Uterine adenosarcoma is a rare gynecological malignancy with no specific symptoms, and the optimal management is still inconclusive. Herein we present a case of uterine adenosarcoma in a 38-year-old woman with a good prognosis and review of literatures. The patient presented with abnormal vaginal bleeding with no special medical history. Sonographic scan revealed a heterogeneous echoic mass in the cavity, indicating a polypus or a submucous myoma. The pathology based on the specimen after the hysteroscopic tumor excision suggested diagnosis of uterine adenosarcoma. Subsequently, the patient received pelvic MRI scan before surgery. MRI identified a patchy lesion at the cervix-lower endometrial cavity with low signal in T1WI and a mixed high T2 signal in T2WI, with no sign of metastasis. Then total abdominal hysterectomy with bilateral salpingo-oopherectomy plus pelvic lymph node dissection was performed and 6 cycles of chemotherapy were administered. The patient remains disease-free on follow-up to date, more than 15 months after chemotherapy.

Chemically engineered nanomaterials have been extensively used in early tumor detection and cancer therapy. Despite the promise shown, their chemical or exogenous nature hinders their application due to their unknown adverse effects. Herein, using a cancer cell environment, fluorescent DNA-gold nanostructures were bio-self-assembled through simple incubation of DNA and Au solutions with cancer cells. In situ, ex vivo, bio-responsive self-assembly of ring-shaped DNA-Au nanostructures is reported for the first time. Subsequently, the exosomes released by the above-mentioned cancer cells were found to carry the self-assembled DNA-Au nanostructures, exhibiting strong in vivo dual fluorescence properties. Interestingly, these exosomes could be immediately taken up in vitro by their parent cells, reaching the nucleus within 10 min after incubation. Taking advantage of the unique endogenous properties of exosomes, and their advanced cargo delivery capacity, we further exploited the DNA-Au nanostructure loaded exosomes with mitoxantrone for accurate cancer theranostics. The in vitro and in vivo results showed that the exosomes could effectively deliver the drug cargo to cancerous cells, hence, displaying an enhanced targeting effect towards parent cancer cells, and a synergistic tumor inhibition effect, while showing great biocompatibility towards normal cells and vital organs. Hence, exosomes carrying the in situ bio-self-assembled DNA-Au nanostructures could be an outstanding delivery system for dye-free targeted cancer detection and therapy.

In this paper, a new hybrid excited axial electrodynamic wheel (HEAEDW)for EDS maglev is proposed and investigated in which propulsion and levitation forces are simultaneously created via moving magnetic rotor above a conductive guideway. The proposed machine is a hybrid excitation machine, which can not only realize control actively and thrust force, but also reduces the risk of PM demagnetization. The field and forces created by this machine are analyzed by using the analytical and finite-element method. Then, the parameters that affect performance were investigated and adjusted to improve performance. Finally, a HEAEDW prototype is manufactured, and experiments are carried out to verify the predicted results. The results show that the proposed machine is capable of producing not only an equivalent suspension force of the basic axial EDWs but also the thrust and controlled mode.

Background: Assessing the effectiveness and safety of Chinese medicine for the mastitis in COVID-19 patients is the main purpose of this systematic review protocol. Methods: The following electronic databases will be searched from inception to April 2020: MEDLINE, Ovid, EMBASE, the Cochrane Library, the Allied and Complementary Medicine Database (AMED), Chinese National Knowledge Infrastructure (CNKI), Chinese Biomedical Literature Database (CBM), VIP Database and Wanfang Database. In addition, Clinical trial registries, like the Chinese Clinical Trial Registry (ChiCTR), the Netherlands National Trial Register (NTR) and ClinicalTrials.gov, will be searched for ongoing trials with unpublished data. No language restrictions will be applied. The primary outcome will be the time of disappearance of main symptoms (including fever, asthenia, cough disappearance rate, and temperature recovery time), and serum cytokine levels. The secondary outcome will be the accompanying symptoms (such as myalgia, expectoration, stuffiness, runny nose, pharyngalgia, anhelation, chest distress, dyspnea, crackles, headache, nausea, vomiting, anorexia, diarrhea) disappear rate, negative COVID-19 results rate on 2 consecutive occasions (not on the same day), CT image improvement, average hospitalization time, occurrence rate of common type to severe form, clinical cure rate, and mortality. Two independent reviewers will conduct the study selection, data extraction and assessment. RevMan V.5.3 will be used for the assessment of risk of bias and data synthesis. Results: The results will provide a high-quality synthesis of current evidence for researchers in this subject area. Conclusion: The conclusion of the study will provide an evidence to judge whether Chinese medicine is effective and safe for mastitis in COVID-19 patients. Ultra-precision polishing is an important processing method for monocrystalline silicon, in order to improve the machining efficiency and obtain good machining quality, it is necessary to investigate the material removal process and process parameters of ultra-precision polishing. Smoothed particle hydrodynamics (SPH) is a meshless method with good self-adaptability, it can be used in the simulation of ultra-precision polishing which has high speed deformation characteristics. The calculation model and SPH analysis model of ultraprecision polishing are established according to the principle of ultra-precision polishing, SPH method is used to simulate and analyze ultra-precision polishing of the monocrystalline silicon. The material removal process of ultra-precision polishing is investigated, the effects of abrasive size and indentation depth on the equivalent plastic strain (PEEQ), Mises stress and the force of abrasive is investigated. The result is that, at the different time of ultra-precision polishing, the maximum PEEQ is different, but the difference is not obvious; the X direction force of the abrasive increases with the indentation depth; the size of abrasive has a great influence on the soft coefficient of stress state in ultra-precision polishing. According to the simulation results, it is possible to optimize the technological parameters of ultra-precision polishing, and provide the theoretical guidance for practical production.

Objective(s): AMP-activated protein kinase/sirtuin 1 (AMPK/SIRT1) signaling pathway has been proved to be involved in the regulation of autophagy in various models. The aim of this study was to evaluate the effect of AMPK/SIRT1 pathway on autophagy after spinal cord injury (SCI). Materials and Methods: The SCI model was established in rats in vivo and the primary spinal cord neurons were subjected to mechanical injury (MI) in vitro. The apoptosis in spinal cord tissue and neurons was assessed by TUNEL staining and Hoechst 33342 staining, respectively. The autophagy-related proteins levels were detected by Western blot. The activation of AMPK/SIRT1 pathway was determined by Western blot and immunohistochemical staining. Results: We found that the apoptosis of spinal cord tissue and cell damage of spinal cord neurons was obvious after the trauma. The ratio of LC3II/LC3I and level of p62 were first increased significantly and then decreased after the trauma in vivo and in vitro, indicating the defect in autophagy. The levels of p-AMPK and SIRT1 were increased obviously after the trauma in vivo and in vitro. Further activation of the AMPK/SIRT1 pathway by pretreatment with resveratrol, a confirmed activator of the AMPK/SIRT1 pathway, alleviated the cell damage and promoted the autophagy flux via downregulation of p62 in spinal cord neurons at 24 hr after MI. Conclusion: Our results demonstrate that regulation of autophagy by AMPK/SIRT1 pathway can restrain spinal cord neurons damage, which may be a potential intervention of SCI.

Considering sophisticated optical path system, low efficiency and accuracy in the traditional screening of food safety, some improvement methods are proposed for the traditional screening equipment of food safety. The equipment is took rear spectroscopic measuring technique and linear array Charge-coupled Device (CCD) which can realize real-time detection within visible range. Automatic liquid surface sensing and tracking technology in the process of sample injection are accepted for improving the test accuracy. Verification experiment through the equipment to accurately determine multicomponent pigment measurement, the results indicate that the equipment showed potential for the rapid testing of adulterated food.

Traditional food safety detection has many short-coming such as the complex light path system, low detection efficiency and low detection precision. In this paper, a new rapid detection equipment of food safety based on rear spectroscopic technology and Charge-coupled Device (CCD) was presented. We the photoelectric colorimetric system was redesigned. The optical path system was simplified by the rear spectroscopic technology, thus the new equipment reduced the volume and weight of the instrument than traditional food safety detection. The flat field holographic concave grating and linear array CCD can collect the plane full spectrum signal at one time, so the new equipment can apply to multicomponent pigment measurement. Experimental results prove its stability which verifies the reasonableness of software and hardware designing.

A novel pitch curve design method for non-circular bevel gear is presented to simplify design of non-circular bevel gear. Pitch curve equation for non-circular bevel gear is deduced based on geometrical relationships between pitch cone surface of non-circular gear with parallel axis and spherical surface. Then, transmission ratio function is transformed into pitch curve design defects modification equation and mathematical expression between modification factor and pitch curve design parameters for non-circular bevel gear is derived. By using the expression, modification value of pitch curve can be determined rapidly and accurately. Simulation results for high-order elliptical bevel gear and Pascal curve bevel gear demonstrate that the proposed modification method can be applied to pitch curve design for other types of non-circular bevel gear. Design and verification efficiency for non-circular bevel gear are improved effectively by employing the proposed pitch curve design defects modification method.

There are a large number of macropores/tubular channels of a few centimeters and plant roots in reclaimed dump soils, which are the main reasons for the formation of soil macropores and soil preferential flow. To systematically study the morphological characteristics and variation of soil preferential flow for different reclaimed vegetations in a dump, a dye-staining experiment and physical and chemical analysis were carried out to investigate the formation mechanism and influencing factors of soil preferential flow in the vegetation restoration process. The results indicate that there were differences in the soil water breakthrough curves for different plots. The macropore effluent rate generally increased at first and then tended to stabilize. The soil steady effluent rate decreased with increasing soil depth, which reached the maximum and minimum values at the depths of 0∼5 cm (0.0193∼0.0315 mm s−1) and 50∼60 cm (0.0028∼0.0035 mm s−1), respectively. Furthermore, the radius of soil macropores under different types of reclaimed vegetation ranged from 0.03 to 4.71 mm, most of which ranged from 0.11 to 2.36 mm. The soil macroporosity of different reclaimed vegetation types ranged from 0.03 to 16.58%, which was significantly greater than 5%. The soil macroporosity determined 65% of the variation in the steady effluent rate and 42% of the variation in the saturated hydraulic conductivity. Furthermore, the dye coverage ratio decreased as the soil layer depth increased in different plots, and there were some differences in each plot. The maximum dye coverage ratio occurred in the 0∼5 cm soil layer, which reached 90.37%. The dye coverage ratio at a depth of 0∼60 cm in six plots followed the order of Robinia pseudoacacia (26.48%) > Ulmus pumila (20.12%) > mixed forest (17.32%) > farmland (15.06%) > shrub (13.97%) > weeds (10.07%). The soil preferential flow mostly occurred in the 0∼40 cm soil depth layer, which occupied more than 93% of the total soil profile (0∼60 cm). Moreover, a Pearson correlation was used to analyze the relationship between environmental factors (soil, water, and plant factors) and the dye coverage ratio. The dye coverage ratio of soil preferential flow under different reclamation vegetations was very significantly or significantly positively correlated with the gravel content, mean radius of soil macropores, soil saturated hydraulic conductivity, root weight density, and root length density, which promoted the formation and development of soil preferential flow. This study will provide a scientific basis for understanding the formation mechanism and perfecting the research system of soil preferential flow, vegetation restoration, and reconstruction in a dump; furthermore, this research offers significance guidance in the construction of green mines and the development of regional economics.

Materials with perpendicular magnetic anisotropy have been intensively investigated due to their potential applications in the nonvolatile magnetic memory and spin-torque oscillators. Hear in this paper, we report a special interesting spin-transfer-driven magnetic behavior in perpendicularly magnetized (Co/Ni) -based spin-valve nano-pillars due to the reduced symmetry of easy axis in the free layer. The micromagnetic simulations indicate that a dip in the average magnetization curve can take place due to the reduced symmetry such as tilt of the magnetic field as well as the easy axis of the free and polarizer layers. In order to further clarify the physics mechanism of the dip, we carry out a series of new simulation studies. In our simulations, we consider a spin-valve nano-pillar with perpendicular anisotropy free layer and a 3 tilted polarizer layer. A negative perpendicular magnetic field and a positive perpendicular current are both applied simultaneously. In the average magnetization curves mz as a function of the magnetic field with various currents, three dips are observed. Note that although the spin-transfer torque is essential to the appearance of the dips, the position of the dips is less affected by the current in a certain current range. For three dips, we notice that the mz values are almost identical at a special magnetic field for different currents. At this special magnetic field, the magnetization oscillation modes in the free layer are similar to each other for different currents. The corresponding frequency spectra show that the amplitude of the main frequency peak decreases with the increasing of current due to the enhanced spin-transfer torque. In addition, the frequency shows a blue-shift with the increasing of applied current. Our simulations show that the main frequency f1 corresponding to the highest peak is approximately equal to the precession frequency of the local magnetization in the free layer. Several high-order frequency peaks are also observed in the frequency spectrum with fn=nf1, where n is an integer. Therefore the periodic oscillation of mz is a harmonic oscillation. Further simulations indicate that the dip appearance is also affected by the thickness of free layer. The spin-transfer torque effect decreases with the thickness of the free layer increasing. As a consequence, the dips shift to a low magnetic field range with the increase of the thickness. And for larger thickness t=8.0 nm, no dip appears. This result suggests that the spin-transfer torque is necessary for the dip, rather than the unique effect factor, to occur. In the dip region, the magnetic oscillation modes of the free layer show interesting frequency spectrum characters:harmonic frequency or inter-harmonic frequency. As a consequence, the periodic oscillation of the free layer is accompanied by the harmonic waves.

The study investigated whether bradykinin (BK) preconditioning could regulate the expression of aquaporin-4 (AQP4) using an in vivo transient spinal cord ischemia model in rats. BK was infused continuously via the left femoral artery with infusion pump for 15 min (10 microg/kg/min) then we induced ischemia for 20 min and reperfusion for 24 and 72 h respectively. The results demonstrated that the central part of the white matter exhibited loss of perivascular AQP4 and showed a partial recovery toward 72 h of reperfusion. The border zone of white matter was different from the central part of the white matter by showing no loss of perivascular AQP4 at 24 h of reperfusion but rather a slight increase. BK significantly reduced the expression level of AQP4 protein in the white matter, but it had none of this effect in the gray matter region at 72 h post-reperfusion. There was no difference in AQP4 protein levels between BK group and control group at the two above-mentioned spinal cord regions at 24 h after reperfusion. In addition, the changes in AQP4 protein induced by BK preconditioning were obvious at 72 h after reperfusion, which were accompanied by a reduction of spinal cord edema. Our results demonstrated that the expression of AQP4 protein after spinal cord ischemia/reperfusion was region-specific, time-dependent and also indicated that the attenuation of AQP4 expression induced by BK could be one of the important molecular mechanisms in physiopathology of spinal cord ischemic edema.

Spinal cord injury (SCI) is a devastating problem of health. Glucagon-like peptide-1 (GLP-1) is glucose-dependent insulinotropic hormone. Exendin-4 is the longacting GLP-1 receptor agonist that used as an anti-diabetic drug. Recent studies demonstrated exendin-4 has a series of beneficial neuroprotective properties. This study was designed to assess the neuroprotective effects of exendin-4 against spinal cord injury. 72 rats were randomly divided into three groups as follows: sham operation group, SCI group, and exendin-4 group (intraperitoneal injection of exendin-4 at 10 μg/rat immediately after SCI). First, a decrease of malodialdehyde (MDA) levels and an increase of glutathione (GSH) levels in the exendin-4 group demonstrated the anti-oxidation effect of exendin-4. Further, the preservation of mitochondrial membrane potential (ΔΨm) and the reduction of cytochrome c release suggested the protection for mitochondria. Moreover, a decline of the expression level of Bax, Bcl-2 and caspase-3 and the results of TUNEL staining indicated its anti-apoptosis role. Finally, behavioral assessment with Basso Beattle Bresnahan locomotor rating scale (BBB) showed that animals in exendin-4 group achieved a significant increase in BBB scores. Our results suggest that exendin-4 prevents against SCI-induced mitochondrial apoptotic pathway and contributes to functional improvement after SCI.

The intention of this paper was to design an effective and feasible method to control the seismic responses of structures within a safe range. Through the Shape Memory Alloy, this is achieved. In this paper, several tests were made to find out the impacts that temperature and max strain values may make on Shape Memory Alloy pseudo-elasticity effects, then, according to the experiment result, 2 kinds of Shape Memory Alloy dampers are designed and their mechanical performances are tested, after that, a passive seismic response control system is design using the dampers, at last, the control system was installed on the model of an actual high-rise structure, and the control effect was tested and proved to be well.

A new high temperature superconductor axial-flux coreless maglev motor (HTS AFIM) is proposed, of which the primary windings are made of HTS tapes and the secondary is a non-magnetic conductor. The main works of this paper are the magnetic-field computation and characteristics analysis of HTS AFIM. For the first one, the reduction of magnetic fields near outer and inner radius of the HTS AFIM is solved by introducing the sub-loop electro-magnetic model along the radial position. For the second one, the AC losses of HTS coils are calculated. The relationships between the device’s characteristics and device parameters are presented, and the results indicate that under certain frequency and current levitation device can output enough lift force. The conclusions are verified by finite element calculations.

The antioxidant activities of extracts and residuum of Herba Leonuri fruits from supercritical CO 2 extraction were determined in vitro. The residuum was extracted in turn with water, propyl alcohol and ethyl acetate. The antioxidant activities of the extracts were assayed with antioxidant capacity in linoleic acid model system, reducing powers, radical scavenging activity using 1, 1-diphenyl-2-picrylhy-drazyl (DPPH) method. The results show that the ethyl acetate extract of Herba Leonuri possesses strongest DPPH radical scavenging activity and reducing power in a concentration-dependent manner.

Large urbanized polders, are always with developed economy and concentrated population, in which the lacking river storage capacity makes the risk of flood and waterlog increase. Therefore, it's much more necessary to study more accurate method of drainage calculation and the relationship between the drainage modulus and the drainage area for the drainage strategy in urbanized polder. According to the analysis of the characteristics of urban drainage, Unsteady Flow Equations was used to calculate in details. In the process, the node with drainage pump stations was treated as the virtual outer river in order to suit the characteristics of pump draining. With the drainage area changing, relationship between the drainage modulus and the area was gained, which can provide the basis for choosing appropriate drainage strategy.

Study design Retrospectively compared 2 reconstructive techniques after the anterior decompression of multilevel cervical spondylotic myelopathy. Objective To clinically compare the biomechanical stability and neurologic results of 2 reconstructive techniques after the anterior decompression of multilevel cervical spondylotic myelopathy retrospectively. Summary of background data Previous studies comparing different reconstruction techniques after the anterior decompression of multilevel cervical spondylotic myelopathy have yielded mixed results. Some studies have reported a high incidence of graft-plate extrusion when 2 or more corpectomies are performed and reconstructed with a long segmental anterior plate fixation that spans the strut graft without supplemental posterior cervical fixation, a standalone cage and segmental plate fixation after combination 1 level discectomy and 1 level corpectomy used to treat multilevel cervical spondylotic myelopathy were reported have no evidence of late-onset instrumentation-related failure, although 2 techniques have never been directly compared in a consecutive series of patients clinically. Methods A retrospective study of 59 patients with multilevel (3 levels) cervical spondylotic myelopathy treated with 1 of 2 anterior decompression and reconstruction methods were compared. Copectomy method (39 patients) is 2-level corpectomies and long segment end-construct plate fixation; hybrid method (20 patients) is standalone cage and segmental plate fixation after 1-level discectomy combined with 1-level corpectomy. Results The follow-up (mean18 mo) results show both methods had similar, satisfactory recovery of neurologic function (P>0.05). There were 7 cases of graft/plate migrations or dislodgments (17.9%) and 4 of these required revision surgery among 39 patients with corpectomy method, as compared with no graft/implant-related complications or nonunion among 20 patients with hybrid method (P Conclusions This study clearly demonstrates that, adequate decompression can be achieved for the multilevel cervical spondylotic myelopathy with both methods, and the hybrid method offers better biomechanical stability and fusion results than the corpectomy method, and obviates the need for staged circumferential procedures.

Linear induction motor has been applied to magnetic suspension maglev and metro vehicle. Its thrust and normal force analyses are important basis of motor design and control. The longitudinal end effects have strong influence to performance of linear induction motor, we consider this influence through equivalent EMF. Aiming to the conductivity of secondary and ferromagnetic secondary, transverse edge effects are considered by different modified coefficients. After got modified coefficients, we can compute thrust and consider end effects. Normal force is computed by one-dimensional analysis and two-dimensional LIM model with multi-layer secondary. Finally, we got the thrust and normal force by a model of a single-sided linear induction motor under different air gap and current frequency. The results are then applied to the single-sided LIMpsilas, and its performances are predicted. Results from analyses are compared with those obtained from tests.

The study was performed to investigate the effects of bradykinin preconditioning on spinal cord ischemic injury using an in vivo transient spinal cord ischemia model in rats. Prior to ischemia, bradykinin was infused continuously via the left femoral artery starting 15 min before ischemia. Neurological functions were evaluated for 7 days postoperatively using modified Tarlov's scores. Tarlov's score outcomes showed a marked improvement in the bradykinin group compared to the ischemia group. The blood-spinal cord barrier (BSCB) permeability was also decreased by bradykinin preconditioning after 72 h reperfusion focal spinal cord in rats, which was greatly reversed by B9430 (bradykinin B2 receptor antagonist). Immunohistochemical and Western blot analysis of spinal cords revealed a significant increase in basic fibroblast growth factor protein (bFGF) levels. The study demonstrated that bradykinin preconditioning induces protection against spinal cord ischemic injury, and this protection is likely due to the protection of the vasculature of the spinal cord and the promotion of neuronal survival.

Electromagnetic environment in the underground has a fatal impact on the reliability and safety of signalling equipments. This paper introduces the ideal differential-mode and common-mode model of linear induction motor (LIM) used in urban rail transit system, and points out the potential EMI problem. For the designed laboratory model, the authors designed the EMI measurement scheme referring to the rotary induction motor regulations. Then the common-mode disturbances and radiation emission experimental results are presented and analyzed. The experimental curves demonstrate that these disturbances are mostly generated by the pulse width modulated (PWM) inverter and the values can be met with CLASS-A standard. The experimental results indicate that the performance of the model of linear traction motor is favorable.

The electronically controlled fuel injection system in locomotive diesel is a complicated nonlinear system. So we lead the NARMAX (nonlinear auto-regressive moving average with exogenous inputs) neural network into its model. In order to overcome the deficiency that the neural network structure relies on one's own personal experience, we used the pruning based on the Hession matrix to optimize the network structure. NDEKF (node-decoupled extend Kalman filter) which was adopted to train networks converges more quickly than the back-propagation algorithm does and assists in the avoidance of local minimum. The experiments showed that the hybrid neural networks of the nonlinear auto-regressive with exogenous outputs are very close to the actual results, and the inputs can identify object ranks precisely.

To facilitate the integration of learning resources categorized under different ontology representations, the techniques of ontology mapping can be applied. Through many algorithms and systems have been proposed for ontology mapping, they do not have an automatic weighting strategy on class features to automate the ontology mapping process. A novel method of computing the feature weights is proposed by feature semantic analysis, defining characteristics of the different entities similarity calculation model and weight calculation model. The results show that it makes the ontology mapping process more automatic while retaining satisfying accuracy. Improve ontology mapping effectiveness.

The modeling of permanent-magnet linear synchronous motor is very important to the control and the static and dynamic characters analysis of the system. In this paper, the model of permanent-magnet linear synchronous motor is presented by using neural networks of the nonlinear autoregressive with exogenous inputs. Based on the same cost function, residual signal analysis is mixed into the networks, and then the networks can identify motor's ranks automatically. First, the nonlinear autoregressive with exogenous inputs model is expanded into the polynomial function, then the condition which true ranks satisfy is presented by using residual signal analysis. Some shortages of BP (back-propagation algorithm) are considered, so NDEKF (node-decoupled extend Kalman filter) is applied to train networks. The experiment results show that the hybrid neural networks of the nonlinear autoregressive with exogenous inputs can identify object's (a vertical transport system driven by permanent-magnet linear synchronous motor) ranks precisely, and the output of networks is very close to the experimental result. In the experiments, the performance of NDEKF is often superior to that of BP, while requiring significantly fewer presentations of training data than BP and less over training time than that of BP.

Objectives PAK5 is a member of P21-activated kinase (PAK) family, which characterised by a highly conserved amino-terminal Cdc42/Rac interactive binding (CRIB) domain and a carboxyl terminal kinase domain. However, the role of PAK5 in apoptosis remains largely unknown. In this study, we constructed pEGFP-C1-PAK5 and established a stably transfected cell line and the effect of PAK5 on cell apoptosis was examined. The results suggest that PAK5 could inhibit the apoptosis induced by staurosporine. Methods A. Materials: All the materials used in this study were listed in Table 1. (B) Designation and synthesis of primer: Primers target to PAK5 gene were designed according to GenBank. Forward primer 5′-GAAGATCTATGTTTGGGAAGAAAAAG-3′ with addition of BglII restriction enzyme recognition site and reverse primer: 5′-CGGAATTCTCAGTGATGCCTGTA TTG-3′ with EcoRI addition of site. (C) Acquisition of PAK5 Coding sequence: We performed RT-PCR (reverse transcription PCR) using total RNA extracting from People fresh placental tissue. Then we used cDNA library as template and the primers to make PCR. Full-length PAK5 cDNA coding sequence (CDS) was cloned. The length is 2160 bp. (D) Construction of eukaryotic expression vector of PAK5 and DNA sequencing. We used T4 DNA ligase to connect the PCR products and pEGFP-C1 plasmid which had both been cut by restriction enzyme EcoRI/BglII. After transforming it into competent cells E.coli DH5α and screening of kanamycin, we picked the positive cloning colony to extract plasmid DNA and made a 0.8% gel electrophoresis identification after EcoRI/BglII cleavage, sequencing by Sangon Ltd. of Shanghai. (E) Establishment of stably transfected cell line: The recombinant plasmid pEGFP-C1-PAK5 was transfected into hela cells by lipofectamine. Then we cultured the transfected cells by G418 (700 mg/l) for 6 weeks 48 h later and propagated hela cells by limited dilution method. Therefore, a stably transfected cell line named hela-PAK5 was established. (F) MTT: We prepared four parallel groups: a: hela cells, b:hela cells, c:hela-PAK5 cells, d:hela-PAK5 cells. Then we added staurosporine into group b and group d after 24 h cultivating. About 5 h later, we added MTT into each group and got OD values of each group by using Microplate Reader. Results (A) Identification of eukaryotic expression vector. We got two bands by electrophoresis after EcoRI and BglII restriction enzyme cleavage. As showned in Fig 1, a clear band about 2100 bp in agrose gel can be seen as expected according to the gene sequence. The other band is about 4700 bp which is the expression vector pEGFP-C1. It suggests that PAK5 gene was introduced into pEGFP-C1. (B) DNA sequence analysis: After sequencing analysis of recombinant plasmid of pEGFP-C1-PAK5, we confirmed that the inserted fragment was PAK5 gene by BLAST. The result of DNA sequence is showed in figure 2. (C) Application MTT method for hela—pak5 cells survival: The cells with staurosporine compared with the control group are different, and the apoptosis are detected p 0.05) (figure 3). Conclusions In our study, we constructed the eukaryotic expression vector pEGFP-C1-PAK5 successfully and transfected the PAK5 gene stably into hela cells. And the experiment proves that PAK5 gene is vital to cell survival by MTT method. It can be seen from the results that hela cells appear obvious apoptosis after adding staurosporine while hela-PAK5 cells grow well after adding staurosporine. PAK5 plays a role in inhibiting cell apoptosis. P21-activated kinase5 (PAK5) is the recently identified member of the group II. P21-activated kinase (PAK) family which have been known from its structure, function, mitochondrial positioning and the nucleus shuttle. Although the mechanisms are not completely clear, it plays extremely important role in signal transduction pathways. It provides a strong basis for the study of related disease in apoptosis and opens up a new research direction. The construction of eukaryotic expression vector of human PAK5 and establishment of stably transfected cell line have laid a good experimental basis for subsequent experiment. We will continue our experiment from mitochondria positioning and signal transduction pathways for a further research.

Apoptosis is a programmed cell death, which plays an important role in normal development and maintenance of homeostasis of a variety of tissues including cardiovascular system. Apop-1 is a novel protein that induces apoptosis of cultured smooth muscle cells. RT-PCR data showed that Apop-1 is expressed in all the organs tested. Western blotting revealed that Apop-1 protein is localised in mitochondria. Transient transfection experiment showed the apoptosis of Apop-1 can induce release of cytochrome c. The apoptosis may contribute to the loss of VSMC in atherosclerosis, which will influence the stability of atherosclerotic plaques.

Objective Exploring the anti-oxidative function of traditional Chinese herbal-Korean Monkshood Root polysaccharide. Methods Using GE Corporation9s KTA explore FPLC purification system, we separate and purify the Korean Monkshood Root polysaccharide. After establishing the anti-oxidation experimental model, experimental study of elimination of the ultra oxygen anion and DPPH free radical are carried out. Results As the active ingredient, Korean Monkshood Root polysaccharides can remove O 2− and DPPH free radical, which shows that Korean Monkshood Root polysaccharide can eliminate free radical and has the function of anti-oxidation. Conclusions Korean Monkshood Root polysaccharides can be used as a natural anti-oxidation for human cardiovascular disease treatment and preventive health care.
