Two time series of financial losses may be observed in different overlapping windows, serially dependent, heteroscedastic, and cross-sectionally dependent. Fitting a regression model to each of the two time series, we construct an improved least squares estimator in one series exploiting the cross-sectional dependence with the other series. We employ a random weight bootstrap method to define the new estimator and to establish its asymptotic normality. The developed inference is robust against heteroscedasticity as we do not estimate the GARCH errors. Simulations confirm the efficiency improvement through substantial variance reduction, especially when the cross-sectional dependence is strong and the second series is longer. We illustrate the usefulness of the method by analyzing mutual funds' returns.

Its pathophysiological mechanism is unclear, which limits the development of therapeutic strategies. Long non-coding RNA (lncRNA) single nucleotide polymorphisms (SNPS) may be related. In this study, we aimed to determine the effects of the rs2242385, rs155979, rs3762983, and rs3762984 polymorphisms in the lncRNA NONHSAT102891 on depression susceptibility in a Chinese population.Methods: We conducted a case-control study in a cohort of 480 patients with depression and 329 healthy controls, and performed genotyping by gene sequencing ii.Results: The rs155979 GC genotype was significantly associated with increased risk of depression compared with healthy controls. Stratified analysis showed a 2.08-fold increased risk of suicide in patients with rs155979 GC or GG genotype. The rs2242385, rs3762983, and rs3762984 polymorphisms were not significantly associated with the risk of depression. Haploid analysis showed linkage disequilibrium between rs155979, rs3762983, and rs3762984, and the CCG haplotype reduced the risk of depression.Limitations: The study sample was relatively small, and was restricted to patients from central and southern China. Further, only peripheral blood was used for DNA extraction.Conclusion: The rs155979 polymorphism may be associated with the occurrence of depression in the Chinese population. However, further studies are needed to verify the reliability of our results in large populations and different ethnic groups.Keywords: depression, SNP, LncRNA

Increasing evidence indicates that glioma topographic location is linked to the cellular origin, molecular alterations and genetic profile. This research aims to (a) reveal the underlying mechanisms of tumor location predilection in glioblastoma multiforme (GBM) and lower-grade glioma (LGG) and (b) leverage glioma location features to predict prognosis. MRI images from 396 GBM and 190 LGG (115 astrocytoma and 75 oligodendroglioma) patients were standardized to construct frequency maps and analyzed by voxel-based lesion-symptom mapping. We then investigated the spatial correlation between glioma distribution with gene expression in healthy brains. We also evaluated transcriptomic differences in tumor tissue from predilection and nonpredilection sites. Furthermore, we quantitively characterized tumor anatomical localization and explored whether it was significantly related to overall survival. Finally, we employed a support vector machine to build a survival prediction model for GBM patients. GBMs exhibited a distinct location predilection from LGGs. GBMs were nearer to the subventricular zone and more likely to be localized to regions enriched with synaptic signaling, whereas astrocytoma and oligodendroglioma tended to occur in areas associated with the immune response. Synapse, neurotransmitters and calcium ion channel-related genes were all activated in GBM tissues coming from predilection regions. Furthermore, we characterized tumor location features in terms of a series of tumor-to-predilection distance metrics, which were able to predict GBM 1-year survival status with an accuracy of 0.71. These findings provide new perspectives on our understanding of tumor anatomic localization. The spatial features of glioma are of great value in individual therapy and prognosis prediction.

The tropical Pacific currents from January 2004 to December 2018 are computed based on the gridded Argo temperatures and salinities using the P-vector method on an f plane and the geostrophic approximation on a β plane. Three branches of the South Equatorial Current (SEC) are identified, i.e., SEC(N) (2°S–5°N), SEC(M) (7°–3°S), and SEC(S) (20°–8°S), with the maximum zonal velocity of −55 cm s−1 and total volume transport of −49.8 Sv (1 Sv ≡ 106 m3 s−1) occurring in the central-east Pacific. The seasonal variability of each branch shows a distinct and different westward propagation of zonal current anomalies, which are well mirrored by the SLA differences between 2°S and 5°N, between 3°S and 6°S, and between 8°S and 15°S, respectively. Most of the seasonal variations are successfully simulated by a simple analytical Rossby wave model, highlighting the significance of the first-mode baroclinic, linear Rossby waves, particularly those driven by the wind stress curl in the central-east Pacific. However, the linear theory fails to explain the SEC(M) variations in certain months in the central-east Pacific, where the first baroclinic mode contributes only around 50% of the explained variance to the equatorial surface currents. A nonlinear model involving higher baroclinic modes is suggested for a further diagnosis. Considering the crucial role played by the tropical Pacific in the natural climate variability via the El Niño–Southern Ocean dynamics and the ocean response to anthropogenic forcing via the ocean heat uptake in the eastern tropical Pacific, advancing the process understanding of the SEC from observations is critical.

Three mononuclear copper(II) and nickel(II) complexes, [Cu(L1)(NCS)(CH3OH)] (1), [Cu(L2)(NCS)] (2) and [Ni(L2) (N3)] (3), where L1 and L2 are the monoanionic forms of the Schiff bases N’-(pyridin-2-ylmethylene)picolinohydrazide (HL1) and 4-methyl-2-(((pyridin-2-ylmethyl)imino)methyl)phenol (HL2), have been prepared and characterized by elemental analysis, IR and UV-Vis spectroscopy, as well as single crystal X-ray diffraction studies. The Cu atom in complex 1 is in a square pyramidal coordination, with the three N atoms of the ligand L and the N atom of the thiocyanate ligand in the basal plane, and with the methanol O atom at the apical position. The Cu and Ni atoms in complexes 2 and 3 are in square planar coordination, with the three donor atoms of the Schiff base ligands and the terminal N atoms of thiocyanate and azide ligands. Complexes 1 and 2 inhibit the Jack bean urease with IC50 value of 0.33 ± 0.12 and 0.39 ± 0.10 μmol L–1, respectively. Molecular docking study was performed to investigate the interaction between the complexes and the enzyme.

Diosgenin, a steroidal saponin, is a natural product found in many plants. Diosgenin has a wide range of pharmacological activities, and has been used to treat cancer, nervous system diseases, inflammation, and infections. Numerous studies have shown that diosgenin has potential therapeutic value for lipid metabolism diseases via various pathways and mechanisms, such as controlling lipid synthesis, absorption, and inhibition of oxidative stress. These mechanisms and pathways have provided ideas for researchers to develop related drugs. In this review, we focus on data from animal and clinical studies, summarizing the toxicity of diosgenin, its pharmacological mechanism, recent research advances, and the related mechanisms of diosgenin as a drug for the treatment of lipid metabolism, especially in obesity, hyperlipidemia, nonalcoholic fatty liver disease, atherosclerosis, and diabetes. This systematic review will briefly describe the advantages of diosgenin as a potential therapeutic drug and seek to enhance our understanding of the pharmacological mechanism, recipe-construction, and the development of novel therapeutics against lipid metabolism diseases.

Microbial electrosynthesis (MES) employs microbial catalysts and electrochemistry to enhance CO2 bioconversion to organics with concurrent waste biorefining capability. The aim of this review is to comprehensively discuss the current state of the art and prospects of medium chain fatty acids (MCFAs) production in MES from CO2 and waste organics. Fundamental mechanisms and development of MCFAs production via conventional fermentation are introduced as well. Studies on MCFAs production in MES are summarized, highlighting the strategy of multiple-electron donors (EDs). Challenges for MCFAs production in MES from CO2 are presented, and the primary discussions included methanogenesis inhibition, adenosine triphosphate (ATP) limitations of acetogens, and production of limited EDs via solventogenesis. Possible applications of electrochemical approaches to promote the bioconversion of actual waste materials with MCFAs production are analyzed. Finally, future directions are explored, including multi-stage reactions, substrate supply, product extraction, and microbial pathways.

Meeting online is becoming the new normal. Creating an immersive experience for online meetings is a necessity towards more diverse and seamless environments. Efficient photorealistic rendering of human 3D dynamics is the core of immersive meetings. Current popular applications achieve real-time conferencing but fall short in delivering photorealistic human dynamics, either due to limited 2D space or the use of avatars that lack realistic interactions between participants. Recent advances in neural rendering, such as the Neural Radiance Field (NeRF), offer the potential for greater realism in metaverse meetings. However, the slow rendering speed of NeRF poses challenges for real-time conferencing. We envision a pipeline for a future extended reality metaverse conferencing system that leverages monocular video acquisition and free-viewpoint synthesis to enhance data and hardware efficiency. Towards an immersive conferencing experience, we explore an accelerated NeRF-based free-viewpoint synthesis algorithm for rendering photorealistic human dynamics more efficiently. We show that our algorithm achieves comparable rendering quality while performing training and inference 44.5% and 213% faster than state-of-the-art methods, respectively. Our exploration provides a design basis for constructing metaverse conferencing systems that can handle complex application scenarios, including dynamic scene relighting with customized themes and multi-user conferencing that harmonizes real-world people into an extended world., Accepted to CVPR 2023 ECV Workshop

In the absence of an authoritative statement about a rumor, people may expose the truth behind such rumor through their responses on social media. Most rumor detection methods aggregate the information of all the responses and have made great progress. However, due to the different backgrounds of users, the responses have different relevance for discovering th suspicious points hidden in a rumor claim. The methods that focus on all the responding tweets would dilute the effect of the critical ones. Moreover, for a multi-modal rumor claim, the focus of a user may be on several words in the text or an object in the image, so the different modalities should be considered to select the relevant responses and verify the claim. In this paper, we propose a novel multi-modal rumor detection model, termed Focal Reasoning Model (FoRM), to filter out the irrelevant responses and further conduct fine-grained reasoning with the multi-modal claim and corresponding responses. Concretely, there are two main components in our FoRM: the coarse-grained selection and the fine-grained reasoning. The coarse-grained selection component leverages the post-level features of the responses to verify the claim and learns a relevant score of each response. Based on the relevant scores, the most relevant responses are reserved as the critical ones to the further reasoning. In the fine-grained reasoning component, we design a relation attention module to explore the fine-grained relations, i.e., token-to-token and token-to-object relations, between the reserved responses and the multi-modal claim for finding out the valuable clues. Hepatocellular carcinoma (HCC) exhibits high invasiveness and mortality rates, and the molecular mechanisms of HCC have gained increasing research interest. The abnormal DNA damage response has long been recognized as one of the important factors for tumor occurrence and development. Recent studies have shown the potential of the protein RING finger and WD repeat domain 3 (RFWD3) that positively regulates p53 stability in response to DNA damage as a therapeutic target in cancers.To investigate the relationship between HCC and RFWD3Compared with adjacent tissues, RFWD3 expression levels were significantly higher in clinical HCC tissues and correlated with tumor size and TNM stage (We provide evidence for the expression and function of RFWD3 in HCC. RFWD3 affects the prognosis, proliferation, invasion, and metastasis of HCC by regulating the Wnt/β-catenin signalling pathway.

In this work, biphenyl was used as carbon source to enrich microorganisms from polychlorinated biphenyls (PCBs)-contaminated paddy soil samples, and the taxonomic structures in both of the soil samples and the fourth-generation enrichments were examined with high-throughput sequencing. Single cells were isolated from the enrichments via single cell sorting technology named Laser Induced Visualized Ejection Separation Technology (LIVEST) and also traditional single colony sorting, and the genera of the isolates were identified using 16S rRNA sequencing. The results from high-throughput sequencing present that enrichment from generation to generation can considerably change the microbial community. Comparing the two sorting methods, the LIVEST is more time-saving and cell-targeted for microbial resource exploration. Based on the further verification of biphenyl degradation, it was found that some strains belonging to genera Macrococcus, Aerococcus and Metabacillus are capable in degrading biphenyl, which have not been reported yet.

To reveal the correlation of dynamic serum tumor markers (STMs) and molecular features of epidermal growth factor receptor-mutated (EGFR-mutated) lung cancer during targeted therapy, we retrospectively reviewed 303 lung cancer patients who underwent dynamic STM tests [neuron-specific enolase (NSE), carcinoembryonic antigen (CEA), carbohydrate antigen 125 (CA125), carbohydrate antigen 153 (CA153), the soluble fragment of cytokeratin 19 (CYFRA21-1), and squamous cell carcinoma antigen (SCC)] and circulating tumor DNA (ctDNA) testing with a panel covering 168 genes. At baseline, patients with EGFR mutation trended to have abnormal CEA, abnormal CA153, and normal SCC levels. Additionally, patients with Thr790Met (T790M) mutation were more likely to have abnormal CEA levels than patients without T790M mutation. Among patients with secondary resistance to EGFR tyrosine kinase inhibitors (TKI), the dynamic STMs showed a descending trend in the responsive stage and a rising trend in the resistant stage. However, the changing slopes differed between T790M subgroup and the non-T790M subgroup in individual STMs. Our study demonstrated that the combination of baseline levels and variations of STMs (including the responsive stage and resistant stage) can be suggestive of secondary EGFR-T790M mutation [area under the curve (AUC) = 0.897] and that changing trends of STMs (within 8 weeks after initiating the TKI therapy) can be potential predictors for the clearance of EGFR ctDNA [AUC = 0.871]. In conclusion, dynamic monitoring STMs can help to predict the molecular features of EGFR-mutated lung cancer during targeted therapy.

Simulating the stochastic traffic load has been an important issue for bridge condition evaluation. This paper proposes an improved cellular automaton for stochastic traffic load simulation with consideration of axle load and microscopic interaction. The cells, neighbors, transition rules and boundary conditions in the improved model are all redefined. The cell length can be manually defined with any value. The vehicle load can be precisely simulated using axle load if the cell length is set small or using concentrated load when a large cell length is defined. Acceleration based transition rules are proposed, and different acceleration for each vehicle can be computed to more reasonably simulate the microscopic behavior. The feasibility of the proposed model is validated through comparisons on traffic flow simulation and load effect quantification with in-field weigh in motion data and other models. The results indicate that the proposed model improves the existing models both in vehicle load simulation and traffic flow simulation and can be used for stochastic traffic load simulation for bridges with any length.

This study aimed to establish a non-invasive radiomics model based on computed tomography (CT), with favorable sensitivity and specificity to predict EGFR mutation status in GGO-featured lung adenocarcinoma subsequently guiding the administration of targeted therapy.Clinical-pathological information and preoperative CT images of 636 lung adenocarcinoma patients (464, 100, and 72 in the training, internal, and external validation sets, respectively) that underwent GGO lesions resection were included. A total of 1476 radiomics features were extracted with gradient boosting decision tree (GBDT).The established radiomics model containing 102 selected features showed an encouraging discrimination performance of EGFR mutation status (mutant or wild type), and the predictive ability was superior to that of the clinical model (AUC: 0.838 vs. 0.674, 0.822 vs. 0.730, and 0.803 vs. 0.746 for the training, internal validation, and external validation sets, respectively). The combined radiomics plus clinical model showed no additional benefit over the radiomics model in predicting EGFR status (AUC: 0.846 vs. 0.838, 0.816 vs. 0.822, and 0.811 vs. 0.803, respectively, in three cohorts). Uniquely, this model was validated in a cohort of lung adenocarcinoma patients who have undertaken adjuvant EGFR-TKI treatment and harbored unresected GGOs during the medication, leading to a significantly improved potency of EGFR-TKIs (response rate: 25.9% vs. 53.8%, p = 0.006; before and after prediction, respectively).This presented radiomics model can be served as a non-invasive and time-saving approach for predicting the EGFR mutation status in lung adenocarcinoma presenting as GGO.• We developed a GGO-specific radiomics model containing 102 radiomics features for EGFR mutation status differentiation. • An AUC of 0.822 and 0.803 in the internal and external validation cohorts, respectively, were achieved. • The radiomics model was utilized in clinical translation in an adjuvant EGFR-TKI treatment cohort with unresected GGOs. A significant improvement in the potency of EGFR-TKIs was achieved (response rate: 25.9% vs. 53.8%, p = 0.006; before and after prediction).

To solve the problem that the K-means algorithm is sensitive to the initial clustering centers and easily falls into local optima, we propose a new hybrid clustering algorithm called the IGWOKHM algorithm. In this paper, we first propose an improved strategy based on a nonlinear convergence factor, an inertial step size, and a dynamic weight to improve the search ability of the traditional grey wolf optimization (GWO) algorithm. Then, the improved GWO (IGWO) algorithm and the K-harmonic means (KHM) algorithm are fused to solve the clustering problem. This fusion clustering algorithm is called IGWOKHM, and it combines the global search ability of IGWO with the local fast optimization ability of KHM to both solve the problem of the K-means algorithm’s sensitivity to the initial clustering centers and address the shortcomings of KHM. The experimental results on 8 test functions and 4 University of California Irvine (UCI) datasets show that the IGWO algorithm greatly improves the efficiency of the model while ensuring the stability of the algorithm. The fusion clustering algorithm can effectively overcome the inadequacies of the K-means algorithm and has a good global optimization ability.

IntroductionEmerging data suggest that perioperative gut dysbiosis is prevalent and may be associated with postoperative neurocognitive disorders (PND). Antibiotics and probiotics are key factors influencing the microbiota. Many antibiotics have anti-microorganisms and direct anti-inflammatory properties, which may have cognitive repercussions. NLRP3 inflammasome activation has been reported to be involved with cognitive deficits. This study aimed to determine the effect and mechanism of probiotics on neurocognitive problems associated with perioperative gut dysbiosis by the NLRP3 pathway.MethodsIn a randomized, controlled trial, adult male Kunming mice undergoing surgery were administered cefazolin, FOS + probiotics, CY-09, or a placebo in four distinct experimental cohorts. Fear conditioning (FC) tests evaluate learning and memory. Following FC tests to evaluate inflammatory response (IR) and the permeability of barrier systems, the hippocampus and colon were extracted, and feces were collected for 16 s rRNA.ResultsOne week after surgery, surgery/anesthesia decreased the frozen behavior. Cefazolin attenuated this declination but aggravated postoperative freezing behavior 3 weeks after surgery. Probiotics ameliorated surgery/anesthesia-induced memory deficits and perioperative cefazolin-induced postoperative memory deficits 3 weeks after surgery. NLRP3, caspase-1, Interleukin-1β (IL-1β), and Interleukin-18 (IL-18) levels were increased 1 week after the hippocampus and colon surgery, which were attenuated by CY-09 and probiotics, respectively.DiscussionProbiotics could correct dysbacteria and IR caused by surgery/anesthesia stress and cefazolin alone. These findings imply that probiotics are an efficient and effective way of maintaining the balance of gut microbiota, which may reduce NLRP3-related inflammation and alleviate PND.

Parthanatos is a type of programmed cell death initiated by over-activated poly (ADP-ribose) polymerase 1 (PARP1). Nuclear translocation of apoptosis inducing factor (AIF) is a prominent feature of parthanatos. But it remains unclear how activated nuclear PARP1 induces mitochondrial AIF translocation into nuclei. Evidence has shown that deoxypodophyllotoxin (DPT) induces parthanatos in glioma cells via induction of excessive ROS. In this study we explored the downstream signal of activated PARP1 to induce nuclear translocation of AIF in DPT-triggered glioma cell parthanatos. We showed that treatment with DPT (450 nM) induced PARP1 over-activation and Tax1 binding protein 1 (TAX1BP1) distribution to mitochondria in human U87, U251 and U118 glioma cells. PARP1 activation promoted TAX1BP1 distribution to mitochondria by depleting nicotinamide adenine dinucleotide (NAD+). Knockdown of TAX1BP1 with siRNA not only inhibited TAX1BP1 accumulation in mitochondria, but also alleviated nuclear translocation of AIF and glioma cell death. We demonstrated that TAX1BP1 enhanced the activity of respiratory chain complex I not only by upregulating the expression of ND1, ND2, NDUFS2 and NDUFS4, but also promoting their assemblies into complex I. The activated respiratory complex I generated more superoxide to cause mitochondrial depolarization and nuclear translocation of AIF, while the increased mitochondrial superoxide reversely reinforced PARP1 activation by inducing ROS-dependent DNA double strand breaks. In mice bearing human U87 tumor xenograft, administration of DPT (10 mg· kg−1 ·d−1, i.p., for 8 days) markedly inhibited the tumor growth accompanied by NAD+ depletion, TAX1BP1 distribution to mitochondria, AIF distribution to nuclei as well as DNA DSBs and PARP1 activation in tumor tissues. Taken together, these data suggest that TAX1BP1 acts as a downstream signal of activated PARP1 to trigger nuclear translocation of AIF by activation of mitochondrial respiratory chain complex I.

Background Sugammadex has been reported to lower the incidence of postoperative residual neuromuscular blockade. Despite the advantages, until recently the effects of sugammadex on postoperative pulmonary complications (PPCs) were controversial. We conducted a systematic review and meta-analysis to determine whether reversal with sugammadex was associated with a lower risk of PPCs compared with neostigmine. Methods PubMed, Embase, and Cochrane Central Register of Controlled Trials were searched from inception to May 2022. Randomized controlled trials (RCTs) and observational studies comparing PPCs in patients receiving sugammadex or neostigmine as reversal agent at the end of surgery were included. The primary outcomes focused on PPCs including desaturation, pneumonia, atelectasis, noninvasive ventilation (NIV) and reintubation. Trial sequential analysis was performed on the primary outcomes to confirm whether firm evidence was reached. Results Meta-analysis of included studies showed that the rate of desaturation (43.2% vs 45.0%, RR = 0.82; 95% CI 0.63 to 1.05; p = 0.11) were comparable between the two groups. When looking at other primary outcomes, significantly lower risk of pneumonia (1.37% vs 2.45%, RR = 0.65; 95% CI 0.49 to 0.85; p = 0.002), atelectasis (24.6% vs 30.4%, RR = 0.64; 95% CI 0.42 to 0.98; p = 0.04), NIV (1.37% vs 2.33%, RR = 0.65; 95% CI 0.43 to 0.98; p = 0.04) and reintubation (0.99% vs 1.65%, RR = 0.62; 95% CI 0.43 to 0.91; p = 0.01) in the sugammadex group were detected compared with the neostigmine group. Conclusions We concluded that sugammadex is more effective at reducing the incidence of PPCs including pneumonia, atelectasis, NIV and reintubation compared with neostigmine. Further evidence, preferably from RCTs, is required to confirm these findings.

Lipid metabolism disorder has become an important hidden danger threatening human health, and various supplements to treat lipid metabolism disorder have been studied. Our previous studies have shown that DHA-enriched phospholipids from large yellow croaker (Larimichthys Crocea) roe (LYCRPLs) have lipid-regulating effects. To better explain the effect of LYCRPLs on lipid regulation in rats, the fecal metabolites of rats were analyzed from the level of metabolomics in this study, and GC/MS metabolomics measurements were performed to figure out the effect of LYCRPLs on fecal metabolites in rats. Compared with the control (K) group, 101 metabolites were identified in the model (M) group. There were 54, 47, and 57 metabolites in the low-dose (GA), medium-dose (GB), and high-dose (GC) groups that were significantly different from that of group M, respectively. Eighteen potential biomarkers closely related to lipid metabolism were screened after intervention with different doses of LYCRPLs on rats, which were classified into several metabolic pathways in rats, including pyrimidine metabolism, the citric acid cycle (TCA cycle), the metabolism of L-cysteine, carnitine synthesis, pantothenate and CoA biosynthesis, glycolysis, and bile secretion. L-cysteine was speculated to be a useful biomarker of LYCRPLs acting on rat fecal metabolites. Our findings indicated that LYCRPLs may regulate lipid metabolism disorders in SD rats by activating these metabolic pathways.
