NADC34-like porcine reproductive and respiratory syndrome viruses (PRRSVs) are emerging globally. Although lineage 1 PRRSVs are emerging in Korea, NADC34-like PRRSV strains have not been detected to date. This study reports the isolation of the first NADC34-like PRRSV strains from samples of MLV-vaccinated animals on a Korean farm with a severe PRRS outbreak causing high mortality and abortion rates in July 2022. Three PRRSV strains with a whole genome length of 15,088 bp were isolated from the outbreaks, and their phylogenetic, recombination properties, and in vitro growth kinetics were evaluated comprehensively. These Korean strains clustered with those of American and Chinese sublineage 1.5 (NADC34-like strains), according to phylogenetic analyses based on the ORF5 gene and the whole genome. All three complete genome sequences originated from recombination between the RespPRRS MLV and NADC34-like strains, according to recombination analysis. Full-length Nsp2 sequence alignment indicated that all three isolates had a continuous deletion in 100 aa, which was similar to that of the IA/2014/NADC34. Based on phylogenetic analysis and the Nsp2 deletion pattern, the isolates are identified as NADC34-like PRRSV. Growth kinetic analysis based on primary porcine alveolar macrophages (PAMs) and MARC-145 cells has revealed that Korean NADC34-like PRRSV strains had better replication ability in primary PAMs than other Korean lineage 1 viruses or the reference strain VR2332, implying higher pathogenicity in pigs. Considering the fact that NADC34-like PRRSV strains were detected in MLV-vaccinated animals on a farm in the area of the highest pig population density in Korea, additional vaccine failure cases and the spread of the virus could be detected in the near future. Collectively, these results aid in comprehending the current epidemic state of PRRSVs in Korea and highlight the need for continuous surveillance.

Demyelination, the loss of the insulating sheath of neurons, causes failed or slowed neuronal conduction and contributes to the neurological symptoms in multiple sclerosis, traumatic brain and spinal cord injuries, stroke, and dementia. In demyelinated neurons, the axonal potassium channels Kv1.1 and Kv1.2, generally under the myelin sheath, become exposed and upregulated. Therefore, imaging these channels using positron emission tomography can provide valuable information for disease diagnosis and monitoring. Here, we describe the novel tracer for Kv1 channels [11C]3-methyl-4-aminopyridine ([11C]3Me4AP). [11C]3Me4AP was efficiently synthesized via Pd(0)-Cu(I) co-mediated Stille cross-coupling of a stannyl precursor containing a free amino group. Evaluation of its imaging properties in rats and nonhuman primates showed that [11C]3Me4AP has a moderate brain permeability and slow kinetics. Additional evaluation in monkeys showed that the tracer is metabolically stable and that a 1-tissue compartment model can accurately model the regional brain time-activity curves. Compared to the related tracers [18F]3-fluoro-4-aminopyridine ([18F]3F4AP) and [11C]3-methoxy-4-aminopyridine ([11C]3MeO4AP), [11C]3Me4AP shows lower initial brain uptake, which indicates reduced permeability to the blood-brain-barrier and slower kinetics, suggesting higher binding affinity consistent within vitrostudies. While the slow kinetics and strong binding affinity resulted in a tracer with less favorable properties for imaging the brain than its predecessors, these properties may make 3Me4AP useful as a therapeutic.

Resistance to hypomethylating agents (HMAs) in myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) is a concerning problem. Polo-like kinase 1 (PLK1) is a key cell cycle modulator and is known to be associated with an activation of the PI3K pathway, which is related to the stabilization of DNA methyltransferase 1 (DNMT1), a target of HMAs. We investigated the effects of volasertib on HMA-resistant cell lines (MOLM/AZA-1 and MOLM/DEC-5) derived from MOLM-13, and bone marrow (BM) samples obtained from patients with MDS (BM blasts5%) or AML evolved from MDS (MDS/AML). Volasertib effectively inhibited the proliferation of HMA-resistant cells with suppression of DNMTs and PI3K/AKT/mTOR and ERK pathways. Volasertib also showed significant inhibitory effects against primary BM cells from patients with MDS or MDS/AML, and the effects of volasertib inversely correlated with

Purpose: To analyze the difference in bony pelvimetry using magnetic resonance imaging (MRI) in women who delivered vaginally and those who underwent cesarean section (CS) due to cephalopelvic disproportion (CPD).Methods: This retrospective case-control study enrolled Korean women who gave birth to their first child between 37 and 42 weeks of pregnancy and underwent pelvic MRI between January 2014 and June 2020. These women were classified into 3 groups: vaginal delivery (VD), CS due to CPD (CPD-CS), and CS due to reasons other than CPD (Other-CS). In MRI images, true conjugate (TC), obstetric conjugate (OC), mid anterior-posterior (AP) distance (MAD), outlet AP distance (OAD), ischial spine distance (ISD), ischial tuberosity distance (ITD), and pubic angle (PA) were measured.Results: In total, 392 women were enrolled. There were no significant differences in maternal height and body mass index at birth among the 3 groups. The ISD was statistically different between the VD group (106.2±6.4 mm) and the Other-CS group (110.4±8.8 mm) (p=0.014). After multiple regression analysis, short ISD (adjusted odds ratio [aOR], 0.92; 95% confidence interval [CI], 0.87-0.97; p=0.002), short MAD (aOR, 0.95; 95% CI, 0.89-0.99; p=0.047), and short ITD (aOR, 0.95; 95% CI, 0.91-0.99; p=0.019) were significantly associated with CS due to CPD.Conclusion: Short ISD, MAD, and ITD, measured using MRI, are important parameters that can predict CS due to CPD. The accumulation of related research could promote the development of an excellent imaging method in terms of cost-effectiveness. Thus, it may be possible to establish a method to reduce obstetric and perinatal complications due to dystonia.

Surgical manipulation of paraganglioma can induce a massive release of catecholamines leading to hypertensive attack. But it has been not known about risk factors to cause hypertensive attack because paragangliomas of urinary bladder and retroperitoneum are notably rare tumors and have been recorded as case report or series. We investigated the relationship between mass size and hypertensive attack during surgery in patients with paraganglioma.Our retrospective chart review included 32 patients who had a pathological diagnosis of paraganglioma between March 2006 and May 2021, in single center. We analyzed the risk factors such as age, sex, height, weight, blood pressure before surgery, history of hypertension, pre-operative symptoms, mass location, and mass in 24 patients with retroperitoneal paragangliomas including urinary bladder. Hypertensive attack was defined as systolic blood pressure180 mmHg during excision of the mass from the electric medical chart. The predictive power was assessed by the area under the curve of the receiver operating characteristic curve.There were 19 retroperitoneal, 5 urinary bladder, 2 middle-ear cavity, 2 mediastinal, 2 neck, 1 spinal cord and 1 duodenal paraganglioma. Seven (29.2%) of the 24 patients had preoperative symptoms such as pain, fluctuation of blood pressure, and palpable mass. Hypertensive attack during surgery occurred in 11 patients (45.8%). There was a significant difference in mass size between groups with (n=11) and without (n=13) fluctuation of blood pressure (P=0.007). The area under curve for predicting surgical complications according to mass size was 0.808 (cutoff size 4.25 cm, sensitivity 72.7%, specificity 76.9%, 95% CI: 0.635-0.981).Mass size impacted occurrence of hypertensive attack during surgery in patients with retroperitoneal paraganglioma. Surgeons have to be watchful regarding of intraoperative hypertension during resection of retroperitoneal masses exceeding 4.25 cm, which are suspected as paraganglioma.

Metabotropic glutamate receptor 2 (mGluR2) has been extensively studied for the treatment of various neurological and psychiatric disorders. Understanding of the mGluR2 function is pivotal in supporting the drug discovery targeting mGluR2. Herein, the positive allosteric modulation of mGluR2 was investigated via the in vivo positron emission tomography (PET) imaging using 2-((4-(2-[11C]methoxy-4-(trifluoromethyl)phenyl)piperidin-1-yl)methyl)-1-methyl-1H-imidazo[4,5-b]pyridine ([11C]mG2P001).Distinct from the orthosteric compounds, pretreatment with the unlabeled mG2P001, a potent mGluR2 positive allosteric modulator (PAM), resulted in a significant increase instead of decrease of the [11C]mG2P001 accumulation in rat brain detected by PET imaging. Subsequent in vitro studies with [3H]mG2P001 revealed the cooperative binding mechanism of mG2P001 with glutamate and its pharmacological effect that contributed to the enhanced binding of [3H]mG2P001 in transfected CHO cells expressing mGluR2. The in vivo PET imaging and quantitative analysis of [11C]mG2P001 in non-human primates (NHPs) further validated the characteristics of [11C]mG2P001 as an imaging ligand for mGluR2. Self-blocking studies in primates enhanced accumulation of [11C]mG2P001 dose- and delivery-dependently. Altogether, these studies show that [11C]mG2P001 is a sensitive biomarker for mGluR2 expression and the binding is affected by the tissue glutamate concentration.

Objectives: This research aimed to develop a guideline for evaluating safety and performance of electronic warm-acupuncture apparatus. With the development of medical devices like electronic warm-acupuncture apparatus with improved performance, convenience and safety measures compared to traditional warm-acupuncture needling, safety and performance guideline is a necessity.Methods: By referring to existing standards and guidelines of other electronic devices for Korean medicine with heating function, guideline for safety and performance assessment of electronic warm-acupuncture apparatus was draftedResults: The guideline, presents explanation for adequate temperature and settings of the apparatus, and safety measurements providing against thermal runaway situations along with guidelines for the manual. Guideline for detailed test method for the performance of the apparatus such as accuracy of temperature increase and the timer, and safety unit was also provided. The test items and suggested test methods for the requirements of biological, electrical and electromagnetic safety were referred to Korean approval documents of ministry of Food and Drug Safety.Conclusion: We proposed the relevant items to verify performance and safety of warm-acupuncture apparatus to assure patient safety and improve the quality of currently developing devices for application in clinical field.

This study was investigated to evaluate the in vitro anti-diabetic activity and lipid accumulation inhibitory effect of the domestic walnut (Junglans regia, Gimcheon 1ho cultivar) in high glucose- and oleic acid-induced hepatic HepG2 cells. Eighty percent of ethanolic extract from Gimcheon 1ho (GC) showed higher inhibitory effects against α-amylase and α-glucosidase activities when compared to other ethanolic extracts. GC significantly inhibited the formation of advanced glycation end products (AGEs). The GC increased cell viability against H2O2- and high glucose-induced cell death determined by 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. GC reduced the formation of reactive oxygen species (ROS) in H2O2- and high glucose-induced HepG2 cells determined by 2’,7’-dichlorofluorescein diacetate (DCF-DA) assay. GC inhibited lipid accumulation in the oleic acid-induced HepG2 cells. The GC increased AMP-activated protein kinase (AMPK) phosphorylation and decreased the expression of lipid metabolism related proteins such as sterol regulatory element-binding proteins 1 (SREBP-1), fatty acid synthase (FAS), sterol regulatory element-binding protein 2 (SREBP-2) and 3-hydroxy-3-methyl-glutaryl-coenzyme A (HMGCR). The results suggest GC is a potentially functional food material which can improve nonalcoholic fatty liver disease by inhibiting lipid accumulation.

The benefits of physical activity (PA) participation are well documented, and yet many students with disabilities remain inactive. Approximately 19 % of U.S. undergraduate students have a disability and this is an 8 % increase since a decade ago. As the number of college/university students with disabilities (CUSD) is consistently increasing, it is important to provide equal opportunities for this population to be physically active. While inactivity raises health concerns for all college students, literature reveals that CUSD are less physically active than those without disabilities and/or CUSD may not know much about college PA opportunities on campus. This suggests that there might not be enough college PA programs available for CUSD. The purpose of this paper is to provide the importance of PA programs/courses for CUSD, college/university students without disabilities, and faculty. In addition, we will identify possible challenges to designing and implementing college PA programs for CUSD and strategies to better manage those challenges based on a review of the existing literature.

The strawberry growth and fruit yield of five Korean cultivars in the tunnel-type greenhouse predicted using their growth. The number of leaves, petiole length, leaf length and width, crown diameter, and the ratio of red and far-red (RFR) of the five Korean cultivars were measured during the cultivation period. The number of leaves of all cultivars exhibited a similar trend during this period; the plant and petiole length of ‘Maehyang’ were the longest, leaf length exhibited similar trends in all five cultivars except for ‘Jukhyang’, the leaf width of ‘Arihyang’, was the longest, and crown diameter of ‘Keumsil’ was the thickest. The leaf length, crown diameter, and RFR were associated with the fruit yield in the multiple linear regression. When a single model was used to predict the yield of all five cultivars, the correlation between expected yield and actual yield was r = 0.53. When cultivar-specific models were built for the prediction, the correlation increased to r = 0.77. The results indicated that the fruit yield of strawberry cultivars could be better predicted by considering cultivar-specific information, so it may be necessary to consider individual cultivars specifically rather than all cultivars simultaneously.

The study aimed to evaluate the morphologic changes in the pharyngeal airway after bimaxillary surgery in skeletal Class III malocclusion patients with or without asymmetry. We also analyzed the correlation between positional changes in the menton, hyoid bone, and changes in the dimensions of the pharyngeal airway.We included 32 patients with skeletal Class III malocclusion who underwent bimaxillary surgery. There were 16 subjects in the symmetry group (10 male, 6 female; mean age, 22.44 ± 5.14 years), and 16 in the asymmetry group (10 male, 6 female; mean age, 21.38 ± 4.62 years). Preoperative and postoperative (2 months after surgery) cone-beam computed tomography scans were taken and then analyzed by comparing postoperative changes in each group.The anteroposterior lengths at the uvula level (P2L) and epiglottis level (P3L) were significantly decreased in the symmetry group. The P2L was also significantly decreased in the asymmetry group, and a difference in the P3L was observed. However, there was no significant change in the width at the uvula level (P2W) and epiglottis level (P3W) in the symmetry group. In contrast, in the asymmetry group, P2W and P3W were significantly decreased. The cross-sectional ratio was significantly decreased P2 (P2L/P2W) and P3 (P3L/P3W) in the symmetry group. However, a statistically significant decrease occurred only at P3 in the asymmetry group. Anteroposterior positional changes of the menton and P2L (r = -0.370; P .05), P3L (r = -0.414; P 0.05), and P3L/P3W (r = -0.361; P 0.05) were correlated.Differences in the morphologic features of the pharyngeal airway after bimaxillary surgery was observed in both the symmetry and asymmetry groups. Bimaxillary surgery with a mandibular setback in patients with skeletal Class III malocclusion worsened morbidity of the elliptical structure of the pharyngeal airway. However, it worsened less in the asymmetry group than in the symmetry group.

The present study aimed to assess the effect of NO• on melanoma A375 cell growth and apoptotic cell death. Trypan blue exclusion assay was employed to detect the cytotoxicity induced by controlled steady-state concentrations (given in µM • min) of NO•. The characteristics of the cellular cell cycle and apoptosis in NO•-treated A375 cells were also analyzed by Annexin V/PI and DNA fragmentation assays. Western blotting was applied to detect the expression of apoptosis-related proteins (p53, Bax, Fas, DR5, caspase-3 and -9, and PARP). When exposed to preformed 100% NO• for 8 h reactor system, a cumulative dose of 3360 μM • min reduced the viability by 22% 24 h after treatment and promoted apoptosis, 2.9- and 12.2-folds 24 and 48 h after treatment higher than the argon control, respectively. Cell cycle analysis 48 h after treatment revealed S-phase arrest in cells treated with 3360 μM • min NO•. It was also observed that the expression of p53, DR5, caspase 9 and PARP increased significantly upon NO• treatment. In addition, the present study assessed the inhibitory effects of endogenous NO• on the proliferation of human melanoma cells by employing specific (AMG, 1400W and/or SMTC) and nonspecific (NMA) NO• synthase (NOS) inhibitors resulting in melanoma cell growth inhibition; the highest cytotoxic effect was seen when inducible NOS inhibition by 1 mM 1400W treatment. Collectively, the present data suggest that NO• is involved in a key mechanism limiting melanoma proliferation and apoptosis, which may play in improving the efficacy of melanoma treatment.

Studies on howmetabolic syndrome affects renal stone progression in untreated asymptomatic patients are lacking. Therefore, we investigated the effect of metabolic syndrome on changes in renal stone size.We retrospectively analyzed 820 patients with renal stones incidentally detected on CT during regular health examinations and who underwent follow-up CT evaluations for 1 year. The patients were divided into two groups according to the presence of metabolic syndrome. Changes in stone size during the follow-up were assessed, and differences were compared according to various factors. Predictors of stone size change on CT were assessed using linear regression analysis.Overall, 820 asymptomatic patients without a history of stone treatments and with a mean follow-up of 52.4 months were included. Of these, 104 (12.7%) had metabolic syndrome and 335 (40.9%) showed stone size increase during the follow-up. The stone size at diagnosis was not significantly different between patients with and without metabolic syndrome (225.3 ± 332.6 vs. 183.9 ± 310.2 mmMetabolic syndrome, fasting glucose level ≥ 100 mg/dl and young age are positively related to renal stone size changes. Therefore, periodic follow-up and metabolic syndrome management are required in asymptomatic patients with renal stones, especially in young age.

Purpose This study aims to introduce a deep neural network (DNN) to estimate the effective thermal conductivity of the flat heat pipe with spreading thermal resistance. Design/methodology/approach A total of 2,160 computational fluid dynamics simulation cases over up to 2,000 W/mK are conducted to regress big data and predict a wider range of effective thermal conductivity up to 10,000 W/mK. The deep neural networking is trained with reinforcement learning from 10–12 steps minimizing errors in each step. Another 8,640 CFD cases are used to validate. Findings Experimental, simulational and theoretical approaches are used to validate the DNN estimation for the same independent variables. The results from the two approaches show a good agreement with each other. In addition, the DNN method required less time when compared to the CFD. Originality/value The DNN method opens a new way to secure data while predicting in a wide range without experiments or simulations. If these technologies can be applied to thermal and materials engineering, they will be the key to solve thermal obstacles that many longing to overcome.

Background: Genotype 4 Eurasian avian-like lineage swine H1N1 viruses, which are reassortants containing the pandemic 2009 H1N1 virus lineage, triple reassortant lineage internal genes, and EA lineage external genes, have been reported in China since 2013. These have been predominant in pig populations since 2016 and have exhibited pandemic potential. Objectives: Examination of the circulation of G4 and its-related EA H1N1 viruses in Korea. Methods: We developed a one-step multiplex RT-qPCR that targeted M, HA1, and PB2 genes to detect G4 and its-related EA H1N1 viruses. We performed the analytical sensitivity and specificity of the multiplex RT-qPCR. The developed multiplex RT-qPCR was applied to swine respiratory samples between 2020 and 2022 in Korea. Results: Among the swine samples, a G4 EA H1N1 virus-related positive sample (6IP) was detected. Phylogenetic analysis based on all eight partial gene segments showed that 6IP contains nucleic acids of IAV segmented genes, which are closely related to G4 EA H1N1 viruses in HA, NA, PB2, NS, and NP genes. Conclusions: The developed multiplex RT-qPCR proved the sensitivity and specificity for the efficient surveillance of G4 and its-related EA H1N1 virus in clinical samples. Considering the phylogenetic analysis, additional screening for G4 EA H1N1 or its related viruses should be performed in future epidemiological studies.

The factors that influence the pathogenicity of African swine fever (ASF) are still poorly understood, and the host’s immune response has been indicated as crucial. Although an increasing number of studies have shown that gut microbiota can control the progression of diseases caused by viral infections, it has not been characterized how the ASF virus (ASFV) changes a pig’s gut microbiome. This study analyzed the dynamic changes in the intestinal microbiome of pigs experimentally infected with the high-virulence ASFV genotype II strain (N = 4) or mock strain (N = 3). Daily fecal samples were collected from the pigs and distributed into the four phases (before infection, primary phase, clinical phase, and terminal phase) of ASF based on the individual clinical features of the pigs. The total DNA was extracted and the V4 region of the 16 s rRNA gene was amplified and sequenced on the Illumina platform. Richness indices (ACE and Chao1) were significantly decreased in the terminal phase of ASF infection. The relative abundances of short-chain-fatty-acids-producing bacteria, such as Ruminococcaceae, Roseburia, and Blautia, were decreased during ASFV infection. On the other hand, the abundance of Proteobacteria and Spirochaetes increased. Furthermore, predicted functional analysis using PICRUSt resulted in a significantly reduced abundance of 15 immune-related pathways in the ASFV-infected pigs. This study provides evidence for further understanding the ASFV–pig interaction and suggests that changes in gut microbiome composition during ASFV infection may be associated with the status of immunosuppression.
