Sella turcica is an intracranial bone depression located in the sphenoid bone that houses the pituitary gland, which is an important gland in the secretion of various hormones. Literature review: Variations in the morphology of the sella turcica can alter the morphology of the pituitary, which can lead to changes in hormone secretion and dental anomalies. There are studies that evaluate the sella turcica using metric methods and other studies that evaluate them with non-metric methods. Researchers use lateral radiographs and/or computed tomography to visualize the sella turcica. The purpose of this brief review was to revise the methods used to study sella turcica morphology in dental research. The literature search for sella turcica morphology was carried out with the following key words (sella turcica, bridging of sella, size, shape of sella turcica and sella turcica morphology) and using the search engines (Pubmed and Google scholar). According to the research, three methods of analyzing the sella turcica were observed, according to the measurements (length, width, diameter, area, depht, sella height posterior, sella height anterior), degree of calcification (class I / type I / group I, class II / type II / group II, class III / type III / group III) and format (normal sella turcica, sella turcica bridge type A – ribbon-like fusion, sella turcica bridge type B – extension of the clinoid processes, incomplete bridge, hypertrophic posterior clinoid process, hypotrophic posterior clinoid process, irregularity in the posterior part of the sella turcica, pyramidal shape of the dorsum sella, double contour of the floor, oblique anterior wall, oblique contour of the floor) of the sella turcica. Conclusion: It is expected that the three methods exposed in this review will help dental researchers to analyze sella turcica.

The aim of this study was to evaluate the color stability and the surface roughness of different composites brushed with toothpastes presenting different levels of abrasivity. Thirty discs of each material were obtained using michohybrid composites (Brilliant NG and Charisma Diamond) and a nanocomposite (Filtek Z350XT). The initial color (CIELab) and surface roughness (confocal laser scanning microscopy) of resin discs were evaluated. Afterwards, 10 specimens per group were brushed with the following dentifrices: Maximum Cavity Protection, Sensodyne Repair & Protect and Colgate Sensitive Pro-Relief. Brushing was performed with an electric toothbrush equipped with soft bristle head, with standard power and weight, for 30 minutes. Every 30 seconds, 1.0 ml of the slurry was injected between the bristles of the brush and the specimen. After abrasive challenge, the samples had their color and roughness reevaluated. Data were submitted to the Kruskal-Wallis test (color change) or the t-test (surface roughness). The level of significance was 5%. Results: Brushing did not significantly change the color of the composites tested in the study herein . On the other hand, the surface roughness of the composites was significantly affected by the abrasive challenge, regardless of the toothpaste used. The surface roughness change was similar for all the composites. The abrasive challenge with the toothpastes Maximum Cavity protection, Sensodyne Repair & Protect and Colgate Sensitive Pro-Relief was not able to significantly change the color of the composite resins. Nevertheless, the abrasive challenges significantly altered the surface roughness of all the evaluated composites. ResumoO objetivo deste estudo foi avaliar a estabilidade de cor e a rugosidade superficial de diferentes resinas compostas escovadas com dentifrícios de diferentes níveis de abrasividade. Trinta discos de cada material foram obtidos utilizando compósitos micro-híbridos (Brilliant NG e Charisma Diamond) e um nanocompósito (Filtek Z350XT). A cor inicial (CIELab) e a rugosidade superficial (microscopia confocal de varredura a laser) dos discos de resina foram avaliadas. Em seguida, 10 amostras por grupo foram escovadas com os dentifrícios Máxima Proteção Anticáries, Sensodyne Repair & Protect e Colgate Sensitive Pro-Alívio. A escovação foi realizada com uma escova elétrica com cabeça de cerdas macias, com potência e peso padronizados, durante 30 minutos. A cada 30 segundos, 1,0 ml da pasta era injetada entre as cerdas da escova e a amostra. Após o desafio abrasivo, as amostras tiveram sua cor e rugosidade reavaliadas. Os dados foram submetidos ao teste de Kruskal-Wallis (alteração de cor) ou ao teste t (rugosidade da superfície) (α=5%). A escovação não alterou significativamente a cor dos compósitos. Por outro lado, a rugosidade superficial dos compósitos foi significativamente afetada pelo desafio abrasivo, independentemente do dentifrício utilizado. A alteração da rugosidade superficial foi semelhante para todos os compósitos. O desafio abrasivo com a Máxima Proteção Anticáries, o Sensodyne Repair & Protect e o Colgate Sensitive Pro-Alívio não foi capaz de alterar significativamente a cor das resinas. Diferentemente, os desafios abrasivos alteraram significativamente a rugosidade superficial de todos os compósitos avaliados. The maternal Immunoglobulin G (IgG) antibodies transferred during the intrauterine life represents the main newborn immune protection against several intestinal and respiratory infection. However, there is little information about the reactivity and function of IgG from cord blood against newborn oral colonization. The aims this study was to evaluate the presence of IgG against Streptococcus mitis, Streptococcus mutans and theirs mains virulence antigens (GbpB, Ag I/II and Gtf) in peripheral blood (PB) samples from mothers with S. mutans or not in the saliva (SA) and compare with umbilical cord blood (CB). PB and SA samples were obtained from healthy women in the Hospital admission and CB was collected after delivery. The specificity of IgG against the bacterial extracts was determined by Western blot. The genetic material detection of S. mutans in the salivas was realized by the quantitative polymerase chain reaction with specific primers. The results showed that the minority of blood samples showed IgG against S. mitis. On the other hand, the majority of samples exhibited IgG anti S. mutans. The number of reactive bands to S. mutans was significantly higher than against S. mitis in both blood samples (p0.05) in the PB from mothers colonized or not by S. mutans. In opposite, in CB sample, it was more frequent to find samples with IgG anti-S. mutans and GbpB in the salivas from mothers with detectable S. mutans (p 0,05) no SP de mães colonizadas ou não por S. mutans. O desenvolvimento dos compósitos permite que o Cirurgião-Dentista realize restaurações dentária resistentes aos esforços mastigatórios e que apresentem estética semelhante ao tecido dental. Para isso, é necessário que as resinas compostas apresentem propriedades satisfatórias, a fim de resistirem aos desafios físicos e químicos que acontecem na cavidade oral. O objetivo deste estudo foi avaliar a microdureza superficial de resinas nanoparticuladas convencional (Filtek Z350 XT) e bulk-fill resina (Filtek One Bulk Fill) após a ciclagem em diferentes bebidas. Foram confeccionados 40 discos de cada resina (4,0 x 2,0 mm). Após a leitura inicial da dureza Vickers, 10 discos de cada resina foram imersos em saliva artificial (grupo controle), refrigerante à base de cola, suco de laranja e vinho tinto. Os espécimes foram mergulhados nas respectivas soluções (1,5 mL/espécime) por cinco minutos em agitação, duas vezes ao dia, com intervalo de 4 horas, por 14 dias consecutivos. Para os grupos controle, os espécimes foram mantidos em saliva artificial a 37ºC. Após a ciclagem, foi realizada leitura final da dureza superficial. Os dados obtidos foram tabulados e analisados pelo teste de ANOVA a 1 critério (comparação entre as resinas) e ANOVA a 2 critérios (comparação entre ar resinas e as bebidas). O nível de significância foi de 5%. Para a resina Filtek Z350 XT, os dados mostraram que a microdureza que todas as soluções reduziram a dureza do material, quando comparadas ao grupo controle. Além disso, a redução da dureza provocada pelo vinho tinto (63,0 ±4,4) foi maior quando comparado aos valores obtidos pela saliva artificial (71,75 ±6,9), mas foi semelhante à microdureza encontrada para a Coca-Cola (68,2±3.5) e para o suco de laranja (67,2 ±4,1). Para a resina Filtek One Bulk Fill, os grupos submetidos ao refrigerante, ao suco de laranja e ao vinho tinto, apresentaram valores semelhantes entre si, mas inferiores ao grupo controle (55,9 ±5,05). Foi concluído que ambas as resinas obtiveram redução da microdureza após serem submetidas à ciclagem nas diferentes bebidas, sendo que o vinho e o suco de laranja foram as bebidas que mais induziram à redução de dureza superficial da resina Bulk-Fill.

The objective of this in vitro study was to evaluate the influence of the Er,Cr:YSGG laser associated or not to a desensitizing agent in the treatment of erosive lesions. Forty specimens with dimensions of 4mm x 4mm and 3mm thickness were divided into 4 groups (n = 10): G1- no treatment; G2- 5% fluoride varnish; G3- Er,Cr: YSGG; G4 - fluoride varnish + laser. The specimens were immersed in erosive drink 3 times a day for 1 minute with an average interval of 2 hours between cycles for a period of 10 days. The treatments were performed according to the groups and the surface roughness and the wear profile were analyzed by scanning confocal microscopy. The normality (Kolmogorov-Smirnov) and homogeneity (Levene's) of the tests were evaluated. After these analyzes, the surface roughness data were submitted to the statistical analysis of Variance Analysis (ANOVA). All tests adopted a significance level of 5% (α = 0.05). At the representative images of the wear profile, the morphology of bovine dental enamel in its control and eroded areas were compared and qualitatively discussed. As regard surface roughness, there was no statistically significant difference between the groups. The qualitative analysis of the loss of volume showed that all experimental groups showed significant superficial morphology differences. Considering the limitations of an in vitro study, it can be concluded that the treatments performed were not able to treat dental erosion satisfactorily, indicating the need for more treatment sessions. ResumoO objetivo desse estudo in vitro foi avaliar a influência do laser Er,Cr:YSGG associado ou não a um agente dessensibilizante no tratamento de lesões erosivas. Foram confeccionados 40 espécimes com dimensões de 4mm x 4mm e 3mm de espessura, divididos em 4 grupos (n=10): G1- nenhum tratamento; G2- verniz fluoretado 5%; G3- aplicação do laser Er,Cr:YSGG; G4- verniz fluoretado + laser. Os espécimes foram imersos em bebida erosiva, 3 vezes por dia, durante 1 minuto, com intervalo médio de 2 horas entre os ciclos, por um período de 10 dias. Os tratamentos foram realizados de acordo com os grupos e que foram analisados a rugosidade superficial e o perfil de desgaste por meio de microscopia confocal de varredura. Avaliou-se a normalidade (Kolmogorov-Smirnov) e homogeneidade (Levene’s) dos dados. Após estas análises, os dados de rugosidade superficial foram submetidos ao teste estatístico de Análise de Variância (ANOVA). Todos os testes adotaram nível de significância de 5% (α=0,05). Através da obtenção das imagens representativas do perfil desgaste, a morfologia do esmalte dental bovino em suas áreas controle e erodida foram comparadas e qualitativamente discutidas. Quanto à rugosidade superficial, não houve diferença estatisticamente significativa entre os grupos. A análise qualitativa da perda de volume mostrou que todos os grupos experimentais apresentaram diferenças significativas na morfologia superficial. Considerando as limitações de um estudo in vitro pode-se concluir que os tratamentos realizados não foram capazes de tratar a erosão dentária de forma satisfatória, indicando a necessidade de mais sessões de tratamento. This study evaluated how Er,Cr:YSGG laser, associated or not with 5% fluoride varnish, influences the surface roughness and volume loss of bovine root dentin submitted to erosive and/or abrasive wear. One hundred and twenty dentin specimens were divided into the groups: without preventive treatment (WPT), 5% fluoride varnish (FV); Er,Cr:YSGG laser irradiation (L), and varnish combined with laser (FV + L). The specimens (n = 10) were subdivided into: 1 = erosion (E); 2 = abrasion (A); and 3 = erosion followed by abrasion (E + A). The erosive solution used was a soft-drink (pH = 2.42 at 4ºC) applied in 5-min cycles twice a day for 10d. Abrasive wear involved brushing for 60s with an electric brush (1,600-oscillations/s) at a load of 2.0N. Surface roughness and volume loss were evaluated using a laser scanning confocal microscope. Roughness data were submitted to one-way ANOVA and Tukey post-hoc test. For volume loss, the Kruskal-Wallis and Dunn’s post-hoc tests were used (α = 5%). The lowest values of roughness were found in the control areas of all subgroups (p > 0.05). In the experimental area, the [(WPT) + (E+A)] subgroup had a significantly higher roughness (5.712 ± 0.163 μm 2 ) than the other subgroups (p < 0.05). The L and (FV + L) groups had statistically similar roughness, regardless of the type of wear. The (FV + L) group had the lowest volume loss, regardless of the type of wear performed: [(FV + L) + (E)] = 7.5%, [(FV + L) + (A) = 7.3%, and [(FV + L) + (E + A)] = 8.1%. The subgroup [(WPT) + (E + A)] had the highest volume loss (52.3%). The proposed treatments were effective in controlling dentin roughness. Laser irradiation can be an effective method to increase root dentin resistance after challenges and limit problems related to non-carious lesions.

The aims of this study were to analyze the presence of Streptococcus mutans (SM)-DNA in cord blood (CB), maternal peripheral blood (PB), and maternal saliva (SA) and compare with data collected in health surveys. Sixty-four healthy women with pregnancies to term and without complications attending for elective cesarean section in the Clinical Hospital of Ribeirao Preto, Sao Paulo were included. Samples of PB and unstimulated SA were obtained on the day of hospitalization and samples of CB were collected after the delivery section. The treatments for dentin hypersensitivity (DH) may change the surface roughness of the root dentin, which can lead to biofilm accumulation, increasing the risk of root caries. The aim was to compare the surface roughness of root dentin after different treatments of DH and the biofilm formation on those surfaces. After initial surface roughness (Sa) assessment, 50 bovine root fragments received the following treatments (n = 10): G 1—no treatment; G2—5% sodium fluoride varnish; G3—professional application of a desensitizing dentifrice; G4—toothbrushing with a desensitizing dentifrice; and G5—diode laser application (908 nm; 1.5 W, 20 s). The Sa was reevaluated after treatments. Afterward, all samples were incubated in a suspension of Streptococcus mutans at 37 °C for 24 h. The colony-forming units (CFU) were counted using a stereoscope, and the results were expressed in CFU/mL. The one-way ANOVA and the Tukey’s tests compared the roughness data and the results obtained on the bacterial adhesion test (α = 5%). Objective To detect Streptococcus mutans in colostrum and saliva of neonates and compare with its detection in saliva of mothers. Methods Forty-three healthy women, full-term gestations with no complications, submitted to elective Cesarean section, and their newborns were included in the study. Samples were investigated by polymerase chain reaction to detect S. mutans in genetic material from the samples. Results Approximately 16% of colostrum samples showed S. mutans , but not correlated with the presence of the bacteria in both samples of saliva. S. mutans was detected in 49 and 30% of saliva samples of mothers and neonates, respectively. There was a positive correlation in S. mutans detection between types of saliva. The number of maternal samples of saliva with detectable S. mutans was smaller in women receiving dental treatment during pregnancy. Tooth brushing, three times a day, influenced the detection of S. mutans in both the saliva and the colostrum. Conclusion Although maternal saliva may present S. mutans , few samples of colostrum present the bacteria. The presence of bacteria in neonate saliva may be related to contact before birth. Dental treatment and hygiene habits seem to influence the detection of S. mutans in samples of maternal saliva and colostrum.

Objectives This study evaluated the effects of a bleaching agent on the composition, mechanical properties, and surface topography of 6 conventional glass-ionomer cements (GICs) and one resin-modified GIC. Materials and Methods For 3 days, the specimens were subjected to three 20-minute applications of a 37% H2O2-based bleaching agent and evaluated for water uptake (WTK), weight loss (WL), compressive strength (CS), and Knoop hardness number (KHN). Changes in surface topography and chemical element distribution were also analyzed by energy-dispersive X-ray spectroscopy and scanning electron microscopy. For statistical evaluation, the Kruskal-Wallis and Wilcoxon paired tests (α = 0.05) were used to evaluate WTK and WL. CS specimens were subjected to 2-way analysis of variance (ANOVA) and the Tukey post hoc test (α = 0.05), and KH was evaluated by one-way ANOVA, the Holm-Sidak post hoc test (α = 0.05), and the t-test for independent samples (α = 0.05). Results The bleaching agent increased the WTK of Maxxion R, but did not affect the WL of any GICs. It had various effects on the CS, KHN, surface topography, and the chemical element distribution of the GICs. Conclusions The bleaching agent with 37% H2O2 affected the mechanical and surface properties of GICs. The extent of the changes seemed to be dependent on exposure time and cement composition.

Eighty dentin specimens were selected and divided into eight groups (n = 10): G1: negative control; G2: positive control (5% fluoride varnish-FV); G3: Er,Cr:YSGG laser; G4: FV + laser; G5: 3% potassium oxalate; G6: 3% potassium oxalate + laser; G7: biphasic calcium silicate/phosphate gel (gel); G8: gel + laser. Laser parameters: 0.5 W, 6.25 J/cm2 at 1-mm distance. The erosive drink used was a cola soft-drink (pH = 2.42 at 4 °C), lasting 5 min, twice a day, with 6-h intervals between the challenges, during 14 days. Kolmogorov-Smirnov and Levene’s tests were satisfied. The surface roughness data were submitted to ANOVA and Tukey post hoc tests. For the wear profile, Kruskal-Wallis and Dunn post hoc tests were used. Afterwards, the Spearman correlation test was performed. All statistical tests assumed a significance level of 5% (α = 0.05). G1 presented the highest surface roughness value after the erosive challenge (3.586 μm2 ± 0.205 μm2) and the G7 presented the lowest surface roughness value after the erosive challenge (1.071 μm2 ± 0.180 μm2). The present study compared IgA specificity against oral streptococci in colostrum and saliva samples. Sixty-two mother-and-child pairs were included; samples of colostrum (C) and saliva (MS) were collected from the mothers and saliva samples were collected from babies (BS). The specificity of IgA against Streptococcus mutans and S. mitis were analyzed by western blot. Only 30% of babies’ samples presented IgA reactivity to S. mutans, while 74 and 80% of MS and C, respectively, presented this response. IgA reactivity to S. mutans virulence antigens (Ag I/II, Gtf and GbpB) in positive samples showed differences between samples for Gtf and especially for GbpB (p < 0.05), but responses to Ag I/II were similar (p > 0.05). The positive response of Gtf-reactive IgA was different between C (90%) and MS (58%) samples (p < 0.05), but did not differ from BS (p > 0.05). GbpB was the least detected, with 48 and 26% of C and MS, and only 5% of BS samples presenting reactivity (p > 0.05). Eight percent of MS and C samples presented identical bands to SM in the same time-point. In conclusion, the differences of IgA response found between C and MS can be due to the different ways of stimulation, proliferation and transportation of IgA in those secretions. The colostrum has high levels of IgA against S. mutans virulence antigens, which could affect the installation and accumulation process of S. mutans, mainly by supplying anti-GbpB IgA to the neonate.

Little is known about the ontogeny of the cytokines in saliva of newborn. Previous studies showed that levels of immunoglobulin A (IgA) in saliva could be influenced by prematurity. So, the aim of this study was to analyze the levels of interleukin 6 (IL-6), interleukin 10 (IL-10), interleukin 12 (IL-12), and interferon gamma (IFN-γ) in sample saliva of fullterm (FT) and preterm (PT) neonates at birth (T0) and after 3 months of age (T3). Saliva from 50 infants (25 FT and 25 PT) were collected at T0 and T3 and analyzed by Luminex Corporation (Austin, Texas, United States) multiplex assay. Clinical characteristics and social-economic data were assessed through questionnaires. All cytokines could be detected at birth in levels higher than found in T3. The mean levels and frequency of detection of cytokines were significantly higher in PT than FT at T0 (P0.05). There were a positive association between IL-10 and infection (P0.05) and IL-6 and stress (P0.005). Salivary cytokines were detected within the first hours after birth and their levels decreased after 3 months. The cytokine levels were different between PT and FT children and appear to be influenced by stress situation and/or antigenic microbial challenge. The results confirm the necessity for further studies about the mucosal immune system by using of saliva as a source of diagnostic by identification of biomarkers of the status of the immune.

Early diagnosis of white spot lesions allows non-invasive treatment to be indicated for the remineralization of active lesions. The goal was to report the clinical Protocol for the treatment of white spot lesion of a patient by the technique of resin infiltration, observing whether there is change of color on the white spot of the dental enamel after its treatment. The selected patient was 16 years old with good overall health, who featured white spot lesion of the dental enamel on tooth 13, classified as ICDAS score 2. The tooth was photographed, and its color was evaluated with the spectrophotometer in accordance with the CIELhC system. For comparison purposes, the color of tooth enamel in an area adjacent to the lesion was evaluated. The treatment of the lesion was performed with the resin infiltrate Icon (DMG, Hamburg, Germany), following the manufacturer's instructions. The color of the tooth was re-evaluated after the application of the resin infiltrate. At the end of the procedure, it was observed that the treatment of white spot lesion by resin infiltration technique reached its goal because, in addition to being minimally invasive, it was highly approved by patient and it provided the masking of the white spot lesion on the dental enamel. ResumoO diagnóstico precoce de lesões de mancha branca permite que um tratamento não invasivo seja indicado para a remineralização das lesões ativas. O objetivo foi relatar o protocolo clínico para tratamento de lesão de mancha branca ativa de um paciente pela técnica do infiltrante resinoso, observando se existe alteração de cor da mancha branca do esmalte dental após o seu tratamento. Foi selecionada uma paciente, de 16 anos de idade, com boa saúde geral e que apresentava lesão de mancha branca no esmalte dental do dente 13, classificada pelo escore 2 do ICDAS. O dente foi fotografado e sua cor foi avaliada com o espectrofotômetro de acordo com o sistema CIELhC. Para fins de comparação, foi avaliada a cor do esmalte dental em uma área adjacente à lesão. O tratamento da lesão foi realizado com a resina infiltrante Icon (DMG, Hamburgo, Alemanha), seguindo as recomendações do fabricante. A cor do dente foi reavaliada após a aplicação do infiltrante. Ao final do procedimento, observou-se que o tratamento da lesão de mancha branca pela técnica da resina infiltrante atingiu seu objetivo pois, além de ter sido minimamente invasiva, foi bem avaliada pela paciente e proporcionou o mascaramento da lesão de mancha branca do esmalte dental. Context: The knowledge of the reasons for the placement of direct restorations makes possible to trace an epidemiological profile of a specific population and to direct the teaching of dentistry to techniques that are commonly used today and will be continued performed in the future. Purpose: The aim of this study was to verify the reasons for placement and replacement of direct restorations in patients treated in the Dental Clinic of the Uberaba University - Brazil. Materials and Methods: This study evaluated 306 restorative procedures carried out on 60 patients. During the treatment planning, a form that contained information about the patient′s gender, tooth number, the classification of restorations, the reasons for placement and replacement of amalgam and tooth-colored restorations, the material that had to be removed and the new material used to fill the cavities was filled for each patient. Statistical analysis was carried out using Chi-square test (α = 0.05). Results: The data showed that most of the patients were female (66.7%). Of all the restorations placed, 60.45% were 1 st -time placements, while 39.55% were replacements. For 1 st -time restorations, the main reason for placement was primary caries (76.76%), followed by non-carious cervical lesions (15.14%). The amalgam restorations were replaced more frequently (67.77%). The primary reason for replacements was the presence of secondary caries (for both previous amalgam (42.68%) and composite (66.67%) restorations (P < 0.05). The resin composite was the most indicated material for the new restorations (98.04%) (P < 0.05). Conclusions: The main reason for placement of direct restorations was primary caries, while secondary caries was the main reason for replacements. In almost all cases, the material used to fill the cavities was the resin composite.

The use of erbium lasers to prevent caries in enamel has shown positive results. However, it is not known if Er,Cr:YSGG laser can also be used to increase acid resistance of root dentine, which is another dental tissue susceptible to the action of cariogenic bacteria. Objective: To analyze the effects of the Er,Cr:YSGG laser (λ=2.78 μm, 20 Hz) irradiation associated with 2% neutral sodium fluoride (NaF) to prevent root dentin demineralization. Material and Methods: One hundred human root dentin samples were divided into 10 groups (G) and treated as follows: G1: no treatment; G2: NaF; G3: laser (4.64 J/cm2) with water cooling (WC=5.4 mL/min); G4: laser (4.64 J/cm2) without WC; G5: laser (8.92 J/cm2) with WC; G6: laser (8.92 J/cm2) without WC; G7: laser (4.64 J/cm2) with WC and NaF; G8: laser (4.64 J/cm2) without WC and NaF; G9: laser (8.92 J/cm2) with WC and NaF; G10: laser (8.92 J/cm2) without WC and NaF. The NaF gel was applied alone or after 4 min of irradiation. After 14 days of acid challenge, the samples were sectioned and the Knoop microhardness (KHN) test was done at different depths (30, 60, 90 and 120 μm) from the outer dentin surface. Data were analyzed by one-way ANOVA and Fisher’s test (α=5%). Summary This research analyzed the influence of bur and erbium, chromium:yttrium-scandium-gallium-garnet (Er,Cr:YSGG) laser caries removal on cavity characteristics and marginal seal of composite resin restorations. One hundred and forty human dental root samples were used. After in vitro root caries induction using Streptococcus mutans, the carious lesions were removed either by a conventional technique using burs (G1=control) or by using an Er,Cr:YSGG laser (λ=2.78 μm, 20 Hz, pulse duration≅140 μs, noncontact mode using a 600-μm tip) with the following power outputs: G2: 1.0 W; G3: 1.25 W; G4: 1.5 W; G5: 1.75 W; G6: 2.0 W; G7: 2.25 W; G8: 2.5 W; G9: 2.75 W; G10: 3.0 W; G11: 3.25 W; G12: 3.5 W; G13: 3.75 W; and G14: 4.0 W. Samples in the 14 groups (n=10) were conditioned with Clearfil SE Bond and restored with a flowable composite. They were then thermocycled (1000 cycles) and immersed into a 2% methylene blue solution for microleakage analysis. The data were statistically compared (analysis of variance or Spearman correlation tests; p≤0.05). The lased groups showed significantly greater microleakage indexes, cavity depths, and presence of residual caries than did those of the control group. There was a strong positive correlation between residual caries and microleakage. The results indicate that Er,Cr:YSGG laser irradiation is not a good alternative to the use of burs for root caries removal since it may cause a significant loss of marginal sealing in composite resin restorations.

The aim was to evaluate the surface roughness and bacterial adhesion on enamel irradiated with high intensity lasers, associated or not to a fluoride varnish. Eighty fragments of bovine enamel were equally divided in 8 groups (n=10). Group 1 was not treated and Group 2 received only a 5% fluoride varnish application. The other groups were irradiated with an Er:Cr:YSGG (8.92 J/cm2), an Nd:YAG (84.9 J/cm2) and a diode laser (199.04 J/cm2), associated or not to a 5% fluoride varnish. The surface roughness was measured before and after treatments. Afterward, all samples were incubated in a suspension of S. mutans at 37 °C for 24 h. The colony-forming units (CFU) were counted by a stereoscope and the results were expressed in CFU/mm2. One-way ANOVA and the Tukey´s test compared the roughness data and the Student´s test compared the results obtained in the bacterial adhesion test (a=5%). The results showed that the irradiated samples without varnish presented the same roughness and the same bacterial adhesion that the non-irradiated samples. However, samples irradiated in the presence of fluoride varnish showed higher surface roughness and higher bacterial adhesion than the non-irradiated samples and those irradiated without varnish. Presence of pigments in the varnish increased the lasers’ action on the enamel surface, which produced ablation in this hard tissue and significantly increased its surface roughness. For this reason, the enamel’s susceptibility to bacterial adhesion was higher when the irradiation of the samples was made in presence of fluoride varnish. Resumo O objetivo foi avaliar a rugosidade superficial e adesão bacteriana no esmalte dental irradiado com lasers de alta intensidade, associados ou não a um verniz fluoretado. Oitenta fragmentos de esmalte foram igualmente divididos em 8 grupos (n= 10). O grupo 1 não recebeu tratamento e o grupo 2 recebeu aplicação de verniz fluoretado a 5%. Os outros grupos foram irradiados com os lasers Er,Cr:YSGG (8,92 J/cm2), Nd:YAG (84,9 J/cm2) e um laser de diodo (199,04 J/cm2), associados ou não ao verniz fluoretado. A rugosidade superficial foi medida antes e após os tratamentos. Depois, todas as amostras foram imersas em suspensão de S. mutans a 37 °C durante 24 h. As unidades formadoras de colônia (UFC) foram contadas utilizando uma lupa estereoscópica, e os resultados foram expressos em UFC/mm2. Os resultados de rugosidade foram analisados por ANOVA e pelo teste de Tukey, e a adesão bacteriana foi analisada pelo teste de Student (a=5%). As amostras irradiadas sem verniz apresentaram a mesma rugosidade superficial e a mesma adesão bacteriana que as amostras não irradiadas. No entanto, as amostras irradiadas na presença do verniz fluoretado apresentaram maior rugosidade superficial e aderência bacteriana do que as amostras não irradiadas e daquelas irradiadas sem o verniz. A presença de pigmentos no verniz aumentou a ação dos lasers na superfície do esmalte, promovendo a ablação do tecido duro e aumentando significativamente a sua rugosidade superficial. Por isso, a adesão bacteriana no esmalte foi maior quando a irradiação foi realizada na presença do verniz fluoretado.

The objective of this study was to evaluate the effects of chromium:yttrium-scandium-gallium-garnet (Er,Cr:YSGG) laser irradiation on the acid resistance of dental enamel. Forty human enamel samples were divided into four groups. They were manually irradiated with an Er,Cr:YSGG laser device (λ = 2.78 µm, 20 Hz, 20 s), in a scanning mode, with and without water cooling, according to the following parameters: Group 1: 0.25 W, 62.5 J/cm(2), no water cooling; group 2: 0.25 W, 62.5 J/cm(2), 5.0 ml/min; group 3: 0.5 W, 125 J/cm(2), no water cooling; group 4: 0.5 W, 125 J/cm(2), 5.0 ml/min. No airflow was used. Afterwards, the samples were submitted to an acid challenge and assessed by cross-sectional Knoop microhardness at different depths (20, 40, 60, 80, and 100 µm) from the outer enamel surface. Average values were obtained for both irradiated and control areas in each sample and they were compared to obtain a percentage of microhardness increase. Data were analyzed by analysis of variance and Fisher's exact test (α = 5%). The percentage of microhardness increase observed in group 1 (+23.58%) was similar to group 3 (+19.12%), but higher than groups 2 (+3.61%) and 4 (10.9%) (p0.05). The comparison of the depths showed that the Er,Cr:YSGG laser acted in the superficial layers of the dental enamel. The findings of the present study suggest that the energy densities of 62.5 and 125 J/cm(2) were capable of increasing the acid resistance of human enamel. The presence of water during irradiation makes it difficult to obtain an enamel surface more resistant to acids.

O objetivo deste trabalho in vitro foi avaliar a influência de diferentes agentes remineralizantes no tratamento de lesões erosivas em esmalte. Foram confeccionados espécimes de 4mmx4mm e 3 mm de espessura a partir da superfície vestibular de incisivos bovinos (n=10) e divididos aleatoriamente em 4 grupos. G1=aplicação do dentifrício remineralizante, G2= aplicação do agente potencializador remineralizante, G3= dentifrício remineralizante + agente potencializador remineralizante, G4=aplicação de verniz fluoretado (controle positivo), G5=nenhum tratamento (controle negativo). Os espécimes foram imersos em refrigerante durante um período de 10 dias. A rugosidade superficial foi analisada por meio de microscopia confocal de varredura a laser. Os dados foram analisados quanto à homogeneidade (Levene’s) e normalidade (Kolmogorov- Smirnov). Foram realizados testes paramétricos com análise de variância a dois critérios: fator tempo e fator tratamento, e pós-teste de Tukey para diferenciação das médias. Todos os testes estatísticos tiveram nível de significância de 5% (α=0,05). Os resultados obtidos mostraram diferenças estatisticamente significantes, demonstrando a redução da rugosidade da superfície do esmalte logo após o primeiro tratamento (G3) e para os demais grupos (G1, G2 e G4) somente após o segundo tratamento. Concluiu-se que a utilização de dentifrício composto por silicato de cálcio e fosfato de sódio influenciou na rugosidade do esmalte erodido do dente bovino.Palavras-chave: Dentifrícios. Erosão Dentária. Esmalte Dentário.Abstract The objective of this in vitro study was to evaluate the influence of different remineralizing agents in the treatment of enamel erosive lesions. Specimens of 4mmx4mm and 3mm thickness were made from the buccal surface of bovine incisors (n=10) and randomly divided into 4 groups. G1 = application of the remineralizing dentifrice, G2 = application of the remineralizing agent, G3 = remineralizing dentifrice + remineralizing agente, G4 = application of fluoride varnish (positive control), G5 = no treatment Specimens were immersed in refrigerant solution during a period of 10 days. The surface roughness was analyzed by means of confocal laser scanning microscopy. The data were analyzed for homogeneity (Levene's) and normality (Kolmogorov-Smirnov). Parametric tests with analysis of variance were performed on two criteria: time factor and treatment factor, and Tukey post-test for differentiation of means. All tests were statistically significant at 5% (α = 0.05). The results showed statistically significant difference, demonstrating the reduction of surface roughness after the first treatment (G3) and the other groups (G1, G2 and G4) only after the second treatment. It was concluded that the use of dentifrice composed of calcium silicate and sodium phosphate influenced the roughness of the eroded tooth enamel of the bovine tooth.Keywords: Dentifrices. Resumo A resina composta e o material restaurador mais utilizado atualmente. Apesar de seu sucesso clinico, o material esta sujeito a alteracoes em suas propriedades fisicas, quando exposto ao meio bucal. O objetivo foi avaliar a influencia da degradacao quimica na estabilidade de cor e na rugosidade superficial de resinas micro-hibrida e nanoparticulada. Foram confeccionados 40 discos (6,0mm x 2,0mm) da resina composta micro-hibrida (Z250XT) e 40 da resina nanoparticulada (Z350XT). Apos a analise da cor inicial e da rugosidade inicial, 10 discos de cada resina foram imersos em saliva artificial (Controle, pH=6,4), suco de laranja (pH= 3,4), refrigerante de limao (pH=2,9) e vinho tinto (pH=3,1), por 4 horas por dia (37oC) durante 30 dias. Apos, foram realizadas as leituras finais de cor e rugosidade. Os dados obtidos para a estabilidade de cor foram analisados pelo Teste de Kruskal-Wallis, seguido pelo Teste de Dunn (α=5%). Para a analise da rugosidade foi utilizado o Teste t para amostras pareadas (α=5%). O manchamento provocado pela saliva artificial, suco de laranja e refrigerante foi similar, mas foi estatisticamente diferente do manchamento produzido pelo vinho, em ambas as resinas testadas. O vinho produziu o mesmo grau de manchamento nas resinas micro-hibrida e nanoparticulada. Foi observado que o suco de laranja, o refrigerante e o vinho promoveram aumento da rugosidade superficial dos discos de resina. Diante disso, concluiu-se que o vinho alterou a cor de resinas compostas micro-hibrida e nanoparticulada, e que a rugosidade superficial dos discos de resina foi alterada por todas as bebidas utilizadas. Despite its clinical success, it is susceptible to changes in its physical properties when exposed to the oral environment. The aim was to evaluate the influence of acidic beverages on the color stability and on the surface roughness of the nanofilled and micro hybrid composite. Forty discs (6.0mm x 2.0mm) of the micro hybrid composite (Z250XT) and 40 of the nanofilled composite (Z350XT) were manufactured. After the measurement of the initial color and the initial roughness, 10 discs of each composite were individually immersed in artificial saliva (control, pH = 6.4), orange juice (pH=3.4), lime soda (pH = 2.9) and red wine (pH = 3.1) during 4 hours per day at 37°C for 30 days. At the end of the experiment, the final analysis of color and surface roughness were performed. The data obtained for the color stability were analyzed by Kruskal-Wallis and Dunn’s test (α= 5%). For the analysis of the surface roughness, the t test for related samples was used (α= 5%). The color changes promoted by artificial saliva, orange juice and soda were similar, but they were statistically different from staining produced by wine, in both tested composites. Wine produced the same degree of staining in both composites. Orange juice, soda and red wine increased the surface roughness of the composite discs. It was concluded that red wine significantly changes the color of micro hybrid and nanofilled composites. This study aimed to analyze in vitro inhibitory effects of restorative materials containing the antibacterial monomer 12-methacryloyloxydodecylpyridinium bromide (MDPB) on the formation of artificial secondary root caries lesions.Class V cavities (2mmx2mm) were prepared in 75 human root fragments. Specimens were randomly divided into five groups (n=15 fragments per group) and restored as follows: (I) MDPB-free adhesive system+MDPB-free composite (negative control); (II) resin modified glass ionomer (RM-GIC; positive control); (III) MDPB-free adhesive system+MDPB-containing composite (2.83% MDPB); (IV) MDPB-containing adhesive system+MDPB-free composite; (V) MDPB-containing adhesive system+MDPB-containing composite. Artificial secondary root caries lesions were produced by a biological artificial caries challenge. The restored specimens were immersed into a culture medium containing Streptococcus mutans and sucrose for 15 days. Histological slices (80+/-20 microm) of the specimens were used for measuring the mean depths of the artificial lesions produced in both margins of the restorations using polarized light microscopy. Results were expressed in percentage related to the mean depth of the negative control, considered 100%. Data were compared by ANOVA followed by the Tukey's test (por =0.05).The depths of lesions adjacent to cavities filled with RM-GIC (GII; 85.17+/-15.2%) were significantly (p0.01) shallower than those adjacent to restorations with MDPB-free composite (GI; 100.00+/-10.04%), despite the presence of MDPB in the adhesive system (GIV; 101.95+/-21.32%). The depths of lesions adjacent to cavities restored with MDPB-containing composite (GIII; 82.68+/-12.81% and GV; 85.65+/-15.42%), despite the adhesive system used, were similar to those of RM-GIC (GII). Mean lesions depths in these groups decreased from 13% (GV) to 17% (GIII) in relation to the negative control (GI).MDPB-containing composite inhibits the progression of artificial secondary root caries lesions regardless of adhesive systems.

The objective of this study was to evaluate the effects of diode laser and a desensitizing dentifrice on dentin permeability. Fifty-two root dentin fragments were obtained (5 × 5mm) and treated with 24% EDTA gel. The samples were divided into 4 groups (n = 13): G1, control (no treatment); G2, diode laser (λ = 908 nm, 1.5 W, continuous mode, 20s); G3, application of abrasive dentifrice for 1 minute (Elmex Sensitive Professional (International Gaba); and G4, application of abrasive dentifrice for 1 minute followed by irradiation with diode laser. Ten samples per group were immersed in 2% methylene blue solution for 4h. The specimens were washed, longitudinally sectioned, observed under optical microscopy, photographed and assessed based on the degree of dye leakage. The remaining samples were observed under scanning electron microscopy (SEM). The leakage data were subjected to ANOVA test, followed by Tukey’s t-test (α = 5%). Groups 2, 3 and 4 showed less dye penetration than the control group (p < 0.05), but were similar among each other. SEM images showed that dentinal tubules were open in G1, and fused and occluded in G2. Group 3 showed dentinal tubules that were occluded by the metal ions from the toothpaste. G4 presented similar characteristics to G3, and the presence of fused dentin. The diode laser and the dentifrice were effective in reducing dentinal permeability, and the combination of the two treatments did not show better results than either one used alone.

Little is known about the physiological mechanisms related to low-intensity laser therapy (LILT), particularly in acute inflammation and subsequent wound healing. The objective of this study was to verify the effect of LILT on mast cell degranulation. Epulis fissuratum tissues from eight patients were used. One part of the lesion was irradiated with an AsGaAl laser (lambda = 670 nm, 8.0 J/cm(2), 5 mW, 4 min). The other part was not irradiated. Then, the specimens were immediately removed, fixed and examined by light microscopy. The number of mast cells was similar in laser-treated samples when compared with non-irradiated specimens. The degranulation indexes of the mast cells observed in the irradiated samples were significantly higher than those of controls (P0.05). LILT with the parameters used increased the number of degranulated mast cells in oral mucosa.

O objetivo desse trabalho foi determinar a freqüência da infecção por Candida sp. em biópsias de lesões da mucosa bucal, assim como associar a presença de Candida sp. com lesões malignas e lesões com vários graus de displasia. Foram utilizadas 832 biópsias da mucosa bucal, previamente incluídas em parafinas, cujos blocos foram obtidos dos arquivos da Disciplina de Patologia da Faculdade de Odontologia de Araraquara da UNESP, no período entre 1990-2001. Três cortes seqüenciais foram corados pelo ácido periódico de Schiff (PAS). Do total de biópsias 27,2% foram PAS positivas, dessas 83,25% eram provenientes de pacientes do sexo masculino. Houve associação positiva entre infecção com displasia epitelial leve, moderada, severa, carcinoma espinocelular e hiperqueratose (p < 0,05). Não houve associação entre hiperplasia fibrosa inflamatória, líquen plano, granuloma piogênico (p < 0,05) com infecções fúngicas. A língua foi o sítio mais acometido por infecções em relação a outros sítios (p < 0,05). A partir dos dados quantitativos, concluiu-se que houve correlação positiva de infecção por fungos, lesões displásicas e carcinoma, sendo mais freqüente no sexo masculino. Estes dados não permitem inferir se o fungo causa displasia epitelial e carcinoma, mas confirmam a maior presença de Candida nessas lesões.Candidosis is the most common fungal infection in the oral cavity, and is usually associated with local and systemic predisposing factors. Histopathology reports issued between 1990 and 2001 inclusive were reviewed. Three sections of each mucosal biopsy were stained using the periodic acid-Schiff (PAS) technique. From the 832 biopsies 27.2% were PAS positive, of which 83.25% were obtained from male patients. There was positive association between fungic infection and mild, moderate and severe epithelial dysplasia, squamous cell carcinoma and hiperqueratosis (p < 0.05). There was no association between fungic infection and inflammatory fibrous hyperplasia, hyperkeratosis, lichen planus and pyogenic granuloma (p < 0.05). The frequency of infection in the tongue was significantly higher (p < 0.05) than in the other sites. Our results do not show a causal relation between Candida sp. and dysplastic lesions and carcinomas, but do confirm the higher presence of that microrganism in those lesions.

Secretory immunoglobulins present in mucosa surfaces represent the first line of defense of the adaptive immune system against infectious challenges. Preterm (PT) neonates' humoral immunity is diminished compared to full-term (FT) newborns. The identification of important antigens (Ags) of virulence of oral species may help in the investigation of the mechanisms of antigenic stimulation and the development of the mucosal immune response. In the present study, we measured saliva levels of immunoglobulins A (IgA) and M (IgM) and characterized the specificity of IgA against Ags of several streptococcal species found early in life.This was a prospective observational study. Salivary IgA (sIgA) antibody responses to bacterial species that are prototypes of pioneer (Streptococcus mitis, S. sanguinis, S. gordonii) and pathogenic (Streptococcus mutans) microorganisms of the oral cavity were studied in FT and PT children in two visits: at birth (T0) and at 3 months of age (T3). Salivas from 123 infants (72 FT and 51 PT) were collected during the first 10h after birth (T0) and again at 3 months of age (T3). Salivary levels of IgA and IgM antibodies were analysed by enzyme-linked immunosorbent assay (ELISA). A subgroup of 26 FT and 24 PT children were compared with respect to patterns of antibody specificities against different streptococci Ags using Western blot assays.No significant differences (P0.05) in salivary levels of IgA and IgM between FT and PT babies were found at birth. At T3, mean sIgA values were similar between groups and sIgM levels were significantly higher in PT than FT (P0.05). Western blot assays identified positive IgA response to streptococci in the majority of children, especially in the FT group. There were some differences between groups in relation to the frequency of children with positive response to Ags and intensity of IgA response. In general, oral streptococci Ags were more frequently detected and bands were more intense in FT than in PT, especially in T3. Prospective analysis of patterns of sIgA against Ags of different streptococcal species revealed an increase in complexity of the sIgA antibody response from the first day of birth (T0) to T3 in PT and FT.The patterns of sIgA response to streptococci Ags appear to be influenced by the gestational age, which might reflect the level of immunological maturity of the mucosal immune system.

Streptococcus mutans (SM) have three main virulence antigens: glucan binding protein B (gbpB), glucosyltransferase (Gtf) and antigens I/II (Ag I/II) envolved in the capacity of those bacteria to adhere and accumulate in the dental biofilm. Also, the glycosyltransferases 153 kDa of Streptococcus gordonii (SGO) and 170kDa of Streptococcus sanguinis (SSA) were important antigens associated with the accumulation of those bacterias. Streptococcus mitis (SMI) present IgA1 protease of 202 kDa. We investigated the specificity and levels IgA against those antigens of virulence in samples of human colostrum. This study involved 77 samples of colostrum that were analyzed for levels of immunoglobulian A, M and G by Elisa. The specificity of IgA against extracts of SM and initials colonizators (SSA, SMI, SGO) were analyzed by the Western blot. The mean concentration of IgA was 2850.2 (±2567.2) mg/100 mL followed by IgM and IgG (respectively 321.8±90.3 and 88.3±51.5), statistically different (p0.05). Results showed that the majority of samples had detectable levels of IgA antibodies to extracts of bacteria antigens and theirs virulence antigens. To SM, the GbpB was significantly lower detected than others antigens of SM (p0.05). High complexities of response to Ags were identified in the samples. There were no significant differences in the mean number of IgA-reactive Ags between the antigens (p0.4). So, the breast milk from first hours after birth presented significant levels of IgA specific against important virulence of antigens those oral streptococci, which can disrupt the installation and accumulation process of these microorganisms in the oral cavity.

SUMMARY The aim of the current study was to assess the acid demineralization susceptibility of bleached dental enamel submitted to different fluoride regimens. One hundred bovine enamel blocks (6×6×3 mm) were randomly divided into 10 groups (n=10). Groups 1 and 2 received no bleaching. Groups 3 to 6 were submitted to an at-home bleaching technique using 6% hydrogen peroxide (HP; G3 and G4) or 10% carbamide peroxide (CP; G5 and G6). Groups 7 to 10 were submitted to an in-office bleaching technique using 35% HP (G7 and G8) or 35% CP (G9 and G10). During bleaching, a daily fluoridation regimen of 0.05% sodium fluoride (NaF) solution was performed on groups 3, 5, 7, and 9, while weekly fluoridation with a 2% NaF gel was performed on groups 4, 6, 8, and 10. The samples in groups 2 to 10 were pH cycled for 14 consecutive days. The samples from all groups were then assessed by cross-sectional Knoop microhardness at different depths from the outer enamel surface. The average Knoop hardness numbers (KHNs) were compared using one-way analysis of variance and Tukey tests (α=0.05). The comparison between groups 1 and 2 showed that the demineralization method was effective. The comparison among groups 2 to 6 showed the same susceptibility to acid demineralization, regardless of the fluoridation method used. A hipersensibilidade dentinária (HD) é caracterizada por uma dor aguda, de curta duração, causada pela movimentação de fluidos no interior de túbulos dentinários expostos ao meio bucal. Recentemente, a utilização dos lasers de alta e baixa intensidade foi proposta como uma alternativa para o tratamento da HD. Este trabalho realizou uma revisão da literatura baseada em artigos publicados entre 2009 e 2013 sobre os efeitos dos lasers baixa e alta intensidade para o tratamento da HD. A base de dados utilizada foi o PubMed utilizando-se as palavras-chave Dentin hypersensitivity, laser, Nd:YAG, Er:YAG, GaAlAs, Er,Cr:YSGG, CO2, HeNe, hydrodinamic theory. Foi observado que o tratamento da HD com lasers é um método biocompatível, não-invasivo e eficiente. Os lasers de baixa intensidade (HeNe e GaAlAs) estimulam as células nervosas do tecido pulpar, interferindo na polaridade das membranas celulares pelo aumento da amplitude do potencial de ação, levando ao bloqueio da transmissão do estímulo ao sistema nervoso central, e estimulam os odontoblastos a produzirem dentina reparadora, o que diminui a permeabilidade da dentina e interrompe a movimentação de fluidos nos túbulos dentinários. Os lasers em alta intensidade (Nd:YAG, Er:YAG, Er,Cr:YSGG e CO2) têm o objetivo de obliterar a embocadura dos túbulos dentinários através da irradiação direta da dentina exposta ao meio bucal, promovendo a dissolução e a ressolidificação dos cristais de hidroxiapatita da dentina, levando à eliminação da sensibilidade dolorosa por um tempo prolongado. Pode-se concluir que os lasers de alta e baixa intensidade são efetivos para o tratamento da hipersensibilidade dentinária cervical.

In this study we compared the microleakage of conventional glass ionomer cement (GIC) restorations following the use of different methods of root caries removal. In vitro root caries were induced in 75 human root dentin samples that were divided in five groups of 15 each according to the method used for caries removal: in group 1 spherical carbide burs at low speed were used, in group 2 a hand-held excavator was used, and in groups 3 to 5 an Er,Cr:YSGG laser was used at 2.25 W, 40.18 J/cm(2) (group 3), 2.50 W, 44.64 J/cm(2) (group 4) and 2.75 W, 49.11 J/cm(2) (group 5). The air/water cooling during irradiation was set to 55%/65% respectively. All cavities were filled with GIC. Five samples from each group were evaluated by scanning electron microscopy (SEM) and the other ten samples were thermocycled and submitted to a microleakage test. The aim of this in vitro study was to evaluate some parameters of dental etching when irradiated with an erbium, chromium:yttrium-scandium-gallium-garnet (Er,Cr:YSGG) laser. One-hundred sound human third molars were selected and randomly distributed into ten groups (n = 10). The class V cavities of group 1 (control) were prepared with a bur and etched with 37% phosphoric acid, while groups G2 to G10, were prepared with laser (5 W, 88.46 J/cm(2), 90/70% air/water) and etched with the following powers: G3 and G4, 0.25 W; G5 and G6, 0.5 W; G7 and G8, 0.75 W; G9 and G10, 1 W. Group G2 received no laser etching. Prior to restoration, G2, G4, G6, G8 and G10 received acid etching. After restoration, all samples were submitted to a microleakage test. According to statistical analysis (Kruskal-Wallis and Dunn's tests), G10 presented the lowest microleakage values (P0.05). The other groups showed no differences between them. Etching with Er,Cr:YSGG laser (1 W) followed by phosphoric acid was effective in reducing the microleakage of class V restorations.

Aim The aim of this study was to evaluate the influence of chlorhexidine on microleakage of composite restorations in class V cavities prepared with a carbide bur (CB) and a erbium:yttrium-aluminum-garnet (Er:YAG) laser. Methods and Materials Cavity preparations were created on the buccal surfaces of 48 bovine incisors using different methods then equally dividing them into four groups. Groups I and II were prepared with a #56 CB in a high speed handpiece while Groups III and IV were prepared with a Er:YAG laser at 350m J/4Hz on enamel and at 80.24 J/cm2 on dentin. The cavity preparations were rinsed with 2% chlorhexidine solution before the application of Clearfill SE Bond system and placement of a Z100 composite resin restoration. After 24 hours, the teeth were polished, thermocycled, and sealed with cosmetic varnish. The samples were then immersed in 2% methylene blue for four hours and sectioned in the buccolingual direction to determine the degree of microleakage. The present study aimed to compare root caries removal by air turbine and Er,Cr:YSGG laser, and examine morphological changes after these caries removal techniques under scanning electron microscopy (SEM). Seventy two extracted human carious-free third molars were used in this study. After the in vitro root caries induction using S. mutans, the carious lesions were removed by the conventional technique, using burs (control), and by the Er,Cr:YSGG laser, using 13 different parameters, between 1 and 4,0 W. During caries removal, preparation time was recorded for all groups. The results appointed out that the conventional method was the fastest one. When only laser treatment was evaluated, the higher parameters seemed to remove caries faster then the lower ones. SEM revealed that the surfaces treated by air turbine were smooth, but with debris. The laser groups demonstrated smooth undulations, with little or absence of smear layer, and no signs of carbonization. These results suggest that caries removal by Er,Cr:YSGG laser was effective. Therefore, this equipment requires training on cavity preparation, in order to avoid damages in dental hard tissues.

This investigation evaluated the variation of the intrapulpal temperature when dentine was irradiated by the Er:YAG laser.The effect of preparation with the Er:YAG laser on the intrapulpal temperature is probably the biggest problem in using the laser for preparation of dental hard tissue.Seventy-two bovine incisors were studied that had the enamel and dentine of the buccal surface polished to a thickness of 2.0 mm. The teeth were divided into three groups, according to the repetition rate used (Group I = 2 Hz, Group II = 4 Hz, and Group III = 6 Hz), and irradiated, with or without water cooling, using 250, 300, and 350 mJ of energy per pulse. Thermocouples were introduced inside the pulp chamber through the palatine opening of the samples and fixed to the vestibular wall of the pulp chamber using a thermal paste.It was verified that there was a decrease of the intrapulpal temperature for all of the parameters in the Group I irradiated with water cooling and for the parameters of 350 mJ/4 Hz with water cooling. The other irradiations showed an increase of the intrapulpal temperature, varying from 0.03 degrees to 2.5 degrees C.We conclude that the use of the Er:YAG laser promoted acceptable temperature increases inside the pulp chamber. However, we do not recommend this procedure without water cooling because macroscopic observations of the dentine irradiated without water cooling showed dark lesions, suggesting carbonization of this tissue.

O objetivo do presente trabalho foi avaliar diferentes tratamentos na remineralização do esmalte desmineralizado em situação de alto desafio cariogênico. Foram utilizados 55 fragmentos de esmalte bovino (4mmX4mmX2mm) planificados e polidos. Os espécimes foram cobertos com resina composta ficando exposta apenas a metade da superfície externa (2mmX4mm) e então, foram imersos em solução tampão de acetato (pH 4,7) por 43 horas para formar a lesão de desmineralização. Os espécimes foram divididos aleatoriamente em 5 grupos (n=11), de acordo com os tratamentos preventivos: 1- Controle (sem tratamento); 2- Verniz fluoretado a 5% Duraphat®; 3- Flúor-gel 1,23%; 4- MI PasteTM; e 5- MI Paste PlusTM. Os tratamentos foram realizados uma vez por semana durante 3 semanas. Nos intervalos das aplicações os espécimes foram submetidos a ciclos de pH (DES/RE) num total de 21 dias. Após a finalização da ciclagem os fragmentos foram seccionados longitudinalmente, sendo que em uma metade foi analisada a microdureza longitudinal, pelo cálculo da variação percentual da dureza e na outra metade foi avaliada a profundidade de desmineralização do esmalte através de microscopia óptica. Para a análise em FTIR, pó do esmalte foi obtido dos espécimes em três diferentes momentos: inicial sem desmineralização da superfície, após a desmineralização por 43 horas e após o tratamento. Para análise da microdureza longitudinal empregou-se a variação da dureza. Os dados foram analisados quanto a sua distribuição, porém como não foram normais realizou-se uma análise não paramétrica comparando grupo contra grupo pelo Mann Whitney test com α=5%. Para análise da profundidade de desmineralização, como os dados se apresentaram normais e homogêneos, realizou-se a análise de variância a 1 critério e teste Holm- Sidak para comparação das médias com α=5%. Para análise em FTIR fez-se apenas uma análise descritiva dos dados. Esse estudo in vitro avaliou a resistência adesiva da dentina preparada com laser de Er:YAG ou broca, reidratada com clorexidina ou água deionizada, após armazenamento em saliva artificial e termociclagem. Cento e vinte terceiros molares humanos hígidos foram seccionados no terço médio da coroa, expondo a superfície dentinária (n=10). Os espécimes foram divididos aleatóriamente de acordo com o tratamento [laser de Er:YAG + reidratação com água deionizada (LW), laser de Er:YAG + reidratação com clorexidina a 2% (LC), turbina de alta-rotação com broca #245 + reidratação com água deionizada (TW) e turbina de alta-rotação com broca #245 + reidratação com clorexidina a 2% (TC)]. Após o preparo da dentina, os espécimes foram condicionados com ácido fosfórico 35%, lavados e secos com ar. A reidratação da dentina foi realizada através da aplicação de 1.5!L de água deionizada ou clorexidina 2%. Um sistema adesivo etch and rinse foi aplicado seguido da restauração com resina composta. Em cada grupo os espécimes foram sub-divididos de acordo com o envelhecimento [24 horas de armazenamento em saliva artificial / sem termociclagem (NA); 6 meses de armazenamento em saliva artificial / 12.000 ciclos térmicos (6A); 12 meses de armazenamento em saliva artificial / 24.000 ciclos térmicos(12A)]. Após cada período de envelhecimento, os dentes foram seccionados a fim de se obter 4 palitos com area de ±1.0 mm2, que foram submetidos ao teste de microtração usando uma máquina universal de testes com velocidade de 0.5 mm/min. Para a análise da interface adesiva em MEV foram utilizados 36 terceiros molares hígidos (n = 3) que foram restaurados e envelhecidos como descrito no protocolo empregado para o teste adesivo. ANOVA a dois critérios mostrou que não houve diferença significante para a interação entre os fatores e para o fator envelhecimento. O teste de Tukey 5% mostrou que o grupo LC apresentou os menores valores de resistência adesiva. A análise da interface adesiva através da MEV demonstrou que para os grupos irradiados houve formação de camada híbrida irregular com presença de fendas e o envelhecimento não interferiu na morfologia da interface adesiva. A reidratação com clorexidina influenciou negativamente a resistência adesiva do prepare com laser de Er:YAG. O envelhecimento em saliva artificial e termociclagem não interferiram na resistência adesiva da dentina. This in vitro study evaluated the bond strength of dentin prepared with Er: YAG laser or turbine, rehydrated with chlorhexidine or deionized water after artificial saliva storage and thermocycling. One hundred twenty human third molars were sectioned in order to expose the dentin surface (n=10). The specimens were equally and randomly divided according to treatment [Er:YAG laser + deionized water rehydration (LW), Er:YAG laser + chlorhexidine 2% rehydration (LC), bur on high-speed turbine + deionized water rehydration (BW) and bur on high-speed turbine + chlorhexidine 2% rehydration (BC)]. After dentin preparation, the specimens were etched with 35% phosphoric acid, washed and dried with air. The dentin rehydration method was performed with deionized water or chlorhexidine 2% by applying 1.5!L of each solution. An etch and rinse adhesive system was then applied and the samples were restored with a composite. In each group, the specimens were sub-divided in accordance with the aging method [24 hours artificial saliva storage, no thermocycling (NA); 6 months of artificial saliva storage, 12.000 thermocycling (6A); 12 months of artificial saliva storage, 24.000 thermocycling (12 A)]. After each aging period, each tooth was sectioned in order to obtain 4 sticks with a cross-sectional area of 1.0 mm2, which were submitted to microtensile test using the Universal Test Machine (0.5 mm/min). For the adhesive interface analysis by SEM, 36 healthy third molars (n=3) were restored and aged as the protocol used for the adhesive test. The two-way ANOVA showed no significant differences for the interaction between the factors and for the aging factor. Tukey test 5% showed that the LC group had the lowest bond strength values. The adhesive interface analysis by SEM showed that the irradiated groups presented irregular hybrid layer and cracks and the aging did not affect the adhesive interface morphology. The rehydration with chlorhexidine negatively influenced the bond strength of the preparation with the Er:YAG laser. The artificial saliva aging and thermocycling did not interfere with dentin bond strength.

Objetivo: Este estudo avaliou o efeito dos agentes dessensibilizantes associados ou não ao laser de diodo na permeabilidade dentinária e na morfologia da superfície. Materiais e Métodos: Para este estudo, 104 espécimes de dentina radicular bovina foram divididos aleatoriamente em 4 grupos e 2 subgrupos (n=13): G1A= nenhum tratamento (controle), G1B= laser de diodo, G2A= Oxagel, G2B= laser de diodo + Oxagel, G3A= Mi Paste, G3B= laser de diodo + Mi Paste, G4A= Sensitive Pró-Alívio dessensibilizante, G4B= laser de diodo + Sensitive Pró-Alívio dessensibilizante. Para análise da permeabilidade, 10 espécimes de cada grupo foram imersos em ácido clorídrico a 1% durante 20 segundos, 3 vezes, com intervalo de 2 horas entre as imersões, para exposição dos túbulos dentinários. A aplicação dos agentes dessensibilizantes foi feita de acordo com as instruções do fabricante e a irradiação com o laser de diodo foi feita no modo contato, comprimento de onda de 970 nm +/- 10 nm, potência máxima de 7 W CW, frequência de 10 Hz, durante 25 segundos. Em seguida, os espécimes foram submetidos à coloração histoquímica e seccionados para serem analisados quanto à permeabilidade por meio de microscopia óptica (MO). A permeabilidade dentinária foi dada pela porcentagem de penetração do corante em relação à área total do espécime seccionado. Os espécimes remanescentes (n = 3) foram utilizados para análise microscópica da superfície que foi dividida em duas áreas (controle e tratada). A exposição dos túbulos dentinários e a aplicação dos tratamentos foram realizadas da forma já descrita e, na sequência, os espécimes foram avaliados em microscopia eletrônica de varredura (MEV). Os valores obtidos de permeabilidade foram analisados através da ANOVA. Resultados: Nenhuma diferença estatística significante foi observada entre os tratamentos. Na microscopia eletrônica de varredura, foi observada uma ligeira diferença entre a área tratada e a área controle, exceto no grupo 4B. Comparando os grupos A e B (sem e associado ao laser, respectivamente), nenhuma mudança significativa foi notada na superfície dentinária. Conclusão: A aplicação dos agentes dessensibilizantes associados ou não ao laser diodo não influenciou na permeabilidade dentinária radicular e na morfologia da superfície dentinária. Objective: This study evaluated the effect of desensitizing agents associated or not with laser diode on dentin permeability and surface morphology. Materials and Methods: For this study, 104 bovine root dentin specimens were randomly divided into 4 groups and 2 subgroups (n = 13): G1A = no treatment (control), G1B= diode laser, G2A= Oxagel, G2B= diode laser + Oxagel, G3A= Mi Paste, G3B= diode laser + Mi Paste, G4A= Sensitive Pro-Relief desensitizing, G4B= diode laser + Sensitive Pro-Relief desensitizing. For permeability analysis , 10 specimens were immersed in 1% hydrochloric acid for 20 seconds, three times, with an interval of 2 hours between them, to expose the dentinal tubules. The application of desensitizing agents was performed according to the manufacturers instructions and the irradiation with the laser diode was made on contact mode, wavelength of 970 nm +/- 10 nm, maximum output 7 W CW, 10 Hz, for 25 seconds. After that, the specimens were imaged in an optical microscope (OM) and permeability was measured in the digitized images as the percentage of cooper ions penetration over the total area. The remaining specimens (n = 3) were used for surface microscopic analysis and they were divided in two areas (control and treated). The dentinal tubules exposure and the treatments application were performed as previously described and then analysed in scanning electron microscopy (SEM). The permeability values were analyzed using ANOVA. Results: No statistical significant difference among the treatments was observed. For scanning electron microscopy, there was a slight difference between the treated and control area, except for group 4B. Comparing groups A and B (without and associated with laser, respectively), no significant change was verified on the dentin surface. Conclusion: The application of desensitizing agents associated or not with diode laser did not affect the root dentin permeability and the dentin surface morphology.
