Cerebral white matter rarefaction (CWMR) was considered by Binswanger and Alzheimer to be due to cerebral arteriolosclerosis. Renewed attention came with CT and MR brain imaging, and neuropathological studies finding a high rate of CWMR in Alzheimer disease (AD). The relative contributions of cerebrovascular disease and AD to CWMR are still uncertain. In 1181 autopsies by the Arizona Study of Aging and Neurodegenerative Disorders (AZSAND), large-format brain sections were used to grade CWMR and determine its vascular and neurodegenerative correlates. Almost all neurodegenerative diseases had more severe CWMR than the normal control group. Multivariable logistic regression models indicated that Braak neurofibrillary stage was the strongest predictor of CWMR, with additional independently significant predictors including age, cortical and diencephalic lacunar and microinfarcts, body mass index, and female sex. It appears that while AD and cerebrovascular pathology may be additive in causing CWMR, both may be solely capable of this. The typical periventricular pattern suggests that CWMR is primarily a distal axonopathy caused by dysfunction of the cell bodies of long-association corticocortical projection neurons. A consequence of these findings is that CWMR should not be viewed simply as “small vessel disease” or as a pathognomonic indicator of vascular cognitive impairment or vascular dementia.

The structure of the title compound, 2C18H19ClN4O·HCl or (CNO)2·HCl (C36H39Cl3N8O2), at 100 K has tetragonal (I4/m) symmetry. The dihedral angle between the benzene rings of the fused ring system of the CNO molecule is 40.08 (6)° and the equivalent angle between the seven-membered ring and its pendant N-oxide ring is 31.14 (7)°. The structure contains a very strong, symmetrical O—H...O hydrogen bond [O...O = 2.434 (2) Å] between two equivalent R 3N+—O− moieties, which share a proton lying on a crystallographic twofold rotation axis. These units then form a (CNO)4·(HCl)2 ring by way of two equivalent N—H...Cl hydrogen bonds (Cl− site symmetry m). These rings are catenated into infinite chains propagating along the c-axis direction by way of shape complementarity and directional C—H...N and C—H...π interactions.

In this paper, we use Large Eddy Simulations (LES) in combination with the Ffowcs Williams - Hawkings method to study the influence of chevrons on the flow field as well as the noise produced by a closely installed M = 0.6 jet. The LES simulations are performed with the spectral/hp element framework Nektar++. Nektar++ uses the high-order discontinuous Galerkin method and an implicit scheme based on the matrix-free Newton-GMRES method to discretize the unfiltered Navier-Stokes equations in space and time, respectively. The far-field noise is computed using Antares. Antares solves the Ffowcs Williams - Hawkings equation for a permeable integration surface in the time-domain using a source-time dominant algorithm. The aerodynamic results show good agreement with experimental data obtained in the Doak Laboratory Flight Jet Rig, located at the University of Southampton. Some discrepancies are observed in terms of the far-field noise levels, especially for higher polar observer angles relative to the downstream jet axis. In terms of noise reduction potential, the simulations predict that the chevrons reduce the OASPL by 1dB compared to an installed round nozzle for all observers located on the unshielded side of the wing. This should be compared to the experiments, which predict a 1.5dB noise reduction for the same chevron nozzle.

IntroductionWith technical advances, confocal and super-resolution microscopy have become powerful tools to dissect cellular pathophysiology. Cell attachment to glass surfaces compatible with advanced imaging is critical prerequisite but remains a considerable challenge for human beta cells. Recently, Phelps et al. reported that human beta cells plated on type IV collagen (Col IV) and cultured in neuronal medium preserve beta cell characteristics.MethodsWe examined human islet cells plated on two commercial sources of Col IV (C6745 and C5533) and type V collagen (Col V) for differences in cell morphology by confocal microscopy and secretory function by glucose-stimulated insulin secretion (GSIS). Collagens were authenticated by mass spectrometry and fluorescent collagen-binding adhesion protein CNA35. ResultsAll three preparations allowed attachment of beta cells with high nuclear localization of NKX6.1, indicating a well-differentiated status. All collagen preparations supported robust GSIS. However, the morphology of islet cells differed between the 3 preparations. C5533 showed preferable features as an imaging platform with the greatest cell spread and limited stacking of cells followed by Col V and C6745. A significant difference in attachment behavior of C6745 was attributed to the low collagen contents of this preparation indicating importance of authentication of coating material. Human islet cells plated on C5533 showed dynamic changes in mitochondria and lipid droplets (LDs) in response to an uncoupling agent 2-[2-[4-(trifluoromethoxy)phenyl]hydrazinylidene]-propanedinitrile (FCCP) or high glucose + oleic acid. DiscussionAn authenticated preparation of Col IV provides a simple platform to apply advanced imaging for studies of human islet cell function and morphology.

Aims Saint John’s wort, Hypericum perforatum, is a medicinally and ecologically important perennial plant species that has a broad global distribution. Despite the species’ importance, little is known about the factors that structure its microbial communities and the identity of microbes that enhance plant growth and fitness. Here we aim to describe the microbial communities associated with Hypericum perforatum and elucidate factors that structure these communities.Methods We collected H. perforatum root samples in three adjacent habitat types: wet and dry alvars (two types of limestone barren) and fallow agricultural fields (i.e. old-fields), in Jefferson County, New York. We used high-throughput amplicon sequencing of the SSU-rRNA gene (16S, bacteria) and the internal transcribed spacer region 1 (ITS1, fungi) to characterize the root microbiome of H. perforatum. At each root sampling location, we quantified aspects of the plant phenotype and soil characteristics to evaluate habitat variables that correlate with root microbial communities.Results Alvars had ~ 13% higher bacterial richness compared to old-fields. In contrast, old-fields had 28% higher fungal richness than dry alvars, but similar fungal richness to wet alvars. Habitat and plant height were important predictors of microbial community composition. We identified two bacterial taxa positively associated with plant height, both belonging to the bacterial phylum Actinobacteria.Conclusions This work contributes to our understanding of the environmental determinants of microbial community composition. Additionally, we were able to identify bacterial taxa that are correlated with plant health and should be investigated further as indicators of soil health or plant-growth promoting rhizobacteria.

Coastal and estuarine ecosystems are environments heavily influenced by natural and anthropogenic activities. Chemicals used for pest control in agriculture and aquaculture may accumulate in natural coastal environments. Pyrethroids are common pesticides that are used on crops as well as applied to aquaculture pens and then may disperse in the surrounding ocean once treatment is complete. This study observed the sublethal effects of two pyrethroids, permethrin and deltamethrin (within commercially available formulations), on post-larval stage IV American lobster (Homarus americanus) using growth parameters and metabolic rate as indicators. Observed effects on growth parameters were a decrease in size increment and specific growth rate as well as an increase in intermolt period in stage IV lobsters exposed to 100 μg/kg permethrin. No significant differences were found for intermolt period, size increment, or specific growth rate in deltamethrin-exposed stage IV lobsters. Metabolic rates were not significantly different between deltamethrin-exposed and control lobsters, however, this sublethal effect warrants further investigation. Collectively, these results represent the first examination of the sublethal effects of exposure to pyrethroids formulations in post-larval lobsters, highlighting the potential for effects on non-target marine organisms.

Bacterial cell growth and division require the coordinated action of enzymes that synthesize and degrade cell wall polymers. Here, we identify enzymes that cleave the d-arabinan core of arabinogalactan, an unusual component of the cell wall of Mycobacterium tuberculosis and other mycobacteria. We screened 14 human gut-derived Bacteroidetes for arabinogalactan-degrading activities and identified four families of glycoside hydrolases with activity against the d-arabinan or d-galactan components of arabinogalactan. Using one of these isolates with exo-d-galactofuranosidase activity, we generated enriched d-arabinan and used it to identify a strain of Dysgonomonas gadei as a d-arabinan degrader. This enabled the discovery of endo- and exo-acting enzymes that cleave d-arabinan, including members of the DUF2961 family (GH172) and a family of glycoside hydrolases (DUF4185/GH183) that display endo-d-arabinofuranase activity and are conserved in mycobacteria and other microbes. Mycobacterial genomes encode two conserved endo-d-arabinanases with different preferences for the d-arabinan-containing cell wall components arabinogalactan and lipoarabinomannan, suggesting they are important for cell wall modification and/or degradation. The discovery of these enzymes will support future studies into the structure and function of the mycobacterial cell wall.

IntroductionChymase is a highly destructive serine protease rapidly neutralized in the circulation by protease inhibitors. Here we test whether pericardial fluid (PCF) chymase activation and other inflammatory biomarkers determine intensive care unit length of stay, and explore mechanisms of chymase delivery by extracellular vesicles to the heart.MethodsPCF was collected from adult patients (17 on-pump; 13 off-pump) 4 h after cardiac surgery. Extracellular vesicles (EVs) containing chymase were injected into Sprague–Dawley rats to test for their ability to deliver chymase to the heart.ResultsThe mean intensive care unit (ICU) stay and mean total length of stay was 2.17 ± 3.8 days and 6.41 ± 1.3 days respectively. Chymase activity and 32 inflammatory markers did not differ in on-pump vs. off-pump cardiac surgery. Society of Thoracic Surgeons Predicted Risk of Morbidity and Mortality Score (STS-PROM), 4-hour post-surgery PCF chymase activity and C-X-C motif chemokine ligand 6 (CXCL6) were all independent predictors of ICU and total hospital length of stay by univariate analysis. Mass spectrometry of baseline PCF shows the presence of serine protease inhibitors that neutralize chymase activity. The compartmentalization of chymase within and on the surface of PCF EVs was visualized by immunogold labeling and transmission electron microscopy. A chymase inhibitor prevented EV chymase activity (0.28 fmol/mg/min vs. 14.14 fmol/mg/min). Intravenous injection of PCF EVs obtained 24 h after surgery into Sprague Dawley rats shows diffuse human chymase uptake in the heart with extensive cardiomyocyte damage 4 h after injection.DiscussionEarly postoperative PCF chymase activation underscores its potential role in cardiac damage soon after on- or off-pump cardiac surgery. In addition, chymase in extracellular vesicles provides a protected delivery mechanism from neutralization by circulating serine protease inhibitors.

Signaling from multiple receptor tyrosine kinases (RTK) contributes to therapeutic resistance in glioblastoma (GBM). Heparan sulfate (HS), present on cell surfaces and in the extracellular matrix, regulates cell signaling via several mechanisms. To investigate the role for HS in promoting RTK signaling in GBM, we generated neural progenitor cells deficient for HS by knockout of the essential HS-biosynthetic enzyme Ext1, and studied tumor initiation and progression. HS-null cells had decreased proliferation, invasion, and reduced activation of multiple RTKs compared with control. In vivo tumor establishment was significantly decreased, and rate of tumor growth reduced with HS-deficient cells implanted in an HS-poor microenvironment. To investigate if HS regulates RTK activation through platelet-derived growth factor receptor α (PDGFRα) signaling, we removed cell surface HS in patient-derived GBM lines and identified reduced cell surface PDGF-BB ligand. Reduced ligand levels were associated with decreased phosphorylation of PDGFRα, suggesting HS promotes ligand–receptor interaction. Using human GBM tumorspheres and a murine GBM model, we show that ligand-mediated signaling can partially rescue cells from targeted RTK inhibition and that this effect is regulated by HS. Indeed, tumor cells deficient for HS had increased sensitivity to EGFR inhibition in vitro and in vivo.Implications:Our study shows that HS expressed on tumor cells and in the tumor microenvironment regulates ligand-mediated signaling, promoting tumor cell proliferation and invasion, and these factors contribute to decreased tumor cell response to targeted RTK inhibition.

International audience; The Carbohydrate-Active Enzyme classification groups enzymes that breakdown, assemble, or decorate glycans into protein families based on sequence similarity. The glycoside hydrolases (GH) are arranged into over 170 enzyme families, with some being very large and exhibiting distinct activities/specificities towards diverse substrates. Family GH31 is a large family that contains more than 20,000 sequences with a wide taxonomic diversity. Less than 1% of GH31 members are biochemically characterized and exhibit many different activities that include glycosidases, lyases, and transglycosidases. This diversity of activities limits our ability to predict the activities and roles of GH31 family members in their host organism and our ability to exploit these enzymes for practical purposes. Here, we established a subfamily classification using sequence similarity networks that was further validated by a structural analysis. While sequence similarity networks provide a sequence-based separation, we obtained good segregation between activities among the subfamilies. Our subclassification consists of 20 subfamilies with sixteen subfamilies containing at least one characterized member and eleven subfamilies that are monofunctional based on the available data. We also report the biochemical characterization of a member of the large subfamily 2 (GH31_2) that lacked any characterized members: RaGH31 from Rhodoferax aquaticus is an α-glucosidase with activity on a range of disaccharides including sucrose, trehalose, maltose, and nigerose. Our subclassification provides improved predictive power for the vast majority of uncharacterized proteins in family GH31 and highlights the remaining sequence space that remains to be functionally explored.

Glycoside hydrolases (GHs) are a diverse group of enzymes that catalyze the hydrolysis of glycosidic bonds. The Carbohydrate-Active enZymes (CAZy) database organizes GHs into families based on sequence data and function, with fewer than 1% of the predicted proteins characterized biochemically. Consideration of genomic context can provide a useful guide to infer possible enzyme activities for proteins of unknown function. We used the MultiGeneBLAST tool to discover a putative gene cluster inMarinovumsp., a member of the marineRoseobacterclade, that encodes homologues of enzymes belonging to the sulfoquinovose monooxygenase pathway. This putative gene cluster lacks a gene encoding a classical family GH31 sulfoquinovosidase (SQase) candidate, but which instead includes an uncharacterized family GH13 protein (MsGH13). We show that recombinantMsGH13 lacks SQase activity but is a broad spectrum α-glucosidase active on a diverse array of α-linked disaccharides, including: maltose (100%), sucrose (7.9%), nigerose (52%), trehalose (20%), isomaltose (6.4%), and kojibiose (3.8%). Using AlphaFold, a 3D model for theMsGH13 enzyme was constructed that predicted its active site shared close similarity with a narrower specificity α-glucosidase fromHalomonassp. H11 of the same GH13 subfamily. This study supports recent findings that bacteria belonging to theRoseobacterclade can metabolize SQ without a classical SQase encoded in their genomes.

Introduction: Visualizing small individual biomolecules at subcellular resolution in live cells and tissues can provide valuable insights into metabolic activity in heterogeneous cells, but is challenging.Methods: Here, we used stimulated Raman scattering (SRS) microscopy to image deuterated methionine (d-Met) incorporated into Drosophila tissues in vivo.Results: Our results demonstrate that SRS can detect a range of previously uncharacterized cell-to-cell differences in d-Met distribution within a tissue at the subcellular level.Discussion: These results demonstrate the potential of SRS microscopy for metabolic imaging of less abundant but important amino acids such as methionine in tissue.

Background Hemangioblastoma of the central nervous system is an uncommon benign neoplasm, with about 25% of cases in patients with von Hippel–Lindau disease. The incidence of metastasis is rare, particularly in patients without von Hippel–Lindau disease. We report a case of hemangioblastoma with leptomeningeal dissemination as a late recurrence. Case presentation A 65-year-old Caucasian man with a history of World Health Organization grade I hemangioblastoma of the cerebellar vermis underwent gross total resection in 1997. In early 2018, he developed intracranial recurrences with diffuse leptomeningeal disease of the entire spine. The patient underwent resection of intracranial recurrence, followed by palliative craniospinal irradiation. The disease progressed quickly, and he died 8 months after recurrence. Conclusions Despite a benign pathology, hemangioblastoma has a low risk of metastasis. The outcome for hemangioblastoma patients with metastasis is poor. Multidisciplinary care for patients with metastatic hemangioblastoma warrants further investigation, and an effective systemic option is urgently needed. Regular lifelong follow-up of at-risk patients is recommended.

Sulfolactate (SL) is a short-chain organosulfonate that is an important reservoir of sulfur in the biosphere. SL is produced by oxidation of sulfolactaldehyde (SLA), which in turns derives from sulfoglycolysis of the sulfosugar sulfoquinovose, or through oxidation of 2,3-dihydroxypropanesulfonate, which are themselves important organosulfur compounds in the biogeochemical sulfur cycle. Oxidation of SLA is catalyzed by SLA dehydrogenases belonging to the aldehyde dehydrogenase superfamily. We report that SLA dehydrogenaseRlGabDfrom the sulfoglycolytic bacteriumRhizobium leguminsarumSRDI565 can use both NAD+and NADP+as cofactor to oxidize SLA, and operates through a rapid equilibrium ordered mechanism. We report the cryo-EM structure ofRlGabD bound to NADH, revealing a tetrameric quaternary structure and supporting proposal of organosulfonate binding residues in the active site, and a catalytic mechanism. Sequence based homology searches identified SLA dehydrogenase homologs in a range of putative sulfoglycolytic gene clusters in bacteria predominantly from the phyla Actinobacteria, Firmicutes, and Proteobacteria. This work provides a structural and biochemical view of SLA dehydrogenases to complement our knowledge of SLA reductases, and provide detailed insights into a critical step in the organosulfur cycle.Graphical abstractSulfolactate is an important species in the biogeochemical sulfur cycle. We estimated the probability of undetected emergence of the SARS-CoV-2 Omicron variant in 25 low and middle-income countries (LMICs) prior to December 5, 2021. In nine countries, the risk exceeds 50%; in Turkey, Pakistan and the Philippines, it exceeds 99%. Risks are generally lower in the Americas than Europe or Asia.

The sulfosugar sulfoquinovose is estimated to be produced on a scale of 10 billion metric tons per annum, making it a major organosulfur species in the biosulfur cycle. The microbial degradation of sulfoquinovose through sulfoglycolysis allows the utilization of its carbon content and contributes to the biomineralization of its sulfur.

Although testing, contact tracing, and case isolation programs can mitigate COVID-19 transmission and allow the relaxation of social distancing measures, few countries worldwide have succeeded in scaling such efforts to levels that suppress spread. The efficacy of test-trace-isolate likely depends on the speed and extent of follow-up and the prevalence of SARS-CoV-2 in the community. Here, we use a granular model of COVID-19 transmission to estimate the public health impacts of test-trace-isolate programs across a range of programmatic and epidemiological scenarios, based on testing and contact tracing data collected on a university campus and surrounding community in Austin, TX, between October 1, 2020, and January 1, 2021. The median time between specimen collection from a symptomatic case and quarantine of a traced contact was 2 days (interquartile range [IQR]: 2 to 3) on campus and 5 days (IQR: 3 to 8) in the community. Assuming a reproduction number of 1.2, we found that detection of 40% of all symptomatic cases followed by isolation is expected to avert 39% (IQR: 30% to 45%) of COVID-19 cases. Contact tracing is expected to increase the cases averted to 53% (IQR: 42% to 58%) or 40% (32% to 47%), assuming the 2- and 5-day delays estimated on campus and in the community, respectively. In a tracing-accelerated scenario, in which 75% of contacts are notified the day after specimen collection, cases averted increase to 68% (IQR: 55% to 72%). An accelerated contact tracing program leveraging rapid testing and electronic reporting of test results can significantly curtail local COVID-19 transmission.

Artificial intelligence (AI) can accelerate materials design by identifying the key parameters correlated with the performance. However, widely used AI methods require big data, and only the smallest part of the available data in heterogeneous catalysis meets the quality requirement for data-efficient AI. Here, we use rigorous experimental procedures, designed to consistently take into account the kinetics of the catalyst active states formation, in order to measure 55 physicochemical parameters as well as the reactivity of 12 catalysts towards ethane, propane, and n-butane oxidation. These catalyst materials are based on vanadium or manganese redox-active elements (RAEs) and present diverse phase compositions, crystallinities, and catalytic behaviors. By applying the sure-independence-screening-and-sparsifying-operator (SISSO) approach to the consistent data set, we identify nonlinear property-function relationships depending on several key parameters, reflecting the intricate interplay of underlying processes governing selective oxidation. This approach indicates the most relevant characterization techniques and shows how the catalyst properties may be tuned in order to achieve the desired performance. For example, to achieve high olefin yields, the catalyst must have a high specific surface area, a low concentration of surface RAE, and the ability to change the surface RAE oxidation states under reaction conditions with respect to vacuum. These parameters are measured by N2 adsorption, x-ray photoelectron spectroscopy (XPS), and near-ambient-pressure in situ XPS. They reflect the relevance of local transport, site isolation, surface redox activity, and the materials dynamical restructuring under reaction conditions. Although the relationship describing the even more challenging oxygenate yields shares similarities with that for olefin yields, a parameter reflecting the importance of specific surface sites, derived from the analysis of the carbon 1s XPS spectra, is additionally identified as key for high selectivity to oxygenates.

We examined whether practice activities adopted by professional youth soccer coaches are modulated through the implementation of and engagement with cocreative evidence-based programs. Across two experiments, we used systematic observation to identify the practice activities of seven coaches across 134 sessions. In Experiment A, drill-based and games-based activities were recorded and quantified. To encourage behaviour change across the study, the systematic observation data were compared with skill acquisition literature to provide coaches with quantitative feedback and recommendations during workshops. Postworkshop systematic observation data indicated that practice activities used by coaches changed in accordance with the evidenced-based information (increase in games-based activities) delivered within the workshop. Interview data indicated that coaches typically stated that the workshop was a key reason for behaviour change. In a follow-up Experiment B, feedback and recommendations were delivered using an interactive video-based workshop. The systematic observation data indicated that coaches increased the use of soccer activities that contained active decision making with coaches citing the workshop as a key reason for behaviour change. These findings indicate that coaching practice activities can be supported and shaped through the implementation of cocreated workshops wherein coaches collaborate with sport scientists and researchers to bridge the gap between science and application.

TVET colleges in South Africa are primarily tasked with preparing students for the workplace. However, employers noted dissatisfaction in the skills and abilities of students entering the workplace. A possible reason for the poor preparation of students may have been found in past research. The curriculum at TVET colleges, as elsewhere in higher education, is very Eurocentric and theoretical in nature. The need for decolonisation of the curriculum thus arises. A decolonized curriculum prepares students to work in a variety of social contexts and considers issues like poverty, inequality and unemployment. A decolonized curriculum needs to include content and context derived from local sources, and need to connect theory with practice. It is thus expected of TVET lecturers to interpret and link theoretical content from the curriculum to the needs of local sources, workplaces and industry. TVET lecturers thus need to have knowledge and experiences from a variety of backgrounds. Past research indicated certain lecturers lack workplace experience and qualifications. It was found that intermediate and FET (grade 10‒11) qualified teachers and graduates from universities who lack workplace experience are employed at TVET colleges. Lecturers who lack workplace experience may find it difficult to link content with the needs of the industry for which they have to prepare their students and may not know how to link theory with practice during lessons. Teaching and learning at TVET colleges require concrete experiences and deliberate encounters. Actual, concrete experiences change the perspectives of students and lecturers and affects them profoundly, which can be seen as a pedagogic piligrimage (Davids and Waghid 2019, 7). To address the latter, SSACI, a Swiss-South African initiative launched a work-integrated learning (WIL) project to identify lecturers that are only equipped with a teaching qualification and lack workplace experience. The work-integrated learning program flared up lecturer’s experience of work-integrated learning and increased motivation and enthusiasm, as they were able to integrate the examples learned from the workplace, to make lessons and lesson preparation more interesting. In addition, lecturers were able to tell students what to expect after graduation when they enter the workplace. A factor that negatively influences the effectiveness of work-integrated learning is the lack of policy supporting the program.

In 1627, at the height of the Bamberg witch-hunt (1595–1631), the prince-bishopric erected the Malefizhaus (“witchcraft-house”), the first cellular prison purpose-built for solitary confinement. This article recovers the history of the Malefizhaus to establish the importance of imprisonment and carceral institutions to the early modern witch-craze. The prison at once concretized the ideology of the hunt and furnished a fearsome weapon of persecution, extracting the confessions without which no inquisitorial campaign could function. By reconstructing the singular architecture and internal regimen of the Malefizhaus, this article demonstrates the sophistication of early modern interrogations, a process distorted by an outsized interest in torture. Having recognized the Malefizhaus as a driver of the witch-hunt, it is possible to recognize the prison’s impact upon Bamberg’s seventeenth-century history—disrupting political and economic relationships, displacing populations, and disciplining social life. The case of the Bamberg witches’ prison counters the modernist slant of the study of the prison, proof that medieval and early modern carceral institutions shaped the history of their societies, despite smaller scales and weaker state apparatuses. In turn, the essay argues that the critical tools of carceral studies, developed to study contemporary mass incarceration, can profitably be applied to premodern practices and institutions, offering insight into patterns of violence, the development of repressive structures, and the problems of “crime” as a historical category.

Levoglucosan is produced in the pyrolysis of cellulose and starch, including from bushfires or the burning of biofuels, and is deposited from the atmosphere across the surface of the earth. We describe two levoglucosan degrading Paenarthrobacter spp. (Paenarthrobacter nitrojuajacolis LG01 and Paenarthrobacter histidinolovorans LG02) that were isolated from soil by metabolic enrichment using levoglucosan as the sole carbon source. Genome sequencing and proteomics analysis revealed the expression of a series of genes encoding known levoglucosan degrading enzymes, levoglucosan dehydrogenase (LGDH, LgdA), 3-keto-levoglucosan β -eliminase (LgdB1) and glucose 3-dehydrogenase (LgdC), along with an ABC transporter cassette and an associated solute binding protein. However, no homologues of 3-ketoglucose dehydratase (LgdB2) were evident, while the expressed genes contained a range of putative sugar phosphate isomerases/xylose isomerases with weak similarity to LgdB2. Sequence similarity network analysis of genome neighbours of LgdA revealed that homologues of LgdB1 and LgdC are generally conserved in a range of bacteria in the phyla Firmicutes, Actinobacteria and Proteobacteria. One group of sugar phosphate isomerase/xylose isomerase homologues (named LgdB3) was identified with limited distribution that is mutually exclusive with LgdB2, and we propose that they may fulfil a similar function. LgdB1, LgdB2 and LgdB3 adopt similar predicted 3D folds, suggesting overlapping function in processing intermediates in LG metabolism. Our findings highlight diversity within the LGDH pathway, through which bacteria utilize levoglucosan as a nutrient source.

Parking sequences (a generalization of parking functions) are defined by specifying car lengths and requiring that a car attempts to park in the first available spot after its preference. If it does not fit there, then a collision occurs and the car fails to park. In contrast, parking assortments generalize parking sequences (and parking functions) by allowing cars (also of assorted lengths) to seek forward from their preference to identify a set of contiguous unoccupied spots in which they fit. We consider both parking sequences and parking assortments and establish that the number of preferences resulting in a fixed parking order $\sigma$ is related to the lengths of cars indexed by certain subsequences in $\sigma$. The sum of these numbers over all parking orders (i.e. permutations of $[n]$) yields new formulas for the total number of parking sequences and of parking assortments., Comment: 9 pages, 2 figures, 1 table

A recently developed computational framework for jet noise is used to compute the noise generated by an isolated and installed jet. The framework consists of two parts. In the first part, the spectral/hp element framework Nektar++ is used to compute the near-field flow. Nektar++ solves the unfiltered Navier-Stokes equations on unstructured grids using the high-order discontinuous Galerkin method. The discrete equations are integrated in time using an implicit scheme based on the matrix-free Newton-GMRES method. In the second part, the Antares library is used to compute the far-field noise. Antares solves the Ffowcs Williams - Hawkings equation for a permeable integration surface in the time domain using a source-time dominant algorithm. The simulations are validated against experimental data obtained in the Doak Laboratory Flight Jet Rig, located at the University of Southampton. For the isolated jet, good agreement is achieved, both in terms of the flow statistics and the far-field noise. The discrepancies observed for the isolated jet are believed to be caused by an under-resolved boundary layer in the simulations. For the installed jet, the flow statistics are also well predicted. In the far-field, very good agreement is achieved for downstream observers. For upstream observers, some discrepancies are observed for very high and very low frequencies.

To conduct a systematic review to summarize cognitive instruments being used in long-term outcome studies of survivors of adult critical illness, as well as evaluate whether these measures are reported as using patient demographic norms, specifically race norms.A comprehensive search was conducted in PubMed (National Center for Biotechnology Information), Excerpta Medica dataBASE (Ovid), Psychological Information Database (ProQuest), and Web of Science (Clarivate) for English language studies published since 2002.Studies were eligible if the population included adult ICU survivors assessed for postdischarge cognitive outcomes.Two independent reviewers screened abstracts, examined full text, and extracted data from all eligible articles.A total of 98 articles (55 unique cohorts: 22 general ICU, 14 Acute respiratory distress syndrome/Acute respiratory failure/Sepsis, 19 COVID-19 and other subpopulations) were eligible for data extraction and synthesis. Among general ICU survivors, the majority of studies (Less than half of the studies measuring cognitive outcomes in ICU survivors reported the use of norming characteristics. There is substantial heterogeneity in how studies reported the use of cognitive instruments, and hence, the prevalence of the use of patient norms may be underestimated. These findings are important in the development of appropriate standards for use and reporting of neuropsychological tests among ICU survivors.
