The systematic study of feather microstructure supports species identification, which is important in cases of illegally traded birds and bird-aircraft strikes. Our study focused on morphometric, macro- and micro-characters of feathers of Common Myna Acridotheres tristis from India. Among macro-characters, silver-colored filoplume feathers with pale black pigmentation on the barbs are specific for A. tristis. Morphometric measurements revealed that primary contour feathers (10.8±0.100 cm) were the longest and bristle feathers (1.26±0.051 cm) the shortest among all feathers. The longest (average) barb is found in primary contour feathers (1.875±0.123 cm), and the shortest in filoplume feathers (0.288±0.017 cm). We observed 3 types of nodal structures, and elongated prongs in bristle and filoplume feathers are significant characteristics of A. tristis. These insights into feather microstructures of A. tristis will aid species identification using plumology.

Earlier research on feather morphology emphasized comprehensively on the body contour feather than various other types of feathers. Therefore, we conducted a systematic study on all feather types of the Indian Pitta Pitta brachyura, a passerine bird native to the Indian subcontinent. Feather barbs from wing contour, tail contour, body contour, semiplume, down, powder down, and bristle feathers were retrieved from the bird and observed under a light microscope. Primary flight feathers from the right and left wing were longest (85.17 mm and 87.32 mm, respectively), whereas bristle feathers were the shortest (5.31 mm). The mean barb length was observed to be the highest (11.37±0.47 mm) in the wing feather followed by body contour (8.31±0.39 mm), semiplume (8.27±0.22 mm), tail feather (7.85±0.50 mm), down (6.45±0.21 mm), powder down (6.04±0.23 mm), and bristle (2.70±0.07 mm). Pearson correlation was found positive for barb length and feather length of down feathers (r= 0.996, p ≤0.05). We observed a novel type of barb the first time from dorsal body contour feather having plumulaceous barbules at the base followed by pennaceous barbules. This unique barbule arrangement is termed ‘sub-plumulaceous’ as it is distinct and analogous to known ‘sub-pennaceous’ type arrangement found absent in passerines.

Though a few in numbers, investigations on feather microstructures from the early 20th century till date, have contributed immensely to various fields such as phylogeny, palaeontology, archaeology, wildlife forensic, biomechanics and so on. However, existing studies on feather micro-structures of birds endemic to the India/Indian sub-continent are few in number and limited in their scope. Also, no study has ever been conducted to compare feather microstructures of different individuals of a species from India. To address this issue, a comparative feather microstructure study of three individuals of Turdoides affinis, a passerine endemic to the Indian sub-continent was done. Select microstructure parameters for five different types of feathers were studied in detail. The molecular sexing method was used to elucidate the sex of T. affinis individuals for gender based differences if any. Results of the study identified that two of T. affinis individuals were female whereas one of them was male. Morphometrically, tail contour was the longest (9.63±0.76 cm) and bristle were the shortest (1.00±0.07 cm) feather. Semiplume had the longest barb length (1.73±0.04 cm) and shortest barbs (0.16±0.01 cm) were present in bristles. Subpennaceous barbs and knob-shaped villi, characteristic of members of the Passeriformes family, was also observed in all three individuals. This study records no significant difference in feather characteristics amongst the three T. affinis individuals irrespective of the differences in their sex and size. Systematically documented feather micro-characteristics of T. affinis in this study could be used as a species identification tool and would provide baseline data for the feather catalogue of Indian bird species being compiled at SACON.

Mitochondrial genome provides useful information about species concerning its evolution and phylogenetics. We have taken the advantage of high throughput next-generation sequencing technique to sequence the complete mitogenome of Yellow-billed babbler (Turdoides affinis), a species endemic to Peninsular India and Sri Lanka. Both, reference-based and de-novo assemblies of mitogenome were performed and observed that de-novo assembled mitogenome was most appropriate. The complete mitogenome of yellow-billed babbler (assembled de-novo) was 17,672 bp in length with 53.2% AT composition. Thirteen protein-coding genes along with two rRNAs and 22 tRNAs were detected. The arrangement pattern of these genes was found conserved among Leiothrichidae family mitogenomes. Duplicated control regions were found in the newly sequenced mitogenome. Downstream bioinformatics analysis revealed the effect of translational efficiency and purifying selection pressure over thirteen protein-coding genes in yellow-billed babbler mitogenome. Ka/Ks analysis indicated the highest synonymous substitution rate in the nad6 gene. Evolutionary analysis revealed the conserved nature of all the protein-coding genes across Leiothrichidae family mitogenomes. Our limited phylogeny results placed T. affinis in a separate group, a sister group of Garrulax. Overall, our results provide a useful information for future studies on the evolutionary and adaptive mechanisms of birds belong to the Leiothrichidae family.

Psittacula cyanocephala is a parakeet endemic to the Indian sub-continent, widespread in the illegal bird trade. Previous studies on Psittacula parakeets have highlighted taxonomic ambiguities, warranting further studies to resolve such issues. Since the mitochondrial genome provides useful information about a species concerning its evolution and phylogenetics, we sequenced the complete mitogenome of P. cyanocephala using NGS, validated 38.86% of the mitogenome using Sanger Sequencing and compared it with other available whole mitogenomes of Psittacula. The complete mitogenome of the species was 16814 bp in length with 54.08% AT composition. P. cyanocephala mitogenome comprises of 13 protein-coding genes, 2 rRNAs and 22 tRNAs. P. cyanocephala mitogenome organization was consistent with other Psittacula mitogenomes. Comparative codon usage analysis indicated the role of natural selection on Psittacula mitogenomes. Strong purifying selection pressure was observed maximum on nad1 and nad4l genes. The mitochondrial control region of all Psittacula species displayed the ancestral avian CR gene order. Phylogenetic analyses revealed the Psittacula genus as paraphyletic nature, containing at least 4 groups of species within the same genus, suggesting its taxonomic reconsideration. Our results provide useful information for developing forensic tests to control the illegal trade of the species, scientific basis for phylogenetic revision of genus Psittacula.

Our limited phylogenetics results suggest thatT. affinisis closer toGarrulax.
