Y900 is one of the top hybrid rice (Oryza sativa) varieties, with its yield exceeding 15 t·hm−2. To dissect the mechanism of heterosis, we sequenced the male parent line R900 and female parent line Y58S using long-read and Hi-C technology. High-quality reference genomes of 396.41 Mb and 398.24 Mb were obtained for R900 and Y58S, respectively. Genome-wide variations between the parents were systematically identified, including 1,367,758 single-nucleotide polymorphisms, 299,149 insertions/deletions, and 4,757 structural variations. The level of variation between Y58S and R900 was the lowest among the comparisons of Y58S with other rice genomes. More than 75% of genes exhibited variation between the two parents. Compared with other two-line hybrids sharing the same female parent, the portion of Geng/japonica (GJ)-type genetic components from different male parents increased with yield increasing in their corresponding hybrids. Transcriptome analysis revealed that the partial dominance effect was the main genetic effect that constituted the heterosis of Y900. In the hybrid, both alleles from the two parents were expressed, and their expression patterns were dynamically regulated in different tissues. The cis-regulation was dominant for young panicle tissues, while trans-regulation was more common in leaf tissues. Overdominance was surprisingly prevalent in stems and more likely regulated by the trans-regulation mechanism. Additionally, R900 contained many excellent GJ haplotypes, such as NARROW LEAF1, Oryza sativa SQUAMOSA PROMOTER BINDING PROTEIN-LIKE13, and Grain number, plant height, and heading date8, making it a good complement to Y58S. The fine-tuned mechanism of heterosis involves genome-wide variation, GJ introgression, key functional genes, and dynamic gene/allele expression and regulation pattern changes in different tissues and growth stages.

STUDY QUESTIONCan long-read amplicon sequencing be beneficial for preclinical preimplantation genetic testing (PGT) workup in couples with a de novo pathogenic variant in one of the prospective parents?SUMMARY ANSWERLong-read amplicon sequencing represents a simple, rapid and cost-effective preclinical PGT workup strategy that provides couples with de novo pathogenic variants access to universal genome-wide haplotyping-based PGT programs.WHAT IS KNOWN ALREADYUniversal PGT combines genome-wide haplotyping and copy number profiling to select embryos devoid of both familial pathogenic variants and aneuploidies. However, it cannot be directly applied in couples with a de novo pathogenic variant in one of the partners due to the absence of affected family members required for phasing the disease-associated haplotype.STUDY DESIGN, SIZE, DURATIONThis is a prospective study, which includes 32 families that were enrolled in the universal PGT program at the University Hospital of Leuven between 2018 and 2022. We implemented long-read amplicon sequencing during the preclinical PGT workup to deduce the parental origin of the disease-associated allele in the affected partner, which can then be traced in embryos during clinical universal PGT cycles.PARTICIPANTS/MATERIALS, SETTING, METHODSTo identify the parental origin of the disease-associated allele, genomic DNA from the carrier of the de novo pathogenic variant and his/her parent(s) was used for preclinical PGT workup. Primers flanking the de novo variant upstream and downstream were designed for each family. Following long-range PCR, amplicons that ranged 5–10 kb in size, were sequenced using Pacific Bioscience and/or Oxford Nanopore platforms. Next, targeted variant calling and haplotyping were performed to identify parental informative single-nucleotide variants (iSNVs) linked to the de novo mutation. Following the preclinical PGT workup, universal PGT via genome-wide haplotyping was performed for couples who proceeded with clinical PGT cycle. In parallel, 13 trophectoderm (TE) biopsies from three families that were analyzed by universal PGT, were also used for long-read amplicon sequencing to explore this approach for embryo direct mutation detection coupled with targeted long-read haplotyping.MAIN RESULTS AND THE ROLE OF CHANCEThe parental origin of the mutant allele was identified in 24/32 affected individuals during the preclinical PGT workup stage, resulting in a 75% success rate. On average, 5.95 iSNVs (SD = 4.5) were detected per locus of interest, and the average distance of closest iSNV to the de novo variant was ∼1750 bp. In 75% of those cases (18/24), the de novo mutation occurred on the paternal allele. In the remaining eight families, the risk haplotype could not be established due to the absence of iSNVs linked to the mutation or inability to successfully target the region of interest. During the time of the study, 12/24 successfully analyzed couples entered the universal PGT program, and three disease-free children have been born. In parallel to universal PGT analysis, long-read amplicon sequencing of 13 TE biopsies was also performed, confirming the segregation of parental alleles in the embryo and the results of the universal PGT.LIMITATIONS, REASONS FOR CAUTIONThe main limitation of this approach is that it remains targeted with the need to design locus-specific primers. Because of the restricted size of target amplicons, the region of interest may also remain non-informative in the absence of iSNVs.WIDER IMPLICATIONS OF THE FINDINGSTargeted haplotyping via long-read amplicon sequencing, particularly using Oxford Nanopore Technologies, provides a valuable alternative for couples with de novo pathogenic variants that allows access to universal PGT. A synergistic photocatalytic system using a bisphosphonium catalyst and a cobalt catalyst has been developed, enabling the selective oxidation of benzylic alcohols under oxidant-free and environmentally benign conditions.

To evaluate the diagnostic performance of radiologists on breast cancer with or without artificial intelligence (AI) support.A retrospective study was performed. In total, 643 mammograms (average age: 54 years; female: 100%; cancer: 62.05%) were randomly allocated into two groups. Seventy-five percent of mammograms in each group were randomly selected for assessment by two independent radiologists, and the rest were read once. Half of the 71 radiologists could read mammograms with AI support, and the other half could not. Sensitivity, specificity, Youden's index, agreement rate, Kappa value, the area under the receiver operating characteristic curve (AUC) and the reading time of radiologists in each group were analyzed.The average AUC was higher if the AI support system was used (unaided: 0.84; with AI support: 0.91; p 0.01). The average sensitivity increased from 84.77% to 95.07% with AI support (p 0.01), but the average specificity decreased (p = 0.07). Youden's index, agreement rate and Kappa value were larger in the group with AI support, and the average reading time was shorter (p 0.01).The AI support system might contribute to enhancing the diagnostic performance (e.g., higher sensitivity and AUC) of radiologists. In the future, the AI algorithm should be improved, and prospective studies should be conducted.

Ammonia (NH3) is the most important alkaline gas in the atmosphere, which has negative effects on biodiversity, ecosystems, soil acidification and human health. China has the largest NH3 emissions globally, mainly associated with agricultural sources including nitrogen fertilizer and livestock. However, there is still a limited number of ground monitoring sites in China, hindering our understanding of both surface NH3 concentrations and emissions. In this study, using the polar-orbiting satellite (Infrared Atmospheric Sounding Interferometer – IASI) and Fengyun-4A Geostationary Interferometric Infrared Sounder (GIIRS), we analyzed the changes in hourly NH3 concentrations and estimated surface NH3 concentrations and NH3 emissions in China. GIIRS-derived NH3 concentrations in the daytime were generally higher than those at night, with high values during 10:00–16:00 local time. Satellite-derived surface NH3 concentrations were generally consistent with the ground observations, with R-square at 0.72 and slope equal to 1.03. Satellite-based NH3 emissions ranged from 12.17 to 17.77 Tg N yr−1 during 2008–2019. Spatially, high values of NH3 emissions mainly occurred in the North China Plain, Northeast China and the Sichuan Basin, while low values were mainly distributed in West China (Qinghai–Tibet Plateau). Our study shows a high predictive power of using satellite data to estimate surface NH3 concentrations and NH3 emissions over multiple temporal and spatial scales, which provides an important reference for understanding NH3 changes over China.

Four new diphenyl ethers, named epicoccethers K-N (1-4), were purified from the fermentation medium of a fungus Epicoccum sorghinum derived from Myoporum bontioides, and identified through HR-ESI-MS and NMR spectral analysis. Except that compound 1 showed moderate antifungal activity against Penicillium italicum and Fusarium graminearum, the other three compounds showed stronger activity against them than triadimefon. All of them showed moderate or weak antibacterial activity towards Staphylococcus aureus and Escherichia coli with O6 and O78 serotypes except that 3 was inactive to E. coli O6.

A theory of everything should not only tell us the laws for matter, gravity, and possibly boundary condition for the universe. In addition, it should specify the relation between theory and experience. Here I argue for a minimal prescription in extracting empirical predictions from path integrals by showing that alternative prescriptions are unjustifiable. In this minimal prescription, the relative probability for one experience is obtained by summing over all configurations compatible with that experience, without any further restriction associated with other experiences of the same or other experiential beings. An application to Wigner's friend settings shows that quantum theory admits objective predictions for subjective experiences. Still, quantum theory differs from classical theory in offering individualized as opposed to collective accounts of experiences. Feeding an increasingly affluent population is a huge challenge facing global agriculture. In contrast to “large-scale farming” in developed economies (e.g. Europe and the United States), developing countries are dominated by “smallholder farms” relying on traditional farming practices but increasingly with substantial nitrogen overuse leading to severe environmental degradation and adverse human health. Here, we explore the potential for better nitrogen use by synthesizing the global relationship between farm size and nitrogen use for 16 major crops, assess the impact of farm size on nitrogen flows, and link these with air quality modelling to produce an integrated assessment of nitrogen-related environmental and health outcomes related to farm size. We find that increasing farm size in developing countries can contribute to more efficient and sustainable farming practices, which could decrease nitrogen overuse, ammonia emissions and nitrogen deposition by 20-25%, increase nitrogen use efficiency by 2-8%, and save over 142,000 premature deaths per year related to PM2.5 air pollution. Although a large one-time investment is required for increasing farm size, there would be substantial progress towards achieving Sustainable Development Goals, associated with food security, a clean environment and improved human health.

The paper presents the phenomenon of groundwater contamination in the mine area during the failure of the coupled grouting composite. Indoor immersion experiments were conducted on standard grouting composite specimens to understand its failure laws. The experimental results showed that with the increase of the experimental time, the mass of the grouting composite decreased linearly. When the experiment was carried out for 180 days, the mass loss of the grouting composite was 19.834g, and the permeability coefficient gradually increased, which all verified the fact that the grouting composite gradually fails. The main groundwater contamination factors and contamination patterns in the study area were analyzed based on the hydrogeological data of the area. A groundwater contaminant transport model was established for the failure process of the coupled grouting composite, and the contamination potential of the 3222-workface goaf water in the shallow aquifer during the failure of the grouting composite was evaluated. The simulation results of groundwater pollutant transport showed that within 9 years after the grouting engineering was completed, the grouting composite could still maintain a certain degree of impermeability, and no characteristic pollutant ions appeared in the fourth aquifer. When the model ran for 10 years, ferrous ions appeared in the fourth aquifer, but the concentration was only 0.0009 mg/L. After 30 years of model operation, the highest concentration of ferrous ions in the fourth aquifer had reached 0.1609 mg/L. It can be seen that with the passage of time, the strength of the grouting composite in the study area is indeed gradually decreasing. After the strength decreases to a certain extent, it loses its original impermeability and becomes a channel for pollutant transport, with the potential to pollute the shallow aquifer.

Beneficial microorganisms can protect crop from phytopathogens, and modify rhizosphere microbiome. However, it is not well understood whether or how do rhizosphere microorganisms which respond to bio-agents contribute to disease suppression. Bacillus velezensis BER1 and tomato bacterial wilt caused by Ralstonia solanacearum were selected as models to disentangle the interactions and mechanisms in the rhizosphere. B. velezensis BER1 greatly suppressed tomato bacterial wilt by over 49.0%, reduced R. solanacearum colonization in the rhizosphere by 36.3%, and significantly enriched two Flavobacterium ASVs (1357 and 2401). A novel colony Loop-Mediated Isothermal Amplification (LAMP) assay system was developed to screen out Flavobacterium from tomato rhizosphere bacterial isolates. In vitro tests revealed that co-cultivating BER1 with Flavobacterium C45 increased biofilm formation by 18.6%. Climate chamber experiment further revealed that Flavobacterium C45 improved the control efficiency of BER1 on tomato bacterial wilt by 46.0%, decreased the colonization of R. solanacearum in the rhizosphere by 43.1% and elevated the transcription of plant defense gene PR1α in tomato by 45.4%. In summary, Flavobacterium C45 boosted the ability of B. velezensis BER1 to prevent bacterial wilt and the colonization of R. solanacearum, highlighting the importance of helper bacteria on elevating the efficiency of biological control.

Background: Noise-induced hearing impairment can mediate delayed injury of spiral neurons (SGNs), resulting in degeneration of nerve fibers, synaptic degeneration and even death of SGNs. We believe that delayed injury is related to mitochondrial energy metabolism disorders. Results: We investigated ATP and the electron transport chain (ETC) in rat SGNs after noise injury and found that with prolonged injury time, ATP synthesis and the expression of complexes II and IV decreased, indicating the functional decline of the ETC. The maintenance of ETC function is related to subunit import and assembly of the complex. The disulfide relay mechanism controlled by the apoptosis inducing factor/coiled-coil-helix-coiled-coil-helix domain-containing protein 4（AIF/CHCHD4) pathway can regulate mitochondrial protein import. The results showed that AIF expression in SGNs decreased after noise exposure, indicating that noise damage to SGNs can restoreintramitochondrial protein input by downregulating the AIF/CHCHD4 pathway, hinderingETC function. Conclusion: After noise injury, the mitochondrial function markers, ATP and complexes II/IV down-expressed , indicating insufficiency of ECT function leaded to delay injured SGNs. And the down-regulation of AIF/CHCHD4 function was a possible mechanism that hindered ECT function.

The DEtection TRansformer (DETR) algorithm has received considerable attention in the research community and is gradually emerging as a mainstream approach for object detection and other perception tasks. However, the current field lacks a unified and comprehensive benchmark specifically tailored for DETR-based models. To address this issue, we develop a unified, highly modular, and lightweight codebase called detrex, which supports a majority of the mainstream DETR-based instance recognition algorithms, covering various fundamental tasks, including object detection, segmentation, and pose estimation. We conduct extensive experiments under detrex and perform a comprehensive benchmark for DETR-based models. Moreover, we enhance the performance of detection transformers through the refinement of training hyper-parameters, providing strong baselines for supported algorithms.We hope that detrex could offer research communities a standardized and unified platform to evaluate and compare different DETR-based models while fostering a deeper understanding and driving advancements in DETR-based instance recognition. As a hypothetical topological defect in the geometry of spacetime, vortex strings play a crucial role in shaping the clusters of galaxies that exist today, and their distinct features can provide observable clues about the early universe's evolution. A key feature of vortex strings is that they can interact with Weyl fermionic modes and support topological chiral-anomaly states with massless dispersions at the core of strings. To date, despite many attempts to detect vortex strings in astrophysics or to emulate them in artificially created systems, observation of these topological vortex-string chiral modes remains experimentally elusive. Here we report the experimental observation of such vortex-string chiral modes using a metamaterial system. This is implemented by inhomogeneous perturbation of a Yang-monopole phononic metamaterial. The measured linear dispersion and modal profiles confirm the existence of topological modes bound to and propagating along the vortex string with the chiral anomaly. Our work not only provides a platform for studying diverse cosmic topological defects in astrophysics but also offers intriguing device applications as topological fibres in signal processing and communication techniques., Comment: 3 Figures

Background More and more studies have shown that circular RNAs (circRNAs) play a critical regulatory role in many cancers. However, the potential molecular mechanism of circRNAs in prostate cancer (PCa) remains largely unknown. Methods Differentially expressed circRNAs were identified by RNA sequencing. The expression of hsa_circ_0003258 was evaluated using quantitative real-time PCR and RNA in situ hybridization. The impacts of hsa_circ_0003258 on the metastasis of PCa cells were investigated by a series of in vitro and in vivo assays. Lastly, the underlying mechanism of hsa_circ_0003258 was revealed by Western blot, biotin-labeled RNA pulldown, RNA immunoprecipitation, luciferase assays and rescue experiments. Results Increased expression of hsa_circ_0003258 was found in PCa tissues and was associated with advanced TNM stage and ISUP grade. Overexpression of hsa_circ_0003258 promoted PCa cell migration by inducing epithelial mesenchymal transformation (EMT) in vitro as well as tumor metastasis in vivo, while knockdown of hsa_circ_0003258 exerts the opposite effect. Mechanistically, hsa_circ_0003258 could elevate the expression of Rho GTPase activating protein 5 (ARHGAP5) via sponging miR-653-5p. In addition, hsa_circ_0003258 physically binds to insulin like growth factor 2 mRNA binding protein 3 (IGF2BP3) in the cytoplasm and enhanced HDAC4 mRNA stability, in which it activates ERK signalling pathway, then triggers EMT programming and finally accelerates the metastasis of PCa. Conclusions Upregulation of hsa_circ_0003258 drives tumor progression through both hsa_circ_0003258/miR-653-5p/ARHGAP5 axis and hsa_circ_0003258/IGF2BP3 /HDAC4 axis. Hsa_circ_0003258 may act as a promising biomarker for metastasis of PCa and an attractive target for PCa intervention.

The molecular structures of hydrocarbon molecules determine the competitive and sequence reactions in the diesel hydrocracking process. In this study, the hydrocracking reactions of typical hydrocarbons with various saturation degrees and molecular weights in diesel fractions synergistically catalyzed by the Ni-Mo-S nanocluster and Al-Si FAU zeolite are investigated. The results show that the two major rate-controlling steps in saturated hydrocarbon hydrocracking are dehydrogenation on the Ni-Mo-S active sites and the cracking of the C-C bonds on the FAU zeolite acid center. Moreover, the major rate-controlling step in cracking the cycloalkyl aromatic hydrocarbons is the protonation of the aromatic ring. Moreover, the aromatic hydrocarbons presented an apparent advantage in competitive adsorption on the Ni-Mo-S active sites, whereas hydrocarbons with higher molecular weights demonstrated a moderate adsorption advantage on both Ni-Mo-S active sites and FAU zeolite acid centers.

The ever-increasing demand for flexible and portable electronics has stimulated research and development in building advanced electrochemical energy devices which are lightweight, ultrathin, small in size, bendable, foldable, knittable, wearable, and/or stretchable. In such flexible and portable devices, semi-solid/solid electrolytes besides anodes and cathodes are the necessary components determining the energy/power performances. By serving as the ion transport channels, such semi-solid/solid electrolytes may be beneficial to resolving the issues of leakage, electrode corrosion, and metal electrode dendrite growth. In this paper, the fundamentals of semi-solid/solid electrolytes (e.g., chemical composition, ionic conductivity, electrochemical window, mechanical strength, thermal stability, and other attractive features), the electrode-electrolyte interfacial properties, and their relationships with the performance of various energy devices (e.g., supercapacitors, secondary ion batteries, metal-sulfur batteries, and metal-air batteries) are comprehensively reviewed in terms of materials synthesis and/or characterization, functional mechanisms, and device assembling for performance validation. The most recent advancements in improving the performance of electrochemical energy devices are summarized with focuses on analyzing the existing technical challenges (e.g., solid electrolyte interphase formation, metal electrode dendrite growth, polysulfide shuttle issue, electrolyte instability in half-open battery structure) and the strategies for overcoming these challenges through modification of semi-solid/solid electrolyte materials. Several possible directions for future research and development are proposed for going beyond existing technological bottlenecks and achieving desirable flexible and portable electrochemical energy devices to fulfill their practical applications. It is expected that this review may provide the readers with a comprehensive cross-technology understanding of the semi-solid/solid electrolytes for facilitating their current and future researches on the flexible and portable electrochemical energy devices.

Vision transformers have recently achieved competitive results across various vision tasks but still suffer from heavy computation costs when processing a large number of tokens. Many advanced approaches have been developed to reduce the total number of tokens in large-scale vision transformers, especially for image classification tasks. Typically, they select a small group of essential tokens according to their relevance with the class token, then fine-tune the weights of the vision transformer. Such fine-tuning is less practical for dense prediction due to the much heavier computation and GPU memory cost than image classification. In this paper, we focus on a more challenging problem, i.e., accelerating large-scale vision transformers for dense prediction without any additional re-training or fine-tuning. In response to the fact that high-resolution representations are necessary for dense prediction, we present two non-parametric operators, a token clustering layer to decrease the number of tokens and a token reconstruction layer to increase the number of tokens. The following steps are performed to achieve this: (i) we use the token clustering layer to cluster the neighboring tokens together, resulting in low-resolution representations that maintain the spatial structures; (ii) we apply the following transformer layers only to these low-resolution representations or clustered tokens; and (iii) we use the token reconstruction layer to re-create the high-resolution representations from the refined low-resolution representations. Laser processing is employed to fabricated zinc-ion battery (ZIB) anodes with state-of-the-art electrochemical performance from commercial zinc foils. Lasers are widely utilized for industrial surface finishing but have received minimal attention for zinc surface modification. Laser lithography patterned zinc foils “LLP@ZF” are hydrophilic, with an electrolyte contact angle of 0°. This is due to the concave-convex surface geometry that enhances wetting (periodic crests, ridges and valleys, roughness 16.5 times planar). During electrodeposition LLP@ZF's surface geometry generates a periodic electric field and associated current density distribution that suppresses tip growth (per continuum simulations). Per Density Functional Theory (DFT) its surface oxide is zincophilic, resulting in low nucleation barriers during plating (e.g. 3.8 mV at 1 mA cm−2). A combination of these attributes leads to stable dendrite-free plating/stripping behavior and low overpotentials at fast charge (e.g. 48.2 mV at 8 mA cm−2 in symmetric cell). Cycling is possible at an unprecedented areal capacity of 50 mA h cm−2, with 400 h stability at 1 mA cm−2. Moreover, exceptional aqueous zinc battery (AZB) performance is achieved, with MnO2-based cathode loading 10 mg cm−2 and corresponding anode capacity 7.6 mA h cm−2. A broad comparison with literature indicates that LLP@ZF symmetric cell and full battery performance are among most favorable.\ud \ud

Potassium–sulfur batteries (KSBs) are regarded as a promising large-scale energy storage technology, owing to the high theoretical specific capacity and intrinsically low cost. However, the commercialization of KSBs is hampered by the low sulfur utilization and notorious shuttle effect. Herein, we employ a porosity engineering strategy to design nitrogen-rich carbon foam as an efficient sulfur host. The tremendous micropores magnify the chemical interaction between sulfur species and the polar nitrogen functionalities decorated carbon surface, which significantly improve the sulfur utilization and conversion. Meanwhile, the abundant mesopores provide ample spaces, accommodating the large volume changes of sulfur upon reversible potassation. Resultantly, the constructed sulfur cathode delivers an ultrahigh initial reversible capacity of 1470 mAh g−1 (87.76% of theoretical capacity) and a superior rate capacity of 560 mAh g−1 at 2 C. Reaching the K2S phase in potassiation is the essential reason for obtaining the ultrahigh capacity. Nonetheless, systematic kinetics analyses demonstrate that the K2S involved depotassiation deteriorates the charge kinetics. The density functional theory (DFT) calculation revealed that the nitrogen-rich micropore surface facilitated the sulfur reduction for K2S but created a higher energy barrier for the K2S decomposition, which explained the discrepancy in kinetics modification effect produced by the porosity engineering.

Noise-induced hearing impairment can mediate delayed injury of spiral neurons (SGNs), resulting in degeneration of nerve fibers, synaptic degeneration and even death of SGNs. We believe that delayed injury is related to mitochondrial energy metabolism disorders. Therefore, we investigated ATP and the electron transport chain (ETC) in rat SGNs after noise injury and found that with prolonged injury time, ATP synthesis and the expression of complexes II and IV decreased, indicating the functional decline of the ETC. The maintenance of ETC function is related to subunit import and assembly of the complex. The disulfide relay mechanism controlled by the apoptosis inducing factor/coiled-coil-helix-coiled-coil-helix domain-containing protein 4(AIF/CHCHD4) pathway can regulate mitochondrial protein import. The results showed that AIF expression in SGNs decreased after noise exposure, indicating that noise damage to SGNs can restore intramitochondrial protein input by downregulating the AIF/CHCHD4 pathway, hindering ETC function; insufficient ETC function is a possible reason for the delayed injury of SGNs.

To compare the clinical application and health economic values of non-invasive prenatal testing (NIPT) and second trimester serum screening (STSS).A retrospective analysis was carried out on 54 026 singleton pregnant women undergoing NIPT and STSS from March 1, 2018 to December 31, 2019 in Changsha Maternal and Child Health Care Hospital. For pregnant women with high-risk results of NIPT, prenatal diagnosis and follow-up of pregnancy outcomes were conducted. The data was grouped to 4 screening models, and their cost-benefit was analyzed.The sensitivity, specificity and positive predictive value of NIPT were all higher than STSS. Screening models 1 to 4 have prevented the birth of 71, 29, 52 and 54 patients with Down syndrome, respectively. The safety index of screening models 1 to 4 were 0.0036, 0.3944, 02215 and 0.1281, respectively. When the price of NIPT was decreased to 600 RMB, the cost-benefit of the screening models 1 to 4 was 0.46, 0.65, 0.44 and 0.40 million RMB, respectively.NIPT has a better detection performance than STSS. When the price of NIPT is 600 RMB, screening model 1 has the best screening effect and the highest accuracy, safety index and health economical value.

One-to-one set matching is a key design for DETR to establish its end-to-end capability, so that object detection does not require a hand-crafted NMS (non-maximum suppression) to remove duplicate detections. This end-to-end signature is important for the versatility of DETR, and it has been generalized to broader vision tasks. However, we note that there are few queries assigned as positive samples and the one-to-one set matching significantly reduces the training efficacy of positive samples. We propose a simple yet effective method based on a hybrid matching scheme that combines the original one-to-one matching branch with an auxiliary one-to-many matching branch during training. Our hybrid strategy has been shown to significantly improve accuracy. In inference, only the original one-to-one match branch is used, thus maintaining the end-to-end merit and the same inference efficiency of DETR. The method is named H-DETR, and it shows that a wide range of representative DETR methods can be consistently improved across a wide range of visual tasks, including DeformableDETR, PETRv2, PETR, and TransTrack, among others. Y900 is one of the top hybrid rice varieties with a yield exceeding 15 t/hm2. To dissect the mechanism of heterosis, the male parent line R900 and female parent line Y58S were sequenced using long-read and Hi-C technology. High-quality reference genomes of sizes of 396.41 Mb and 398.24 Mb were obtained for R900 and Y58S, respectively. Genome-wide variations between the parents were systematically identified, including 1,367,758 SNPs and 299,149 Indels. No megabase level structural variations exist. >75% of genes exhibited variation between the two parents. Compared with other two-line hybrids sharing the same female parent, the Geng/japonica-type genetic components from different male parents showed an increasing trend from phase 2-4 super-hybrid rice; Transcriptome analysis revealed that additive and dominance effects are the main genetic effects that constitute the heterosis of Y900. Allele-specific expression patterns and expression regulation patterns are quite dynamic in different tissues. For young panicle tissues, cis-regulation is dominant, while trans-regulation is more popular in leaf issues. Overdominance is more likely regulated by the trans-regulation mechanism. The differential gene expression and regulation pattern are closely related to Geng/japonica introgression. Additionally, R900 contained several excellent japonica haplotypes, such as NAL1, OsSPL13, Ghd8, OsBRI1, and DTH2, which make a good complement to Y58S. The fine tune mechanism through dynamic expression or regulation pattern change, especially on some key functional genes, is the base for heterosis.

PurposeTo assess the value of novel deep learning (DL) scores combined with complementary lung imaging reporting and data system 1.1 (cLung-RADS 1.1) in managing the risk stratification of ground-glass nodules (GGNs) and therefore improving the efficiency of lung cancer (LC) screening in China.Materials and MethodsOverall, 506 patients with 561 GGNs on routine computed tomography images, obtained between January 2017 and March 2021, were enrolled in this single-center, retrospective Chinese study. Moreover, the cLung-RADS 1.1 was previously validated, and the DL algorithms were based on a multi-stage, three-dimensional DL-based convolutional neural network. Therefore, the DL-based cLung-RADS 1.1 model was created using a combination of the risk scores of DL and category of cLung-RADS 1.1. The recall rate, precision, accuracy, per-class F1 score, weighted average F1 score (F1weighted), Matthews correlation coefficient (MCC), and area under the curve (AUC) were used to evaluate the performance of DL-based cLung-RADS 1.1.ResultsThe percentage of neoplastic lesions appeared as GGNs in our study was 95.72% (537/561) after long-period follow-up.Compared to cLung-RADS 1.1 model or DL model, The DL-based cLung-RADS 1.1 model achieved the excellent performance with F1 scores of 95.96% and 95.58%, F1weighted values of 97.49 and 96.62%, accuracies of 92.38 and 91.77%, and MCCs of 32.43 and 37.15% in the training and validation tests, respectively. The combined model achieved the best AUCs of 0.753 (0.526–0.980) and 0.734 (0.585–0.884) for the training and validation tests, respectively.ConclusionThe DL-based cLung-RADS 1.1 model shows the best performance in risk stratification management of GGNs, which demonstrates substantial promise for developing a more effective personalized lung neoplasm management paradigm for LC screening in China.
