1. Modulation of Visual Contrast Sensitivity with tRNS across the Visual System, Evidence from Stimulation and Simulation

Abstract

Transcranial random noise stimulation (tRNS) has been shown to significantly improve visual perception. Previous studies demonstrated that tRNS delivered over cortical areas acutely enhances visual contrast detection of weak stimuli. However, it is currently unknown whether tRNS-induced signal enhancement could be achieved within different neural substrates along the retino-cortical pathway. In three experimental sessions, we tested whether tRNS applied to the primary visual cortex (V1) and/or to the retina improves visual contrast detection. We first measured visual contrast detection threshold (VCT; N = 24, 16 females) during tRNS delivery separately over V1 and over the retina, determined the optimal tRNS intensities for each individual (ind-tRNS), and retested the effects of ind-tRNS within the sessions. We further investigated whether we could reproduce the ind-tRNS-induced modulation on a different session (N = 19, 14 females). Finally, we tested whether the si-multaneous application of ind-tRNS to the retina and V1 causes additive effects. Moreover, we present detailed simulations of the induced electric field across the visual system. We found that at the group level tRNS decreases VCT compared with baseline when delivered to the V1. Beneficial effects of ind-tRNS could be re-plicated when retested within the same experimental session but not when retested in a separate session. Applying tRNS to the retina did not cause a systematic reduction of VCT, regardless of whether the individu-ally optimized intensity was considered or not. We also did not observe consistent additive effects of V1 and retina stimulation. 12. Functionalized Anatomical Models for Computational Life Sciences

Abstract

The advent of detailed computational anatomical models has opened new avenues for computational life sciences (CLS). To date, static models representing the anatomical environment have been used in many applications but are insufficient when the dynamics of the body prevents separation of anatomical geometrical variability from physics and physiology. Obvious examples include the assessment of thermal risks in magnetic resonance imaging and planning for radiofrequency and acoustic cancer treatment, where posture and physiology-related changes in shape (e.g., breathing) or tissue behavior (e.g., thermoregulation) affect the impact. Advanced functionalized anatomical models can overcome these limitations and dramatically broaden the applicability of CLS in basic research, the development of novel devices/therapies, and the assessment of their safety and efficacy. Various forms of functionalization are discussed in this paper: (i) shape parametrization (e.g., heartbeat, population variability), (ii) physical property distributions (e.g., image-based inhomogeneity), (iii) physiological dynamics (e.g., tissue and organ behavior), and (iv) integration of simulation/measurement data (e.g., exposure conditions, “validation evidence” supporting model tuning and validation). Although current model functionalization may only represent a small part of the physiology, it already facilitates the next level of realism by (i) driving consistency among anatomy and different functionalization layers and highlighting dependencies, (ii) enabling third-party use of validated functionalization layers as established simulation tools, and (iii) therefore facilitating their application as building blocks in network or multi-scale computational models. Integration in functionalized anatomical models thus leverages and potentiates the value of sub-models and simulation/measurement data toward ever-increasing simulation realism. In our o2S2PARC platform, we propose to expand the concept of functionalized anatomical models to establish an integration and sharing service for heterogeneous computational models, ranging from the molecular to the organ level. The objective of o2S2PARC is to integrate all models developed within the National Institutes of Health SPARC initiative in a unified anatomical and computational environment, to study the role of the peripheral nervous system in controlling organ physiology. The functionalization concept, as outlined for the o2S2PARC platform, could form the basis for many other application areas of CLS. Abstract

Neuromodulation is a new therapeutic pathway to treat inflammatory conditions by modulating the electrical signalling pattern of the autonomic connections to the spleen. However, targeting this sub-division of the nervous system presents specific challenges in translating nerve stimulation parameters. Firstly, autonomic nerves are typically embedded non-uniformly among visceral and connective tissues with complex interfacing requirements. Secondly, these nerves contain axons with populations of varying phenotypes leading to complexities for axon engagement and activation. Thirdly, clinical translational of methodologies attained using preclinical animal models are limited due to heterogeneity of the intra- and inter-species comparative anatomy and physiology. Here we demonstrate how this can be accomplished by the use of in silico modelling of target anatomy, and validation of these estimations through ex vivo human tissue electrophysiology studies. 15. A modeling framework to understand and optimize ultrasound neuromodulation

Abstract

Over the last decade, Low Intensity Focused Ultrasound Stimulation (LIFUS) has emerged as an attractive technology to modulate the activity of deep neural targets without invasive procedures. However, the underlying mechanisms by which ultrasonic waves can excite neurons are still unclear, which prevents a reliable and targeted application of the LIFUS technology and hinders its translation into clinical settings. The Neuronal Intramembrane Cavitation Excitation (NICE) model hypothesizes that ultrasound excites neurons through the nucleation and resonance of nanoscale membrane structures (bilayer sonophores) that can alter membrane capacitance and induce depolarizing currents. The model predictions of LIFUS neuromodulatory effects match a wide range of empirical observations in the brain. However, because it neglects cellular morphology, the NICE model cannot address important questions on the interaction of ultrasonic waves with neural structures. In this thesis, I propose several strategies to address these limitations, and to bridge the gap towards an experimental validation of this mechanism. I begin with an introduction establishing the background, describing the state-of-the-art and issues plaguing the field of ultrasound neuromodulation, and motivating the global objectives that will be pursued throughout the thesis. In a second part, I provide a mathematical description of the two pillars of the NICE model, namely a cavitation model describing mechanical membrane oscillations, and a point-neuron model describing the membrane electrical response. In doing so, I also discuss the inherent model assumptions, the potential implications for its validity range, and its sensitivity to key parameters. In a third part, I present a novel multi-Scale Optimized Neuronal Intramembrane Cavitation (SONIC) model that alleviates the stiffness of the NICE model by numerically separating its two constituent time scales. I demonstrate how this approach drastically reduces computational costs and confers interpretability to LIFUS neuromodulatory effects in terms of effective membrane dynamics. In a fourth part, I present a morphological expansion of the SONIC model (termed morphoSONIC) allowing to simulate intramembrane cavitation in a wide variety of realistic neuron models.With this framework, I investigate LIFUS neuromodulatory effects in peripheral nerve fibers. I predict that myelinated and unmyelinated axons can be distinctively and selectively recruited by LIFUS, thereby opening exciting avenues for peripheral neuromodulation. In a fifth part, I present the results of a parallel collective effort to track down the mechanisms of ultrasound neuromodulation in sensory neurons extracted from the medicinal leech.We found that LIFUS can reliably induce spiking activity within an optimal intensity range, and identified common and differing response features between acoustically and electrically evoked spikes.This chapter ends with a discussion of the implications of our findings for the validity of the intramembrane cavitation hypothesis. The achievements presented in this thesis provide an increased understanding of the mechanisms by which ultrasound modulates neural activity, as well as computational tools for their investigation. Moreover, they pave the way towards the development of reliable modeling frameworks to simulate ultrasound neuromodulatory effects across spatial, temporal and functional scales, helping to propel LIFUS into the clinics.
