Soft glucocorticoids are compounds that are biotransformed to inactive and non-toxic metabolites and have fewer side effects than traditional glucocorticoids. A new class of 17-carboxamide steroids has been recently introduced by our group. In this study, local anti-inflammatory activity of these derivatives was evaluated by use of the croton oil-induced ear edema test. Glucocorticoids with the highest maximal edema inhibition (MEI) were pointed out, and the systemic side effects of those with the lowest EC50 values were significantly lower in comparison to dexamethasone. A 3D-QSAR model was created and employed for the design of 27 compounds. By use of the sequential combination of ligand-based and structure-based virtual screening, three compounds were selected from the ChEMBL library and used as a starting point for the design of 15 derivatives. Molecular docking analysis of the designed derivatives with the highest predicted MEI and relative glucocorticoid receptor binding affinity (20, 22, 24-1, 25-1, 27, VS7, VS13, and VS14) confirmed the presence of interactions with the glucocorticoid receptor that are important for the activity.

Molecular docking studies were performed on 18 17-carboxamide steroids in order to select compounds with potential local anti-inflammatory activity. These derivatives are amides of cortienic acids (obtained from hydrocortisone, prednisolone, and methylprednisolone) with methyl or ethyl esters of six amino acids. Interactions with the glucocorticoid receptor (GR), binding energies and ligand efficiency values of these compounds were compared with dexamethasone and cortienic acid obtained from prednisolone (inactive metabolite). On the basis of molecular docking studies, seven compounds were selected and their binding affinities for the GR were predicted by use of the exponential model created in this study. Subsequently, selected compounds were synthesized in good yields by use of modified N,N-dicyclohexylcarbodiimide (DCC)/1-hydroxybenzotriazole (HOBt) coupling procedure. Finally, the local anti-inflammatory activity of the synthesized compounds was examined by use of the croton oil-induced ear edema test. In vivo evaluation of systemic side effects as well as in silico prediction of metabolism were performed on the derivative with the best local anti-inflammatory activity. The combination of molecular docking studies and the exponential model for the GR binding affinity prediction could be used as an in silico tool for the rational design of novel 17-carboxamide steroids with potentially better biological profile than dexamethasone.

In this assay, the evaluation of lipophilicity of four ACE-inhibitors and hydrochlorothiazide (HCTZ) with RP-TLC on cellulose layers was described using three binary solvent systems. The selected ACE inhibitors had sufficiently different structures which can indicate the method suitability for their lipophilicity evaluation as the model substances in comparison with HCTZ. In addition, the linear relationship between the R-M-values and composition of mobile phases was established in the current study. From the regression data of the plots, the hydrophobicity parameters, R-M(0) and m, were determined and C-0 parameter was calculated. The correlations between the experimentally obtained hydrophobicity parameters and calculated log p values were studied. Furthermore, the obtained results were compared with those previously obtained on RP-18 modified silica gel. Very good correlation (r = 0.91; water-ethanol solvent system) between the chromatographically obtained hydrophobicity parameters and calculated log p values confirmed the selection of ACE inhibitors since lisinopril and quinapril were on the opposite sites of linear relationship. The results indicate that cellulose as an easily available sorbent can be successfully used for the lipophilicity investigation of examined substances with RP-TLC.

Topical anti-inflammatory corticosteroids are "drug of choice" in treatment of many inflammatory and allergic diseases in dermatology, pulmology, ophthalmology rheumatology and gastroenterology. Their using is limited because some serious systematic effects have been noted by longtime using of these drugs. In this manuscript, it was made a review of the relationship of structural changes on molecules of corticosteroids and intensity and specificity of their activity. Some of recently trends in design of newly anti-inflammatory steroids are based on retrometabolic design of "soft drugs", which are biotransformed to the inactive metabolits in body of patient after their applications. In manuscript, design and synthesis of new diastereoisomeric esters of steroid, as potentional antiinflammatory agents, was shown. Lokalni antiinflamatorni steroidi su "lekovi izbora" u lečenju raznih inflamatornih i alergijskih oboljenja u dermatologiji, pulmologiji, oftalmologiji, reumatologiji, gastroenterologiji. Dugotrajnom primenom ovih lekova mogu nastati veoma ozbiljni sistemski neželjeni efekti koji ograničavaju njihovu upotrebu. U ovom radu, dat je pregled uticaja strukturnih promena, u molekulama lokalnih antiinflamatornih steroida, na jačinu i specifičnost antiinflamatornog dejstva, a sve u cilju dobijanja novih bezbednijih lekova. Jedan od savremenih trendova u dizajnu antiinflamatornih steroida zasniva se na retrometaboličkom pristupu "soft" lekova, koji se nakon ispoljavanja dejstva, u organizmu pacijenta biotransformišu do neaktivnih i netoksičnih metabolita. U radu je prikazan dizajn i sinteza novih diastereoizomernih estara antiinflamatornih steroida kao potencijalnih lekova u lokalnoj terapiji.

Today with the development of combinatorial chemistry hundreds and hundreds of compounds that have potential biological activity are synthesized. The studies which include the selection of drug candidates and the study of their pharmacological properties are time consuming, expensive and usually require the use of experimental animals. For ethical and/or economical reasons, a great deal effort is currently being made to develop in vitro systems and provide primary information about the capability of new compounds in the first steps of drug development. Chromatographic models to predict drug absorption are experimentally easier than membrane-based permeability assays, because of their simplicity, accuracy and avoidance of experimental animals. Different chromatographic systems have been proposed to predict oral drug absorption. The use of conventional reversed-phase columns only has proven to provide adequate correlations for homologous series of compounds. The inclusion of amphiphilic structures in the stationary and/or mobile phases is a pre-requisite to emulate interactions of drugs with the phospholipids bilayers in the membranes. Savremeni razvoj kombinatorijalne hemije omogućio je sintezu velikog broja jedinjenja sa potencijalnom biološkom aktivnošću. Ispitivanja koja uključuju izbor jedinjenja i studije farmakoloških osobina su dugotrajna, skupa i obično zahtevaju primenu eksperimentalnih životinja. Iz etičkih i/ili ekonomskih razloga, veliki napori ulažu se u razvoj in vitro sistema koji mogu da pruže značajne informacije u ranim fazama razvoja leka. Hromatografski modeli za predviđanje apsorpcije leka su eksperimentalno jednostavni, tačni i ne zahtevaju primenu eksperimentalnih životinja. Primena reverznofaznih hromatografskih sistema dala je dobre korelacije samo za homologe serije jedinjenja. Uvođenje amfifilnih struktura u stacionarnu i/ili mobilnu fazu je bitan uslov za simulaciju interakcija farmakološki aktivnih jedinjenja sa fosfolipidnim dvoslojem u membrani.

Agonists of estrogen receptors, such as esters of estradiol, conjugated estrogens and esterified estrogens have been widely used in estrogen replacement therapy. Partial agonists of estrogen receptors, derivatives of triarylethylene such as, tamoxifen and toremifene posses weak estrogen-like effects on endometrium, bone and lipids. Recent advances in molecular pharmacology and estrogen receptors have resulted in the development of selective receptor modulators (SEMSs) that activate the estrogen receptor but that also exhibit tissue-specific estrogen-agonist or -antagonist activity. The first SERM marked is the benzothiophene, raloxifene which was introduced to maintain bone density in controlling osteoporosis postmenopausal women. Bisphosphonates have been used as second-line agents for prevention and treatment of osteoporosis in postmenopausal women. U supstitucionoj terapiji kod žena koriste se hormoni steroidne strukture/ agonisti ERa i ERp estrogenih receptora: derivati estrana, estri estradiola, metaboliti estardiola - prirodni proizvodi konjugovani estrogeni i polusintetski -esterifikovani estrogeni. Pored agonista u terapiji se koriste i parcijalni agonisti steroidnih receptora, derivati trifeniletilena ili trifeniletana kao što su tamoksifen i toreksifen. Derivat rigidne biciklične strukture-benzotiofena, raloksifen, je selektivni modulator estrogenih receptora, agonista estrogenih receptora u osteoblastima i osteoklastima, a antagonista na estrogenim receptorima uterusa i dojke. U terapiji osteoporoze se koriste hemijski stabilni bisfosfonati kao što su alendronat, risendronat i dr.

The lipophilicity expressed as the partition coefficient of newly synthesized IDA derivatives (DIETHYLIODIDA and DIIODIDA) and commercial IDA derivatives as well as theirs corresponding complexes with 99mTc was calculated using two computer programs: PACO and ChemOffice Ultra 2003 and determinated experimentally by extraction method. The correlation between: log P calculated by two programs; ChemOffice program and experimental log P; and PACO program and experimental log P, is r=0.76, r=0.81 and r=0.91 respectively. The involvement of more neurotrasmiters, hormones and autocoids in the control of gastric acid secretion has resulted in a number of therapeutic approches directed toward achieving its inhibition. Although significant progress in obtaining CCK2 gastrin receptor antagonists as vell as EP3 receptor agonists has been made in recent years, compounds from either of these groups seem unlikely to dislodge the irreversibile proton-pump inhibitors and histamine H2-angonists as the preferred treatment in acid-related disoreders at present. All of the potencially antiulcerative useful prostanoids are in general synthetically produced closely related analogs of natural prostaglandins PGE1 and PGE2. For example, it was hoped that EP3 receptor agonists such as misoprostol and newly sinthetised, selectivelly and more active GR63799X, might prove beneficial as cytoprotective agents. Učešće brojnih endokrinih hormona i neurotransmitera u kontroli gastrične sekrecije pruža mogućnost različitih terapijskih pristupa u tretiranju ulkusne bolesti i stanja hipersekrecije. Inhibitori protonske pumpe, H2 antagonisti histamina i muskarinskih receptora su veoma efikasni i bezbedni antiulkusni i antihipersekretorni lekovi. Iz ove grupe lekova u kliničkoj fazi ispitivanja su novi, reverzibilni inhibitori protonske pumpe i antagonisti H2 receptora druge generacije. U kliničkoj fazi ispitivanja nalaze se i brojni antagonisti CCK2-receptora kao potencijalni inhibitori gastrične sekrecije. Pored inhibicije gastrične sekrecije ovi potencijalni lekovi imaju i antianksiozni efekat. Citoprotektivna zaštita prostanoida, agonsta EP3 receptora za per os primenu je noviji pristup naročito u hroničnoj terapiji nesteroidnim antireumaticima koji mogu biti uzročnici ozbiljnih neželjenih efektata kao što su hipersekrecija gastritis, ulkus i gastrične hemoragije.

The ionization constants of 2,4-diiodo-6-methylphenylcarbamoylmethyl iminodiacetic acid were determined: pK1 1.3 (the first carboxylic group), pK2 2.52 (the second carboxylic group), pK3 5.86 (amino group) and pK4 10.85 (amide group). Heparin is being replaced in many of its applications by currently available alternative parenteral agents such as fondaparinux, leperudin, bivalirudin and argatromban. These alternative agents offer advantages in terms of more predictable pharmacokinetics and improved safety, reducing or eliminating the need for patient monitoring and reducing or eliminating the risk of heparin induced thrombocytopenia. Orally bioavialable direct thrombin and FXa inhibitors now under development, seem poised to become available over the next years and will represent viable alternative to both the parenteral anticoaguants and some currently available oral anticagulants such as warfarin, fenprocumon and acenokumarol. Oboljenja kardiovaskularnog sistema kao što su infarkt miokarda, različiti oblici tromboza i embolija, predstavljaju najznačajnije uzročnike mortaliteta i morbiteta. U savremenoj antikoagulantnoj terapiji pored klasičnih peroralnih antiagregatika kao što su: aspirin, dipiridamol i sulfinpirazon, i ne selektivnih inhibitora tkivnog faktora plazminogena, prećišćenog bakterijskog proteina, streptokinaze, reteplaze i urokinaze za parenteralnu primenu, su uvedeni selektivniji aktivatori plazminogena: alteplaza i tenekteplaza koji predstavljaju proizvode rekombinatne DNK tehnologije. Pored razvoja parenteralnih fibrinolitika, proizvoda rekombinantne tehnologije, sulfonskih polisaharidnih anjona male molekulske mase sa aktivnim pantasaharidnim sulfonatim anjonima, u prevenciji i terapiji postoperativno se koriste i peroralni antagonisti faktora koagulacije kao sto je varfarin i njegovi derivati koji predstavljaju antagoniste vitamina K. U savremenoj terapiji učinjen je napredak u prevenciji i terapiji embolija i dubokih tromboza uvođenjem u kliničku praksu lekova iz grupe malih molekula poznatih pod nazivom peptidomimetika. Dizajniranje peptidomimetika je omogućeno razvojem molekularne biologije, genetskog inženjeringa, imunohemije, biohemije i fiziologije. Razvoj ovih naučnih disciplina doveo je do utvrđivanja preciznih .

Chemistry of some non-insulin dependent diabetes mellitus (type 2) such as sulfonylurea derivatives, glinides, biguanides and thiazolidinedione is shown. The relationship between chemical structures and the molecular mechanism of action are presented. U terapiji insulin nezavisnog dijabetesa (tip 2) koriste se derivati sulfonilureje, bigvanidina, tiazolidindiona i inhibitori alfa glukozidaze. U ovom radu je prikazan odnos hemijske strukture i aktivnosti nekih sintetskih antihiperglikemika.

Many novel types of asthma therapeutic procedures have been investigated in the past decade. A vast amount of work in the 5-LO inhibitor and LT1-receptor antagonist area has resulted in a various of compounds being identified for clinical trails, and several compounds are used in the clinic. The impact of the LT1-receptor antagonist and 5-LO inhibitors on alergy therapy will be significant, if these compounds have an antiinflammatory effect within their mechanism. The area with possibly the most promise is selective inhibitors of phosphodiesterase enzymes (PDEs). Selective inhibitors of three isoenzymes of PDEs (PDE III, IV and V) may be possess both activity; bronchodilatory and anti-inflammatory effects. This results imply that this agents may lead to a re-examination and further subclassification of various types of asthma diseases.

The proposed method for the determination of fluocortolone is based on the formation of a stable, yellow coloured hydrazone product with 1,4-dihydrazinophthalazine as the reagent. Heating at 85 °C for 2 h was found to be necessary to ensure optimal hydrazone formation in acidified 1-propanol as the solvent. The detection limit was 1.2 μg/ml. This method when applied for the determination of fluocortolone in pharmaceutical formulations gave precise and reproducible results. Predložena spektrofotometrijska metoda određivanja fluokortolona se zasniva na građenju žuto obojenog proizvoda tipa hidrazona sa 1,4-dihidrazinoftalazinom kao reagensom. Metoda je reproduktivna kada se reakcija izvodi iz 1-propanola kome je dodata koncentrovana kiselina, uz zagrevanje na 85 °C u toku 2 sata. Limit detekcije iznosi 1,2 μg/ml. Predložena spektrofotometrijska metoda je primenjena za određivanje sadržaja fluokortolona u farmaceutskim oblicima.

Potential and generally selective antihypertensive and antianginal activities of nisoldipine have initiated the study of its molecular structure. The single crystals, which are the potential photodegradation products, were crystallized from methanol solution of nisoldipine, which was exposed to the action of the daylight. They were assumed to be the nitrozo derivatives of nisoldipine with the melting point 151-152°C. The crystal and molecular structure was determined by the X-ray diffraction method (MoKα, λ=0.71073A). On the basis of 3858 independent reflection intensities, the structure was solved by the direct method using the program SHELXS83 and refined by the least squares method (SHELXL93) up to final R-factor R=0.0578 for 1953 reflection (F0>4σ(F0)) and 334 parameters. The molecular structure obtained was that of: isobutyl methyl 2,6-dimethyl-4-(2-nitrophenyl)-1,4-dihydro pyridine -3,5 dicarboxylate which confirmed that we are dealing with nisoldipine, so one should not speak about the presence of any assumed photodegradation product. . Potencijalna i u opštem slučaju selektivna antihipertenzivna i antianginozna aktivnost nizoldipina kao i njegova izuzetna fotosenzitivnost na dnevnom svetlu, inicirala je istraživanje njegove molekulske strukture kao i molekulske strukture njegovih produkata. Iz metanolnog rastvora nizoldipina izloženog dejstvu dnevne svetlosti dobijeni su kristalizacijom potencijalni produkti fotodegradacije. Pretpostavljeno je da se radi o nitrozo derivatu nizoldipina, tačke topljenja 151-152°C. Kristalna i molekulska struktura određena je metodom difrakcije rendgenskih zraka (MoKα, λ=0.71073A). Na osnovu 3858 izmerenih intenziteta refleksa rešena je molekulska struktura, direktnom metodom primenom programa SHELXS83 i utačnjena metodom najmanjih kvadrata (SHELXL93) do konačnog R-faktora R=0.0578 za 1953 refleksa (F0>4σ(F0)) i 334 parametra. Dobijena molekulska struktura: izobutil metil-2,6-dimetil-4-(2-nitrofenil)-1,4-dihidropiridin-3,5-dikarboksilat, potvrdila je da se radi o nizoldipinu i da nema osnova govoriti da kristali pripadaju nekom od pretpostavljenih produkata fotodegradacije. .

Estri (S,S)-1,2-etandiamin-N,N'-di-2-(3-cikloheksil)propanske kiseline (EDCP) sa metanolom (DM-EDCP), etanolom (DE-EDCP), propanolom (DPEDCP), butanolom (DB-EDCP) i izobutanolom (DIB-EDCP) i estri (S,S)-1,3- propandiamin-N,N'-di-2-(3-cikloheksil)propanske kiseline (PDCP) sa metanolom (DM-PDCP), etanolom (DE-PDCP), propanolom (DP-PDCP), butanolom (DB-PDCP), izobutanolom (DIB-PDCP), pentanolom (DPE-PDCP) i izopentanolom (DIPE-PDCP) dizajnirani su kao ligandi za Pt(IV) komplekse. U in vitro ispitivanjima utvrđena je značajna citotoksična aktivnost za navedene komplekse, ali i za ligande u nevezanom obliku. Ispitivana supstanca DE-EDCP pokazala je najveću citotoksičnu aktivnost. U ovoj doktorskoj disertaciji opisan je razvoj i validacija savremene bioanalitičke metode - ultra visoko efikasne tečne hromatografije u sprezi sa masenom detekcijom (UHPLC-MS/MS). Metoda je razvijena i validirana za potrebe pretkliničkih ispitivanja, odnosno za određivanje ispitivane supstance DE-EDCP u biološkom materijalu, kao i njenog potencijalnog metabolita EDCP. Kao interni standard primenjen je strukturni analog DB-PDCP. Tokom in vitro fizičko-hemijske biofarmaceutske karakterizacije ispitivanih supstanci određena je i ocenjena njihova rastvorljivost u vodenom rastvoru, lipofilnost i permeabilnost na veštačkim membranama. Profili rastvorljivosti kiselina i njihovih estara se značajno razlikuju, dok su profili rastvorljivosti obe kiseline (EDCP i PDCP) slični, kao i profili rastvorljivosti svih estara. Kiseline se dobro rastvaraju u izrazito kiseloj sredini i u izrazito baznoj sredini, dok je rastvorljivost estara najveća u izrazito kiseloj sredini... In this doctoral dissertation, there is presented a development and validation of a new ultra-high-performance liquid chromatography tandem mass spectrometry bioanalytical method (UHPLC-MS/MS). The developed method is supposed for determination of DE-EDCP and its potential metabolit EDCP in biological materials during non-clinical and clinical studies. The structural analogue DB-PDCP was used as an internal standard. During the in vitro biopharmaceutical characterization of the investigated substances, there was performed a determination of solubility, lipophilicity and membrane permeability. Profiles of solubility of the observed acids and their corresponding esters are significantly different, while the profiles of solubility for two acids (EDCP and PDCP) are similar as well as the profiles of solubility for all the esters...

Nesteroidni antiinflamatorni lekovi (NSAIL) se koriste za ublažavanje simptoma akutne i hronične inflamacije već više od pedeset godina. Grupa NSAIL je veoma brojna i obuhvata različite hemijske strukture. Potreba za pronalaženjem novih NSAIL i dalje postoji jer neselektivni NSAIL često izazivaju neželjene gastrointestinalne efekte, a neki noviji selektivni lekovi se dovode u vezu sa ozbiljnim neželjenim kardiovaskularnim efektima. U ovoj disertaciji opisani su dizajniranje, sinteza, ispitivanje fizičko-hemijskih i bioloških osobina β-hidroksi-β-arilalkanskih kiselina. U doking studijama je ispitan uticaj α supstitucije jednom ili dvema metil grupama bočnog niza šest prethodno već sintetisanih β-hidroksi-β-arilalkanskih kiselina koje su derivati β-hidroksi-β- bifenilbuterne kiseline ili β-hidroksi-β,β-difenilpropionske kiseline na inhibiciju enzima ciklooksigenaze kao i na selektivnost prema izoformi ciklooksigenaza-2. Predmet ove doktorske disertacije je proučavanje stabilnosti donepezilhidrohlorida, ispitivanjem razgradnih proizvoda nakon različitih uslova forsirane razgradnje. Stabilnost donepezil-hidrohlorida kako u čistom tako i u komercijalnom obliku je ispitivana sprovođenjem studije forsirane razgradnje. Uzorci za ispitivanje su radi toga bili podvrgnuti kiseloj i alkalnoj hidrolizi, oksidaciji i razgradnji pri zagrevanju kao i elektrohemijskoj razgradnji. Ispitivana je i elektrohemijska aktivnost donepezil-hidrohlorida u čistom obliku. Elektrohemijske reakcije su praćene pomoću različitih elektrohemijskih metoda (ciklične voltametrije i voltametrije sa pravougaonim impulsima) na elektrodi od zlata. Za analizu elektrolita u elektrohemijskim eksperimentima korišćene su i druge analitičke metode (HPLC, MS i NMR) radi potvrde podataka dobijenih elektrohemijskim ispitivanjima i objašnjenja procesa elektrohemijske oksidacije i predviđanja stabilnosti donepezil-hidrohlorida. Naučni cilj ovog rada je da se ispitivanjem stabilnosti odgovarajućim transformacijama donepezil-hidrohlorida razviju metode za kvalitativno i kvantitativno određivanje donepezil-hidrohlorida u rastvorima i da se analizom dobijenih rastvora nakon određenih reakcija forsirane razgradnje utvrdi priroda transformacija ispitivanog molekula. U tom cilju izvršena je karakterizacija dobijenih razgradnih proizvoda korišćenjem HPLC tehnika, IR i UV-Vis spektrofotometrije, LC-MS merenjima i 1D i 2D NMR spektroskopskih metoda. Proizvodi dobijeni oksidativnom razgradnjom koji su identifikovani predstavljaju proizvode metabolizma u ljudskom organizmu. Takođe, dobijeni eksperimentalni rezultati su upoređeni sa in-silico predviđenim razgradnim proizvodima uz pomoć softvera za predviđanje hemijskih reakcija. U ovom radu su ispitivani i razgradni proizvodi, koji se literaturno ne pominju u ranijim radovima kao razgradni proizvodi, a proizvodi su transformacije donepezila u oksidaciji u prisustvu oksidacionog sredstva i posebno proizvodi nastali elektrohemijskim transformacijama. The subject of this doctoral thesis is investigation of donepezil hydrochloride stability, by examination of the degradation products formed under different forced degradation conditions. The stability of donepezil hydrochloride both in pure form and in the commercial form is examined by conducting forced degradation studies. Samples were subjected to an acidic and alkaline hydrolysis, oxidation, degradation under thermal conditions as well as the electrochemical degradation. Electrochemical activity of donepezil hydrochloride in a pure form was also investigated. Electrochemical reactions are monitored using various electrochemical methods (cyclic voltammetry and square wave voltammetry) on a gold electrode. For the analysis of electrolytes in electrochemical experiments other analytical methods (HPLC, MS and NMR) were used to confirm the obtained data in electrochemical experiments, to evaluate the involved process of electrochemical oxidation and to predict donepezil hydrochloride stability. The scientific objective of this paper is to investigate the stability and transformations of donepezil hydrochloride involved in order to develop methods for qualitative and quantitative determination of donepezil hydrochloride in solutions and by analyzing the obtained solutions under a certain forced degradation conditions to determine the nature of the transformation of the tested molecule. In this intention the characterization of degradation products was performed using the HPLC technique, IR and UV-Vis spectrophotometry, LC-MS measurements and 1D and 2D NMR spectroscopic methods. Products obtained by the oxidative degradation which have been identified are products of metabolism in the human body. Also, the obtained experimental results were compared with in-silico predicted degradation products with the help of software for predicting chemical reactions. Predmet ove doktorske disertacije je ispitivanje elektrohemijske aktivnosti amlodipina i nifedipina odnosno proučavanje odabranih 1,4-dihidropiridinskih lekova koji se kao blokatori Ca2+-kanala koriste u terapiji kardiovaskularnih oboljenja. Ispitivano je elektrohemijsko ponašanje amlodipina i nifedipina u čistim (kao aktivni farmaceutski ingredienti) i različitim komercijalnim oblicima, kao i u kompleksima sa različitim ciklodekstrinima. Elektrohemijske reakcije su praćene pomoću različitih elektrohemijskih metoda (prvenstveno ciklične voltametrije i voltametrije sa pravougaonim impulsima) na elektrodi od zlata i na elektrodi od zlata modifikovanoj o-MWCNT. Za analizu elektrolita korišćene su druge analitičke metode (HPLC, LC-MS) radi potvrde podataka dobijenih elektrohemijskim ispitivanjima. Takođe je ispitivana stabilnost ovih preparata, koji su radi toga bili podvrgnuti kiseloj i alkalnoj hidrolizi, oksidaciji, fotolizi i degradaciji pri zagrevanju kao i elektrohemijskoj degradaciji. U tom cilju, radi proučavanja povećanja stabilnosti sintetizovani su i analizirani ciklodekstrinski kompleksi amlodipina i nifedipina. Naučni cilj ovog rada je da se ispitivanjem elektrohemijske aktivnosti i odgovarajućim transformacijama amlodipina i nifedipina razviju metode za njihovo kvalitativno i kvantitativno određivanje i da se analizom dobijenih smeša nakon određenih reakcija utvrdi priroda transformacija ispitivanih molekula. Rezultati koji su proistekli iz istraživanja ove doktorske disertacije treba da doprinesu razvoju novih elektrohemijskih metoda za kvalitativno i kvantitativno određivanje amlodipina i nifedipina. Proučavana je elektrohemijska aktivnost inkluzionih kompleksa amlodipina i nifedipina sa ciklodekstrinima i uticaj strukture dobijenih ciklodekstrinskih kompleksa na elektrohemijsko ponašanje, što je od izuzetnog značaja pri ispitivanju aktivnih farmaceutskih supstanci i farmaceutskih preparata. The subject of this PhD thesis is examination of electrochemical activity of amlodipine and nifedipine, a 1,4-dihydropyridine type of Ca2+ channel blockers used in treatment of cardiovascular diseases. It was investigated the electrochemical behavior of amlodipine and nifedipine as active pharmaceutical ingredients, in different commercial pharmaceutical dosage forms as well as in complexes with different cyclodextrins. Electrochemical reactions were followed by using of different electrochemical methods (primary cyclic voltammetry and square wave voltammetry) on gold electrode and Au/o-MWCNT. For analysis of the electrolytes other analytical methods (HPLC, LC-MS) were used in order to confirm the data obtained by electrochemical investigation. It was also investigated stability of these preparations which were therefore exposed to hydrolysis in acid and alkaline conditions, oxidation, photolysis, temperature degradation and electrochemical degradation. In order to study stability enhancement the appropriate cyclodextrin complexes of amlodipine and nifedipine were synthesized and analyzed. The scientific aim of this study was to test the electrochemical activity and by appropriate transformations to develop methods for qualitative and quantitative determination of amlodipine and nifedipine as well as by analysis of the obtained mixture after certain reactions to determine the nature of transformations of examined molecules. The results arising from the research of this PhD thesis should contribute to development of new electrochemical methods for qualitative and quantitative determination of amlodipine and nifedipine. The electrochemical activity of inclusion complexes of amlodipine and nifedipine with cyclodextrins and influence of the structure of the obtained complexes on electrochemical behavior were studied which is of great importance in the study of active pharmaceutical ingredients and pharmaceutical preparations.

U ovoj doktorskoj disertaciji opisana je sinteza sest halkonskih intermedijera kao prekursora za sintezu sest novih aminoalkoksi derivata fenilpropiofenona i flavanona. Aminoalkoksi derivati fenilpropiofenona su strukturni analozi postojeceg antiaritmika Ic grupe, propafenona. U položaje 2 i 4 terminalnog benzenovog prstena uvođeni su supstituenti koji modifikuju hidrofobne i elektronske osobine molekula u cilju povecanja selektivnosti i potentnosti prema određenom tipu jonskog kanala. Sinteza propafenonskih derivata obuhvata pet faza: aldolnu kondenzaciju; kataliticku hidrogenizaciju; formiranje epoksida; aminolizu i građenje hidrohloridnih soli. Ako se alkalni, vodeno-etanolni rastvor trifluorometil halkona podvrgne dejstvu povisene temperature, nastaje flavanon koji hlađenjem kristalise u vidu iglicastih kristala. Cistoca i struktura sintetisanih jedinjenja potvrđena je hromatografskim i spektroskopskim metodama, dok je struktura sintetisanog flavanona potvrđena i kristalografski. Vazorelaksantna aktivnost propafenona i novosintetisanih derivata (1-100 μM) ispitana je na izolovanoj aorti pacova sa i bez endotela, koja je prethodno prekontrahovana rastvorom fenilefrina (10 μM). Svih sest sintetisanih derivata, ukljucujuci i propafenon, pokazali su doznozavisni vazorelaksantni efekat za koji je eksperimentalno potvrđeno da nije endotel zavisan. Ucesce jonskih kanala u vazorelaksantnom mehanizmu dejstva derivata propafenona ispitano je u prisustvu: lidokaina, antagoniste Na+ kanala (3 mM), nifedipina, selektivnog antagoniste LCa2+ kanala (1 μM) i 4-aminopiridina (4-AP), neselektivnog antagoniste K+ kanala (3 mM). Dobijeni rezultati ukazuju da su jonski kanali ukljuceni u mehanizam vazorelaksantnog dejstva i da uvedene strukturne modifikacije u molekulu propafenona doprinose postizanju izvesnog stepena selektivnosti. 4-AP antagonizovao je efekat derivata koji u para položaju terminalnog benzenovog prstena ima –CH3 grupu (5PCH3). Srednja efektivna koncentracija za derivat 5PCH3 (pEC50 4,41) tri puta je veca od koncentracije propafenona (pEC50 4,97) i dva puta od koncentracije sledeceg u nizu po kompeticiji, para fluoro supstituisanog derivata (5PF). U prisustvu nifedipina, najbolji antagonizam postignut je kod orto trifluorometil supstituisanog derivata (5OCF3), sto ukazuje na vecu selektivnost ovog derivata (pEC50 4,55) u odnosu na propafenon (pEC50 4,90), ali i ostale derivate (pEC50 4,70 - 4,97). Lidokain je antagonizovao efekat 5PF derivata. Srednja efektivna koncentracija 5PF derivata je dva i po puta veca u odnosu na propafenon (pEC50 4,89) i druge derivate (pEC50 4,86-5,19). Docking studije koje su sprovedene uz pomoc programa AutoDock v. 4.0.1 doprinele su razjasnjenju rezultata dobijenih u in vitro ispitivanjima antagonistickog dejstva 4-AP-a i nifedipina. Docking eksperimenti sa kristalografskom strukturom bakterijskog KcsA kanala su izvedeni pod pretpostavkom da se ispitivana jedinjenja vezuju za istu aminokiselinsku sekvencu pore kanala (Thr 107, Ala 108 i Ala 111) kao i 4-AP. Na osnovu dobijenih rezultata zakljucuje se, da 5OF derivat ciji vazorelaksantni efekat nije antagonizovan 4-AP-om, pokazuje najveci afinitet za vezivno mesto 4-AP-a u pori kanala. Ako se uporede rezultati dobijeni u docking eksperimentima na izdvojenom peptidu L-Ca2+ kanala, uocava se da 5OCF3 i 5OCl derivati pokazuju veci afinitet prema vezivnom mestu u poređenju sa ostalim orto i para supstituisanim derivatima. Docking eksperimenti na homologom modelu hERG kanala prethodili su in vivo eksperimentima. Najveca pažnja tokom eksperimenata bila usmerena ka interakcijama propafenona i derivata sa aminokiselinama Phe 656 i Tyr 652 za koje se smatra da su odgovorne za neželjene efekte mnogih lekova. Na osnovu dobijenih rezultata izabrana su tri halogenovana derivata (5OCl, 5OF i 5PF) za in vivo ispitivanja. Primenom akonitinskog testa, u in vivo eksperimentima na pacovima u dubokoj anesteziji, ispitivan je antiaritmijski potencijal halogenovanih derivata propafenona...

U cilјu prоučаvаnjа еfеkаtа sоlvаtаciје nа fаrmаkоlоškа svојstvа dеrivаtа hidаntоinа u оvоm rаdu su sintеtisаnе dvе sеriје јеdinjеnjа: а) 5-supstituisаni-5-fеnilhidаntоini, kојu čini 11 dеrivаtа, kоd kојih su rаzličitе аlkil-, ciklоаlkil- i аril-grupе (mеtil, еtil, n-prоpil, izоprоpil, n-butil, izоbutil, terc-butil, ciklоpеntil, ciklоhеksil, fеnil i bеnzil) uvеdеnе u pоlоžај 5 hidаntоinskоg prstеnа; b) 3,5-disupstituisаni-5-fеnilhidаntоini, kојu čini 25 dеrivаtа, kоd kојih su rаzličitе аlkil-, ciklоаlkil-, аlkеnil- i аril-grupе (mеtil, еtil, n-prоpil, izоprоpil, аlil, n-butil, izоbutil, terc-butil, ciklоpеntil, ciklоhеksil, fеnil i bеnzil) uvеdеnе u pоlоžаје 3 i 5 hidаntоinskоg prstеnа. Svа јеdinjеnjа su sintеtisаnа prеmа pоstupcimа оpisаnim u litеrаturi ili njihоvim mоdifikаciјаmа i оkаrаktеrisаnа оdrеđivаnjеm tеmpеrаturе tоplјеnjа, kао i UV, FTIR, 1H i 13C NMR spеktrоskоpiјоm, а njihоvа čistоćа је utvrđеnа pоmоću HPLC-а. Kvаntitаtivnа prоcеnа uticаја rаstvаrаčа nа pоmеrаnjе аpsоrpciоnih mаksimumа izvršеnа је primеnоm mеtоdа linеаrnе kоrеlаciје еnеrgiје sоlvаtаciје (LSER), оdnоsnо Kаmlеt–Таftоvоm јеdnаčinоm. Оsnоvа zа rаzumеvаnjе еfеkаtа sоlvаtаciје nа fаrmаkоlоškа svојstvа prоučаvаnih dеrivаtа hidаntоinа pоstаvlјеnа је kоrеlisаnjеm pоdаtаkа, kојi izrаžаvајu uticај rаstvаrаčа nа pоmеrаnjе аpsоrpciоnih mаksimumа, i pаrаmеtrа lipоfilnоsti (log P), kао i mоlеkulskih dеskriptоrа, kојi оpisuјu intеstinаlnu аpsоrpciјu (%Abs.), spоsоbnоst prоdirаnjа krоz krvnо–mоždаnu bаriјеru (log BB) i vеzivаnjе zа prоtеinе plаzmе (log kA). Vrеdnоsti log P su izrаčunаtе primеnоm prоgrаmskоg pаkеtа ACD Solaris v. 4.67, а mоlеkulski dеskriptоri, kојi оpisuјu оdgоvаrајućа ADMET svојstvа, primеnоm prоgrаmskоg pаkеtа ChemSilico. Оslаnjајući sе nа Аbrаhаmоv pristup u prоcеni еfеkаtа sоlvаtаciје, prikаzаnа је i diskutоvаnа vеzа izmеđu fаrmаkоlоški rеlеvаntnih svојstаvа i rаzličitih tipоvа intеrаkciја izmеđu mоlеkulа rаstvаrаčа i rаstvоrеnе supstаncе. Dоbiјеni rеzultаti su pоkаzаli dа su prоučаvаni dеrivаti hidаntоinа ispunili fаrmаkоkinеtičkе uslоvе dа budu kаndidаti zа lеkоvе i kvаlifikоvаli sе zа fаrmаkоdinаmičku fаzu ispitivаnjа. Аntikоnvulzivnа аktivnоst čеtiri 3,5-disupstituisаnа-5-fеnilhidаntоinа, čiје su lipоfilnоsti sličnе lipоfilnоsti fеnitоinа, оdrеđеnа је nа pаcоvimа Vistаr sоја u s.c. PTZ i i.v. PTZ tеstоvimа. Rеndgеnskа strukturnа аnаlizа tri јеdinjеnjа pоkаzаlа је dа pоstојi kvаlitаtivnа kоrеlаciја izmеđu mеđumоlеkulskih intеrаkciја u kristаlnоm stаnju i intеrаkciја sа biоlоškim rеcеptоrоm, kоје su оdgоvоrnе zа njihоvu аntikоnvulzivnu аktivnоst. Јеdinо kоd 3-n-prоpil-5-еtil-5-fеnilhidаntоinа, vеličinа tоrziоnоg uglа izmеđu fеnil-grupе i hidаntоinskоg јеzgrа nаlаzi sе u оpsеgu vrеdnоsti kојi је kаrаktеrističаn zа nајаktivniје dеrivаtе hidаntоinа i оvо јеdinjеnjе ispоlјаvа аntikоnvulzivnu аktivnоst kоја sе mоžе pоrеditi sа fеnitоinоm i, pri tоmе, izаzivа mаnjе sеdаciје. Prоučаvаnа је i аntiprоlifеrаtivna аktivnоst dvаnаеst 3-supstituisаnih-5,5-difеnilhidаntоinа prеmа ćеliјskој liniјi humаnоg kаrcinоmа kоlоnа HCT–116. Utvrđеnо је dа uvоđеnjе supstituеnаtа u pоlоžај 3 prоizvоdi trеnd prоmеnе аntiprоlifеrаtivnih učinаkа prоučаvаnih јеdinjеnjа, kојi је аnаlоgаn prоmеnаmа njihоvе аntikоnvulzivnе аktivnоsti u оdnоsu nа fеnitоin. Dоdаtnо је оdrеđеnа i njihоvа аntibаktеriјskа аktivnоst prеmа Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923 i kliničkim izоlаtimа Escherichia coli, Proteus mirabilis, Pseudomonas aeruginosa, Enterococcus faecalis i Staphylococcus aureus. Pоkаzаnо је dа јеdinо 3-izоprоpil i 3-bеnzil dеrivаti pоsеduјu slаbu аntibаktеriјsku аktivnоst prеmа grаm-pоzitivnој bаktеriјi E. faecalis i grаm-nеgаtivnim bаktеriјаmа E. coli ATCC 25922 i E. coli. Тo examine solvation effects on the pharmacological properties of hydantoin derivatives, two series of compounds have been synthesized: a) 5-substituted-5-phenylhydantoins include 11 compounds with different alkyl, cycloalkyl and aryl groups (methyl, ethyl, n-propyl, isopropyl, n-butyl, isobutyl, tert-butyl, cyclopentyl, cyclohexyl, phenyl and benzyl) in position 5 of the hydantoin ring; b) 3,5-disubstituted-5-phenylhydantoins include 25 compounds with different alkyl, cycloalkyl, alkenyl and aryl groups (methyl, ethyl, n-propyl, isopropyl, allyl, n-butyl, isobutyl, tert-butyl, cyclopentyl, cycloheksyl, phenyl and benzyl) in positions 3 and 5 of the hydantoin ring. All compounds have been synthesized according to procedures given in the literature or their modifications and characterized by UV, FTIR, 1H and 13C NMR spectroscopy and their purity has been confirmed by HPLC. The absorption spectra of the investigated hydantoin derivatives have been recorded in fifteen solvents of different polarities in the range from 200 to 400 nm. In order to determine a spectroscopic assignment of the absorption bands, the quantum chemical calculations have been performed using Gausian03 for DFT calculations and MOPAC 2009 for semiempirical calculations. The quantitative assessment of solvent effects on the absorption maxima shifts of the investigated molecules has been carried out by means of the linear solvation energy relationship (LSER) using the Kamlet–Taft equation. To provide a framework for an analyze of solvation effects on the pharmacological properties of the investigated hydantoin derivatives, the data, which interpret solvent effects on the absorption maxima shifts, have been correlated with the lipophilicity parameter (log P) and the in silico calculated bioactivity descriptors, in particular %Abs. (human intestinal absorption), log BB (blood–brain barrier permeation) and log kA (protein binding affinities) parameters. The log P values have been estimated with the ACD Solaris v. 4.67 and molecular descriptors for predicting ADMET properties have been obtained with the ChemSilico program. Drawing on Abraham's approach in assessing solvation effects, the relationship between pharmacologically relevant properties and different types of solvent–solute interactions has been presented and disscussed. In view of the results of this study, the investigated hydantoin derivatives have met the pharmacokinetic criteria for pre-selection as drug candidates and qualified them for the pharmacodynamic phase of antiepileptic drug development. The anticonvulsant activities of four 3,5-disubstituted-5-phenylhydantoins, the lipophilicities of which have been estimated to be similar to the lipophilicity of phenytoin, have been evaluated on male Wistar rats by the s.c. pentylenetetrazol (PTZ) seizure test and i.v. PTZ threshold test and the spontaneous locomotor activity test was used to assess the possible sedative effects. The X-ray analysis of three compounds has suggested that certain analogies might be drawn between interactions in crystal packing and biological interactions, which are responsible for their anticonvulsant activity. Only the value of the torsion angle of the phenyl ring in 3-n-propyl-5-ethyl-5-phenylhydantoin is in the range, which is characteristic for the most active hydantoin derivatives, and it exerts anticonvulsant activity comparable to phenytoin with lower liability for the induction of sedation in rats. The antiproliferative effects of a series of eleven 3-substituted-5,5-diphenylhydantoins, including some whose anticonvulsant activities have already been reported in the literature, against HCT–116 human colon carcinoma cells have been evaluated. It has been found that the introduction of a substituent at position 3 produces a trend of changes in the antiproliferative potencies of compounds analogous to that in their anticonvulsant activities. Moreover, their antibacterial activities in vitro against Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923 and clinical isolates of Escherichia coli, Proteus mirabilis, Pseudomonas aeruginosa, Enterococcus faecalis and Staphylococcus aureus have been evaluated. Only the 3-isopropyl and 3-benzyl derivatives show weak antibacterial activities against the Gram-positive bacterium E. faecalis and the Gram-negative bacteria E. coli ATCC 25922 and E. coli.

Cilj ove disertacije bio je da se ispitivanjem elektrohemijske aktivnosti makrolidnih antibiotika roksitromicina i midekamicina i poredjenjem dobijenih rezultata sa rezultatima ranije ispitanih azitromicina, klaritromicina i eritromicina izvede opšti zaključak o elektrohemijskom ponašanju ispitanih makrolidnih antibiotika, njihovim mogućim elektrohemijskim transformacijama kao i o mogućnosti njihovog kvalitativnog i kvantitativnog odredjivanja. Posebno je razmatrana mogućnost primene novih voltametrijskih metoda za odredjivanja makrolidnih antibiotika kao sastojaka komercijalnih i bioloških uzoraka. Na osnovu detaljnog ispitivanja elektrohemijskog ponašanja roksitromicina metodama ciklične voltametrije i diferencijalne pulsne voltametrije i midekamicina metodom ciklične voltametrije na elektrodi od zlata, simultano praćeno HPLC i LC/MS metodama kvalitativno i kvantitativno analitički su određene farmaceutske supstance roksitromicin i midekamicin, a roksitromicin i kao sastojak komercijalnog roksitromicina (Runac® tablete) i biološke tečnosti (humani urin). Poredjenjem sa ranije ispitivanim makrolidnim antibioticima pod istim uslovima, utvrdjeno je da su azitromicin, roksitromicin i midekamicin kvalitativno i kvantitativno analitički određeni. Eritromicin i klaritromicin su određeni kvalitativno a transformacije kojima podležu, za razliku od azitromicina, roksitromicina i midekamicina su praćene FTIR i HPLC metodama. The aim of the present PhD thesis was to examinate the electrochemical activity of macrolide antibiotics roxithromycin and midecamyin and to compare with the previously investigated azithromycin, clarithromycin and erithromycin in order to have general conclusion concerning electrochemical behavior of macrolide antibiotics. That means, their possible electrochemical transformations as well as the possibility of their quantitative and qualitative determination. Separetely was analysed the possibility of the new voltammetric methods application for the determination of macrolide antibiotics and as a content of commercial an biological samples. Taking into account the detailed investigation of the electrochemical behavior of roxithromycin by cyclic voltammetry and differential pulse voltammetry and a midecamyin by cyclic voltammetry on gold electrode by simultaneous detection of antibiotics by HPLC and LC/MS methods. Their standards were quantitatively and qualitatively were determined and roxithromycin was determined in the same manner and as a content of commercial tablets Runac® and as content of humane urine. The comparison with the previously investigated macrolide antibiotics under the identical experimental conditions, shown that azithromycin, roxithromycin and midecamyin are succesfully quantitatively and qualitatively determined. Clarithromycin and erithromycin undergo to the electrochemical transformations during the qualitative determination and these transformations were simultaneosly followed by HPLC and FTIR methods.
