Wire rope is a necessary tool in practical applications especially in crane, elevator and bridge construction, which plays an important role in the national economy and daily life, and safety inspection for wire rope is the key to ensure people’s life and property. However, detection signals are usually complicated due to the twining structures, which make the wire rope defect signal and strand signal mix together. What’s more, no reports and studies have appeared to solve this problem. In view of the situation and challenges above, this paper proposes a combined signal processing method based on notch filtering and wavelet denoising to process detected wire rope signals. Basic time domain, frequency domain and joint time–frequency analysis are first conducted, thereafter, conventional signal processing methods such as low-pass filtering and adaptive analysis are presented according to the signal characterizations. These comparisons and results demonstrate that a conventional single method is incapable of wire-rope-detection signal identification and differentiation. Nonetheless, after the notch filter design and calculation, the processing results for the typical wire rope inspection signals in the experiments indicate that the combined methods can not only distinguish steel wire rope defect signal and strand signal effectively but also with high detection accuracy, even for the inner defect. Finally, the feasibility and reliability are verified by a series of signal processing results and comparisons, which demonstrate that this new method has great application potential and is of vital significance to the development of wire rope safety inspection.

Land is a finite natural resource, the efficient management of which is vital for economic growth and development of the country. It is required for both agriculture and non-agricultural purposes. An analysis of structural changes in the land use pattern over a period of time provides the scope for planned and judicious management of land. In this connection, the present study on ‘An Economic analysis of temporal changes in land use pattern in Western Maharashtra’ was undertaken. For analysis the secondary sources and the necessary regional level time series data on area under land use categories were compiled from various issues of statistical abstract of Maharashtra and other publications of Government. The entire period of 1960-61 to 2019-20 was decomposed into three periods, viz; pre-liberalization, post-liberalization and overall period. A temporal change in land use pattern was estimated with the per cent change and compound growth rates. The temporal changes in land use dynamics of Western Maharashtra region revealed that, land put to non-agricultural uses, miscellaneous tree crops and groves, current fallow and fallows other than current fallow showed an increased trend while, area under forest, barren and uncultivable land, cultivable waste and net area sown registered a decline during Period-II. It is suggested that, during Period-II, the average proportion of forest land is 14.72 per cent, which is far less than the necessary percentage i.e., 33 per cent. Therefore, necessary steps may be taken by the state government in order to restore and maintain ecological balance. Land put to non-agricultural uses inclined throughout all the periods under study indicating an alarming situation. Majority of this conversion to land put to non-agricultural uses has taken place of agricultural land. Hence, the state government may prepare suitable policies controlling conversion of agricultural land, particularly.

Introduction Sarin is a highly toxic organophosphorus nerve agent that irreversibly inhibits neuronal enzyme acetylcholinesterase. In the prevailing scenario, it is of paramount importance to develop early diagnosis and medical countermeasures for sarin exposure. A deeper understanding of the molecular mechanism of sarin intoxication and perturbations in the associated cellular processes is likely to provide valuable clues for the elucidation of diagnostic markers and therapeutic targets for sarin exposure. Methods Present study, uncovered the changes in phosphorylation patterns of multiple proteins in different rat brain regions after sarin intoxication using 2-DE/MS approach. It provided a holistic view of the phosphorylation-mediated changes in the cellular proteome and highlighted various signaling and response pathways affected at an early time point of sarin intoxication. Results We found total 22 proteins in the cortex, 25 proteins in the corpus striatum, and 17 proteins in the hippocampus, showed ≥1.5 fold changes (hyper- or hypo- phosphorylated) with respect to control, either at 2.5 h or 1 d after sarin exposure. These results indicated the differential expression of phosphoproteins involved in protein folding in the endoplasmic reticulum, carbon metabolism, metabolic function, and energy metabolism. Conclusion Four candidates (protein disulfide-isomerase A3, heat shock cognate 71 kDa protein, alpha-enolase, and creatine kinase B-type), hyperphosphorylated in all three brain regions, can be further studied to understand the molecular mechanism behind neurodegenerative changes mediated by sarin exposure. The study sheds light on major pathogenic processes initiated during sarin intoxication and provides putative diagnostic markers/therapeutic targets for further validation.

Present work reports details of the design, fabrication, and laboratory testing of a natural draft metal biomass cookstove for a community kitchen. Heat transfer and fluid flow considerations along with experimental data available in the literature are used for the design of the community cookstove. The summation of pressure drop in different sections of the cookstove is compared with the outside pressure drop for the system using buoyancy considerations. The cookstove is fabricated as per the dimensions finalized during the design stage. Laboratory testing of the cookstove is conducted according to Bureau of Indian Standards (BIS) protocol. The tests are conducted under different conditions of main air hole closing viz. 0%, 25%, 75% and 100%. Each test is replicated thrice and an analysis of data is performed using a student’s t-test. The efficiency of cookstove is found to be about 42% i.e., well above the BIS limit of 25% for natural draft metal cookstove. The effect of insulation on the outer body of cookstove is studied experimentally. It is found that the efficiency enhances by about 5.3% due to insulation. The authors found that provision of primary and secondary air holes play a very important role in the performance of the cookstove. When 100% main air holes are closed, the average efficiency of the cookstove is found to be poor (27.3%) due to no supply of secondary air to the combustion chamber of the cookstove.

In view of this issue, there is need to identify a suitable alternative for its management. In this context, semiochemicals serve as one of the best eco-friendly options. These chemical substances or mixtures broadcast messages for the purpose of communication. Semiochemicals may be released from eggs, faecal pellets, tarsal segments, scales, anal tufts and/or any insect components that reduce the intra/interspecific competition for shared resources such as food, mating, oviposition site etc. Accurate identification of the semiochemical compounds and their proportion in natural blends that arouse a strong behavioural response in insects, requires a thorough knowledge. Therefore in the present study, we tried to explore the chemicals present in eggs, faecal pellets and whole-body wash extracts of mated female. Seven compounds were identified from a methanolic extract of faecal-pellet samples, namely hexadecanoic acid, 9,12-octadecadienoic acid, 9-octadecenoic acid and their methyl esters along with methyl stearate. However, only four methyl-esterified forms were found in common in eggs and female body wash extract. This clearly shows that the compounds remain constant and conserved for the species but the role of these compounds needs to be demonstrated.

Endoglucanase (EG) is a key enzyme during enzymatic preparation of cellulose nanocrystals (CNCs). Myceliophthora thermophila is a thermophilic fungus that has thermal properties and a high secretion of endoglucanases (EGs), and could serve as potential sources of EGs for the preparation of CNCs. In this work, four different GH families (GH5, GH7, GH12, and GH45) of EGs from M.thermophila were expressed and purified, and their enzymatic characteristics and feasibility of application in CNC preparation were investigated. It was shown that the MtEG5A from M. thermophila has good potential in the enzymatic preparation of CNCs using eucalyptus dissolving pulp as feedstock. It was also observed that there was a synergistic effect between the MtEG5A and other MtEGs in the preparation of CNCs, which improved the yield and properties of CNCs obtained by enzymatic hydrolysis. This study provides a reference for understanding the enzymatic characteristics of different families of EGs from M. thermophile and their potential application in nanocellulose production.

A systematic DoE and Analytical Quality by Design (AQbD) approach was utilized for the development and validation of a novel stability indicating high-performance thin–layer chromatographic (HPTLC) method for Rivaroxaban (RBN) estimation in bulk and marketed formulation. A D-optimal design was used to screen the effect of solvents, volume of solvents, time from spotting to development and time for development to scanning. ANOVA results and Pareto chart revealed that toluene, methanol, water and saturation time had an impact on retention time. The critical method and material attributes were further screened by Box-Behnken design (BBD) to achieve optimal chromatographic condition. A stress degradation study was carried out and structure of major alkaline degradant was elaborated. According to the design space, a control strategy was used with toluene: methanol: water (6:2:2) and the saturation time was 15 min. A retention factor (RF) of 0.59 ± 0.05 was achieved for RBN using chromatographic plate precoated with silica gel at detection wavelength 282 nm with optimized conditions. The linear calibration curve was achieved in the concentration range of 200–1,200 ng/band with r2 > 0.998 suggesting good coordination between analyte concentration and peak areas. The quadratic model was demonstrated as the best fit model and no interaction was noted between CMAs. The optimized HPTLC method was validated critically as stated in International Conference on Harmonization (ICH) Q2 (R1) guideline and implemented successfully for stress degradation study of RBN. The developed HPTLC method obtained through AQbD application was potentially able to resolve all degradants of RBN achieved through forced degradation study. The obtained results demonstrate that a scientific AQbD approach implementation in HPTLC method development and stress degradation study drastically minimizes the number of trials in experiments, ultimately time and cost of analysis could be minimized.

SAP2000 is a tool used for creating and analysing structural models. It offers many features across a single, customizable user interface, allowing engineers working on transportation, industrial, public works, or other facilities to generate complex models and run comprehensive tests. Earthquakes shake the ground surface, can cause buildings to collapse, disrupt transport and services, and can cause fires. They can trigger landslides and tsunami. Earthquakes occur mainly as a result of plate tectonics, which involves blocks of the Earth moving about the Earth's surface.

Earthquakes in active seismic regions usually occur in a series of medium to strong intensity ground motions at small intervals of times. A large intensity mainshock (MS) is often followed by a series of aftershocks (AS) or even preceded by smaller foreshocks. This sequence type of mainshock-aftershock (MS-AS) ground motions with varying intensity pose major seismic hazard as there is limited scope of repair and retrofitting between seismic events. Due to continuous and repeated seismic ground motions over a brief period of time, the damages in the structure gets accumulated and structure collapse. This study navigates the behaviour of reinforced concrete structures under such seismic sequences. For this purpose, the nonlinear response of three 12 storey reinforced concrete buildings (regular plan, mass irregularity and diaphragm irregularity) is evaluated. The buildings are subjected to five real seismic sequences from previous earthquakes. Nonlinear dynamic analyses are carried out to study the response of buildings under MS and MS-AS sequences considering: a) material and geometric non linearities and b) irregularities. A single highest aftershock is considered in the present study. The results in this study indicates that MS-AS seismic sequence considering both material and geometric nonlinearities has significant effect on the response of structure. It also showed that seismic sequences significantly alter the response of irregular structure.

Liquid storage tanks are strategically important due to their essential requirement of service in the post-earthquake situation. Numerical modeling of the liquid storage tank needs special attention and cannot be done in the same manner as that of the conventional buildings. In the present paper, a numerical simulation of the RC elevated liquid storage tank is presented. The staging of the tank is modeled as a multi-degree freedom system, and the container with contained liquid is modeled as a two-mass system. Free vibration analysis of the tank is carried out, and mode shapes are extracted. Further, to study the seismic response of the tank, nonlinear time history analysis is carried out. The tank is subjected to time histories of real earthquake ground motions. The varying level of the liquid in the container is another characteristic feature of tanks. The filled condition of the tank is taken into account by considering the aspect ratio (S), defined as the ratio of height of the liquid to the radius of the container. The response of the tanks with two different aspect ratios viz. 0.5 (broad) and 2.0 (slender) is studied. The linear modal analysis also carried out to understand the significance of nonlinear analysis, particularly in liquid storage tanks. Displacement, velocity, and acceleration response at the bracing levels, as well as at container levels, are obtained. Additionally, the base shear response is also obtained. The effect of aspect ratio on the free vibration analysis and the seismic response of the tanks are presented. Liquid storage tanks are special structures that have typically low fundamental natural frequencies. The nonlinear time history response of the tank showed that the higher displacement and velocity response occurs at the convective level. It is found that the linear modal analysis significantly underestimates the response of the liquid storage tank.

An earthquake is a catastrophe which is always a topic of concern for structural designers and civil technocrats. Not only it causes physical damage to the structures but also successful to impart psychological disorder in human minds for long-run. In order to mitigate the devastating effects of earthquake on society, it is eminent to study the dynamic characteristics in detailed manner i.e. to examine how the structure responds owing to the seismic characteristics. The various ground motion characteristic includes time-duration and velocity, frequency content and amplitude, displacement, incremental velocity and incremental displacement, peak ground accelerations (PGAs), etc. Out of these, effect of frequency content and maximum amplitude value of earthquake ground motions on the seismic response of structures is often underestimated. This paper investigates the effect of frequency content and maximum amplitude value on the seismic response of structures which is dominant of all characteristics. The study proceeds with recording time-history of acceleration obtained from conventional unidirectional harmonic shake table. Further, FFT (Fast Fourier Transform) analysis of applied time-history is done and the effect of frequency content and the maximum amplitude value of applied time-history on the seismic responses of structures is investigated and studied. For this, structures with varying dimensions (height, length, and width) are modelled and time-history analysis of structural models has been carried out in SAP2000v19. Seismic responses of analysed structures are represented in the form of fundamental natural frequency, storey displacement, and base shear. It is reported that out of many earthquake indices, frequency content and amplitude of earthquake ground motions are the most dominating. It is reported that the resonance phenomena occurs for the less height structure and thus it shows maximum displacement and base shear responses. Also, as height, length, and width of structure increases; displacement, base shear, and fundamental time-period of structure increases.

In recent years, intensive research works are concentrated on the development of semi-active control devices. A semi-active control system can also be explained as a system that mainly requires a small external power source for functioning and applies the motion of the structure to generate the control forces. A semi-active control system typically begins from a passive control system, which is followed by a change to permit the flexibility of mechanical properties. The mechanical properties of the semi-active control devices may be modified depending upon the response from the excitation and/or from the calculated reaction. Magneto-rheological (MR) dampers are a type of a semi-active control device that, in the presence of Magneto-rheological (MR) fluid, responds to the applied vibrations. This type of damper has copper winding in it, forming an electronic circuit internally with a combination of MR fluids. The effectiveness of MR damper greatly depends on the command voltage to the copper winding. In the present study, the response of MR Damper to the command voltage, and the excitation frequency is experimentally evaluated. MR damper is supported by a wall mounting rigid fixture and connected to a shake table by a fixing unit. The voltage is varied from 0 V - 10 V with an interval of 2 V. Command voltages is applied to the MR damper with the help of voltage regulator and digital multi meter. The effect of excitation frequency is also studied (1 Hz – 8 Hz). While conducting the experimental work, acceleration values are obtained through the Data acquisition system. These acceleration values are integrated to get the velocity and displacement response. The relation between the displacement, velocity, acceleration of the MR damper, and the frequency of shake table and command voltage is studied. From the experimental work, it is observed that the displacement response of the damper is effectively controlled by increasing the command voltage.

Background In clinical practice, decision about management of choledocholithiasis is driven by availability of resources and expertise, patients and healthcare professional preferences. This survey is aimed to describe the approach of physicians and surgeons for the management of choledocholithiasis. Method A 36-question online survey was conducted using Google Forms on various aspects of management of choledocholithiasis. Results The responses from 323 participants were included, of which 202 (62.54%) were physicians and 121 (37.46%) were surgeons. The proportion of responders who do not follow American or European Society of Gastrointestinal Endoscopy guidelines is associated with increasing age and experience of responders (p = 0.0001), while place of work (private vs. teaching) and broad specialty (physician vs surgeon) are not associated (p >0.05). For patients with high likelihood of choledocholithiasis, 123 (38.1%) participants prefer to do endoscopic ultrasound/magnetic resonance cholangiopancreatography (EUS/MRCP) rather than directly performing endoscopic retrograde cholangiopancreatography/intraoperative cholangiography (ERCP/IOC). For intermediate likelihood, MRCP is more commonly preferred compared with EUS, due to local availability (44%), expertise (39.6%), healthcare professionals preference (30.7%), and patients preference (17.3%). For difficult common bile duct (CBD) stones, short biliary sphincterotomy with large balloon sphincteroplasty (59.4%), followed by laparoscopic CBD exploration are commonly used approaches. Prophylactic CBD stent placement after ERCP and CBD clearance is common practice. Preoperative ERCP followed by cholecystectomy is more preferred approach than cholecystectomy and CBD exploration. Conclusion There is considerable variability in the management of choledocholithiasis. The practices such as use of EUS/MRCP for high likelihood group, use of prophylactic CBD stent placement after ERCP and CBD clearance, and use of single stage approach especially in patient with intermediate likelihood group should be addressed in future studies.

It is a cultured and automated form of Thin Layer Chromatography (TLC) and is based on the use of an optimized silica gel 60 with a significantly smaller particle size than which is used for TLC. The previously stated analytical methods are employed for purity control of chemicals, steroids, pesticides, and water analysis, water-soluble food dyes, vitamins, pesticides in vegetables, fruits, and other foodstuffs. The Performance of The RCC Building During Earthquake Depends Upon the Distribution of Stiffness Amongst The Storeys. If The Distribution Is Non- Uniform To A Large Extend, The Performance of The Building is Poor. The Storey Stiffness is Contributed By The Columns As Well As The Brick Infill. If The Effective Storey Stiffness of A Floor is Less Than 70% of the Storey Above Or Below It , Then It Is Called The Soft Storey. It Is Learnt From The Past Earthquakes That Soft Storey Are More Vulnerable To Failure. The Study of Building Performance During Past Earthquakes Has Revealed That Maximum Failures Have Occurred In The Parking Storey. Most of the buildings were damaged and collapse during in strong earthquake hence we will have to confirm safety against the earthquake force that are affecting the structure, and determine seismic responses of such building. In this project we will analyse the G+5 building through response spectrum analysis to develop the economic design. Computer software’s are also being used for the calculation of forces, bending moment, stress, strain & deformation or deflection for a complex structural system.
