Autophagy (‚self-eating‘) is a major catabolic pathway. It plays a role in energy metabolism, but also in the development of different diseases. One of the first complexes to assemble during autophagy is the Atg1-kinase complex. For this study, all subunits of the Atg1-kinase complex were recombinantly expressed and analyzed. Assembly, disassembly and architecture of the complex could be investigated in vitro. Furthermore, it was shown how assembly of the complex is involved in regulating and promoting autophagy. Die Autophagozytose ist ein zentraler Abbaumechanismus in der Zelle. Sie spielt nicht nur eine Rolle im Stoffwechsel, sondern auch bei der Entstehung von Krankheiten. Der Atg1-kinase Komplex ist essentiell für ihre Regulation. Seine Untereinheiten wurden für diese Arbeit rekombinant exprimiert und analysiert. Die (Dis)assemblierung, sowie die Architektur des Komplexes konnten im Reagenzglas untersucht werden. Außerdem konnte gezeigt werden, wie der Zusammenbau des Komplexes die Autophagozytose reguliert und antreibt.
