Las áreas naturales protegidas buscan la conservación de la biodiversidad. El objetivo del estudio fue describir la diversidad de especies arbóreas y su importancia ecológica en el Parque Ecológico El Tecuán en Durango, México. Se realizó un muestreo sistemático estratificado con 168 sitios circulares de 1,000 m² cada uno, distribuidos en 511.16 hectáreas del Parque, representando una intensidad de muestreo del 3.2%. En cada sitio se midieron todos los individuos con diámetro normal mayor o igual a 7.5 cm. La diversidad de especies arbórea se estimó con el índice de Shannon, mientras que la importancia ecológica de las especies se describió con los parámetros abundancia relativa basada en la densidad o número de árboles por hectárea, dominancia relativa de acuerdo con el área basal, frecuencia relativa y el valor de importancia ecológica (VIE). Los resultados mostraron la presencia de cuatro géneros: Arbutus (Ericaceae), Juniperus (Cupressaceae), Pinus (Pinacea) y Quercus (Fagacea). El índice de Shannon fue de 1.89 sugiriendo una diversidad arbórea de media a alta. Las existencias reales registradas fueron de 153.73 m3 ha-1 y las clases de diámetro y de altura dominantes fueron de 10 cm y 5 m, respectivamente. El género Pinus tuvo mayor riqueza especies (nueve); de las cuales P. cooperi presentó los valores de densidad (32.15%), dominancia (37.77%) y de valor de importancia ecológica más elevados (28.23%), sin embargo, Quercus sideroxyla registró la mayor frecuencia relativa (18%). Los resultados obtenidos permitirán desarrollar acciones de sustentabilidad para la conservación de la diversidad arbórea del área de estudio.

Las distribuciones diamétricas son un factor importante en la caracterización del rodal, ya que el diámetro, generalmente, está correlacionado con otras variables de interés como la altura, volumen, biomasa, etcétera, esto permite conocer el tipo de productos que pueden obtenerse del bosque. El objetivo de la presente investigación fue desarrollar una estrategia para ajustar las FDP Weibull, Beta y SB Johnson, así como reconstruir (modelación) la distribución diamétrica futura con el método de recuperación de parámetros. En una primera etapa, se estudió comparativamente la calidad de ajuste de tres funciones de distribución de probabilidad (FDP: Weibull, SB Johnson y Beta) mediante los métodos de momentos y máxima verosimilitud para recuperar los parámetros y describir la distribución diamétrica de 2 252 parcelas temporales de muestreo, ubicadas en bosques naturales de Pueblo Nuevo, Durango, México. En general, los mejores resultados, en términos de precisión y parsimonia en la etapa de ajuste, evaluados mediante el sesgo medio y la raíz del error medio cuadrático se obtuvieron con la FDP Weibull ajustada con el método de los momentos; mientras que, las distribuciones SB Johnson y Beta se ubicaron en segundo y tercer lugar, respectivamente. Por tanto, la FDP Weibull biparamétrica fue seleccionada para modelar las distribuciones diamétricas de los rodales estudiados. La técnica de la recuperación de parámetros evidenció que 62 % de las parcelas modeladas siguen una distribución teórica tipo Weibull con 20 % de nivel de significancia en la prueba de Kolmogorov-Smirnov.

Background Deficiency of antithrombin increases risk of venous thromboembolism. We hypothesized that antithrombin deficiency affects fibrin clot structure and function. Methods We evaluated 148 patients (age: 38 [32–50] years; 70% women) with genetically confirmed antithrombin deficiency and 50 healthy controls. Fibrin clot permeability (Ks) and clot lysis time (CLT) along with thrombin generation capacity were assessed before and after antithrombin activity normalization in vitro. Results Antithrombin-deficient patients had lower antithrombin activity (−39%) and antigen levels (−23%) compared with controls (both p Conclusions Our study suggests that enhanced thrombin generation and prothrombotic plasma fibrin clot phenotype can contribute to increased risk of thrombosis in patients with antithrombin deficiency.

Multiplex ligation-dependent probe amplification (MLPA) identifies genetic structural variants in SERPINC1 in 5% of cases with antithrombin deficiency (ATD), the most severe congenital thrombophilia. Our aim was to unravel the utility and limitations of MLPA in a large cohort of unrelated patients with ATD (N = 341). MLPA identified 22 structural variants (SVs) causing ATD (6.5%). MLPA did not detect SVs affecting introns (four cases), and the diagnosis was inaccurate in two cases according to long-range PCR or nanopore sequencing. MLPA was used to detect possible hidden SVs in 61 cases with type I deficiency with single nucleotide variations (SNVs) or small insertion/deletion (INDEL). One case had a false deletion of exon 7, as the 29-bp deletion affected an MLPA probe. We evaluated 32 variants affecting MLPA probes: 27 SNVs and 5 small INDELs. In three cases, MLPA gave false-positive results, all diagnosed as deletions of the affected exon: a small INDEL complex, and two SNVs affecting MLPA probes. Our study confirms the utility of MLPA to detect SVs in ATD, but also shows some limitations in detecting intronic SVs. MLPA renders imprecise and false-positive results for genetic defects which affect MLPA probes. Our results encourage the validation of MLPA results.

The pace of population ageing is increasing and is currently becoming one of the challenges our society faces. The introduction of Cyber-Physical Systems (CPS) has fostered the development of e-Health solutions that ease the associated economic and social burden. In this work, a CPS-based solution is presented to partially tackle the problem: a Deep Multimodal Habit Tracking system. The aim is to monitor daily life activities to alert in case of life-threatening situations improving their autonomy and supporting healthy lifestyles while living alone at home. Our approach combines video and heart rate cues to accurately identify indoor actions, running the processing locally in embedded edge nodes. Local processing provides inherent protection of data privacy since no image or vital signs are transmitted to the network, and reduces data bandwidth usage. Our solution achieves an accuracy of more than 80% in average, reaching up to a 95% for specific subjects after adapting the system. Adding heart-rate information improves F1-score by 2.4%. Additionally, the precision and recall for critical actions such as falls reaches up to 93.75%. Critical action detection is crucial due to their dramatic consequences, it helps to reduce false alarms, leading to building trust in the system and reducing economic cost. Also, the model is optimized and integrated in a Nvidia Jetson Nano embedded device, reaching real-time performance below 3.75 Watts. Finally, a dataset specifically designed for indoor action recognition using synchronized video and heart rate pulses has been collected.

The study of above-ground biomass (AGB) is important for monitoring the dynamics of the carbon cycle in forest ecosystems. The emergence of remote sensing has made it possible to analyze vegetation using land surface temperature (LST), Vegetation Temperature Condition Index (VTCI) and evapotranspiration (ET) information. However, relatively few studies have evaluated the ability of these variables to estimate AGB in temperate forests. The aim of the present study was to evaluate the relationship of LST, VTCI and ET with AGB in temperate forests of Durango, Mexico, regarding each season of the year and to develop a AGB estimation model using as predictors LST, VCTI and ET, together with topographic, reflectance and Gray-Level Co-Occurrence Matrix (GLCM) texture variables. A semi-parametric model was generated to analyze the linear and non-linear responses of the predictive variables of AGB using a generalized linear model (GAM). The results show that the best predictors of AGB were longitude, latitude, spring LST, ET, elevation VTCI, NDVI (Normalized Difference Vegetation Index), slope and GLCM mean (R2 = 0.61; RMSE = 28.33 Mgha−1). The developed GAM model was evaluated with an independent dataset (R2 = 0.58; RMSE = 31.21 Mgha−1), suggesting the potential of this modeling approach to predict AGB for the analyzed temperate forest ecosystems.

Serine protease inhibitors (serpins) include thousands of structurally conserved proteins playing key roles in many organisms. Mutations affecting serpins may disturb their conformation, leading to inactive forms. Unfortunately, conformational consequences of mutations affecting serpins are difficult to predict. In this study we integrate experimental data of patients with mutations affecting one serpin with the predictions obtained by AlphaFold and molecular dynamics. FiveSERPINC1mutations causing antithrombin deficiency, the strongest congenital thrombophilia, (p.Arg79Cys; p.Pro112Ser, p.Met283Val; p.Pro352insValPheLeuPro, and p.Glu241_Leu242delinsValLeuValLeuValAsnThrArgThr-Ser) were selected from a cohort of 350 unrelated patients based on functional, biochemical, and crystallographic evidence supporting a folding defect. AlphaFold gave an accurate prediction for the wild-type antithrombin structure. However, it only produced native structures for all variants, regardless of its complexity andin-vivoconformational consequences. Similarly, molecular dynamics of up to 1000 ns at temperatures that caused conformational transitions did not show significant changes in the native structure of wild-type or variants. In conclusion, one predictive tool of protein folding, AlphaFold, and a simulation method for analyzing the physical movements of atoms and molecules, molecular dynamics, force predictions into the native stressed conformation at conditions with experimental evidence supporting a conformational change to relaxed structures. It is necessary to improve predictive strategies for serpins that consider the conformational sensitivity of these molecules.

Antithrombin deficiency, the most severe thrombophilia, might be underestimated, since it is only investigated in cases with consistent functional deficiency or family history. We have analyzed 444 consecutive, unrelated cases, from 1998 to 2021, with functional results supporting antithrombin deficiency in at least one sample. Plasma antithrombin was evaluated by functional and biochemical methods in at least two samples. SERPINC1 gene was analyzed by sequencing and MPLA. Hypoglycosylation was studied by electrophoresis and high-performance liquid chromatography (HPLC). In 260 of 305 cases (85.2%) with constitutive deficiency (activity < 80% in all samples), a SERPINC1 (N = 250), or N-glycosylation defect (N = 10) was observed, while 45 remained undetermined. The other 139 cases had normal antithrombin activity (≥ 80%) in at least one sample, what we called transient deficiency. Sixty-one of these cases (43.9%) had molecular defects: 48 had SERPINC1 variants, with two recurrent mutations (p.Ala416Ser[Cambridge II], N = 15; p.Val30Glu[Dublin], N = 12), and 13 hypoglycosylation. Thrombotic complications occurred in transient deficiency, but were less frequent, latter-onset, and had a higher proportion of arterial events than in constitutive deficiency. Two mechanisms explained transient deficiency: The limitation of functional methods to detect some variants and the influence of external factors on the pathogenic consequences of these mutations. Our study reveals a molecular defect in a significant proportion of cases with transient antithrombin deficiency, and changes the paradigm of thrombophilia, as the pathogenic effect of some mutations might depend on external factors and be present only at certain timepoints. Antithrombin deficiency is underestimated, and molecular screening might be appropriate in cases with fluctuating laboratory findings.

Hereditary antithrombin deficiency is caused by SERPINC1 gene mutations and predisposes to recurrent venous thromboembolism that can be life-threatening. Therefore, lifelong anticoagulation is required, which has side effects and may not be effective. In this study, peripheral blood mononuclear cells from a patient with severe antithrombin deficiency were reprogrammed into induced pluripotent stem cells (iPSCs). The mutation was corrected using CRISPR-Cas9 and Cre/LoxP genome editing. iPSCs were differentiated into hepatocytes, which were injected into the spleen of antithrombin knockout mice to restore the activity of antithrombin and reduce the thrombophilic state. Human iPSC-differentiated hepatocytes colonized mice and secreted antithrombin stably, normalizing antithrombin in plasma (activity: from 46.8 ± 5.7% to 88.6 ± 7.6%, P P P = 0.0025) and showed no notable difference compared with that in wild-type mice (10.41 ± 2.91 milligrams). The activity and concentration of antithrombin remained high for 3 weeks after injection. The liver and kidney function markers showed no obvious abnormality during the observation period. This study provides a proof of principle for correction of mutations in patient-derived iPSCs and potential therapeutic applications for hereditary thrombophilia.

Whole-genome sequencing using nanopore technologies can uncover structural variants, which are DNA rearrangements larger than 50 base pairs. Nanopore technologies can also characterize their boundaries with single-base accuracy, owing to the kilobase-long reads that encompass either full variants or their junctions. Other methods, such as next-generation short read sequencing or PCR assays, are limited in their capabilities to detect or characterize structural variants. However, the existing software for nanopore sequencing data analysis still reports incomplete variant sets, which also contain erroneous calls, a considerable obstacle for the molecular diagnosis or accurate genotyping of populations.We compared multiple factors affecting variant calling, such as reference genome version, aligner (minimap2, NGMLR, and lra) choice, and variant caller combinations (Sniffles, CuteSV, SVIM, and NanoVar), to find the optimal group of tools for calling large (50 kb) deletions and duplications, using data from seven patients exhibiting gross gene defects on SERPINC1 and from a reference variant set as the control. The goal was to obtain the most complete, yet reasonably specific group of large variants using a single cell of PromethION sequencing, which yielded lower depth coverage than short-read sequencing. We also used a custom method for the statistical analysis of the coverage value to refine the resulting datasets.We found that for large deletions and duplications (50 kb), the existing software performed worse than for smaller ones, in terms of both sensitivity and specificity, and newer tools had not improved this. Our novel software, disCoverage, could polish variant callers' results, improving specificity by up to 62% and sensitivity by 15%, the latter requiring other data or samples.We analyzed the current situation of50-kb copy number variants with nanopore sequencing, which could be improved. The methods presented in this work could help to identify the known deletions and duplications in a set of patients, while also helping to filter out erroneous calls for these variants, which might aid the efforts to characterize a not-yet well-known fraction of genetic variability in the human genome.

Even though the site index is a popular method for describing forest productivity, its use is limited in uneven-aged multispecies forests. Accordingly, the site form (SF) is an alternative measure of productivity to the site index based on the tree height–diameter relationship. Our study aims to evaluate SF as a measure of productivity in the temperate uneven-aged multispecies forests of Durango, Mexico, applying three methods to estimate SF: (i) as the mean height of dominant trees at a reference diameter (SFH-D); (ii) as the expected mean height of dominant trees at a reference mean diameter (SFMH-MD), and (iii) as the expected height at a reference diameter for a given site (SFh-dbh). We assess the effectiveness of the SF based on two hypotheses: (i) the SF correlates to the total volume production, and (ii) the SF is independent of stand density. The SFH-D and the SFh-dbh showed a high correlation with productivity. However, they also did so with density. Contrary to this, the SFMH-MD had a weak correlation with density and productivity. We conclude that the SF is a suitable approach to describe site quality. Nonetheless, its effectiveness as a site quality indicator may be affected according to the method used.

Atresia of inferior vena cava (IVC) is a rare congenital malformation associated with high risk of venous thrombosis that still has unknown etiology, although intrauterine IVC thrombosis has been suggested to be involved. The identification of IVC atresia in a case with early idiopathic venous thrombosis and antithrombin deficiency caused by the homozygous SERPINC1 c.391C > T variant (p.Leu131Phe; antithrombin Budapest 3) encouraged us to evaluate the role of this severe thrombophilia in this vascular abnormality. We have done a cross-sectional study in previously identified cohorts of patients homozygous for the Budapest 3 variant (N = 61) selected from 1118 patients with congenital antithrombin deficiency identified in two different populations: Spain (N = 692) and Hungary (N = 426). Image analysis included computed tomography and phlebography. Atresia of the IVC system was observed in 17/24 cases (70.8%, 95% confidence interval [CI]: 48.9%-87.3%) homozygous for antithrombin Budapest 3 with available computed tomography (5/8 and 12/16 in the Spanish and Hungarian cohorts, respectively), 16 had an absence of infrarenal IVC and one had atresia of the left common iliac vein. All cases with vascular defects had compensatory mechanisms, azygos-hemiazygos continuation or double IVC, and seven also had other congenital anomalies. Short tandem repeat analysis supported the specific association of the IVC system atresia with SERPINC1. We show the first evidence of the association of a severe thrombophilia with IVC system atresia, supporting the possibility that a thrombosis in the developing fetal vessels is the reason for this anomaly. Our hypothesis-generating results encourage further studies to investigate severe thrombophilic states in patients with atresia of IVC.

The identification of inherited antithrombin deficiency (ATD) is critical to prevent potentially life-threatening thrombotic events. Causal variants in SERPINC1 are identified for up to 70% of cases, the majority being single-nucleotide variants and indels. The detection and characterization of structural variants (SVs) in ATD remain challenging due to the high number of repetitive elements in SERPINC1. Here, we performed long-read whole-genome sequencing on 10 familial and 9 singleton cases with type I ATD proven by functional and antigen assays, who were selected from a cohort of 340 patients with this rare disorder because genetic analyses were either negative, ambiguous, or not fully characterized. We developed an analysis workflow to identify disease-associated SVs. This approach resolved, independently of its size or type, all eight SVs detected by multiple ligation-dependent probe amplification, and identified for the first time a complex rearrangement previously misclassified as a deletion. Remarkably, we identified the mechanism explaining ATD in 2 out of 11 cases with previous unknown defect: the insertion of a novel 2.4 kb SINE-VNTR-Alu retroelement, which was characterized by de novo assembly and verified by specific polymerase chain reaction amplification and sequencing in the probands and affected relatives. The nucleotide-level resolution achieved for all SVs allowed breakpoint analysis, which revealed repetitive elements and microhomologies supporting a common replication-based mechanism for all the SVs. Our study underscores the utility of long-read sequencing technology as a complementary method to identify, characterize, and unveil the molecular mechanism of disease-causing SVs involved in ATD, and enlarges the catalogue of genetic disorders caused by retrotransposon insertions.

The remote sensing of fire severity and burned area is fundamental in the evaluation of fire impacts. The current study aimed to: (i) compare Sentinel-2 (S2) spectral indices to predict field-observed fire severity in Durango, Mexico; (ii) evaluate the effect of the compositing period (1 or 3 months), techniques (average or minimum), and phenological correction (constant offset, c, against a novel relative phenological correction, rc) on fire severity mapping, and (iii) determine fire perimeter accuracy. The Relative Burn Ratio (RBR), using S2 bands 8a and 12, provided the best correspondence with field-based fire severity (FBS). Tree aboveground biomass (e.g., bole, branches, and foliage), M, plays key roles in forest management as it is the basis for evaluating the sink and flux of, for example, carbon and nitrogen, stand productivity, dendro-energy, litter & root biomass, hydrological parameters, among others. With the aim of further simplifying and understanding M, the central objective of this research was to review available techniques to develop, test, and validate two independent novel non-destructive, semi-empirical models using four major M datasets: (i) the shape dimensional bio-physical, MSD; and (ii) the restrictive mathematical, MNR, models. The proposed models advance and test how each of both approaches: (i) constant or (ii) variable scalar coefficients perform when predicting M with major assumptions bearing bio-physical principles. The aim of this study was to review and to assess the quality of the scientific articles regarding early and late anastomotic leak (AL) after colorectal surgery and their risk factors. An electronic systematic search for articles on Colorectal Surgery, AL and its timing was undertaken using the MEDLINE database via PubMed, Cochrane and Embase. The selected articles were thoroughly reviewed and assessed for methodological quality using a validated methodology quality score (MINCIR score). This review was registered in the PROSPERO registry under ID: CRD42022303012. 9 articles were finally reviewed in relation to the topic of early and late anastomotic leak. There is a lack of consensus regarding the exact cut-off in time to define early and late anastomotic leak, but it is clear that they are two differentiated entities. The first, occurring in relation to technical factors; whereas the latter, is related to impaired healing.

Hereditary angioedema due to pathogenic FXII variants (HAE-FXII) is a rare dominant disease caused by increased activation of the plasma contact system. The most prevalent HAE-FXII variant, c.1032C > A p.Thr309Lys (FXII309Lys), results in a smaller FXII protein with increased sensitivity to fluid-phase activation by poorly understood mechanisms. We aimed to investigate the functionality of the FXII309Lys variant in 33 HAE-FXII patients, 25 healthy controls and 46 patients with congenital disorders of glycosylation (CDG). Activation of the plasma contact system was assessed by western blot and amidolytic assay in basal conditions or after treatment with either artificial or physiological activators. Recombinant wild-type and FXII309Lys variants were expressed in S2 insect (Drosophila) cells. Amidolytic and fibrin generation assays were performed in fresh plasma samples. FXII309Lys samples exhibited an increased electrophoretic mobility comparable with N-glycan-deficient FXII from CDG patients and asialo-FXII generated by neuraminidase treatment. They presented increased sensitivity to activation by dextran sulphate and silica which resulted in the generation of an aberrant 37-kDa heavy chain. We did not observe increased susceptibility of FXII309Lys to proteolysis by exogenous or tPA-generated plasmin. However, both exogenous and endogenous thrombin cleaved the FXII309Lys variant, releasing a 37-kDa fragment and resulting in enhanced proteolytic activation on the fluid phase. This model supports a sequential proteolytic activation process involving thrombin priming of FXII309Lys, followed by kallikrein cleavage and generation of active βFXIIa. The present results and the observation that angioedema episodes in HAE-FXII patients occur predominantly during hypercoagulable situations suggest a key role for thrombin.

Introduction Wood is a natural resource used for construction and the manufacture of many products. This material is exposed to damage due to biotic and abiotic factors. An important biotic factor is wood-degrading fungi that generate large economic losses. The objectives of this study were to determine the effect of xylophagous fungi (Coniophora puteana and Trametes versicolor) on the natural durability of six timber species in southern Durango, Mexico, and to establish differences between fungal effects on each tree species. Materials and Methods Samples of Pinus durangensis, P. cooperi, P. strobiformis, Juniperus deppeana, Quercus sideroxyla, and Alnus acuminata were exposed to fungi for 4 months under laboratory conditions according to European Standard EN350-1. Samples of Fagus sylvatica were used as control. Durability was determined as the percentage of wood mass loss for each species. Welch ANOVA tests were performed to establish differences among tree species. Welch t-tests were used to prove loss mass differences between fungi for each tree species. Results The most resistant species to C. puteana were P. durangensis, J. deppeana, P. cooperi and P. strobiformis, showing mean mass losses lower than 8.08%. The most resistant species to T. versicolor were J. deppeana, P. strobiformis and P. durangensis (mean mass losses lower than 7.39%). Pinus strobiformis and Q. sideroxyla were more susceptible to C. puteana effect; in contrast, P. durangensis and P. cooperi showed more damage due to T. versicolor degradation. Conclusions Woods of P. durangensis, P. cooperi, P. strobiformis and Juniperus deppeana are well adapted to infection by these xylophagous fungi and are therefore highly recommended for commercial use in southern Durango, Mexico.

Recurrent problems have been observed for biomass measurement in tree sprouts, mainly dueto differences in patterns of biomass distribution and the bias generated by using models for mature trees.The objective of this research was to evaluate models to estimate aboveground biomass in sprouts from two-year-old stumps of Short Rotation Woody Crops (SRWC) used for energetic purposes. The study was conducted in Central Chile (Bío-Bío region) under a complete block design and four replications.Three plant species were included: Eucalyptus globulus, E. denticulata, and Acacia dealbata, established in three population densities (5,000, 10,000 and 15,000 trees per ha). Two groups of nonlinear models wereused on three independent variables: root collar diameter (squared mean of two dominant stems), height (mean of two dominant stems) and number of epicormic stems. In the allometric model, the total aboveground biomass and the aboveground biomass by component were explained as the product of the mean square of the root collar diameter, mean height of the two dominant new sprouts and total number of epicormic sprouts as the predictive variable. Values of coefficient of determination (R2) ranged from 0.78 to 0.95 and RMSE ranged from 168 to 913 g. The species of E. globulus showed higher RMSE for biomass estimation in the all components except the branch component. The inclusion of dummy variables to identify the differences in each parameter of the models explained the seasonalityof the biomass accumulation in tree sprouts at 10, 21, and 31 months of age, improving the model goodness of fit RMSE by 27%. Accurate prediction of sprouts aboveground biomass was obtained by the models only evaluating the two dominant epicormic sprouts in the stump, avoiding the need of individual sprouts evaluation or using destructive methods for biomass measurement.

Se realizó un análisis para evaluar el efecto, en un periodo de 10 años, de la aplicación del Método Silvícola de Selección sobre la diversidad y estructura en un bosque templado de la Sierra de Durango, México. Se compararon doce Sitios Permanentes de Investigación Forestal y de Suelos (SPIFyS), los cuales fueron remedidos. El análisis comparativo se hizo mediante el índice de Diversidad de Shannon-Wiener; asimismo, se estimó el Índice de Valor de Importancia (IVI) mediante la densidad, el área basal y la frecuencia de las especies en los sitios de muestreo; también se calcularon tres índices de estructura para conocer la mezcla de especies, distribución espacial y dominancia del arbolado. Los índices de estructura tampoco se modificaron significativamente en el intervalo de 10 años después de la extracción forestal, por lo que se determina que el tratamiento silvícola aplicado no modifica la diversidad ni los componentes de estructura del estrato arbóreo.

This dataset contains the reports of the series12 local and a pan-European onlineco-creation workshops, organised in the frame of the agroBRIDGES H2020 project, which aims to better connect producers and consumers through Short Food Supply Chains (SFSCs). The local co-creation workshops were organised during the period of August and September 2021 in 12 focal regions and countries of the projects (Beacon Regions), involving 150+agrifood stakeholders of the project's local communities(agroBRIDGES Multi Actor Platform members). The workshops were organised by the project partners responsible for the stakeholder engagement and research activities of the project in each focal region (Beacon Region Leaders). 3 core topics were discussed in local workshops for co-creation: Ranking and evaluation of good practices and success cases of SFSCs in the regions Identification of barriers impeding the development and growth of regional SFSCs along with proposed solutions Development and improvement of sustainable business models for SFSCs. Following the local co-creation workshops, an online pan-European workshop followed to discuss, compare and generate macro-trends from regional insights. Inherited antithrombin deficiency, the most severe form of thrombophilia, is predominantly caused by variants in SERPINC1. Few causal structural variants have been described, usually detected by multiplex ligation-dependent probe amplification or cytogenetic arrays, which only define the gain or loss and the approximate size and location. This study has done a complete dissection of the structural variants affecting SERPINC1 of 39 unrelated patients with antithrombin deficiency using multiplex ligation-dependent probe amplification, comparative genome hybridization array, long-range PCR, and whole genome nanopore sequencing. Structural variants, in all cases only affecting one allele, were deleterious and caused a severe type I deficiency. Most defects were deletions affecting exons of SERPINC1 (82.1%), but the whole cohort was heterogeneous, as tandem duplications, deletion of introns, or retrotransposon insertions were also detected. Their size was also variable, ranging from 193 bp to 8 Mb, and in 54% of the cases involved neighboring genes. All but two structural variants had repetitive elements and/or microhomologies in their breakpoints, suggesting a common mechanism of formation. This study also suggested regions recurrently involved in structural variants causing antithrombin deficiency and found three structural variants with a founder effect: the insertion of a retrotransposon, duplication of exon 6, and a 20-gene deletion. Las variables geográficas determinan en gran medida la estructura y diversidad de especies. El objetivo del estudio fue conocer si existen diferencias entre los componentes que conforman la estructura, diversidad y composición florística con relación a la exposición y altitud en bosques templados del estado de Durango. Los datos dasométricos se obtuvieron de 30 sitios permanentes (2 500 m2), se consideraron: diámetro normal (Dn)>7.5 cm (1.30 m), altura total, y el registro por especie. Se definieron seis áreas distribuidas en tres intervalos altitudinales (500 m) con exposición sur y norte. Se caracterizó la estructura por medio del Índice de Valor de Importancia (IVI), diversidad alfa (α), riqueza de especies (S) y el Índice de Diversidad Verdadera de Shannon (1D), así como la diversidad beta (β) por medio del análisis de similitud de Bray-Curtis. Se realizó una prueba estadística de ANOVA de dos factores para determinar diferencias significativas entre intervalos de altitud en las áreas; las de mayor altitud tuvieron más densidad y área basal en ambas exposiciones, con excepción del área 5 (1 500 a 1 800 m sur) que mostró valores similares en área basal. Los géneros Pinus y Quercus presentaron un IVI superior en todas las áreas. Los sitios en exposición sur registraron una mayor riqueza de especies, sobre todo los que se ubicaron en el intervalo de menor altitud. La similitud entre áreas comprendió las zonas con más altitud y exposición norte, y se aislaron las tierras bajas con intervalos altitudinales diferentes.

BackgroundAortic valve replacement is the gold standard treatment for severe symptomatic aortic stenosis, but thrombosis of bioprosthetic valves (PVT) remains a concern.ObjectiveTo analyze the factors involved in the contact pathway during aortic valve replacement and to assess their impact on the development of thromboembolic complications.MethodsThe study was conducted in 232 consecutive patients who underwent: transcatheter aortic valve replacement (TAVR, N = 155), and surgical valve replacement (SAVR, N = 77) (MUVITAVI project). Demographic and clinical data, outcomes including a combined end point (CEP) of thrombotic events, and imaging controls were recruited. Samples were collected 24 h before and 48 h after valve replacement. FXII, FXI and (pre)kallikrein were evaluated by Western Blot and specific ELISA with nanobodies.ResultsThe CEP of thrombotic events was reached by 19 patients: 13 patients presented systemic embolic events and 6 patients subclinical PVT. Valve replacement did not cause FXII activation or generation of kallikrein. There was a significant reduction of FXI levels associated with the procedure, which was statistically more pronounced in SAVR than in TAVR. Cases with reductions of FXI below 80% of basal values had a lower incidence of embolic events during the procedure than patients in whom FXI increased above 150%: 2.7 vs. 16.7%; p: 0.04.ConclusionTAVR or SAVR did not significantly activate the contact pathway. A significant reduction of FXI, was observed, particularly in SAVR, associated with lower incidence of thrombotic events. These results encourage evaluating the usefulness and safety of FXI-directed antithrombotic treatments in these patients.

El objetivo fue estimar el volumen forestal (m3 ha-1) mediante información obtenida del Sistema de Planeación Forestal (SiPlaFor) y datos espectrales de imágenes de satélite del sensor Landsat 8 OLI en el Ejido La Victoria Pueblo Nuevo, Durango, México. Se utilizó la técnica no paramétrica Random Forest para la estimación del volumen forestal. Los resultados mostraron que empleando un modelo con 400 árboles de decisión, la raíz cuadrada del cuadrado medio del error (RMSE) se mantiene estable. La variable predictiva de mayor importancia fue el Índice de Vegetación de Área Foliar Especifica (SLAVI), con una RMSE de 12.99%. El modelo de Random Forest presentó un coeficiente de determinación (R2) de 0.84 y un valor de RMSE de 28.8 m3 ha-1. La información espectral de un sensor de media resolución en combinación con datos de campo es una alternativa viable para estimar volumen forestal en bosques templados mixtos.

Over the last 40 years, the learning process in university studies has gradually shifted from a teacher-centered approach to a student-centered approach. Economic and Business studies are a suitable sector for active teaching-methodologies such as Problem-Based Learning (PBL) and it is becoming popular in business programmes. Nevertheless, they cause students stress. The aim of this work is to study the fears that PBL produces in students and the perception whether it is worthwhile to overcome them in view of their professional development. It is applied to a Faculty of Economics and Business in order to detect the main factors that influence the success of PBL. One of the conclusions is that the main cause that hinders PBL materialization is the difficulty that students find in making their opinions known in the classroom. So, it is necessary to offer teachers educational programs for learning emotional methods to face those fears to successfully develop PBL among students.

Thanks to high-performance computing (HPC), it is possible to solve all kinds of highly complex projects from multiple scientific disciplines that require computationally intensive tasks to be undertaken and which otherwise could not be addressed. Unfortunately, since the development of parallel codes requires highly specific knowledge, it can become a challenge for beginners and non-expert programmers, especially when it comes to making adequate and efficient use of the available computing resources. To this end, we developed a transcompiler for helping researchers and inexperienced users who do not have the necessary skills in the use of parallel programming, and aimed at improving the performance of their HPC routines and tasks. Current efforts are focused on an additional module for optimizing code fragments in order to reduce their running times. In order to achieve this, twenty-six software techniques were selected from the literature to be integrated into this new module, all of them aimed at improving execution times of HPC programs by directly writing efficient code. Their effectiveness is analyzed and discussed in the current manuscript through a complete set of tests designed and conducted to measure and evaluate benefits achieved when applying these techniques.

Splanchnic vein thromboses (SVT) are a rare condition that can be life-threatening. The most severe thrombophilia associated to SVT is antithrombin (AT) deficiency, usually caused by SERPINC1 mutations. Although transitory AT deficiencies and congenital disorders of the N-glycosylation pathways (CDG) have been recently reported as causes of AT deficiency, the current AT clinical screening still only includes anti-FXa activity. This study aims to 1) improve the detection of antithrombin deficiency in SVT and 2) characterize the features of antithrombin deficiency associated with SVT.The study was performed in 2 cohorts: 1) 89 SVT patients with different underlying etiologies but in whom AT deficiency had been ruled out by classical diagnostic methods; and 2) 271 unrelated patients with confirmed AT deficiency and venous thrombosis. Antithrombin was evaluated by functional (anti-FXa and anti-FIIa) and immunological methods (ELISA, crossed immunoelectrophoresis, western blot), and SERPINC1 sequencing was performed.In 4/89 patients (4.5%) additional alterations in AT were found (two had SERPINC1 mutations, one had a specific variant causing transient AT deficiency and one patient had CDG). In 11 of the 271 patients (4.1%) with AT deficiency and thrombosis, thrombosis was located at the splanchnic venous territory.AT deficiency may be underdiagnosed by current clinical screening techniques. Therefore, a comprehensive AT evaluation should be considered in cases of rethrombosis or doubtful interpretation of anti-FXa activity levels. A study by Elechiguerra, Corral-Lage and Maguregui in 2015 shows how a model of social and economic indicators can help provide more transparent information on the Not-For-Profit area. Based on this premise, the objective of this work is to corroborate, using Cronbach´s Alpha, the degree of reliability of the set of social indicators related to the area of communication and accountability, with the desire and eagerness to favour the processes of issuing clearer and more homogeneous information related to this area within the Not-For-Profit Sector. It identifies which are the dimensions that the Not-For-Profit Organizations (NFPO) themselves think they must do when it comes to communicate information and accountability in an effective and efficient way. To this end, a survey has been carried out in which the opinion of 122 Spanish NFPO is gathered. Subsequently, based on the correlation study, the data obtained were analysed making it clear that the indicators these organizations grant a higher degree of reliability in the area studied are those related to anti-corruption measures, stakeholders´ participation in the entity process, the auditors´ insights and their committees. And those linked to penalties-sanctions and complaints-claims are discarded as they do not add value to the indicators as a whole.

Radiation-induced dermatitis is a common complication of radiotherapy. In Spain, the manufactured products used to prevent and treat that condition consist of emollient creams (which are often ineffective) or corticosteroids (which can cause dermal atrophy and other adverse effects). However, we have found that topically applied melatonin, a product that to our knowledge is not commercially available in Spain, is safe and effective in the prevention and treatment of radiodermatitis and a viable substitute for corticosteroid therapy. In this report, we describe the use of compounded melatonin in Beeler cream (the formulation for which is provided) in a 46-year-old woman who underwent surgery and radiotherapy for early stage adenocarcinoma of the breast. The patient applied that cream as directed throughout the prescribed course of radiotherapy and continued its use for 3 weeks after she last underwent irradiation. She experienced no adverse effects from the use of the compounded formulation and tolerated that treatment well. Radiation dermatitis did not develop at the irradiated site until approximately 1 week after the final radiotherapy session, at which time mild erythema of the treated skin emerged. That condition, which was successfully treated by application of the melatonin-containing cream as previously prescribed, resolved after 3 weeks of its use. We suggest that when manufactured products designed to prevent or treat the adverse dermal effects of radiotherapy are unavailable, fail to be effective, or prove objectionable to patients, a melatonin-containing topical compound can provide a successful alternative.

In the United States, CRC is the third most common type of cancer and the second leading cause of cancer-related death. Although the incidence of CRC among the Hispanic population has been declining, recently, a dramatic increase in CRC incidents among HL younger than 50 years of age has been reported. The incidence of early-onset CRC is more significant in HL population (45%) than in non-Hispanic Whites (27%) and African-Americans (15%). The reason for these racial disparities and the biology of CRC in the HL are not well understood. We performed this study to understand the biology of the disease in HL patients. We analyzed formalin-fixed paraffin-embedded tumor tissue samples from 52 HL patients with mCRC. We compared the results with individual patient clinical histories and outcomes. We identified commonly altered genes in HL patients (APC, TP53, KRAS, GNAS, and NOTCH). Importantly, mutation frequencies in the APC gene were significantly higher among HL patients. The combination of mutations in the APC, NOTCH, and KRAS genes in the same tumors was associated with a higher risk of progression after first-line of chemotherapy and overall survival. Our data support the notion that the molecular drivers of CRC might be different in HL patients.

The latitudinal diversity gradient (LDG) is one of the most recognized global patterns of species richness exhibited across a wide range of taxa. Numerous hypotheses have been proposed in the past two centuries to explain LDG, but rigorous tests of the drivers of LDGs have been limited by a lack of high-quality global species richness data. Here we produce a high-resolution (0.025° × 0.025°) map of local tree species richness using a global forest inventory database with individual tree information and local biophysical characteristics from ~1.3 million sample plots. We then quantify drivers of local tree species richness patterns across latitudes. Generally, annual mean temperature was a dominant predictor of tree species richness, which is most consistent with the metabolic theory of biodiversity (MTB). However, MTB underestimated LDG in the tropics, where high species richness was also moderated by topographic, soil and anthropogenic factors operating at local scales. Given that local landscape variables operate synergistically with bioclimatic factors in shaping the global LDG pattern, we suggest that MTB be extended to account for co-limitation by subordinate drivers. Forest plantations and natural forests perform a relevant role in capturing CO2 and reducing greenhouse gas concentrations. The objective of this study was to compare the diameter increment, biomass and carbon accumulation in a plantation of Pinus durangensis and a naturally regenerated stand. The data were collected from 32 circular plots of 100 m2 (16 plots in the planted site and 16 in naturally regenerated area). At each plot, the diameter at the base (cm) and height (m) of all seedlings were measured using a Vernier and tape measure, and a seedling was destructively sampled collecting one cross-section at the base of the stump. The annual ring-width increment of each sampled seedling was recorded to obtain its diameter at the base and estimate annual aboveground biomass and carbon accumulation through allometric equations. The response variables were evaluated using mixed-effects ANOVA models. Results indicated that there were significant differences (P ≤ 0.05) on annual tree-ring width growth, biomass and carbon accumulation. The plantation seedlings showed significantly higher growth rates, biomass and carbon accumulation at most evaluated years. After 7 years of growth the lines of current annual increment (CAI) and mean annual increment (MAI) in basal diameter for both the plantation and the natural regeneration have not yet intersected. Both forest plantations and naturally regenerated stands of the studied tree species may be suitable alternatives to promote CO2 capture and increase timber production.

Introduction: Predictions of dominant or co-dominant height are an important element in planning forest management with timber production objectives. Objective: To develop dominant height growth and site index (SI) equations for Quercus sideroxyla Bonpl. Materials and methods: The height current annual increment model was fitted, and the age of the sections was estimated from a database of 29 stem-mapped plots. A taper analysis data of 37 trees was also used to reconstruct dominant height growth trajectories. Three equations based on algebraic difference approach (ADA; one anamorphic and two polymorphic) and an equation based on generalized algebraic difference approach (GADA) were used to simultaneously model the dominant height and SI. Results and discussion: The ADA polymorphic equations were statistically better than the anamorphic equation, according to the adjusted coefficient of determination, root mean square error, mean bias, Akaike’s information criterion and log-likelihood; however, the GADA equation was better than the ADA equations. The growth curves were biologically realistic and showed SI classes at a base age of 60 years. Conclusions: The developed equations can be used for decision making in forest management when Q. sideroxyla shows dominance over Pinus species and the main objective is timber production.

El presente estudio evaluó el efecto de tratamientos silvícolas en la diversidad y estructura de especies en ecosistemas forestales de clima templado en el Municipio de Pueblo Nuevo del Estado de Durango, México; se realizó con la finalidad de conocer si el aprovechamiento forestal modifica la diversidad, mezcla de especies, distribución espacial y diferenciación dimensional de los individuos en estos ecosistemas. La evaluación se realizó comparando 10 parcelas con historial de manejo, las cuales se midieron antes de la aplicación del tratamiento y cinco años después. Se compararon los índices de diversidad de Shannon, Simpson y Margalef, así como índices de estructura de mezcla de especies, distribución espacial y diferenciación dimensional. De acuerdo con los tratamientos silvícolas aplicados, los valores de los índices no presentan diferencias significativas en sus evaluaciones (p > 0.05), lo que indica que el aprovechamiento forestal no modifica la diversidad y estructura de especies del estrato arbóreo de esta comunidad vegetal.

Forest certification is a private, voluntary and market-driven instrument designed to promote responsible forest management. This paper focused on the FSC and the NOM NMX-AA-143-SCFI-2008 schemes used in Mexico for the certification of sustainable forest management. In this paper we used the analytical hierarchical process (AHP) to study the factors that determine the main impacts of forest certification in México. A panel of 30 experts was selected as decision-makers to find which principles, criteria and indicators are considered as the most relevant while implementing forest certification. For decision-makers, the environmental principle occupied the first place with 40.26% of importance in the implementation of forest certification, followed by the social principle, and the economic principle with 32.15% and 27.59% of importance, respectively. Regarding the criteria, forest management and production, biodiversity, and forest protection were considered to be the most relevant. Regarding the indicators, the results indicated that forest certification in Mexico can have a positive impact on the existence of educational institutions, community services such as water, energy, medical services and drainage, the quality of the forest management plans, investment in forest management, machinery and equipment, environmental services, recreation, tourism, research, development and community education, planning for the conservation of biological diversity, and planning for biodiversity conservation.

Background: Previously published retrospective series show a high prevalence of fecal incontinence (FI) in stroke patients. We aimed to analyze in a prospective series the current incidence of FI in acute stroke in functionally independent patients and its evolution over time and the patient characteristics associated with the appearance of FI in acute stroke.Methods: We included consecutive patients with acute stroke admitted in our stroke unit who fulfilled the following inclusion criteria: a first episode of stroke, aged >18 years, with no previous functional dependency [modified Rankin Scale (mRS) ≤ 2] and without previous known FI. FI was assessed by a multidisciplinary trained team using dedicated questionnaires at 72 ± 24 h (acute phase) and at 90 ± 15 days (chronic phase). Demographic, medical history, clinical and stroke features, mortality, and mRS at 7 days were collected.Results: Three hundred fifty-nine (48.3%) of 749 patients (mean age 65.9 ± 10, 64% male, 84.1% ischemic) fulfilled the inclusion criteria and were prospectively included during a 20-month period. FI was identified in 23 patients (6.4%) at 72 ± 24 h and in 7 (1.9%) at 90 days ± 15 days after stroke onset. FI was more frequent in hemorrhagic strokes (18 vs. 5%, p 0.007) and in more severe strokes [median National Institute of Health Stroke Scale (NIHSS) 18 (14–22) vs. 5 (3–13), p < 0.0001]. No differences were found regarding age, sex, vascular risk factors, or other comorbidities, or affected hemisphere. Patients with NIHSS ≥12 (AUC 0.81, 95% CI 0.71 to 0.89) had a 17-fold increase for the risk of FI (OR 16.9, IC 95% 4.7–60.1) adjusted for covariates.Conclusions: At present, the incidence of FI in acute stroke patients without previous functional dependency is lower than expected, with an association of a more severe and hemorrhagic stroke. Due to its impact on the quality of life, it is necessary to deepen the knowledge of the underlying mechanisms to address therapeutic strategies.

Tropical forests are some of the most biodiverse ecosystems in the world, yet their functioning is threatened by anthropogenic disturbances and climate change. Global actions to conserve tropical forests could be enhanced by having local knowledge on the forestsʼ functional diversity and functional redundancy as proxies for their capacity to respond to global environmental change. Here we create estimates of plant functional diversity and redundancy across the tropics by combining a dataset of 16 morphological, chemical and photosynthetic plant traits sampled from 2,461 individual trees from 74 sites distributed across four continents together with local climate data for the past half century. Our findings suggest a strong link between climate and functional diversity and redundancy with the three trait groups responding similarly across the tropics and climate gradient. We show that drier tropical forests are overall less functionally diverse than wetter forests and that functional redundancy declines with increasing soil water and vapour pressure deficits. Areas with high functional diversity and high functional redundancy tend to better maintain ecosystem functioning, such as aboveground biomass, after extreme weather events. Our predictions suggest that the lower functional diversity and lower functional redundancy of drier tropical forests, in comparison with wetter forests, may leave them more at risk of shifting towards alternative states in face of further declines in water availability across tropical regions.

El manejo forestal a través de la aplicación de tratamientos silvícolas adecuados permite lograr la persistencia, el rendimiento sostenido y la máxima producción de los bosques. El objetivo de este trabajo fue evaluar el incremento en volumen fustal de árboles de Pinus durangensis, a partir del análisis de los anillos de crecimiento mediante la técnica del análisis troncal en rodales sometidos a cuatro tratamientos silvícolas; los cuales fueron cortas de regeneración con árboles Padre (realizada en 2007), matarrasa, selección y de aclareo aplicados en el año 2010. Se estudiaron 16 árboles muestra de Pinus durangensis recolectados en sitios de 100 m2, distribuidos al azar por cada tratamiento. Para el análisis troncal, por individuo se obtuvo una rodaja a la base, otra a 1.3 m sobre el nivel del suelo y posteriormente cada metro hasta llegar a la punta. Se estimó el diámetro y la altura de los árboles cada dos anillos de crecimiento y se estimó el volumen, para finalmente conocer los incrementos (ICA e IMA) en intervalos de dos años. Los resultados de la prueba no paramétrica de Kruskal Wallis mostraron diferencias significativas (p < 0.05) en el ICA e IMA en la mayoría de los tratamientos silvícolas evaluados. La matarrasa resultó ser la práctica con los mayores valores de ICA e IMA para todas las edades estudiadas.

The bleeding phenotype of factor XI (FXI) deficiency is unpredictable. Bleeding is usually mild and mostly occurs after injury. Although FXI deficiency renders antithrombotic protection, some patients might eventually develop thrombosis or atrial fibrillation, requiring anticoagulant therapy. There is almost no evidence on the bleeding risk in this scenario. Our retrospective study of 269 white FXI-deficient subjects (1995-2021) identified 15 cases requiring anticoagulation. They harbored 8 different F11 variants, mainly in heterozygosis (1 case was homozygote), and had mild to moderate deficiency (FXI:C: 20% to 70%). Two subjects (13.3%) had bleeding history before anticoagulation. Atrial fibrillation was the main indication (12/15; 80%). Fourteen patients started therapy with vitamin K antagonists (VKA), but 4 subjects were on direct oral anticoagulants (DOACs) at the end of follow-up. Over >1000 months of anticoagulation, 2 mild bleeding episodes in 2 patients (13.3%, 95% confidence interval: 3.7% to 37.9%) were recorded. No major/fatal events were reported. “Pre-post” bleeding localization and severity did not change despite treatment. On VKA, drug dosing and management were also standard, unaltered by FXI deficiency. We provide the largest description of anticoagulant use in FXI deficiency, and the first cases receiving DOACs. Although further studies are needed, our observations suggest that moderate FXI deficiency does not interfere with anticoagulant management nor bleeding risk.
