1. Detection of grain yield QTLs in the durum population Lahn/Chaml tested in contrasting environments

Abstract

Durum wheat (Triticum turgidum L. var durum) is tetraploid wheat (AABB); it is the main source of semolina and other pasta products. Grain yield in wheat is quantitatively inherited and influenced by the environment. The genetic map construction constitutes the essential step in identifying quantitative trait loci (QTLs) linked to complex traits, such as grain yield. The study aimed to construct a genetic linkage map of two parents that are widely grown durum cultivars (Lahn and Chaml) in the Mediterranean basin, which is characterized by varying climate changes. The genetic linkage map of Lahn/Chaml population consisted of 112 recombinant inbred lines (RILs) and was used to determine QTLs linked to the grain yield in 11 contrasting environments (favorable, cold, dry, and hot). Simple sequence repeat (SSR) molecular markers were used to construct an anchor map, which was later enriched with single nucleotide polymorphisms (SNPs). The map was constructed with 247 SSRs and enriched with 1425 SNPs. The map covered 6122.22 cM. One hundred and twenty-six QTLs were detected on different chromosomes. Chromosomes 2A and 4B harbored the most significant grain yield QTLs. Furthermore, by comparison with several wheat mapping populations, all the A and B chromosomes of Lahn/Cham 1 QTLs contributed to grain yield. The results showed that the detected QTLs can be used as a potential candidate for marker-assisted selection in durum breeding programs.

Abstract

In major crops, panels of germplasm accessions have been assembled for allele mining at targeted candidate genes and for whole-genome association mapping studies. A germplasm collection of 189 cultivated durum wheat accessions representing the major breeding groups was tested in a broad range of Mediterranean environments under rainfed and irrigated conditions with an average yield ranging from 0.9 to 6.8 t/ha. Yield, yield components, agronomic and physiological traits were recorded in 15 trials and tested for significant association with the allelic profile of 180 microsatellite markers (simple sequence repeats, SSRs). Population structure was evaluated and the clustering data were used as covariates in the association test. SSRs showing significant associations in several environments were identified for the traits with the highest heritability values, i.e. plant height, heading date, peduncle length and kernel weight, with R2 values ranging from 5 to 10%. As to yield and yield components, most associations were detected only in two to four environments, with an average R2 value lower than 5%.

Abstract

Coping with reduced inputs, especially in terms of yield stability under the various stress conditions that frequently occur in the drought-prone areas of the Mediterranean basin, is a main objective for durum wheat improvement. A total of 249 RILs (Kofa × Svevo) were evaluated in 16 trials conducted in 2004-2005 under a broad range of water input and yield potential. Two major, epistatic QTLs on chrs. 2BL and 3BS influenced grain yield and related physiological traits in a wide range of environments. In both cases, coincidence between the QTLs for grain yield and those for plant height, peduncle length, SPAD, NDVI index and kernel weight was observed. Epistasis favoured the parental genotypes. On a mean basis, the R2 values for grain yield of the 2BL and 3BS QTLs were equal to 21.5 and 13.8%, respectively. The effects of these two QTLs were validated in a set of 11 trials conducted in 2006 using 40 selected RILs.

Abstract

Vernalization and photoperiod response genes play a significant role in the geographical adaptation, agronomic performance and yield potential of crops. Therefore, understanding the distribution pattern and allelic diversity for vernalization and photoperiod genes are important in any wheat breeding program. In this study, we screened 63 bread wheat cultivars and 7 bread wheat landraces from Turkey for photoperiod (Ppd-D1) and vernalization genes (Vrn-A1, Vrn-B1, Vrn-D1 and Vrn-B3) using diagnostic molecular markers. The photoperiod insensitive dominant allele, Ppd-D1a, was present in 60% of wheat cultivars and 42% of landraces, whereas, all other genotypes carried the photoperiod sensitive allele Ppd-D1b as recessive allele. Twenty-four cultivars and two landraces contained recessive alleles for all four VRN loci, whereas 39 wheat cultivars and 6 landraces contained one or more dominant VRN alleles. The highest percentage of Turkish wheat cultivars contained the dominant Vrn-B1 allele followed by Vrn-D1 and Vrn-A1. Information for vernalization and photoperiod alleles in Turkish germplasm will facilitate the planning and implementation of molecular markers in wheat breeding programs. This information will be helpful to develop elite wheat cultivars carrying suitable vernalization and photoperiod alleles with higher grain yield potential and better quality suitable for different production environments through marker assisted selection.

A collection of 191 durum wheat accessions representing Mediterranean Basin genetic diversity was grown in nine different environments in four countries, with productivities ranging from 0.99 to 6.78 t ha(-1). Abstract

Grain yield is a major goal for the improvement of durum wheat, particularly in drought-prone areas. In this study, the genetic basis of grain yield (GY), heading date (HD), and plant height (PH) was investigated in a durum wheat population of 249 recombinant inbred lines evaluated in 16 environments (10 rainfed and 6 irrigated) characterized by a broad range of water availability and GY (from 5.6 to 58.8 q ha−1). Among the 16 quantitative trait loci (QTL) that affected GY, two major QTL on chromosomes 2BL and 3BS showed significant effects in 8 and 7 environments, with R2 values of 21.5 and 13.8% (mean data of all 16 environments), respectively. In both cases, extensive overlap was observed between the LOD profiles of GY and PH, but not with those for HD. QTL specific for PH were identified on chromosomes 1BS, 3AL, and 7AS. Additionally, three major QTL for HD on chromosomes 2AS, 2BL, and 7BS showed limited or no effects on GY. For both PH and GY, notable epistasis between the chromosome 2BL and 3BS QTL was detected across several environments.

Abstract

Until now, only a few genetic and physical maps had been constructed for durum wheat, mostly from the gene pools of only few geographic regions. The Fertile Crescent, particularly southeastern Turkey, is thought to be the primary center of wheat domestication and diversity. In spite of the importance of the genetic diversity from this area, a severe lack of information on the genetic structure of the durum wheat gene pool from this region is evident. For example, thus far, no efforts have been made to understand the genetic structure of the Anatolian durum wheat gene pool, despite its importance in durum wheat breeding. This study aimed to provide a new insight into the genetic structure of the durum wheat from Anatolia. The present study represents the first genetic linkage map of durum wheat from Anatolia, containing one famous Turkish local cultivar Kunduru-1149. The genetic linkage map was constructed using a population of 141 recombinant inbred lines (RILs) derived from a cross between the Turkish cultivar Kunduru-1149 and ICARDA cultivar Cham1. Linkage analysis resulted in a final linkage map spanning 4853.8 cM, which comprised 395 polymorphic marker loci (213 simple sequence repeats [SSRs], 146 amplified fragment length polymorphisms [AFLPs], and 36 seed storage proteins [SSPs]). The detected loci were distributed over 15 linkage groups on 14 whole chromosomes of durum wheat. Most of the markers (70.7 %) used in this map segregated according to Mendelian laws. This linkage map represents a key starting point toward understanding the genome structure of Anatolian durum wheat to genetically evaluate important agromorphological and qualitative characteristics. © 2014, Springer Science+Business Media New York.

Abstract

Association mapping was used to dissect the genetic basis of drought-adaptive traits and grain yield (GY) in a collection of 189 elite durum wheat accessions evaluated in 15 environments highly different for water availability during the crop cycle (from 146 to 711 mm) and GY (from 9.9 to 67.3 q ha(-1)). For highly heritable traits (e.g. heading date, kernel weight, etc.) several significant experiment-wise marker-trait associations were detected across five or more (up to 13 for kernel weight) environments, with R(2) values ranging from ca. 5 to 10%. As to GY, significant associations (R(2) from 2.5 to 4.2%) were mostly detected in one environment only (56 markers), while decreasing rapidly from two to five environments (from 20 to three markers, respectively) and with only one marker (Xbarc197 on chr. 5A) found significant in six environments (ranging from low- to high-yielding). These results are probably due to the complex genetic basis of GY and its interaction with environmental conditions. The number of markers significantly affecting GY decreased considerably under drought conditions, suggesting a limited effectiveness of association mapping to identify loci for GY under low-moisture conditions, most likely because different genotypes can attain similar phenotypes via different morpho-physiological traits and corresponding gene networks. Our study confirmed the role of major loci for phenology previously described in biparental mapping populations, highlighted a novel set of loci for drought-adaptive traits, and provided information on the agronomic value of the alleles at such loci across a broad range of soil moisture conditions.
