Abstract

International audience; From an easy and scalable synthetic access to quinoline derivatives substituted by fluorinated groups in both C2-and C4-positions developed in our laboratory, we devised the synthesis of a new series of unprecedented 2,4-bis(fluoroalkyl)quinoline-3-carboxylates in two steps only. After standard saponification, the latter afforded their corresponding 2,4-bis(fluoroalkyl)quinoline-3-carboxylic acids, which served as pivotal intermediates for post-functionalization reactions. Indeed, the carboxylic function could then be derived according to known procedures and allowed the introduction of chemical diversity in C3-position of these unprecedented structures. The resulting highly functionalized quinolines will then serve as platform in the development of ingredients with strong potential for agrochemical research.

Abstract

The use of TFEDMA, a fluoroalkyl amino reagent, for the difluoromethylation and difluoroacylation of arenes, heteroarenes, and C-H acidic compounds is reported. This approach allows for an efficient access to difluoromethylated products of high added value in good to excellent yields and with scale-up possibilities.

Abstract

A novel approach towards highly functionalized fluoroalkyl pyrazoles was developed by using fluoroalkyl amino reagents in combination with a variety of fluorinated ketimines. Tuneable introduction of fluoroalkyl groups in the 3- and 5-positions was possible by using vinamidinium intermediates or the corresponding enamino ketones after hydrolysis. These high-value building blocks can give rise to a large number of analogues for bioactivity screening and discovering new heterocyclic bioactive ingredients in various life science fields.

Abstract

International audience; The regioselective Baeyer-Villiger oxidation of a wide range of α,α−disubstituted β-ketoesters has been developed to synthesize, in good yields, α,α-disubstituted α-acetoxy esters. The reactions were performed using m-chloroperbenzoic acid in the presence of triflic acid. The rearrangement was shown to occur with retention of configuration of the migrating group. The corresponding α,α−disubstituted α-hydroxy acids were obtained in good yields, after hydrolysis.

Abstract

Three tetrapeptides incorporating a 14-membered (Ri+1, Si+2) cycloisodityrosine at the i + 1 and i + 2 positions were designed and synthesized. Conformational anal. by 1H NMR and CD spectra as well as mol. modeling indicated that they all adopt a beta -turn conformation. While the CD spectrum of peptide I is characteristic of the typical type-II beta -turn (max. at .apprx.200 nm and a min. at .apprx.220 nm), that of peptide II (atropisomer of I) is opposite in sign to the expected spectrum of the type-II beta -turn.
