Background: Laser additive manufacturing has been used for surface repair and remanufacturing due to fast laser processing speed, high energy density, and dense microstructure. However, the properties of coating samples produced by laser additive manufacturing of ironbased alloys vary considerably, resulting in a large amount of data that needs to be accumulated and analyzed. Methods: The coating properties of iron-based alloy powders manufactured by laser cladding are studied. The optimal process parameters of the laser cladding are determined by exploring and comparing the macroscopic appearance, hardness, and conductivity of the junction of the cladding. Results: From the macroscopic appearance, when the ratio of the height to the width of the cladding layer is 3.615, the surface of the cladding layer has a smooth surface and is closely combined with the substrate. Conclusions: The hardness of the cladding layer is found to increase significantly, with an average hardness of 663 HV. Besides, it is found that the blackhead's hole causes the conductivity change. The ratio of the largest hole area to the smallest hole area is 8.29 times, and the depth ratio is 1.91 times, but the average resistance ratio is about 1.6 times.

Background: Data from the US National Health and Nutrition Examination Survey are freely available and can be analyzed to produce hypertension statistics for the noninstitutionalized US population. The analysis of these data requires statistical programming expertise and knowledge of National Health and Nutrition Examination Survey methodology. Methods: We developed a web-based application that provides hypertension statistics for US adults using 10 cycles of National Health and Nutrition Examination Survey data, 1999 to 2000 through 2017 to 2020. We validated the application by reproducing results from prior publications. The application’s interface allows users to estimate crude and age-adjusted means, quantiles, and proportions. Population counts can also be estimated. To demonstrate the application’s capabilities, we estimated hypertension statistics for noninstitutionalized US adults. Results: The estimated mean systolic blood pressure (BP) declined from 123 mm Hg in 1999 to 2000 to 120 mm Hg in 2009 to 2010 and increased to 123 mm Hg in 2017 to 2020. The age-adjusted prevalence of hypertension (ie, systolic BP≥130 mm Hg, diastolic BP≥80 mm Hg or self-reported antihypertensive medication use) was 47.9% in 1999 to 2000, 43.0% in 2009 to 2010, and 44.7% in 2017 to 2020. In 2017 to 2020, an estimated 115.3 million US adults had hypertension. The age-adjusted prevalence of controlled BP, defined by the 2017 American College of Cardiology/American Heart Association BP guideline, among nonpregnant US adults with hypertension was 9.7% in 1999 to 2000, 25.0% in 2013 to 2014, and 21.9% in 2017 to 2020. After age adjustment and among nonpregnant US adults who self-reported taking antihypertensive medication, 27.5%, 48.5%, and 43.0% had controlled BP in 1999 to 2000, 2013 to 2014, and 2017 to 2020, respectively. Conclusions: The application developed in the current study is publicly available at https://bcjaeger.shinyapps.io/nhanesShinyBP/ and produced valid, transparent and reproducible results.

Semantic segmentation of remote sensing (RS) images plays a vital role in a variety of fields, including urban planning, natural disaster monitoring, and land resource management. Due to the complexity and low resolution of RS images, many approaches have been proposed to handle the related task. However, these previously developed approaches dedicate to contextual interaction but ignore the cross-scale semantic correlation and multi-scale boundary information. Therefore, we propose a Cross-scale Graph Interaction Network (CGIN) to address semantic segmentation problems of RS images, which consists of a semantic branch and a boundary branch. In the semantic branch, we first apply atrous convolution to extract multi-scale semantic features of RS images. Particularly, based on the multi-scale semantic features, a Cross-scale Graph Interaction (CGI) module is introduced, which establishes cross-scale graph structures and performs adaptive graph reasoning to capture the cross-scale semantic correlation of RS objects. In the boundary branch, we propose a Multi-scale Boundary Feature Extraction (MBFE) module that utilizes atrous convolutions with different dilation rates to extract multi-scale boundary features. Finally, to address the problem of sparse boundary pixels in the fusion process of the two branches, we propose a Multi-scale Similarity-guided Aggregation (MSA) module by calculating the similarity of semantic features and boundary features at the corresponding scale, which can emphasize the boundary information in semantic features. Our proposed CGIN outperforms state-of-the-art approaches in numerical experiments conducted on two benchmark remote sensing datasets.

This research examines what companies can learn from consumer statements made about them over social media, specifically, how the valence (positive or negative statements) and tweet character (emotional or rational statements) of consumer statements coupled with the type of CSR/CSR related fraud impacts consumers’ beliefs about firm CSR use as well as firm confidence. This was conducted via an experimental design manipulating the type of CSR used and related fraud, and then participants responded to the events via simulated tweets and responded to questions about their beliefs in firms engaging in CSR as well as their overall confidence in a firm. Through applying how individuals assess moral judgments via ethical philosophies (idealism vs. relativism) and how those apply to the types of “tweets” made (emotional vs. rational), it was found that the valence and character of a tweet coupled with the CSR type and the related CSR fraud can be used to understand CSR beliefs and confidence in the firm. Additionally, it was found that tweet valence (positive or negative) moderated by the tweet character (emotional vs. rational) impacts the beliefs that firms should engage in CSR. In certain instances, some consumers will not like that a firm engages in CSR, therefore, after a fraud and related negative buzz, it is recommended that firms withhold broadcasting their CSR activities.

Background. Peripheral nerve injury (PNI) is one of the most debilitating injuries, but therapies for PNI are still far from satisfactory. Pyroptosis, a recently identified form of cell death, has been demonstrated to participate in different diseases. However, the role of pyroptosis of Schwann cells in PNI remains unclear. Methods. We established a rat PNI model, and western blotting, transmission electron microscopy, and immunofluorescence staining were used to confirm pyroptosis of Schwann cells in PNI in vivo. In vitro, pyroptosis of Schwann cells was induced by lipopolysaccharides (LPS)+adenosine triphosphate disodium (ATP). An irreversible inhibitor of pyroptosis, acetyl (Ac)-Tyr-Val-Ala-Asp-chloromethyl ketone (Ac-YVAD-cmk), was used to attenuate Schwann cell pyroptosis. Moreover, the influence of pyroptotic Schwann cells on the function of dorsal root ganglion neurons (DRGns) was analyzed by a coculture system. Finally, the rat PNI model was intraperitoneally treated with Ac-YVAD-cmk to observe the effect of pyroptosis on nerve regeneration and motor function. Results. Schwann cell pyroptosis was notably observed in the injured sciatic nerve. LPS+ATP treatment effectively induced Schwann cell pyroptosis, which was largely attenuated by Ac-YVAD-cmk. Additionally, pyroptotic Schwann cells inhibited the function of DRGns by secreting inflammatory factors. A decrease in pyroptosis in Schwann cells promoted regeneration of the sciatic nerve and recovery of motor function in rats. Conclusion. Given the role of Schwann cell pyroptosis in PNI progression, inhibition of Schwann cell pyroptosis might be a potential therapeutic strategy for PNI in the future.

Background: Stroke is a leading cause of morbidity and mortality worldwide. Neuroinflammation plays a key role in acute brain injury of ischemic stroke. MicroRNA-210 (miR210) is the master hypoxamir and regulates microglial activation and inflammation in a variety of diseases. In this study, we uncovered the mechanism of miR210 in orchestrating ischemic stroke–induced neuroinflammation through repression of TET2 (ten-eleven translocation methylcytosine dioxygenase 2) in the adult mouse brain. Methods: Ischemic stroke was induced in adult WT (wild type) or miR210 KO (miR210 deficient) mice by transient intraluminal middle cerebral artery occlusion. Injection of TET2 silencing RNA or miR210 complementary locked nucleic acid oligonucleotides, or miR210 KO mice were used to validate miR210-TET2 axis and its role in ischemic brain injury. Furthermore, the effect of TET2 overexpression on miR210-stimulated proinflammatory cytokines was examined in BV2 microglia. Post assays included magnetic resonance imaging scan for brain infarct size; neurobehavioral tests, reverse transcription-quantitative polymerase chain reaction, and Western blot for miR210; and TET2 levels, flow cytometry, and ELISA for neuroinflammation in the brain after stroke or microglia in vitro. Results: miR210 injection significantly reduced TET2 protein abundance in the brain, while miR210 complementary locked nucleic acid oligonucleotides or miR210 KO preserved TET2 regardless of ischemic brain injury. TET2 knockdown reversed the protective effects of miR210 inhibition or miR210 KO on ischemic stroke–induced brain infarct size and neurobehavioral deficits. Moreover, flow cytometry and ELISA assays showed that TET2 knockdown also significantly dampened the anti-inflammatory effect of miR210 inhibition on microglial activation and IL (interleukin)-6 release after stroke. In addition, overexpression of TET2 in BV2 microglia counteracted miR210-induced increase in cytokines. Conclusions: miR210 inhibition reduced ischemic stroke–induced neuroinflammatory response via repression of TET2 in the adult mouse brain, suggesting that miR210 is a potential treatment target for acute brain injury after ischemic stroke.

The present meta-analysis investigated the safety and effectiveness of this hybrid anticoagulants management model.Methods: The PubMed, Embase, Cochrane, and Web of Science databases were searched for papers published before May 1, 2022. To reduce bias, only randomized controlled trials were included. RevMan 5.3 (Cochrane) software was used to evaluate and analyze clinical outcomes, including the effectiveness and safety of patient management approaches, determined by the time in the therapeutic range (TTR) and occurrence of thrombotic and bleeding events.Results: Eight studies, comprising 3853 patients, were selected. The meta-analysis showed that anticoagulant management combining portable coagulometers and telemedicine significantly improved frequency of testing (mean difference [MD]= 12.95 days; 95% CI, 8.77– 17.12; I2= 92%; P< 0.01) and TTR (MD= 9.50%; 95% CI, 3.16– 15.85; I2= 87%; P< 0.01). Thromboembolism events were reduced (RR= 0.72; 95% CI, 0.51– 1.01; I2= 0%; P= 0.05), but the results were not statistically significant. And no significant differences in major bleeding events, rehospitalization rate, mortality, or overall treatment cost existed between the two groups.Conclusion: Although the safety of remote cardiovascular disease management is not superior to that of conventional outpatient anticoagulant management, it provides a more stable monitoring of coagulation status.Keywords: VKA therapy, warfarin, telemedicine, self-testing, randomized controlled studies, meta-analysis

The excellent remote sensing ability of synthetic aperture radar (SAR) will be misled seriously when it encounters deceptive jamming which possesses high fidelity and fraudulence. In this paper, the dynamic synthetic aperture (DSA) scheme is used to extract the difference between the true and false targets. A simultaneous deceptive jamming suppression and target reconstruction method is proposed for a single channel SAR system to guarantee remote sensing ability. The system model is formulated as a sparse signal recovery problem with an unknown parametric dictionary to be estimated. An iterative re-weighted method is employed to jointly handle the dictionary parameter learning and target reconstruction problem in an majorization-minimization framework, where a surrogate function majorizing the Gaussian entropy in the objective function is introduced to circumvent its non-convexity. After dictionary parameter learning, the grid mismatching problem in a fixed grid based method is avoided. Therefore, the proposed method can reap a super resolution result. Besides, a simple yet effective DSA section scheme is developed for the SAR data excerpting, in which only two DSAs are required. Experimental results about location error and reconstruction power error reveal that the proposed method is able to achieve a good performance in deceptive jamming suppression.

Proprotein convertase subtilisin/kexin type 9 can mediate the intracellular lysosomal degradation of the low-density lipoprotein receptor protein in hepatocytes and decrease the liver’s ability to scavenge low-density lipoprotein cholesterol from circulation, resulting in high levels of cholesterol in the circulatory system. Current studies have primarily focused on the relationship between PCSK9 and blood lipid metabolism; however, the biological function of PCSK9 in hepatocytes is rarely addressed. In this study, we evaluate its effects in the human hepatoma carcinoma cell line HepG2, including proliferation, migration, and free cholesterol transport. PCSK9-D374Y is a gain-of-function mutation that does not affect proliferation but significantly suppresses the migration and cholesterol efflux capacity of these cells. The suppression of the transmembrane outflow of intracellular-free cholesterol regulates small G proteins and the suppression of extracellular signal-regulated kinase. In summary, PCSK9-D374Y affects hepatocyte features, including their migration and free cholesterol transport capabilities.
