microRNAs (miRNAs) are promising biomarkers for different pathological models because of their stable and detectable characters in biofluids. Here, we collected urine samples from 5 beagle dogs on the 3th, 6th, and 12th day in an acute kidney injury (AKI) caused by gentamycin. miRNA levels were measured with high-throughput sequencing and the results were then differentially investigated. Gene Ontology (GO) and KEGG pathway analysis were performed to analyze potential target genes corresponding to the differentially expressed miRNAs (DE-miRNAs). Relationships between hub genes and DE-miRNAs were analyzed with STRING and Cytoscape. We identified 234 DE-miRNAs 3, 6, and 12 days after gentamycin treatment (p < 0.05). Top 10 up- and down-regulated candidate target genes of DE-miRNAs were predicted by overlapping TargetScan and miRanda results). GO and KEGG analyses for DE-miRNAs demonstrated that the DE-miRNAs target genes are mainly involved in kidney injury-related pathways, such as the insulin signaling pathway, oxytocin signaling pathway, and hedgehog signaling pathway. The network of miRNA-hub genes suggests that miR-452, miR-106a, and 106b participate in regulating the largest number of hub genes. We evaluated the miRNA signature via a canine model built by gentamycin-caused acute kidney injury. Our results represent a valuable resource for evaluating miRNAs as biomarkers of renal toxicity.

Membrane protein engineering exhibits great potential for cell functionalization. Although genetic strategies are sophisticated for membrane protein engineering, there still exist some issues, including transgene insertional mutagenesis, laborious, complicated procedures, and low tunability. Herein, we report a DNA-templated anchoring of exogenous proteins on living cell membranes to realize programmable functionalization of living cells. Using DNA as a scaffold, the model cell membranes are readily modified with proteins, on which the density and ratio of proteins as well as their interactions can be precisely controlled through predictable DNA hybridization. Then, the natural killer (NK) cells were engineered to gain the ability to eliminate the immune checkpoint signaling at the NK-tumor synapse, which remarkably promoted NK cell activation in immunotherapy. Given the versatile functions of exogenous proteins and flexible designs of programmable DNA, this method has the potential to facilitate membrane-protein-based cell engineering and therapy.

A type of Al-MCM-41 supported dual imidazolium ionic liquids were constructed and efficiently used as catalysts for the synthesis of 2-oxazolidinones from epoxides, amines, and CO2. The influence of the different catalysts and reaction parameters on the catalytic behaviours was investigated. Al-MCM-41@ILTiCl5 was identified as the most excellent catalyst because it could efficiently promote the three-component cycloaddition of CO2, epoxide, and amines to form the corresponding 2-oxazolidinones in high to excellent yields (84∼96%) with excellent selectivities (98∼99.7%). In addition, the recovery and reuse performances of Al-MCM-41@ILTiCl5 were examined. The catalyst could be recovered by simple filtration and reused six times without a change in the catalytic activity. Green reaction conditions, operational simplicity, feasibility, and sustainability of the functionalized catalyst are the main highlights of the present protocol.

Transcutaneous electrical cranial-auricular acupoint stimulation (TECAS) is a novel non-invasive therapy that stimulates acupoints innervated by the trigeminal and auricular vagus nerves. An assessor-blinded, randomized, non-inferiority trial was designed to compare the efficacy of TECAS and escitalopram in mild-to-moderate major depressive disorder.468 participants received 2 TECAS sessions per day at home (n = 233) or approximately 10-13 mg/day escitalopram (n = 235) for 8 weeks plus 4-week follow-up. The primary outcome was clinical response, defined as a baseline-to-endpoint ≥50% reduction in Montgomery-Åsberg Depression Rating Scale (MADRS) score. Secondary outcomes included remission rate, changes in the severity of depression, anxiety, sleep and life quality.The response rate was 66.4% on TECAS and 63.2% on escitalopram with a 3.2% difference (95% confidence interval [CI], -5.9% to 12.9%) in intention-to-treat analysis, and 68.5% versus 66.2% with a 2.3% difference (95% CI, -6.9% to 11.4%) in per-protocol analysis. The lower limit of 95% CI of the differences fell within the prespecified non-inferiority margin of -10% (P ≤ 0.004 for non-inferiority). Most secondary outcomes did not differ between the two groups. TECAS-treated participants who experienced psychological trauma displayed a markedly greater response than those without traumatic experience (81.3% versus 62.1%, P = 0.013). TECAS caused much fewer adverse events than escitalopram.TECAS was comparable to escitalopram in improving depression and related symptoms, with high acceptability, better safety profile, and particular efficacy in reducing trauma-associated depression. Early diagnosis and therapy are clinically crucial in decreasing mortality from breast carcinoma. However, the existing probes have difficulty in accurately identifying the margins and contours of breast carcinoma due to poor sensitivity and specificity. The shoulders with critical shoulder angle (CSA) of greater than 33-35° are associated with rotator cuff tears, whereas a CSA of less than 30° is likely to be osteoarthritic. However, anterior acromioplasty or lateral acromioplasty could not reduce high CSAs to the desired range (30-33°), with satisfactory accuracy and efficacy. Thus, we introduce a computer image-guided precise acromioplasty (CIG-PAP) technique, an individualized treatment based on three-dimensional planning. We believe that the introduction of this technique will provide an alternative approach to reduce a large CSA to the desired range (30-33°).

Multiple studies have demonstrated that neoadjuvant chemotherapy (NACT) can prolong the overall survival of pancreatic ductal adenocarcinoma (PDAC) patients. However, most studies have focused on open surgery following NACT.To investigate the efficacy and safety of laparoscopic radical resection following NACT for PDAC.We retrospectively analyzed the clinical data of 15 patients with pathologically confirmed PDAC who received NACT followed by laparoscopic radical surgery in our hospital from December 2019 to April 2022. All patients underwent abdominal contrast-enhanced computed tomography (CT) and positron emission tomography-CT before surgery to accurately assess tumor stage and exclude distant metastasis.All 15 patients with pancreatic cancer were successfully converted to surgical resection after NACT, including 8 patients with pancreatic head cancer and 7 patients with pancreatic body and tail cancer. Among them, 13 patients received the nab-paclitaxel plus gemcitabine regimen (gemcitabine 1000 mg/mLaparoscopic radical resection of PDAC after NACT is safe and effective if it is performed by a surgeon with rich experience in LPD and in a large center of pancreatic surgery.

Volatile organic compounds (VOCs) are key precursors of ozone and particulate matter, which are the two dominant air pollutants in urban environments. However, compositions and sources of VOCs in urban air aloft have rarely been reported so far. To address this matter, highly time-resolved measurements of VOCs were made by a proton-transfer-reaction time-of-flight mass spectrometer (PTR-ToF-MS) at a 450 m platform on the Canton Tower in Guangzhou, China. A combination of in situ measurements and modeling techniques was used to characterize variations in and sources of VOCs. Five sources were identified from positive matrix factorization (PMF) analysis, namely daytime mixed (e.g., biogenic emissions and secondary formation), visitor-related (e.g., human breath, cooking, and volatilization of ethanol-containing products), vehicular–industrial, regional transport, and volatile chemical product (VCP) (i.e., volatilization of personal care products), contributing on average to 21 %, 30 %, 28 %, 10 %, and 11 % of total VOC (TVOC) mixing ratios, respectively. We observe that contributions of the visitor-related source, mainly composed of ethanol, followed with the variation in visitor numbers on the tower well. The VCP-dominated source only had an average contribution of ∼5.7 ppb during the campaign, accounting for a small fraction (11 %) of TVOC mixing ratios but a large fraction (22 %) of the total OH reactivity. However, large fractions of reactive VOC species, e.g., monoterpenes (49 %), were attributed to the VCP-dominated source, indicating important contributions of VCPs to ambient concentrations of these species in urban environments. Vertical profiles of air pollutants (namely NOx, ozone, Ox, and PM2.5), measured at 5, 118, 168, and 488 m, exhibited more evident gradients at night than in the daytime owing to the stronger stability of the nocturnal boundary layer. Mixing ratios of VOC species during the nighttime generally decreased with time when the 450 m platform was located in the nocturnal residual layer and markedly increased when impacted by emissions at ground level. The results in this study demonstrated composition characteristics and sources of VOCs in urban air aloft, which could provide valuable implications in making strategies for control of VOCs and secondary air pollutants.

Bone Marrow Mesenchymal Stem Cells (BMSCs), multidirectional cells with self-renewal capacity, can differentiate into many cell types and play essential roles in tissue healing and regenerative medicine. Cell experiments and in vivo research in animal models have shown that BMSCs can repair degenerative discs by promoting cell proliferation and expressing Extracellular Matrix (ECM) components, such as type II collagen and protein-polysaccharides. Delaying or reversing the Intervertebral Disc Degeneration (IDD) process at an etiological level may be an effective strategy. However, despite increasingly in-depth research, some deficiencies in cell transplantation timing and strategy remain, preventing the clinical application of cell transplantation. Exosomes exhibit the characteristics of the mother cells from which they are secreted and can inhibit Nucleus Pulposus Cell (NPC) apoptosis and delay IDD through intercellular communication. Furthermore, the use of exosomes effectively avoids problems associated with cell transplantation, such as immune rejection. This manuscript introduces almost all of the BMSCs and exosomes derived from BMSCs (BMSCs-Exos) described in the IDD literature. Many challenges regarding the use of cell transplantation and therapeutic exosome intervention for IDD remain to be overcome.

Vehicular emissions are an important source for volatile organic compounds (VOCs) in urban and downwind regions. In this study, we conducted a chassis dynamometer study to investigate VOC emissions from vehicles using gasoline, diesel, and liquefied petroleum gas (LPG) as fuel. Time-resolved VOC emissions from vehicles are chemically characterized by a proton-transfer-reaction time-of-flight mass spectrometer (PTR-ToF-MS) with high frequency. Our results show that emission factors of VOCs generally decrease with the improvement of emission standards for gasoline vehicles, whereas variations in emission factors for diesel vehicles with emission standards are more diverse. Mass spectrum analysis of the PTR-ToF-MS suggests that cold starts significantly influence VOC emissions of gasoline vehicles, while the influences are less important for diesel vehicles. Large differences in VOC emissions between gasoline and diesel vehicles are observed with emission factors of most VOC species from diesel vehicles being higher than gasoline vehicles, especially for most oxygenated volatile organic compounds (OVOCs) and heavier aromatics. These results indicate quantification of heavier species by the PTR-ToF-MS may be important in the characterization of vehicular exhausts. Our results suggest that VOC pairs (e.g., C14 aromatics / toluene ratio) could potentially provide good indicators for distinguishing emissions from gasoline and diesel vehicles. The fractions of OVOCs in total VOC emissions are determined by combining measurements of hydrocarbons from canisters and online observations of the PTR-ToF-MS. We show that OVOCs contribute 9.4 % ± 5.6 % of total VOC emissions for gasoline vehicles, while the fractions are significantly higher for diesel vehicles (52 %–71 %), highlighting the importance of detecting these OVOC species in diesel emissions. Our study demonstrated that the large number of OVOC species measured by the PTR-ToF-MS are important in the characterization of VOC emissions from vehicles.

Hypothalamic agouti-related peptide and neuropeptide Y-expressing (AgRP) neurons have a critical role in both feeding and non-feeding behaviors of newborn, adolescent, and adult mice, suggesting their broad modulatory impact on brain functions. Here we show that constitutive impairment of AgRP neurons or their peripubertal chemogenetic inhibition resulted in both a numerical and functional reduction of neurons in the medial prefrontal cortex (mPFC) of mice. These changes were accompanied by alteration of oscillatory network activity in mPFC, impaired sensorimotor gating, and altered ambulatory behavior that could be reversed by the administration of clozapine, a non-selective dopamine receptor antagonist. The observed AgRP effects are transduced to mPFC in part via dopaminergic neurons in the ventral tegmental area and may also be conveyed by medial thalamic neurons. Our results unmasked a previously unsuspected role for hypothalamic AgRP neurons in control of neuronal pathways that regulate higher-order brain functions during development and in adulthood.

In order to overcome the demerits of traditional city air terminals (CAT), a brand-new service mode based on urban mobile stations (UMS) is introduced in this article for providing the luggage check-in service in urban area. Different from CAT, for each service time period, the UMSs are located based on the real-time passenger distributions. The objectives are to improve the service quality and enhance the competitiveness of air transportation. First, the system components and operation mode of UMS are described. Second, a mathematical model is set up for locating UMSs and assigning passengers to service stations. Three aspects, including the average path length from passengers to UMSs, the maximum acceptable path length for passengers, and the maximum service capacity of a station are considered. Third, to meet the strict time requirement of the UMS system, an effective evolutionary algorithm for optimizing UMS locations in real time is further developed. In the method, the ripple-spreading algorithm is applied for solving the many-to-many path optimization problem and a two-stage adaptive genetic algorithm is utilised for efficiently locating UMSs. Finally, a test case is set up based on the city centre of Tianjin, China, and passenger source locations are obtained by surveying passengers at Tianjin Airport. The reported method is then applied to dynamically locate UMSs in Tianjin urban area and it is compared to some other existing methods to show its effectiveness and efficiency. The service quality of UMS is compared to CAT in both cases with realistic data and randomly generated data to show its advantages.
