The vehicular edge computing (VEC) can cache contents in different RSUs at the network edge to support the real-time vehicular applications. In VEC, owing to the high-mobility characteristics of vehicles, it is necessary to cache the user data in advance and learn the most popular and interesting contents for vehicular users. Since user data usually contains privacy information, users are reluctant to share their data with others. To solve this problem, traditional federated learning (FL) needs to update the global model synchronously through aggregating all users' local models to protect users' privacy. However, vehicles may frequently drive out of the coverage area of the VEC before they achieve their local model trainings and thus the local models cannot be uploaded as expected, which would reduce the accuracy of the global model. In addition, the caching capacity of the local RSU is limited and the popular contents are diverse, thus the size of the predicted popular contents usually exceeds the cache capacity of the local RSU. Hence, the VEC should cache the predicted popular contents in different RSUs while considering the content transmission delay. In this paper, we consider the mobility of vehicles and propose a cooperative Caching scheme in the VEC based on Asynchronous Federated and deep Reinforcement learning (CAFR). We first consider the mobility of vehicles and propose an asynchronous FL algorithm to obtain an accurate global model, and then propose an algorithm to predict the popular contents based on the global model. In addition, we consider the mobility of vehicles and propose a deep reinforcement learning algorithm to obtain the optimal cooperative caching location for the predicted popular contents in order to optimize the content transmission delay. Argostemma ehuangzhangense, a new Rubiaceae species from E’huangzhang Nature Reserve, Guangdong Province, China, is here described and illustrated. A morphological comparison between the new species and its putative relatives, A. lamxayanum, A. laotica and A. verticillatum, is presented. The new species is mostly similar to A. laotica, but they can be distinguished from each other since Argostemma ehuangzhangense presents solitary flower (vs. 2-flowered inflorescences), flower lobes 4 (vs. 5) and anthers opening by longitudinal slits (vs. apical pores). In a preliminary IUCN Red List status of Argostemma ehuangzhangense this species is assigned as Vulnerable (VU).

Paraphlomis jinggangshanensis (Lamiaceae), a new species from Jiangxi Province, China, is described and illustrated. The new species is morphologically similar to P. intermedia, but can be easily distinguished from the latter by its cordate leaf base (vs. cuneate, decurrent), stem and calyx tube with glandular hairs (vs. short pubescent), and glabrous anthers (vs. ciliate anthers). A phylogenetic analysis, based on ITS regions, suggests that P. jinggangshanensis represents a separate branch in Paraphlomis and is closely related to Clade II. It is currently known only from Jinggangshan National Natural Reserve. Because of its limited distribution and small population size, the species was assessed as Near Threatened (NT) according to the IUCN Red List Categories and Criteria.

Vehicular edge computing (VEC) is envisioned as a promising approach to process the explosive computation tasks of vehicular user (VU). In the VEC system, each VU allocates power to process partial tasks through offloading and the remaining tasks through local execution. During the offloading, each VU adopts the multi-input multi-out and non-orthogonal multiple access (MIMO-NOMA) channel to improve the channel spectrum efficiency and capacity. However, the channel condition is uncertain due to the channel interference among VUs caused by the MIMO-NOMA channel and the time-varying path-loss caused by the mobility of each VU. In addition, the task arrival of each VU is stochastic in the real world. The stochastic task arrival and uncertain channel condition affect greatly on the power consumption and latency of tasks for each VU. It is critical to design an optimal power allocation scheme considering the stochastic task arrival and channel variation to optimize the long-term reward including the power consumption and latency in the MIMO-NOMA VEC. Different from the traditional centralized deep reinforcement learning (DRL)-based scheme, this paper constructs a decentralized DRL framework to formulate the power allocation optimization problem, where the local observations are selected as the state. The deep deterministic policy gradient (DDPG) algorithm is adopted to learn the optimal power allocation scheme based on the decentralized DRL framework. Background The factors that maintain phenotypic and genetic variation within a population have received long-term attention in evolutionary biology. Here the genetic basis and evolution of the geographically widespread variation in twig trichome color (from red to white) in a shrub Melastoma normale was investigated using Pool-seq and evolutionary analyses. Results The results show that the twig trichome coloration is under selection in different light environments and that a 6-kb region containing an R2R3 MYB transcription factor gene is the major region of divergence between the extreme red and white morphs. This gene has two highly divergent groups of alleles, one of which likely originated from introgression from another species in this genus and has risen to high frequency (> 0.6) within each of the three populations under investigation. In contrast, polymorphisms in other regions of the genome show no sign of differentiation between the two morphs, suggesting that genomic patterns of diversity have been shaped by homogenizing gene flow. Population genetics analysis reveals signals of balancing selection acting on this gene, and it is suggested that spatially varying selection is the most likely mechanism of balancing selection in this case. Conclusions This study demonstrate that polymorphisms on a single transcription factor gene largely confer the twig trichome color variation in M. normale, while also explaining how adaptive divergence can occur and be maintained in the face of gene flow.

During the flotation of metal sulfide minerals, due to the interference of unavoidable ions, the quartz also partially floats in some cases. The research on the mechanism of quartz being activated and floating up is still insufficient. In this study, the influence of unavoidable ions Cu2+ and Ni2+ on the floatation of quartz was studied by micro-flotation experiments, adsorption detection, zeta potential measurement, solution composition calculation, infrared spectroscopy (FT-IR) analysis, X-ray photoelectron spectroscopy (XPS) analysis, and atomic force microscopy (AFM) observation. This provides a theoretical reference for further understanding the mechanism of sodium ethylxanthogenate and quartz surface, as well as the development of new quartz depressant. The results of flotation showed that after activation by Cu2+(1×10-4 mol/L) and Ni2+(5×10-5 mol/L), quartz was captured by sodium ethylxanthogenate (EX:1.4×10-4 mol/L) under alkaline conditions, while the best recoveries were 80% and 43%, respectively. The results of adsorption measurement and Zeta potential measurement showed that the precipitation rate of Cu2+ was greater than that of Ni2+ under alkaline conditions. In addition, both Cu2+ and Ni2+ were electrostatically adsorbed on the quartz surface and changed the Zeta potential of quartz. The solution composition calculation further showed that Cu(OH)+, Cu(OH)2(s), and Ni(OH)+, Ni(OH)2(s) were the main components in the solution under alkaline conditions. FT-IR analysis, XPS analysis, and AFM observations revealed that Cu species and Ni species were adsorbed on O atoms on the quartz surface, providing active sites for EX adsorption.

Phylogenetic studies in the phylogenomics era have demonstrated that reticulate evolution greatly impedes the accuracy of phylogenetic inference, and consequently can obscure taxonomic treatments. However, the systematics community lacks a broadly applicable strategy for taxonomic delimitation in groups identified to have pervasive reticulate evolution. The red-fruit genus,Stranvaesia, provides an ideal model for testing the effect of reticulation on generic circumscription when hybridization and allopolyploidy define a group’s evolutionary history. Here, we conducted phylogenomic analyses integrating data from hundreds of single-copy nuclear (SCN) genes and plastomes, and interrogated nuclear paralogs to clarify the inter/intra-generic relationship ofStranvaesiaand its allies in the framework of Maleae. Analyses of phylogenomic discord and phylogenetic networks showed that allopolyploidization and introgression promoted the origin and diversification of theStranvaesiaclade, a conclusion further bolstered by cytonuclear and gene tree discordance. The well-inferred phylogenetic backbone revealed an updated generic delimitation ofStranvaesiaand a new genus,Weniomeles, characterized by purple-black fruits, trunk and/or branches with thorns, and fruit core with multilocular separated by a layer of sclereids and a cluster of sclereids at the top of the locules. Here, we highlight a broadly-applicable workflow for inferring how analyses of reticulate evolution in phylogenomic data can directly shape taxonomic revisions.

Sedum jinglanii, a new species of Crassulaceae from Mount Danxia in Guangdong, China, is described and illustrated. Phylogenetic analysis based on the internal transcribed spacer (ITS) region of nrDNA suggests that the new species belongs to S. sect. Sedum sensu Fu and Ohba (2001) in the “Flora of China”, and is sister to a clade comprising S. alfredi and S. emarginatum with high support values (SH-aLRT = 84, UFBS = 95) but is distantly related to S. baileyi. The new species is morphologically similar to S. alfredi but it can be distinguished from the latter in its opposite leaves (vs. alternate leaves), its usually wider leaves (0.4–1.2 cm vs. 0.2–0.6 cm), its usually shorter petals (3.4–4.5 mm vs. 4–6 mm), its shorter nectar scales (0.4–0.5 mm vs. 0.5–1 mm), its shorter carpels (1.5–2.6 mm vs. 4–5 mm), and its shorter styles (0.6–0.9 mm vs. 1–2 mm). The new species can be easily distinguished from S. emarginatum which both have opposite leaves by its short, erect or ascending rhizome (vs. long and prostrate rhizome in the latter), shorter petals (3.4–4.5 mm vs. 6–8 mm) and shorter carpels (1.5–2.6 mm vs. 4–5 mm). It can also be easily distinguished from S. baileyi by its short, erect or ascending rhizome (vs. long and prostrate rhizome) and its shorter style (0.6–0.9 mm vs. 1–1.5 mm).

The diverse members of the genus Daphne are prized for their fragrant flowers. Despite being promising ornamental plants in many countries, genetic information of Daphne is scarce. In this study, the plastomes of four species and one variety of Daphne were sequenced and analyzed. The plastomes were typical and contained a pair of inverted repeat (IR) regions that separated the large single-copy (LSC) region from the small single-copy (SSC) region. With a length ranging from 132,869 bp (D. genkwa) to 174,773 bp (Daphne championii), 106 to 141 genes were predicted. Comparative plastome analysis of the newly sequenced plastomes with four publicly available Daphne plastomes identified an expansion of the IRs, sequence variations, and mutational hotspots. Phylogenetic analyses indicated that the genus Daphne in its current circumscription is polyphyletic. Daphne genkwa was nested within the genus Wikstroemia, while D. championii was well resolved as sister to Edgeworthia. These findings concurred with results from our study that used nuclear ribosomal internal transcribed spacer sequence data. The conflicts on the molecular placement of D. championii and D. genkwa and the present taxonomic classification in Daphne suggest that a new intergeneric classification system of Daphneae warrants consideration.

This work reveals that the anisotropic layer Ir2Cl2O2 exhibits high TE performance, which confirms that it is feasible to screen excellent TE materials via high-throughput calculations.

Chymotrypsin-like elastase 1 (CELA1) is a serine protease that is neutralized by α1-antitrypsin (AAT) and prevents emphysema in a murine antisense oligonucleotide model of AAT-deficient emphysema. Mice with genetic ablation of AAT do not have emphysema at baseline but develop emphysema with injury and aging. We tested the role of CELA1 in emphysema development in this genetic model of AAT-deficiency following tracheal lipopolysacharide (LPS), 8 months of cigarette smoke (CS) exposure, aging, and a low-dose tracheal porcine pancreatic elastase (LD-PPE) model. In this last model, we performed proteomic analysis to understand differences in lung protein composition. We were unable to show that AAT-/- mice developed more emphysema than wild type with LPS. In the LD-PPE model, AAT-/- mice developed progressive emphysema from which Cela1-/-&AAT-/- mice were protected. In the CS model, Cela1-/-&AAT-/- mice had worse emphysema than AAT-/-, and in the aging model, 72-75 week-old Cela1-/-&AAT-/- mice had less emphysema than AAT-/- mice. Proteomic analysis of AAT-/- vs. wildtype lungs in the LD-PPE model showed reduced amounts of AAT proteins and increased amounts of proteins related to Rho and Rac1 GTPases and protein oxidation. Similar analysis of Cela1-/-&AAT-/- vs. AAT-/- lungs showed differences in neutrophil degranulation, elastin fiber synthesis, and glutathione metabolism. Thus, Cela1 prevents post-injury emphysema progression in AAT-deficiency, but it has no effect and potentially worsens emphysema in response to chronic inflammation and injury. Prior to developing anti-CELA1 therapies for AAT-deficient emphysema, an understanding of why and how CS exacerbates emphysema in Cela1 deficiency is needed.

Paraphlomis yingdeensis (Lamiaceae), a new species from the limestone area in northern Guangdong Province, China, is described and illustrated. Phylogenetic analyses, based on two nuclear DNA regions (ITS and ETS) and three plastid DNA regions (rpl32-trnL, rps16 and trnL-trnF), suggest that P. yingdeensis represents a distinct species in Paraphlomis. Morphologically, P. yingdeensis is similar to P. foliata subsp. montigena and P. nana, but can be distinguished from the former by its densely villous lamina and calyx, not decurrent base of lamina and bristle-like-acuminate apex of calyx teeth, and distinguished from the latter by its significantly taller plant (15–20 cm vs. 1–5 cm) and larger lamina (6.2–16.5 × 4–11.5 vs. 2–7 × 1.5–4 cm), densely villous stem, lamina and calyx and yellow corolla.

Abnormal nuclear morphology is suggested to be a hallmark of aging and one such abnormality is nuclear blebbing. However, little is known about whether and how nuclear blebbing participates in animal aging, and what regulates it. In this study, we show that the frequency of nuclear blebbing in the hypodermis increases during aging in wild-type C. elegans. These nuclear blebs are enveloped by the nuclear lamina, the inner and the outer nuclear membrane, and 42% of them contain chromatin. Although nuclear blebbing could lead to DNA loss if chromatin-containing blebs detach and fuse with lysosomes, we find by time-lapse imaging that nuclear blebs rarely detach, and the estimated lifetime of a nuclear bleb is 772 h or 32 days. The amount of DNA lost through nuclear blebbing is estimated to be about 0.1% of the total DNA loss by adult Day 11. Furthermore, the frequency of nuclear blebbing does not correlate with the rate of aging in C. elegans. Old age does not necessarily induce nuclear blebbing, neither does starvation, heat stress, or oxidative stress. Intriguingly, we find that proliferation of germ cells promotes nuclear blebbing.

Background Although laparoscopic total extraperitoneal (TEP) inguinal hernia repair has the advantages of less bleeding, less trauma, less pain, and fast recovery, there are several issues that need to be addressed. This study aims to evaluate the effectiveness of preperitoneal closed‑suction drainage on reducing postoperative complications in TEP inguinal hernia repair. Methods A retrospective analysis of 122 patients who underwent TEP inguinal hernia repair between June 2018 and June 2021 was performed. The patients were divided into the drainage group and the non-drainage group according to whether the drainage tube was placed or not. Clinical data, surgical procedures and outcome of these patients were collected and analyzed to assess the effectiveness of drainage. Results A total of 122 patients undergoing TEP surgery were screened, of which 22 were excluded. Most of the patients were male with right indirect inguinal hernia. There was no difference in the mean length of hospital stay between the two groups. Postoperative pain was alleviated by preperitoneal closed‑suction drainage 24 h after operation (p = 0.03). The rate of complications such as scrotal edema, seroma and urinary retention in the drainage group was significantly lower than that in the non-drainage group (p 10 cm3 might receive more clinical benefits by placing drainage tube. Conclusion In TEP inguinal hernia repair, placing drainage tube is a simple and feasible traditional surgical treatment, which can promote postoperative recovery without increasing the risk of infection, especially in patients with large hernia sac volume.

Commelina danxiaensis (Commelinaceae), a remarkable new species from Mount Danxia, Guangdong Province, China, is described and illustrated. This species is similar to C. communis in inflorescences and flowers but readily distinguishable in its nearly erect stems, larger flowers, and different petal colouration.

Global climate changes during the Miocene could have created ample opportunities for hybridization between members of tropical and subtropical biomes at the boundary between those zones, yet very few studies have tested this hypothesis. The Yunnan-Guizhou Plateau (YGP) in Southwest China is a biodiversity hotspot of vascular plants, located at a transitional area between the floristic regions of tropical Southeast Asia and subtropical East Asia. The genus Eriobotrya (Rosaceae) comprises both tropical and subtropical taxa, and has 12 species recorded from the YGP, making it a good system to test the hypothesis of between-biome hybridization. Therefore, we surveyed the evolutionary history of Eriobotrya by examining three chloroplast regions plus five nuclear genes for 817 individuals (47 populations) of 23 Eriobotrya species (including 19 populations of 12 species in the YGP), plus genome re-sequencing of 33 representative samples. We concluded that: 1) phylogenetic positions for 16 species (lineages) exhibited strong cyto-nuclear conflicts, most likely due to ancient hybridization; 2) The YGP is a hotspot for hybridization, where 11 species show clear evidence of chloroplast capture; 3) Eriobotrya likely originated in tropical Asia during the Eocene. From the Miocene onwards the intensification of the Eastern Asia Monsoon and global cooling may have caused shifts of the tropical-subtropical boundary and secondary contact between species, and thus provided ample opportunity to hybridization and diversification of Eriobotrya species, especially in the YGP. Our study highlights the significant role that paleo-climate changes likely played in driving hybridization and generating rich species diversity in climate transition zones.

A gold nanocluster Au17Cd2(PNP)2(SR)12 (PNP = 2,6-bis(diphenylphosphinomethyl)pyridine, SR = 4-MeOPhS) consisting of an icosahedral Au13 kernel, two Au2CdS6 staple motifs, and two PNP pincer ligands has been designed, synthesized and well characterized. This cadmium and PNP pincer ligand co-modified gold nanocluster showed high catalytic efficiency in the KA2 reaction, featuring high TON, mild reaction conditions, broad substrate scope as well as catalyst recyclability. Comparison of the catalytic performance between Au17Cd2(PNP)2(SR)12 and the structurally similar single cadmium (or PNP) modified gold nanoclusters demonstrates that the co-existence of the cadmium and PNP on the surface is crucial for the high catalytic activity of the gold nanocluster. This work would be enlightening for developing efficient catalysts for cascade reactions and discovering the catalytic potential of metal nanoclusters in organic transformations.

An Au4Ni2 nanocluster containing a square-planar [PPh2–Au–S–Au]2 ring and two nickel-pincer arms was found to be assembled by noncovalent interactions. The assembly-dependent properties that are distinct from the octahedral M6 clusters were shown.

Background The commonly used technique for treating unstable pelvic fractures with sacroiliac screws and anterior internal fixator (INFIX) is prone to complications, such as injury to the pelvic vasculature and nerves, life-threatening bleeding, lateral femoral cutaneous neuritis, and wound infection. This study investigated the clinical effects of using a modified percutaneous iliosacral screw and INFIX technique for treating unstable pelvic fractures. Methods A retrospective analysis of minimally invasive internal fixation using modified incision of an anterior-ring INFIX application combined with modified percutaneous iliosacral screw placement was performed for 22 cases of unstable pelvic fractures from January 2017 to December 2018. Based on the Tile classification, there were 4 type B1, 7 type B2, 5 type B3 and 6 type C1 injuries. Preoperatively, the length and orientation of the internal fixation were computer-simulated and measured. On postoperative day 3, pelvic radiographs and three-dimensional computed tomograms were used to assess fracture reduction and fixation. All patients were regularly followed up at 4 weeks, 12 weeks, 6 months, 12 months, 24 months and annually thereafter. Fracture healing, complications, visual analogue scale (VAS) scores, the quality of fracture repositioning and Majeed score were assessed during follow-up. Results All patients were followed up for a mean of 25.23 ± 1.48 months. All fractures healed without loss of reduction and no patient showed evidence of delayed union or nonunion. Two years postoperatively, the mean VAS score was 0.32 ± 0.09 and the mean Majeed score was 94.32 ± 1.86. Conclusion The modified percutaneous iliosacral screw technique increases the anterior tilt of the sacroiliac screw by shifting the entry point posteriorly to increase the safety of the screw placement. Downward modification of the INFIX incision reduces the risk of lateral femoral cutaneous nerve injury. This technique is safe, effective and well tolerated by patients.

Wearable electronics have enriched daily lives by providing smart functions as well as monitoring body health conditions. However, the realization of wearable electronics with personal healthcare and thermal comfort management of the human body is still a great challenge. Furthermore, manufacturing such on-skin wearable electronics on traditional thin-film substrates results in limited gas permeability and inflammation. Herein, we proposed a personal healthcare and thermal management smart textile with a three-dimensional (3D) interconnected conductive network, formed by silver nanowires (AgNWs) bridging lamellar structured transition-metal carbide/carbonitride (MXene) nanosheets deposited on nonwoven fabrics. Benefiting from the interconnected conductive network synergistic effect of one-dimensional (1D) AgNWs bridging two-dimensional (2D) MXene, the strain sensor exhibits excellent durability (>1500 stretching cycles) and high sensitivity (gauge factor (GF) = 1085) with a wide strain range limit (∼100%), and the details of human body activities can be accurately recognized and monitored. Moreover, thanks to the excellent Joule heating and photothermal effect endowed by AgNWs and MXene, the multifunctional smart textile with direct temperature visualization and solar-powered temperature regulation functions was successfully developed, after further combination of thermochromic and phase-change functional layers, respectively. The smart textiles with a stretchable AgNW-MXene 3D conductive network hold great promise for next-generation personal healthcare and thermal management wearable systems.

Lespedeza danxiaensis (Fabaceae), a new species from Danxiashan National Nature Reserve in Guangdong Province, is described and illustrated. The new species is morphologically similar to Lespedeza pilosa, but it can be easily distinguished by its thin leathery leaflets and long peduncles. Phylogenetic analysis based on ITS confirmed that the new species belongs to Lespedeza subg. Macrolespedeza. The new species is the first known species of Lespedeza endemic to Danxia landform and is currently only known from Mount Danxia, Guangdong.

Background Lung adenocarcinoma (LUAD) is a common subtype of lung cancer with high recurrence rate and fatality. Circ_0001361 has been recognized as key regulators in various malignancies, but its roles in LUAD remain ambiguous. Methods Circ_0001361, miR-525-5p, and VMA21 levels were assessed by RT-qPCR. The growth and metastasis of LUAD cells were detected by MTT, colony formation, wound scratch, and transwell assays, respectively. The interaction between circ_0001361/VMA21 and miR-525-5p was detected by dual luciferase, RNA immunoprecipitation, and RNA pull-down assays. VMA21 protein level was detected by Western blotting. Nude mouse xenograft model was established to determine the role of circ_0001361 in tumor growth in vivo. Results Circ_0001361 was up-regulated, while miR-525-5p was down-regulated in LUAD tissues and cells. Functional experiments demonstrated that circ_0001361 drove LUAD cell growth and metastasis. Mechanistically, circ_0001361 functioned as a sponge of miR-525-5p to up-regulate downstream target VMA21 level. MiR-525-5p/VMA21 axis was involved in circ_0001361-mediated malignant phenotypes of LUAD cells. Finally, inhibition of circ_0001361 restrained in vivo xenograft tumor growth via regulating miR-525-5p/VMA21 axis. Conclusion Our findings elucidate that circ_0001361 facilitates the tumorigenesis and development of LUAD through miR-525-5p/VMA21 axis, providing evidence for circ_0001361 as a potential prognosis biomarker and therapeutic target for clinical treatment of LUAD.

After fertilization, zygotic genome activation (ZGA) marks the beginning of the embryonic program for a totipotent embryo, which further gives rise to the pluripotent embryonic lineages and extraembryonic trophectoderm after the first lineage commitment. While much has been learned about pluripotency regulation, how ZGA is connected to the pluripotency commitment in early embryos remains elusive. Here, we investigated the role of nuclear receptor1family transcription factors (TFs) in mouse pre-implantation embryos, whose motifs are highly enriched in accessible chromatin at the 2-cell (2C) to 8-cell (8C) stages after ZGA. We found NR5A2, an NR TF highly induced upon ZGA, is required for early development, as both the knockdown and knockout ofNr5a2from 1C embryos led to morula arrest. While the zygotic genome was largely activated at the 2C stage, 4-8C-specific gene activation (mid-preimplantation activation) was substantially impaired. Genome-wide chromatin binding and RNA-seq analyses showed NR5A2 preferentially regulates its binding targets including a subset of key pluripotency genes (i.e.,Nanog, Pou5f1, andTdgf1). Finally, NR5A2-occupied sites at the 2C and 8C stages predominantly reside in accessible B1 elements where its motif is embedded. Taken together, these data identify NR5A2 as a key regulator that connects ZGA to the first lineage segregation in early mouse development.

The new species Eriobotrya crassifolia, collected from Yunnan Province, China, is characterised and illustrated. A phylogeny based on chloroplast genomes supported its closest affinity with E. tengyuehensis, while a phylogeny based on 197 single-copy nuclear genes supported its closest affinity with E. fragrans and E. deflexa. Morphologically, however, it resembles E. angustissima. Nevertheless, it can be easily distinguished from E. angustissima by its abaxially retroflexed and sharply serrate leaf margins, densely rusty tomentose inflorescences, and oblong or elliptic leaves.

Translational regulation plays a critical role during the oocyte-to-embryo transition (OET) and zygotic genome activation (ZGA). Here, we integrated ultra-low-input ribosome profiling (Ribo-lite) with messenger RNA sequencing to co-profile the translatome and transcriptome in human oocytes and early embryos. Comparison with mouse counterparts identified widespread differentially translated gene functioning in epigenetic reprogramming, transposon defense, and small RNA biogenesis, in part driven by species-specific regulatory elements in 3′ untranslated regions. Moreover, PRD-like homeobox transcription factors, including TPRXL , TPRX1 , and TPRX2 , are highly translated around ZGA. TPRX1/2/L knockdown leads to defective ZGA and preimplantation development. Ectopically expressed TPRXs bind and activate key ZGA genes in human embryonic stem cells. These data reveal the conservation and divergence of translation landscapes during OET and identify critical regulators of human ZGA.

Abnormal nuclear morphology is suggested to be a hallmark of aging. One type of such abnormalities is nuclear blebbing, but little is known about whether and how nuclear blebbing participates in animal aging. What regulates nuclear blebbing is also unknown. In this study, we show that the frequency of nuclear blebbing in the hypodermis increases during aging in wild-type C. elegans. These nuclear blebs are enveloped by the nuclear lamina, the inner and the outer nuclear membrane, and 42% of them contain chromatin. Detachment of a bleb from the nucleus is rare but does happen, thereby generating cytoplasmic chromatin. Cytoplasmic chromatin-containing lysosomes juxtaposing the nucleus are detected in old worms. Therefore, nuclear blebbing contributes to the age-associated chromatin loss. However, the frequency of nuclear blebbing does not correlate with the rate of aging in C. elegans. Old age does not necessarily induce nuclear blebbing, neither does starvation, heat stress, or oxidative stress. To prevent the scale formation in the equipments and pipelines after pre-treated coal gasification gray water (CGGW) entering the reuse system and reduce the influence of various pollutants in the effluent on subsequent biochemical treatment, this study presented a coupled use of pulse electrocoagulation (PEC) and chemical precipitation (CP) coupling method for the pretreatment of coal gasification gray water (CGGW). In addition, the operation parameters of PEC and the reaction conditions of PEC-CP were optimized based on iron plate as electrode and total hardness, turbidity and sludge yield as assessment indicators.
