Abstract Pandemics like SARS-Cov-2 very frequently have their origin in different animals and in particular herds of camels could be a source of zoonotic diseases. This study took advantage on a highly sensitive and adaptable method for the fast and reliable detection of viral antibodies in camels using low-cost equipment. Magnetic nanoparticles (MNP) have high variability in their functionalization with different peptides and proteins. We confirm that 3-aminopropyl triethoxysilane (APTES)-coated MNP could be functionalized with viral proteins. The protein loading could be confirmed by simple loading controls using FACS-analysis (p φn on selected critical point of the fifth higher harmonic (n = 5th). Here, positive sera display highly significant signal increase over the control or negative sera. Furthermore, a clear distinction could be made in antibody detection as an immune response to closely related viruses (SARS-CoV2 and MERS). Using modified MNPs along with COMPASS offers a fast and reliable method that is less cost intensive than current technologies and offers the possibility to be quickly adapted in case of new occurring viral infections. Key points • COMPASS (critical offset magnetic particle spectroscopy) allows the fast detection of antibodies. • Magnetic nanoparticles can be adapted by exchange of the linked bait molecule. • Antibodies could be detected in camel sera without washing steps within seconds.

After peptide functionalization of superparamagnetic iron oxide nanoparticles (SPIONs) with an appropriate interface, they can be used for the separation of sepsis-associated yeasts like Candida albicans. Due to their magnetic properties, the magnetic extraction of the particles in the presence of an external magnetic field ensures the accumulation of the targeted yeast.Materials and Methods: In this study, we used SPIONs coated with 3-aminopropyltriethoxysilane (APTES) and functionalized with a peptide originating from GP340 (SPION-APTES-Pep). For the first time, we investigate whether this system is suitable for the separation and enrichment of Candida albicans, we investigated its physicochemical properties and by thermogravimetric analysis we determined the amount of peptide on the SPIONs. Further, the toxicological profile was evaluated by recording cell cycle and DNA degradation. The separation efficiency was investigated using Candida albicans in different experimental settings, and regrowth experiments were carried out to show the use of SPION-APTES-Pep as a sample preparation method for the identification of fungal infections.Conclusion: SPION-APTES-Pep can magnetically remove more than 80% of the microorganism and with a high selective host-pathogen distinction Candida albicans from water-based media and about 55% in blood after 8 minutes processing without compromising effects on the cell cycle of human blood cells. Long-term functional storage of therapeutic proteins at room temperature has been an eternal challenge. Inspired by the cellular cooperativity of proteins, we have taken a step forward to address this challenge by cohabitating Immunoglobulin G (IgG1) with a food protein gelatin in the solid-state at room temperature. Interestingly, IgG1 remained functionally active for a record 14 months revealed from the western-blot assay. Further quantification by HP-LC analysis showed 100% structural integrity of IgG1 with no degradation in the gelatin matrix during this period. The developed formulation has a direct application in oral medical nutrition therapy to cure gastrointestinal microbial infections. Also the strategy provides a robust energy economic alternative to the protein engineering methods for long-term functional storage of therapeutic proteins at room temperature.

Epilepsy surgery can potentially cure drug-resistant epilepsy, but careful presurgical evaluation is vital to select patients who will profit from such an intervention. Many epilepsy surgery programs offer extensive presurgical evaluation including several days of video-EEG monitoring. Non-lesional epilepsy cases are rare among epilepsy surgery patients. We set up a lesion-orientated paediatric epilepsy surgery program for patients with clearly localized lesions with limited presurgical diagnostics, in particular, with a maximum of 48 hours of non-invasive EEG monitoring that did not necessarily include ictal EEGs.We retrospectively evaluated the outcome of patients who were operated on within our epilepsy surgery program with respect to seizure freedom.Fifty-two children and adolescents with MRI lesions at a mean age of 8.27 ±4.83 years (range: 0.17-18.87) underwent a resective procedure. The most frequent surgery was a hemispherotomy. Overall seizure freedom was 81.8% after 12 months and 85.6% after a median observation period of 20.45 months. Seizure frequency was reduced50% in all other patients. Preoperative recording of an ictal EEG on the side of surgery had no effect on postoperative seizure outcome (p= 0.697), nor did recording of epileptiform discharges on the ipsilateral (p= 0.538) and contralateral side (p= 0.147).Our findings highlight the high success rate using a lesion-orientated epilepsy surgical approach with reduced presurgical video-EEG monitoring in the paediatric epilepsy population. Our data show that it is possible to reduce the complex pre-surgical work-up for epilepsy in children and adolescents by asking the basic question: "Is there any reason why the lesion should not be resected".

Coprolites are one of the sources of evidence most commonly used in paleoparasitological studies. However, the zoological origin identification is not always possible. Parasites of the genus Eimeria (Apicomplexa: Eimeriidae) are obligate protozoan which have evolved to exhibit a great diversity of host range, and present strict host specificity. Usually, the identification of Eimeria spp. can be done by morphological characteristics such as size and shape of the oocysts. Moreover, molecular diagnostic methods such as PCR have proven to be very useful methodologies for an accurate taxonomic identification. In recent years, molecular techniques have started to be applied in paleoparasitological studies. Here, we provide the first integrative approach that combine light microscopic studies and ancient DNA (aDNA) in order to detect and identify Eimeria spp. on artiodactyl coprolites recovered from an archeological context from northwestern Patagonia, Argentina. The main objective is to discuss the advantages of complementing these two approaches as a promising tool to identify the zoological origin of coprolites. Furthermore, we provide paleoparasitological data about the presence of Eimeria spp. in coprolites of South American ungulates. Light microscopic studies allowed the identification of oocysts attributed to Eimeria spp. in 13 of the 40 coprolites. Amplification and sequencing of the aDNA ITS1 detected the presence of Eimeria spp. in 5 of the 10 samples, and allowed the taxonomic identification of these sequences as belonging to Eimeria arloingi. Furthermore, the results obtained from cytochrome b gene (Cytb) analysis confirmed the identity of the host as Capra hircus. Results displayed the presence of coccidiosis in goats from historical times. This study highlights that both techniques are complementary and promotes the use of the identification of Eimeria spp. in ancient samples in order to identify the zoological origin.

This paper critically examines the metaphorical use of medical terms in philosophy. Three examples selected from distinct philosophical contexts demonstrate that such terms have been employed as metaphors both to describe the practice of philosophising and historically to diagnose philosophical positions. The selected examples are (i) the title of Avicenna's main philosophical work, The Book of Healing, (ii) the criticism of medical metaphors in Enlightenment philosophy, and (iii) recent historical diagnoses in philosophy. The underlying epistemological assumptions of all three contexts are reconstructed to critically analyse the medical metaphors. Through this tripartite synopsis, I arrive at a normative conclusions medical metaphors, such as the "healing of the soul" or "pathology of reason", do not stand up to the critique of Enlightenment and are obsolete against the theoretical background of my reference texts.

ACL reconstruction tunnel location is an important predictor for outcomes after surgery. The aim was to establish 3D and 2D MRI radiological measurements for native ACL tibial footprint that can provide information to facilitate pre-operative planning for anatomical graft placement. The measurements were also correlated in a subset of patients on arthroscopy.Retrospective evaluation of a consecutive series of knee MRIs with both 2D and 3D MR imaging was performed in 101 patients with 43 men and 66 women and ages 39.5 ± 11.9 years. Two measurements were obtained, tibial to ACL and intermeniscal ligament to ACL (T-ACL) and (IM-ACL), respectively. In a cohort of 18 patients who underwent knee arthroscopy, the T-ACL and IML-ACL distances were also determined by an orthopedic surgeon using a standard scale. ICC, Pearson correlation, and Bland-Altman plot were generated.For readers 1 and 2, the mean differences between 2D and 3D measurements of T-ACL and IM-ACL were 1.17 and 1.03 mm and 0.65 and 0.65 mm, respectively. The 2D measurements of T-ACL and IM-ACL were larger than the 3D measurements for both readers. The inter-reader reliability was excellent on 2D (0.81-0.96) and fair to excellent on 3D MRI (0.59-0.90). The mean arthroscopic IML-ACL was closer to that of 3D MRI compared to 2D MRI. The mean arthroscopic T-ACL was closer to 2D MRI than 3D MRI.Both 2D and 3D MRI show inter-reader reliability with small inter-modality mean differences in the measurements from the tibial or inter-meniscal ligament margins.• The mean differences between 2D and 3D measurements of tibia-ACL and intermeniscal ligament-ACL are small (1.2 mm). • As compared to arthroscopy, the mean T-ACL and IML-ACL were closer to measurements from 2D and 3D MRI, respectively. • Both 2D and 3D MRI can be reliably used to delineate ACL foot plate anatomy.

Although urbanization poses one of the largest threats for biodiversity, only few studies have so far examined its impact in large Central European cities. Our study aimed at investigating the effects of urbanization on bird diversity using two methods to describe the urban environment. The first measure used the degree of sealing, portion of traffic infrastructure, number of floors of the tallest building at a site scale (100-m radius), and the distance from the city center, while the second one relied on land-cover data at a local scale (1000-m radius). We conducted bird surveys at 761 sites across Hamburg. Bird diversity was assessed as species richness, abundance, and Shannon index. Additionally, evenness was calculated. Both urbanization measures represented a rural-to-urban gradient and were negatively correlated with bird diversity as well as evenness. At the site scale, the degree of sealing had the strongest negative effect on bird diversity followed by the portion of traffic infrastructure and the tallest building. At the local scale, artificial surfaces proved to have the strongest negative effect, while agricultural areas and forests were positively associated with bird diversity. In general, models using the site scale for measuring urbanization showed higher associations with diversity. The results emphasize the importance of unsealed areas in the urban environment and of natural habitats in the periphery for bird diversity in Central European cities with millions of human inhabitants. Considering this for future city planning can help to preserve biodiversity in the urban environment, increase bird diversity in development areas, and thus improve living conditions for people.

Critical Offset Magnetic PArticle SpectroScopy (COMPASS) is a new method that allows to detect slight changes in the mobility of particles in a solution very sensitively. While first applications in detecting biomarkers such as SARS CoV 2 antibodies show very promising results, the full potential of the technique and the underlying physical effects were yet only considered superficially. Current bioassays for detection of antibodies or antigens such as ELISA (Enzyme-linked Immunosorbent Assay) are relatively inflexible, expensive and time-consuming. Upcoming methods, such as ACS (AC susceptometry) or MPS (Magnetic Particle Spectroscopy), exploit the magnetization response of functionalized MNP ensembles to assess specific information about the MNP mobility as well as conjugations of chemical or biological compounds on their surface. Both methods have shown promising results in the past but cannot reach the sensitivity of above-mentioned techniques. We used a novel method based on a modified MPS being sensitive to minimal changes in mobility of MNP ensembles. This facilitates robust and easy-to-handle measurements of minimal changes in the diameter of MNPs. As an example, we detected SARS-CoV-2 antibodies binding to the S1 antigen on the surface of functionalized MNPs. Emergency Psychiatry is evolving. In an environment that lacks a clear evidence base, and where a constellation of factors is driving up Emergency Department presentation rates and lengths of stay, several stakeholders are working towards and clamouring for change. With the goal of collaborating with such parties, we believe Emergency Psychiatrists should position themselves to establish and advocate for best-practice change in culture, research, clinical care and training, and funding in the provision of mental health crisis care. To this end, we have formed the NSW Emergency Psychiatry Network, a group of Emergency Psychiatrists with a broad experience in a range of settings, from tertiary metropolitan emergency facilities with access to subspecialty psychiatric services, to rural and remote emergency settings with sporadic in-reach from local mental health services and telehealth. We unanimously recognise the need to upskill both Emergency Department and Mental Health clinicians in crisis care, and the need for committed, evidence-based Mental Health resourcing within Emergency Departments.

Textile reinforced concrete was developed in recent years into a construction technique that has its benefits and advantages in applications where conventional types of reinforcement have their limits. The current, minimum slab thickness for steel-reinforced concrete façade slabs is 7.0 cm; this is due to the minimum, required concrete cover to ensure adequate corrosion protection. Façade slab anchors for these slab thicknesses are building authority approved. As corrosion protection is not an issue for textile reinforced concrete, the minimum thickness for the concrete cover can be significantly reduced. The requirement for component thickness is now determined by the load-bearing capacity and by production-related boundary conditions. For practical building reasons, panel thicknesses of 3.0 cm have proven to be the best choice. Compared to steel-reinforced façade panels, this is a weight and thickness reduction of almost 60%. Thin concrete elements are of great interest in cases when the thickness or the weight of the panels is largely limited e.g. because of adjoining concrete elements in renovation or upgrade projects, retrofitting or improvements. Compared to other building materials, concrete has characteristic advantages in building physics and fire protection properties, irrespective of the thickness. Obviously, minimal thicknesses place extra demands on planning and construction. Especially effects on concrete, punching, splitting and concrete breakout must be examined in experiments. This is an overview of calculation and test methods. Results are provided to show the bearing behaviour of fixings in thin, textile reinforced concrete slabs. The design rules are explained and the results are illustrated.

Nearly one-third of all infants with epilepsy develop drug-resistant epilepsy. Although epilepsy surgery is a well-established therapy across all age groups, there might be a reluctance to operate on infants in the first six months of life due to unique surgical and anesthesiologic difficulties.We performed a meta-analysis and systematic review to assess the outcome and complication rate of epilepsy surgery in infants operated on ≤ six months of life.158 infants underwent epilepsy surgery, most frequently a hemispherotomy rather than focal surgery. Overall seizure freedom after surgery was 65.6% [CI: 0.5785; 0.7261], with higher seizure-free rates following hemispherotomy (71%) than after focal surgery (58%). Complications occurred in 27.7% [0.1794; 0.4004] of patients. Most prevalently, a hydrocephalus developed in 20 out of 136 cases (14.71%). Anti-seizure medication (ASM) was discontinued in 21.5% [0.1431; 0.3100] and reduced in 85.9% [0.515; 0.9721] of 93 patients postoperatively. 84.6% of infants displayed cognitive impairment (development quotient (DQ)85) preoperatively. After surgery, there was a trend toward a cognitive gain. However, cognitive gain was seen almost exclusively in seizure-free patients.Excellent seizure control can be achieved with epilepsy surgery in the first six months of life, a large proportion of patients are able to reduce or discontinue ASM. Data regarding cognitive outcome are promising, but also show that the primary goal should be to achieve seizure freedom. Given the more difficult surgical conditions, epilepsy surgery in the first six months of life should only be performed in specialized epilepsy centers.

Introduction Optic pathway gliomas are often asymptomatic tumors occurring in children with neurofibromatosis type 1 (NF1 + OPG) or sporadically (spOPG). Treatment is usually prompted by visual loss and/or tumor progression on MRI. The aim of this study was to investigate the relationship between visual acuity (VA), tumor growth, and contrast enhancement to provide more distinct indications for the administration of gadolinium-based contrast agents. Methods Tumor load was retrospectively measured and enhancement semi-quantitatively scored on 298 MRIs of 35 patients (63% NF1 + OPG). Spearman rank correlation between tumor load and enhancement was calculated and a linear mixed model used to examine the influence of tumor load and enhancement on corresponding VA tests (LogMAR). Results The optic nerve width in NF1 + OPGs was strongly associated with VA (regression coefficient 0.75; confidence interval 0.61—0.88), but weakly with enhancement (0.06; −0.04—0.15). In spOPGs, tumor volume and optic nerve width were more relevant (0.31; −0.19—0.81 and 0.39; 0.05—0.73) than enhancement (0.09; −0.09—0.27). Conclusions Tumor load measures may be more relevant for the surveillance of optic pathway gliomas than enhancement, given that VA is the relevant outcome parameter. Regular contrast administration should therefore be questioned in these patients.

Top and bottom surfaces of polymer films are used to construct interfaces in heterojunction based devices, affecting device figure of merit significantly with their different aggregation states.

Vapor-phase film deposition of metal-organic frameworks (MOFs) would facilitate the integration of these materials into electronic devices. We studied the vapor-phase layer-by-layer deposition of zeolitic imidazolate framework 8 (ZIF-8) by consecutive, self-saturating reactions of diethyl zinc, water, and 2-methylimidazole on a substrate. Two approaches were compared: (1) Direct ZIF-8 "molecular layer deposition" (MLD), which enables a nanometer-resolution thickness control and employs only self-saturating reactions, resulting in smooth films that are crystalline as deposited, and (2) two-step ZIF-8 MLD, in which crystallization occurs during a postdeposition treatment with additional linker vapor. The latter approach resulted in a reduced deposition time and an improved MOF quality, i.e., increased crystallinity and probe molecule uptake, although the smoothness and thickness control were partially lost. Both approaches were developed in a modified atomic layer deposition reactor to ensure cleanroom compatibility.

The critical balance between intended and adverse effects in allogeneic hematopoietic stem cell transplantation (alloHSCT) depends on the fate of individual donor T-cells. To this end, we tracked αβT-cell clonotypes during stem cell mobilization treatment with granulocyte-colony stimulating factor (G-CSF) in healthy donors and for six months during immune reconstitution after transfer to transplant recipients. More than 250 αβT-cell clonotypes were tracked from donor to recipient. These clonotypes consisted almost exclusively of CD8+ effector memory T cells (CD8TEM), which exhibited a different transcriptional signature with enhanced effector and cytotoxic functions compared to other CD8TEM. Importantly, these distinct and persisting clonotypes could already be delineated in the donor. We confirmed these phenotypes on the protein level and their potential for selection from the graft. Thus, we identified a transcriptional signature associated with persistence and expansion of donor T-cell clonotypes after alloHSCT that may be exploited for personalized graft manipulation strategies in future studies.

Superparamagnetic iron oxide nanoparticles (SPION) are being investigated for many purposes, e.g., for the amplification of ionizing radiation and for the targeted application of therapeutics. Therefore, we investigated SPIONs coated with (3-Aminopropyle)-Triethoxysilane (SPION-APTES) for their influence on different head and neck squamous cell carcinoma (HNSCC) cell lines, as well as for their suitability as a radiosensitizer. We used 24-well microscopy and immunofluorescence microscopy for cell observation, growth curves to determine cytostatic effects, and colony formation assays to determine cytotoxicity. We found that the APTES-SPIONs were very well taken up by the HNSCC cells. They generally have a low cytotoxic effect, showing no significant difference in clonogenic survival between the control group and cells treated with 20 µg Fe/mL (p > 0.25) for all cell lines. They have a cytostatic effect on some cell lines cells (e.g., Cal33) that is visible across different radiation doses (1, 2, 8 Gy, p = 0.05). In Cal33, e.g., SPION-APTES raised the doubling time at 2 Gy from 24.53 h to 41.64 h. Importantly, these findings vary notably between the cell lines. However, they do not significantly alter the radiation effect: only one out of eight cell lines treated with SPION-APTES showed a significantly reduced clonogenic survival after ionizing radiation with 2 Gy, and only two showed significantly reduced doubling times. Thus, although the APTES-SPIONs do not qualify as a radiosensitizer, we were still able to vividly demonstrate and analyze the effect that the APTES-SPIONs have on various cell lines as a contribution to further functionalization.

This exploratory study provides an overview of circular economy related intellectual property (CEIP) challenges that actors in circular economy innovation ecosystems (CEIEs) face. Therefore, findings from a literature review are combined with empirical evidence from semi-structured interviews with different actors in CEIEs, including original equipment manufacturers as well as end-of-life solution providers of different company sizes and manufacturing industries. The key contributions of this paper to the literature are: (i) a preliminary framework to study IP challenges in CEIEs, (ii) the CEIP challenges framework which extends findings from the literature by categorising CEIP challenges mentioned in the conducted interviews and (iii) a visual representation of CEIP challenges in the aerospace industry.

In the field of international relations and transnational cooperation, normative attitudes and values of foreign cultural and educational policy (FCEP) sometimes precede practice in Germany and within internationally active organisations. However, in order to be able to act credibly to the outside world, cultural mediator organisations in particular should also implement structures of diversity and inclusion. In this study, the concepts of diver-sity and inclusion will be explained, a selection of existing measures will be presented and concrete steps for implementing diversity and inclusion will be shown. Half of all mental health disorders start occurring by the age of 14, with depression being the fourth most common disorder among adolescents worldwide. The prevalence of depression among German adolescents has nearly doubled in recent years. When it comes to mental health information sources, the internet has become a common medium for adolescents. Hence, to raise awareness of depression among this group, their specific expectations for online information and services must be met. Due to a lack of mixed-methods studies, this study therefore compares adolescents' expectations of online information and support services about depression (Study I), and information provided on the internet (Study II). Based on a literature review, qualitative interviews with adolescents were conducted (N=34). Moreover, the multi-platform online communication of nine German non-profit organizations (NPOs) that aim to improve information and care for people suffering from depression was analyzed using quantitative content analysis (N=1,435). Comparing the information gained from both studies, results indicate that expectations for fact-based communication were met by the NPOs frequently providing information on depression and requested experience reports were often communicated. However, discrepancies are apparent in the use of communication channels and videos, and the particular importance of personalization is evident.
