BACKGROUND AND OBJECTIVE: Acne vulgaris is one of the most common skin diseases that imposes too much mental pressure and high costs on patients. The existing treatments have low efficacy and include antibiotics and anti-inflammatory drugs, which have many complications due to the chronic nature of the disease, especially at young age, including antibiotic resistance and allergic reactions. For this reason, one of the new therapeutic approaches is the use of a vaccine with the help of the bacterium or its components. The aim of this study is to produce nanoparticles carrying the recombinant CAMP – Sialidase protein as a new chimeric antigen to be used in acne vaccine. METHODS:To express the recombinant CAMP – Sialidase protein, E. coli BL21 DE3 was used as the host. Purification of protein was done through combined urea/imidazole method and using a nickel-nitroacetic acid column. The recombinant protein was confirmed using Western Blotting by Anti – Histidine Antibody. Then, the loaded nanoparticles were prepared by recombinant protein using ionic gelation technique and tripolyphosphate. Finally, the size and zeta potential of the nanoparticles were determined by the DLS device. FINDINGS: The recombinant CAMP – Sialidase protein was confirmed after expression and purification. The size and zeta potential of nanoparticles containing recombinant CAMP – Sialidase protein at a concentration of 0.6 mg / ml were determined to be 80 nm and +27 mV, respectively, using the DLS device. The loading rate of the protein in the nanoparticles was found to be 88%. CONCLUSION: The results show that the recombinant protein is expressed completely and is successfully encapsulated in the chitosan nanoparticles.

Because vitamin D deficiency and as well as the prevalence of allergy is the important issues in the health field, this study was aimed to investigate the relationship between vitamin D level and and IgE in children in Tehran. Background and aims: Vitamin D deficiency is a common problem of the world that has many complications, especially in adults. Studies suggest a relationship between low levels of vitamin D and allergic diseases. Regarding to the role of vitamin D in the regulation of immune system, Vitamin D deficiency can be one of the effective factors in the development and exacerbation of allergic diseases. Hence, the present study was aimed to examine the relationship between vitamin D levels and the amount of IgE in adults with allergies (type 1) in Tehran. Methods: In a cross-sectional study, 80 people with allergies (28 males and 52 females) older than 16 years old referred to the Khorshid Allergy and immunology Clinic were evaluated. Vitamin D and total IgE levels were determined by ELISA. Three levels of vitamin D deficiency (less than 20 ng/ml), insufficient (30-20 ng/ml) and sufficient (more 30 than ng/ml) were classified. In order to confirm the presence of specific IgE, skin prick test was used. Finally, the data were analyzed by SPSS software version 20. Results: The mean level of vitamin D in the all subjects was 21.59 ng/ml that fall into insufficient range (less than 30 ng/ml). Based on the results, the abnormal level of vitamin D was present in 76.25% (65% insufficient and 11.25% deficient) and only 23.75% of them had sufficient vitamin D levels. The results also showed abnormal level of vitamin D in women was more than men (67.21% to 32.78%). In addition, vitamin D deficiency was significantly associated with female group (P=0.01). Conclusion: Because of the prevalence of vitamin D deficiency, identification of effective factors on vitamin D deficiency and also its impact on the incidence of allergic diseases, it seems necessary to study vitamin D deficiency and how to prevent it.

Sulfur mustard (SM, bis- (2-chloroethyl) sulphide) is a chemical warfare agent that causes DNA alkylation, protein modification and membrane damage. SM can trigger several molecular pathways involved in inflammation and oxidative stress, which cause cell necrosis and apoptosis, and loss of cells integrity and function. Epigenetic regulation of gene expression is a growing research topic and is addressed by DNA methylation, histone modification, chromatin remodeling, and noncoding RNAs expression. It seems SM can induce the epigenetic modifications that are translated into change in gene expression. Classification of epigenetic modifications long after exposure to SM would clarify its mechanism and paves a better strategy for the treatment of SM-affected patients. In this study, we review the key aberrant epigenetic modifications that have important roles in chronic obstructive pulmonary disease (COPD) and compared with mustard lung.

Objectives: Enterotoxigenic Escherichia coli (ETEC) is one of the most common agents of diarrhea among other bacterial agents. Designing and producing vaccine against these bacteria is one of the major purposes of World Health Organization (WHO). Due to presence of diverse clones of ETEC strains in the world, the use of global vaccines for ETEC infection is controversial. B subunit of heat labile toxin (LTB) was introduced as a vaccine candidate molecule by several investigators. The expression of LTB gene isolated from a local bacterial strain and investigation of its immunological property was the objective of this study. Materials and Methods: LTB gene was isolated from a local isolated ETEC, cloned and expressed using pET28a expression vector. For LTB gene expression, the three main expression parameters (IPTG concentration, time and temperature of induction) were investigated. The recombinant protein was purified ( > 95%) with Ni-NTA column using 6XHis-tag and used as an antigen in ELISA test. Results: The immunological analyses showed production of high titer of specific antibody in immunized mice. Anti LTB Antibody could bind to whole toxin and neutralize the toxin through inhibition of its binding to the Ganglioside M1 receptor. Conclusion: The recombinant LTB protein is a highly immunogenic molecule. Considering the LTB role in ETEC pathogenesis, it can be taken into account as one of the most important components of vaccines against local ETEC.
