Background: As the number of nuclear facilities nearing their pre-determined design life increases, demand is increasing for technology and infrastructure related to the decommissioning and decontamination (D&D) process. It is necessary to consider the nature of the dismantling environment constantly changing and the worker doing new tasks. A method was studied that can calculate the effect of learning and the change in work time on the work process, according to the learning-forgetting curve model (LFCM).Materials and Methods: The LFCM was analyzed, and input values and scenarios were analyzed for substitution into the D&D process of a nuclear facility.Results and Discussion: The effectiveness and efficiency of the training were analyzed. It was calculated that skilled workers can receive a 16.9% less collective radiation dose than workers with only basic training.Conclusion: Using these research methods and models, it was possible to calculate the change in the efficiency of workers performing new tasks in the D&D process and the corresponding reduction in the work time and collective dose.

Regulatory agencies and civil society organizations concerned with radioactive waste management are demanding maximized safety management of radioactive-waste-containing drums using advanced technology for more efficient and safe management. In the present paper, we propose a sensor system that can track and monitor drums containing low- and intermediate-level radioactive waste using advanced technology based on the Internet of Things (IoT). The proposed system consists of drum nodes, pallet nodes, and gateways. A drum node and magnetic sensor installed on a drum can be used to remotely check whether the drum lid is open or closed, and the location of the drum can be tracked using a near-field communication reader and an infrared module installed on the pallet node. Considering that radioactive waste is stored for a long time period, the proposed sensors are designed for low power consumption. Moreover, a pilot test involving 48 drums and 12 pallets was conducted to verify the system prototype. Accordingly, from the results of the pilot test, drawbacks were noted and solutions were proposed to improve the system in future work.

Two new berkeley meroterpenoids (1 and 2), along with seven known compounds (3‒9) were isolated from a fungus, Penicillium sp. SSW03M2 GY derived from a sediment at Seosan bay, South Korea. Chemical structures of the isolated compounds were elucidated on the basis of 1D, 2D NMR, HRESIMS, and optical rotation. All the isolated compounds, 1 showed anti-virulence activity by significantly inhibiting α-toxin (Hla) secreted by methicillin-resistant Staphylococcus aureus without its growth inhibition.

We investigated the quality characteristics and microbial community found in traditional outdoor-type doenjang fermented at different temperatures. The quality characteristics that were highly correlated with fermentation temperature were analyzed. The average temperature of doenjang in the summer was highest in the order of fermentation by field in Damyang (JDF), fermentation by smart management facility in Damyang (JDS), and fermentation by field in Pyeongchang (JDFP), and in winter was the highest in the order JDS, JDF, and JDFP. The b* value and the degree of browning were highest in the order of JDF, JDS, and JDFP. Among the quality characteristics that affect the degree of fermentation and taste, samples with high summer temperature showed high ammonia nitrogen content, α-amylase activity, sugar content, organic acid content, and acidity and samples with high winter temperature showed high protease activity, amino nitrogen content, biogenic amine content, and alcohol content. Regarding the microbial community, the level of the Enterococcus genus was high in the samples with low winter temperatures, while the level of the Bacillus genus was high in the samples with high winter temperatures. Additionally, the genus Wickerhamiella and Zygosaccharomyces were highest in the samples with the lowest temperatures in summer and winter. Furthermore, acidity, biogenic amine content, glucose content, and lactic acid content were analyzed as major factors influencing the quality characteristics of doenjang according to the ripening region and location.

The repair of large bone defects remains a significant challenge in clinical practice and requires bone grafts or substitute materials. In this study, we developed a unique hybrid bone scaffold comprising a three dimensional (3D)-printed metal plate for weight bearing and a biodegradable polymer tube serving as bone conduit. We assessed the long-term effect of the hybrid bone scaffold in repairing radial bone defects in a beagle model.Bone defects were created surgically on the radial bone of three beagle dogs and individually-tailored scaffolds were used for reconstruction with or without injection of autologous bone and decellularized extracellular matrix (dECM). The repaired tissue was evaluated by X-ray, micro-computed tomography, and histological observation 6 months after surgery. The functional integrity of hybrid bone scaffold-mediated reconstructions was assessed by gait analysis.In vivo analysis showed that the hybrid bone scaffolds maintained the physical space and bone conductivity around the defect. New bone was formed adjacent to the scaffolds. Addition of autologous bone and dECM in the polymer tube improved healing by enhancing bone induction and osteoconduction. Furthermore, the beagles' gait appeared normal by 4 months.The future of bone healing and regeneration is closely related to advances in tissue engineering. Bone production using autologous bone and dECM loaded on 3D-printed hybrid bone scaffolds can successfully induce osteogenesis and provide mechanical force for functional bone regeneration, even in large bone defects.

Plasmodium vivax is the most widespread cause of malaria, especially in subtropical and temperate regions such as Asia-Pacific and America. P. vivax lactate dehydrogenase (PvLDH), an essential enzyme in the glycolytic pathway, is required for the development and reproduction of the parasite. Thus, LDH from these parasites has garnered attention as a diagnostic biomarker for malaria and as a potential molecular target for developing antimalarial drugs. In this study, we prepared a transformed Escherichia coli strain for the overexpression of PvLDH without codon optimization. We introduced this recombinant plasmid DNA prepared by insertion of the PvLDH gene in the pET-21a(+) expression vector, into the Rosetta(DE3), an E. coli strain suitable for eukaryotic protein expression. The time, temperature, and inducer concentration for PvLDH expression from this E. coli Rosetta(DE3), containing the original PvLDH gene, were optimized. We obtained PvLDH with a 31.0 mg/L yield and high purity (>95%) from this Rosetta(DE3) strain. The purified protein was characterized structurally and functionally. The PvLDH expressed and purified from transformed bacteria without codon optimization was successfully demonstrated to exhibit its potential tetramer structure and enzyme activity. These findings are expected to provide valuable insights for research on infectious diseases, metabolism, diagnostics, and therapeutics for malaria caused by P. vivax.

Botrytis cinerea is a necrotrophic fungal pathogen with an extremely broad host range, causing significant economic losses in agricultural production. In this study, we discovered a culture filtrate of bacterial strain HK235, which was identified as Chitinophaga flava, exhibiting high levels of antifungal activity against B. cinerea. From the HK235 culture filtrate, we isolated a new antimicrobial peptide molecule designated as chitinocin based on activity-guided fractionation followed by characterization of the amino acid composition and spectroscopic analyses. The HK235 culture filtrate and chitinocin completely inhibited both conidial germination and mycelial growth of B. cinerea at a concentration of 20% and 200 μg/mL, respectively. In addition to antibiosis against B. cinerea, the active compound chitinocin had a broad antifungal and antibacterial activity in vitro. When tomato plants were treated with the culture filtrate and chitinocin, the treatment strongly reduced the development of gray mold disease in a concentration-dependent manner compared to the untreated control. Here, considering the potent antifungal property in vitro and in vivo, we present the biocontrol potential of C. flava HK235 for the first time.

A multi-physics-analysis-based topology optimization (TO) method is proposed to optimally design the internal partition layout of a muffler integrated with a thermoelectric generator (TEG). The basic equations governing the acoustical behavior, heat transfer, and fluid flow in the muffler are introduced, and their interaction is designated for exact numerical analysis in terms of acoustics, heat transfer, and fluid mechanics. To implement density-based TO, one design variable is assigned to each finite element in the design domain, and interpolation functions suitable for each physics phenomenon are employed. In the TO problem formulation, the sum of the squared acoustic pressures at the outlet of the muffler for multi-target frequencies is selected as an objective function to achieve broadband noise attenuation. The temperature of the TEG and the pressure drop are constrained for high energy recovery efficiency and fluid passage, respectively. The optimization problem formulated for the muffler design is solved for various design conditions. Optimal partition layouts are obtained depending on the location and length of the TEG, the upper limit value of the pressure drop, and the number of target frequencies in the same frequency band. The noise attenuation performances of each partition layout are compared, and their expected recovery energies are calculated. One optimal partition layout is discussed in terms of acoustics, heat transfer, and fluid mechanics. The numerical results strongly support the validity of our proposed method for the optimal design of a muffler integrated with a TEG.

Few studies have investigated long-term clinical outcomes of a mobile-bearing total ankle arthroplasty (TAA) system. This study analyzed long-term outcomes of TAA using the HINTEGRA prosthesis at a single, non-developer center.Primary TAAs were performed on 213 ankles in 194 patients, and 151 consecutive ankles [71%] in 136 patients with a minimum follow-up of 10 years after the primary TAA were included in this study. Clinical results were assessed using a visual analog scale (VAS) pain score, the American Orthopaedic FootAnkle Society (AOFAS) Ankle-Hindfoot Scale score, the Ankle Osteoarthritis Scale (AOS) pain and disability subscores, and ankle range of motion. Prosthesis survivorship, reoperations, and risk factors were also evaluated.The mean follow-up was 135.5 months (range, 120.0 to 204.0 months). All clinical scores and ankle range of motion improved significantly from preoperatively to 2 years, 4 to 6 years, and ≥10 years after TAA (p0.001). A total of 43 ankles (28.5%) required revision procedures, with the most common reason being periprosthetic osteolysis (32 ankles [21.2%]). The overall implant survivorship was 93.5% in Kaplan-Meier survival analysis at the mean follow-up of 11.3 years after the TAA.TAA using the HINTEGRA prosthesis with careful follow-up observation and appropriate adjunct procedures for the treatment of end-stage ankle arthritis produced satisfactory clinical results, which were maintained at a follow-up of ≥10 years, and resulted in 93.5% of implant survivorship.Prognostic Level IV . Background: The evidence supporting best practice guidelines in the field of cartilage repair of the ankle is based on both low quality and low levels of evidence. Therefore, an international consensus group of experts was convened to collaboratively advance toward consensus opinions based on the best available evidence on key topics within cartilage repair of the ankle. The purpose of this article is to report the consensus statements on “terminology for osteochondral lesions of the ankle” developed at the 2019 International Consensus Meeting on Cartilage Repair of the Ankle. Methods: Forty-three international experts in cartilage repair of the ankle representing 20 countries were convened and participated in a process based on the Delphi method of achieving consensus. Questions and statements were drafted within four working groups focusing on specific topics within cartilage repair of the ankle, after which a comprehensive literature review was performed, and the available evidence for each statement was graded. Discussion and debate occurred in cases where statements were not agreed on in unanimous fashion within the working groups. A final vote was then held, and the strength of consensus was characterised as follows: consensus, 51%–74%; strong consensus, 75%–99%; unanimous, 100%. Results: A total of 11 statements on terminology and classification reached consensus during the 2019 International Consensus Meeting on Cartilage Repair of the Ankle. Definitions are provided for osseous, chondral and osteochondral lesions, as well as bone marrow stimulation and injury chronicity, among others. An osteochondral lesion of the talus can be abbreviated as OLT. Conclusions: This international consensus derived from leaders in the field will assist clinicians with the appropriate terminology for osteochondral lesions of the ankle.

Discovering cell–surface markers based on a comprehensive understanding of development is utilized to isolate a particular cell type with high purity for therapeutic purposes. Given that latrophilin-2 (Lphn2) substantially contributes to cardiac differentiation, we examined whether Lphn2 regulates functional significance in heart development and repair. We performed whole-mount immunostaining followed by clearing technique of embryo, RNA sequencing related to Lphn2-knockout (KO) embryo, and in vivo functional analyses of Lphn2+ cells using echocardiography. After immunostaining the cleared embryo sample, Lphn2 was exclusively observed in cardiac cells expressing α-sarcomeric actinin at embryonic days E9.5 and E10.5. Homozygous Lphn2-KO mice were embryonically lethal and showed underdevelopment of the ventricular myocardium. However, Lphn2 was not required to develop vessels, including endothelial cells and smooth muscle cells. For the purpose of cardiac regeneration, we transplanted pluripotent stem cell (PSC)–derived Lphn2+ cells into the infarcted heart. PSC–derived Lphn2+ cells differentiated into cardiomyocytes and regenerated the myocardium when transplanted into the infarcted heart, unlike Lphn2− cells. Transplanted Lphn2+ cells improved left-ventricle systolic function and reduced infarct size. We demonstrated that Lphn2 exhibits potential as a cardiomyogenic marker to facilitate targeted stem cell therapy for heart repair in clinical practice.

Background Although mRNA dysregulation can induce changes in mesenchymal stem cell (MSC) homeostasis, the mechanisms by which post-transcriptional regulation influences MSC differentiation potential remain understudied. PUMILIO2 (PUM2) represses translation by binding target mRNAs in a sequence-specific manner. Methods In vitro osteogenic differentiation assays were conducted using human bone marrow-derived MSCs. Alkaline phosphatase and alizarin red S staining were used to evaluate the osteogenic potential of MSCs. A rat xenograft model featuring a calvarial defect to examine effects of MSC-driven bone regeneration. RNA-immunoprecipitation (RNA-IP) assay was used to determine the interaction between PUM2 protein and Distal-Less Homeobox 5 (DLX5) mRNA. Ovariectomized (OVX) mice were employed to evaluate the effect of gene therapy for postmenopausal osteoporosis. Results Here, we elucidated the molecular mechanism of PUM2 in MSC osteogenesis and evaluated the applicability of PUM2 knockdown (KD) as a potential cell-based or gene therapy. PUM2 level was downregulated during MSC osteogenic differentiation, and PUM2 KD enhanced MSC osteogenic potential. Following PUM2 KD, MSCs were transplanted onto calvarial defects in 12-week-old rats; after 8 weeks, transplanted MSCs promoted bone regeneration. PUM2 KD upregulated the expression of DLX5 mRNA and protein and the reporter activity of its 3'-untranslated region. RNA-IP revealed direct binding of PUM2 to DLX5 mRNA. We then evaluated the potential of adeno-associated virus serotype 9 (AAV9)-siPum2 as a gene therapy for osteoporosis in OVX mice. Conclusion Our findings suggest a novel role for PUM2 in MSC osteogenesis and highlight the potential of PUM2 KD-MSCs in bone regeneration. Additionally, we showed that AAV9-siPum2 is a potential gene therapy for osteoporosis.

Background The endometrium, the inner lining of the uterine cavity, plays essential roles in embryo implantation and its subsequent development. Although some positive results were preliminarily archived, the regeneration of damaged endometrial tissues by administrating stem cells only is very challenging due to the lack of specific microenvironments and their low attachment rates at the sites of injury. In this context, various biomaterial-based scaffolds have been used to overcome these limitations by providing simple structural support for cell attachment. However, these scaffold-based strategies also cannot properly reflect patient tissue-specific structural complexity and thus show only limited therapeutic effects. Method Therefore, in the present study, we developed a customizable Lego-like multimodular endometrial tissue architecture by assembling individually fabricated tissue blocks. Results Each tissue block was fabricated by incorporating biodegradable biomaterials and certain endometrial constituent cells. Each small tissue block was effectively fabricated by integrating conventional mold casting and 3D printing techniques. The fabricated individual tissue blocks were properly assembled into a larger customized tissue architecture. This structure not only properly mimics the patient-specific multicellular microenvironment of the endometrial tissue but also properly responds to key reproductive hormones in a manner similar to the physiological functions. Conclusion This customizable modular tissue assembly allows easy and scalable configuration of a complex patient-specific tissue microenvironment, thus accelerating various tissue regeneration procedures.

There is a direct link between widespread prevalence of clinical giardiasis and poverty. This may be one of the reasons why giardiasis is a significant contributor to diarrheal morbidity, stunting, and death of children in resource-limited communities around the world.

Degradation of the kinetically trapped bulk heterojunction film morphology in organic solar cells (OSCs) remains a grand challenge for their practical application. Herein, we demonstrate highly thermally stable OSCs using multicomponent photoactive layer synthesized via a facile one-pot polymerization, which show the advantages of low synthetic cost and simplified device fabrication. The OSCs based on multicomponent photoactive layer deliver a high power conversion efficiency of 11.8% and exhibit excellent device stability for over 1000 h (>80% of their initial efficiency retention), realizing a balance between device efficiency and operational lifetime for OSCs. In-depth opto-electrical and morphological properties characterizations revealed that the dominant PM6-b-L15 block polymers with backbone entanglement and the small fraction of PM6 and L15 polymers synergistically contribute to the frozen fine-tuned film morphology and maintain well-balanced charge transport under long-time operation. These findings pave the way towards the development of low-cost and long-term stable OSCs.

We report a new molecular design of sequentially regular polymerized small-molecule-acceptors containing flexible spacer (FS) units (PYFS-Reg) to achieve all-polymer solar cells with both high-performance and mechanical stretchability.

We develop a series of polymer donors containing carbazole-based units (CBT) to achieve high-performance (PCE of 15.54%) and thermally stable PSCs.

We summarize a variety of examples of regioregularity-controlled conjugated polymers and describe their impacts on the polymer properties and performances in polymer solar cells.

We prospectively investigated the changes of liver stiffness (LS) and the occurrence of hepatocellular carcinoma (HCC) after hepatitis C virus (HCV) eradication using direct antiviral agents (DAA) over three years. LS measurement using transient elastography and serum fibrosis surrogate markers before treatment and at 48, 96, 144 weeks after starting direct-acting antivirals (DAA) according to the protocol were evaluated. Patients were also compared with historical cohort treated with pegylated interferon (peg-IFN). Sustained viral response (SVR) was observed in 95.8%. New polymer acceptors (PYSiO-X (X = 0–30%)) containing siloxane-based flexible spacers (SiO-FSs) are developed. The resulting halogen-free solvent processed all-polymer solar cells show high photovoltaic efficiency and mechanical robustness.

We develop a non-covalently fused ring acceptor, CPDT–ICMe, to achieve P3HT-based organic solar cells with high-performance (8.17%) and excellent thermal stability.

Cancer stem cells (CSCs) are a small subset of cancer cells with stem cell-like properties, self-renewal potential, and differentiation capacity into multiple cell types. Critical genetic alterations or aberrantly activated signaling pathways associated with drug resistance and recurrence have been observed in multiple types of CSCs. In this context, CSCs are considered to be responsible for tumor initiation, growth, progression, therapeutic resistance, and metastasis. Therefore, to effectively eradicate CSCs, tremendous efforts have been devoted to identify specific target molecules that play a critical role in regulating their distinct functions and to develop novel therapeutics, such as proteins, monoclonal antibodies, selective small molecule inhibitors, and small antisense RNA (asRNA) drugs. Similar to other CSC types, oral CSCs can be characterized by certain pluripotency-associated markers, and oral CSCs can also survive and form 3D tumor spheres in suspension culture conditions. These oral CSC-targeting therapeutics selectively suppress specific surface markers or key signaling components and subsequently inhibit the stem-like properties of oral CSCs. A large number of new therapeutic candidates have been tested, and some products are currently in the pre-clinical or clinical development phase. In the present study, we review new oral CSC-targeted therapeutic strategies and discuss the various specific CSC surface markers and key signaling components involved in the stem-like properties, growth, drug resistance, and tumorigenicity of oral CSCs.

Background/aims No information is available regarding the influence of besifovir (BSV) on the occurrence of hepatocellular carcinoma (HCC) in patients with chronic hepatitis B (CHB). This study aimed to assess the reduced risk of HCC in patients undergoing BSV treatment. Methods Overall, 188 patients with CHB were treated with BSV for up to 8 years. We assessed the incidence of HCC during follow-up and compared it with the predictive numbers of HCC using models developed from untreated CHB patients. Additionally, we compared the performance of various HCC prediction models developed for patients with CHB receiving antiviral therapy. Results During the follow-up period of 8 years, five patients developed HCC; one of 139 patients with non-cirrhotic CHB, and four of 49 patients with liver cirrhosis. We compared the HCC incidence in non-cirrhotic patients with the predicted number derived from the REACH-B model. The standardized incidence ratio (SIR) was 0.128 (P = 0.039) at 7 years, suggesting a significant decrease in HCC incidence in non-cirrhotic CHB patients. The incidence of HCC in patients with cirrhosis was compared using the GAG-HCC model, and the SIR was 0.371 (P = 0.047) at 7.5 years, suggesting a significantly decreased HCC incidence. When we compared several HCC prediction models developed for CHB patients under antiviral therapy, the HCC-RESCUE model showed the highest area under the curve (0.924). Conclusions BSV decreases the risk of HCC in patients with CHB, with or without liver cirrhosis. HCC prediction was available for BSV-treated patients using the existing prediction models.
