Wolfiporia cocos is commonly used as a traditional Chinese medicine for its diuretic, tonifying, and invigorating effects on the spleen. However, the epidermis of W. cocos is discarded as scrap during harvesting because of its low price, resulting in a great waste of resources and environmental pollution. In this work, the epidermis of W. cocos was studied and three new lanostane triterpenoids were isolated. The structures were determined using NMR and HRESIMS, with absolute configurations established by comparison of the calculated and experimental ECD spectra. The three new compounds were evaluated for their antimicrobial activities in vitro against Staphylococcus aureus, Escherichia coli, and Saccharomyces cerevisiae. None of the tested compounds showed inhibition against these three strains of indicator microbes at a concentration of 128 μg/ml. This study provides a reference for further medicinal development and the utilization of the epidermis of W. cocos.

Alpha-pinene is an important forest chemical resource, and the cis-pinane obtained by its hydrogenation reaction is used in spices, medicine, and other industries. A hollow nanospheric material with amphiphilic properties in a two-sided structure was prepared, with a nitrogen-doped carbon layer towards the inside and mesoporous silica in the outer layer of the nanospheres. The amphiphilic catalyst with non-precious nickel as the active component was synthesized by loading nickel onto the nanospheres using a simple impregnation method and applied to the aqueous phase hydrogenation reaction of α-pinene. The mesoporous structure of the catalyst shortened the mass transfer distance and thus reduced the mass transfer resistance. The nitrogen atoms doped in the carbon matrix provided anchor points for stabilizing the metal nanoparticles; the hydrophilic outer surface and hydrophobic hollow cavity enabled the catalyst to be well dispersed in water while still enriching the organic matter in aqueous solution. The effect of various reaction conditions on the catalytic reaction was investigated. The catalysts showed excellent activity and good stability, suggesting a new green and efficient method for more effective exploitation of biomass pine resin resources, and providing a reference for expanding the application of non-precious metal catalysts.

P-N junctions or heterostructures are commonly used in the fabrication of self-driven photodetectors because they serve critical roles in influencing the electrical characteristics of two-dimensional (2D) materials. For stable 2D heterostructures, planar WSe2 monolayers have attracted a lot of interest. Using first-principles energetic and dynamic calculations, we find that the transition from the indirect band gap of WSe2 bulk phase to the direct band gap of a two-dimensional plane is caused by the formation of p-n hetero junctions. Furthermore, it affects the carrier concentration transport mode and results in a significant orbital spin coupling. There is no virtual frequency in the phonon dispersion curve across the Brillouin zone at zero pressure. The interaction of the nearest neighbour atoms causes the frequency of the two acoustic modes of the G-point to be zero, as well as the frequency of the two optical films to merge. The theoretical analysis can provide support for the transport of two-dimensional planar carriers in electronics and photonics.

Development of solid oxide fuel cell (SOFC) anode with high resistance to coking and sulfur poisoning is highly desirable for the direct application of natural gas in SOFC. Herein, a (Cu, Sm)CeO2 anode with anchored Cu nanoparticles has been prepared. Most of Cu nanoparticles particle size ranges from 20 to 50 nm, which can increase the conductivity and catalytic activity of the anode. The Cu/CSCO10 supported cell exhibits a maximum power density of 404.6 mW/cm2 at 600 ⁰C when dry methane is used as fuel while its ohmic resistance is only 0.39 Ω·cm2. The single SOFC shows good stability when H2S content in the fuel is less than 150 ppm. Up to 900 hours of continuous stable operation with simulated natural gas and commercial natural gas as fuel prove the advantages and application potential of this anode.

Background: Primary gallbladder gastrinoma is an exceptionally uncommon tumor which is a rare form of neuroendocrine neoplasm. Until now, no cases of primary gallbladder gastrinoma and rare cases of primary gastrinoma from the biliary system have been reported. Case presentation: We reported a 50-year-old female with watery diarrhea and intermittently received proton pump inhibitors as treatment. Enhanced computed tomography (CT) imaging and Octreotide imaging uncovered a solitary tumor at the hepatic hilar region. The tumor was found originated from the wall of the gallbladder during the surgery and a subsequent laparoscopic cholecystectomy was exerted. Histological analysis revealed a primary neuroendocrine tumor from the gallbaldder. Conclusion: This case confirmed that primary gallbladder gastrinoma represents a distinct nosological entity. Immunohistochemical analysis plays a pivotal role in the diagnosis process. Due to limited knowledge regarding primary gallbladder gastrinoma, we aim to contribute valuable insights into this rare condition by providing unique information and present the therapeutic value of surgery.

Recent years have witnessed tremendous progresses in creating and manipulating ground-state ultracold polar molecules. However, the two-body loss regardless of the chemical reactivities is still a hurdle for many future explorations. Here, we investigate the loss suppression of non-reactive bosonic $^{23}$Na$^{87}$Rb molecules with a circular polarized microwave blue-detuned to the rotational transition. We achieve suppression of the loss by two orders of magnitude with the lowest two-body loss rate coefficient reduced to $3\times10^{-12}~\rm{cm^3/s}$. Meanwhile, the elastic collision rate coefficient is increased to the $10^{-8}~\rm{cm^3/s}$ level. The large good-to-bad collision ratio has allowed us to carry out evaporative cooling of $^{23}$Na$^{87}$Rb with an efficiency of 1.7(2), increasing the phase-space density by a factor of 10. With further improvements, this technique holds great promises for creating a Bose-Einstein condensate of ultracold polar molecules., 11 pages, 8 figures. BACKGROUND: Pulmonary carcinoids (PC), including typical (TC) and atypical carcinoids (AC), are low-grade neuroendocrine tumors (NETs) which account for 1–5% of all lung tumors. Due to the low prevalence of PC and extreme rarity of anaplastic lymphoma kinase (ALK) rearrangements in patients with PC, the advances in targeted therapy development in PC are still limited and there is no standard treatment. Even though in patients with PC harboring ALK rearrangements there is a room for a success in targeted therapy. To our knowledge, case 1 was the first report to detect ALK gene p.I1171N mutation after taking alectinib and sensitive to ceritinib in patients with atypical carcinoid. CASE DESCRIPTION: Herein, we report the cases of 2 non-smoking patients, 51 year-old female with tumor in left lower lobe and 49 year-old female with tumor in right upper lobe, both with metastatic PC who harbored EML4-ALK fusion and were sensitive to small-molecule ALK inhibitors. The first patient initially received alectinib, then therapy was switched to ceritinib after developing drug resistance due to the missense mutation of ALK gene p.I1171N mutation in exon 22 detected by next-generation sequencing (NGS), and finally died of intracranial disease progression. The second patient also received alectinib, and her treatment is currently ongoing with good effect and tolerance. After conducting comprehensive review of literature, we found that 14 lung NETs with ALK rearrangements have been reported to date. The clinical outcome was partial response for 6 NETs patients and 5 patients exhibited stable disease after treatment with ALK inhibitors. CONCLUSIONS: According to the effectiveness of ALK inhibitors in our cases and previous articles, we recommend alectinib for the first-line treatment of metastatic PC with EML4-ALK fusion and highlight the need for molecular profiling of metastatic lung NETs patients and that ALK inhibitors are feasible in the treatment for metastatic lung NETs patients with ALK rearrangements. Finally, further studies to assess the real prevalence of ALK gene fusions and their spectrum of sensitivity to different ALK inhibitors are needed in larger cohorts.

Gradually aggravated disease caused by phytopathogenic bacteria severely restricts food security and crop yield, and few pesticides can relieve this severe situation. Thus, development and excavation of new agrochemicals with high bioactivity and novel action mechanism may be a feasible strategy to control intractable bacterial diseases. As a privileged molecular framework, steroid molecules exhibit diversiform bioactivities. Herein, a series of novel androst-4-ene derivatives were designed, synthesised and investigated for their antibacterial behaviour to excavate novel agrochemicals on the base of steroid molecules.Bioassay results indicated that target compounds displayed high bioactivities toward three destructive phytopathogenic bacteria, including Xanthomonas oryzae pv. oryzae (Xoo), Xanthomonas axonopodis pv. citri (Xac) and Pseudomonas syringae pv. actinidiae (Psa). Compound IIIGiven these profiles of target compounds, we highlight a new strategy for controlling intractable plant bacterial diseases by inducing plant resistance and targeting the bacterial cell membrane. © 2022 Society of Chemical Industry.

By theoretical calculation, the crystal structure, stability, and electron-phonon interactions of calcium dinitride (CaN2) have been described, and CaN2 is predicted to be a superconducting material. The action mechanism of thermodynamics about CaN2 are induced through the Debye-Grüneisen model. The dispersion curves of CaN2 show large pseudo-gaps in the 400–1300 cm−1 range. The total of the electron-phonon coupling constants for CaN2 is given by λ = 0.37, with a corresponding estimated superconducting temperature of Tc ≈ 1 K. The estimated superconducting temperature of CaN2 rose to 28.8 K as the pressure increased to 10 GPa, and then decreased monotonically at higher pressures to almost zero at 50 GPa.

Background: As the complexity and diversity of the tumor immune microenvironment (TIME) are becoming better understood, burgeoning researches got progress in this field. This study sought to investigate the development pattern of TIME-related research in recent years from a bibliometric perspective. Methods: We acquired both articles and reviews related to TIME from the Web of Science Core Collection (WoSCC) (retrieved on September 14, 2022). R package “Bibliometrix” was used to calculate the basic bibliometric features, present the collaborative conditions of countries and authors and generate a three-field plot to show the relationships among authors, affiliations, and keywords. VOSviewer was utilized for co-authorship analysis of country and institution and keyword co-occurrence analysis. CiteSpace was used for citation burst analysis of keywords and cited references. Results: A total of 2545 publications on TIME were included. China, the USA, and Japan had the highest number of publications. Frontiers in Oncology had the highest number of publications. Apart from “expression,” “cancer,” and “cells,” the terms “survival,” “immunotherapy,” and “T-cells” were coequally the most frequently mentioned keywords. Conclusions: This bibliometric study analyzed 16 years of TIME-related research and sketched out a basic knowledge framework that includes publications, countries, journals, authors, institutions, and keywords. The results provided a thorough overview on the broad landscape of this research area.

Renal fibrosis (RF) is the common pathological manifestation of virtually all chronic kidney diseases (CKD) and one of the major causes of end-stage renal disease (ESRD), but the pathogenesis of which is still unclear. Renal tubulointerstitial lesions have been identified as a key pathological hallmark of RF pathology. Renal tubular epithelial cells are the resident cells of the tubulointerstitium and play an important role in kidney recovery versus renal fibrosis following injury. Studies in recent years have shown that senescence of renal tubular epithelial cells can accelerate the progression of renal fibrosis. Oxidative stress(OS), telomere attrition and DNA damage are the major causes of renal tubular epithelial cell senescence. Current interventions and therapeutic strategies for cellular senescence include calorie restriction and routine exercise, Klotho, senolytics, senostatics, and other related drugs. This paper provides an overview of the mechanisms and the key signaling pathways including Wnt/β-catenin/RAS, Nrf2/ARE and STAT-3/NF-κB pathway involved in renal tubular epithelial cell senescence in RF and therapies targeting renal tubular epithelial cell senescence future therapeutic potential for RF patients. These findings may offer promise for the further treatment of RF and CKD.

Elemental fragmentation cross sections (EFCSs) of stable and unstable nuclides have been investigated with various projectile-target combinations at a wide range of incident energies. These data are critical to constrain and develop the theoretical reaction models and to study the propagation of galactic cosmic rays (GCR). In this work, we present a new EFCS measurement for $^{28}$Si on carbon at 218~MeV/nucleon performed at the Heavy Ion Research Facility (HIRFL-CSR) complex in Lanzhou. The impact of the target thickness has been well corrected to derive an accurate EFCS. Our present results with charge changes $\Delta Z$ = 1-6 are compared to the previous measurements and to the predictions from the models modified EPAX2, EPAX3, FRACS, ABRABLA07, NUCFRG2, and IQMD coupled with GEMINI (IQMD+GEMINI). All the models fail to describe the odd-even staggering strength in the elemental distribution, with the exception of the IQMD+GEMINI model, which can reproduce the EFCSs with an accuracy of better than 3.5\% for $\Delta Z\leq5$. The IQMD+GEMINI analysis shows that the odd-even staggering in EFCSs occurs in the sequential statistical decay stage rather than in the initial dynamical collision stage. Background/Aims The activation of the complement system and subsequent inflammatory responses are important features of myocardial ischemia/reperfusion (I/R) injury. Exosomes are nanoscale extracellular vesicles that play a significant role in remote ischemic preconditioning (RIPC) cardioprotection. The present study aimed to test whether RIPC-induced plasma exosomes (RIPC-Exo) exert protective effects on myocardial I/R injury by inhibiting complement activation and inflammation and whether exosomal heat shock protein 90 (HSP90) mediates these effects. Methods Rat hearts underwent 30 min of coronary ligation followed by 2 h of reperfusion. Plasma exosomes were isolated from RIPC rats and injected into the infarcted myocardium immediately after ligation. Sixty rats were randomly divided into Sham, I/R, I/R + RIPC-Exo (50 µg/µl), and RIPC-Exo + GA (geldanamycin, 1 mg/kg, administration 30 min before ligation) groups. Cardiomyocyte apoptosis, the release of myocardial markers (LDH, cTnI and CK-MB), infarct size, the expression of HSP90, complement component (C)3, C5a, c-Jun N-terminal kinase (JNK), interleukin (IL)-1β, tumor necrosis factor (TNF)-alpha and intercellular adhesion molecule -1 (ICAM-1) were assessed. Results RIPC-Exo treatment significantly reduced I/R-induced cardiomyocyte apoptosis, the release of myocardial markers (LDH, cTnI and CK-MB) and infarct size. These beneficial effects were accompanied by decreased C3 and C5a expression, decreased inflammatory factor levels (IL-1β, TNF-α, and ICAM-1), decreased JNK and Bax, and increased Bcl-2 expression. Meanwhile, the expression of HSP90 in the exosomes from rat plasma increased significantly after RIPC. However, treatment with HSP90 inhibitor GA significantly reversed the cardioprotection of RIPC-Exo, as well as activated complement component, JNK signalling and inflammation, indicating that HSP90 in exosomes isolated from the RIPC was important in mediating the cardioprotective effects during I/R. Conclusion Exosomal HSP90 induced by RIPC played a significant role in cardioprotection against I/R injury, and its function was in part linked to the inhibition of the complement system, JNK signalling and local and systemic inflammation, ultimately alleviating I/R-induced myocardial injury and apoptosis by the upregulation of Bcl-2 expression and the downregulation of proapoptotic Bax.

Ultracold polyatomic molecules offer intriguing new opportunities in cold chemistry, precision measurements, and quantum information processing, thanks to their rich internal structure. However, their increased complexity compared to diatomic molecules presents a formidable challenge to employ conventional cooling techniques. Here, we demonstrate a new approach to create ultracold polyatomic molecules by electroassociation in a degenerate Fermi gas of microwave-dressed polar molecules through a field-linked resonance. Starting from ground state NaK molecules, we create around $1.1\times 10^3$ tetratomic (NaK)$_2$ molecules, with a phase space density of $0.040(3)$ at a temperature of $134(3)\,\text{nK}$, more than $3000$ times colder than previously realized tetratomic molecules. We observe a maximum tetramer lifetime of $8(2)\,\text{ms}$ in free space without a notable change in the presence of an optical dipole trap, indicating these tetramers are collisionally stable. The measured binding energy and lifetime agree well with parameter-free calculations, which outlines pathways to further increase the lifetime of the tetramers. Moreover, we directly image the dissociated tetramers through microwave-field modulation to probe the anisotropy of their wave function in momentum space. Our result demonstrates a universal tool for assembling ultracold polyatomic molecules from smaller polar molecules, which is a crucial step towards Bose--Einstein condensation (BEC) of polyatomic molecules and towards a new crossover from a dipolar Bardeen-Cooper-Schrieffer (BCS) superfluid to a BEC of tetramers. Additionally, the long-lived FL state provides an ideal starting point for deterministic optical transfer to deeply bound tetramer states.

Optical mirrors determine cavity properties by means of light reflection. Imperfect reflection gives rise to open cavities with photon loss. We study an open cavity made of atom-dimer mirrors with a tunable reflection spectrum. We find that the atomic cavity shows anti-$\mathcal{PT}$ symmetry. The anti-$\mathcal{PT}$ phase transition controlled by atomic couplings in mirrors indicates the emergence of two degenerate cavity supermodes. Interestingly, a threshold of mirror reflection is identified for realizing strong coherent cavity-atom coupling. This reflection threshold reveals the criterion of atomic mirrors to produce a good cavity. Moreover, cavity quantum electrodynamics with a probe atom shows mirror-tuned properties, including reflection-dependent polaritons formed by the cavity and probe atom. Our work presents a non-Hermitian theory of an anti-$\mathcal{PT}$ atomic cavity, which may have applications in quantum optics and quantum computation., Comment: 7 pages, 5 figures

Diffuse large B-cell lymphoma (DLBCL) is the most prevalent subtype of non-Hodgkin lymphoma and is a very aggressive malignancy with tumor growing rapidly in organs like lymph nodes. The pathogenesis of DLBCL is not clear and the prognosis of DLBCL requires improvement. Here, we investigated the mechanisms of DLBCL, with the focus on lncRNA PVT1/miR-34b-5p/Foxp1 axis. Human DLBCL tissues from diagnosed DLBCL patients and four human DLBCL cell lines, one normal human B lymphoblastoid cell line were used. qRT-PCR and western blotting were employed to measure expression levels of lncRNA PVT1, Foxp1, miR-34b-5p, β-catenin, and proliferation-related proteins. MTT assay and colony formation assay were performed to determine cell proliferation. Flow cytometry was used to examine cell apoptosis. ChIP and Dual-luciferase assay were utilized to validate interactions of Foxp1/promoters, PVT1/miR-34b-5p and miR-34b-5p/Foxp1. Mouse tumor xenograft model was used to determine the effect of sh-PVT1 on tumor growth in vivo. In this study, we found PVT1 and Foxp1 were elevated in DLBCL tissues and cells while miR-34b-5p was decreased. Knockdown of PVT1, overexpression of miR-34b-5p, or Foxp1 knockdown repressed DLBCL cell proliferation but enhanced cell apoptosis. PVT1 directly bound miR-34b-5p to disinhibit Foxp1/β-catenin signaling. Foxp1 regulated CDK4, CyclinD1, and p53 expression via binding with their promoters. Knockdown of Foxp1 partially reversed the effects of miR-34b-5p inhibitor on DLBCL cell proliferation and apoptosis. Inhibition of PVT1 through shRNA suppressed DLBCL tumor growth in vivo. All in all, lncRNA PVT1 promotes DLBCL progression via acting as a miR-34b-5p sponge to disinhibit Foxp1/β-catenin signaling.

A photo-switchable hetero-complementary quadruple H-bonding array, which consists of an azobenzene-derived ureidopyrimidinone (UPy) module (Azo-UPy) and a nonphotoactive diamidonaphthyridine (DAN) derivative (Napy-1), is constructed based on a reversible photo-locking approach. Upon UV (390 nm)/Vis (460 nm) light irradiations, photo-switchable quadruple H-bonded dimerization between Azo-UPy and Napy-1 can be achieved with exhibiting 4.8×104 -fold differences in binding strength (ON/OFF ratios). Furthermore, smart polymeric gels with unique photo-controlled macroscopic self-assembly behavior can be fabricated by introducing such quadruple H-bonding array as photo-regulable noncovalent interfacial connections.

The transition from glacial to interglacial periods has been hypothesized to be linked to millennial-scale changes in oceanic/atmospheric circulation, but the relationships between these phenomena remain poorly constrained. Here we present a speleothem oxygen isotope record from Yongxing Cave, China, spanning 40.9 to 33.1 ka and compare this to existing Antarctic proxy records. We find that decadal-to-centennial rapid shifts in the Asian summer monsoon, Antarctic temperature, atmospheric methane and carbon dioxide are all coupled together during Dansgaard–Oeschger cycles, which may suggest an important role of the Intertropical Convergence Zone and Southern Ocean in driving the global greenhouse gas changes. Analogous to millennial-scale variations in trend, amplitude and internal sub-centennial-scale structures during Dansgaard–Oeschger 8 and Heinrich Stadial 4, the Younger Dryas and Heinrich Stadial 1 during the last ice termination provided critical positive feedbacks to changes in terrestrial vegetation and northern ice volume, and may have contributed to glacial to interglacial transition.

In addition to “immune-related adverse events”, “immune checkpoint inhibitors”, “immunotherapy”, and “nivolumab” were the most frequently used keywords.ConclusionsA bibliometric analysis of 17 years of irAEs research was conducted to map a basic knowledge structure including countries, institutions, authors, journals, and publications. The findings provided a comprehensive perspective on the broad future of this research area.

Background CD4+ T cell counts in certain human immunodeficiency virus (HIV)-infected patients called immunological non-responders (INRs) could not return to a normal level even with sustained antiretroviral therapy (ART) because of persistent immune activation, which is associated with pro-inflammatory cytokines production and an altered intestinal microbiome profile. Changes in gut bacterial composition have been linked to low CD4+ T cell counts in HIV-infected individuals. However, the association between CD4+ T cell counts and gut microbiota community composition and cytokines levels in INRs (CD4+ T cell counts Methods To address this issue, we carried out a cross-sectional study of 34 HIV-infected INRs. The patients were divided into CD4 count > 200 cells/μL group and CD4 count 16S rRNA gene sequencing. We also compared CD8+ T cell counts, pro-inflammatory cytokines levels, and nutritional status between the two groups. Results Compared to INRs with CD4 count > 200 cells/μL, those with CD4 count Ruminococcaceae was less abundant in the CD4 count 200 cells/μL group, and difference in alpha diversity was observed between the two groups. Moreover, CD4+ T cell counts were negatively associated with TNF-α and IL-1α levels and positively associated with the relative abundance of Ruminococcaceae. Conclusions Our study demonstrated that lower CD4+ T cell counts in INRs are associated with a reduced abundance of Ruminococcaceae in the gut and elevated serum pro-inflammatory cytokines levels. Thus, interventions targeting gut microbiota to increase CD4+ T cell counts are a potential strategy for promoting immune reconstitution in HIV-infected INRs.

Background: Microgrid is an effective integration mode of distributed generation (DG). It can eliminate the negative effect caused by the intermittent DG and consume the energy locally. Methods: For a public distribution network, a microgrid performs as a controllable and flexible unit. However, when the public distribution network has the demand of flexible regulation, there is no reasonable quantitative indexes to evaluate the flexibility of microgrid for dispatching decision. For this problem, the operational flexibility indexes of a microgrid are defined in this paper. Results: The indexes are quantified according to the maximum energy and active power regulation margin of each flexible equipment in the microgrid. The operational flexibility of microgrid is closely related to the scheduling strategy. Therefore the corresponding dispatching model of a microgrid is also provided in this paper. Conclusion: A case study shows how to calculate and use these flexibility indexes in an operation cycle of a microgrid system with energy storage (ES) facility and demonstrates the availability and practicability of the evaluation indexes and the method.

Pine wilt disease has caused huge losses in ecological and economic values in China, especially in the southern parts. We develop a non-Gaussian variational approach that enables us to study both equilibrium and far-from-equilibrium physics of the two-dimensional Fermi polaron. This method provides an unbiased analysis of the polaron-to-molecule phase transition without relying on truncations in the total number of particle-hole excitations. Our results -- which include the ground state energy and quasiparticle residue -- are in qualitative agreement with the known Monte Carlo calculations. The main advantage of the non-Gaussian states compared to conventional numerical methods is that they enable us to explore long-time polaron evolution and, in particular, study various spectral properties accessible to both solid-state and ultracold atom experiments. We design two types of radiofrequency spectroscopies to measure polaronic and molecular spectral functions. Depending on the parameter regime, we find that these spectral functions and fermionic density profiles near the impurity display either long-lived oscillations between the repulsive and attractive polaron branches or exhibit fast relaxational dynamics to the molecular state., 12 pages, 7 figures

BackgroundCardiovascular complications in patients with acute upper gastrointestinal bleeding (AUGIB) have been associated with a high-risk of subsequent adverse consequences. This study aimed to analyze the risk factors for myocardial injury in AUGIB patients, predict the risk of myocardial injury, and explore the clinical prognosis and influencing factors in AUGIB patients with myocardial injury.Materials and methodsA retrospective case-control study based on AUGIB patients in the First Affiliated Hospital of Xi’an Jiaotong University from 2016 to 2020 was performed. We divided the enrolled patients into a myocardial injury group and a control group according to whether they developed myocardial injury. The variables significant in the univariate analysis were subjected to binary logistic regression for risk factor analysis and were used to establish a nomogram for predicting myocardial injury. In addition, logistic regression analysis was performed to better understand the risk factors for in-hospital mortality after myocardial injury.ResultOf the 989 AUGIB patients enrolled, 10.2% (101/989) developed myocardial injury. Logistic regression analysis showed that the strong predictors of myocardial injury were a history of hypertension (OR: 4.252, 95% CI: 1.149–15.730, P = 0.030), blood urea nitrogen (BUN) (OR: 1.159, 95% CI: 1.026–1.309, P = 0.018) and left ventricular ejection fraction (LVEF) P = 0.037). The patients with a tumor history (digestive system tumors and non-digestive system tumors) had no significant difference between the myocardial injury group and the control group (P = 0.246). A prognostic nomogram model was established based on these factors with an area under the receiver operator characteristic curve of 0.823 (95% CI: 0.730–0.916). The patients with myocardial injury had a much higher in-hospital mortality rate (10.9% vs. 2.0%, P < 0.001), and an elevated D-dimer level was related to in-hospital mortality among the AUGIB patients with myocardial injury (OR: 1.273, 95% CI: 1.085–1.494, P = 0.003).ConclusionA history of hypertension, renal dysfunction, and cardiac function with LVEF

Background Patients with non-alcoholic fatty liver disease (NAFLD) may be more susceptible to coronavirus disease 2019 (COVID-19) and even more likely to suffer from severe COVID-19. Whether there is a common molecular pathological basis for COVID-19 and NAFLD remains to be identified. The present study aimed to elucidate the transcriptional alterations shared by COVID-19 and NAFLD and to identify potential compounds targeting both diseases. Methods Differentially expressed genes (DEGs) for COVID-19 and NAFLD were extracted from the GSE147507 and GSE89632 datasets, and common DEGs were identified using the Venn diagram. Subsequently, we constructed a protein–protein interaction (PPI) network based on the common DEGs and extracted hub genes. Then, we performed gene ontology (GO) and pathway analysis of common DEGs. In addition, transcription factors (TFs) and miRNAs regulatory networks were constructed, and drug candidates were identified. Results We identified a total of 62 common DEGs for COVID-19 and NAFLD. The 10 hub genes extracted based on the PPI network were IL6, IL1B, PTGS2, JUN, FOS, ATF3, SOCS3, CSF3, NFKB2, and HBEGF. In addition, we also constructed TFs–DEGs, miRNAs–DEGs, and protein–drug interaction networks, demonstrating the complex regulatory relationships of common DEGs. Conclusion We successfully extracted 10 hub genes that could be used as novel therapeutic targets for COVID-19 and NAFLD. In addition, based on common DEGs, we propose some potential drugs that may benefit patients with COVID-19 and NAFLD.

Objective To summarize the clinical, muscle pathology and molecular biological features of late⁃onset glycogen storage disease type Ⅱ (GSDⅡ). Methods and Results Five patients with late⁃onset GSD Ⅱ diagnosed and treated in The Fifth Affiliated Hospital of Zhengzhou University from January 2013 to January 2020 were selected. The main clinical manifestations of 5 patients were weakness of raising the head, weakness of the proximal extremities, decreased muscle tone, fatigue intolerance, and dyspnea. The activity of acid α⁃glucosidase (GAA) was significantly reduced. HE staining of muscular tissues in 5 patients showed vacuole⁃like changes of different sizes, different numbers, and irregular shapes in most muscle fibers. Modified Gomori trichrome (MGT) staining showed a large number of blue and purple particles deposited in the vacuole. Periodic acid⁃Schiff (PAS) staining showed the glycogen content in the vacuoles were increased in 4 cases, and the glycogen content in the vacuoles were lost in one case. GAA gene testing showed that 9 mutations were detected in 5 patients, and 4 cases were compound heterozygous mutations, which were derived from father and mother respectively. The c.1320_1322delGAT (p.Met440del) was a deletion mutation, c.2331G＞C (p.Thr777Thr) was a synonymous mutation, c.2237G＞A (p.Trp746*) was a nonsense mutation, c.877G＞A (p.Gly293Arg) was a missense mutation, c.2238G＞C (p.Trp746Cys) was a missense mutation, c.784G＞A (p.Glu262Lys) was a missense mutation, c.2014C＞T (p.Arg672Trp) was a missense mutation, and c. 2332⁃2A＞G was a splicing mutation. One case was homozygous mutation of c. 1432G＞A (p. Gly478Arg), which originated from the mother. Among them, c. 2331G＞C, c. 1432G＞A and c. 2332⁃2A＞G were reported for the first time at home and abroad. Conclusions The clinical manifestations of late⁃onset GSD Ⅱ are weakness of proximal limbs and dyspnea. The activity of GAA in peripheral serum is decreased significantly. Muscle tissue pathology is characteristic. GAA gene mutations are mainly compound heterozygous mutation, c.2331G＞C, c.1432G＞A and c.2332⁃2A＞G are new mutations, which extended the mutation spectrum of GAA gene.
