Background: Reporting standards, such as PRISMA aim to ensure that the methods and results of systematic reviews are described in sufficient detail to allow full transparency. Flow diagrams in evidence syntheses allow the reader to rapidly understand the core procedures used in a review and examine the attrition of irrelevant records throughout the review process. Recent research suggests that use of flow diagrams in systematic reviews is poor and of low quality and called for standardised templates to facilitate better reporting in flow diagrams. The increasing options for interactivity provided by the Internet gives us an opportunity to support easy-to-use evidence synthesis tools, and here we report on the development of a tool for the production of PRISMA 2020-compliant systematic review flow diagrams.\ud \ud Methods and Findings: We developed a free-to-use, Open Source R package and web-based Shiny app to allow users to design PRISMA flow diagrams for their own systematic reviews. Our tool allows users to produce standardised visualisations that transparently document the methods and results of a systematic review process in a variety of formats. In addition, we provide the opportunity to produce interactive, web-based flow diagrams (exported as HTML files), that allow readers to click on boxes of the diagram and navigate to further details on methods, results or data files. We provide an interactive example here; https://prisma-flowdiagram.github.io/.\ud \ud Conclusions: We have developed a user-friendly tool for producing PRISMA 2020-compliant flow diagrams for users with coding experience and, importantly, for users without prior experience in coding by making use of Shiny (https://estech.shinyapps.io/prisma_flowdiagram/). This free-to-use tool will make it easier to produce clear and PRISMA 2020-compliant systematic review flow diagrams. Significantly, users can also produce interactive flow diagrams for the first time, allowing readers of their reviews to smoothly and swiftly explore and navigate to further details of the methods and results of a review. We believe this tool will increase use of PRISMA flow diagrams, improve the compliance and quality of flow diagrams, and facilitate strong science communication of the methods and results of systematic reviews by making use of interactivity. We encourage the systematic review community to make use of the tool, and provide feedback to streamline and improve their usability and efficiency.

Studies of individual quantum systems, which have led to considerable progress in our understanding of quantum physics, have traditionally been associated with atomic gases. In the last decades however, the emphasis has shifted towards solid-state systems, which are much more practical for applications. In particular, a new field has recently emerged that is concerned with the study of quantum systems based on single spins localized near point defects in crystalline solids. One such system is the nitrogen-vacancy (NV) defect in diamond. Initially used as an experimental breadboard for testing concepts of quantum physics and quantum computation, the NV defect was soon proposed as a sensitive magnetometer, capable of detecting minute magnetic fields, down to ultimate level of single spins. This atomic-sized magnetometer can be used as a standalone sensor, or integrated into an imaging system providing spatial resolution down to the atomic scale. Diamond-based instruments thus offer new pathways to probe the magnetism of matter from the mesoscale down to the nanoscale. This book chapter gives an overview of the field of diamond-based magnetic sensing and imaging, with an emphasis on already demonstrated applications of this technology. The chapter is divided into three main sections. In Section 2, the underlying physics and methods of diamond-based magnetometry are described. Section 3 is devoted to various experimental implementations that employ this new class of sensors for magnetic sensing and imaging. Lysosomes have traditionally been viewed as degradative organelles, though a growing body of evidence suggests that they can function as Ca2+ stores. Here, we examined the function of these stores in hippocampal pyramidal neurons. We found that back-propagating action potentials (bpAPs) could elicit Ca2+ release from lysosomes in the dendrites. This Ca2+ release triggered the fusion of lysosomes with the plasma membrane, resulting in the release of Cathepsin B. Cathepsin B increased the activity of matrix metalloproteinase 9 (MMP-9), an enzyme involved in extracellular matrix (ECM) remodelling and synaptic plasticity. Inhibition of either lysosomal Ca2+ signalling or Cathepsin B release prevented the maintenance of dendritic spine growth induced by Hebbian activity. This impairment could be rescued by exogenous application of active MMP-9. Our findings suggest that activity-dependent exocytosis of Cathepsin B from lysosomes regulates the long-term structural plasticity of dendritic spines by triggering MMP-9 activation and ECM remodelling.

The aim of the research was to describe and explain the barriers to, and opportunities for, outdoor play and recreation that exist in residential child care services. The research focussed on six residential facilities which represented a cross-section of the sector. Interviews were conducted with two young people, one manager and two basic grade staff members from each of the facilities. The findings were analysed using data display and reduction and the following key findings emerged.
