Background: Peptides and peptide-based therapeutics are biomolecules that demarcate a significant chemical space to bridge small molecules with biological therapeutics, such as antibodies, recombinant proteins, and protein domains. Introduction: Cyclooligopeptides and depsipeptides, particularly cyanobacteria-derived thiazoline-based polypeptides (CTBCs), exhibit a wide array of pharmacological activities due to their unique structural features and interesting bioactions, which furnish them as promising leads for drug discovery. Methods: In the present study, we comprehensively review the natural sources, distinguishing chemistries, and pertinent bioprofiles of CTBCs. We analyze their structural peculiarities counting the mode of actions for biological portrayals which render CTBCs as indispensable sources for emergence of prospective peptide-based therapeutics. In this milieu, metal organic frameworks and their biomedical applications are also briefly discussed. To boot, the challenges, approaches, and clinical status of peptide-based therapeutics are conferred. Results: Based on these analyses, CTBCs can be appraised as ideal drug targets that have always remained a challenge for traditional small molecules, like those involved in protein- protein interactions or to be developed as potential cancer-targeting nanomaterials. Cyclization-induced reduced conformational freedom of these cyclooligopeptides contribute to improved metabolic stability and binding affinity to their molecular targets. Clinical success of several cyclic peptides provokes the large library-screening and synthesis of natural product-like cyclic peptides to address the unmet medical needs. Conclusion: CTBCs can be considered as the most promising lead compounds for drug discovery. Adopting the amalgamation of advanced biological and biopharmaceutical strategies might endure these cyclopeptides to be prospective biomolecules for futuristic therapeutic applications in the coming times.

Reaction of thiazoline fused 2-pyridones with alkyl halides in the presence of cesium carbonate opens the thiazoline ring via S-alkylation and generates N-alkenyl functionalized 2-pyridones. In the reaction with propargyl bromide, the thiazoline ring opens and subsequently closes via a [2 + 2] cycloaddition between an in situ generated allene and the α,β-unsaturated methyl ester. This method enabled the synthesis of a variety of cyclobutane fused thiazolino-2-pyridones, of which a few analogues inhibit amyloid β1–40 fibril formation. Furthermore, other analogues were able to bind mature α-synuclein and amyloid β1−40 fibrils. Several thiazoline fused 2-pyridones with biological activity tolerate this transformation, which in addition provides an exocyclic alkene as a potential handle for tuning bioactivity.

Triple-negative breast cancer (TNBC) is an aggressive cancer insensitive to hormonal and human epidermal growth factor receptor 2 (HER2)-targeted therapies and has a poor prognosis. Therefore, there is a need for the development of convenient anticancer strategies for the management of TNBC. In this paper, we evaluate the antitumoral potential of a platinum(II) complex coordinated with the ligand 2-(3,5-diphenylpyrazol-1-yl)-2-thiazoline (DPhPzTn), hereafter PtDPhPzTn, against the TNBC cell line MDA-MB-231, and compared its effect with both cisplatin and its less lipophilic counterpart PtPzTn, the latter containing the ligand 2-(pyrazol-1-yl)-2-thiazoline (PzTn). Then, the putative potentiating actions of melatonin, a naturally occurring antioxidant with renowned antitumor properties, on the tumor-killing ability of PtDPhPzTn were also checked in TNBC cells. Our results show that PtDPhPzTn presented enhanced cytotoxicity compared to both the classical drug cisplatin and PtPzTn. In addition, PtDPhPzTn was able to induce apoptosis, being more selective for MDA-MB-231 cells when compared to non-tumor breast epithelial MCF10A cells. Likewise, PtDPhPzTn produced moderate S phase arrest and greatly impaired the migration ability of MDA-MB-231 cells. Most importantly, the co-stimulation of TNBC cells with PtDPhPzTn and melatonin substantially enhanced apoptosis and markedly improved the anti-migratory action compared to PtDPhPzTn alone. Altogether, our findings provide evidence that PtDPhPzTn and melatonin could be potentially applied to breast cancer treatment as powerful synergistic agents.

The shift in consumer landscape towards vegan, vegetarian and flexitarian diets has created an unprecedented challenge in creating meat aroma from plant-based alternatives. The search for potential vegan solutions has thus led to a renewed interest in authentic meat flavour profiles. To gain a better understanding of the qualitative odour differences between boiled beef and boiled chicken, aroma extracts were isolated using Likens-Nickerson simultaneous distillation-extraction (SDE), selected expressly because the in-situ heating of the sample facilitates the capture of aroma intermediates during the cooking process, thereby mimicking the cooking of meat in stocks and stews. The extracts were then analysed by Gas Chromatography-Mass Spectrometry (GC-MS) and GC-Olfactometry (GC-O). Most of the volatiles identified in this study were sulfur-containing compounds, such as sulfides, thiols, mercaptoaldehydes and mercaptoketones, which are derived from the Maillard reaction. Meanwhile, lipid oxidation results in the formation of unsaturated aldehydes, such as alkenals and alkadienals. Families of thiazoles and 3-thiazolines were found in the extracts. Two novel 3-thiazolines (5-ethyl-2,4-dimethyl-3-thiazoline and 2-ethyl-4,5-dimethyl-3-thiazoline) which may also contribute to the meaty aroma were identified in this work and synthesised from their respective aldehyde and mercaptoketone precursors.

The siderophore piscibactin is a key virulence factor involved in the iron uptake of pathogenic bacteria Photobacterium damselae subsp. piscicida and Vibrio anguillarum, responsible for the fish diseases photobacterioisis (pasteurellosis) and vibriosis, respectively. A convergent total synthesis of its Ga3+ complex using l-/d-cysteine as chiral agents and Meldrum's acid is described. A Staudinger reduction/Aza-Wittig process in the synthesis of the acid-sensitive β-hydroxy-2,4-disubstituted thiazoline moiety and the convenient protecting groups was a key step in this synthesis.

Chagas disease, a century-old disease that mainly affects the impoverished population in Latin America, causes high morbidity and mortality in endemic countries. The available drugs, benznidazole (Bz) and nifurtimox, have limited effectiveness and intense side effects. Drug repurposing, and the development of new chemical entities with potent activity against Trypanosoma cruzi, are a potential source of therapeutic options. The present study describes the biological activity of two new series of pyrazole-thiazoline derivatives, based on optimization of a hit system 5-aminopyrazole-imidazoline previously identified, using structure–activity relationship exploration, and computational and phenotype-based strategies. Promising candidates, 2c, 2e, and 2i derivatives, showed good oral bioavailability and ADMET properties, and low cytotoxicity (CC50 > 100 µM) besides potent activity against trypomastigotes (0.4–2.1 µM) compared to Bz (19.6 ± 2.3 µM). Among them, 2c also stands out, with greater potency against intracellular amastigotes (pIC50 = 5.85). The selected pyrazole-thiazoline derivatives showed good permeability and effectiveness in the 3D spheroids system, but did not sustain parasite clearance in a washout assay. The compounds’ mechanism of action is still unknown, since the treatment neither increased reactive oxygen species, nor reduced cysteine protease activity. This new scaffold will be targeted to optimize in order to enhance its biological activity to identify new drug candidates for Chagas disease therapy.

The structural and functional characterization of natural products is vastly outpaced by the bioinformatic identification of biosynthetic gene clusters (BGCs) that encode such molecules. Uniting our knowledge of bioinformatics and enzymology to predict and synthetically access natural products is an effective platform for investigating cryptic/silent BGCs. We report the identification, biosynthesis, and total synthesis of a minimalistic class of ribosomally synthesized and post-translationally modified peptides (RiPPs) with the responsible BGCs encoding a subset of enzymes known from thiopeptide biosynthesis. On the basis of the BGC content, these RiPPs were predicted to undergo enzymatic dehydration of serine followed by [4+2]-cycloaddition to produce a trisubstituted, pyridine-based macrocycle. These RiPPs, termed "pyritides", thus contain the same six-membered, nitrogenous heterocycle that defines the thiopeptide RiPP class but lack the ubiquitous thiazole/thiazoline heterocycles, suggesting that thiopeptides should be reclassified as a more elaborate subclass of the pyritides. One pyritide product was obtained using an 11-step synthesis, and the structure verified by an orthogonal chemoenzymatic route using the precursor peptide and cognate pyridine synthase. This work exemplifies complementary bioinformatics, enzymology, and synthesis to characterize a minimalistic yet structurally intriguing scaffold that, unlike most thiopeptides, lacks growth-suppressive activity toward Gram-positive bacteria.

We report a single‐molecule mechanistic investigation into 2‐cyanobenzothiazole (CBT) chemistry within a protein nanoreactor. When simple thiols reacted reversibly with CBT, the thioimidate monoadduct was approximately 80‐fold longer‐lived than the tetrahedral bisadduct, with important implications for the design of molecular walkers. Irreversible condensation between CBT derivatives and N‐terminal cysteine residues has been established as a biocompatible reaction for site‐selective biomolecular labeling and imaging. During the reaction between CBT and aminothiols, we resolved two transient intermediates, the thioimidate and the cyclic precursor of the thiazoline product, and determined the rate constants associated with the stepwise condensation, thereby providing critical information for a variety of applications, including the covalent inhibition of protein targets and dynamic combinatorial chemistry., Better off alone: A nanoreactor approach was used to investigate the chemistry of2‐cyanobenzothiazole (CBT), an important bioorthogonal reagent, at the single‐molecule level. Cyclic peptides are excellent drug candidates, placing macrocyclization reactions at the apex of drug development. PatG and related dual-action proteases from cyanobactin biosynthesis are responsible for cleaving off the C-terminal recognition sequence and macrocyclizing the substrate to provide cyclic peptides. This reaction has found use in the enzymatic synthesis of diverse macrocycles. However, these enzymes function best on substrates that terminate with the non-proteinogenic thiazole/thiazoline residue, complicating synthetic strategies. Here, we biochemically characterize a new class of PatG-like macrocyclases that natively use proline, obviating the necessity of additional chemical or biochemical steps. We experimentally define the biochemical steps involved in synthesizing the widespread prenylagaramide-like natural products, including macrocyclization and prenylation. Using saturation mutagenesis, we show that macrocyclase PagG and prenyltransferase PagF are highly promiscuous, producing a library of more than 100 cyclic peptides and their prenylated derivatives in vitro. By comparing our results to known cyanobactin macrocyclase enzymes, we catalog a series of enzymes that collectively should synthesize most small macrocycles. Collectively, these data reveal that, by selecting the right cyanobactin macrocyclase, a large array of enzymatically synthesized macrocycles are accessible.

A photocatalyst- and additive-free, visible-light-mediated chemoselective domino protocol was devised to access fully substituted thiazoline derivatives from β-ketothioamides and α-diazo 1,3-diketones at moderate temperature in open air. The reaction proceeds through in situ generation of electrophilic carbenes from α-diazo 1,3-diketones by a low-energy blue LED (448 nm), which undergoes selective coupling with nucleophilic β-ketothioamides to give thiazolines by successive formation of C-S and C-N bonds in one stretch. Notably, the benign and clean conditions, operational simplicity, sustainability, 100 % carbon economy, high yields, and wide functional-group tolerance are further attributes of the strategy. A mechanistic rationale for this cascade reaction sequence is well supported by control experiments.

An operationally simple and efficient one-pot protocol for the synthesis of highly functionalized thiazolidin-4-ones and thiazolines has been devised via Rh(OAc)2-catalyzed annulative coupling of β-ketothioamides with diazo compounds under mild reaction conditions for the first time. This double functionalization of diazo compounds proceeds via selective S-alkylation followed by intramolecular N-cyclization enabling the formation of C-S and C-N bonds at moderate temperature. Notably, the products possess Z-stereochemistry with regard to the exocyclic C═C double bond at the 2-position of the ring. Further, the synthetic utility of the strategy has been revealed to access 2,3-dihydrobenzo[d]thiazoles. Remarkably, atom economy and tolerance of a wide range of functional groups are added characteristics to this strategy.

The emergence of antibiotic resistance among microbial pathogens is a serious public health problem, resulting in a constant need of discovering new antibiotics. For this purpose, we synthetized and determined the antimicrobial activity of copper (Cu), cobalt (Co), and tin (Sn) complexes coordinated with scorpionate ligand in a tridentate mode against microorganisms of clinical importance. New complexes with the sodium hydrotris(2-mercaptothiazolyl)borate ligand were characterized by physicochemical and spectroscopic techniques (UV-vis, IR, 1H and 13C NMR). Moreover, their antimicrobial activity was evaluated using microdilution broth method. Cytotoxicity was determined in Vero cells and the selectivity index (SI) was calculated. The coordination with metals increased the ligand antimicrobial activity, which showed no activity when evaluated alone (MIC>1,080 µM). Cu-complex (1) had activity against Candida albicans and C. krusei, with MICs of 130 and 66 µM, respectively. The compounds had low toxicity on Vero cells and chemo-informatics assays corroborated a low systemic toxic effect. The SI revealed that the compound 1 was 16 times more selective for C. krusei and 8 times more selective for C. albicans in relation to Vero cells. The Cu-complex stands out as a potential prototype for the development of new antifungal agents.

A series of novel sulfur-containing cycliclipopeptides named thioamycolamides A-E, with thiazoline, thioether rings, and fatty acid moieties, were identified from the culture broth of the rare actinomycete Amycolatopsis sp. 26-4. The planar structural elucidation was accomplished by HRMS and 1D/2D NMR spectroscopic data analyses. The absolute configurations were unambiguously determined by Marfey's method, CD spectroscopy, and synthesis of partial structures. Moreover, their growth inhibitory activities against human tumor cell lines were investigated.

Lewis base/Brønsted acid cocatalysis for electrophilic thiocyanation of olefins is reported. Using a combination of triphenylphosphine selenide and diphenyl phosphate as a catalyst, a wide range of unsaturated amides and thioamides underwent thiocyanation to furnish thiocyanated thiazoline and oxazoline derivatives in high yields (up to 97%).

A series of xanthene-based thiazoles was synthesized and characterized by different scpectroscopic methods, i.e. Proton nuclear magnetic resonance (H-1 NMR), carbon nuclear magnetic resonance (C-13 NMR), infrared spectroscopy, carbon hydrogen nitrogen analysis, and X-ray crystallography. The inhibition potencies of 18 newly synthesized thiazole derivatives were investigated on the activities of acetylcholinesterase (AChE), butyrylcholinesterase (BChE), alpha-amylase (alpha-Amy), and alpha-glycosidase (alpha-Gly) enzymes in accordance with their antidiabetic and anticholinesterase ability. The synthesized compounds have the highest inhibition potential against the enzymes at low nanomolar concentrations. Among the 18 newly synthesized molecules, 3b and 3p were superior to the known commercial inhibitors of the enzymes and have a much more effective inhibitory potential, with IC50: 2.37 and 1.07 nM for AChE, 0.98 and 0.59 nM for BChE, 56.47 and 61.34 nM for alpha-Gly, and 152.48 and 124.84 nM for alpha-Amy, respectively. Finally, the optimized 18 compounds were subjected to molecular docking to describe the interaction between thiazole derivatives and AChE, BChE, alpha-Amy, and alpha-Gly enzymes in which important interactions were monitored with amino acid residues of each target enzyme. Iron overload disorder and diseases where iron mismanagement plays a crucial role require orally available iron chelators with favourable pharmacokinetic and toxicity profile. Desferrithiocin (DFT), a tridentate and orally available iron chelator has a favourable pharmacokinetic profile but its use has been clinically restricted due to its nephrotoxic potential. The chemical architecture of the DFT has been naturally well optimized for better iron chelation and iron clearance from human biological system. Equally they are also responsible for its toxicity. Hence, subsequent research has been devoted to develop a non-nephrotoxic analogue of DFT without losing its iron clearance ability.:The review has been designed to classify the compounds reported till date and to discuss the structure activity relationship with reference to modifications attempted at different positions over pyridine and thiazoline ring of DFT. Compounds are clustered under two major classes: (i) Pyridine analogues and (ii) phenyl analogue and further each class has been further subdivided based on the presence or absence and the number of hydroxy functional groups present over pyridine or phenyl ring of the DFT analogues. Finally a summary and few insights into the development of newer analogues are provided.

Ferrocene and 2-thiazolines, in combination or separately, are important structures to design new bi-, tri-, or polydentate ligands with application in organometallic catalysis. Recently, our research group developed a new methodology for the synthesis of 2-ferrocenyl-2-thiazolines. These compounds were used to get a new bidentate ligand [N,S], in racemic and enantiopure version, in excellent yields. These ligands were successfully used in metal-catalyzed C-C coupling reactions, ethylene oligomerization, and asymmetric allylic alkylation. In order to extend the use in synthesis and catalysis of 2-ferrocenyl-2-thiazolines, we were interested to build a new Schiff base type tridentate ligand [N,N,O] to use it in the transfer hydrogenation reaction catalyzed by ruthenium. This reaction allows the preparation of secondary alcohols, key molecules in the synthesis of compounds used in pharmaceutical, agrochemical, and food industries. The strategy to accomplish the synthesis of a [N,N,O] ligand is based on the ortho-metallation of ferrocene directed by 2-thiazoline. The ortho-lithiation of 2-ferrocenyl-2-thiazoline, followed by the addition of DMF as an electrophile, allows the introduction of carbaldehyde in the ferrocene core. Afterward, the condensation of this aldehyde with 2-aminophenol, provides a new tridentate ligand in 77% global yield, after two reaction steps. The scope of this method was studied in the preparation of, the enantiomeric version of this ligand. The ortho-lithiation occurs in a diastereoselective way thanks to the influence of the enantiopure thiazoline moiety. The enantiopure ligand combines the central and planar chirality. This ligand was obtained in a global yield of 65%. The coordination study of the tridentate ligand was done using a ruthenium(II) source, to obtain a new ruthenium complex. After isolation of this complex and characterization by UV-vis, IR, and EPR, we found the presence of a Ru(III) species in equilibrium with its reduced counterpart of Ru(II). This ruthenium complex was tested as an efficient catalytic precursor for the reduction of acetophenone in transfer hydrogenation. After getting the optimal conditions for this reaction, we revealed the reaction scope submitting 18 aromatics and aliphatic ketones. The enantiopure ruthenium complex, obtained from the enantiopure ligand, was evaluated in the reduction of acetophenone in transfer hydrogenation as well. In the optimal conditions, both complexes were found to be equally reactive. Nevertheless, the ligand and the enantiopure complex showed low selectivities. With these results, we proved the importance of 2-ferrocenyl-2-thiazolines to the design of news-efficient ligands in catalysis.; Le ferrocène et les 2-thiazolines, en combinaison ou séparément, sont des structures de base importantes pour la conception de nouveaux ligands bi-, tri- ou poly-dentées performants en catalyse organométallique. Récemment notre groupe de recherche a développé une nouvelle méthodologie pour la synthèse des 2-ferrocényl-2-thiazolines. Ces composés ont été utilisés pour la synthèse d'un nouveau ligand bidenté [N,S], en version racémique et énantiopure, avec d'excellents rendements. Ils ont été utilisés avec succès dans des réactions métallo-catalysées de couplage C-C, d'oligomérisation d'éthylène et d'alkylation allylique asymétrique. Afin d'étendre l'utilisation en synthèse et catalyse des 2-ferrocényl-2-thiazolines, nous nous sommes intéressés dans ce projet à la construction d'un nouveau ligand tridenté [N,N,O] de type base de Schiff, pour une application dans des réactions d'hydrogénation par transfert catalysées par le ruthénium. Cette réaction permet obtenir des alcools secondaires qui sont des grands intermédiaires clés de synthèse de nombreuses cibles pour les industries pharmaceutiques, agrochimiques et alimentaires. La stratégie de synthèse du ligand [N,N,O] est basée sur l'ortho-métallation dirigée de la 2-thiazoline. L'ortho lithiation du 2-ferrocényl-2-thiazoline suivie de l'addition du DMF comme réactif électrophile, permet d'introduire la fonction aldéhyde sur le noyau ferrocénique. La condensation de cet aldéhyde avec le 2-aminophénol conduit à un nouveau ligand tridenté avec un rendement global de 77% sur ces deux étapes de synthèse. Nous avons étendu notre méthode de synthèse à la préparation la version énantiopure de cette ligand L'ortholithiation se produit de manière diastérosélective grâce à l'influence de la thiazoline énantiopure. Le ligand énantiopur combine la chiralité centrale et planaire. Ceci est obtenu avec un rendement global de 65%. La capacité de coordination du ligand tridenté au ruthénium(II) a été mise en évidence et le complexe résultant a été isolé et analysé. Une caractérisation fine, effectuée par UV-Vis, IR et EPR, a montré la présence d'une espèce de Ru(III) en équilibre avec sa contrepartie réduite de Ru(II). Ce complex s'est révelé être un précurseur catalytique efficace pour la réduction de l'acétophénone par hydrogénation par transfert. Après optimisation des conditions de réactions, son utilisation a été étendue à un panel de 18 cétones aromatiques et aliphatiques. Le complexe de ruthénium énantiopure, obtenu à partir du ligand énantiopure, a aussi été évalué dans la réduction de l'acétophénone par hydrogénation par transfert. Dans les conditions optimisées, les deux complexes se sont avérés être également réactifs. Cependant, le ligand et le complexe énantiopure ont montré de faibles sélectivités. Les résultats obtenus montrent l'importance de la structure des 2-ferrocényl-2-thiazolines pour la conception de nouveaux ligands efficaces en catalyse organométallique.

Oxo- and thio-substituted quinazolines are promising biologically active objects. A special place in a number of quinazolines is occupied by its condensed analogues, namely thiazoloquinazolines, which exhibit antioxidant, antimicrobial and antitumor activity. An effective method for the synthesis of condensed quinazoline derivatives is electrophilic intramolecular cyclization of its unsaturated derivatives. The usage of pharmacoformic chalcogen-containing electrophiles in these reactions has been insufficiently studied. Organochalcogen-containing heterocyclic compounds are the promising biologically active substances with different pharmacological action. Therefore, the study of reactions of electrophilic intramolecular cyclization of N(3)-alkenyl derivatives of 2-thioxo-2,3-dihydroquinazolin-4(1H)-one under the action of chalcogen tetrahalides is an urgent task. N (3)-alkenyl derivatives of 2,3-dihydroquinazolin-4(1H)-one were used as the object of study. It was found that the interaction of tellurium tetrabromide with N(3)-alkenyl-2-thioxoquinazolin-4-ones in glacial acetic acid leads to linear hydrobromides of 2-[(tribromotelluro) methyl]-2,3-dihydro-5H-[1,3]thiazolo[2,3-b]quinazolin-5-one. In the reaction with N(3)-methallyl-2-thioxo-2,3-dihydroquinazolin-4(1H)-one, individual tellurium tetrachloride was used, which was obtained by passing chlorine over the tellurium. It is proved that halotellurium-induced cyclization of N-methallyl quinazolone with tellurium tetrachloride occurs regioselectively with the formation of 2-methyl-2-[(trichlorotelluro)methyl]-2,3-dihydro-5H- [1,3]thiazolo[2,3-b]quinazoline-5-one hydrochloride of linear structure. In order to study the influence of the nature of chalcogen in the electrophilic reagent on the regiochemistry of cyclization in the reaction with N(3)-methallyl-2-thioxo-2,3-dihydroquinazolin-4(1H)-one, obtained in situ selenium tetrachloride was used. The selenochlorination reaction of N(3)-methallyl-2-thioxo-2,3-dihydroquinazolin-4(1H)-one in chloroform gave a linear hydrochloride of 2-methyl-2-[(trichloroseleno)methyl]-2,3-dihydro-5H-[1,3]thiazolo[2,3-b]quinazolin-5-one. Thus, the regiochemistry of electrophilic intramolecular cyclization of N(3)-alkenyl-2-thioxo-2,3-dihydroquinazolin-4(1H)-ones by chalcogentetrahalides was investigated and it was proved that the annulationof the thiazoline cycle occurs with the formation of bioperspective bundles of tricyclic heterocycles of linear constitution independently from nature and origin of electrophilic reagent.

In nature, azolines produced by YcaO cyclodehydratases during the biosynthesis of ribosomally synthesized and post-translationally modified peptides (RiPPs) are generally limited to thiazoline, oxazoline, and methyloxazoline, which are derived from the proteinogenic Cys, Ser, and Thr residues, respectively. To investigate whether YcaO cyclodehydratases precisely recognize the common structural characteristics and chirality of the modifiable residues, the "reprogrammed FIT-PatD system" has been established by combining a YcaO cyclodehydratase (PatD) with genetic code reprogramming powered by the flexible in vitro translation (FIT) system, in which precursor peptides bearing non-proteinogenic Cys/Ser/Thr analogues could be expressed through a reprogrammed genetic code and subsequently cyclodehydrated by PatD. The study has revealed remarkable stereo-, chemo-, and regioversatility for modifiable residues in PatD-catalyzed cyclodehydration, expanding the repertoire of backbone heterocycles in RiPPs to exotic azoline analogues.

This review provides a generalized and systematized analysis of literature data from the last 15 years regarding the methods of synthesis and properties of bi- and tricyclic assemblies constructed from 1,3-thiazolidine/thiazoline rings that are linked by an exocyclic С=С double bond. Examples of such assemblies which are of interest as biologically active compounds, sensitizing dyes for solar cells, and semiconductors are considered in details. The literature data have been arranged according to the types of chemical structures.

На основі несиметричних тіосечовин синте- зовані нові похідні N-(R`-феніл)-3-(4-метил-1-піперазиніл)-1,3- тіазол-2(3H)-іміну за реакцією Ганча. Структуру цільових компонентів підтверджено елементим аналізом та ЯМР спектроскопією. Досліджено антимікробну активність похід- них 1,3-тіазоліну з N-метилпіперазиновим фрагментом щодо сульфатвідновлювальних бактерій штаму Desulfovibrio sp. М 4.1. Встановлено, що сполуки з галогензаміщеним та незаміще- ним фенільним фрагментом в 4-му положенні тіазолінового циклу виявляють виразну протимікробну активність. New derivatives of N-(R-phenyl)-3-(4-methyl- 1-piperazinyl)-1,3-thiazole-2(3H)- imine with the medium to high yields were synthesized by the Hantzsch reaction in the ethanol medium. The structure of target compounds was confirmed by elemental analysis and NMR spectroscopy. The antimicrobial activity of 1,3-thiazoline derivatives with the N-methylpiperazine moiety against sulfate-reducing bacteria of Desulfovibrio sp. М.4.1 strain was studied. It was found that substances containing the halo- and unsubstituted phenyl fragment in the position 4 of the thiazoline cycle showed the potent antimicrobial activity.

It is known that the quinazoline fragment is a structural element of many natural alkaloids. A promising method for the synthesis of condensed derivatives of quinazoline is the method of electrophilic intramolecular heterocyclization. The purpose of this work is to investigate the regioselectivity of electrophilic heterocyclization of S-allyl derivatives of quinazolone with aryltellurium trichlorides. N-(3)-phenyl-2-S-alkenyl derivatives of quinazolone with a non-terminal allyl substituent are used as substrates for aryltellurohalogenation. The choice of such objects is motivated by the presence of one additional nucleophilic center for cyclization – the endocyclic nitrogen atom in the first position of the pyrimidine cycle. Interaction of allyl(methallyl) substituted thioquinazolones with p methoxyphenyltellurium trichloride was carried out in acetic acid or tetrahydrofuran (THF) at room temperature for 8 hours stirring the reaction mixture. It has been established that irrespective of the ratio of reagents, the non-terminal alkenyl thioether, under the action of p-methoxyphenyltellurium trichloride, undergoes annulation of the thiazoline ring to form tricyclic condensed systems of the angular structure. It should be noted that the molecular complex of p-methoxyphenyltellurium trichloride with Sulfur atom of thiazolinoquinazolone is formed. The formation of such a complex was not observed under aryltellurohalogenation of N-alkenyl 2-thioxoquinazolones, where, obviously, there is intramolecular coordination of the Sulfur atom to the Tellurium atom. Instead, the use of the terminal-substituted dimethylallyl thioether in the reaction with p methoxyphenyltellurium trichloride in similar conditions changes the direction of the reaction and leads to the formation of the molecular complex. The impossibility of the cyclization is due to the steric shielding of the reaction center with respect to the tellurim-containing electrophile. Thus, thioethers of quinazolone with a terminal non-substituted allyl substituent under the action of p-methoxyphenyltellurium trichloride undergo cyclization, while the introduction of two methyl groups to the terminal carbon atom of the allyl fragment changes the direction of the reaction and leads to the formation of the molecular complex.

A thiazoline and pyrene containing sensor 1 was synthesized via one-pot reaction and utilized as a highly selective and sensitive fluorescence “turn-on” sensor for Cu2+ detection, via the fluorescence enhancement of pyrene monomer emission. The 2:1 stoichiometry of 1 and Cu2+ was calculated from Job's plots based on fluorescent titrations, and the complexation of 1 with Cu2+ was also supported by mass spectra, Fourier transform infrared spectra, proton NMR spectra and density functional theory analysis. The fluorescence intensity of 1 at 389 nm and 410 nm increased significantly upon the addition of Cu2+. Limit of detection and association constant value of 1–Cu2+ were calculated using standard deviations and linear fittings, respectively. Results showed that 1 was active for Cu2+ detection in the wide pH range of 2.0–11.0.

Novel findings during the course of the total synthesis of sulfur-containing natural products are described. These natural products include sporidesmin A, dehydrogliotoxin, tantazole B, and leinamycin.

Iron(III) phthalocyaninate decorated with crown ether substituents, [(15C5)4PcFe]Cl, efficiently catalyzed the insertion of carbene derived from ethyl diazoacetate to six amines functionalized with thiazole, thiazoline and thiadiazole heterocycles. The reactions were carried out under practical conditions using EDA:amine stoechiometric ratio with 0.05 mol% catalyst loading. Turnover numbers up to 3360 have been achieved. The aminoacid derivatives bearing heterocyclic moieties were obtained under catalytic conditions for the first time with 36–69% yields in the case of single N–H insertion products and up to 77% in the case of double N–H insertion products.

A new series of achiral pyrimidine derivatives based on nifedipine-like structure was designed and synthesized. These pyrimidyl derivatives contained hydrazine, hydrazones, acetohydrazide, differently substituted benzylidene functionalities, benzosulfohydrazine, various heterocycles such as pyrazole, pyrazolidinedione, thiazoline, and thiazolidinone rings, and fused ring systems such as triazolopyrimidine and pyrimidotriazine rings. Compounds 5a, 5b, 11b, 8b, 9b–d, and 15b showed a decrease in mean arterial rabbit blood pressure (MABP) ranging from 51.4 to 78.2 mmHg in rabbits in comparison with nifedipine-treated rabbits. Among these derivatives, compounds 5a, 5b, 9b, and 9c were found to exhibit calcium channel blockade activity on preparations of rabbit aortae. They exhibited relaxation in the range of 89.2% to 74.4% in comparison to nifedipine (57.6%) as well as a decrease in heart rate. Histopathological effect of compounds 5a,b on the expression of endothelial nitric oxide synthase (eNOS) was also examined on rat aorta. An intense expression of eNOS immune staining in aortic endothelium was seen for compound 5b indicating that it lowered blood pressure via activation of eNOS expression in aorta.

We have engineered an intein which spontaneously and reversibly forms a thiazoline ring at the native N-terminal Lys-Cys splice junction. We identified conditions to stablize the thiazoline ring and provided the first crystallographic evidence, at 1.54 A resolution, for its existence at an intein active site. The finding bolsters evidence for a tetrahedral oxythiazolidine splicing intermediate. In addition, the pivotal mutation maps to a highly conserved B-block threonine, which is now seen to play a causative role not only in ground-state destabilization of the scissile N-terminal peptide bond, but also in steering the tetrahedral intermediate toward thioester formation, giving new insight into the splicing mechanism. We demonstrated the stability of the thiazoline ring at neutral pH as well as sensitivity to hydrolytic ring opening under acidic conditions. A pH cycling strategy to control N-terminal cleavage is proposed, which may be of interest for biotechnological applications requiring a splicing activity switch, such as for protein recovery in bioprocessing.

Aerucyclamide A (ACA) is an emerging cyanopeptide toxin produced by cyanobacteria, and its transformation pathway has rarely been reported. In the present study, ACA was purified from cyanobacterial extracts, and photodegradation processes were investigated in dissolved organic matter (DOM) solutions. Under simulated solar irradiation, the photodegradation of ACA was dominated by •OH oxidation, accounting for ~72% of the indirect photodegradation. The bimolecular reaction rate constant of ACA with •OH was (6.4 ± 0.2) × 109 M − 1 s − 1. Our results indicated that the major reactive sites of ACA toward •OH are thiazoline and thiazole moieties. Product analysis via high-resolution mass spectrometry suggested that hydrogen abstraction and gradual hydroxylation are the main photodegradation pathways. The acute toxicity assessment indicate that the products generated in photolysis process did not show any measurable toxicity to Thamnocephalus platyurus. Photodegradation experiments with various DOM-phycocyanin mixtures demonstrated that the half-life of ACA is much longer than that of microcystin-LR.

Inhibition of aldose reductase (ALR2) by using small heterocyclic compounds provides a viable approach for the development of new antidiabetic agents. With our ongoing interest towards aldose reductase (ALR2) inhibition, we have synthesized and screened a series of thiazoline derivatives (5a–k, 6a–f, 7a–1 & 8a–j) to find a lead as a potential new antidiabetic agent. The bioactivity results showed the thiazoline-based compound 7b having a benzyl substituent and nitrophenyl substituent-bearing compound 8e were identified as the most potent molecules with IC50 values of 1.39 ± 2.21 μM and 1.52 ± 0.78 μM respectively compared with the reference sorbinil with an IC50 value of 3.14 ± 0.02 μM. Compound 7b with only 23.4% inhibition for ALR1 showed excellent selectivity for the targeted ALR2 to act as a potential lead for the development of new therapeutic agents for diabetic complications.

The cyanobacterial oligopeptides are recognized for being highly selective, efficacious and relatively safer compounds with diverse bioactivities. Azoline-based natural compounds consist of heterocycles which are reduced analogues of five-membered heterocyclic azoles. Among other varieties of azoline-based natural compounds, the heteropeptides bearing oxazoline or thiazoline heterocycles possess intrinsic structural properties with captivating pharmacological profiles, representing excellent templates for the design of novel therapeutics. The specificity of heteropeptides has been translated into prominent safety, tolerability, and efficacy profiles in humans. These peptidic congeners serve as ideal intermediary between small molecules and biopharmaceuticals based on their typically low production complexity compared to the protein-based biopharmaceuticals. The distinct bioproperties and unique structures render these heteropeptides one of the most promising lead compounds for drug discovery. The high degree of chemical diversity in cyanobacterial secondary metabolites may constitute a prolific source of new entities leading to the development of new pharmaceuticals. This review focuses on the azoline-based natural oligopeptides with emphasis on distinctive structural features, stereochemical aspects, biological activities, structure activity relationship, synthetic and biosynthetic aspects as well as mode of action of cyanobacteria-derived peptides.

International audience; A method for the synthesis of a new phosphine-free [N,N,O]-tridentate Schiff base ligand L1 using the 2-Ferrocenyl-2-thiazoline as scaffold was developed. The 1,2-disubstituted ferrocene-based ligand was assembled using as key strategy the directed ortho-metalation (DoM) in 2-ferrocenyl-2-thiazoline. L1 was successfully obtained in 83% of overall yield after two-step synthesis. The coordination ability of L1 towards Ru(II) was evidenced and the resulting complex was characterized by IR, UV-vis and EPR. Its catalytic performance was tested in transfer hydrogenation of a variety of substrates giving moderate to excellent conversions.

Bottromycins are ribosomally synthesized and post-translationally modified peptide natural product antibiotics that are effective against high-priority human pathogens such as methicillin-resistant Staphylococcus aureus. The total synthesis of bottromycins involves at least 17 steps, with a poor overall yield. Here, we report the characterization of the cytochrome P450 enzyme BotCYP from a bottromycin biosynthetic gene cluster. We determined the structure of a close BotCYP homolog and used our data to conduct the first large-scale survey of P450 enzymes associated with RiPP biosynthetic gene clusters. We demonstrate that BotCYP converts a C-terminal thiazoline to a thiazole via an oxidative decarboxylation reaction and provides stereochemical resolution for the pathway. Our data enable the two-pot in vitro production of the bottromycin core scaffold and may allow the rapid generation of bottromycin analogues for compound development.

Thioamycolamide A (1) is a biosynthetically unique cytotoxic cyclic microbial lipopeptide that bears a d-configured thiazoline, a thioether bridge, a fatty acid side chain, and a reduced C-terminus. It has gained attention for its unique structure, and very recently we reported the total synthesis of 1 via a biomimetic route. The NMR spectra of synthetic 1 agreed with those of natural 1. However, structural identity between peptidic natural and synthetic compounds is often difficult to confirm by comparison of NMR spectra because their NMR spectra vary depending on the conditions in the NMR tube, which often result in the structural misassignment of peptidic compounds. Especially, our total synthesis based on the putative biomimetic route potentially gives 1 as a diastereomixture at the final step. The problem is that the diastereomers of peptidic mid-sized molecules often exhibit similar properties (such as NMR spectra and bioactivities), and their separation procedures are often laborious. Herein we report the structural confirmation of synthetic 1 by the LC–MS-based chromatographic comparison with the use of our highly sensitive labeling reagent l-FDVDA; the highly sensitive-advanced Marfey’s method (HS-advanced Marfey’s method). This work demonstrated the utility of our highly sensitive labeling reagent for the structural determination of not only scarce natural products but also readily isomerizable synthetic compounds.

Thioamycolamide A is a biosynthetically unique cytotoxic cyclic microbial lipopeptide that bears a d-configured thiazoline, a thioether bridge, a fatty acid side chain, and a reduced C-terminus. Based on the biosynthetic insights, a concise total synthesis of thioamycolamide A was accomplished.

Malaria treatment is based on a reduced number of antimalarial drugs, and drug resistance has emerged, leading to the search for new antimalarial drugs incorporated into pharmaceutical formulations. In this study, 10-(4,5-dihydrothiazol-2-yl)thio)decan-1-ol) (thiazoline), a synthetic analog of 3-alkylpiridine marine alkaloid, and a potent antimalarial substance, was incorporated into O/W nanoemulsion. This formulation was prepared by a 23 factorial design. It was characterized by globule diameter, polydispersity index, zeta potential, encapsulation efficiency, in vitro thiazoline release at pH 2 and 6.86, and accelerated stability. In vitro and in vivo antimalarial activity was determined against P. falciparum and P. berghei, respectively. Thiazoline nanoemulsion showed 248.8 nm of globule diameter, 0.236 of polydispersity index, -38.5 mV of zeta potential, 96.92% encapsulation efficiency, and it was stable for 6 months. Thiazoline release profiles differed in acidic and neutral media, but in both cases, the nanoemulsion controlled and prolonged the thiazoline delivery. Thiazoline nanoemulsion exerted in vitro antimalarial activity against the parasite (IC50 = 1.32 µM), and it significantly reduced the in vivo parasitemia for 8 days without increasing the survival time of animals. Therefore, the thiazoline nanoemulsion represents a strategy to treat malaria combining an antimalarial candidate and a new nanocarrier.

In this research, a new series of thiazoline-iridium (III) complexes (4–7) derived from cysteine were prepared and fully characterized by conventional methods. The molecular structure of complex 5 was also determined by single-crystal X-ray diffraction. These complexes were evaluated as catalysts for hydrogen-borrowing reactions of amines with alcohols. In particular, complex 5 showed the best activity as catalyst. Various amines have been alkylated with alcohols affording moderate to good yield (33–99%). Moreover, the immobilized nanomaterials (M1,2) were fabricated by sonication process from the best catalyst 5 with the multi-walled carbon nanotubes (MWCNTs) and graphene oxide (GO), respectively, and characterized by X-ray diffraction (XRD), Fourier transform infrared (FT-IR) spectroscopy, field emission scanning electron microscopy (FE-SEM), energy dispersive X-ray (EDX) spectroscopy, and inductively coupled plasma-mass spectrometry (ICP-MS). The M1,2 nanomaterials were also tested as catalysts in model catalytic reaction for N-alkylation. The M1 nanomaterial showed significantly higher activity than the M2 nanomaterial. An efficient and general method for the synthesis of 2-substituted thiazolines and 5,6-dihydro-4H-1,3-thiazines is developed via microwave-assisted ring closure of ω-thioamidoalcohols promoted by ethyl polyphosphate (PPE). The cyclization reaction involves an SN2-type mechanism and features the advantages of very short reaction times, high yields and a predictable stereochemical outcome. The acyclic precursors are prepared in high overall yields by an improved diacylation–thionation–saponification sequence from commercially available ω-amino­alcohols. The whole process is metal-free and operationally simple.

The synthesis, reactivity, spectroscopic characterization and catalytic activities of a series of Pd(II) complexes bearing the chelating ligands 2-(4,5-dihydrothiazol-2-yl)aniline (2) and 2-(4,5-dihydro-1H-imidazol-2-yl)aniline (3) are reported. The reactions of 2 with [Pd(COD)Cl2] in 1:1 and 1:2 (ligand into metal and metal into ligand) molar ratios afforded the mononuclear complexes, PdCl2{κ2−N,S − 2−(4,5-dihydrothiazol-2-yl)aniline} (4), Pd{κ2−N,N’−2−(4,5-dihydrothiazol-2-yl)aniline}2 (5) and Pd{κ12−N,N’−κ22−N,S − 2−(4,5-dihydrothiazol-2-yl)aniline}2 (6), respectively, in good yields. The neutral mononuclear palladium complexes PdCl2{κ2−N,N’−(4,5-dihydro-1H-imidazol-2-yl)aniline} (7) and Pd{κ2−N,N’−2−(4,5-dihydro-1H-imidazol-2-yl)aniline}2 (8) were synthesized using [Pd(COD)Cl2] with appropriate molar ratios. The palladium complexes 4–8 were evaluated for their catalytic activities in base assisted cross coupling reactions between acid chlorides and aryl halide with boronic acids. The complex 4 was found to be an efficient catalyst for producing unsymmetrical ketones (TOF = 19.8 min−1) while complex 7 was effective for making biaryls (TOF = 19.8 min−1).

Aim: Many heterocyclic compounds derived from natural steroids exhibited broad activities, so this work focused on the investigations on a series of steroidal thiazoline conjugates as antiviral agents. Materials & methods: A series of steroid derivatives containing thiazoline heterocycles were designed and synthesized via a convenient condensation procedure. The compounds were screened for their potential antivirus activities against Enterovirus 71 (EV71) and Coxsackie Virus Type B (CVB3). Results and Conclusion: The in vitro bioassay indicated that compounds 5b, 5g and 5i exhibited excellent antiviral effects on EV71, and compounds 5b, 5e, 6c and 6g presented better antiviral activities against CVB3 compared with the controls ribavirin or pirodavir. These results indicate that these steroidal thiazoline conjugates might be feasible therapeutic candidates against EV71 infection, which might also be considered as promising compounds for optimization of potential antivirus agents.

The dipeptidyl peptidase-4 (DPP-4) inhibitor vildagliptin (VG) is used to treat type 2 diabetes. In rare cases, VG-induced liver injury has been reported. One case report suggested that immune responses were involved in the hepatotoxicity. However, the underlying mechanisms of VG-induced hepatotoxicity are uncertain. In the present study, we investigated whether VG has the potential to covalently bind to macromolecules in cells, a process that could initiate immune-mediated hepatotoxicity. For comparison, M20.7, a major metabolite of VG, and other DPP-4 inhibitors were also evaluated. We found that VG and anagliptin (ANG), which both contain a cyanopyrrolidine moiety, rapidly reacted in non-enzymatic manners on co-incubation with l -cysteine. Both VG and ANG had half-lives of 20–30 min. In contrast, incubation with GSH, rather than l -cysteine, failed to decrease the concentrations of VG or ANG. M20.7, sitagliptin, linagliptin, and alogliptin, having no cyanopyrrolidine moiety, were stable on incubation with l -cysteine or GSH. Structural analysis of the VG– and ANG–cysteine adducts, designated M407 and M487, respectively, revealed that the nitrile moieties of VG and ANG were irreversibly converted to a thiazoline acid. In conclusion, we found that VG and ANG have the potential to covalently bind to a thiol residue of l -cysteine in proteins. Such binding may lead to unpredictable immune responses in humans. l -Cysteine, rather than GSH, would likely be useful to detect the potential for covalent binding that could initiate immune-mediated hepatotoxicity.

A newly designed thiazoline iminopyridine ligand for enantioselective cobalt-catalyzed sequential Nazarov cyclization/electrophilic fluorination was developed. Various chiral α-fluorocyclopentenones were prepared with good yields and diastereo- and enantioselectivities. Further derivatizations could be easily carried out to provide chiral cyclopentenols with three contiguous stereocenters. Furthermore, a direct deesterification of fluorinated products could afford chiral α-single fluorine-substituted cyclopentenones.

Aim. To study the effect of blood thiazoline ammonium 4-chlorophenyl-2-hydroxy-4-oxo-2-butenoate content on the manifestation of anticoagulant effect upon subcutaneous administration to rabbits. Materials and methods. The studies were fulfilled on outbred rabbits. The substance was administered subcutaneously in the dose of 35 mg/kg. The time of blood coagulation was determined using coagulometer “Minilab 701”, and blood concentration - by means of liquid micro-column chromatograph “Milichrom A-02”. Results. The obtained results prove that maximum concentration of the compound in blood and maximum anticoagulant effect is reached in 30 minutes after subcutaneous administration. The substance anticoagulant effect lasted for 120 minutes after its administration. At present, the compound concentration in blood is equal to 519.8 ± 145.41 ng/ml. Conclusions. The studied compound, when administered subcutaneously, is quickly absorbed and begins to render an anticoagulant effect. Efficacy of action of this compound depends on its blood concentration.
