Astroviruses are considered the cause of gastroenteritis in humans and animals. Studies in recent years show avian astroviruses are also associated with duckling hepatitis, gosling gout, and chicken nephritis. In this study, a GAstV strain, designated as JS2019/China, was detected in dead goslings from a commercial goose farm in Jiangsu province of China. Viral strain was proliferated in goose embryos and sequence analysis showed the isolated strain had a classical structure arrangement and a series of conserved regions compared with other GAstVs. Sequence comparison and phylogenetic analysis of whole genome and ORF2 revealed that JS2019/China belongs to the GAstV-1 group, which consists of most of the GAstV strains. Amino acid analysis indicated that some mutants might have an impact on viral protease capacity, such as V505I and K736E of ORF1a and T107I, F342S, and S606P of ORF2. Taken together, a novel GAstV strain was isolated and genomic analysis and protein polymorphism analysis indicated that some amino acid mutants might affect the viral virulence.

Objective: To investigate the prognostic value of DNA methylation of tumor suppressor genes for hepatocellular carcinoma (HCC) recurrence after liver transplantation. Methods: APC gene was selected according to The Cancer Genome Atlas dataset. Tumor tissues and clinical data of 85 HCC patients who received a liver transplantation were retrospectively enrolled and next-generation methylation sequencing was performed. Risk factors were determined using the Cox proportional-hazard-regression model. Results: The APC methylation site (chromosome 5, position 112043544) was an independent predictor of post-transplant HCC recurrence. Patients with hyper-methylated APC112043544 experienced superior recurrence-free survival (p = 0.021) and had a decreased proportion of microvascular invasion (p = 0.017). APC112043544 also predicted recurrence risk in patients beyond selection criteria. Conclusions: APC112043544 methylation may serve as a potential biomarker for post-transplant HCC recurrence.

6. MIIP downregulation promotes colorectal cancer progression via inducing adjacent adipocytes browning

- Abstract
The enrichment of peri-cancerous adipocytes is one of the distinctive features of colorectal cancer (CRC), which accelerates the disease progression and worsens the prognosis. The communication between tumor cells and adjacent adipocytes plays an important role in this process. However, the detailed mechanisms remain largely unknown. Here we demonstrated MIIP is downregulated in high-grade CRC, and revealed its role and mechanism in tumor cell-adipocyte communication. By detecting MIIP expression in CRC tissues and adjacent normal tissues, we found MIIP was significantly decreased in CRC, and was closely related to adjacent adipocytes browning. In detail, MIIP reduction altered the N-linked glycosylation modification of AZGP1 and thus alleviated the inhibition of secretion. AZGP1, a critical lipid mobilization factor, led to the intensification of adipocytes browning and the release of free fatty acids (FFAs), which in turn fueled for CRC progression. Our data demonstrate that MIIP plays a key regulatory role in the communication between CRC and neighboring adipocytes by mediating AZGP1 secretion, and MIIP reduction leads to adipose browning-induced cancer rapid progression and poor prognosis.

Bamboo and wood are widely used as food contact materials (FCMs). Fungicides are often applied during the manufacture of these products to prevent mildew; thus, monitoring for fungicide residues is essential because of the possible risks to food safety. However, it is not clear whether residual fungicides in treated bamboo and wood products migrate into food to cause harm to humans, and there is an absence of procedures for these analyses. In this study, the simultaneous determination of fungicides was validated in bamboo and wood FCMs by high-performance liquid chromatography–triple quadrupole tandem mass spectrometry (HPLC–MS/MS). The analytes were extracted with methanol or 1:1 methanol/acetonitrile and purified using 25 mg primary secondary amine adsorbent with 2.5 mg graphitized carbon black or 25 mg PSA with 25 mg octadecylsilane (C18). The recoveries for wood and bamboo FCMs were from 60.0% to 100.7% and 70.0% to 97.5%, respectively. The intra-day precision of wood and bamboo FCMs were from 0.1% to 5.3% and 0.1 to 3.0%, and the inter-day precision from 0.3% to 8.5% and 1.2% to 7.8%, respectively. The limits of detection ranged from 0.11 to 15.38 for wood and 0.25–6.19 μg kg−1 for bamboo. This method provides a reference for the determination of fungicides in bamboo and wood FCMs.

9. Analysis of the Impact of Carbon Emission Trading on Social and Economic Development under the Change of International Oil Price

The drastic fluctuation of international oil price has a great impact on social and economic development. It is of great significance for social and economic development to analyze the impact of carbon emissions trading under the fluctuation of international oil price on social and economic development. Johansen test and state space model are used to study the spillover effect of international oil price fluctuation on carbon market price change and yield. VAR model, impulse response and variance decomposition are used to explore the impact of carbon emissions trading on energy price and consumer price index fluctuation, so as to analyze the social and economic development of carbon emissions trading under international oil price fluctuation. The results show that there is a long-term equilibrium relationship between international oil price and carbon price, and the spillover of international oil price to carbon market and yield shows that oil price can lead to the change of carbon price, but because of the change of EU energy consumption structure and the continuous improvement of carbon trading mechanism, the spillover effect shows a downward trend. The fluctuation of carbon trading price under the change of international oil price constitutes energy. Source price fluctuation causes, but the impact on consumer price index (CPI) is small, which does not constitute the cause of affecting social and economic development. The carbon price has little impact on domestic oil price fluctuation, and has a positive effect.

β-galactoside α-2,3-sialyltransferase 2 (ST3GAL2) is a member of the sialyltransferase family that mediates terminal modification of glycoproteins and glycolipids. ST3GAL2 has been found to play a role in obesity, aging, and malignant diseases. In this study, we cloned porcine ST3GAL2 (pST3GAL2) from porcine alveolar macrophages (PAMs), and its role in porcine reproductive and respiratory syndrome virus (PRRSV) infection was investigated by transcriptome analysis. pST3GAL2 was found to be located in the Golgi apparatus, and it was expressed at high levels in PRRSV-infected PAMs. Overexpression of pST3GAL2 resulted in a slight increase in PRRSV proliferation, and the interaction between pST3GAL2 and GP2a of PRRSV was detected by coimmunoprecipitation and confocal microscopy. The expression of pro-inflammatory cytokines (IFN-β, IL-2, IL-6, IL-18, IL-1β and TNF-α) was significantly inhibited in pST3GAL2-overexpressing, PRRSV-infected cells and upregulated in PRRSV-infected pST3GAL2-knockout cells, while the pattern of expression of anti-inflammatory cytokines (IL-4 and IL-10) was diametrically opposite. Our results demonstrate that the regulation of pST3GAL2 plays an important role in PRRSV proliferation and functional alterations in virus-infected cells. These results contribute to our understanding of the role of β-galactoside α-2,3-sialyltransferase 2 in antiviral immunity.

Porcine reproductive and respiratory syndrome (PRRS) is an important infectious disease caused by porcine reproductive and respiratory syndrome virus (PRRSV), leading to significant economic losses in swine industry worldwide. Although several studies have shown that PRRSV can affect the cell cycle of infected cells, it is still unclear how it manipulates the cell cycle to facilitate its proliferation. In this study, we analyzed the mRNA expression profiles of transcription factors in PRRSV-infected 3D4/21 cells by RNA-sequencing. The result shows that the expression of transcription factor DP2 (TFDP2) is remarkably upregulated in PRRSV-infected cells. Further studies show that TFDP2 contributes to PRRSV proliferation and the PRRSV nucleocapsid (N) protein induces TFDP2 expression by activating C/EBPβ. TFDP2 positively regulates cyclin A expression and triggers a less proportion of cells in the S phase, which contributes to PRRSV proliferation. This study proposes a novel mechanism by which PRRSV utilizes host protein to regulate the cell cycle to favor its infection. Findings from this study will help us for a better understanding of PRRSV pathogenesis. 14. Toward Sustainable Development: Unleashing the Mechanism Among International Technology Spillover, Institutional Quality, and Green Innovation Capability

Under the background of sustainable development, China’s economic growth engine becomes innovation-driven, and it is an important way for China to rapidly improve its green innovation capability by opening up to the outside world and utilizing the spillover effect of international technology. In this article, the system quality evaluation system is reconstructed by the method of fully arranged polygonal graphical indicators, and the provincial system quality in China is measured and added into the model as a regulating variable. The dynamic panel method and the dynamic threshold panel method are used to test the direct effects of foreign direct investment (FDI) and foreign trade on green innovation capability, the interaction effect of institutional quality, and the threshold effect. Empirical results show that the three technology spillovers have significantly promoted China’s green innovation capability. System quality will affect the determining coefficient of international technology spillovers on China’s green innovation capability. The positive promoting effects of FDI and foreign trade on China’s green innovation capability, all increase with the improvement of China’s system quality. Therefore, when utilizing FDI and foreign trade to promote green innovation in each region, each region should consider creating a good institutional environment for the emergence of international technological effects.

Nanosecond pulsed electric field (nsPEF) treatment has emerged as a promising and effective approach for pancreatic cancer. Tumor-infiltrating myeloid cells are crucial tumor regulators and potential immunotherapy targets. Understanding the effect of nsPEF on the myeloid cells in tumors is necessary for grasping the anti-tumor impact of nsPEF therapy. This study describes the phenotype and function of myeloid cells in Panc02 pancreatic cancer mouse models on day three after nsPEF using single-cell RNA sequencing (scRNA-Seq). Defining comparable myeloid cells in Panc02 tumors enabled characterization of their response to nsPEF treatment. Treatment with nsPEF increased infiltration by monocytes/macrophages, which participated in forming a immunosuppressive tumor microenvironment. NsPEF also promoted the recruitment of dendritic cells to tumors. Our comprehensive investigation of crucial myeloid subsets and significant cellular interactions regulating tumor immunity indicated that the nsPEF induced a compartmental remodeling of tumor-infiltrating myeloid cells in pancreatic cancer. These results provide information for interpreting the complex immune changes after nsPEF treatment in pancreatic cancer and may guide future therapeutic interventions.

The organizational development following the philosophy of sustainability is becoming the prerequisite for companies, which urges and emphasizes them to incorporate social, economic, and environmental aspects into their manufacturing and operations. Despite plethora of researches exploring lean practices as the solution for improving productivity, it is limited to the operational aspects only, thus ignoring the other aspects of sustainability. Moreover, there is a dearth of studies that explored the combined effect of sustainable innovation, process innovation, and lean practices on the three aspects of sustainability. The current study is an attempt and contribution in the existing literature through the sample of 431 respondents from ISO 14001 certified Chinese organizations during COVID-19 pandemic conditions. The estimations were performed through the Partial Least Squares–Structural Equation Modeling. The results revealed a positive association among the proposed hypothesis of aforementioned studied phenomena. In addition, the current study explores the role of sustainable innovation as a mediator between lean and three dimensions of the sustainability, which is reportedly found to be a partial mediator as both direct and indirect effects are found to be statistically significant. Through the findings of the current study, the managers and decision makers can comprehend the potential of the possible benefits which can be reaped by incorporating both lean and sustainability within their operations.

A recent study showed that patients with coronavirus disease 2019 (COVID-19) have gastrointestinal symptoms and intestinal flora dysbiosis. Yeast probiotics shape the gut microbiome and improve immune homeostasis. In this study, an oral candidate of yeast-derived spike protein receptor-binding domain (RBD) and fusion peptide displayed on the surface of the yeast cell wall was generated. The toxicity and immune efficacy of oral administration were further performed in Institute of Cancer Research (ICR) mice. No significant difference in body weights, viscera index, and other side effects were detected in the oral-treated group. The detectable RBD-specific immunoglobulin G (IgG) and immunoglobulin A (IgA) of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and more complex microbiota were detected from oral administration mice compared with those of the control group. Interestingly, the recombinant yeast was identified in female fetal of the high-dose group. These results revealed that the displaying yeast could fulfill the agent-driven immunoregulation and gut microbiome reconstitution. The findings will shed light on new dimensions against SARS-CoV-2 infection with the synergistic oral agents as promising non-invasive immunization and restoring gut flora.

Selenium (Se) is critical for human health, but human intake of Se is often inadequate. Organic forms of dietary Se are considered safe and more bioavailable than inorganic forms. Along with a generally high nutritional value, sprouts are sensitive to Se treatment. This study used selenomethionine and methylselenocysteine solutions to cultivate Se-enriched sprouts under an optimized hydroponic condition. Content change and transformations of the selenoamino acids were analyzed by a developed HPLC-ESI-MS/MS method. Uptake of both selenomethionine and methylselenocysteine was dose-dependent and involved active transport and passive diffusion, as demonstrated by the respiratory and aquaporin inhibition assays. Passive diffusion played a dominant role. Free methylselenocysteine was the predominant form in samples. Selenomethionine and methylselenocysteine were capable of mutual transformation. Moreover, the selenoprotein generation was associated with the increasing Se concentration of the culture solutions. The results provided scientific references for the efficient utilization of organic Se in sprouts.

25. Integrating Remote Sensing and Meteorological Data to Predict Wheat Stripe Rust

Wheat stripe rust poses a serious threat to wheat production. An effective prediction method is important for food security. In this study, we developed a prediction model for wheat stripe rust based on vegetation indices and meteorological features. First, based on time-series Sentinel-2 remote sensing images and meteorological data, wheat phenology (jointing date) was estimated using the harmonic analysis of time-series combined with average cumulative temperature. Then, vegetation indices were extracted based on phenological information. Meteorological features were screened using correlation analysis combined with independent t-test analysis. Finally, a random forest (RF) was used to construct a prediction model for wheat stripe rust. The results showed that the RF model using the input combination (phenological information-based vegetation indices and meteorological features) produced a higher prediction accuracy and a kappa coefficient of 88.7% and 0.772, respectively. The prediction model using phenological information-based vegetation indices outperformed the prediction model using single-date image-based vegetation indices, and the overall accuracy improved from 62.9% to 78.4%. These results indicated that the method combining phenological information-based vegetation indices and meteorological features can be used for wheat stripe rust prediction. The results of the prediction model can provide guidance and suggestions for disease prevention in the study area.

Image restoration methods are commonly used to improve the quality of astronomical images. In recent years, developments of deep neural networks and increments of the number of astronomical images have evoked a lot of data--driven image restoration methods. However, most of these methods belong to supervised learning algorithms, which require paired images either from real observations or simulated data as training set. For some applications, it is hard to get enough paired images from real observations and simulated images are quite different from real observed ones. In this paper, we propose a new data--driven image restoration method based on generative adversarial networks with option--driven learning. Our method uses several high resolution images as references and applies different learning strategies when the number of reference images is different. For sky surveys with variable observation conditions, our method can obtain very stable image restoration results, regardless of the number of reference images., Comment: 11 pages. 27. The effect of migration policy on growth, structural change, and regional inequality in China

Between 2000 and 2015, China’s aggregate income quadrupled, its provincial income inequality fell by a third, and its share of employment in agriculture fell by a half. Internal migration is central to this transformation, with the number of internal migrant workers reaching 300 million by 2015. Combining rich data on migration with a spatial general equilibrium model of China’s economy, we quantify the size and the impact of internal migration cost reductions in China between 2000 and 2015. During the 15-year period, China’s internal migration costs fell by forty-five percent. In addition to contributing substantially to growth, these migration cost changes account for the majority of the reallocation of workers out of agriculture and the drop in regional inequality. We compare the effect of migration policy changes with other important economic changes, including changes in trade costs, capital market distortions, average cost of capital, and productivity. While each contributes meaningfully to growth, migration policy changes are central to China’s structural change and regional income convergence. We also find that the recent slow-down in aggregate economic growth between 2010 and 2015 is associated with smaller reduction in inter-provincial migration costs and a larger role of capital accumulation.

The Oriental migratory locust is a destructive agricultural pest in China. Large-scale locust area (the area possessing suitable breeding habitat for locusts and has locust infestation) extraction and its evolution analysis are essential for locust ecological control. Existing methods seldom consider the spatial differences in the locust development and habitat landscape structures in large areas. To analyze these effects, our study proposed a landscape-based habitat suitability model (LHS model) for large-scale locust area extraction based on remote sensing data, taking the middle and lower reaches of the Yellow River (MLYR) as an example. Firstly, the DD model was used to simulate locust development and obtain habitat factors of the corresponding dates; secondly, the patch distribution of different land cover classes and their adjacent landscape characteristics were analyzed to determine the landscape-based factors memberships; finally, the habitat suitability index was calculated by combining the factors memberships and weights to extract the locust area. Compared with the patch-based model using moving windows (patch based-analytic hierarchy process model, R2 = 0.77), the LHS model accuracy improved significantly (R2 = 0.83). Our results showed that the LHS model has a better application prospect in large-scale locust area extraction. By analyzing the locust areas evolution along the MLYR extracted using the LHS model, we found human activities were the main factors affecting the locust areas evolution from 2016 to 2020, including: (1) planting the plants that locusts do not like and urbanization caused the decrease of the locust area; (2) the wetland protection policies may cause the increase of the locust area. The model and research results help locust control and prevention to realize the sustainable development of agriculture.

29. Dynamic Forecast of Desert Locust Presence Using Machine Learning with a Multivariate Time Lag Sliding Window Technique

Desert locust plagues can easily cause a regional food crisis and thus affect social stability. Preventive control of the disaster highlights the early detection of hopper gregarization before they form devastating swarms. However, the response of hopper band emergence to environmental fluctuation exhibits a time lag. To realize the dynamic forecast of band occurrence with optimal temporal predictors, we proposed an SVM-based model with a temporal sliding window technique by coupling multisource time-series imagery with historical locust ground survey observations from between 2000–2020. The sliding window method was based on a lagging variable importance ranking used to analyze the temporal organization of environmental indicators in band-forming sequences and eventually facilitate the early prediction of band emergence. Statistical results show that hopper bands are more likely to occur within 41–64 days after increased rainfall; soil moisture dynamics increasing by approximately 0.05 m³/m³ then decreasing may enhance the chance of observing bands after 73–80 days. While sparse vegetation areas with NDVI increasing from 0.18 to 0.25 tend to witness bands after 17–40 days. The forecast model combining the optimal time lags of these dynamic indicators with other static indicators allows for a 16-day extended outlook of band presence in Somalia, Ethiopia, and Kenya. Monthly predictions from February to December 2020 display an overall accuracy of 77.46%, with an average ROC-AUC of 0.767 and a mean F-score close to 0.772. The multivariate forecast framework based on the lagging effect can realize the early warning of band presence in different spatiotemporal scenarios, supporting early decisions and response strategies for desert locust preventive management.

Leaves are the primary photosynthetic structures, while photosynthesis is the direct motivation of crop yield formation. As a legume plant, peanut (Arachis hypogaea) is one of the most economically essential crops as well as an important source of edible oil and protein. The leaves of A. hypogaea are in the tetrafoliate form, which is different from the trifoliate leaf pattern of Medicago truncatula, a model legume species. In A. hypogaea, an even-pinnate leaf with a pair of proximal and distal leaflets was developed; however, only a single terminal leaflet and a pair of lateral leaflets were formed in the odd-pinnate leaf in M. truncatula. In this study, the development of compound leaf in A. hypogaea was investigated. Transcriptomic profiles revealed that the common and unique differentially expressed genes were identified in a proximal leaflet and a distal leaflet, which provided a research route to understand the leaf development in A. hypogaea. Then, a naturally occurring mutant line with leaf developmental defects in A. hypogaea was obtained, which displayed a pentafoliate form with an extra terminal leaflet. The characterization of the mutant indicated that cytokinin and class I KNOTTED-LIKE HOMEOBOX were involved in the control of compound leaf pattern in A. hypogaea. These results expand our knowledge and provide insights into the molecular mechanism underlying the formation of different compound leaf patterns among species.

Ubiquitination plays a major role in immune regulation after viral infection. An alternatively spliced porcine E3 ubiquitin ligase RNF122 promoted PRRSV infection and upregulated in PRRSV-infected PAM cells was identified. We characterized the core promoter of RNF122, located between −550 to −470 bp upstream of the transcription start site (TSS), which displayed significant differential transcriptional activities in regulating the transcription and expression of RNF122. The transcription factor HLTF was inhibited by nsp1α and nsp7 of PRRSV, and the transcription factor E2F complex regulated by nsp9. Together, they modulated the transcription and expression of RNF122. RNF122 could mediate K63-linked ubiquitination to raise stability of PRRSV nsp4 protein and thus promote virus replication. Moreover, RNF122 also performed K27-linked and K48-linked ubiquitination of MDA5 to degrade MDA5 and inhibit IFN production, ultimately promoted virus proliferation. In this study, we illustrate a new immune escape mechanism of PRRSV that enhances self-stability and function of viral nsp4, thus, regulating RNF122 expression to antagonize IFNα/β production. The present study broadens our knowledge of PRRSV-coding protein modulating transcription, expression and modification of host protein to counteract innate immune signaling, and may provide novel insights for the development of antiviral drugs.

Polyploidy is a fascinating characteristic of the liver and correlates with HCC development and progression. This study aims to investigate the association between hepatocyte polyploidy spectrum and HCC recurrence after LT.Patients and Methods: Thirty-two paired HCC, peritumoral, cirrhotic, and normal liver specimens were employed to examine the hepatocyte polyploidization pattern during liver tumorigenesis. Clinicopathological implications of polyploidy spectrum for LT recipients with HCC were investigated in 205 patients from two transplant centers. Immunofluorescence staining was performed on paraffin-embedded tissue sections to determine the ploidy profiles in situ. Expression levels of CD4, CD8, forkhead box protein 3 (Foxp3) and programmed death-ligand 1 (PD-L1) were measured using immunohistochemistry. An array-based multiplex ELISA system was used for the quantitative measurement of 40 unique inflammatory cytokines.Results: The fraction of mononuclear polyploidy increased, whereas that of binuclear polyploidy reduced during hepatocarcinogenesis. Recipients with highly mononuclear polyploid HCC (HMPâHCC) had inferior recurrence-free survival and HCC-specific survival than poorly mononuclear polyploid HCC recipients. These two groups differed in abundance of infiltrative CD8+ cytotoxic T cells and FoxP3+ Treg cells, and PD-L1 expression, as well as circulating granulocyteâmacrophage colony-stimulating factor, interferon-γ and interleukin-10 levels. HMPâHCC constituted an independent recurrence predictor and could improve the discriminative efficacy of clinical prediction models (Milan criteria, AFP model, and Metroticket 2.0 criteria). Strong lensing in galaxy clusters probes properties of dense cores of dark matter halos in mass, studies the distant universe at flux levels and spatial resolutions otherwise unavailable, and constrains cosmological models independently. The next-generation large scale sky imaging surveys are expected to discover thousands of cluster-scale strong lenses, which would lead to unprecedented opportunities for applying cluster-scale strong lenses to solve astrophysical and cosmological problems. However, the large dataset challenges astronomers to identify and extract strong lensing signals, particularly strongly lensed arcs, because of their complexity and variety. Hence, we propose a framework to detect cluster-scale strongly lensed arcs, which contains a transformer-based detection algorithm and an image simulation algorithm. We embed prior information of strongly lensed arcs at cluster-scale into the training data through simulation and then train the detection algorithm with simulated images. We use the trained transformer to detect strongly lensed arcs from simulated and real data. Results show that our approach could achieve 99.63 % accuracy rate, 90.32 % recall rate, 85.37 % precision rate and 0.23 % false positive rate in detection of strongly lensed arcs from simulated images and could detect almost all strongly lensed arcs in real observation images. Besides, with an interpretation method, we have shown that our method could identify important information embedded in simulated data. Pancreatic cancer is one of the leading causes of cancer-related deaths worldwide. Poor prognosis and low survival rates have driven the development of novel therapeutic strategies. Nanosecond pulsed electric field has emerged as a novel, minimal invasive and non-thermal treatment for solid tumors. It is of great significance to study the combination therapy of nsPEF and other treatment strategies for pancreatic cancer. We developed neutrophil membrane-wrapped liposomal nanoparticles loaded with gemcitabine (NE/Lip-GEM) and investigated their use as a complementary agent for nsPEF treatment. Our results showed that neutrophil-mediated delivery of liposomal-gemcitabine (NE/Lip-GEM) efficiently inhibited the growth of pancreatic tumors in mice whose has been treated with incomplete nsPEF ablation. The combination of nsPEF and NE/Lip-GEM may be a promising synergistic strategy for pancreatic cancer therapy.

38. Liver transplantation for Hepatocellular Carcinoma: A prognostic model incorporating pretransplant inflammatory cytokines

Liver transplantation (LT) remains an optimal treatment for selected hepatocellular carcinoma (HCC). Cytokines can be obtained by minimally invasive techniques and are associated with the development of HCC. The purpose of our investigation was to explore the prognostic value of pretransplant serum inflammatory cytokine profiles in HCC patients for LT.We detected forty inflammatory cytokines in pre-LT serum from 42 HCC patients by using an inflammation-related antibody array. A pretransplant serum inflammatory cytokine-associated risk assessment model (pre-SCRAM) was developed and was validated in an external cohort of 213 HCC patients who underwent LT and were then followed prospectively.The pre-LT factors independently associated with recurrence-free survival (RFS) were as follows: B-lymphocyte chemoattractant (BLC), interleukin (IL)-12p40 and maximum tumor diameter. High IL-12P40 level was associated with a significantly smaller maximum tumor diameter (p = 0.021), decreased proportion of nodules ≥ 3 (p = 0.001), lower platelet counts (p = 0.011) and lower portal vein tumor thrombus (p = 0.031). Conversely, recipients with poor BLC level had higher alpha-fetoprotein (AFP) levels (p = 0.016). Kaplan-Meier analyses revealed that high pre-LT BLC or IL-12p40 level was associated with superior RFS. The pre-SCRAM stratified recipients into three risk groups: high risk, intermediate risk and low risk. In the validation cohort, for patients in the high, intermediate, and low risk groups, the 3-year RFS rates were 29.3%, 58.7%, and 82.2%, respectively, the 3-year HCC-specific survival rates were 54.5%, 73.8%, and 86%, respectively, and the 3-year overall survival rates were 44.4%, 60.9%, and 79.9%, respectively. The pre-SCRAM model performed well and remained significant in optimizing the risk stratification of recurrence in patients beyond the Milan criteria or the AFP model.Pretransplant cytokine profiles can provide powerful prognostic information in the setting of LT for HCC. A pre-LT risk model incorporating cytokines showed excellent efficiency in recurrence prediction for HCC patients, which could ultimately stratify the prognosis in patients beyond the Milan criteria or the AFP model.

39. Analyzing the mechanism among rural financing constraint mitigation, agricultural development, and carbon emissions in China: A sustainable development paradigm

Agricultural development in developing countries generally faces increasingly severe financing constraints, and China's practice provides a model for other countries. This article investigates the impact of the policy to alleviate rural financing constraints on agricultural carbon emissions in developing countries by using a quasi-natural experiment, the “Incremental Incentives for Agricultural Loans from County Financial Institutions,” implemented in China, with data from 2005 to 2018, and with a DID approach. It is found that under the influence of this policy, the alleviation of agricultural financing constraints will significantly generate an increase in the intensity of agricultural carbon emissions, which is triggered by the fact that the alleviation of financing constraints increases the production factors invested per unit area in the agricultural production process, while the increase in the mechanization and scale of agriculture will assist to reduce the contribution of the alleviation of financing constraints to carbon emissions. The impact of financing constraint alleviation on agricultural carbon emissions may differ among places with different economic and social development and agricultural bases. Additionally, the alleviation of rural financing constraints can promote agricultural development and increase farmers' income, enabling other developing countries to adjust their policy orientation when adopting such policies, thus promoting agricultural development while controlling agricultural carbon emissions and contributing to the global emission reduction cause.

FOLFOX (oxaliplatin, fluorouracil and calcium folinate) is the first-line chemotherapy regimen for colon cancer therapy in the clinic. It provides superior efficacy than oxaliplatin alone, but the underlying mechanism remains unclear. In the present study, pharmacomicrobiomics integrated with metabolomics was conducted to uncover the role of the gut microbiome behind this. First, in vivo study demonstrated that FOLFOX exhibited better efficacy than oxaliplatin alone in colon cancer animal models. Second, 16S rDNA gene sequencing analysis showed that the abundance of Akkermansia muciniphila (A. muciniphila) remarkably increased in the FOLFOX treated individuals and positively correlated with the therapeutic effect. Third, further exploration confirmed A. muciniphila colonization significantly enhanced the anti-cancer efficacy of FOLFOX. Last, metabolomics analysis suggested dipeptides containing branched-chain amino acid (BCAA) might be responsible for gut bacteria mediated FOLFOX efficacy. In conclusion, our study revealed the key role of A. muciniphila in mediating FOLFOX efficacy, and manipulating A. muciniphila might serve as a novel strategy for colon cancer therapy.

1. Organic anion transporting polypeptides (OATPs) belong to the superfamily of solute carriers (SLC), which are important membrane transporters in animals and humans. Liver is an important organ for drug disposition. In human liver, three OATPs, namely OATP1B1, 1B3 and 2B1, are expressed on the basolateral membrane of hepatocytes.2. OATPs have multiple substrate specificity, mediating transport of a wide range of endo- and exogenous substances such as bile salts, bilirubin, hormones and their conjugates, toxins and various drugs. Therefore, they are important for drug disposition in human body. In this review, we compiled a complete list of the substrates for human hepatic OATPs.3. OATP genes have single-nucleotide polymorphisms (SNPs), which could lead to the alteration of their function, and thus might result in the change of pharmacokinetic properties for their substrate drugs. In this review, we summarized the genetic polymorphisms of the three hepatic OATPs and their effect on in vitro transport function and in vivo pharmacokinetics of substrate drugs.4. Finally, some concerns and perspectives on OATP polymorphism research are discussed.

This paper is concerned with theoretical and practices approach for overall quality-related fault detection and identification in process industries. Fault detection and fault tracing can help engineers to take corrective actions and recover the process operations. A novel diagnostic method is proposed based on stacked automatic encoder-canonical correlation analysis (SAE-CCA) and least absolute shrinkage selection operator (Lasso). First, a quality monitoring scheme based on SAE-CCA is proposed, which establishes a relationship model among quality and characteristic variables to detect faults. Then, Lasso is used for locating the root causes, according to the process state and fault information. Finally, the experiments are conducted with typical industry process data, i.e., a hot strip mill process (HSMP), in order to demonstrate the effectiveness of the whole diagnosis method.

Most actual industrial processes are multivariable and constrained complex systems. The state and output of the system also have uncertainty due to the existence of random disturbances. The output of the system is easier to be measured than the state and can more intuitively reflect the running state of the system. Considering the limitations of industrial equipment and the benefits of production, it is generally allowed to have a small portion of the output that can exceed the constraints. As a result, the outputs can be represented as corresponding single probabilistic constraints. In this paper, therefore, an output probabilistic constrained optimal control algorithm based on multivariable model algorithm control (MMAC) is proposed. First, the feedback correction link of the MMAC algorithm is improved, and the predicted outputs are corrected by taking the weighted average of the errors. Then, assuming that the disturbances obey Gaussian distribution, the output probabilistic constraints are transformed into deterministic ones. Next, the optimal control problem is solved as a quadratic programming (QP) problem after combining them with the performance index function of the MMAC. Finally, the proposed algorithm is applied to the looper control system in hot strip rolling process and compared with the single MMAC algorithm to verify its effectiveness.

As a result of emerging climate change trends, businesses and organizations are becoming more concerned about environmental sustainability. In this context, green supply chain management (GSCM) approaches are expanding globally. Based on institutional and sociotechnical systems theory, the conceptual model of this research highlights a mediation of two GSCM types, namely technological and behavioral practices, as well as the moderation of organizational pressure on institutional performance. The hypotheses were evaluated using the structural equation modeling (SEM) method, obtaining data from 563 Chinese firms. This study's categorization of technological and behavioral GSCM practices and results add to the literature on GSCM. The empirical findings indicate that the technological GSCM practices considerably affect behavioral practices. Moreover, GSCM technological and behavioral practices have a substantial effect on institutional performance. GSCM behavioral practices mediate the link between technological GSCM practices and institutional performance. The outcomes also reveal that organizational pressure moderates the association between GSCM technological GSCM practices and institutional performance. These results show that companies in emerging economies should prioritize the behavioral GSCM practices to successfully apply the technological GSCM practices to achieve compelling financial, social, and environmental performance.

As transcriptional co-activator of AP-1/Jun, estrogen receptors and NF-κB, nuclear protein RBM39 also involves precursor mRNA (pre-mRNA) splicing. Porcine reproductive and respiratory syndrome virus (PRRSV) causes sow reproductive disorders and piglet respiratory diseases, which resulted in serious economic losses worldwide. In this study, the up-regulated expression of RBM39 and down-regulated of inflammatory cytokines (IFN-β, TNFα, NF-κB, IL-1β, IL-6) were determined in PRRSV-infected 3D4/21 cells, and accompanied with the PRRSV proliferation. The roles of RBM39 altering phosphorylation of c-Jun to inhibit the AP-1 pathway to promote PRRSV proliferation were further verified. In addition, the nucleocytoplasmic translocation of RBM39 and c-Jun from the nucleus to cytoplasm was enhanced in PRRSV-infected cells. The three RRM domain of RBM39 are crucial to support the proliferation of PRRSV. Several PRRSV RNA (nsp4, nsp5, nsp7, nsp10-12, M and N) binding with RBM39 were determined, which may also contribute to the PRRSV proliferation. Our results revealed a complex mechanism of RBM39 by altering c-Jun phosphorylation and nucleocytoplasmic translocation, and regulating binding of RBM39 with viral RNA to prompt PRRSV proliferation. The results provide new viewpoints to understand the immune escape mechanism of PRRSV infection.

Interferon (IFN)-mediated antiviral responses are central to host defense against viral infection. Porcine viral infection has emerged as a serious hazard for the pig industry. The construction of an engineered Saccharomyces cerevisiae strain that efficiently produces porcine IFN has demonstrated several advantages. It can be easily fed to pigs, which helps in reducing antibiotic residues in pork and improve meat quality. In this study, the stable expression of several porcine IFN molecules (pIFN-α1, pIFN-β, pIFN-λ1, pIFN-λ1-β, pIFN-λ1-β-α1) were determined using an engineered S. cerevisiae system. With the YeastFab assembly method, the complete transcriptional units containing promoter (GPD), secretory peptide (α-mating factor), target gene (IFN) and terminator (ADH1) were successfully constructed using the characteristics of type II restriction endonuclease, and then integrated into the chromosomes Ⅳ and XVI of ST1814 yeast host strain, respectively. The expression kinetics of recombinant pIFNs were further analyzed. Synergism in the expression level of IFN receptor, antiviral protein, and viral loading was observed in viral-cell infection model treated with different porcine IFN subtypes. The porcine reproductive and respiratory syndrome viral load and antibody titer in serum decreased significantly after oral administration of IFN expression yeast fermentation broth. These findings indicate the potential efficacy of multi-valent pIFNs expressing S. cerevisiae as a potent feed material to prevent viral infections of pigs.

The viral proliferation process of envelop viruses including invasion, assembly, and release process is impacted by glycosylation. Sialic acid modification mainly exists in the terminal glycosylation of proteins and lipids. In this study, the effect of porcine sialylglycosyltransferase (ST3GAL2) on virus proliferation, as well as regulation of inflammatory factor in PRRSV-infected 3D4/21 cell model were analyzed. The expression of ST3GAL2 was up-regulated in PRRSV-infected 3D4/21 cells. The expression of ST3GAL2 that promote the proliferation of PRRSV was determined by overexpression and interference experiment. The ST3GAL2 localizes to Golgi apparatus and interacts with PRRSV structural protein GP2a was verified by confocal microscopy and co-immnoprecipitation array, which may play roles in the assembly of PRRSV envelope and virus proliferation. The expression of ST3GAL2 also impacts the expression of inflammatory cytokines, which regulate cellular immunity and promote the immune escape of PRRSV from innate immunity. Our results highlighted a potential mechanism whereby ST3GAL2 promotes PRRSV proliferation and offers the anti-inflammatory role of ST3GAL2 in cellular immunity.
