Synthesis of high-performance single crystal LiNi0.8Mn0.1Co0.1O2 (NMC811) in the absence of molten salt is challenging with no success yet. An innovative drop-in approach is discovered to synthesize single crystal NMC811 by controlling the morphology of transition metal hydroxide TM(OH)2 precursors followed by a simple decomposition step to form transition metal oxide (TMO) intermediates. Ni redistribution in TMO, as a result of the concurrent formation of mixed spinel and rock salt phases, helps deagglomerate the later formed NMC811 clusters of single crystals. As-prepared single crystal NMC811 is validated in a 2Ah pouch cell demonstrating 1000 stable cycling. The fundamentally new reaction mechanism of single crystal growth and segregation without molten salt provides a new direction towards cost-efficient manufacturing of single crystal NMC811 cathode for advanced lithium-based batteries., Comment: 13 pages, 5 figures

Background Melanocortin 3 receptor (MC3R), a rhodopsin-like G protein-coupled receptor, is an important regulator of metabolism. Although MC3R knock-out (KO) mice and rats were generated in earlier studies, the function of MC3R remains elusive. Since pig models have many advantages over rodents in metabolism research, we generated an MC3R-KO pig using a CRSPR/Cas9-based system combined with somatic cell nuclear transfer (SCNT) technology. Method Four CRSPR/Cas9 target vectors were constructed and then their cleavage efficiency was tested in porcine fetal fibroblasts (PFFs). The pX330-sgRNA1 and pX330-sgRNA4 vectors were used to co-transfect PFFs to obtain positive colonies. PCR screening and sequencing were conducted to identify the genotype of the colonies. The biallelically modified colonies and wild-type control colonies were used simultaneously as donor cells for SCNT. A total of 1203 reconstructed embryos were transferred into 6 surrogates, of which one became pregnant. The genotypes of the resulting piglets were determined by PCR and sequencing, and off-target effects in the MC3R KO piglets were detected by sequencing. Then, offspring were obtained through breeding and six male KO pigs were used for the growth performance analysis. Results Four vectors were constructed successfully, and their cleavage efficiencies were 27.96, 44.89, 32.72 and 38.86%, respectively. A total of 21 mutant colonies, including 11 MC3R−/− and 10 MC3R+/− clones, were obtained, corresponding to a gene targeting efficiency of 29.17%, with 15.28% biallelic mutations. A total of 6 piglets were born, and only two MC3R KO piglets were generated, one with malformations and a healthy one. No off-target effects were detected by sequencing in the healthy mutant. Six male MC3R KO pigs were obtained in the F2 generation and their body weight and body fat were both increased compared to wild-type full siblings. Conclusion A MC3R KO pig strain was generated using the CRSIPR/Cas9-based system, which makes it possible to study the biological function of MC3R in a non-rodent model. Based on the Internet of things technology, this paper proposes to develop an intelligent management system of big data center computer room using HTML5. In a variety of intelligent mobile terminal systems, it receives real-time early warning information such as temperature and humidity environment in the machine room of the big data center, power, server cloud operating system and so on, and realizes the intelligent management function of the power system such as suction fan, anti-theft, air conditioning, power distribution cabinet and UPS power supply. Through the practical application of the system, the management level of the machine room of the big data center is improved effectively with the advantages of the unattended intelligent management and reducing the workload of the machine room administrators. It provides strong security for the computer room of big data center.

State-of-the-art lithium (Li)-ion batteries are approaching their specific energy limits yet are challenged by the ever-increasing demand of today’s energy storage and power applications, especially for electric vehicles. Li metal is considered an ultimate anode material for future high-energy rechargeable batteries when combined with existing or emerging high-capacity cathode materials. However, much current research focuses on the battery materials level, and there have been very few accounts of cell design principles. Here we discuss crucial conditions needed to achieve a specific energy higher than 350 Wh kg −1 , up to 500 Wh kg −1 , for rechargeable Li metal batteries using high-nickel-content lithium nickel manganese cobalt oxides as cathode materials. We also provide an analysis of key factors such as cathode loading, electrolyte amount and Li foil thickness that impact the cell-level cycle life. Furthermore, we identify several important strategies to reduce electrolyte-Li reaction, protect Li surfaces and stabilize anode architectures for long-cycling high-specific-energy cells.

To enable next-generation high-energy-density lithium (Li)-metal batteries (LMBs), an electrolyte that has simultaneous high Li-metal Coulombic efficiency (CE) and high anodic stability on cathodes is of significant importance. Sulfones are known for strong resistance against oxidation, yet their application in LMBs is restricted because of their poor compatibility with Li-metal anodes, high viscosity, and poor wettability. Here, we demonstrate that a high Li CE over 98% can be achieved in concentrated sulfone-based electrolytes. Furthermore, the viscosity and wettability issues of sulfones are resolved by the addition of a fluorinated ether, 1,1,2,2-tetrafluoroethyl-2,2,3,3-tetrafluoropropyl ether, to form a localized high-concentration electrolyte (LHCE), which not only provides further improvement in Li CE (98.8%) but also remains anodically stable with high-voltage cathodes, suppresses Al corrosion, and enables LMBs to operate in a wide temperature range. As a result, significantly improved cycling performance of LMBs has been realized with sulfone-based LHCE. For high-voltage rechargeable lithium (Li)-metal batteries (LMBs), electrolytes with good stabilities on both the highly oxidative cathodes and the highly reductive Li-metal anodes are urgently desired. Sulfones have excellent oxidative stability, yet their high viscosity, poor wettability, and, in particular, incompatibility with Li anodes greatly hinder their applications in LMBs. Here, we demonstrate that a high Li Coulombic efficiency (CE) of 98.2% during repeated Li plating and stripping cycles can be realized in concentrated lithium bis(fluorosulfonyl)imide (LiFSI)-tetramethylene sulfone electrolyte. More importantly, the localized high-concentration electrolyte, formed by the dilution of the high-concentration electrolyte with a non-solvating fluorinated ether, 1,1,2,2-tetrafluoroethyl-2,2,3,3-tetrafluoropropyl ether, solves the viscosity and wettability issues, further improves Li CE (98.8%), and improves the high-voltage (4.9 V) performance of LMBs with effective Al protection. High-voltage batteries with Li-metal anodes can offer desirable high energy densities. Despite their excellent oxidative stability, sulfones have various limitations to be useful in Li-metal batteries, in particular their instability with Li metal. Here, we achieved a high Li Coulombic efficiency of nearly 99% in a sulfone-based localized high-concentration electrolyte (LHCE) with the addition of a non-solvating co-solvent. In addition, this co-solvent is highly beneficial for realizing stable battery cycling up to 4.9 V.

International audience; AbstractDiarrhea remains one of the leading causes of morbidity and mortality globally, with enterotoxigenic Escherichia coli (ETEC) constituting a major causative pathogen. The development of alternative treatments for diarrhea that do not involve chemotherapeutic drugs or result in antibiotic resistance is critical. Considering that lysozyme is a naturally occurring antimicrobial peptide, in a previous study we developed a transgenic pig line that expresses recombinant human lysozyme (hLZ) in its milk. In the present study, we examined the protective effects of the consumption of this milk against ETEC infection in neonatal piglets. We found that consuming hLZ milk facilitated faster recovery from infection and decreased mortality and morbidity following an ETEC oral inoculation or infection acquired by contact-exposure. The protective effect of hLZ was associated with the enrichment of intestinal bacteria that improve gut health, such as Lactobacillus, and the enhancement of the mucosal IgA response to the ETEC-induced diarrhea. Our study revealed potential protective mechanisms underlying the antimicrobial activity of human lysozyme, validating the use of lysozyme as an effective preventive measure for diarrhea.

Genome-wide association studies (GWAS) have identified numerous common prostate cancer (PrCa) susceptibility loci. We have fine-mapped 64 GWAS regions known at the conclusion of the iCOGS study using large-scale genotyping and imputation in 25 723 PrCa cases and 26 274 controls of European ancestry. We detected evidence for multiple independent signals at 16 regions, 12 of which contained additional newly identified significant associations. A single signal comprising a spectrum of correlated variation was observed at 39 regions; 35 of which are now described by a novel more significantly associated lead SNP, while the originally reported variant remained as the lead SNP only in 4 regions. We also confirmed two association signals in Europeans that had been previously reported only in East-Asian GWAS. Based on statistical evidence and linkage disequilibrium (LD) structure, we have curated and narrowed down the list of the most likely candidate causal variants for each region. Functional annotation using data from ENCODE filtered for PrCa cell lines and eQTL analysis demonstrated significant enrichment for overlap with bio-features within this set. By incorporating the novel risk variants identified here alongside the refined data for existing association signals, we estimate that these loci now explain ∼38.9% of the familial relative risk of PrCa, an 8.9% improvement over the previously reported GWAS tag SNPs. This suggests that a significant fraction of the heritability of PrCa may have been hidden during the discovery phase of GWAS, in particular due to the presence of multiple independent signals within the same region

The hospitalization cost of patients with colorectal cancer was studied to reveal influencing factors to explore traditional Chinese medicine (TCM) hospital performance characteristics, distribution law of factors affecting costs, and proposal of reducing medical costs. Retrospective analysis was conducted on the data of 3046 cases with colorectal cancer on basic medical insurance in Beijing in 2008. Factors of medical quality and costs were investigated by single and multivariate analysis. The relation of different factors and hospitalization cost was discussed. The results showed that major factors influencing hospitalization costs were hospitalization days, hospital category, main diag- nosis, hospital rank, operation or not, combination with cardiocerebral diseases, etc. It was concluded that hospitalization costs can be reduced by promoting applied rate of TCM, shortening hospitalization duration, promoting level of diagnosis and treatment, and service quality.

Atherosclerosis is a leading cause of death worldwide, and its mechanisms are still unclear. However, various animal models have significantly advanced our understanding of the mechanisms involved in atherosclerosis and have allowed the evaluation of therapeutic options. The aim of this paper is to review those animal models (i.e., rabbits, mice, rats, guinea pigs, hamsters, avian, carnivores, swine, and, non-human primates) that have been used to study atherosclerosis. Though there is no single perfect animal model that completely replicates the stages of human atherosclerosis, cholesterol feeding and mechanical endothelial injury are two common features shared by most models of atherosclerosis. Further, with the development of genetically modified animals, these models are significantly broadening our understanding of the pathogenesis of atherosclerosis.

Focusing on the new power reform, behavioral models of market participants are established in this paper to describe China's power market in the future and risk factors are listed based on the models. For the risk assessment of market participants, "Stress testing" in financial field is applied to China's power market to present how the risk factors impact on the tested factors.

AADL supports standard accurate modeling of embedded systems and has been widely used in the embedded field. However, as a new modeling language, modeling and analysis tools of AADL are not mature enough. In the field of software engineering, the method and tool about UML model have been developed widely, so we consider designing a transformation method. This method define a complete meta­model for AADL error model and implement the automatic transformation from the AADL model to UML model. Our research makes the method and tool for UML model reused in the AADL model, expands the application scope of AADL model, and reduces the development cost of AADL modeling. Finally, the potential research directions are discussed.
