This study aimed to analyze the time-course of the physical properties of the psoas major muscle (PM) before and after exercise using magnetic resonance elastography (MRE). Muscle stiffness is one of the important properties associated with muscle function. However, there was no research on the stiffness of the PM after exercise. In this study, we investigated time-course changes of the shear modulus of the PM after exercise. Furthermore, T2 values and apparent diffusion coefficient (ADC), as the additional information associated with muscular physical properties, were also measured simultaneously. Healthy young male volunteers were recruited in this study (n = 9) and they were required to perform a hand-to-knee isometric and unilateral exercise (left side). At each time-point before and after exercise, a set of 3 types of MR scans to measure multiple physical properties of the PM [shear modulus (MRE), T2 values, and ADC] were repeatedly taken. On day 1, a single set MR scan was taken before exercise (pre-exercise MR scan), and 6 sets MR scans were taken (5.5 to 38.0 min after exercise). After about 10-min rest (46.0 to 56.0 min after exercise), 4 sets MR scans were taken (57.5 to 77.0 min after exercise). About 10-min rest was taken again (85.0-95.0 min after exercise), 4 sets MR scans were taken (96.5 to 116.0 min after exercise). On days 2 and 7, a single set MR scan (MRE, T2 value, and ADC) was taken on each experimental day. The data were analyzed as relative changes (%) of the given parameters to the pre-exercise values. The results indicated significant decreases in PM shear modulus up to about 30 min after exercise. Then, it gradually increased and showed significant increases at about 100 min after exercise compared to that before exercise. T2 values and ADC showed significant increases up to about 65 min after exercise compared to those before exercise, and then returned to the pre-exercise values. On days 2 and 7, all values showed no significant changes compared to the pre-exercise values. This study is the first to report the time-course of the physical properties of the PM after exercise.

Dynamic X-ray elastography using propagating shear waves is a novel technique to visualize the spatial distribution of elasticity in soft materials. We demonstrate that it can be applied to a gel sample with a fine structure. We prepared a viscoelastic gel sample with a two-dimensional lattice structure filled with ZrO2 particles, and applied the technique to map storage and loss moduli. The obtained storage modulus map successfully distinguished the harder lattice structure from the soft gel matrix with a spatial resolution much higher than those of other conventional elastography techniques using magnetic resonance and ultrasound.

This study aimed to develop a refined actuator suitable for psoas major muscle (PM) MR elastography (MRE) and evaluate its measurement precision. MRE was performed on nine healthy volunteers. Discomfort under PM-MRE examinations was evaluated by discomfort scores using a four-point subjective scale. The performance of refined actuator was evaluated in detail; repeatability was evaluated using repeatability coefficients (%RC) according to the Quantitative Imaging Biomarker Alliance (QIBA) and reproducibility with different operators was evaluated using interclass correlation coefficients (ICC). The discomfort score was 1.4 ± 0.53 and it indicated refined actuator was sufficiently comfortable (no pain). In the repeatability study (n = 9), the %RC was approximately 19%, which was comparable to the limit required for clinical liver MREs by the QIBA (19%). In the reproducibility study, the ICCs were > 0.90 (n = 3), indicating that the refined actuator had almost perfect reproducibility. The refined actuator was suitable for PM-MREs and had acceptable repeatability and reproducibility. Refined actuator was expected to meet the standard set by the QIBA (for liver MRE).

The purpose of this study was to determine an optimal condition (vibration frequency and image filtering) for stiffness estimation with high accuracy and stiffness measurement with high repeatability in magnetic resonance elastography (MRE) of the supraspinatus muscle. Nine healthy volunteers underwent two MRE exams separated by at least a 30 min break, on the same day. MRE acquisitions were performed with a gradient-echo type multi-echo MR sequence at 75, 100, and 125 Hz pneumatic vibration. Wave images were processed by a bandpass filter or filter combining bandpass and directional filters (bandpass-directional filter). An observer specified the region of interest (ROI) on clear wave propagation in the supraspinatus muscle, within which the observer measured the stiffness. This study assessed wave image quality according to two indices, as a substitute for the assessment of the accuracy of the stiffness estimation. One is the size of the clear wave propagation area (ROI size used to measure the stiffness) and the other is the qualitative stiffness resolution score in that area. These measurements made by the observer were repeated twice at least one month apart after each MRE exam. This study assessed the intra-examiner and observer repeatability of the stiffness value, ROI size and resolution score in each combination of vibration frequency and image filter. Repeatability of the data was analyzed using the intraclass correlation coefficient (ICC) and 95% limits-of-agreement (LOA) in Bland-Altman analysis. The analyses on intra-examiner and observer repeatability of stiffness indicated that the ICC and 95% LOA were not varied greatly depending on vibration frequency and image filter (intra-examiner repeatability, ICC range, 0.79 to 0.88; 95% LOA range, ±23.95 to ±32.42%, intra-observer repeatability, ICC range, 0.98 to 1.00; 95% LOA range, ±5.10 to ±10.99%). In the analyses on intra-examiner repeatability of ROI size, ICCs were rather low (ranging from: 0.03 to 0.69) while 95% LOA was large in all the combinations of vibration frequency and image filter (ranging from: ±62.66 to ±83.33%). In the analyses on intra-observer repeatability of ROI size, ICCs were sufficiently high in the total combination of vibration frequency and image filter (ranging from 0.80 to 0.87) while the 95% LOAs were better (lower) in the bandpass-directional filter than the bandpass filter (bandpass directional filter vs. bandpass filter, ±28.81 vs. ±54.83% at 75 Hz; ±25.63 vs. ±37.83% at 100 Hz; ±34.51 vs. ±43.36% at 125 Hz). In the analyses on intra-examiner and observer repeatability of resolution score, the mean difference (bias) between the two exams (or observations) was significantly low and there was almost no difference across all the combinations of vibration frequency and image filter (range of bias: −0.11–0.11 and −0.17–0.00, respectively). Additionally, effects of vibration frequency and image filter on wave image quality (ROI size and resolution score) were assessed separately in each exam. Both mean ROI size and resolution score in the bandpass-directional filter were larger than those in the bandpass filter. Among the data in the bandpass-directional filter, mean ROI size was larger at 75 and 100 Hz, and mean resolution score was larger at 100 and 125 Hz. Taking into consideration with the results of repeatability and wave image quality, the present results suggest that optimal vibration frequency and image filter for MRE of the supraspinatus muscles is 100 Hz and bandpass-directional filter, respectively.

In this study, the longitudinal wave speed (LWS) and shear wave speed (SWS) of the homogeneous agar-glycerol phantoms were ultrasonically measured, and the relationship between the compositions of phantoms and these two speeds was investigated. From the viewpoint of the measurement sensitivity of LWS and SWS, the LWS had the sensitivity to discriminate the differences in the glycerol weight concentration, while the SWS did not exhibit such sensitivity. These results indicate that the LWS can discriminate the composition of tissues different from that by the SWS in clinical diagnosis.

Magnetic resonance elastography (MRE) can visualize the shear wave propagation of in vivo tissues, which can be mapped into viscoelastic properties. No study has measured the biomechanical properties of the PM muscle in vivo using MRE. In this study, we evaluated stiffness values calculated by local frequency estimate (LFE) and algebraic inversion of differential equation (AIDE) in PM-MRE. The PM muscles of 17 healthy male volunteers were scanned in supine position by MRE. The Laplacian-based estimate (LBE) phase wrapped image data were filtered by gaussian-bandpass filter (GBF), and by both directional and GBF. LFE (MREWave) and AIDE wave inversion methods were used to calculate the respective elastograms. The wave interferences were removed by directional filtering, and smooth wave fields were obtained. The stiffness values calculated by LFE of non-DF images were 1.39 ± 0.25 kPa and 1.33 ± 0.22 kPa for right and left PM respectively, whereas for DF images, they were 1.26 ± 0.20 kPa for right PM and 1.18 ± 0.28 kPa for left PM. The stiffness values calculated by AIDE of non-DF images were 0.78 ± 0.10 kPa and 0.78 ± 0.13 kPa for right and left PM respectively, whereas for DF images, they were 0.73 ± 0.12 kPa for right PM and 0.74 ± 0.11 kPa for left PM. There was no statistically significant difference in mean values of stiffness with/without applying directional filter whereas there was a statistically significant difference in mean values of stiffness between LFE and AIDE. Both LFE and AIDE could be used for psoas major MR Elastography.

The present study aimed to develop vibration techniques for magnetic resonance (MR) elastography (MRE) of the psoas major muscle (PM). Seven healthy volunteers were included. MRE was performed with motion-encoding gradient (MEG)-less multi-echo MRE sequence, which allows clinicians to perform MRE using conventional MR imaging. In order to transmit mechanical vibration of the pneumatic type to the PM, a long narrow vibration pad was designed using a 3D printer, and the optimum vibration techniques were verified. The vibration pad was placed under the lower back, with the volunteers in the supine position. The results indicated that the PM vibrated well through the transmitted vibration from the lumbar spine, which suggests that the placement of a narrow vibration pad under the supine body, along the lumbar spine, allows the vibration of the PM. The shear modulus of the PM (n = 7) was 1.23 ± 0.09 kPa (mean ± SEM) on the right side and 1.22 ± 0.15 kPa on the left side, with no significant difference (t-test, P > 0.05). Increased stiffness of the muscle due to continuous local contraction may be an important cause of non-specific low back pain (LBP). The present vibration techniques for MRE of the PM provide a quantitative diagnostic tool for changes in muscle stiffness associated with non-specific LBP.

BACKGROUND: Digital radiography (DR) is grayscale adjustable and it can be unclear whether an acquired DR image is captured with the minimum radiation dose required. It is necessary to make an image of the amount of noise when taken at a lower dose than the acquired image, without increased exposure. OBJECTIVE: To examine whether an image of unacquired dose can be created from two types of dose DR images acquired using a phantom. METHODS: To create an additive image from two images of different doses, the pixel value of one image is multiplied by a coefficient and added to the other. The normalized noise power spectra (NNPS) of the normal image and the additive image with the same signal-to-noise ratio (SNR) are compared. The image noise of the unacquired doses is estimated from the graph changes of the pixel values and standard deviations of two images. The error between the SNR of the image obtained by changing the dose and the estimated SNR is measured. We propose a multiplication coefficient calculation formula that theoretically adjusts the additive image to the target SNR. The SNR error of the image created based on this formula is measured. RESULTS: The NNPS curves of the additive and normal images show a difference on the high frequency side. According to the statistics considering the preset of mAs value, there is no significant difference at 85%. The SNR estimation error is approximately 1%. The SNR error of the additive image created based on the formula is approximately 5%. CONCLUSION: The noise of the image of unacquired dose can be estimated, and the additive image adjusted to this value can be considered equivalent to the image taken at the actual dose.

This study aims to develop and assess a new automated processing technique in MR elastography (MRE), namely coherent-wave auto-selection (CHASE). CHASE enables automatic selection of the region of interest (ROI) for stiffness measurement by extraction of the coherent wave region (CHASE ROI), and it improves the reconstruction of stiffness by a directional filter oriented along the main wave in each pixel (CHASE filtering). In this study, MRE of a phantom and of the liver of four healthy volunteers was performed. To investigate the potential of CHASE, this study assessed the CHASE according to three indices through the phantom study: 1) agreement on the ROI settings between CHASE and expert observers, 2) noise dependency, and 3) effect of the CHASE on stiffness variability within the CHASE ROI. The agreements on the ROI settings were analyzed by Cohen's kappa coefficient (κ). The noise dependency was analyzed by the mean absolute percentage errors (MAPEs) within the ROI between low (20%-80% amplitudes) and high vibration amplitudes (100% amplitude). The stiffness variability was assessed by standard deviation (SD) within the ROI. In the volunteer study, agreements on the ROI settings (or stiffness value) and stiffness variability within the CHASE ROI were assessed using κ-value (or intraclass correlation coefficient: ICC) and coefficient of variation, respectively. The results showed close agreement on the ROI settings and stiffness (κ-value: greater than 0.61 in both the phantom and volunteer studies, ICC: 0.97 in the volunteer study). The MAPEs within the CHASE ROI were much smaller than those in the whole region of the phantom (CHASE ROI vs. the whole region at 20% amplitude: 10.3% vs. 50.8%). Moreover, in both the phantom and volunteer studies, the stiffness variation within the CHASE ROI was smaller in the elastogram processed with CHASE filtering than in the unprocessed one. Our results demonstrated that the CHASE has high robustness against noise and the potential to provide ROI settings for stiffness measurement comparable to expert observers, as well as improve the reconstruction of stiffness.

The purpose of this study was to investigate an influence of vibration waveform on magnetic resonance elastography (MRE). MRE is an innovative imaging technique for the non-invasive quantification of the elasticity of soft tissues through the direct visualization of propagating shear waves in vivo using a special phase-contrast magnetic resonance imaging sequence. Since the elasticity of soft tissue calculates from the wavelength of propagating shear waves, it is necessary to propagate sine-wave-shape shear wave at the target soft tissue. However, due to the various factors; i.e. overload of vibration generator, poor contact between imaging object and vibration pad, etc.; it may be difficult to generate a simple sine wave. This work was focused on change vibration waveforms; i.e. square wave, triangle wave, saw-tooth wave; which is induced by the various factors. Phantom experimental results demonstrated that when square and saw-tooth waveforms of 25 Hz vibration frequency, into the phantom, the waveform of propagating wave was not similar to sine waveform. It may influence on the MRE that in case of the waveforms has low frequency and square or saw-tooth like waveforms.

The elastic modulus of tissue as a useful biomarker of disease detection can be quantitatively evaluated based on shear wave speed (SWS) measurements in shear wave elastography. Although the longitudinal wave speed (LWS) is also expected to be a promising biomarker for disease detection, the elasticity is not always dominant because the LWS is affected by the bulk modulus. In other words, LWS and SWS may reflect different tissue properties. Therefore, in this study, based on the improvement in LWS measurement, the relationship between the composition of a phantom mixed with agar and glycerol and ultrasonically measured LWS and SWS was investigated. The LWS had a good sensitivity in detecting glycerol, while the SWS had a good sensitivity in detecting agar. The calculated Poisson’s ratio had a better sensitivity in detecting agar. In conclusion, a simultaneous measurement of LWS and SWS may help identify the tissue composition.

Phase Unwrapping (PU) is an ill-posed problem in Magnetic Resonance Elastography (MRE). The phase information is not usable until the phases are retrieved by using PU algorithms. In this present study, we attempt to determine the ideal PU method for MRE using both phantom and volunteer psoas major (PM) muscle images. All the MRE experiments were carried out in Philips MRI (Achieva 3.0 T, Best, The Netherlands). A multi-echo gradient-echo MRE pulse sequence was employed and the four PU methods were considered based on their easy user platform. They are namely, Minimum Discontinuity (MD), Laplacian-Based Estimate (LBE), Region Growing (RG) and Dilate-Erode (DE) Propagate. Phantom images were successfully unwrapped by all four methods, whereas MD and LBE could only unwrap PM muscle images properly. RG and DE failed to unwrap the PM muscle images.

We report star-trail artifacts of advanced-putrefied brain on postmortem CT (PMCT) of a 77-year-old male approximately one month after death. After CT calibration, routine PMCT (tube current time product: 400 mAs) showed star-trail artifacts, which arose due to cerebral cavitation, but were hardly visible on low-dose PMCT (100 mAs) due to noise. Low-dose PMCT with 4-time-image summation also showed star-trail artifacts. Those are not due to mechanical X-ray detector problems.

Palpation is a standard clinical tool to diagnose abnormal stiffness changes in soft tissues. However, it is difficult to palpate the supraspinatus muscle because it locates under the trapezius muscle. The magnetic resonance elastography (MRE) uses harmonic mechanical excitation to quantitatively measure the stiffness (shear modulus) of both the superficial and deep tissues. The purpose of this study was to build a vibration system for applying the MRE to the supraspinatus muscle. In this study, a power amplifier and a pneumatic pressure generator were used to supply vibrations to a vibration pad. Six healthy volunteers underwent MRE. We investigated the effects of position (the head of the humerus and the trapezius muscle) of the vibration pad on the patterns of wave propagation (wave image). When the vibration pad was placed in the trapezius muscle, the wave images represented clear wave propagation. On the other hand, when the vibration pad was placed in the head of the humerus, the wave images represented unclear wave propagation. This result might be caused by wave interferences resulting from the vibrations from bones and an intramuscular tendon of the supraspinatus muscle. The mean shear modulus also was 8.12 ± 1.83 (mean ± SD) kPa, when the vibration pad was placed in the trapezius muscle. Our results demonstrated that the vibration pad should be placed in the trapezius muscle in the MRE of the supraspinatus muscle.

Magnetic resonance elastography (MRE) can measure tissue stiffness quantitatively and noninvasively. Supraspinatus muscle injury is a significant problem among throwing athletes. The purpose of this study was to develop an MRE technique for application to the supraspinatus muscle by using a conventional magnetic resonance imaging (MRI). MRE acquisitions were performed with a gradient-echo type multi-echo MR sequence at 100 Hz pneumatic vibration. A custom-designed vibration pad was used as a pneumatic transducer in order to adapt to individual shoulder shapes. In a gradient-echo type multi-echo MR sequence, without motion encoding gradient (MEG) that synchronizes with vibrations, bipolar readout gradient lobes achieved a similar function to MEG (MEG-like effect). In other words, a dedicated MRE sequence (built-in MEG) is not always necessary for MRE. In this study, 7 healthy volunteers underwent MRE. We investigated the effects of direction of the MEG-like effect and selected imaging planes on the patterns of wave propagation (wave image). The results indicated that wave images showed clear wave propagation on a condition that the direction of the MEG-like effect was nearly perpendicular to the long axis of the supraspinatus muscle, and that the imaging plane was superior to the proximal supraspinatus muscle. This limited condition might be ascribed to specific features of fibers in the supraspinatus muscle and wave reflection from the boundaries of the supraspinous fossa. The mean stiffness of the supraspinatus muscle was 10.6 ± 3.17 kPa. Our results demonstrated that using MRE, our method can be applied to the supraspinatus muscle by using conventional MRI.

In cartilage regenerative medicine, autologous chondrocyte implantation (ACI) has been applied clinically for partial defects of joint cartilage or nasal augmentation. To make treatment with ACI more effective and prevalent, modalities to evaluate the quality of transplanted constructs noninvasively are necessary. In this study, we compared the efficacy of several noninvasive modalities for evaluating the maturation of tissue-engineered auricular cartilage containing a biodegradable polymer scaffold. We first transplanted tissue-engineered cartilage consisting of human auricular chondrocytes, atelocollagen gel, and a poly-l-lactic acid (PLLA) porous scaffold subcutaneously into the back of athymic nude rats. Eight weeks after transplantation, the rats were examined by magnetic resonance imaging (MRI), X-ray, and ultrasound as noninvasive modalities. Then, the excised constructs were examined by histological and biochemical analysis including toluidine blue (TB) staining, glycosaminoglycans content, and enzyme-linked immunosorbent assay of type II collagen. Among the modalities examined, transverse relaxation time (T2) and apparent diffusion coefficient of MRI showed quite a high correlation with histological and biochemical results, suggesting that these can effectively detect the maturation of tissue-engineered auricular cartilage. Since these noninvasive modalities would realize time-course analysis of the maturation of tissue-engineered auricular cartilage, this study provides a substantial insight for improving the quality of tissue-engineered cartilage, leading to improvement of the quality and technique in cartilage regenerative medicine.

We measured standard deviation (SD), noise power spectrum (NPS), and modulation transfer function (MTF) of computed tomography obtained by image summation method (fused CT) using phantoms. SDs, NPSs and MTFs of fused CT up to 25 times summation were calculated. Increased number of image summation reduced SD and NPS, although the effect became small after 10 times summation. MTF remained the same regardless of the number of summation. Fused CT is effective in reducing noise.

The speed of sound (SOS) of a longitudinal wave propagating in the living body reflects the tissue properties and is expected as a promising evaluation index for detection of disease, diagnosis and monitoring. However, in general, the interpretation of SOS is complex, because it is influenced mostly by compressibility and multiple factors. In this study, as a study for the interpretation of SOS, the SOS of cartilage was measured in vivo by using a fusion of ultrasound and magnetic resonance imaging (MRI), and the correlations between the SOS and other quantities were investigated. In summary, as a possible interpretation of SOS of cartilage tissue used in this study, it was suggested that the SOS was partly related to water content.

We developed a Magnetic Resonance elastography (MRE) technique using a conventional magnetic resonance imaging (MRI), which allows a simultaneous elastography of the supraspinatus and trapezius muscles, by designing a new wave transducer (vibration pad) and optimizing the mechanical vibration frequency. Five healthy volunteers underwent an MRE. In order to transmit the mechanical vibration (pneumatic vibration) to the supraspinatus and trapezius muscles, a new vibration pad was designed using a three-dimensional (3D) printer. The vibration pad was placed on the skin 2 cm medial and 2 cm cephalad the deltoid tubercle. MRE acquisition was performed with a multi-slice gradient-echo type multi-echo MR sequence, which allows MREs even in a conventional MRI; two oblique axial images of the supraspinatus and trapezius muscles were obtained simultaneously. Vibration frequencies were set at 50–150 Hz, with a 25 Hz step. Wave image quality in each frequency was analyzed using a phase-to-noise ratio (PNR) and clarity of propagating wave that was assessed by two readers qualitatively. In the supraspinatus muscle, the wave images were of good quality especially at frequencies >75 Hz. In the trapezius muscle, the wave images were of better quality at low frequencies (50 and 75 Hz) compared with high frequencies (100–150 Hz). The PNR of both muscles were higher at low frequencies. The mean stiffness in the trapezius muscle (7.26 ± 2.13 kPa at 75 Hz) was larger than those in the supraspinatus muscle (4.16 ± 0.50 kPa at 75 Hz). The results demonstrated that our MRE technique allows simultaneous assessment of the stiffness in the supraspinatus and trapezius muscles using a conventional MRI, and that optimal vibration frequency for simultaneous MRE of these muscles is 75 Hz. This technique provides a new means for early detection of abnormality in the shoulder.

Diffusion-magnetic resonance elastography (dMRE) is an emerging practical technique that can acquire diffusion magnetic resonance imaging and MRE simultaneously. However, a signal loss attributable to intravoxel phase dispersion (IVPD) interferes with the calculation of the apparent diffusion coefficient (ADC). This study presents an approach to dMRE that reduces the influence of IVPD by introducing a new pulse sequence. The existing and proposed techniques were performed using a phantom comprising five rods with different elasticities at 60 Hz vibration to investigate the accuracy of previous and proposed dMRE techniques. The measures of ADC and stiffness, obtained by using both dMRE techniques, were compared with conventional spin-echo (SE) diffusion and SE-MRE. Then, we evaluated those differences by using the mean of absolute differences (MAD) in each rod within the phantom. The results of the MAD of the stiffness from both dMRE techniques showed almost no difference. In contrast, the value of the ADC MAD (MAD ≒ 0.16 × 10−3 mm2/s), obtained in the soft region within the phantom with the previous dMRE technique, was large. This value was about 2.7 times that of the value produced by the proposed dMRE technique. This difference must reflect the degree of influence of IVPD in both techniques. These results demonstrate that our dMRE technique is a robust method for addressing the signal loss attributable to IVPD.

To demonstrate the feasibility of a novel MR elastography (MRE) technique based on a conventional gradient-echo type multi-echo MR sequence which does not need additional bipolar magnetic field gradients (motion encoding gradient: MEG), yet is sensitive to vibration. In a gradient-echo type multi-echo MR sequence, several images are produced from each echo of the train with different echo times (TEs). If these echoes are synchronized with the vibration, each readout's gradient lobes achieve a MEG-like effect, and the later generated echo causes a greater MEG-like effect. The sequence was tested for the tissue-mimicking agarose gel phantoms and the psoas major muscles of healthy volunteers. It was confirmed that the readout gradient lobes caused an MEG-like effect and the later TE images had higher sensitivity to vibrations. The magnitude image of later generated echo suffered the T2 decay and the susceptibility artifacts, but the wave image and elastogram of later generated echo were unaffected by these effects. In in vivo experiments, this method was able to measure the mean shear modulus of the psoas major muscle. From the results of phantom experiments and volunteer studies, it was shown that this method has clinical application potential.

We aimed to investigate the association between mobility and skeletal muscle strength by using magnetic resonance diffusion tensor imaging (DTI). This study included 20 healthy male volunteers (mean age, 21.8 ± 1.1 years). The maximum voluntary strength (MVC) of each participant was measured with the ankle joint in plantar and dorsal flexion using an instrument for measuring muscle strength. Moreover, magnetic resonance imaging (MRI) was performed with the ankle joint at rest, in plantar flexion, and in dorsal flexion. For imaging, a 1.5-T MRI device was used, and a diffusion-weighted stimulated echo-planar imaging pulse sequence. Tensor eigenvalues (λ), fractional anisotropy (FA), and the apparent diffusion coefficient (ADC) were calculated from data obtained by DTI. The resulting MRI data were compared to the data on muscle mobility or strength and statistically analyzed. Regarding changes in DTI indices during muscle movements, anisotropy of the tibialis anterior was significantly increased from rest to plantar flexion (P < 0.01), whereas no significant change was observed in dorsal flexion (n.s.). In contrast, the extent of significant changes in anisotropy of the medial gastrocnemius (mGC) and soleus (SOL) was small at plantar flexion (mGC, P < 0.01; SOL, n.s.), whereas the indices were significantly increased at dorsal flexion (P < 0.01). Regarding the correlation between MVC of each skeletal muscle and the DTI indices, FA and λ3 were significantly correlated in movements involving the muscles, whereas no significant correlation was observed in movements not involving them. Changes in intramuscular water molecules by elongation and contraction of the skeletal muscle fibers could be assumed to affect changes in diffusional anisotropy. When muscles contract, the space between myocytes was reduced and they might become increasingly dense. Moreover, diffusional anisotropy increased with increasing MVC, whereas ADC remained unchanged. DTI was suggested to produce measurements similar to the degree of muscle strength.

We developed a facile one-pot fabrication process for magnetic iron oxide–calcium phosphate (IO–CaP) composite nanoparticles via coprecipitation in labile supersaturated CaP solutions containing IO nanocrystals. All the source solutions used were clinically approved for injection, including water and magnetic IO nanocrystals (ferucarbotran, used as a negative magnetic resonance imaging (MRI) contrast agent). This ensured that the resulting nanoparticles were pathogen- and endotoxin-free. The dispersants used were clinically approved heparin sodium (heparin) or adenosine triphosphate disodium hydrate (ATP), which were added to the IO-containing labile supersaturated CaP solutions. Both heparin and ATP coprecipitated with CaP and ferucarbotran to form heparin- and ATP-modified IO–CaP nanoparticles, respectively, with a hydrodynamic diameter of a few hundred nanometers. Both the resulting nanoparticles exhibited relatively large negative zeta potentials, caused by the negatively charged functional groups in heparin and ATP, which improved the particle dispersibility when compared to non-modified IO–CaP nanoparticles. The heparin-modified IO–CaP nanoparticles were effectively ingested by murine macrophages (RAW264.7) without showing significant cytotoxicity but barely ingested by non-phagocytotic human umbilical vein endothelial cells, indicating the potential of these nanoparticles for targeted delivery to macrophages. The heparin-modified IO–CaP nanoparticles exhibited a negative contrast enhancing ability for MRI. Our results show that IO–CaP nanoparticles have potential as delivery and MRI contrast agents for macrophages.

Purpose: Commonly used diffusion weighted (DW) imaging such as DW spin echo (SE) type echo planar imaging (DW-SE-EPI) is known to be a snapshot-like acquisition and to have a relatively high signal-to-noise ratio. Spiral MRI sequence (SPIRAL) has characteristics similar to these of EPI, but it has rarely been used for diffusion-weighted imaging (DWI). In vivo DW-SPIRAL of the rat brain has almost never been reported. Our purpose in this study was to examine the potential of SE-type two-dimensional (2D) multi-shot spiral acquisition MRI for apparent diffusion coefficient (ADC) mapping of the rat brain in vivo. Materials and Methods: We made an SE-type DW-2D-spiral MRI sequence (DW-SPIRAL) which was prepared on a 2.0-T animal-experiment MR scanner. Comparing the phantom experimental result of DW-SPIRAL with the phantom experimental result of DW SE-type echo-planar imaging (DW-SE-EPI) and conventional DW spin echo imaging (DW-SE), we estimated the characteristics of DW-SPIRAL and assessed the clinical application of DW-SPIRAL in an animal experiment on the rat brain. Results: There was not much difference between the calculated water/glycerol phantom diffusion coefficient of DW-SPIRAL and the calculated diffusion coefficient of DW-SE. This result shows that the DW-SPIRAL sequence is appropriate for use in diffusion weighted imaging. There were fewer phantom image distortions and ghosting artifacts with DW-SPIRAL than with DW-SE-EPI, and this tendency was similar in the animal experiment on the rat brain. Conclusion: The DW-SPIRAL sequence had been successfully tested in phantom experiments and rat brain experiments. It has been demonstrated that the DW-SPIRAL sequence is capable of producing in vivo rat brain DWI.

Objective Using an anthropomorphic phantom, we have investigated the feasibility of digital tomosynthesis (DT) of flat-panel detector (FPD) radiography to reduce radiation dose for sinonasal examination compared to multi-detector computed tomography (MDCT). Materials and methods A female Rando phantom was scanned covering frontal to maxillary sinus using the clinically routine protocol by both 64-detector CT (120 kV, 200 mA s, and 1.375-pitch) and DT radiography (80 kV, 1.0 mA s per projection, 60 projections, 40° sweep, and posterior–anterior projections). Glass dosimeters were used to measure the radiation dose to internal organs including the thyroid gland, brain, submandibular gland, and the surface dose at various sites including the eyes during those scans. We compared the radiation dose to those anatomies between both modalities. Results In DT radiography, the doses of the thyroid gland, brain, submandibular gland, skin, and eyes were 230 ± 90 μGy, 1770 ± 560 μGy, 1400 ± 80 μGy, 1160 ± 2100 μGy, and 112 ± 6 μGy, respectively. These doses were reduced to approximately 1/5, 1/8, 1/12, 1/17, and 1/290 of the respective MDCT dose. Conclusion For sinonasal examinations, DT radiography enables dramatic reduction in radiation exposure and dose to the head and neck region, particularly to the lens of the eye.

Magnetic resonance elastography (MRE) is designed for imaging the mechanical properties of soft tissues. However, the interpretation of shear modulus distribution is often confusing and cumbersome. For reliable evaluation, a common practice is to specify the regions of interest and consider regional elasticity. Such an experience-dependent protocol is susceptible to intrapersonal and interpersonal variability. In this study we propose to remodel shear modulus distribution with piecewise constant level sets by referring to the corresponding magnitude image. Optimal segmentation and registration are achieved by a new hybrid level set model comprised of alternating global and local region competitions. Experimental results on the simulated MRE data sets show that the mean error of elasticity reconstruction is 11.33% for local frequency estimation and 18.87% for algebraic inversion of differential equation. Piecewise constant level set modeling is effective to improve the quality of shear modulus distribution, and facilitates MRE analysis and interpretation.

Background 18q-Syndrome is a chromosomal disorder exhibiting various symptoms arising from the central nervous system. Brain magnetic resonance imaging (MRI) of patients with this syndrome usually demonstrates abnormal white matter intensities. This is widely believed to be due to impaired myelin formation because this syndrome involves the deletion of the myelin basic protein (MBP) gene in 18q23. However, this hypothesis has not been confirmed by actual pathology because early death is unusual and autopsy rarely performed. Patient A 6-year-old boy with ring chromosome 18 syndrome was examined by genetic analysis for the MBP gene, brain MRI, and autopsy. Results Haploinsufficiency of the MBP gene was confirmed. T2-weighted MRI revealed diffuse high intensities throughout the cerebral white matter. Pathological examination showed the cerebral white matter to be uniformly stained by Kluver–Barrera and MBP immunohistochemical staining. Oligodendrocytes were immunoreactive for proteolipid protein and ferritin but not MBP. Electron microscopy revealed clusters of axons wrapped in compact myelin sheaths with distinct major dense lines. Holzer and immunohistochemical staining for glial fibrillary acidic protein showed extensive staining of the white matter and an increased number of glial filaments. Conclusions This pathological study demonstrated that in this disorder, the brain was well myelinated, contrary to established hypotheses about this disorder. The MRI signal abnormalities in 18q-syndrome could be attributed to gliosis and not to dysmyelination.

Objective Quantitative MR imaging techniques of degenerative cartilage have been reported as useful indicators of degenerative changes in cartilage extracellular matrix, which consists of proteoglycans, collagen, non-collagenous proteins, and water. Apparent diffusion coefficient (ADC) mapping of cartilage has been shown to correlate mainly with the water content of the cartilage. As the water content of the cartilage in turn correlates with its viscoelasticity, which directly affects the mechanical strength of articular cartilage, ADC can serve as a potentially useful indicator of the mechanical strength of cartilage. The aim of this study was to investigate the correlation between ADC and viscoelasticity as measured by indentation testing. Materials and methods Fresh porcine knee joints (n = 20, age 6 months) were obtained from a local abattoir. ADC of porcine knee cartilage was measured using a 3-Tesla MRI. Indentation testing was performed on an electromechanical precision-controlled system, and viscosity coefficient and relaxation time were measured as additional indicators of the viscoelasticity of cartilage. For security, boards containing lead are commonly used to partition off rooms adjacent to radiation facilities. From the viewpoints of environmental safety and of material recycling during the process of remodeling or repair, however, the trend is toward avoiding the use of lead-containing radiation-protection board. Consequently, we developed a lead-free board(W 910xH 1820xD 12.5 mm)composed of a mixed material incorporating calcium sulfate and barium sulfate to be used as a partition at a diagnostic X-ray institution. The mass attenuation coefficients of the lead-free board were lower by about 12% and 60% for energy ranges of 25 approximately 37 keV and 38 approximately 41 keV, respectively, as compared with those of lead. Lead equivalents for the newly developed board(12.5 mm thickness)were 0.87+/-0.01 mmPb at 100 kV tube voltage, 0.60+/-0.01 at 125 kV, and 0.47+/-0.01 at 150 kV. A double-layer lead-free board proved to have enough shielding ability even at usual photographic frequencies.

In this work we report on the development of a novel technique for fat-saturated three-dimensional (3D) diffusion-weighted (DW) MRI sequence based upon 3D magnetization-prepared rapid gradient-echo (3D-MP-RAGE). In order to saturate fat, two kinds of procedures were competed CHESS-DW-3D-MP-RAGE sequence (CHESS -3D-DWI) and DW-3D-MP-WE-RAGE sequence (WE-3D-DWI) “chemical shift selective: CHESS method vs. water-excitation: WE method”. The CHESS-3D-DWI sequence and WE-3D-DWI sequence were compared in terms of their degree of fat suppression. In CHESS-3D-DWI sequence a preparation phase with a “CHESS-90ºRF-motion probing gradient: MPG-180ºRF-MPG-90ºRF” pulse-train was used to sensitize the magnetization to fat-saturated diffusion. In contrast, WE-3DDWI sequence a RAGE-excitation pulse with a “binominal -pulse 1-1 or 1-2-1” was selected to water-excited (fat-saturated) diffusion imaging. These imaging were done during in vivo studies using an animal experiment. From experimental results obtained with a phantom, the effect of diffusion weighting and the effect of fat-saturation were confirmed. Fat-saturation was much better in the WE-3D-DWI sequence than CHESS-3D-DWI sequence. From rat experimental results, fat-saturated diffusion-weighted image data were obtained. It is important to evaluate the elasticity of tissue-engineered cartilage, for evaluating its structural strength. However, in vivo evaluation of tissue-engineered cartilage elasticity has not been established. On the other hand, since the speed of sound (SOS) is available for elasticity evaluation, we have proposed in vivo measurement method of SOS using ultrasound and MR images. This method determines the SOS based on the thickness measurement using the MR image and the time-of-flight (TOF) measurement using the ultrasound. In this study, this method was applied to the SOS measurement in tissue-engineered cartilage, and then relation between in vivo SOS and elasticity was investigated. As the result, in vivo SOS had a high correlation with Young's modulus. This result reveals that in vivo evaluation of tissue-engineered cartilage elasticity is available through in vivo SOS measurement.

BACKGROUND Transmission and tomographic X-ray measurements are useful in assessing bone structures, but only a few studies have examined cartilage growth because of the poor contrast in conventional X-ray imaging. OBJECTIVE In this study, we attempted to use the linear attenuation coefficient (LAC) as a metric of tissue-engineered cartilage development, which would be useful in high-throughput screening of cartilage products. METHODS Assuming that the LAC is related to the amount of extracellular matrix (ECM) in terms of the density and its atomic components, we measured X-ray absorption through tissue-engineered cartilage constructs. Characteristic X-ray beams from a molybdenum microfocus X-ray tube were employed to avoid beam hardening. The correlation of the LAC with mechanical properties was analyzed for verification. RESULTS The LAC was higher for chondrocyte constructs and lower for fibroblast-dominant constructs and was consistent with the quantification of toluidine blue staining, which is a proof of ECM production. The LAC was positively correlated with the bending modulus but negatively correlated with the dynamic elastic modulus and stiffness, possibly because of the remaining scaffold. CONCLUSIONS The LAC has the potential to be used as a metric of development of tissue-engineered cartilage. However, the calcified regions should be excluded from analysis to avoid decreasing the correlation between the LAC and the amount of ECM.

The advantages of three-dimensional (3D) acquisition are that you obtain thinner and more slices with better profiles, and better signal-to-noise ratio for an equivalent slice thickness. Three-dimensional acquisition is preferable for obtaining contiguous thin-slice MR images. However, the acquisition time extends compared with the two-dimensional acquisition because the second phase-encode axis is applied by the 3D acquisition. Therefore, 3D acquisition should be a high-speed imaging method. In this paper, a new diffusion-sensitive 3D magnetization-prepared rapid gradient-echo (3D MP-RAGE) sequence was studied. In this sequence, a preparation phase with a 90 degrees RF-motion proving gradient (MPG): MPG-180 degrees RF-MPG-90 degrees RF pulse train (diffusion-weighted driven-equilibrium Fourier transform) was used to sensitize the magnetization to diffusion. Centric k-space acquisition order is necessary to minimize saturation effects from tissues with short relaxation times. From phantom experimental results, the effect of the diffusion weighting was changed by the centric vs. sequential k-space acquisition order. The effect of centric k-space acquisition order was larger than the effect of sequential k-space acquisition order. The contrast of centric k-space acquisition order became equal to the contrast of conventional diffusion-weighted spin echo. From rat experimental results, small isotropic diffusion-weighted image data (voxel size: 0.625 x 0.625 x 0.625 mm3) were obtained. This sequence was useful in vivo.

In order to detect the disease and degeneration of cartilage early, evaluations of acoustic properties are expected, but the physical meaning should be clarified. On the other hand, we have proposed a multimodal method using ultrasound (US) and magnetic resonance (MR) images for in vivo SOS measurement. In this study, the method was applied for the tissue-engineered cartilage, and the SOS was compared with T1, T2, apparent diffusion coefficient (ADC), attenuation coefficient (ATT), and Young’s modulus (YM). All procedures were approved by the Animal Care and Use Committee of our institution. 12-week-old male rats were used, and five different types of tissue-engineered cartilage was subcutaneously implanted in the back of rat (n = 30). Since the speed of sound (SOS) reflects tissue characteristics and is expected as an evaluation index of elasticity and water content, the noninvasive measurement of SOS is eagerly anticipated. However, it is difficult to measure the SOS by using an ultrasound device alone. Therefore, we have presented a noninvasive measurement method of SOS using ultrasound (US) and magnetic resonance (MR) images. By this method, we determine the longitudinal SOS based on the thickness measurement using the MR image and the time of flight (TOF) measurement using the US image. The accuracy of SOS measurement is affected by the accuracy of image registration and the accuracy of thickness measurements in the MR and US images. In this study, we address the accuracy improvement in the latter thickness measurement, and present an image-processing-based method for improving the accuracy of thickness measurement. The method was investigated by using in vivo data obtained from a tissue-engineered cartilage implanted in the back of a rat, with an unclear boundary.

In this study, the mutual fusion of chondrocyte pellets was promoted in order to produce large-sized tissue-engineered cartilage with a three-dimensional (3D) shape. Five pellets of human auricular chondrocytes were first prepared, which were then incubated in an agarose mold. After 3 weeks of culture in matrix production-promoting medium under 5.78g/cm(2) compression, the tissue-engineered cartilage showed a sufficient mechanical strength. To confirm the usefulness of these methods, a transplantation experiment was performed using beagles. Tissue-engineered cartilage prepared with 50 pellets of beagle chondrocytes was transplanted subcutaneously into the cell-donor dog for 2 months. The tissue-engineered cartilage of the beagles maintained a rod-like shape, even after harvest. Histology showed fair cartilage regeneration. Furthermore, 20 pellets were made and placed on a beta-tricalcium phosphate prism, and this was then incubated within the agarose mold for 3 weeks. The construct was transplanted into a bone/cartilage defect in the cell-donor beagle. After 2 months, bone and cartilage regeneration was identified on micro-computed tomography and magnetic resonance imaging. This approach involving the fusion of small pellets into a large structure enabled the production of 3D tissue-engineered cartilage that was close to physiological cartilage tissue in property, without conventional polyper scaffolds.

Liver has complex structure of blood vessels. There are two inlets of the blood flow in a liver. One is hepatic artery in which the blood flow from a heart to supply oxygen. Another is portal vein in which the blood flow from the digestive organs to maintain the metabolic and the detoxification processes. These vessels join at hepatic lobule which is consisted by hepatocyte. The hepatic lobule has a central vein and the central veins drain the blood flow into a hepatic vein. Radiofrequency ablation (RFA) is an important modality in tumor treatment. One of the challenging issues is how to control and evaluate RFA treatment. This study is one of the first systems to examine magnetic resonance elastography (MRE) for RFA evaluation. The ex vivo experiments were designed and performed on bovine and porcine liver tissues respectively. It is found that the contrast ratios of the common imaging protocols were 1:1.09 (2.44:2.65) only for bovine tissues and 1:1.12 (2.19:2.45) for porcine tissues, but they were enhanced to 1:3.03 (25.74:78.04) and 1:3.66 (9.16:33.55) respectively on MRE. In conclusion, MRE is a good solution for RFA evaluation.

The purpose of this study was to develop a new technique for a powerful compact MR elastography (MRE) actuator based on a pneumatic ball-vibrator. This is a compact actuator that generates powerful centrifugal force vibrations via high speed revolutions of an internal ball using compressed air. This equipment is easy to handle due to its simple principles and structure. Vibration frequency and centrifugal force are freely adjustable via air pressure changes (air flow volume), and replacement of the internal ball. In order to achieve MRI compatibility, all parts were constructed from non-ferromagnetic materials. Vibration amplitudes (displacements) were measured optically by a laser displacement sensor. From a bench test of displacement, even though the vibration frequency increased, the amount of displacement did not decrease. An essential step in MRE is the generation of mechanical waves within tissue via an actuator, and MRE sequences are synchronized to several phase offsets of vibration. In this system, the phase offset was detected by a four-channel optical-fiber sensor, and it was used as an MRI trigger signal. In an agarose gel phantom experiment, this actuator was used to make an MR elastogram. This study shows that the use of a ball actuator for MRE is feasible.

The aim of our study was to simulate an effective diffusion-weighted imaging (DWI) pulse sequence and to evaluate the best b value in skeletal muscle. The evaluated pulse sequences were spin echo (SE), stimulated echo (STE), and gradient-recalled echo (GRE). The signal intensity changed in some DWI pulse sequences when the b value was changed. Moreover, the static magnetic field of 1.5 T was compared with that of 3.0 T. Next, diffusion tensor imaging of the human skeletal muscle was measured. The experimental subjects were healthy male volunteers (n = 7; age 21.8 ± 1.1 years). We changed the b value in steps of 100 s/mm2 from 0 to 2000 s/mm2, and the diffusion values [fractional anisotropy (FA), λ 1, λ 2, and λ 3] were calculated based on the data. The STE method could maintain signals up to b values of 1300 s/mm2, but the SE and GRE methods suffered from high noise. In the human study, the mean FA (±SD) was 0.41 ± 0.02 in the tibialis anterior muscle (TA) and 0.31 ± 0.02 in the soleus muscle (SOL) at a b value of 1000 s/mm2. The purpose of this study was to examine the potential of diffusion-weighted (DW) three-dimensional (3D) MP-RAGE sequence for cerebral ischemia of a rat brain in vivo. The prototype of a DW-3D-MP-RAGE (3D-DWI) sequence was implemented at 2.0T animal experiment MR imager. The advantage of 3D imaging are that you obtain thinner and more slices with better profiles, and better signal-to-noise ratio for an equivalent slice thickness. Since 3D imaging is preferable for obtaining contiguous thin-slice MR images, image data with 3D imaging are expected to volume estimate of imaging area. Three-dimensional calculated absolute apparent diffusion coefficient (ADC) maps were compared with coronal stained (triphenyltetrazolium chloride: TTC) brain sections. From rat experimental results, the region of cerebral ischemia with 3D-DWI and the region of stained brain sections were well in agreement. Applying tissue-engineered cartilage in a clinical setting requires noninvasive evaluation to detect the maturity of the cartilage. Magnetic resonance imaging (MRI) of articular cartilage has been widely accepted and applied clinically in recent years. In this study, we evaluated the negative fixed-charge density (nFCD) of tissue-engineered cartilage using gadolinium-enhanced MRI and determined the relationship between nFCD and biomechanical properties. To reconstruct cartilage tissue, articular chondrocytes from bovine humeral heads were embedded in agarose gel and cultured in vitro for up to 4 weeks. The nFCD of the cartilage was determined using the MRI gadolinium exclusion method. The equilibrium modulus was determined using a compressive stress relaxation test, and the dynamic modulus was determined by a dynamic compression test. The equilibrium compressive modulus and dynamic modulus of the tissue-engineered cartilage increased with an increase in culture time. The nFCD value—as determined with the [Gd-DTPA2−] measurement using the MRI technique—increased with culture time. In the regression analysis, nFCD showed significant correlations with equilibrium compressive modulus and dynamic modulus. From these results, gadolinium-enhanced MRI measurements can serve as a useful predictor of the biomechanical properties of tissue-engineered cartilage.

A q-space diffusion MR imaging is able to detect microstructure (10μm ∼ ) dynamically. Recently this method is suggested as investigating neuronal generation and degeneration in spinal cord. The q-space data sets are measured by two-dimensional (2D) multi-slice imaging. In order to perform 2D multi-slice imaging, it is necessary to have a few millimeters of slice thickness and slice gap. Multi-slice imaging by an isotropic voxel in the submicrometer range is difficult. While on the other hand, three-dimensional (3D) MR imaging has the advantage of very thin slices with no slice gap (contiguous slices), and it allows acquisition in a small isotropic voxel compared with 2D multi-slice imaging. For detecting microstructural changes in neuron, q-space data sets should be measured by 3D diffusion imaging which have small isotropic voxel. The purpose of this study was to examine a 3D diffusion-weighted MP-RAGE MR sequence (3D-DWI) for q-space imaging of the plant phantom. In post-processing, q-space analysis were performed and we had several analytic volume map of fresh celery (average displacement, zero-displacement probability, kurtosis). This study we had accomplished to get high resolution q-space map of plant phantom strongly reflected diffusion restriction. We would now like to go on to develop this method by applying rat spinal cord for effectiveness in vivo.
