GOALS No established methods exist to predict who will require a higher number of endoscopic necrosectomy sessions for walled-off necrosis (WON). We aim to identify radiologic predictors for requiring a greater number of necrosectomy sessions. This may help to identify patients who benefit from aggressive endoscopic management. MATERIALS AND METHODS This is a multicenter retrospective study of patients with WON at 3 tertiary care centers. WON characteristics on preintervention computed tomography imaging were evaluated to determine if they were predictive of requiring more endoscopic necrosectomy. RESULTS A total of 104 patients were included. Seventy patients (67.3%) underwent endoscopic necrosectomy, with median of 2 necrosectomies. In this paper, ZnO thin film transistors were simulated and fabricated. The TFTs fabricated are of bottom gate configuration. Initially, the structure of the TFT is simulated by using Silvaco TCAD tool and the input and output characteristics are determined and the theoretical threshold voltage of the device is also calculated. Material characterization techniques such as SEM, XRD and EDS were performed to optimize the gate oxide layer and the oxide layer. The different materials are used in the fabrication of the thin film are Aluminium (Al) as the gate, source, and drain material. Aluminium oxide (Al$_{2} O_{3}$) as the gate oxide layer and Zinc Oxide (ZnO) as channel layers. PVD techniques such as sputtering and electron beam deposition are used to deposit the gate layer, gate oxide layer and the channel layer. Thermal evaporation is used to deposit the drain and source layer. After the deposition, the device is tested at a probing station to find out the input and output characteristics and verify if they match with the simulated values obtained from TCAD simulations.

Duplication of chromosome 22q11.2 (LCR A-D) has been reported at higher frequencies in clinical samples than the general population, but phenotypes vary widely. Triplication (4 copies) is rare, but studying the associated phenotype may provide insight into dosage-sensitivity of the genes in this chromosomal interval. We describe a proband with a triplication, specifically a “double duplication” (two copies per chromosome) of the 22q11.2 region, while his parents and two siblings each have a single duplication (3 copies). The proband had a heart malformation, dysmorphic features, and learning and socialization deficits, whereas the other family members did not. This family illustrates that while duplication of the 22q11.2 may not be sufficient to cause clinically significant neurodevelopmental or health-related phenotypes, triplication of the same region may result in a phenotype characterized by a mild neurodevelopmental disorder, facial dysmorphism, and possibly cardiac anomalies.

Although the prognostic value of p53 abnormalities in Stage III microsatellite stable (MSS) colorectal cancers (CRCs) is known, the gene expression profiles specific to the p53 status in the MSS background are not known. Therefore, the current investigation has focused on identification and validation of the gene expression profiles associated with p53 mutant phenotypes in MSS Stage III CRCs. Genomic DNA extracted from 135 formalin-fixed paraffin-embedded tissues, was analyzed for microsatellite instability (MSI) and p53 mutations. Further, mRNA samples extracted from five p53-mutant and five p53-wild-type MSS-CRC snap-frozen tissues were profiled for differential gene expression by Affymetrix Human Genome U133 Plus 2.0 arrays. Differentially expressed genes were further validated by the high-throughput quantitative nuclease protection assay (qNPA), and confirmed by quantitative real-time polymerase chain reaction (qRT-PCR) and by immunohistochemistry (IHC). Survival rates were estimated by Kaplan-Meier and Cox regression analyses. A higher incidence of p53 mutations was found in MSS (58%) than in MSI (30%) phenotypes. Both univariate (log-rank, P = 0.025) and multivariate (hazard ratio, 2.52; 95% confidence interval, 1.25-5.08) analyses have demonstrated that patients with MSS-p53 mutant phenotypes had poor CRC-specific survival when compared to MSS-p53 wild-type phenotypes. Gene expression analyses identified 84 differentially expressed genes. Of 49 down-regulated genes, LPAR6, PDLIM3, and PLAT, and, of 35 up-regulated genes, TRIM29, FUT3, IQGAP3, and SLC6A8 were confirmed by qNPA, qRT-PCR, and IHC platforms. p53 mutations are associated with poor survival of patients with Stage III MSS CRCs and p53-mutant and wild-type phenotypes have distinct gene expression profiles that might be helpful in identifying aggressive subsets.

Background: Diabetes mellitus refers to a group of common metabolic disorders that share the phenotype of hyperglycemia resulting from defects of reduced insulin secretion, decreased glucose utilization and increase in glucose production. It is estimated that there are currently 285 million people worldwide and this number is set to increase to 438 million by the year 2030. India has the highest number of patients with known diabetes worldwide, with a prevalence of 11.6%. The aim of the study was to evaluate the anti diabetic activity of ethanolic extract of leaves of plant Ocimum sanctum in alloxan induced diabetes in rats. Methods: The study was conducted on 4 groups of 6 rats each to evaluate the hypoglycaemic effect of ethanolic extract of Ocimum sanctum. Glibenclamide was used as a standard drug and the results were compared in reference to it. Tween 80 was given for both normal and diabetic control groups. The fasting blood sugar levels were recorded on 1 st , 3 rd , 5 th , 7 th , 10 th days by glucometer. Resul ts: The results indicate that the test compound ethanolic extract of Ocimum sanctum has significant and sustained oral hypoglycaemic activity, comparable with the hypoglycaemic effect of glibenclamide, a sulfonylurea. Conclusion: The hy poglycaemic potential of the test compound is found to be comparable with that of the standard drug glibenclamide.
