Four different leaf extracts of B. microstachya var. massambabensis were studied to evaluate their antioxidant capacity by using three in vitro methods, with Ginkgo biloba and Trolox® as the standards. With the DPPH and ABTS·+ methods, the antioxidant activity of the extracts was in the following order, from maximum to minimum: AcEt > WAc > raw EtOH > EtOH CA > EGb, while with the ORAC method, it was as follows: EtOH CA > raw EtOH > AcEt > WAc > EGb. Phototoxic analysis was performed in yeast cultures of Saccharomyces cerevisiae. From the ethyl acetate extract, 2 flavonoids kaempferol-3-O-rhamnoside and astragalin-2”,6”-O-digallate were isolated and identified by HPLC and 1H- and 13C-NMR; to our knowledge, this is the first report of the occurrence of astragalin-2”,6”-O-digallate in the Bauhinia genus., Colegio de Farmacéuticos de la Provincia de Buenos Aires

Nanoparticles are widely utilized to overcome drugs insolubility problems and sustain release improving the bioavailability. Zinc phthalocyanine, a hydrophobic photosensitizer with solubility problems, was loaded in PLA nanoparticles. Photosensitizer loaded in polymeric nanoparticles was produced with the following characteristics: size in the 200-300 nm range, negative zeta potential (-15 to -19 mV), low polydispersity index (< 0.1), satisfactory encapsulation efficiency (70-80%), low residual PVA, smooth surface and spherical shape. The photosensitizer release from nanoparticles was sustained and the kinetic followed Higuchi’s model. ZnPc loaded in polymeric nanoparticles exhibited higher phototoxicity than free photosensitizer. Phototoxicity of the ZnPc loaded in Resomer® R203 nanoparticles was improved for increasing photosensitizer concentration (1 to 4 μg/ml), light dose (10 to 30 J/cm2 ) or incubation time (2 to 4 h). Apresenta-se método simples de cromatografia líquida de alta eficiência (CLAE) para análise da lidocaína em meio aquoso, após estudo de liberação in vitro. A lidocaína foi analisada usando-se coluna LichroCART RP-18 (5 mm, 125x4 mm), fase móvel constituída de acetonitrila: tampão fosfato de sódio 0,05 M, pH 6 (35:65), adicionada de 0,05% de dietilamina com fluxo de 1 mL/min. O tempo de retenção foi de 7,9 min. O comprimento de onda de análise utilizado foi de 210 nm. A linearidade do método foi de 1,25 a 25 µg/mL com coeficiente de variação intraensaio e inter-ensaio menor que 3,5 %. A metodologia desenvolvida e validada mostrou sensibilidade e especificidade para a realização dos estudos propostos, considerando-se que as amostras obtidas a partir dos estudos de liberação in vitro contêm concentrações muito baixas do fármaco, além de outras substâncias do meio de dissolução que podem interferir no doseamento. A quantificação do fármaco e dos interferentes pode não ser possível se for efetuada por outras metodologias analíticas convencionais. Assim, o método desenvolvido é de grande importância para a quantificação do fármaco nas alíquotas obtidas nos ensaios de liberação in vitro. A simple high performance liquid chromatography method to assay lidocaine hydrochloride in aqueous receiving media, following in vitro release, is presented. Lidocaine hydrochloride was analysed using a 5 mm LichroCART® RP-18 column (125 x 4 mm i.d.). The mobile phase was acetonitrile: 0.05 M sodium phosphate buffer, pH 6.0 (35:65) and 0.05% of diethylamine at a flow rate of 1 mL/min. The retention time was 7.9 min. Detection was carried out at 210 nm at room temperature (28 ºC). The method was found to be linear in the range 1.25 to 25 mg/mL, showing average intraassay and inter-assay coefficients of variation below 3.5%. The proposed method was validated for linearity, specificity, precision and accuracy and was shown to be useful for the analysis of lidocaine hydrochloride in in vitro release studies.

Among its tumors, there is the Pancreatic Polypeptide-Secreting Tumor of the Distal Pancreas (PPoma). A rare tumor that belongs to the malignancy rate (90%), occurring mainly with bad breath and poor prognosis, with survival rates in 5 years of less than 10%. In the absence of diagnosis, this should be done in an intensive or accidental manner or with mass effect, which causes non-specific abdominal pain due to hepatomegaly or symptoms suggestive of obstruction of the pancreatic or biliary duct. PPoma is classified as a non-functional pancreatic neuroendocrine tumor that overexpresses SSTR 2 (Somatostatin Receptor of subtype 2). Octreotide has a high affinity for SST 2, but its labeling with Technetium-99 meta-stable (99mTc) by direct mode has been shown to be low yield. With the advent of nanotechnology, the problems in the targeting of tumor cells in the production and in the adhesion to the radiopharmaceutical are not favorable agents to those overcome. Thus the present work has as main objective the production of nanorradiopharmaceuticals based on polymeric nanoparticles of polylactic acid(PLA) loaded with Octreotide and radiolabeled with 99mTc to obtain a diagnostic agent of the PPoma. Octreotide was encapsulated in alcohol nanoparticles by the emulsion emulsion method using the PLA polymer and the poly(vinyl alcohol) surfactant (PVA) at 4% concentration of the drug. The characterization of the nanoparticles was performed using the technique of Atomic Force Microscopy (AFM) and Dynamic Light Scattering (DLS) and the efficiency of the encapsulation of the indirect analysis in the spectrophotometer at 279 nm for the reading of Octreotide. Evaluation of cytotoxicity by the MTT assay (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) at various concentrations of the nanosystem against a human pancreatic carcinoma cell line (MIA Paca-2) SSTR 2 by mimicking the PPoma and by direct method radiolabeled the nanosystem with 99mTc and then determined its yield and radiolabel stability up to 24 hours by paper chromatography using a Whatmam n° 1 paper stationary phase and for a mobile acetone phase. The bioavailability study was used in males of Balb/c induced by the MIA Paca-2 xenograft divided into two groups each with 6 animals for the effect of nanoparticles with and without an Octreotide. The results were as nanoparticles of size around 189 nm, by size as well as by AFM and with Polydispersity Index (PDI) 0.097 corroborating the monodispersive behavior. The efficiency of the Octetotide encapsulation was 60%. As nanoparticles were successfully radiolabeled and stable for 24 hours with 99mTc, demonstrating an excellent induced tumor. The MTT assay corroborated a safety of the nanosystem for the cells. Therefore, the use of 99mTc-labeled PLA/PVA/Octreotide nanoparticles can be used as an image agent for PPoma. O câncer de pâncreas está preste a se tornar a segunda causa de morte por câncer no mundo. Dentre os seus tipos tumorais, tem-se o Tumor Secretor de Polipeptídeo Pancreático (PPoma). Tumor raro que apresenta alta taxa de malignidade (90%) ocorrendo preferencialmente metástase no fígado e de prognóstico ruim, com taxas de sobrevida em 5 anos menores do que 10%. Isto se deve, principalmente pela demora no diagnóstico, que geralmente é feito de modo acidental ou pelo efeito de massa que causa uma dor abdominal inespecífica pela hepatomegalia ou sintomas sugestivos de obstrução do ducto pancreático ou biliar. O PPoma é classificado como um tumor neuroendócrino pancreático não funcional que superexpressa o SSTR 2 (Receptor de Somatostatina do subtipo 2). A Octreotida possui alta afinidade pelo SST 2, mas sua marcação com Tecnécio-99 metaestável (99mTc) pelo modo direto tem-se demonstrado com baixo rendimento. Com o advento da nanotecnologia, problemas no direcionamento para células tumorais na produção e na adesão do radiofármaco no órgão alvo têm sido superados. Assim o presente trabalho tem como objetivo principal a produção de um nanorradiofármaco com base de nanopartículas poliméricas de ácido poliláctico(PLA) carregado com Octreotida e radiomarcadas com 99mTcpara obtenção de um agente diagnóstico do PPoma. A Octreotida foi encapsulada na forma de nanopartículas pelo método de dupla emulsificação utilizando o polímero PLA e o tensoativo álcool polivinílico (PVA) na concentração de 4% do fármaco. A caracterização das nanopartículas foi realizada pela técnica de Microscopia de Força Atômica (AFM) e Espalhamento Dinâmico da Luz (DLS) e a eficiência da encapsulação pela análise indireta no espectrofotômetro a 279 nm para leitura da Octreotida. A avaliação da citotoxicidade realizada pelo ensaio de MTT (brometo de 3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazólio) em várias concentrações do nanossistema frente a linha celular de carcinoma pancreático humano(MIA Paca-2) que expressão SSTR 2 mimetizando o PPoma. Pelo método direto fez-se aradiomarcação do nanossitema com 99mTc. Em seguida determinou o seu rendimento e estabilidade da radiomarcação até 24 horas pela cromatografia em papel, utilizando a fase estacionária papel de Whatmam nº 1 e para a fase móvel a acetona. No estudo de biodistribuição foram utilizados camundongos machos de Balb/c induzidos pelo xenoenxerto de MIA Paca-2divididos em dois grupos cada um com 6 animais para avaliar nanopartículas com e sem a Octreotida. Após duas horas da administração do nanorradiofármaco, os animais foram sacrificados e dissecados para contagem dos órgãos no contador gama. Os resultados mostraram que as nanopartícula eram de tamanho em torno de 189 nm, tanto pelo DLS quanto pelo AFM e com Índice de Polidispersão (PDI) de 0,097 corroborando o comportamento monodispersivo. A eficiência do encapsulamento da Octetotida foi de 60%. As nanopartículas foram radiomarcadas com sucesso e estáveis por 24 h com 99mTc, demostrando uma excelente absorção pelo tumor induzido. O ensaio MTT corroborou a segurança do nanossistema para as células. Portanto, o uso de nanopartículas de PLA/PVA/Octreotida marcadas com 99mTc podem ser utilizadas como agente de imagem para o PPoma.

Among all cancer treatment and diagnosis options, the development of nanorradiopharmaceuticals is among the most promising techniques for the early detection of tumors. The advances of nanotechnology in the last decades have presented important contributions to the improvement of the diagnostic and therapeutic processes, in particular the overcoming of several molecular barriers, thus enabling better targeting, as well as increasing the chances of early diagnosis and efficient treatment. This work aimed at the production, characterization and in vivo evaluation of the biodistribution of polymeric nanoparticles of 5-Fluorouracil labeled with Technetium-99m for the detection of adenocarcinoma. The results corroborated expectations, in particular tumor targeting and imaging. O desenvolvimento de novos nanorradiofármacos é uma atividade essencial ao aperfeiçoamento da medicina nuclear e fundamental para o diagnóstico precoce e eficaz das doenças oncológicas, assim como para o avanço da radiofarmácia. Dentre todas as opções de tratamento e diagnóstico do câncer, o desenvolvimento de nanorradiofármacos figura entre as técnicas mais promissoras na detecção precoece de tumores. Os avanços da nanotecnologia, nas últimas décadas, apresentaram importantes contribuições para a melhoria dos processos diagnósticos e terapêuticos, em especial a superação de diversas barreiras moleculares, possibilitando, assim, o melhor direcionamento, bem como aumentando as chances de diagnóstico precoce e tratamento eficiente. Este trabalho teve como objetivo a produção, caracterização e avaliação in vivo da biodistribuição de nanopartículas poliméricas de 5- Fluorouracil marcadas com Tecnécio-99m para a detecção de adenocarcinoma. Os resultados corroboraram as expectativas, em particular o direcionamento e o imagiamento tumoral.

One of the most prevalent barriers in the oncology area that corresponds to the high morbidity rate is the absence of early detection of the carcinogenic processes, especially in cases of triple negative breast cancer. Non-discovery of a cancer below the level detectable by current screening tests, including by means of blood samples, points to the real need for innovative technological tools that are specifically targeted to the tumor microenvironment, with minimal disruption to healthy tissues. The use of aptamers is an evolution of target-directed therapy, presenting a considerable level of sensitivity to particular substrates, such as mucin 1 (MUC1). In this study, the anti-MUC1 aptamer was used as a drug delivery system (DDS) for a radioactive polymeric nanoparticle (NP) in the imaging of TNBCs. Thus, poly(lactic-co-glycolic acid) NPs loaded with the anti-MUC1 aptamer and labeled with technetium-99m were used for a biodistribution study and imaging of TNBC. The results confirmed that the NP was successfully obtained, with a mean size of 262 nm, according to the dynamic light scattering data. The biodistribution assay in induced animal models with TNBC showed that although there was a high capture by intestine (30%), the DDS developed had a high tumor uptake (5%) and with great in vivo imaging properties, corroborating the possibility of use of this DDS as an imaging drug for TNBC. O câncer de mama é o tipo de neoplasia que mais causa mortes entre as mulheres em todo o mundo. Um dos entraves mais prevalentes na área oncológica que correspondem a alta taxa de morbidade é a ausência de detecção precoce dos processos carcinogênicos, especialmente em casos de câncer de mama triplo negativo (TNBC). A não descoberta de um câncer abaixo do nível detectável por testes de rastreios atuais, inclusive por meio de amostras sanguíneas, aponta para a real necessidade de ferramentas tecnológicas inovadoras, que sejam direcionadas especificamente ao microambiente tumoral, com o mínimo de perturbações aos tecidos saudáveis. A utilização de aptâmeros é uma evolução da terapia alvo-dirigida, apresentando considerável nível de sensibilidade a substratos particulares, como a mucina 1 (MUC1). Neste estudo, o aptâmero anti-MUC1 foi o direcionador utilizado em nanopartículas poliméricas radioativas para geração de imagem de TNBCs. Assim, as NPs de poli (ácido láctico-co-glicólico) carregadas com o aptâmero anti-MUC1 e radiomarcadas com tecnetium-99m foram utilizadas para realização de estudo de biodistribuição e imagem de TNBC. Os resultados confirmaram que as NPs foram obtidas com sucesso, com um tamanho médio de 262 nm, de acordo com os dados dinâmicos de dispersão da luz. O teste de biodistribuição em modelos animais induzidos com TNBC mostrou que, embora tenha havido uma alta captura pelo intestino (30%), o sistema de entrega de fármaco (DDS) desenvolvido apresentou uma alta absorção no tumor (5%) e excelentes propriedades de imagem in vivo, corroborando a possibilidade de uso desse DDS como radiofármaco para imagem em TNBC.

The targeting and controlling release of these nanoradiopharmaceuticals ensures greater efficiency and decreases side effects and toxicity. Cutting-edge nanoparticles have specific recognition surface ligands, such as (monoclonal) antibodies, allowing increased recognition of tumor tissue. The nanoparticles of iron oxide (Fe3O4) as magnetite present high surface chemical reactivity, favoring the biomolecules binding as diffusion through tissues and lower toxicity. In this context, and interconnecting last generation technologies such as nuclear and nanotechnology, this work had the objective of producing, characterizing and studying the biodistribution of magnetic nanoparticles decorated with trastuzumab and labeled with technetium-99m radionuclide for the early and accurate detection of breast cancer. The magnetic nanoparticles were prepared by the thermodecomposition method and characterized by transmission electron microscopy (TEM) and high resolution transmission electron microscopy (HRTEM), showing its spherical shape and its diameter of 7 nm. The conjugation of the monoclonal antibody to the surface of the magnetite nanoparticle was performed by means of a carbodiimide reaction. Labeling with technetium-99m was performed by the indirect method and its yield determined by Whatman 1 paper chromatography. The result showed a labeling yield greater than 97%. The SPECT image shows the uptake corresponding to the lesion, which has its location confirmed with the respective bioluminescence image. In the biodistribution study, healthy and induced human breast tumor xenograft-induced MCF-7 Balb/c micewere sacrificed two hours after the administration of the nanoradiopharmaceutical and dissected for counting the organs in the gamma counter. The xenoenxerted tumor had a large caption (> 10% of the total injected dose), corroborating the use of the nanoradiopharmaceutical. The biodistribution demonstrated that the nanoradiopharmaceutical has an excellent renal clearance, and little recognition by the macrophagic system as well as it was not able to permeate the blood-brain barrier, consubstantiating its safety of use. Finally, the nanoradiopharmaceutical showed high affinity to plasma proteins and low accumulation in the cardiac striatum, thus reducing its cardiotoxic effect. The data set establishes a profile of use as image agent for diagnostic purposes in breast cancer. O advento do desenvolvimento de radiofármacos em escala nanométrica, propicia um novo paradigma para a medicina nuclear e desponta como alternativa viável ao tratamento e diagnóstico de tumores. A liberação controlada, específica e direcionada ao alvo correto, garante maior eficiência e diminui os efeitos colaterais e tóxicos. As nanopartículas de última geração possuem ligantes de superfície de reconhecimento específicos, como anticorpos (monoclonais), permitindo o aumento do reconhecimento do tecido tumoral. As nanopartículas de óxido de ferro (Fe3O4) como magnetita apresentam alta reatividade química superficial, favorecendo a ligação de biomoléculas; apresentam difusão através de tecidos e baixa toxicidade. Nesse contexto, e interligando tecnologias de ponta, tais como a nuclear e nanotecnologia, este trabalho teve como objetivo a produção, caracterização e estudo da biodistribuição de nanopartículas magnéticas decoradas com Trastuzumabe marcadas com o radionuclídeo Tecnécio-99m para detecção precoce e precisa de tumores de mama. As nanopartículas magnéticas foram preparadas pelo método de termodecomposição e caracterizadas por microscopia eletrônica de transmissão (MET) e Microscopia eletrônica de transmissão de alta resolução (HRTEM), evidenciando sua forma esférica e seu diâmetro de 7 nm. A conjugação do anticorpo monoclonal à superfície da nanopartícula de magnetita foi realizada por meio de uma reação de carbodiimida. A marcação com Tecnécio-99m foi realizada pelo método indireto e seu rendimento determinado pela cromatografia em papel Whatman nº 1. O resultado demonstrou um rendimento de marcação maior que 97%. A imagem SPECT mostra a captação correspondente à lesão, que tem sua localização confirmada com a respectiva imagem de bioluminescência. No estudo de biodistribuição foram utilizados camundongos Balb/c nude sadios e induzidos com xenoenxerto de tumor de mama humano de linhagem MCF-7, que foram sacrificados duas horas após a administração do nanorradiofármaco e dissecados para contagem dos órgãos no contador gama. O tumor xenoenxertado teve grande captação (> 10% da dose total injetada), corroborando o uso do nanorradiofármaco. A biodistribuição demonstrou que o nanorradiofármaco possui um clearance renal excelente, e pouco reconhecimento pelo sistema macrofágico assim como não foi capaz de permear a barreira hematoencefálica, consubstanciando sua segurança de uso. Por fim, o nanorradiofármaco apresentou grande afinidade às proteínas plasmáticas e baixa acumulação no tecido estriado cardíaco, reduzindo assim seu efeito cardiotóxico. O conjunto dos dados estabelece um perfil de uso como agente de imagem para fins diagnóstico em câncer de mama.

The monomers used were isophorone diisocyanate (IPDI), dimethylolpropionic acid (DMPA) and two types of polyether polyol: poly(propylene glycol) (PPG) and four block copolymers of poly(ethylene glycol) and poly(propylene glycol) (PEG-b-PPG), with different PEG-segments ratio (7%, 17%, 25% and 38%, in terms of copolymer molar mass). Urea groups were formed on the main chains due to the use of hydrazine as a chain extender. The ratio between soft and hard segments was kept the same (NCO/ OH=2.3) to all systems. The incorporation of two different drugs: hydroxyzine (HDZ) and ketoconazole (CTZ), of hydrophilic and hydrophobic natures, respectively, to WPU was evaluated. The WPU were characterized in terms of solids content, pH, viscosity and average particle size, by light scattering (LS). The cast films were characterized by Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA) and scanning electron microscopy (SEM). Release tests were carried out at 37±0.5ºC in different environments similar to physiological conditions of the gastrointestinal system: simulated gastric juice (HCl solution 0.1 N, pH 1.2) and simulated intestinal fluids (phosphate buffered saline solution - PBS, pH 7.4). The content of the released drug was determined by ultraviolet-visible (UV-Vis) spectroscopy. The mechanisms involved and the kinetics of drug release were assessed employing mathematical models and linear regression. The drugs retained their chemical integrity after the dispersion in WPU systems, and the dispersions remained stable by means of sedimentation throughout the study. No significant differences were observed in the morphology of films containing drugs. Polyurethane films studied released HDZ and CTZ for a period of 8h without detecting peaks of drug release, which represents an advantage in the use of these systems. In general, a combination of drug diffusion and polymer erosion was observed. According to kinetic models, HDZ systems followed zero order models and the CTZ systems followed zero order and Higuchi models. WPU systems containing higher amounts of PEG segments (25 and 38%) showed greater efficiency in terms of dissolution and release of the drugs. Os monômeros empregados foram: diisocianato de isoforona (IPDI), ácido dimetilolpropiônico (DMPA) e dois tipos de poliol poliéter: poli (glicol propilênico) (PPG) e quatro copolímeros em bloco à base de poli(glicol etilênico) e de poli(glicol propilênico) (PEG-b-PPG), com variação na proporção de segmentos à base de PEG (7%, 17%, 25% e 38%, em relação à massa molar do copolímero). A hidrazina, como extensor de cadeia, deu origem a grupos ureia. A proporção entre os segmentos flexíveis e rígidos foi mantida a mesma (NCO/OH=2,3) para todos os sistemas. A melhor forma de incorporação dos fármacos hidroxizina (HDZ) e cetoconazol (CTZ), de naturezas hidrofílica e hidrofóbica, respectivamente, nas dispersões pré-formadas foi avaliada. As WPU foram caracterizadas quanto ao teor de sólidos, pH, viscosidade aparente e tamanho médio de partículas por espalhamento de luz (LS). Os filmes produzidos a partir do vazamento das dispersões foram caracterizados por espectroscopia de infravermelho com transformada de Fourier (FTIR), análise termogravimétrica (TGA) e microscopia eletrônica de varredura (SEM). Os testes de liberação in vitro dos fármacos foram efetuados a 37±0,5ºC por dissolução em diferentes meios similares às condições fisiológicas do sistema gastrointestinal: suco gástrico simulado (solução HCl 0,1 N, pH 1,2) e suco entérico simulado (solução de tampão fosfato-salino - PBS, pH 7,4). A quantificação dos fármacos foi determinada por espectroscopia na região do ultravioleta visível (UV-Vis). Os mecanismos envolvidos e a cinética de liberação dos fármacos foram determinados através da aplicação de modelos matemáticos e análise da regressão linear. Os resultados indicaram que os fármacos mantiveram sua integridade química após dispersão nos sistemas WPU, que se mantiveram estáveis em termos de sedimentação ao longo do período do estudo. Não foram verificadas diferenças significativas na morfologia dos filmes contendo os fármacos. Os filmes poliuretânicos propiciaram a liberação da HDZ e do CTZ por um período de 8 h, sem que picos de liberação dos fármacos fossem detectados, indicando vantagem no emprego desses sistemas. Em geral, obteve-se uma combinação de difusão do fármaco e erosão do polímero. De acordo com os modelos cinéticos, sistemas contendo HDZ obedeceram ao modelo de ordem zero e os sistemas com CTZ obedeceram aos modelos de ordem zero e de Higuchi. Os sistemas WPU contendo maiores quantidades de segmentos à base de PEG (25 e 38%) apresentaram maior eficiência em termos de dissolução e liberação dos fármacos. No entanto, as dispersões contendo menores quantidades (7 e 17%) se mostraram mais apropriadas, visto que a liberação ocorreu de forma mais lenta
