesenchymal characteristics of HA22T/VGH cells, which were significantly alleviated by HGF in a dose-dependent manner. CTAB has the potential to evolve as a therapeutic agent for HCC.

Bacterial RNA polymerase (RNAP) is a validated drug target for broad-spectrum antibiotics, and its "switch region" is considered as the promising binding site for novel antibiotics. Based on the core scaffold of dithiolopyrrolone, a series of N-aryl pyrrothine derivatives was designed, synthesized, and evaluated for their antibacterial activity. Compounds generally displayed more active against Gram-positive bacteria, but less against Gram-negative bacteria. Among them, compound 6e exhibited moderate antibacterial activity against clinical isolates of rifampin-resistant Staphylococcus aureus with minimum inhibition concentration value of 1-2 μg/ml and inhibited Escherichia coli RNAP with IC50 value of 12.0 ± 0.9 μM. In addition, compound 6e showed certain degree of cytotoxicity against HepG2 and LO2 cells. Furthermore, molecular docking studies suggested that compound 6e might interact with the switch region of bacterial RNAP in a similar conformation to myxopyronin A. Together, the N-aryl pyrrothine scaffold is a promising lead for discovery of antibacterial drugs acting against bacterial RNAP.

DNA methyltransferases are primary enzymes for cytosine methylation at CpG sites of epigenetic gene regulation in mammals. De novo methyltransferases DNMT3A and DNMT3B create DNA methylation patterns during development, but how they differentially implement genomic DNA methylation patterns is poorly understood. Here, we report crystal structures of the catalytic domain of human DNMT3B–3L complex, noncovalently bound with and without DNA of different sequences. Human DNMT3B uses two flexible loops to enclose DNA and employs its catalytic loop to flip out the cytosine base. As opposed to DNMT3A, DNMT3B specifically recognizes DNA with CpGpG sites via residues Asn779 and Lys777 in its more stable and well-ordered target recognition domain loop to facilitate processive methylation of tandemly repeated CpG sites. We also identify a proton wire water channel for the final deprotonation step, revealing the complete working mechanism for cytosine methylation by DNMT3B and providing the structural basis for DNMT3B mutation-induced hypomethylation in immunodeficiency, centromere instability and facial anomalies syndrome.

Hepatocellular carcinoma (HCC) is common and causes many deaths worldwide. The aim of this study is to explore the mechanism by which long non-coding RNA FGD5-AS1 regulates HCC cell proliferation and stemness.Tumor and normal adjacent tissues were harvested from HCC patients. Real-time quantitative reverse transcription-PCR was applied to examine the expression of FGD5-AS1, miR-223, Epithelial cell transforming sequence 2 (ECT2) and FAT1. The protein levels of ECT2, FAT1, proliferating cell nuclear antigen (PCNA), OCT4, CD133 and CD90 were analyzed by western blot. The localization of FGD5-AS1 was examined by Fluorescence in situ hybridization. Cell proliferation was analyzed with CCK-8 and colony formation assays. Spheroid formation was used for analyzing cell stemness. Gene interaction was examined by RNA immunoprecipitation and luciferase activity assays. A subcutaneous xenograft mouse model was established to analyze HCC growth and stemness in vivo. Immunohistochemistry staining was used to analyze the expression PCNA and OCT4 in subcutaneous tumors.FGD5-AS1 was upregulated in HCC and its high expression indicated poor prognosis of patients. High expression of FGD5-AS1 enhanced HCC cell proliferation and stemness. Knockdown of FGD5-AS1 restrained tumor growth and stemness in mice. FGD5-AS1 directly sponged miR-223 and promoted the expression of ECT2 and FAT1 in HCC. Both knockdown of miR-223 and overexpression of ECT2 and FAT1 reversed FGD5-AS1 silencing-mediated suppression of HCC cell proliferation and stemness.FGD5-AS1 directly sponged miR-223 and promoted the expression of ECT2 and FAT1 in HCC, thus enhancing HCC cell proliferation and stemness. Our study identifies potential prognostic biomarkers and therapeutic targets for HCC.

Oxidative stress is closely related to the occurrence of depression. Acupuncture has been proved to be an effective method for treating depression. In order to explore the mechanism of the antidepressant effect of acupuncture, this study performed acupuncture prevention on chronic unpredictable mild stress (CUMS) depression model rats, and observed the effect of acupuncture on hippocampal oxidative stress and Nrf2 signaling pathway. Male SD rats were randomly divided into control group, CUMS group, acupuncture group, and fluoxetine group (n = 10/group). Fluoxetine, a common antidepressant, was used as a positive control drug in this study. In the fluoxetine group, rats were given fluoxetine (2.1 mg/kg) intragastrically once a day for 28 days. The acupoints of Shangxing (GV23) and Fengfu (GV16) were applied in acupuncture group, once every other day for 14 times in total. Behavioral tests and biological detections were used to evaluate the effects of the interventions and the changes of factors related to oxidative stress, Nrf2 pathway, and neuronal apoptosis. The results showed that both acupuncture and fluoxetine could increase sugar preference rate in SPT and decrease immobility time in FST in depression model rats. It also significantly decreased oxidative stress products such as ROS and H2O2, and elevated the protein and mRNA expressions of Nrf2 and HO-1. From Nissl’s staining, there were more abundant nerve cells in two intervention groups compared with CUMS group. Plus, acupuncture down-regulated the expression levels of Bax and caspase-3 and up-regulated the expression of Bcl-2. Our findings indicate that acupuncture improved depression-like behaviors of CUMS rats. And CUMS-induced depression-like behaviors in rats were related to oxidative stress and neuronal apoptosis in hippocampus. Acupuncture showed antidepressant effects in reducing oxidative stress products via regulating the Nrf2/HO-1 signaling pathway so that prevented neuronal apoptosis.

The Infectious Disease Surveillance of Pediatrics (ISPED) program was established in 2015 to monitor and analyze the trends of bacterial epidemiology and antimicrobial resistance (AMR) in children. Clinical bacterial isolates were collected from 11 tertiary care children’s hospitals in China in 2016 to 2020. Antimicrobial susceptibility testing was carried out using the Kirby-Bauer method or automated systems, with interpretation according to the Clinical and Laboratory Standards Institute 2019 breakpoints. A total of 288,377 isolates were collected, and the top 10 predominant bacteria were Escherichia coli, Streptococcus pneumoniae, Staphylococcus aureus, Haemophilus influenzae, Klebsiella pneumoniae, Moraxella catarrhalis, Streptococcus pyogenes, Staphylococcus epidermidis, Pseudomonas aeruginosa, and Acinetobacter baumannii. In 2020, the coronavirus disease 2019 (COVID-19) pandemic year, we observed a significant reduction in the proportion of respiratory tract samples (from 56.9% to 44.0%). A comparable reduction was also seen in the primary bacteria mainly isolated from respiratory tract samples, including S. pneumoniae, H. influenzae, and S. pyogenes. Multidrug-resistant organisms (MDROs) in children were commonly observed and presented higher rates of drug resistance than sensitive strains. The proportions of carbapenem-resistant K. pneumoniae (CRKP), carbapenem-resistant A. baumannii (CRAB), carbapenem-resistant P. aeruginosa (CRPA), and methicillin-resistant S. aureus (MRSA) strains were 19.7%, 46.4%%, 12.8%, and 35.0%, respectively. The proportions of CRKP, CRAB, and CRPA strains all showed decreasing trends between 2015 and 2020. Carbapenem-resistant Enterobacteriaceae (CRE) and CRPA gradually decreased with age, while CRAB showed the opposite trend with age. Both CRE and CRPA pose potential threats to neonates. MDROs show very high levels of AMR and have become an urgent threat to children, suggesting that effective monitoring of AMR and antimicrobial stewardship among children in China are required. IMPORTANCE AMR, especially that involving multidrug-resistant organisms (MDROs), is recognized as a global threat to human health; AMR renders infections increasingly difficult to treat, constituting an enormous economic burden and producing tremendous negative impacts on patient morbidity and mortality rates. There are many surveillance programs in the world to address AMR profiles and MDRO prevalence in humans. However, published studies evaluating the overall AMR rates or MDRO distributions in children are very limited or are of mixed quality. In this study, we showed the bacterial epidemiology and resistance profiles of primary pathogens in Chinese children from 2016 to 2020 for the first time, analyzed MDRO distributions with time and with age, and described MDROs’ potential threats to children, especially low-immunity neonates. Our study will be very useful to guide antiinfection therapy in Chinese children, as well as worldwide pediatric patients.

Aim To measure the prevalence of compassion fatigue among Chinese clinical nurses and to examine the effects of resilience and self-efficacy on compassion fatigue. Design A cross-sectional descriptive survey was conducted in accordance with the STROBE guidelines. Methods Participants were recruited from three tertiary hospitals in central China from October 3 to December 15, 2019, using convenience sampling. Clinical nurses (n = 992) from different nursing departments completed the General Information Questionnaire, Professional Quality of Life Scale, Connor-Davidson Resilience Scale, and General Perceived Self-Efficacy Scale. Descriptive statistics, t-tests, one-way analysis of variance, Pearson or Spearman's correlation analyses, and multiple linear regression models were used. Results Nurses experienced moderate levels of compassion fatigue (burnout and secondary traumatic stress). Resilience and self-efficacy were significantly negatively correlated with burnout but not with secondary traumatic stress. Linear regression analysis showed that resilience, self-efficacy, exercise, and physical conditions were the main predictors of burnout. Only physical conditions and marital status significantly predicted secondary traumatic stress. Conclusion Nurses are vulnerable to compassion fatigue in China. Resilience and self-efficacy significantly negatively predicted nurses' compassion fatigue. Physical conditions, healthy lifestyles, and social support are also important factors for compassion fatigue. Impact Our findings can be used to generate targeted intervention and coping strategies for nurses to improve their resilience and self-efficacy to alleviate compassion fatigue.

Global food insecurity is becoming more severe under the threat of rising global carbon dioxide concentrations, increasing population, and shrinking farmlands and their degeneration. We acquired the ISI Web of Science platform for over 31 years (1988–2018) to review the research on how climate change impacts global food security, and then performed cluster analysis and research hotspot analysis with VosViewer software. We found there were two drawbacks that exist in the current research. Firstly, current field research data were defective because they were collected from various facilities and were hard to integrate. The other drawback is the representativeness of field research site selection as most studies were carried out in developed countries and very few in developing countries. Therefore, more attention should be paid to developing countries, especially some African and Asian countries. At the same time, new modified mathematical models should be utilized to process and integrate the data from various facilities and regions. Finally, we suggested that governments and organizations across the world should be united to wrestle with the impact of climate change on food security.

Persistent organic pollutant (POPs) contamination was analyzed in samples collected from wild and captive giant pandas to characterize seasonal variation in concentrations of POPs and possible sources. POP concentrations in bamboo and fecal samples collected from captive pandas showed significant fluctuations compared with those collected from wild pandas in each season. The highest polychlorinated biphenyl (PCB) and organochlorine pesticide (OCP) concentrations were 1380 pg g−1 dw and 3140 pg g−1 dw, respectively, which were observed in captive bamboo samples in the summer. PCBs varied seasonally, whereas OCPs did not show apparent seasonal variation. Based on the seasonal variability, component analysis, and the positive matrix factorization results, we determined that the secondary volatilization of POPs during periods of high temperatures was the leading cause of the exposure of pandas to pollutants (45%), and atmospheric transport played a crucial role in the secondary distribution of pollutants in panda food. The other two sources of pollution were historical residues transmitted over long distances to protected areas (28%), as well as UP-POPs and new inputs from agricultural activities (27%). The concentrations of pollutants in bamboo shoots were significantly lower than those in bamboo. Therefore, bamboo shoots should be incorporated into the diet of captive pandas in the spring to reduce their exposure to pollutants. The absorption capacity of pollutants associated with the consumption of bamboo shoots was significantly lower than that associated with the consumption of bamboo. The diet of young captive pandas in the summer should also be managed with caution given their slightly stronger ability to absorb pollutants.

To understand the history and research status of monsoon scholarly publication, a scientometrics study based on the Science Citation Index-Expanded has been made from 1900 to 2018. HistCite was used to analyze the annual outputs, countries, institutions, categories, journals, citations, and high-impact authors in the field. Results revealed that the annual number of outputs showed an increasing trend over the past decades, with an especially rapid increase after 1990. The country with the greatest academic impact was the USA, while the institution with the highest research outputs and citations was the Chinese Academy of Sciences. Meteorology Atmospheric Sciences and Geology were the most dominant categories. Journal of Climate was the most active journal, of which both in outputs and citations. Through citation visualization analysis, monsoon research mainly focuses on two aspects: modern monsoon and paleomonsoon.

To mitigate air pollution problem, the government has been planning to build more liquefied petroleum gas stations to motivate drivers to use liquefied petroleum gas vehicles in Taiwan. Such facility allocation problem is a multi-objective optimization process considering spatial variation in the need of refueling. This study presents a stochastic multi-objective optimization model for liquefied petroleum gas station allocation (SMOMLSA) that integrates a nondominated sorting genetic algorithm II with a Monte Carlo simulation to optimally allocate liquefied petroleum gas stations according to three trade-off objectives, including investment performance, energy conversion, and business opportunity. Monte Carlo simulation procedure generates the starting location of a taxicab car in need of refueling in the spatial grid based on a probability distribution. Nondominated sorting genetic algorithm II resolves the station location problem with these multi-objectives. The SMOMLSA was validated by conducting a real-world case study. Result depicts that the SMOMLSA can provide information on the optimal allocation of liquefied petroleum gas stations for minimizing construction costs, minimizing average refueling distance for vehicles, and maximizing potential customers.

Crested ibis (Nipponia nippon), one of the rarest birds in the world, was almost extinct in the historically widespread areas partly due to the environmental pollution. Therefore, non-invasive indicators of feather, eggshell, and excrement were used to investigate the exposure of this endangered bird to eleven trace elements in this study. The results indicated that crested ibises under in situ and ex situ conservations were diversely exposed to trace elements, with higher exposure levels of As, Cd, and Mn in the wild, but higher exposure levels of Hg, Se, and Zn in the captive breeding center. In addition, concentrations of As, Co, Cr, and Ni were significantly greater in the sediments of three types of foraging habitats for wild crested ibis, but concentration of Se was greater in the soil of captive cages. Feather and eggshell of crested ibis exhibited a very consistent indication for most of the trace elements, and concentrations of almost all of the elements in the excrements were very consistent with the results in the environmental samples (sediments or soils). Concentrations of As, Hg, Mn, and Zn in feathers, and Mn and Zn in eggshells of wild and captive crested ibis were greater than those in other similar species. Moreover, As, Cd, Cu, and Mn concentrations in excrement of wild crested ibises were greater than that in captive individuals and other species, but Se and Zn concentrations in excrement of captive crested ibises were greater than that of the wild and other species. The present study provided evidence that both of the wild and captive crested ibis were exposed to trace elements, which may be harmful to their health.

Grassland afforestation strongly influences the structure and function of soil microorganisms. Yet the mechanisms of how afforestation could simultaneously alter both the soil fungal and bacterial communities and its implications for ecosystem management are poorly understood, especially in nitrogen-limited ecosystems. Using high-throughput sequencing of 16S rRNA and ITS rRNA genes, the present study investigated the changes in soil properties and soil microorganisms after afforestation of natural grasslands with Chinese pine (Pinus tabuliformis) on the Loess Plateau in China. Results showed that soil bacterial diversity had no significant differences among the grassland (GL), forest-grassland transition zone (TZ), and forestland (FL), while soil fungal diversity in the GL was significantly higher than that in the FL and TZ (P 0.05). The proportion of shared OTUs in the soil bacterial community was higher than that in the soil fungal community among the three land use types. The dominant bacterial phylum shifted from Proteobacteria to Actinobacteria, while the dominant fungal phylum shifted from Ascomycota to Basidiomycota after the GL conversion to the FL. The functional groups of ECM fungi increased significantly while biotrophic fungi decreased significantly after grassland afforestation. Both the soil bacterial and fungal communities in the TZ showed great similarity with those in the FL. In addition, among all examined soil properties, soil nitrogen (N) showed a more significant effect on the soil microbial communities. The reduction of soil N after grassland afforestation resulted in both the structure and function changes in soil microbial communities. Our results demonstrated simultaneously differential changes in the composition and diversity of both soil bacterial and fungal communities after afforestation from grasslands to planted forests.

A megaproject, “Gully Land Consolidation” (GLC), was launched on the Chinese Loess Plateau in 2011 to combat land degradation and create new farmlands. However, the newly-claimed farmlands have suffered soil salinization, and the mechanism of salinization has been inadequately addressed. In this study, a creek valley that completed the GLC project in 2014 was selected and a total of 17 new farmlands along the reshaped creek valley were investigated in 2016. Moreover, the hillslope farmlands without conducting the GLC project, the loess parent material that used to fill the stream channel, and the water samples from reservoirs, irrigation wells and spring outlets in the reshaped valley were sampled and chemically analyzed. We found that the new farmlands experienced a significant rise in the water table that played a key role on soil salinization. After GLC, >80% of the sampling sites (14 out of 17) showed a shallow depth (≤3 m) from the soil surface down to the groundwater influence zone. Moreover, the closer the downstream farmlands to the newly-built dams, the shallower the water table, indicating considerable impacts of constructing dams on the water table depth in the near-dam farmlands. At the study site, more than half of the shallow soil samples (20 cm depth) underwent light soil salinization, and the most significant soil salinity occurred in the upper stream farmlands. The interplay between a near-surface water table, a high evaporation rate, landform features, and the alkaline properties of the loessial soil has led to the significant soil salinization in the upper stream of the valley. To mitigate soil salinization and ensure the sustainability of the newly-created farmlands in GLC, drainage trenches in the downstream near-dam farmlands and the drainage canals on both sides of the reshaped valley are suggested, which improve the drainage efficiency and control the water table depth.

Human RNA exoribonuclease 2 (Rexo2) is an evolutionarily conserved 3′-to-5′ DEDDh-family exonuclease located primarily in mitochondria. Rexo2 degrades small RNA oligonucleotides of Escherichia coli Oligoribonuclease (ORN), suggesting that it plays a role in RNA turnover in mitochondria. However, how Rexo2 preferentially binds and degrades nanoRNA remains elusive. Here, we show that Rexo2 binds small RNA and DNA oligonucleotides with the highest affinity, and it is most robust in degrading small nanoRNA into mononucleotides in the presence of magnesium ions. We further determined three crystal structures of Rexo2 in complex with single-stranded RNA or DNA at resolutions of 1.8–2.2 Å. Rexo2 forms a homodimer and interacts mainly with the last two 3′-end nucleobases of substrates by hydrophobic and π−π stacking interactions via Leu53, Trp96, and Tyr164, signifying its preference in binding and degrading short oligonucleotides without sequence specificity. Crystal structure of Rexo2 is highly similar to that of the RNA-degrading enzyme ORN, revealing a two-magnesium-ion-dependent hydrolysis mechanism. This study thus provides the molecular basis for human Rexo2, showing how it binds and degrades nanoRNA into nucleoside monophosphates and plays a crucial role in RNA salvage pathways in mammalian mitochondria.

Validation in vehicle engineering identifies and quantifies the differences between simulation models and experiment data. In this work we consider these differencesthe lack of ability to model uncertainties and to identify unknown parameters values, especially for coupled complex systems such as vehicles. Effects of unknown model parameters vary under different maneuvers and the ability to excite a source of uncertainty is the focus of this study. We propose an optimization method to generate a proper maneuver that maximize the sensitivity of uncertain parameters based on global sensitivity analysis (GSA). Sensitivities with respect to individual uncertain parameters and those that consider coupled effects are all included. We utilize Kriging-based metamodels to improve the efficiency of the GSA problems with computationally expensive simulations. The optimal design of excitation maneuvers to create the most sensitive performances can then be obtained. The applicability and the accuracy of the proposed method are assessed via a math model and a practical application on a x-by-wire autonomous tricycle. Results show that our proposed method can assist in providing a suitable maneuver as an alternative validation to uncertain parameters in a vehicle system.

Diastolic dysfunction is an emerging challenge among hemodialysis (HD) patients, and the associations between serum zinc with echocardiographic parameters and diastolic function remain uncertain. A total of 185 maintenance HD patients were stratified by the tertiles of serum zinc level to compare their clinical characteristics and echocardiography. Correlations of serum zinc levels with echocardiographic parameters were examined using Pearson’s analysis. Univariate and multivariate logistic regression analyses were performed to investigate the determinants of E/e’ ratio &gt, 15 and left atrial volume index (LAVI) &gt, 34 mL/m2, both indicators of diastolic dysfunction. Patients belonging to the first tertile of serum zinc level had a significantly higher E/e’ ratio and LAVI. Serum zinc levels were negatively correlated with E (r = −0.204, p = 0.005), E/e’ ratio (r = −0.217, p = 0.003), and LAVI (r = −0.197, p = 0.007). In a multivariate analysis, older age, diabetes, coronary artery disease, and lower serum zinc levels (OR = 0.974, 95% CI = 0.950–0.999, p = 0.039) were significantly associated with E/e’ ratio &gt, 15. Furthermore, diabetes and lower serum zinc levels (OR = 0.978, 95% CI = 0.958–0.999, p = 0.041) were significantly associated with LAVI &gt, 34 mL/m2. Reduced serum zinc level was significantly associated with diastolic dysfunction among HD patients. Further prospective studies are warranted to investigate whether zinc supplementation can attenuate cardiac dysfunction in maintenance HD patients.

This paper proposed an intelligent plant disease detection system that uses the modified Alexnet. As a convolutional neural network (CNN), Alexnet is good at objection detection from images. However, plant disease detection by the leaf image is a difficult task compared to traditional object detection. Therefore, Alexnet is modified for the different size images and angles to fit the requirement of a practical scenario. A training corpus gathered from the internet and with human leveling is used to train the detection models. According to the experimental results, the proposed approach achieves a good performance.

Fifteen chalcone derivatives 3a-3o were synthesized, and evaluated as multifunctional agents against Alzheimer's disease. In vitro studies revealed that these compounds inhibited self-induced Aβ1-42 aggregation effectively ranged from 45.9-94.5 % at 20 μM, and acted as potential antioxidants. Their structure-activity relationships were summarized. In particular, (2E)-3-[4-(dimethylamino)phenyl]-1-(pyridin-2-yl)prop-2-en-1-one (3g) exhibited an excellent inhibitory activity of 94.5 % at 20 μM, and it could disassemble the self-induced Aβ1-42 aggregation fibrils with ratio of 57.1 % at 20 μM concentration. In addition, compound 3g displayed good chelating ability for Cu2+ , and could effectively inhibit and disaggregate Cu2+ -induced Aβ aggregation. Moreover, compound 3g exerted low cytotoxicity, significantly reversed Aβ1-42 -induced SH-SY5Y cell damage. More importantly, compound 3g remarkably ameliorated scopolamine-induced memory impairment in mice. In summary, all the results revealed compound 3g was a potential multifunctional agent for AD therapy.

Simulation models play crucial roles in efficient product development cycles, therefore many studies aim to improve the confidence of a model during the validation stage. In this research, we proposed a dynamic model validation to provide accurate parameter settings for minimal output errors between simulation models and real model experiments. The optimal operations for setting parameters are developed to maximize the effects by specific model parameters while minimizing interactions. To manage the excessive costs associated with simulations of complex systems, we propose a procedure with three main features: the optimal excitation based on global sensitivity analysis (GSA) is done via metamodel techniques, for estimating parameters with the polynomial chaos-based Kalman filter, and validating the updated model based on hypothesis testing. An illustrative mathematical model was used to demonstrate the detail processes in our proposed method. We also apply our method on a vehicle dynamic case with a composite maneuver for exciting unknown model parameters such as inertial and coefficients of the tire model; the unknown model parameters were successfully estimated within a 95% credible interval. The contributions of this research are also underscored through multiple cases.

BACKGROUND Acupuncture, which has many good effects and few adverse effects, is widely recognized as an alternative therapy for depression in clinical practice. This study aimed to explore the mechanism of acupuncture in antidepressant treatment. MATERIAL AND METHODS In this experiment, Sprague-Dawley rats were randomly divided into 4 groups: control, chronic unpredictable mild stress (CUMS), acupuncture, and fluoxetine groups. The CUMS, acupuncture, and fluoxetine groups were orphaned and subjected to chronic unpredictable stress for 6 weeks, and the acupuncture and fluoxetine groups were treated with their respective intervention in weeks 4-6. The body weight of rats was monitored weekly. After behavioral tests were completed, serum, feces, and hippocampal tissue of rats were collected. RESULTS The results showed that the acupuncture and fluoxetine treatments could alleviate the behavioral changes caused by CUMS. The treatments increased the total distance of rat crossing in the open-field test, prolonged the activity time of the open cross maze in the open arm, and improved the rate of sucrose consumption in the sucrose preference test. In addition, both the decreased level of dopamine (DA) and 5-hydroxytryptamine (5-HT) in serum and hippocampus caused by CUMS were improved after the treatments with acupuncture and fluoxetine, and the decreased expression of brain-derived neurotrophic factor signaling and the astrocytes in the hippocampus caused by CUMS were increased after the treatments with acupuncture and fluoxetine. Acupuncture and fluoxetine also decreased the ß isoform of calmodulin-dependent protein kinase II in the hippocampus, which was increased by CUMS. Furthermore, acupuncture regulated intestinal microbial disorders caused by CUMS, which reduced the relative abundance ratio of Bacteroidetes/Firmicutes in rats. CONCLUSIONS Our experimental results indicate that acupuncture can alleviate depression-like performance in CUMS rats by regulating intestinal microbes and neurotransmitters.

The SARS-CoV-2 replication and transcription complex (RTC) comprising nonstructural protein (nsp) 2-16 plays crucial roles in viral replication, reducing the efficacy of broad-spectrum nucleoside analog drugs such as remdesivir and in evading innate immune responses. Most studies target a specific viral component of the RTC such as the main protease or the RNA-dependent RNA polymerase. In contrast, our strategy is to target multiple conserved domains of the RTC to prevent SARS-CoV-2 genome replication and to create a high barrier to viral resistance and/or evasion of antiviral drugs. We show that clinically-safe Zn-ejector drugs, disulfiram/ebselen, can target conserved Zn2+-sites in SARS-CoV-2 nsp13 and nsp14 and inhibit nsp13 ATPase and nsp14 exoribonuclease activities. As the SARS-CoV-2 nsp14 domain targeted by disulfiram/ebselen is involved in RNA fidelity control, our strategy allows coupling of the Zn-ejector drug with a broad-spectrum nucleoside analog that would otherwise be excised by the nsp14 proofreading domain. As proof-of-concept, we show that disulfiram/ebselen, when combined with remdesivir, can synergistically inhibit SARS-CoV-2 replication in Vero E6 cells. We present a mechanism of action and the advantages of our multi-targeting strategy, which can be applied to any type of coronavirus with conserved Zn2+-sites.

Background: Acupuncture therapy in Traditional Chinese Medicine has been widely believed as a complementary treatment for depression with numerous clinic and research reports. However, mechanisms of neural plasticity about the positive effects of acupuncture still rarely reported. The purpose of our paper aim to investigate the efficiency of acupuncture in curing depressive-like behaviors and discuss the mechanism underling neural plasticity and its related proteins BDNF/PSD95/SYN/PKMZ in prefrontal cortex.Methods: 32 rats with the same baseline were divided into 4 groups at random: control group(8rats), model group(8rats), acupuncture group(8rats) and flouxetine group(8rats). Depression rat models were induced by chronic unpredictable mild stress combined with single cage isolation for 6 weeks. Acupuncture group and fluoxetine group then give 3-weeks treatment during the later 3weeks of modeling procedure. The open field test (OFT), elevated-plus-maze (EPM) and sucrose preference test (SPT) were executed to estimate the depressive behaviors. The number of nerve cells, the length of dendrites, and the spines density in prefrontal cortex were observed by Golgi staining. The expression of prefrontal cortex BDNF, PSD95, SYN and PKMZ protein were detected by western blot and their related mRNA were detected by RT-PCR.Results: Acupuncture could alleviate depressive-like behaviors and promote the recovery of the neural plasticity functions in prefrontal cortex, which exerted similar antidepressant effect as fluoxetine, increasing cells number, prolonging the dendrites length, enhancing the spine density. Conclusion: Acupuncture therapy could alleviate depressive-like behaviors which may contribute to the recovery of the neural plasticity functions and the repair of neural plasticity-related proteins BDNF/PSD95/SYN/PKMZ in the prefrontal cortex of CUMS-induced depressed rats.

Microbiome plays an important role in evaluating soil quality for sustainable agriculture. However, the suitability of biological indicators in reclaimed farmland is less understood. Using high-throughput sequencing, we evaluated the soil microbial community of the newly created farmland (NF) after reclamation with two local high-yield farmlands (slope farmland (SF), check-dam farmland (CF)) on the Loess Plateau. Soil enzyme activities and the amount of culturable microorganism were also quantified to assess the soil quality. Results showed that the microbial diversity, cultural microorganism abundance, and soil enzyme activities indicated poor soil quality in NF. The dominant bacterial phyla were Proteobacteria, Bacteroidetes, Acidobacteria, and Cyanobacteria. The abundance of Acidobacteria was significantly lower in NF (13.31%) than in SF (27.25%) and CF (27.91%). Soil enzyme activities had a significant correlation with the abundance of culturable microorganism, Proteobacteria and Bacteroidetes, soil organic matter, total nitrogen, cation exchange capacity, and pH, suggesting that soil microbes have driven the formation of nutrition and further mediated crop growth. Therefore, the application of bacterial fertilizers could be a potential way to improve the soil quality of reclaimed farmland for crop growth.
