Abstract

We have investigated the influence of dietary n-6/n-3 (ù-6/ù-3) polyunsaturated fatty acid-balance on the tendency to arterial thrombosis and the progress of atherosclerosis in apoE-/- LDLR-/- double knockout mouse. Homozygous apoE-/- LDLR-/- double knockout mouse (DKO mice, 129XC57BL/6J background) and male C57BL/6 mice aged 6 weeks were divided into four groups. Each group was fed a diet containing a different n-6/n-3 ratio (Group l: 0.29; Group 2: 1.43; Group 3: 5.00; Group 4: 8), prepared with high linolenic (LNA) flaxseed oil (n-3 rich) and high linoleic (LA) safflower oil (n-6 rich). There were no statistical differences in the gain in body weight between the four groups. After 16 weeks, plasma triglyceride and LDL levels in Group 1 were significantly lower than in the other groups. Conversely, HDL was the highest. After 8 and 16 weeks, the tendency to arterial thrombosis was assessed using a He-Ne laser-induced thrombosis model. The degree of atherosclerosis was measured using the entire aorta method employing image analysis software. The n-6/n-3 ratio had a dose-dependent antithrombotic effect (thrombus volume decreased 23%, Group 1 vs. Group 4), In addition, the extent of atherosclerosis was less in the animals fed a low n-6/n-3 ratio compared with the high n-6/n-3 ratio group (atherosclerotic area decreased 40%, Group 1 vs. Group 4). The lowest n-6/n-3 ratio tested (0.29) was the most effective in suppressing the thrombotic and atherosclerotic parameters in these DKO mice.

Abstract

For many years the vascular endothelium was believed simply to provide a passive lining between circulating blood and extravascular tissue. It is now clear, however, that this monolayer of cells on the luminal surface of all blood vessels, provides a selective barrier that responds dynamically to various stimuli, and controls a complex series of cellular reactions and interactions. The current presentation describes the use of computer enhanced video recording to study interactions between endothelial cells and circulating blood cells, especially leucocytes. Subsequently, modern assays for soluble cell adhesion molecules and other cell receptors were assessed for potential use in routine clinical practice. The results demonstrated that adhesive mechanisms involving leucocytes and endothelial cells involve a range of interrelationships that cut across conventional views of haemostasis and leucocyte function. The findings also suggest that interplay between the vascular lumen and circulating blood cells might be vitally important in clinically demanding pathologies, such as life-threatening sepsis, ischaemic heart disease, atherosclerosis and cancer. The concepts provide challenging strategies for further investigation.

Abstract

Red blood cells (RBCs) from human, equine, canine, feline, and bovine whole blood samples have been separated and characterized by high-speed flow/hyperlayer field-flow fractionation (Fl/HyFFF). The elution-based separation of RBCs by this method is based mainly on the size and shape of the cell particles. The typical separation time for RBCs is less than 3 min. Size distributions can be derived from the fractograms of cell samples using a calibration plot based on retention data for uniform polystyrene beads. The method is shown to be effective both to separate and to characterize cell populations, including those with cells of abnormal shape and size. In order to investigate differences in cell morphology, shape and size changes induced by 500,000 Da Dextran were monitored. The changes in the Fl/HyFFF elution profiles indicate that the RBCs decrease in size but become partially aggregated in the presence of the dextran. These changes were found to depend on polymer concentration and specific blood samples. Some of the results from Fl/HyFFF were compared with those from the Coulter counter and from microscopy.

Abstract

Photon proton cross sections for elastic light vector meson production, σel νp, inelastic diffractive production, σnd νp, non-diffractive production, σd νp, as well as the total cross section, σtot νp, have been measured at an average υp center of mass energy of 180 GeV with the ZEUS detector at HERA. The resulting values are σel νp = 18 ± 7 μb, σd νp = 33 ± 8 μb, σnd νp = 91 ± 11 μb, and σtot νp 143 ± 17 μb, where the errors include statistical and systematic errors added in quadrature. © 1994 Springer-Verlag.

Abstract

This paper presents our first measurement of the F 2 structure function in neutral-current, deep inelastic scattering using the ZEUS detector at HERA, the ep colliding beam facility at DESY. The data correspond to an integrated luminosity of 24.7 nb −1 . Results are presented for data in range of Q 2 from 10 GeV 2 to 4700 GeV 2 and Bjorken x down to 3.0 × 10 −4 . The F 2 structure function increases rapidly as x decreases.

Abstract

This paper reports a search for excited electrons at the HERA electron-proton collider. In a sample corresponding to an integrated luminosity of 26 nb−, no evidence was found for any resonant state decaying into e−γ, νW− or e−Z0. Limits on the coupling strength of an excited electron have been determined for masses between 45 and 225 GeV. This study also reports the observation of the wide-angle eγ Compton scattering process.

Abstract

The work done on this project is divided into two principal areas. The first involves the application of sedimentation/steric FFF to metaphase chromosomes in an attempt to fractionate the chromosomes according to their size. The preparation of chromosomes from a number of organisms was attempted; procedures were finally worked out in collaboration with Los Alamos National Laboratory for the preparation of metaphase chromosomes from Chinese hamster cells. After extensive experimental work was done to identify suitable operating conditions, the partial fractionation of the Chinese hamster chromosomes was achieved. In the second component of the project, flow FFF was applied to the separation of DNA fragments. Figures are provided that show considerable success in the separation of plasmid digests and in the separation of single from double stranded DNA under 10{sup 4} base pairs. Preliminary work was done on DNA fragments having a size greater than 10{sup 4} base pairs. This work has served to establish the inversion point for DNA.

Abstract

The first topic of this project involved the preparation, fractionation by sedimentation/steric Field Flow Fractionation (FFF), and modeling of metaphase chromosomes. After numerous unsuccessful attempts to prepare chromosomes, we have implemented a procedure (in collaboration with Los Alamos National Laboratory) to prepare metaphase chromosomes from Chinese hamster cells. Extensive experimentation was necessary to identify a suitable FFF channel surface to minimize chromosome adsorption and a carrier liquid to stabilize and disperse the chromosomes. Under suitable operating conditions, the Chinese hamster chromosomes were purified from cell debris and partially fractionated. The purified, preenriched chromosomes that can be prepared by sedimentation/steric FFF or produced continuously by continuous SPLITT fractionation provide an enriched feed material for subsequent flow cytometry. In the second project component, flow FFF permitted successful separations of single- from double-stranded circular DNA, double-stranded circular DNAs of various sizes, and linear double-stranded DNA fragments of various lengths. Diffusion coefficients extracted from retention data agreed well with literature data as well as predictions of major polymer theories. The capacity of FFF separations was evaluated to examine potential applications to long DNA chains.

15. High-speed separation of large (>1 .mu.m) particles by steric field-flow fractionation

Abstract

It is postulated, based on known elements of the steric field-flow fractionation mechanism, that separation speed can be increased without resolution loss by increasing the channel flow rate and simultaneously increasing the sedimentation field strength. The validity of this approach was confirmed in a general way by two series of runs, one with increasing flow at constant field strength (7 g) and one with increasing field strength at constant flow (21 mL/min). The limits of this approach were searched for (but not found) in a special high-flow set of experiments run at 38 mL/min. With a field strength of 343 g, seven different sizes of latex particles (2-45 ..mu..m diameter) were base-line resolved in about 3.5 min from the start-up of the centrifuge and 3.0 min from the initiation of flow.

Abstract

In this paper theoretical and experimental results are obtained relating to the perturbation in retention in sedimentation field-flow fractionation due to particle-wall electrostatic and van der Waals interactions. These perturbations are described in relationship to standard retention theory, an ideal theory whose basic assumptions are summarized. A general equation in integral form is given for retention ratio R, and it is shown how the standard retention theory and sterically corrected retention theory are thereby obtained. Expressions are given for the potential energy of a colloidal particle near a wall resulting from electrostatic and van der Waals interactions; these interactions alter the concentration profile in the field-flow fractionation channel in a way that requires numerical integration to get R. By the use of estimated interaction parameters, R is plotted against field strength and ionic strength for several wall materials including stainless steel and fluorocarbon resin. Experimental results are reported for five different carrier solutions including distilled water. The agreement between theory and experimental results is very good considering the approximate nature of the parameters used. Both the calculations and the measurements show that the retentive perturbations are smaller for fluorocarbon resin than for stainless steel, Hastelloy C, and polyimide surfaces. Anmore » intermediate ionic strength also appears to be optimal. A new separation technique based on the combination of field-flow fractionation and potential barrier chromatography is suggested, and its possible advantages are discussed.« less

Abstract

Coupled column field-flow fractionation (FFF), in which a narrow fraction collected from one run is reinjected for a second FFF run under different conditions, is shown to be a multidimensional technique analogous to coupled column chromatography. Bu using different carrier densities in successive sedimentation FFF stages, this approach is theoretically capable of unraveling both the size and density characteristics of the components of complex colloids. This concept is tested here by preparing simple binary mixtures of polystyrene (PS) and poly(vinyl chloride) (PVC) latex particles; coupled column FFF is then used to recover size and density information on the PS latex despite the presence of the PVC contaminant. The problem of transferring enough of a sample fraction from the first stage to the second to provide an adequate detector signal is approached by three concentration methods: split-outlet concentration, on-channel concentration, and centrifugation. All work reasonably well, but improvements in some of these techniques would greatly simplify the practical usage of coupled column FFF.

Flow/hyperlayer field-flow fractionation (flow/hyperlayer FFF), a recently developed method capable of the rapid separation of particles over 1 μm in diameter according to their size, was applied to the characterization of ground coal and limestone samples. The results of the analysis were found to be consistent and reproducible, in agreement with those obtained from microscopic examination and from the elution profiles of latex particle standards. The results were also in agreement with particle size distributions obtained by laser light scattering. The validity of the size-dependent but density-independent separation by flow/hyperlayer FFF was demonstrated by the comparison of elution profiles for ground coal, ground limestone, glass beads, and polystyrene latex samples, which differ substantially in density. These results show that flow/hyperlayer FFF is a promising tool for the size analysis of ground minerals and similar particulate materials, perhaps even in a process control environment.

Abstract

A new form of field-flow fractionation (FFF) is described in which the field strength is cycled up and down many times during a run. It is shown that this method, termed cyclical FFF (CFFF), leads to migration rates dependent on transport coefficients. After development of the general theory, results are obtained for coherent (no diffusion) square-wave operation. For the latter, migration rates are shown to depend on the generalized mobility of the particles and upon system parameters such as cycle time and field strength. Other variants of CFFF are noted and several advantages are indicated. A gravitational CFFF system operating according to sedimentation coefficients is described and certain programming options are discussed.

Abstract

In spite of recent advances in knowledge concerning the detailed biochemistry of blood coagulation, the diagnosis of haemostatic disturbances remains an important problem of clinical judgement in many instances. Laboratory support relies initially on a series of screening tests designed to investigate the general nature of blood clotting. Recent interest in these aspects is centred on standardization and quality assurance of methods and results. Procedures have been recommended in an attempt to unify data. Several aspects of conventional laboratory investigations have been modified and the reliability of diagnostic information has been improved. Some relatively recent findings have extended the application to coagulation studies. For example, the discovery of protein C, a potent physiological inhibitor of blood coagulation, has clarified the nature of the clotting disorder in some patients with hereditary thrombosis disease. In addition, close analysis of plasma from patients with systemic lupus erythematosus has stimulated interest in the association between the haemostatic, neurological and immunological abnormalities recorded in these patients. More recently, sophisticated techniques for the diagnosis of many coagulation factor defects have been developed. Carrier detection of the sex-linked disorders is undertaken widely with reasonable success and reliable prenatal diagnosis procedures have been established in specialized centres. Unequivocal information regarding the diagnosis of carrier status in some families is obtained by the use of gene analysis and linked polymorphisms. Precise details of the genes for several clotting factors have been recorded. Future development in this field is likely to improve the clinical course of many coagulation disorders.

The levels of procoagulant factor VIII activity in the plasma of four patients with von Willebrand's disease (v Wd) obtained 24 hours after treatment with cryoprecipitate or fresh-frozen plasma were higher than those of factor-VIII-related antigen. Analysis of the plasma by gel chromatography on sepharose 60 showed two peaks of factor-VIII procoagulant activity. A lower-molecular weight peak was not seen in normal plasma or that of treated haemophiliacs. In one patient hitherto diagnosed as v Wd an intermediate pattern of results was observed but this patient was otherwise not typical and may have been suffering from a variant of haemophilia or v Wd. In the patients tested the lower-molecular weight procoagulant factor VIII did not precipitate with rabbit antiserum to factor-VIII-related antigen and a plasma sample which contained excess lower-molecular weight procoagulant activity did not neutralise the precipitating capacity of rabbit antiserum. It did however neutralise the procoagulant inhibitory activity of rabbit antiserum and human inhibitor. The findings are consistent with the hypothesis that “factor VIII” consists of a high-molecular weight unit (HMW) and a lower-molecular weight unit (LMW). It is suggested that HMW, autosomally controlled, gives precipitin reactions with the rabbit antisera and stimulates the formation or release of LMW. It is further suggested that LMW, controlled by the X-chromosome, is complexed to or carried by HMW. The evidence indicates that LMW has procoagulant activity alone or when carried by HMW and the appearance of free LMW in treated v Wd supports the proposed hypothesis.

Abstract

Five different antisera raised against “factor VIII” have been compared with respect to their activity in inhibiting procoagulant factor VIII, in immunofluorescent studies and by immunoelectrophoretic techniques. A method for titrating precipitating antibody to factor-VIII related protein using counter-immuno-electrophoresis is described. Four of the antisera possessed both precipitating antibody to factor VIII related protein and inhibitory activity against procoagulant factor VIII. These antisera reacted by immunofluorescence or immunoprecipitation methods with a factor VIII related antigen or antigens in normal platelets and in the platelets of a patient with severe von Willebrand's disease although antigen could not be detected in the patient's plasma. The other antiserum had specific precipitating activity against factor VIII related protein but it did not inhibit procoagulant factor VIII. It reacted with normal platelets by both immunoprecipitation and immunofluorescent techniques but it did not react with the severe von Willebrand's disease platelets. The results show that although the antisera appeared to contain a single antibody giving reactions of identity when examined by simple immunodiffusion some of them in fact contained antibodies with different characteristics.

Abstract

Plasma samples were obtained from patients of two families with inherited combined deficiency of factor V and factor VIII and examined by coagulation and immunological methods for the presence of factor V and factor VIII related antigens. A deficiency of procoagulant factor V was confirmed in all patients although a factor V related antigen was consistently demonstrated in all plasma samples by an inhibitor neutralisation technique using a non precipitating rabbit antibody. A deficiency of procoagulant factor VIII was also confirmed in all patients and factor VIII related antigens were detected by inhibitor neutralisation using human antibody and by electroimmunoassay using a precipitating rabbit antibody. The amounts of factor V and factor VIII related antigens present in the patient’s plasma samples were similar to those found in normal human plasma. Ristocetin co-factor assay using washed, fixed human platelets was normal in all patients. The results suggest that inactive antigenic determinants of procoagulant factor V and factor VIII are present in the plasma of these patients and are consistent with the possibility that a common precursor of factor V and factor VIII is defective in these families.
