A focus of recent research is to understand the role of our own response goals in the selection of information that will be encoded in episodic memory. For example, if we respond to a target in the presence of distractors, an important aspect under study is whether the distractor and the target share a common response (congruent) or not (incongruent). Some studies have found that congruent objects tend to be grouped together and stored in episodic memory, whereas other studies found that targets in the presence of incongruent distractors are remembered better. Our current research seems to support both views. We used a Tulving-based definition of episodic memory to differentiate memory from episodic and non-episodic traces. In this task, participants first had to classify a blue object as human or animal (target) which appeared in the presence of a green one (distractor) that could belong to the same category as the target (congruent); to the opposite one (incongruent); or to an irrelevant one (neutral). Later they had to report the identity (What), location (Where) and time (When) of both target objects (which had been previously responded to) and distractors (which had been ignored). Episodic memory was inferred when the three scene properties (identity, location and time) were correct. The measure of non-episodic memory consisted of those trials in which the identity was correctly remembered, but not the location or time. Our results show that episodic memory for congruent stimuli is significantly superior to that for incongruent ones. In sharp contrast, non-episodic measures found superior memory for targets in the presence of incongruent distractors. Our results demonstrate that response compatibility affects the encoding of episodic and non-episodic memory traces in different ways.

Intuitive interpretation tables and a decision tree will help strengthen international data comparison for the identification of reported missing individuals discovered outside of their national borders.

Background and purpose Recent crystal structures of GPCRs have emphasized the previously unappreciated role of the second extracellular (E2) loop in ligand binding and gating and receptor activation. Here, we have assessed the role of the E2 loop in the activation of the melatonin MT1 receptor and in the inactivation of the closely related orphan receptor GPR50. Experimental approach Chimeric MT1 -GPR50 receptors were generated and functionally analysed in terms of 2-[125 I]iodomelatonin binding, Gi /cAMP signalling and β-arrestin2 recruitment. We also used computational molecular dynamics (MD) simulations. Key results MD simulations of 300 ns revealed (i) the tight hairpin structure of the E2 loop of the MT1 receptor (ii) the most suitable features for melatonin binding in MT1 receptors and (iii) major predicted rearrangements upon MT1 receptor activation, stabilizing interaction networks between Phe179 or Gln181 in the E2 loop and transmembrane helixes 5 and 6. Functional assays confirmed these predictions, because reciprocal replacement of MT1 and GPR50 residues/domains led to the predicted loss- and gain-of-melatonin action of MT1 receptors and GPR50 respectively. Conclusions and implications Our work demonstrated the crucial role of the E2 loop for MT1 receptor and GPR50 function by proposing a model in which the E2 loop is important in stabilizing active MT1 receptor conformations and by showing how evolutionary processes appear to have selected for modifications in the E2 loop in order to make GPR50 unresponsive to melatonin. Dans la lignee des travaux de Dominique Cotte et Marie Despres-Lonnet, l’article pose la question du lieu de la donnee au sein des ressources numeriques et ambitionne d’y repondre en procedant a une description de ce que le mot locator de la ressource designe, etant a la fois son adresse et le moyen d’y acceder. L’adresse du serveur, le chemin a l’interieur du disque puis l’ensemble des moyens de denomination et de description fine des markup languages permettent de construire une image fidele de l’information documentaire au sein de collections qui autorisent de nouvelles strategies pour la fouille de donnees. Quoi qu’il en soit, la fouille textuelle et l’analyse quantitative demeurent des positionnements ideologiques quand seuls l’intelligence et le discernement humains peuvent en dernier lieu en apprecier la richesse et en valider la pertinence.

There is some debate as to whether responding to objects in our environment improves episodic memory or does not impact it. Some authors claim that actively encoding objects improves their representation in episodic memory. Conversely, episodic memory has also been shown to improve in passive conditions, suggesting that the action itself could interfere with the encoding process. This study looks at the impact of attention and action on episodic memory using a novel what-where-when (WWW) task that includes information about object identity (what) and spatial (where) and temporal (when) properties. With this approach, we studied the episodic memory of 2 types of objects: a target, where attention or an action is defined, and a distractor, an object to be ignored, following 2 selective states: active versus passive selection. When targets were actively selected, we found no evidence of episodic memory enhancement compared to passive selection; instead, memory from irrelevant sources was suppressed. The pattern was replicated across a 2-D static display and a more realistic 3-D virtual environment. This selective attention effect on episodic memory was not observed on nonepisodic measures, demonstrating a link between attention and the encoding of episodic experiences. Massively parallel sequencing (MPS) applications in forensic science highlight the advantages of this technique compared to capillary electrophoresis (CE). The multiplexing of a wide range of genetic markers and access to the full amplicon sequence, allowing the detection of isoalleles, make it a very promising tool which could be applied to the most challenging casework DNA samples. However, the complexity of the manual library preparation protocol, potential DNA contamination and sample tracking issues are the main reasons why forensic scientists still hesitate to implement MPS analytical workflows in their laboratory. In the criminal framework, the analysis of approximately 20 DNA microsatellites enables the establishment of a genetic profile with a high statistical power of discrimination. This technique gives us the possibility to establish or exclude a match between a biological trace detected at a crime scene and a suspect whose DNA was collected via an oral swab. However, conventional techniques do tend to complexify the interpretation of complex DNA samples, such as degraded DNA and mixture DNA. The aim of this review is to highlight the powerness of new forensic DNA methods (including high-throughput sequencing or single-cell sequencing) to facilitate the interpretation of the expert with full compliance with existing french legislation.

The relevance of CB(2)-mediated therapeutics is well established in the treatment of pain, neurodegenerative and gastrointestinal tract disorders. Recent works such as the crystallization of class-A G-protein-coupled receptors in a range of active states and the identification of specific anchoring sites for CB(2) agonists challenged us to design a reliable agonist-bound homology model of CB(2) receptor. Docking-scoring enrichment tests of a high-throughput virtual screening of 140 compounds led to 13 hits within the micromolar affinity range. Most of these hits behaved as CB(2) agonists, among which two novel full agonists emerged. Although the main challenge was a high-throughput docking run targeting an agonist-bound state of a CB(2) model, a prior 2D ligand-based Bayesian network was computed to enrich the input commercial library for 3D screening. The exclusive discovery of agonists illustrates the reliability of this agonist-bound state model for the identification of polar and aromatic amino acids as new agonist-modulated CB(2) features to be integrated in the wide activation pathway of G-protein-coupled receptors.

Au cours des années 1960 et 1970, deux émissions diffusées à la radio et à la télévision ont attiré l’attention du grand public sur le patrimoine et l’environnement. Les Chefs-d’œuvre en péril de Pierre de Lagarde et La France défigurée de Louis Bériot et Michel Péricard ont adopté un ton alarmiste et polémique adapté à la violence croissante du conflit entre modernité et respect du passé. Malgré la culture et le style différents de leurs réalisateurs, les deux émissions ont pris pour cibles les vandales à l’œuvre dans les administrations, les mairies et les entreprises. En utilisant les techniques du journalisme d’investigation, elles ont donné à voir les ravages causés par l’ignorance, la cupidité ou une conception étroite du progrès. Mais elles ont aussi cherché à promouvoir de bons exemples : propriétaires de vieilles demeures ou bénévoles sur les chantiers de restauration pour Pierre de Lagarde, architectes novateurs ou élus éclairés pour Louis Bériot et Michel Péricard. Interpellés par un public nombreux et curieux, les journalistes ont souhaité prolonger leur action à travers des publications, des concours et des associations. Le ministère des affaires culturelles, craignant le ton revendicatif des émissions, a cherché à leur donner un contenu plus positif. Les relations ont été particulièrement tendues avec Pierre de Lagarde, qui a repris des arguments souvent utilisés contre le service des monuments historiques et ses architectes. Le succès de ces émissions, très regardées jusqu’à la fin des années 1970, en ont fait des références pour le traitement du thème du patrimoine dans les médias audiovisuels. Chefs-d’œuvre en péril by Pierre de Lagarde and La France défigurée by Louis Bériot and Michel Péricard adopted an alarmist and polemical tone. Both accused the vandals in the administration, the town councils and the industry. With the tools of investigative reporting, they showed the devastation caused by ignorance, greed or a narrow conception of progress. But they also tried to promote good examples: owners of castles or volunteers for restoration, good architects or clever representatives. Beyond the TV screen, the journalists tried to share their ideas through publications, games and associations. The Culture ministry didn’t like the bitter tone of the emissions and tried to give them a more positive content. The relations were particularly bad with Pierre de Lagarde, because of his critics against heritage service and its architects. These emissions were popular till the end of 1970s. They still are reference points for the journalists who deal with the theme of heritage. The present article shows the originality of the old french translation of the Bible by exposing many linguistic facts of the adptation of the "Lingua sacra" concept to the old french language., Le présent article expose la composition originale de la page traduite du latin en ancien français en mettant en valeur les phénomènes linguistiques de traduction du texte biblique sacré en langue vulgaire.

Le présent article illustre les travaux du groupe d'études consacré à l'ancien français né dans le cadre de la réflexion de l'équipe LDI autour des problèmes de résolution automatique des problèmes linguistiques. Nous voudrions particulièrement présenter les prémices théoriques qui fondent notre travail de création de ressources au sein de la base G.R.A.A.L consacré à l'étude et au traitement automatique de l'ancien français.

La présente contribution publiée dans le rapport général du corrigé de l'Agrégation de lettre modernes propose une définition linguistique de "l'expression de l'ordre et du souhait".

Le présent article s'inscrit dans la continuité de la réflexion que nous avions entreprise en Italie alors que nous posions les bases d'une étude linguistique des procédés à l'oeuvre dans le discours des traductions des Bibles au Moyen Âge, et de leur rapport avec la lexicographie bilingue. Nous souhaiterions en effet poser le postulat suivant: c'est à travers le travail « sur le mot » que se manifeste la pensée « du mot » particulièrement opérante dans les traductions des clercs de l'Université médiévale. A partir de ce postulats fondamental, nous souhaiterions défendre l'idée qu'il existe un concept opératoire du mot dont nous nous efforcerons de montrer qu'elle est théorisée de manière indirecte dans l'oeuvre grammaticale d'Isidore de Séville, elle-même tributaire de la tradition grammaticale d'Aristote et de Donat. Enfin, nous nous efforcerons d'illustrer notre propos à travers le répertoire d'exemples de phénomènes de traduction propres à la traduction de la Bible en prose au Moyen Âge particulièrement propres à souligner cette pensée issue d'une tradition antique. Il existe donc une tradition du mot depuis l'Antiquité, mais une tradition qui ne dit pas son nom. En effet, l'idée même du « mot » conçue comme une unité minimale porteuse de sens indifféremment de sa nature et de sa relation aun discours ne se manifeste pas en tant que telle, en tant que théorie du sens.

A number of potentially bioactive molecules can be found in nature. In particular, marine organisms are a valuable source of bioactive compounds. The activity of an α-galactosylceramide was first discovered in 1993 via screening of a Japanese marine sponge (Agelas mauritanius). Very rapidly, a synthetic glycololipid analogue of this natural molecule was discovered, called KRN7000. Associated with the CD1d protein, this α-galactosylceramide 1 (KRN7000) interacts with the T-cell antigen receptor to form a ternary complex that yields T helper (Th) 1 and Th2 responses with opposing effects. In our work, we carried out molecular dynamics simulations (11.5 µs in total) involving eight different ligands (conducted in triplicate) in an effort to find out correlation at the molecular level, if any, between chemical modulation of 1 and the orientation of the known biological response, Th1 or Th2. Comparative investigations of human versus mouse and Th1 versus Th2 data have been carried out. A large set of analysis tools was employed including free energy landscapes. One major result is the identification of a specific conformational state of the sugar polar head, which could be correlated, in the present study, to the biological Th2 biased response. These theoretical tools provide a structural basis for predicting the very different dynamical behaviors of α-glycosphingolipids in CD1d and might aid in the future design of new analogues of 1., Author Summary To modulate the natural immune response toward aggressive (Th1) or protective (Th2) profiles remains a difficult challenge, but can also offer great therapeutic opportunities, particularly for the treatment of cancer or auto-immune diseases. It has been demonstrated that a particular type of cells, named invariant Natural Killer T (iNKT) cells, are able to induce both protective and aggressive response profiles, depending on the antigen that is presented to it by CD1d proteins. Since this discovery, efforts have been made to find synthetic compounds that would selectively induce Th1 or Th2 immune response. KRN7000 was the first to selectively induce a Th1 response, and for two decades many analogues of this compound were synthesized. Some of them effectively induce Th1- or Th2-biased responses. But, unfortunately, the Th1/Th2 selectivity mechanism remains unclear. That is the reason why we have undertaken large-scale molecular modeling (molecular dynamics) simulations of various CD1d-ligand systems with the aim to find out correlation between chemical modulation and the orientation of the biological response for a variety of known ligands.

Le présent article, après avoir exposé les liens qui existent entre certaines assertions du texte et la toponymie parisienne contemporaine de l'auteur démontre que "Les trois livre de la vie" de Ficin sont une de ses sources d'inspiration.

Le présent article abordera les particularités du fonctionnement de l'adverbe « derechief » dans la traduction du Livre de la Genèse de Guyart-des-Moulins au regard des particules présentes dans les textes antérieurs de la Bible, afin de démontrer que la dynamique du discours traduit réside dans l'expression d'une voix spécifiquement française introduite au sein de la polyphonie du texte sacré. Cette description participe à la définition du travail opéré par les traducteurs pour couler la poétique des textes anciens dans le « carcan de la syntaxe »

Since the discovery in 1995 of α-galactosylceramide 1 (α-GalCer), also known as KRN7000,1 hundreds of compounds have been synthesized in order to activate invariant natural killer T (iNKT) cells. Such keen interest for this lymphocyte cell type is due to its ability to produce different cytokines that bias the immune response toward a Th1 or Th2 profile. Thus, an understanding of the immune polarization mechanism via iNKT activation may pave the way toward new therapeutics in various domains including cancer and infectious and autoimmune diseases. In this review, we propose an up-to-date analysis of iNKT activators associated with a structure-activity relationship (SAR) study aimed at complementing available reviews by highlighting molecular bases for a selective immune response.
