Experimental autoimmune uveoretinitis (EAU) is a cell-mediated model of retinal autoimmunity that is negatively regulated by interleukin (IL)-10. The antidepressant drug rolipram, a type IV phosphodiesterase inhibitor, enhances IL-10 production by monocyte/macrophages. The effect of rolipram on induction of EAU and its associated immunologic responses was investigated.Mice were challenged for EAU induction by immunization with the retinal antigen interphotoreceptor retinoid-binding protein (IRBP) or by adoptive transfer of uveitogenic T cells and were treated with rolipram. EAU severity and immunologic responses to IRBP were analyzed. In addition, the effect of rolipram added to the culture on antigen-driven responses of primed lymph node cells was tested.Rolipram treatment from days -1 to 7 after immunization (afferent phase) was not protective, but severity of EAU was reduced to 50% by treatment from days 8 to 16 after immunization or when EAU was induced by adoptive transfer (efferent phase). Antigen-specific proliferation and interferon (IFN)-gamma production ex vivo by lymph node cells of protected mice were not reduced. However, the addition of rolipram directly to the culture suppressed IRBP-driven proliferation and IFN-gamma production by primed lymph node cells. Freshly explanted lymph node cells of treated mice showed inhibition of IFN-gamma mRNA but no parallel enhancement of IL-10 mRNA by quantitative polymerase chain reaction. Rolipram inhibited EAU in IL-10 knockout mice equally well compared with controls and suppressed their primed lymph node cells in culture.Rolipram appears to inhibit the expansion and effector function of uveitogenic T cells, raising the possibility that it may be useful for treatment of established disease. Contrary to expectations based on in vitro studies, the protective effects in vivo appear to be independent of IL-10. The observation that suppression of antigen-specific responses is demonstrable only in the physical presence of the drug suggests that, in a clinical setting, continuous administration of rolipram might be needed to sustain its therapeutic effect.

The binding and trans-endothelium migration of inflammatory cells is believed to play a critical role in a variety of inflammatory conditions. This study investigates the ability of the experimental drug suramin to block the activation of HUVEC by endotoxin and by the proinflammatory cytokines IL-1 and TNF. We demonstrate that the inducible expression of several adhesion molecules by LPS and IL-1 beta but not by TNF-alpha is prevented by suramin. In a dose-dependent manner, suramin inhibits the binding of neutrophils and T lymphocytes to LPS and IL-1 beta but not to TNF-alpha-activated HUVEC. The inhibitory effect of the drug on IL-1 beta-induced but not on LPS-induced cell stimulation can be completely reversed by the addition of excess cytokine but not by excess LPS. Because LPS activation of HUVEC is known to depend on serum/plasma-derived soluble CD14, we set out to determine whether suramin inhibition involves interference with the action of the CD14-LPS complex on HUVEC. Indeed, the drug prevents the binding of radioactive LPS in the presence of serum and inhibits LPS-induced cell activation in serum-free medium supplemented with recombinant soluble CD14. The results suggest that suramin interferes with the CD14-dependent activation of HUVEC and that it also may be a useful agent in blocking infectious endotheliopathies in vivo.

Tissue-specific cytokine mRNA expression was examined in mice that received LPS. In the liver, IL-6, IL-10, IL-12 (p40), and TNF-alpha were induced by 30 min after injection with LPS. In the spleen, IL-6 and TNF-alpha were induced by 30 min after LPS challenge, while increases in IL-10 and IL-12 (p40) were delayed in onset. GM-CSF, IFN-gamma, and IL-12 (p35) were not induced in the liver or spleen until 60 to 90 min after LPS injection. Mice were depleted of macrophages in their liver and spleen by i.v. injection of liposome-encapsulated dichloromethylene bisphosphonate (Cl2MBP). Induction of IL-1 beta, IL-6, IL-10, and IL-12 (p40) mRNA by LPS was reduced by > 95% in the liver of macrophage-depleted mice, implicating macrophages as the primary producers of these cytokines. Macrophage depletion resulted in a 50 to 75% reduction in TNF-alpha mRNA in the liver. The results from Cl2MBP-liposome-treated mice also suggested that splenic macrophages were the primary producers of LPS-induced IL-1 beta, IL-6, IL-12 (p40), and IL-1 receptor antagonist (IL-1ra) mRNA, but not IL-10 and TNF-alpha mRNA. Mice treated with Cl2MBP-liposomes were more susceptible to ionizing irradiation than control mice, whether or not they were administered a radioprotective dose of LPS. These findings suggest that depletion of liver and splenic macrophages results in a dysregulation of basal and LPS-induced cytokine responses that can be associated with an altered biologic response.
